Isodon wightii (Bentham) H. Hara is well known for aromatic essential oil. This is the first study aimed to evaluate antioxidant, antiacetylcholinesterase and apoptotic potential of I. wightii oil. DPPH scavenging, metal ion chelation and antiacetylcholinesterase activities were carried out for the extracted essential oil from the leaves of I. wightii . Cytotoxic effect on HeLa cells was studied by MTT assay and its apoptotic potential was determined by Acridine Orange/Ethidium Bromide and Hoechst33258 stains. A significant free radical scavenging and metal ion chelation were determined for oil. Acetylcholinesterase inhibition was recorded with IC 50 value of 7.864 µg/mL. The cytotoxic effect of oil on HeLa cells was calculated with IC 50 value of 13.264 µg/mL. High level of apoptosed cells were observed at 100 µg/mL oil treated cells. The results of the study emphasise the medicinal importance of I. wightii essential oil which could be a prominent plant based cancer therapeutic agent.

Plastic contamination in aquatic ecosystems all around the world is on the rise. Microplastics (MPs) interfere with the ecological functions of different aquatic animals, including zooplankton. Other contaminants can also be transported by microplastics, resulting in higher toxicity. Here, we evaluated the effect of microplastics alone and in combination with cyanotoxins (CT) on the demographic and feeding responses of the freshwater rotifer Brachionus calyciflorus. The microplastics used were polystyrene spheres of 30 μm diameter. The cyanobacteria were obtained from a sample collected from the Valle de Bravo reservoir in Central Mexico, and the secondary metabolites were obtained after 5 cycles of freezing, thawing and sonication. We conducted acute (LC 50 , 24h bioassay) and chronic toxicity tests involving population growth, life table experiments, and feeding experiments. The LC 50 value using microplastics was 21.91 mg/L, and for the cyanobacterial crude extract it was 2.56 μg/L, but the combination of both resulted in a higher adverse effect (16.64 mg/L) due to the microplastics. Rotifer survival, reproduction, and feeding decreased in the presence of either MPs or CT but the adverse effect was more in the presence of both contaminants. Microplastics (MPs) are becoming increasingly common in freshwater ecosystems. Rotifers may involuntarily ingest MPs through non-selective filtration, thus causing non-real satiety, leading to adverse effects on their demography. Here, we evaluated the acute and chronic effects of MPs (30 µm) on two successive generations of Brachionus calyciflorus s.s. at 20 and 25 °C. Demographic variables and ingestion rates were compared in the absence and presence of microplastics (550 spheres mL -1 ). For the life table experiments, cohorts of twenty neonates were introduced into 20 mL medium with four replicates per treatment. Ingestion rates of the rotifer were tested at both test temperatures in the absence and presence of the microplastics in 20 mL containers with 1 ind. mL -1 per container. The rotifer population growth rates ranged from 0.55 to 1.05 d -1 depending on the treatment. MPs decreased the fecundity by 20-24 %. The average lifespan of rotifers exposed to MPs in the F 0 generation was lower than that in the F 1 generation at both temperatures, indicating that pre-exposure of mothers may offer some protection to the offspring. The population growth rate was higher at 25 °C. The ingestion rates were significantly lower in the presence of the microplastics. The study of fish larval nutrition is important as dietary requirements change significantly with growth. Vieja zonata is a cichlid species that is endemic to Mexico. In this study, we investigated the ingestion rate, prey selectivity, and growth of V. zonata larvae that were fed on prey cultured with and without probiotics. We conducted three experiments to test the acceptability of the prey offered, determine the optimal density of the prey items, and observe the effect of probiotics on the larvae's growth. The first experiment tested the acceptability of the prey offered to 5 days post-hatched (dph) V. zonata larvae, and the second experiment determined the optimal density of the prey items. In the third experiment, we individually placed 5 dph larvae (SL = 5.97 ± 0.13 mm; 8.5 ± 0.25 mg) and fed them for 10 days with three different prey items: two rotifer species ( Brachionus angularis and Plationus patulus at a density of 20 ind/mL) and a cladoceran species ( Moina cf. macrocopa at 1 ind/mL), both cultures with and without probiotics. We counted the prey items consumed daily and provided fresh media with new prey at the above density. We determined the total counts, ingestion rates, and Manly's selectivity index ( α i ) and measured and weighed the larvae at the beginning and end of the experiment. The endogenous feeding period with the yolk sac lasted until 5 dph, a mixed period with endogenous and exogenous feeding occurred from 5 to 7 dph, and an exogenous feeding period in which they fed on zooplankton was observed from day 7 to 15 dph. Brachionus angularis and Plationus patulus were accepted as prey after 5 dph, and Moina cf. macrocopa was accepted on 11 dph. During the first days of feeding, the preferred prey item was P. patulus , which later switched to M . cf. macrocopa on day 11. We found that the use of prey produced with NanoCrusta probiotics resulted in a significant increase in the somatic growth and weight of V. zonata . Plastic debris is thought to be widespread in freshwater ecosystems globally 1 . However, a lack of comprehensive and comparable data makes rigorous assessment of its distribution challenging 2,3 . Here we present a standardized cross-national survey that assesses the abundance and type of plastic debris (>250 μm) in freshwater ecosystems. We sample surface waters of 38 lakes and reservoirs, distributed across gradients of geographical position and limnological attributes, with the aim to identify factors associated with an increased observation of plastics. We find plastic debris in all studied lakes and reservoirs, suggesting that these ecosystems play a key role in the plastic-pollution cycle. Our results indicate that two types of lakes are particularly vulnerable to plastic contamination: lakes and reservoirs in densely populated and urbanized areas and large lakes and reservoirs with elevated deposition areas, long water-retention times and high levels of anthropogenic influence. Plastic concentrations vary widely among lakes; in the most polluted, concentrations reach or even exceed those reported in the subtropical oceanic gyres, marine areas collecting large amounts of debris 4 . Cyanobacterial blooms have been recognized as a problem in fresh water for about 150 years. Over the past 50 years, experimental studies on the subject have gained importance considering the increasing need to control toxic cyanobacterial blooms. This article presents information on the different lines of research that have been undertaken on zooplankton-cyanobacteria interactions over the past 50 years. These include information on filtering/ingestion rates and phytoplankton preferences of small and large rotifers, cladocerans, and copepods; growth rates of zooplankton on cyanobacterial diets; feeding rates of other freshwater invertebrates on cyanobacteria; role of zooplankton in top-down biomanipulation efforts; effect of cyanotoxins on zooplankton; bioaccumulation of cyanotoxins; and physical and chemical control of cyanobacterial blooms. We also highlight measures that have led to successful lake management and improvement of water quality in selected waterbodies.

