Abstract: Syphilis is a sexually transmitted disease that can present a wide variety of clinical and histopathological manifestations. We present the case of a 75-year-old patient with a persistent maculopapular rash on the trunk and extremities, in which the biopsy showed an exclusively perifollicular granulomatous infiltrate. In the immunohistochemical staining with anti-Treponema pallidum antibodies, spirochetes scattered within the perifollicular inflammatory infiltrate were identified. These findings together with the serological analysis allowed the diagnosis of secondary syphilis. Folliculotropic nonalopecic syphilis is rare, and the pathogenic mechanisms that cause this specific tropism are unknown. On the other hand, granulomatous inflammation is a pattern typically described in tertiary syphilis but also occasionally found in secondary syphilis. Mycosis fungoides (MF) is defined as the most common cutaneous T‑cell lymphoma (CTCL). The bullous form is considered one of its numerous variants. Only a few cases of this rare entity have been described. This study investigates how amorphous silica (ASi) influences soil-plant-water interactions in distinct soil textures. A sandy loam and silty clay soil were mixed with 0 and 2% ASi, and their impact on soil retention and soil hydraulic conductivity curves were determined. In parallel, tomato plants (Solanum lycopersicum L.) were grown in experimental pots under controlled conditions. When plants were established, the soil was saturated, and a controlled drying cycle ensued until plants reached their wilting points. Soil water content, soil water potential, plant transpiration rate, and leaf water potential were monitored during this process. Results indicate a positive impact of ASi on the sandy loam soil, enhancing soil water content at field capacity (FC, factor of 1.3 times) and at permanent wilting point (PWP, a factor of 3.5 times), while its effect in silty clay loam was negligible (< 1.05 times). In addition, the presence of ASi prevented a significant drop in soil hydraulic conductivity ( K h ) at dry conditions. The K h of ASi-treated sandy loam and silty clay at PWP were 4.3 times higher than their respective control. Transpiration rates in plants grown in ASi-treated sandy loam soil under soil drying conditions were higher than in the control, attributed to improved soil hydraulic conductivity. At the same time, no significant difference was observed in the transpiration of plants treated with ASi in silty clay soil. Predicted climate warming and prolonged droughts pose a threat to the soil structure as organic carbon losses weaken the stability of soil aggregates. Well-structured soils are important for storage and movement of water, solutes, and air, the development of plant roots, as habitat for soil organisms, and the microbial activity. Structural stability is measured in terms of hydro-mechanical properties. This study compares effects of amorphous silica with those of organic carbon on stability parameters during drying of aggregates from relatively finer- and coarser-textured soils. Silica amendment enhanced the positive effect of organic carbon on structural stability in terms of the tensile strength. Synergistic effects between silica and organic carbon in soil colloids appear to dynamically alter aggregate density and friability (i.e., ability to crumble) during drying. Background and Objectives: Previous work has demonstrated that hydrochlorothiazide (HCTZ) is a risk factor for squamous cell carcinomas (SCC) and basal cell carcinomas (BCC) due to pro-photocarcinogenic effects. Atypical fibroxanthoma (AFX) and pleomorphic sarcoma (PDS), both ultraviolet-induced cancers, display a rare but rising cutaneous tumor entity. This study aimed to evaluate if the use of HCTZ is higher in patients with AFX/PDS than in patients with SCC/BCC and subsequently may be a risk factor for AFX/PDS-development., Patients and Methods: In a retrospective study of four German skin cancer centers, AFX/PDS cases and SCC/BCC controls were sex and age matched (1:3) over a time-period of 7 years (2013-2019) to evaluate the use of HCTZ, immunosuppressive medication, second malignancies, and presence of diabetes mellitus., Results: Overall, 146 AFX/PDS and 438 controls (SCC/BCC) were included in the study. The use of HCTZ was significantly higher in patients with AFX/PDS (44.5%) compared to patients with SCC/BCC (25.3%). Additionally, the presence of diabetes mellitus was significantly higher in AFX/PDS patients., Conclusions: This study demonstrates a significantly higher use of HCTZ in patients