Excess sodium intake poses health risks, prompting the exploration of taste modulators to reduce the salt content in low-sodium foods yet maintain salty perception. Previous research found a subthreshold synergistic effect among pyroglutamyl dipeptides on saltiness enhancement. This study investigated the subthreshold synergistic effect of pyroglutamyl peptides and organic acids on saltiness perception. Pyroglutamyl dipeptides (pgluE, pgluV), pyroglutamyl tripeptides (pgluVL and pgluVC), and organic acids (malic acid and succinic acid) were explored in a model system and subsequently in commercial brown onion sauce. The detection thresholds of peptides (pgluE, pgluV, pgluVL, and pgluVC) were determined to be 646, 77, 273, and 221 μmol/L, respectively, and the subthreshold synergistic effect of the pyroglutamyl tripeptides and organic acids was determined using the isobologram method. One of the eight combinations of pyroglutamyl tripeptides with pyroglutamyl dipeptide (pgluV) showed a subthreshold synergistic effect, whereas four combinations of tripeptides with malic acid and one combination with succinic acid exhibited a subthreshold synergistic effect. In commercial brown onion sauce, 25 and 30% salt reductions were achieved using the combinations of the tripeptides with malic acid and succinic acid, respectively. This research lays the foundation for future investigations into the potential combinations of pyroglutamyl peptides and organic acids for saltiness enhancement in low-sodium foods.

In this work, we describe the CRIMSON (CardiovasculaR Integrated Modelling and SimulatiON) software environment. CRIMSON provides a powerful, customizable and user-friendly system for performing three-dimensional and reduced-order computational haemodynamics studies via a pipeline which involves: 1) segmenting vascular structures from medical images; 2) constructing analytic arterial and venous geometric models; 3) performing finite element mesh generation; 4) designing, and 5) applying boundary conditions; 6) running incompressible Navier-Stokes simulations of blood flow with fluid-structure interaction capabilities; and 7) post-processing and visualizing the results, including velocity, pressure and wall shear stress fields. A key aim of CRIMSON is to create a software environment that makes powerful computational haemodynamics tools accessible to a wide audience, including clinicians and students, both within our research laboratories and throughout the community. The overall philosophy is to leverage best-in-class open source standards for medical image processing, parallel flow computation, geometric solid modelling, data assimilation, and mesh generation. It is actively used by researchers in Europe, North and South America, Asia, and Australia. Numerical simulations of cardiovascular mass transport pose significant challenges due to the wide range of Péclet numbers and backflow at Neumann boundaries. In this paper we present and discuss several numerical tools to address these challenges in the context of a stabilized finite element computational framework. To overcome numerical instabilities when backflow occurs at Neumann boundaries, we propose an approach based on the prescription of the total flux. In addition, we introduce a "consistent flux" outflow boundary condition and demonstrate its superior performance over the traditional zero diffusive flux boundary condition. Lastly, we discuss discontinuity capturing (DC) stabilization techniques to address the well-known oscillatory behavior of the solution near the concentration front in advection-dominated flows. We present numerical examples in both idealized and patient-specific geometries to demonstrate the efficacy of the proposed procedures. It is well known that blood has non-Newtonian properties, but it is generally accepted that blood behaves as a Newtonian fluid at shear rates above 100 s-1. However, in transient conditions, there are times and locations where the shear rate is well below 100 s-1, and it is reasonable to infer that non-Newtonian effects could become important. In this study, purely viscous non-Newtonian (generalized Newtonian) properties of blood are incorporated into the simulation-based framework for cardiovascular surgery planning developed by Taylor et al. (1999, "Predictive Medicine: Computational Techniques in Therapeutic Decision Making," Comput. Aided Surg., 4, pp. 231-247; 1998, "Finite Element Modeling of Blood Flow in Arteries," Comput. Methods Appl. Mech. Eng., 158, pp. 155-196). Equations describing blood flow are solved in a patient-based abdominal aortic aneurysm model under steady and physiological flow conditions. Direct numerical simulation (DNS) is used, and the complex flow is found to be constantly transitioning between laminar and turbulent in both the spatial and temporal sense. It is found for the case simulated that using the non-Newtonian viscosity modifies the solution in subtle ways that yield a mesh-independent solution with fewer degrees of freedom than the Newtonian counterpart. It appears that in regions of separated flow, the lower shear rate produces higher viscosity with the non-Newtonian model, which reduces the associated resolution needs. When considering the real case of pulsatile flow, high shear layers lead to greater unsteadiness in the Newtonian case relative to the non-Newtonian case. This, in turn, results in a tendency for the non-Newtonian model to need fewer computational resources even though it has to perform additional calculations for the viscosity. It is also shown that both viscosity models predict comparable wall shear stress distribution. This work suggests that the use of a non-Newtonian viscosity models may be attractive to solve cardiovascular flows since it can provide simulation results that are presumably physically more realistic with at least comparable computational effort for a given level of accuracy.

In this study, we present an adaptive anisotropic finite element method (FEM) and demonstrate how computational efficiency can be increased when applying the method to the simulation of blood flow in the cardiovascular system. We use the SUPG formulation for the transient 3D incompressible Navier-Stokes equations which are discretised by linear finite elements for both the pressure and the velocity field. Given the pulsatile nature of the flow in blood vessels we have pursued adaptivity based on the average flow over a cardiac cycle. Error indicators are derived to define an anisotropic mesh metric field. Mesh modification algorithms are used to anisotropically adapt the mesh according to the desired size field. We demonstrate the efficiency of the method by first applying it to pulsatile flow in a straight cylindrical vessel and then to a porcine aorta with a stenosis bypassed by a graft. We demonstrate that the use of an anisotropic adaptive FEM can result in an order of magnitude reduction in computing time with no loss of accuracy compared to analyses obtained with uniform meshes.