Background and Aim: Complete removal of gutta-percha and sealer is an important step during retreatment. The aim of this study was to assess the efficacy and time taken by full counterclockwise rotary motion of a reciprocating file in the removal of gutta-percha and BioRoot™ RCS bioceramic sealer., Materials and Methods: Thirty-six extracted human mandibular premolars were instrumented with Mtwo rotary files and obturated using lateral condensation technique with gutta-percha and BioRoot™ RCS bioceramic sealer. The teeth were divided into three retreatment groups ( n = 12) based on the kinematics used: Group I, Mtwo retreatment (Mtwo-R) files; Group II, Reciproc files used in reciprocation mode (Reciproc-R); and Group III, Reciproc files used in full counterclockwise motion (Reciproc-CCW). The efficacy of gutta-percha and bioceramic sealer removal was assessed by volume of gutta-percha removed using cone-beam computed tomography and area of remaining gutta-percha and bioceramic sealer using ImageJ software after longitudinal sectioning. Statistical analysis was done by one-way analysis of variance test and Tukey's multiple post hoc tests., Results: Reciproc-CCW motion was significantly efficient in the removal of gutta-percha and bioceramic sealer in comparison to Mtwo-R files and Reciproc-R. The Aeromonads are ubiquitous Gram-negative bacilli that cause community acquired, and healthcare associated infections. In this retrospective study we analysed clinical and microbiological characteristics of thirty-six culture proven Aeromonas infections. The most common species isolated was A.hydrophila. Clinical presentation included syndromes like skin and soft tissue infections (SSTI), urinary tract infections, and central line associated bloodstream infections (CLABSI). Most of the isolates were sensitive to aminoglycosides [97.2%], followed by 3 rd generation cephalosporins, quinolones and carbapenems. Overall mortality was 13.88% (5 out of 36 patients). Brain tumor classification is a very important and the most prominent step for assessing life-threatening abnormal tissues and providing an efficient treatment in patient recovery. To identify pathological conditions in the brain, there exist various medical imaging technologies. Magnetic Resonance Imaging (MRI) is extensively used in medical imaging due to its excellent image quality and independence from ionizing radiations. The significance of deep learning, a subset of artificial intelligence in the area of medical diagnosis applications, has macadamized the path in rapid developments for brain tumor detection from MRI to higher prediction rate. For brain tumor analysis and classification, the convolution neural network (CNN) is the most extensive and widely used deep learning algorithm. In this work, we present a comparative performance analysis of transfer learning-based CNN-pretrained VGG-16, ResNet-50, and Inception-v3 models for automatic prediction of tumor cells in the brain. Pretrained models are demonstrated on the MRI brain tumor images dataset consisting of 233 images. Our paper aims to locate brain tumors with the utilization of the VGG-16 pretrained CNN model. The performance of our model will be evaluated on accuracy. Background: Tuberculosis is a chronic infectious disease that affects one-third of the global population. The emergence of Multi-resistant (MDR) strains and high susceptibility of human immunodeficiency virus (HIV) infected persons to the disease forced to develop novel antituberculosis agents and preferably have a novel mechanism of action as to avoid cross-resistant with other agents. A literature survey indicated that Pyridine, Thiadiazole, Benzimidazole and Acetyl thiophene derivatives exhibit various pharmacological activities, including anti-mycobacterial activity., Methods: Thus, a series of Pyridine, Thiadiazole, Benzimidazole and Acetyl thiophene based molecules were designed and docked against crucial mtb enzyme target InhA (Enoyl Acyl Carrier Protein Reductase) Enzyme. The docked molecules were screened against good docking-score and multiple interactions and opted for synthesis. Synthesized molecules were recrystallized to obtain purity. All the purified compounds were characterized by various spectral analyses and evaluated for antimycobacterial activity against tuberculosis H37RV strain by Microplate Alamar Blue Assay (MABA) method., Results: The experimental results showed that Schiff bases of Pyridine (Compounds 'd') and Benzimidazole derivatives (Compounds 'i' ) possess good anti-tubercular activity with an MIC below 1.6 μg /mL. The emerging contaminant 4-nonylphenol is an active ingredient in many surfactants. Though 4-nonylphenol has been reported in Mexican waterbodies, no stringent law is available to establish its permissible limits. Most ecotoxicological assays using zooplankton species are based on previously unexposed populations, and multigenerational experiments are rare. Plationus patulus and Brachionus havanaensis are widely distributed species in Mexico and have been used in ecotoxicological assessments. In this work, the median lethal concentration of 4-nonylphenol (LC 50 , 24 h) for both rotifer species was derived. Based on our acute toxicity data (24 h LC 50 of 4-nonylphenol for P. patulus and B. havanaensis were 500 and 250 µg L -1 , respectively), three sublethal concentrations (µg L -1 ) (7.81, 15.82 and 31.25 for B. havanaensis and 15.62, 31.25 and 62.5 for P. patulus ) were used in the life table demography tests for two successive generations (F0 and F1) of either rotifer species. Regardless of the concentrations used, 4-nonylphenol had an adverse effect on both rotifer species over two successive generations. P. patulus was more sensitive than B. havanaensis in chronic toxicity tests. F1 generation of both rotifer species was far more adversely affected than the F0 in all toxicant treatments.