with AFX/PDS compared to SCC/BCC. Elevated leaf silicon (Si) concentrations improve drought resistance in cultivated plants, suggesting Si might also improve drought performance of wild species. Tropical tree species, for instance, take up substantial amounts of Si, and leaf Si varies markedly at local and regional scales, suggesting consequences for seedling drought resistance. Yet, whether elevated leaf Si improves seedling drought performance in tropical forests is unknown. To manipulate leaf Si concentrations, seedlings of seven tropical tree species were grown in Si-rich and -poor soil, before exposing them to drought in the forest understorey. Survival, growth and wilting were monitored. Elevated leaf Si did not improve drought survival and growth in any of the species. In one species, drought survival was reduced in seedlings previously grown in Si-rich soil, contrary to our expectation. Our results suggest that elevated leaf Si does not improve drought resistance of wild tropical tree species. Elevated leaf Si may even reduce drought performance, suggesting differences in soil conditions influencing leaf Si may contribute to soil-related variation of tropical seedling performance. Furthermore, our results are at odds with most studies on cultivated species and show that alleviative effects of Si in crops cannot be generalized to wild plants in natural systems.

Rice (Oryza sativa L.) cultivation in regions marked by elevated arsenic (As) concentrations poses significant health concerns due to As uptake by the plant and its subsequent entry into the human food chain. With rice serving as a staple crop for a substantial share of the global population, addressing this issue is critical for food security. In flooded paddy soils, where As availability is pronounced, innovative strategies to reduce As uptake and enhance agricultural sustainability are mandatory. Silicon (Si) and Si nanoparticles have emerged as potential candidates to mitigate As accumulation in rice. However, their effects on As uptake exhibit complexity, influenced by initial Si levels in the soil and the amount of Si introduced through fertilization. While low Si additions may inadvertently increase As uptake, higher Si concentrations may alleviate As uptake and toxicity. The interplay among existing Si and As availability, Si supplementation, and soil biogeochemistry collectively shapes the outcome. Adding water-soluble Si fertilizers (e.g., Na 2 SiO 3 and K 2 SiO 3 ) has demonstrated efficacy in mitigating As toxicity stress in rice. Nonetheless, the expense associated with these fertilizers underscores the necessity for low cost innovative solutions. Silicate-solubilizing bacteria (SSB) resilient to As hold promise by enhancing Si availability by accelerating mineral dissolution within the rhizosphere, thereby regulating the Si biogeochemical cycle in paddy soils. Promoting SSB could make cost-effective Si sources more soluble and, consequently, managing the intricate interplay of Si's dual effects on As accumulation in rice. This review paper offers a comprehensive exploration of Si's nuanced role in modulating As uptake by rice, emphasizing the potential synergy between As-resistant SSB and Si availability enhancement. Background: Institutes of dermatopathology are faced with considerable challenges including a continuously rising numbers of submitted specimens and a shortage of specialized health care practitioners. Basal cell carcinoma (BCC) is one of the most common tumors in the fair-skinned western population and represents a major part of samples submitted for histological evaluation. Digitalizing glass slides has enabled the application of artificial intelligence (AI)-based procedures. To date, these methods have found only limited application in routine diagnostics. The aim of this study was to establish an AI-based model for automated BCC detection., Patients and Methods: In three dermatopathological centers, daily routine practice BCC cases were digitalized. The diagnosis was made both conventionally by analog microscope and digitally through an AI-supported algorithm based on a U-Net architecture neural network., Results: In routine practice, the model achieved a sensitivity of 98.23% (center 1) and a specificity of 98.51%. The model generalized successfully without additional training to samples from the other centers, achieving similarly high accuracies in BCC detection (sensitivities of 97.67% and 98.57% and specificities of 96.77% and 98.73% in centers 2 and 3, respectively). Silicon-based defenses deter insect herbivores in many cultivated and wild