Diurnal temperature fluctuations affect ectothermic species more than endothermic taxa. We tested the effect of three fixed temperatures (20, 25 and 30 °C) and a 24 h variable (20-30 °C) on the population growth and fatty acid profiles of the common rotifer Brachionus calyciflorus. Depending on the temperature treatment, the peak population abundances of B. calyciflorus varied from 65 to 80 ind./ml, the lowest being on variable temperature range. The rate of population increase varied from 0.31 to 0.52 per day, highest being at 30 °C. There was a curvilinear relationship between the population density and the egg ratio (number of eggs/female) in all the tested temperature regimes. The egg ratio was higher (>0.6) for treatments involving fixed temperatures, but for variable temperature regime, the egg ratios were lower (<0.5). Temperature also induced changes in the fatty acid content of B. calyciflorus. While the total saturated fatty acids increased, both mono-unsaturated and poly-unsaturated fatty acids decreased with increasing temperature regime. Many urban lakes in Mexico City such as Lake Chapultepec are infested with high densities of cyanobacteria, particularly Microcystis. We tested the effect of cyanotoxins from cyanobacterial crude extracts on the demographic variables of zooplankton. The rotifers Brachionus havanaensis and Brachionus calyciflorus, and the cladocerans Ceriodaphnia dubia and Moina macrocopa were used for the assays. Temperature effects on the response of B. calyciflorus and 2 clones of M. macrocopa were tested. We hypothesized that with an increase in cyanotoxin concentration and temperature there would be an increase in the adverse effect on the test species and that the clone of Moina previously exposed to cyanobacteria from Lake Chapultepec would be more resistant to the cyanotoxins. Demography experiments showed that B. havanaensis was more sensitive than C. dubia. The negative effect of the cyanobacterial crude extract on B. calyciflorus was greater at 30 °C than at 20 °C or 25 °C. The strain of M. macrocopa isolated from Lake Chapultepec was more resistant to the cyanotoxins than the strain that had not been previously exposed to the cyanobacteria. Several abiotic factors influence the ecological responses of aquatic invertebrates to metal toxicity. We examined the effect of salinity (10, 20, and 30 psu) and temperature (25 and 32 °C) on acute and chronic arsenic (As) toxicity to the euryhaline rotifers, Proales similis and Brachionus ibericus. In general, higher salinities and low temperature resulted in lower arsenic toxicity. The population growth studies indicated that P. similis was more sensitive than B. ibericus to As. Arsenic toxicity intensified the vulnerability of P. similis to B. ibericus competition. Life table parameters decreased with increasing As levels in the medium. Chronic toxicity bioassays were more sensitive than acute toxicity tests for determining the adverse effect of As to rotifers. Our findings provide useful insights on the effect of arsenic on rotifer populations exposed to different temperature and salinity scenarios. Proliferating cyanobacterial blooms due eutrophication in reservoirs is a major global problem. The production of cyanotoxins often increases with grazing pressure and temperature while the sensitivity of zooplankton to cyanotoxins is directly related to temperature. Here we evaluate the effect of different concentrations of the crude extract of cyanobacteria from Valle de Bravo reservoir during dry (January) and rainy (September) seasons at 20 and 25 °C on the rotifer Brachionus calyciflorus based on acute and chronic toxicity tests. We filtered 20 or 150 l of lake water, depending on the intensity of the bloom, and estimated the density and diversity of the cyanobacteria. The crude extracts, after 5 cycles of freezing, thawing and sonication at 14 MHz, were filtered and the microcystin concentration quantified based on ELISA. The extracts were used to conduct the acute and chronic toxicity tests, all in quadruplicate. Acute toxicity tests were based on 24 h mortality. Chronic toxicity tests (population growth and life table experiments) were conducted at 5 and 10% of the median lethal concentration. The field samples were dominated by Microcystis sp. (January) or Woronichinia naegeliana (September). The microcystin concentration in lake water was 9.57 μg/l and 0.097 μg/l and the median lethal concentration was 5.34 μg microcystin/L and 0.35 μg microcystin/L in January and September, respectively. Survival and reproduction of B. calyciflorus were lower in the presence of the cyanobacteria crude extract, more so at 20° than at 25 °C. Epilepsy is a neurological disorder characterized by an enduring predisposition to generate and aggravate epileptic seizures affecting around 1% of global population making it a serious health concern. Despite the recent advances in epilepsy research, no disease-modifying treatment able to terminate epileptogenesis have been reported yet reflecting the complexity in understanding the disease pathogenesis. To overcome the current treatment gap against epilepsy, one effective approach is to explore anti-epileptic effects from a drug that are approved to treat non-epileptic diseases. In this regard, Metformin emerged as an ideal candidate which is a first line treatment option for type 2 diabetes mellitus (T2DM), has conferred neuroprotection in several in vivo neurological disorders such as Alzheimer's diseases (AD), Parkinson's disease (PD), Stroke, Huntington's diseases (HD) including epilepsy. In addition, Metformin has ameliorated cognitive alteration, learning and memory induced by epilepsy as well as in animal model of AD. Herein, we review the promising findings demonstrated upon Metformin treatment against animal model of epilepsy however, the precise underlying mechanism of anti-epileptic potential of Metformin is not well understood. However, there is a growing understanding that Metformin demonstrates its anti-epileptic effect mainly via ameliorating brain oxidative damage, activation of AMPK, inhibition of mTOR pathway, downregulation of α-synuclein, reducing apoptosis, downregulation of BDNF and TrkB level. These reflects that Metformin being non-anti-epileptic drug (AED) has a potential to ameliorate the cellular pathways that were impaired in epilepsy reflecting its therapeutical potential against epileptic seizure that might plausibly overcome the limitations of today epilepsy treatment., (Copyright © 2019 Elsevier Inc. Charcot-Marie-Tooth disease (CMT) is the most common peripheral neuromuscular disorder worldwide. The axonal degeneration in CMT causes distal muscle weakness and atrophy, resulting in gait problems and difficulties with basic motor coordination skills. A mutation in the cytoplasmic dynein heavy chain (DHC) gene was discovered to cause an autosomal dominant form of the disease designated Charcot-Marie-Tooth type 2O disease (CMT2O) in 2011. The mutation is a single amino acid change of histidine into arginine at amino acid 306 (H306R) in DHC. We previously generated a knock-in mouse carrying the corresponding CMT2O mutation (H304R) and examined the heterozygous H304R/+offspring in a variety of motor skills and histological assays. Here we report the initial characterization of the homozygous H304R/R mouse, which is the first homozygous mutant DHC mouse to survive past the neonatal stage. We show that H304R/R mice have significantly more severe disease symptoms than the heterozygous H304R/+mice. The H304R/R mice have significant defects in motor skills, including grip strength, motor coordination, and gait and also related defects in neuromuscular junction architecture. Furthermore, the mice have defects in sensation, another aspect of CMT disease. Our results show that the H304R/+ and H304R/R mice will be important models for studying the onset and progression of both heterozygous and homozygous CMT disease alleles.

Cyanotoxins from toxic blooms in lakes or eutrophic reservoirs are harmful to several organisms including zooplankton, which often act as vectors of these secondary metabolites, because they consume cyanobacteria, bioaccumulate the cyanotoxins and pass them on along the food chain. Microcystins are among the most commonly found cyanotoxins and often cause zooplankton mortality. Although cyanobacterial blooms are common and persistent in Mexican water bodies, information on the bioaccumulation of cyanotoxins is scarce. In this study we present data on the bioaccumulation of cyanotoxins from Planktothrix agardhii, Microcystis sp., Cylindrospermopsis raciborskii and Dolichospermum planctonicum blooms in the seston (suspended particulate matter more than 1.2 μm) by zooplankton and fish (tilapia (Oreochromis niloticus) and mesa silverside (Chirostoma jordani) samples from Lake Zumpango (Mexico City). The cyanotoxins were extracted from the seston, zooplankton and fish tissue by disintegration using mechanical homogenization and 75% methanol. After extraction, microcystins were measured using an ELISA kit (Envirologix). Concentration of microcystins expressed as equivalents, reached a maximum value of 117 μg g -1 on sestonic samples; in zooplankton they were in the range of 0.0070-0.29 μg g -1 . The dominant zooplankton taxa included Acanthocyclops americanus copepodites, Daphnia laevis and Bosmina longirostris. Our results indicate twice the permissible limits of microcystins (0.04 μg kg -1 d -1 ) for consumption of cyanobacterial products in whole fish tissue of Chirostoma jordani. Cladocerans are constantly exposed to humic substances in nature, yet the effects of these substances on their survival and reproduction are not well known. Here, the effects of humic substances (20 and 40 mg L -1 ) (HS) on the life history variables of three common cladocerans, Ceriodaphnia dubia , Moina macrocopa , and Daphnia pulex were evaluated. The results showed that the effect of humic substances on the tested cladocerans is species-specific, affecting either survival, reproduction or both. For M. macrocopa , exposed to HS at a concentration of 40 mg L -1 , the average lifespan and the life expectancy at birth were significantly reduced as compared to controls, but for C. dubia and D. pulex these parameters were increased. Gross reproductive rate was unaffected by the HS level for both D. pulex and M. macrocopa , but it was significantly higher for C. dubia . When compared to the corresponding controls, for HS-exposed cladocerans, the rate of population increase was significantly reduced in case of D. pulex while it was stimulated for both C. dubia and M. macrocopa . It appears that humic substances had a slightly stronger influence on survivorship than on reproduction of the tested cladocerans.

The demographic response of the brackish-water rotifers Proales similis and Brachionus plicatilis to mercury (0.5, 2, 8 and 32 μg L -1 of HgCl 2 ) at different salinity levels (10 and 20‰) and two temperature (25 °C and 32 °C) regimes were evaluated. Median lethal concentration (LC 50 ) for P. similis and B. plicatilis was 10 and 16 μg L -1 , respectively, showing that Proales similis was more sensitive to mercury than B. plicatilis. The rate of population increase (r) for both species was greater at 10‰ salinity and 32 °C (ranged from 0.6 to 0.95 d -1 ). The r-value decreased as the concentration of mercury in the medium increased. Regardless of the temperature, at lower salinity and higher mercury concentration (32 μg L -1 ), P. similis died within six days. The survivorship of P. similis and B. plicatilis was higher at 25 °C than at 32 °C (ranged from 5 to 8 and 7-13 d, respectively). Fecundity was higher at 32 °C than at 25 °C for both rotifers species. There was a significant effect of the interaction among salinity, temperature, and mercury in both species on the reproductive variables such as net and gross reproductive rates, generation time and the rate of population increase. Aberrant hedgehog (Hh) signaling contributes to the pathogenesis of multiple cancers. Available inhibitors target Smoothened (Smo), which can acquire mutations causing drug resistance. Thus, compounds that inhibit Hh signaling downstream of Smo are urgently needed. We identified dynarrestin, a novel inhibitor of cytoplasmic dyneins 1 and 2. Dynarrestin acts reversibly to inhibit cytoplasmic dynein 1-dependent microtubule binding and motility in vitro without affecting ATP hydrolysis. It rapidly and reversibly inhibits endosome movement in living cells and perturbs mitosis by inducing spindle misorientation and pseudoprometaphase delay. Dynarrestin reversibly inhibits cytoplasmic dynein 2-dependent intraflagellar transport (IFT) of the cargo IFT88 and flux of Smo within cilia without interfering with ciliogenesis and suppresses Hh-dependent proliferation of neuronal precursors and tumor cells. As such, dynarrestin is a valuable tool for probing cytoplasmic dynein-dependent cellular processes and a promising compound for medicinal chemistry programs aimed at development of anti-cancer drugs., (Copyright © 2018 Elsevier Ltd. Charcot-Marie-Tooth disease (CMT) is a peripheral neuromuscular disorder in which axonal degeneration causes progressive loss of motor and sensory nerve function. The loss of motor nerve function leads to distal muscle weakness and atrophy, resulting in gait problems and difficulties with walking, running, and balance. A mutation in the cytoplasmic dynein heavy chain (DHC) gene was discovered to cause an autosomal dominant form of the disease designated Charcot-Marie-Tooth type 2 O disease (CMT2O) in 2011. The mutation is a single amino acid change of histidine into arginine at amino acid 306 (H306R) in DHC. In order to understand the onset and progression of CMT2, we generated a knock-in mouse carrying the corresponding CMT2O mutation (H304R/+). We examined H304R/+ mouse cohorts in a 12-month longitudinal study of grip strength, tail suspension, and rotarod assays. H304R/+ mice displayed distal muscle weakness and loss of motor coordination phenotypes consistent with those of individuals with CMT2. Analysis of the gastrocnemius of H304R/+ male mice showed prominent defects in neuromuscular junction (NMJ) morphology including reduced size, branching, and complexity. Based on these results, the H304R/+ mouse will be an important model for uncovering functions of dynein in complex organisms, especially related to CMT onset and progression.

Triclosan is a personal care product widely used in North America, Europe and Asia as antimicrobial ingredient in many consumer chemical products. In Mexico concentrations of triclosan have been reported in aquatic systems. However, there is no law regulating the presence of chemicals such as triclosan, in aquatic systems. The scarce data about this chemical has increased concern among ecotoxicologists regarding possible effects on aquatic organisms. Moreover, multigenerational studies are rarely studied and the results vary depending on the contaminant. Rotifers, are a dominant group of zooplankton, and have been used in aquatic risk assessments of personal care products due to their sensitivity and high reproductive rates. Plationus patulus and Brachionus havanaensis are common rotifers distributed in aquatic ecosystems of Mexico and have been used in ecotoxicological bioassays. In this study, the median lethal concentration (LC50, 24h) of P. patulus and B. havanaensis exposed to triclosan was determined. Based on the LC50, we tested three sublethal concentrations of triclosan to quantify the demographic responses of both rotifers for two successive generations (F0, and F1). The 24h LC50 of triclosan for P. patulus and B. havanaensis were 300 and 500µgL -1 respectively. Despite the concentration, triclosan had an adverse effect on both Plationus patulus and Brachionus havanaensis in both generations exposed. Experiments show that P. patulus was more sensitive than B. havanaensis when exposed to triclosan. Unlike temperate regions, tropical ecosystems are characterized by high temperatures (>18 °C) all year, promoting blooms of cyanobacteria which often produce secondary metabolites toxic to zooplankton. Nabor Carillo and the Recreational Lake are part of the saline, Lake Texcoco, in Central Mexico which is filled nowadays with treated waste water. Both water bodies are dominated by Planktothrix, Anabaenopsis, Spirulina and Microcystis. In this study we present the concentration of microcystins in these waterbodies over an annual cycle. We also evaluated the chronic effects of cyanobacterial crude extracts from both lakes on two clones of the rotifer Brachionus calyciflorus, one from Nabor Carrillo Lake and the other from a canal in the shallow, Lake Xochimilco. The experiments on population growth were performed, beginning with 10 individuals per container for each of the following treatments: control (no crude extract), concentrated crude extract, and diluted crude extract (50:50) with moderately hard water and Chlorella vulgaris in a concentration of 0.5 × 10 6  cells ml -1 . The cyanotoxin levels were measured using an ELISA test and ranged between 0.20 and 2.4 μg L -1 in the lake water. The results showed that the Recreational Lake extracts were more toxic, killing the rotifers in less than five days. The r values ranged from -1.74 to 0.48 in the presence of the crude extracts and 0.16 and 0.24 in the controls. In this study, we quantified intergenerational, demographic variability of Moina macrocopa subjected to cadmium stress. Exposure of M macrocopa to cadmium (0.2, 0.3, and 0.4 mg/L as CdCl 2 ) through 4 consecutive generations revealed changes in demographic responses not only in survivorship variables but also in reproductive parameters. Long-term demographic responses varied differently, depending on the demographic trait and the concentration of heavy metal in the medium. With the exception of generation time, all life history traits were significantly and adversely influenced due to increase in Cd concentrations. The average life span of M macrocopa varied up to 40% depending on Cd level and the generation of exposure. The highest gross reproductive rates were recorded in controls, while the lowest (∼30% less) were recorded at the highest Cd level. Survival-weighted net reproductive rates were reduced by nearly 50% due to Cd toxicity. The rate of population increase per day of M macrocopa was also significantly affected (∼25%) by Cd as compared to controls. Aluminum (Al) use has increased greatly during the last two decades, yet little information is available on its toxic effects in relation to pH particularly on zooplankton. In this work, we determined the acute toxicity (LC 50 ) and life table responses for Moina micrura exposed to 0.008, 0.016 and 0.08 mg of Al L -1 at pH of 5, 6 and 7. The age-specific survivorship and reproduction showed a steep decline (80% mortality by the second day) at pH 5, independent of Al level. Both gross and net reproductive rates were significantly lower at pH 6 compared to pH 7, regardless of Al concentration. At pH 7 the rate of population increase of M. micrura was not significantly influenced by the Al level, while at pH 6 it was significantly lower (p < 0.05), suggesting that M. micrura is sensitive to changes in Al under slightly acidic conditions.