grass species. Furthermore, in some of these species, silicon (Si) uptake and defense can be induced by herbivory. Tropical trees also take up Si and leaf Si concentrations vary greatly across and within species. As herbivory is a major driver of seedling mortality and niche differentiation of tropical tree species, understanding anti-herbivore defenses is pivotal. Yet, whether silicon is a constitutive and inducible herbivory defense in tropical forest tree species remains unknown. We grew seedlings of eight tropical tree species in a full factorial experiment, including two levels of plant-available soil Si concentrations (-Si/+Si) and a simulated herbivory treatment (-H/+H). The simulated herbivory treatment was a combination of clipping and application of methyl jasmonate. We then carried out multiple-choice feeding trials, separately for each tree species, in which leaves of each treatment combination were offered to a generalist caterpillar ( Spodoptera frugiperda ). Leaf damage was assessed. Three species showed a significant decrease in leaf damage under high compared to low Si conditions (by up to 72%), consistent with our expectation of Si-based defenses acting in tropical tree species. In one species, leaf damage was increased by increasing soil Si and in four species, no effect of soil Si on leaf damage was observed. Opposite to our expectation of Si uptake and defense being inducible by herbivory damage, simulated herbivory increased leaf damage in two species. Furthermore, simulated herbivory reduced Si concentrations in one species. Our results showed that tropical tree seedlings can be better defended when growing in Si-rich compared to Si-poor soils, and that the effects of Si on plant defense vary strongly across species. Furthermore, Si-based defenses may not be inducible in tropical tree species. Overall, constitutive Si-based defense should be considered part of the vast array of anti-herbivore defenses of tropical tree species. Increased crop production is a main goal to feed the predicted human population in future. The current management practice is, however, not sustainable as it depends on high amounts of fertilizer application and is highly vulnerable to decreased soil water availability. At the same time it becomes more and more crucial to reduce or even mitigate anthropogenic greenhouse gas (GHG) emissions. A possible way to enable this, might be the increase of the soil C sequestration and thus the C sink function of arable lands. A recent and potentially more sustainable idea is the single time fertilization with amorphous silicon (ASi) which is known to increase both nutrient and water availability. Here we show for the first time on the basis of a field plot experiment how a fertilization with ASi is affecting both, crop yield and the C sequestration of the soils in an agricultural system cultivating wheat. We found a strong increase in wheat yield and biomass production after ASi fertilization by increasing soil moisture during the whole growing season. Additionally, despite a relatively short growing season, Si fertilization increased the net C uptake by soils, i.e., C sequestration with both Si fertilized treatments showing a negative net ecosystem C balance (soil C gain) during the measurement period, while the control showed a small positive net ecosystem C balance (soil C loss). To our best knowledge, this is the first time such effect has ever been observed. Background: In melanoma patients, surgical excision of the first draining lymph node, the sentinel lymph node (SLN), is a routine procedure to evaluate lymphogenic metastases. Metastasis detection by histopathological analysis assesses multiple tissue levels with hematoxylin and eosin and immunohistochemically stained glass slides. Considering the amount of tissue to analyze, the detection of metastasis can be highly time-consuming for pathologists. The application of artificial intelligence in the clinical routine has constantly increased over the past few years., Methods: In this multi-center study, a deep learning method was established on histological tissue sections of sentinel lymph nodes collected from the clinical routine. The algorithm was trained to highlight potential melanoma metastases for further review by pathologists, without relying on supplementary immunohistochemical stainings (e.g. anti-S100, anti-MelanA)., Results: The established method was able to detect the existence of metastasis on individual tissue cuts with an area under the curve of 0.9630 and 0.9856 respectively on two test cohorts from different laboratories. The