Objective: The purpose of this study was to assess the role of cross section on cyclic fatigue resistance of One Shape, Revo-S SU, and Mtwo rotary files in continuous rotation and reciprocating motion in dynamic testing model., Materials and Methods: A total of 90 new rotary One Shape, Revo-S SU, and Mtwo files (ISO size 25, taper 0.06, length 25 mm) were subjected to continuous rotation or reciprocating motion. A cyclic fatigue testing device was fabricated with 60° angle of curvature and 5 mm radius. The dynamic testing of these files was performed using an electric motor which permitted the reproduction of pecking motion. All instruments were rotated or reciprocated until fracture occurred. The time taken for each instrument to fracture was recorded. All the fractured files were analyzed under a scanning electron microscope (SEM) to detect the mode of fracture. Statistical analysis was performed using one-way ANOVA, followed by Tukey's honestly significant difference post hoc test., Results: The time taken for instruments in reciprocating motion to fail under cyclic loading was significantly longer when compared with groups in continuous rotary motion. There was a statistically significant difference between Mtwo rotary and the other two groups in both continuous and reciprocating motion. One Shape rotary files recorded significantly longer duration to fracture resistance when compared with Revo-S SU files in both continuous and reciprocating motion. SEM observations showed that the instruments of all groups had undergone a ductile mode of fracture., Conclusion: Reciprocating motion improved the cyclic fatigue resistance of all tested groups., Competing Interests: There are no conflicts of interest.

This manuscript reports a new approach for the synthesis of one dimensional gold nanostructure (AuNs) and its application in the development of cholesterol biosensor. Au nanostructures have been synthesized by exploiting β-diphenylalanine (β-FF) as an sacrificial template, whereas the Au nanoparticles (AuNPs) were synthesized by ultrasound irradiation. X-ray diffractometer (XRD), scanning electron microscope (SEM) and energy dispersive analysis of X-rays (EDAX) have been employed to characterize the morphology and composition of the prepared samples. With the aim to develop a highly sensitive cholesterol biosensor, cholesterol oxidase (ChOx) was immobilized on AuNs which were appended on the graphite (Gr) electrode via chemisorption onto thiol-functionalized graphene oxide (GO-SH). This Gr/GO-SH/AuNs/ChOx biosensor has been characterized using cyclic voltammetry (CV), electrochemical impedance spectroscopy and chronoamperometry. CV results indicated a direct electron transfer between the enzyme and the electrode surface. A new potentiostat intermitant titration technique (PITT) has been studied to determine the diffusion coefficient and maxima potential value. The proposed biosensor showed rapid response, high sensitivity, wide linear range and low detection limit. Furthermore, our AuNs modified electrode showed excellent selectivity, repeatability, reproducibility and long term stability. Demographic responses and fatty acid profiles of Moina macrocopa were quantified under different temperature regimes (20°C, 25°C and 30°C and diurnally variable 20-30°C) and at fixed ration (10.65µgDWml(-1)) of Chlorella. Highest constant temperature (30°C) reduced the density of M. macrocopa. The cladocerans under the fluctuating temperature regime too had lower population growth (about 50% lower than that at constant 25°C). The survivorship of M. macrocopa was higher at 20°C than that at 25°C and 30°C or at variable temperature regime. Gross and net reproductive rates were higher at 25°C. At 20°C, neonates had the highest proportion (67%) of myristic, palmitic and stearic acids while the adults had the lowest (26%) proportion. For both adults and neonates, palmitoleic, linoleic and linolenic comprised of 15-35% of the total fatty acids. Higher percentage (19%) of linoleic acid was present in adults than neonates (7%). Adults had linolenic acid level which was 3-times higher than in neonates. Linoleic and linolenic fatty acids decreased with increasing temperature for neonates and adults from 20°C to 30°C. In this study we evaluated the toxicological effects of crude extracts of Dolichospermum planctonicum on Plationus patulus (Rotifera) and Ceriodaphnia cornuta (Cladocera). The active compound from the laboratory-cultured cyanobacteria was extracted by lyophilizing it at -70 °C for 48 h, after which the sample was thawed. The cycle was repeated five times, and between each cycle the cells were sonicated to lyse them. Using the extract of Anabaena, experiments of population growth and life table demography of the two zooplankton species were compared in media with and without (controls) the cyanotoxins. In both experiments two toxic concentrations diluted in US EPA medium were used, one in a 5:15 proportion, and the other in a 2.5:17.5. We found that the rotifers were more resistant to the toxicants than the cladocerans. Their population growth rates ranged from 0.11 d(-1) in controls to 0.08 d(-1) in the presence of the crude extracts for P. patulus and from 0.18 in controls to -0.27 in the presence of the crude extracts for C. cornuta. The crude extract also had a significant negative impact on the demographic parameters, adversely affecting reproduction more than survival. The data have been discussed with emphasis on the importance of further research using cyanobacterial crude extracts.

In this paper, a simple and innovative electrochemical hydrogen peroxide biosensor has been proposed using catalase (CATpp) derived from Pichia pastoris as bioelectrocatalyst. The model biocomponent was immobilized on gold nanoparticle nanotubes (AuNPNTs) and polythiophene composite using 1-ethyl-3-(3-dimethylaminopropyl)-carbodiimide and N-hydroxysuccinimide (EDC-NHS) coupling reagent. In this present work, we have successfully synthesized gold nanoparticles (AuNPs) by ultrasonic irradiation. The tubular gold nanostructures containing coalesced AuNPs were obtained by sacrificial template synthesis. The assembly of AuNPNTs onto the graphite (Gr) electrode was achieved via S-Au chemisorption. The latter was pre-coated with electropolymerized thiophene (PTh) to enable S groups to bind AuNPNTs. The combination of AuNPNTs-PTh, i.e., an inorganic-organic hybrid, provides a stable enzyme immobilization platform. The physical morphology of the fabricated biosensor Gr/PTh/AuNPNTs/EDC-NHS/CATpp was investigated using scanning electron microscopy and energy-dispersive microscopy. The analytical performance of the bioelectrode was examined using cyclic voltammetry, differential pulse voltammetry and chronoamperometry. Operational parameters such as working potential, pH, and thermal stability of the modified electrode were examined. The beneficial analytical characteristics of the proposed electrode were demonstrated. Our results indicate that the Gr/PTh/AuNPNTs/EDC-NHS/CATpp bioelectrode exhibits a wide linear range from 0.05 mM to 18.5 mM of H2O2, fast response time of 7 s, excellent sensitivity of 26.2 mA mM(-1) cm(-2), good detection limit of 0.12 μM and good Michaelis-Menten constant of 1.4 mM. In addition, the bioelectrode displayed good repeatability, high stability and acceptable reproducibility, which can be attributed to the AuNPNTs-PTh composite that provides a biocompatible micro-environment.

Microcystis aeruginosa is generally dominant in many Mexican freshwater ecosystems interacting with zooplankton species. Hence, feeding and filtration rates were quantified for three cladoceran (Daphnia pulex, Moina micrura and Ceriodaphnia dubia) and three rotifer species (Brachionus calyciflorus, Brachionus rubens and Plationus patulus) using sonicated M. aeruginosa alone or mixed with Scenedesmus acutus in different proportions (25, 50 and 75%, based on cell density), offering a combined initial density of 100,000 cells·ml(-1). All the three cladoceran species ingested M. aeruginosa (100-300 cells ind(-1) min(-1)) when fed exclusively with cyanobacterium. When green alga offered as exclusive diet, the number of cells ingested by the tested cladocerans varied from 80 to 400 cells ind(-1) min(-1). Compared to cladocerans, rotifers in general consumed much lower quantity (< 200 cells ind(-1) min(-1)) of M. aeruginosa and S. acutus. The filtration rate for Daphnia pulex was inversely related to the proportion of green alga in the diet. For other tested cladocerans, no such clear trend was evident. In mixed treatments containing M. aeruginosa, the filtration rate of Daphnia was highest (about 220 μl ind(-1) min(-1)) when the medium contained 75% of S. acutus. Among the rotifer species, P. patulus filtered highest volume (100 μl ind(-1) min(-1) from mixed diets containing higher proportions (50 or 75%) of M. aeruginosa. Thus, there were species-specific differences in the filtration and feeding rates of zooplankton when offered mixed diets of green algae and toxic cyanobacteria. These probably explain the coexistence of different zooplankton species in Microcystis-dominant waterbodies.

Introduction: The purpose of this study was to evaluate and compare the cyclic fatigue resistance of RaCe (FKG Dentaire, La Chaux-de-Fonds, Switzerland) and Mtwo (VDW, Munich, Germany) rotary files in continuous rotation and reciprocating motion., Methods: A total of 60 new rotary Mtwo and RaCe files (ISO size = 25, taper = 0.06, length = 25 mm) were selected and randomly divided into 4 groups (n = 15 each): Mtc (Mtwo NiTi files in continuous rotation), Rc (RaCe NiTi files in continuous rotation), Mtr (Mtwo NiTi files in reciprocating motion), and Rr (RaCe NiTi files in reciprocating motion). A cyclic fatigue testing device was fabricated with a 60° angle of curvature and a 5-mm radius. All instruments were rotated or reciprocated until fracture occurred. The time taken for each instrument to fracture and the length of the broken fragments were recorded. All the fractured files were analyzed under a scanning electron microscope to detect the mode of fracture. The Kolmogorov-Smirnov test was used to assess the normality of samples distribution, and statistical analysis was performed using the independent sample t test., Results: The time taken for the instruments of the Mtr and Rr groups to fail under cyclic loading was significantly longer compared with the Mtc and Rc groups (P < .001). Scanning electron microscopic observations showed that the instruments of all groups had undergone a ductile mode of fracture. The length of the fractured segments was between 5 and 6 mm, which was not statistically significant among the experimental groups., Conclusions: Mtwo and RaCe rotary instruments showed a significantly higher cyclic fatigue resistance in reciprocating motion compared with continuous rotation motion., (Copyright © 2014 American Association of Endodontists. This article presents a case of a palatogingival groove (PPG) in tooth No. 12 that was misdiagnosed as a mutilated root fracture. A PPG is a developmental anomaly that predisposes a tooth to a severe periodontal defect. The initial diagnosis of mutilated root fracture on tooth No. 12 was made solely from radiographs that showed a vertical and horizontal radiolucent line with a radiolucent halo-shaped bone resorption in relation to the root apex. Transillumination, bite test, surgical visualization of the site, and the use of high-end diagnostic modalitieswere not considered prior to the final diagnosis. PPG was confirmed only after the tooth was extracted and observed under a dental operating microscope.

Objective: The aim of this study was to assess the efficacy of apical negative pressure (ANP), manual dynamic agitation (MDA), passive ultrasonic irrigation (PUI) and needle irrigation (NI) as final irrigation activation techniques for smear layer removal in curved root canals., Materials and Methods: Mesiobuccal root canals of 80 freshly extracted maxillary first molars with curvatures ranging between 25° and 35° were used. A glide path with #08-15 K files was established before cleaning and shaping with Mtwo rotary instruments (VDW, Munich, Germany) up to size 35/0.04 taper. During instrumentation, 1 ml of 2.5% NaOCl was used at each change of file. Samples were divided into 4 equal groups (n=20) according to the final irrigation activation technique: group 1, apical negative pressure (ANP) (EndoVac); group 2, manual dynamic agitation (MDA); group 3, passive ultrasonic irrigation (PUI); and group 4, needle irrigation (NI). Root canals were split longitudinally and subjected to scanning electron microscopy. The presence of smear layer at coronal, middle and apical levels was evaluated by superimposing 300-μm square grid over the obtained photomicrographs using a four-score scale with X1,000 magnification., Results: Amongst all the groups tested, ANP showed the overall best smear layer removal efficacy (p < 0.05). Removal of smear layer was least effective with the NI technique., Conclusion: ANP (EndoVac system) can be used as the final irrigation activation technique for effective smear layer removal in curved root canals.

Two non-steroidal anti-inflammatory commonly used in Mexico are paracetamol and diclofenac. These drugs reach natural waterbodies transport through the hydrological areas of agriculture and effluent treatment plants and can affect plankton. In this study, we quantified the chronic effects of paracetamol and sodium diclofenac on the population dynamics of the rotifer Plationus patulus and the cladoceran Moina macrocopa under laboratory conditions. Both these species of zooplankton are common in freshwaters and are sensitive to chemicals in the medium. Our results showed that densities of both P. patulus and M. macrocopa decreased with increasing levels of both the analgesic drugs. Both zooplankton species did not survive beyond when paracetamol was applied at 32 mg I(-1) in the medium. Diclofenac in general had more adverse effect than paracetamol for either zooplankton species. P. patulus was more sensitive than M. macrocopa to both analgesic drugs. When diclofenac was present in the medium at > or = 12.5 mg I(-1), rotifer reproduction was inhibited, while the tested cladocerans continued to grow but to lower densities compared to control. The rate of population increase (r) per day of P. patulus and M. macrocopa was significantly and inversely related to the concentration of paracetamol and diclofenac in the medium. However, the relationship between r and drug concentration differed depending on the zooplankton species and drug. In controls, the r of P. patulus was 0.18 d(-1), for M. macrocopa under similar conditions, it was slightly lower (0.16 d(-1)). The rvalues of both zooplankton populations became negative (-0.10 to -0.15 d(-1)) when exposed to paracetamol at 32 mg l(-1) or diclofenac at 25 mg I(-1).

Malathion is a common pesticide used to control insects in agricultural, domestic and industrial sectors in different parts of the world. In this work we evaluated the effects of sublethal concentrations of malathion on the survivorship and reproductive variables of two cladoceran species Daphnia pulex and Diaphanosoma birgei using standard life table demography method. Based on preliminary tests, we selected four sublethal concentrations of malathion for each cladoceran species. For D. pulex, the malathion concentrations were 0. 225, 0.45, 0.9 and 1.8ng I(-1) and for D. birgei, these were 0.0281, 0.0562, 0.1125, 0.225ng I(-1). Our results showed that in general, Daphnia pulex was less sensitive than Diaphanosoma birgei to malathion. The average lifespan of Daphnia pulex in controls was about 19 days while under similar conditions, that of D. birgei was about 21 days. For either cladoceran species, increased pesticide concentration resulted in decreased survival; this was more evident in the treatment containing the highest concentration of malathion (0.225 ng l(-1)) for D. birgei. Fecundity of D. pulex and D. birgei also decreased with increase in the concentration of malathion. For a given cladoceran species, compared to controls, the gross reproductive, net reproductive rates and the population growth rate significantly decreased due to malathion. Compared to the survivorship variables (age-specific survival, life expectancy and average lifespan), the reproductive parameters (gross reproductive rate, net reproductive rate and the rate of population increase) of the two cladoceran species were decreased by the pesticide.

Background: Over the years, acyclovir has been the oral antiviral agent approved for the treatment of patients with acute herpes zoster,Its effectiveness in lessening the acute signs and symptoms of herpes zoster has been established but the effects on post herpetic neuralgia are less clear cut. Famciclovir is a new member of guanine nucleoside family of drugs. It is a well absorbed oral form of penciclovir with longer half life. This was a open comparative randomized study carried out to compare the safety and efficacy of famciclovir administered at 250mg thrice daily with acyclovir 800mg five times daily for the treatment of acute uncomplicated herpes zoster in immunocompetent individuals aged above 40 years., Aim: To assess the clinical profile of Herpes zoster, compare the efficacy and safety of acyclovir and famciclovir in the treatment of herpes zoster and to describe the effectiveness of acyclovir and famciclovir preventing post herpetic neuralgia., Methods: A total of 100 newly zoster were randomized in 1:1 ratio into acyclovir and famciclovir groups after inclusion criteria were satisfied.Treatment was initiated within 72 hrs of onset of symptoms and was continued for 7 days and evaluated at the end of each week up to six weeks period for full crusting of the lesions, complete healing of the lesion and loss of acute pain., Results: It was observed that famciclovir was as effective as acyclovir with no significant difference in time taken for full crusting, complete healing of lesions or loss of acute pain. Famciclovir was well tolerated with a better safety profile comparable to that of acyclovir. Constipation, headache, nausea and vomiting were the most commonly reported adverse effects, but constipation was considered to have a possible relationship to treatment., Conclusion: In conclusion, oral famciclovir administered three times daily for 7 days during acute zoster infection is as effective as acyclovir, administered 800mg five times daily.In addition it offers significant benefit by providing a well tolerated, cost effective, convenient dosage regime and accelerated rate of lesion resolution and a reduced duration of PHN.

Background: There has been a lot of recent search on consideration of psoriasis as a systemic disease, with researchers being of the view that dermatological manifestations represent only a part of the spectrum. Although, there have been plenty of studies from the west reporting an association of psoriasis with the metabolic syndrome, there are no large-scale Indian studies evaluating Asian patients. The present study is an endeavour in this regard., Aim: To investigate the prevalence of metabolic syndrome in Indian patients with newly diagnosed psoriasis at the onset of the disease., Methods: The study is a prospective clinical case control study, with 100 patients of psoriasis and 100 age-matched healthy controls. 5ml plain venous blood after overnight fasting was obtained by venepuncture. Plasma glucose was tested by glucose oxidase method. Serum cholesterol and triglycerides was estimated by enzymatic method. Metabolic syndrome was diagnosed by the presence of three or more criterion of the National Cholestrol Education Programme's Adult Panel (ATP). The statistical software SAS 9.2 and SPSS 15.0 was used for the analysis of the data., Results: Metabolic syndrome was diagnosed in 8 out of 100 cases and 9 out of 100 controls (p-value: 0.811). We did not find any association of psoriasis with metabolic syndrome in our study. The age of onset of the disease, the duration of the disease and the severity of the disease activity were also not found to be associated with the likelihood of developing metabolic syndrome., Conclusion: Our study refuted any association of psoriasis with metabolic syndrome at the onset of disease activity in Indian patients. The plenty of reports from west approving such an association can be explained by increased risk factors like smoking, alcohol consumption, obesity and stress levels. Further, most such studies have been conducted with patients on treatment, while ours is the first study on newly diagnosed patients prior to the initiation of any therapy.

We evaluated different strains of the rotifer Asplanchna brightwellii collected from central Mexico using morphology, morphometry and molecular tools (CO1 and ITS). Three distinct clonal populations from each of the 3 regions (Mexico City, State of Mexico and State of Guerrero) were established under laboratory conditions. For a given waterbody, morphometric comparisons within the populations of A. brightwellii showed almost stable measurements of trophi and with no statistically significant differences among them (p > 0.05). However, asplanchnid body length and width as well as the cyst diameter varied significantly depending on the waterbody from which A. brightwellii was collected. The smallest adults (about 700 microm) were from Valerio Trujano lake (Guerrero State) samples while the largest were from Xochimilco lake. Similar tendencies were reflected in the diameter of resting eggs. In addition, morphologically the cysts of A. brightwellii from the three waterbodies showed slightly different pattern. The number of globular structures on the surface of cysts was smaller for Valerio Trujano strain, while these were larger and less numerous for both Xochimilco and Zumpango strains. The ITS region tree displayed two groups Xochimilco and Valerio Trujano -Zumpango, this analysis did not reflect the morphological grouping; on the contrary the CO1 gene tree separated the populations according to morphological clusters and location (Xochimilco, Valerio Trujano and Zumpango lakes). When the tree was built using the combination of both ITS and CO1 sequences, the phylogenetic relationships observed on CO1 gene were consistent; but showed differences with the relationships observed on ITS region tree (only two groups).

Aim: To evaluate the effect of an apical negative pressure system, a passive ultrasonic irrigation system and a combination of both apical negative pressure and passive ultrasonic irrigation on the penetration of the irrigating contrast solution (ICS) up to working length and into simulated lateral canals., Methodology: The root canals of 64 single-rooted teeth were instrumented using the ProTaper rotary system. In each sample, three simulated lateral canals were created at 2, 4 and 6 mm levels from the root apex using a 06-size C+ file (Dentsply Maillefer, Ballaigues, Switzerland). Samples were randomly assigned into 4 experimental groups (n = 16): group I - conventional needle irrigation, group II - passive ultrasonic irrigation, group III - apical negative irrigation system and group IV - combination of passive ultrasonic irrigation and apical negative pressure irrigation system. To examine irrigating solution penetration, Indian ink was mixed with 5.25% NaOCl and delivered into the root canals. Samples were then assessed by direct observation of the images taken using Canon EOS rebel T3. The depth of penetration of ICS up to the working length and into the simulated lateral canals was analysed using chi-squared tests., Results: The combination (ANP and PUI) and ANP group had significantly deeper ICS penetration up to the working length (P < 0.001). The combination (ANP and PUI) and the PUI group exhibited significantly greater ICS penetration into lateral canals at the 6 mm level (P < 0.001). At the 4 and 2 mm levels, the combination of ANP and PUI had significantly greater ICS penetration into the lateral canals than the other groups (P < 0.001)., Conclusions: The combination of ANP and PUI was the only group able to achieve irrigating contrast solution penetration both up to the working length and into lateral canals., (© 2013 International Endodontic Journal. Endodontic management of teeth with aberrant root canal morphology can be challenging. This article presents a case in which multiple angulated radiographs and a dental operating microscope clearly revealed the presence of 3 root canals in a right mandibular central incisor with 2 different canal patterns. This case report emphasizes the importance of utilizing a dental operating microscope to understand unusual root canal morphology.

Rotifers have attracted the attention of biologists for well over 200 years. Interest in these exquisite animals rests in their diverse morphology, short generation time resulting in high growth rates, ability to withstand desiccation, and wide distribution, coupled with evidence of cryptic speciation. Moreover, three modes of reproduction are present in the phylum: obligatory sexuality, cyclical parthenogenesis, and obligatory ameiotic parthenogenesis. Thus, this phylum offers a rich field of study. Recognizing the need to share advances in knowledge, a triennial meeting, the International Rotifer Symposium (IRS), was begun in 1976. The most recent symposium (13(th) IRS) was held at Shillong (India) from 18-24, November 2012. In this commentary we considered the development of rotifer research as viewed through the lens of more than 35 years of IRS. Initially papers presented at the IRS focused on ecology, morphology, and pure taxonomic problems, with little applied work being reported. However, after more than three decades, the emphasis has swung to a balance of both basic (e.g., aging, ecology, genetics, and taxonomy) and applied (aquaculture and ecotoxicology) research.

This case report describes the nonsurgical endodontic management of a maxillary first molar with the unusual morphology of three separate buccal roots. An accurate assessment of this morphology was made with the help of cone-beam computed tomography (CBCT). This report also describes the varied root morphology associated with maxillary first molars and the role of CBCT as a diagnostic tool for managing these complex cases successfully.

We demonstrate that the prophage status of bacteria plays a critical role in achieving homogenous population of a phage preparation. When a lytic Staphylococcus bacteriophage 44AHJD was propagated in a Staphylococcus clinical isolate, the enriched phage showed 44AHJD phage virions along with the released prophages from the baiting host. The released prophage was identified as a siphophage by transmission electron microscopy. To obtain a phage preparation free of prophages, when we carried out multiplication of the 44AHJD phage in a prophage free Staphyloccoccus aureus host namely RN4220, we were surprised not to see any phage plaques in spite of the phage exhibiting >99.9% adsorption to such cells. Since RN4220 host is devoid of restriction modification system and prophages, we hypothesized that in spite of successful infection and multiplication, the phage virions might have failed to show plaques due to its insignificant release from the cell possibly due to insufficient endolysin expressed from phage virions during phage development and assembly. Our hypothesis was confirmed when we observed plaques of 44AHJD phage in RN4220 cells where additional phage endolysin protein was supplemented via a plasmid. Endolysin protein from various types of Staphylococcus phages showed plaques of 44AHJD in RN4220 cells confirming our hypothesis. Also, we demonstrate for the first time that propagation of 44AHJD phage with endolysin supplementation in prophage free RN4220 host yields pure phage preparation., (Copyright © 2012 Elsevier B.V. Cytoplasmic dynein is responsible for the transport and delivery of cargoes in organisms ranging from humans to fungi. Dysfunction of dynein motor machinery due to mutations in dynein or its activating complex dynactin can result in one of several neurological diseases in mammals. The mouse Legs at odd angles (Loa) mutation in the tail domain of the dynein heavy chain has been shown to lead to progressive neurodegeneration in mice. The mechanism by which the Loa mutation affects dynein function is just beginning to be understood. In this work, we generated the dynein tail mutation observed in Loa mice into the Neurospora crassa genome and utilized cell biological and complementing biochemical approaches to characterize how that tail mutation affected dynein function. We determined that the Loa mutation exhibits several subtle defects upon dynein function in N. crassa that were not seen in mice, including alterations in dynein localization, impaired velocity of vesicle transport, and in the biochemical properties of purified motors. Our work provides new information on the role of the tail domain on dynein function and points out areas of future research that will be of interest to pursue in mammalian systems., (2012 Wiley Periodicals, Inc)