method was able to accurately identify tumour deposits>0.1 mm and, by automatic tumour diameter measurement, classify these into 0.1 mm to -1.0 mm and>1.0 mm groups, thus identifying and classifying metastasis currently relevant for assessing prognosis and stratifying treatment., Conclusions: Our results demonstrate that AI-based SLN melanoma metastasis detection has great potential and could become a routinely applied aid for pathologists. For the majority of higher plants, silicon (Si) is considered a beneficial element because of the various favorable effects of Si accumulation in plants that have been revealed, including the alleviation of metal(loid) toxicity. The accumulation of non-degradable metal(loid)s in the environment strongly increased in the last decades by intensified industrial and agricultural production with negative consequences for the environment and human health. Phytoremediation, i.e., the use of plants to extract and remove elemental pollutants from contaminated soils, has been commonly used for the restoration of metal(loid)-contaminated sites. In our viewpoint article, we briefly summarize the current knowledge of Si-mediated alleviation of metal(loid) toxicity in plants and the potential role of Si in the phytoremediation of soils contaminated with metal(loid)s. In this context, a special focus is on metal(loid) accumulation in (soil) phytoliths, i.e., relatively stable silica structures formed in plants. The accumulation of metal(loid)s in phytoliths might offer a promising pathway for the long-term sequestration of metal(loid)s in soils. As specific phytoliths might also represent an important carbon sink in soils, phytoliths might be a silver bullet in the mitigation of global change. Thus, the time is now to combine Si/phytolith and phytoremediation research. This will help us to merge the positive effects of Si accumulation in plants with the advantages of phytoremediation, which represents an economically feasible and environmentally friendly way to restore metal(loid)-contaminated sites.

Arctic soils are the largest pool of soil organic carbon worldwide. Temperatures in the Arctic have risen faster than the global average during the last decades, decreasing annual freezing days and increasing the number of freeze-thaw cycles (temperature oscillations passing through zero degrees) per year as the temperature is expected to fluctuate more around 0 °C. At the same time, proceeding deepening of seasonal thaw may increase silicon (Si) and calcium (Ca) concentrations in the active layer of Arctic soils as the concentrations in the thawing permafrost layer might be higher depending on location. We analyzed the importance of freeze-thaw cycles for Arctic soil CO 2 fluxes. Furthermore, we tested how Si (mobilizing organic C) and Ca (immobilizing organic C) interfere with the soil CO 2 fluxes in the context of freeze-thaw cycles. Our results show that with each freeze-thaw cycle the CO 2 fluxes from the Arctic soils decreased. Our data revealed a considerable CO 2 emission below 0 °C. We also show that pronounced differences emerge in Arctic soil CO 2 fluxes with Si increasing and Ca decreasing CO 2 fluxes. Furthermore, we show that both Si and Ca concentrations in Arctic soils are central controls on Arctic soil CO 2 release, with Si increasing Arctic soil CO 2 release especially when temperatures are just below 0 °C. Our findings could provide an important constraint on soil CO 2 emissions upon soil thaw, as well as on the greenhouse gas budget of high latitudes. Despite seminal papers that stress the significance of silicon (Si) in plant biology and ecology, most studies focus on manipulations of Si supply and mitigation of stresses. The ecological significance of Si varies with different levels of biological organization, and remains hard to capture. We show that the costs of Si accumulation are greater than is currently acknowledged, and discuss potential links between Si and fitness components (growth, survival, reproduction), environment, and ecosystem functioning. We suggest that Si is more important in trait-based ecology than is currently recognized. Background and Objectives: In recent years, considerable insight has been gained into the pathogenesis, diagnosis and treatment of cutaneous sarcomas, including atypical fibroxanthoma (AFX) and pleomorphic dermal sarcoma (PDS). Both entities have shown increasing incidence rates in the last decade. This study was initiated to evaluate how these new insights impact the number of diagnoses of AFX/PDS compared to other cutaneous sarcoma entities., Patients and Methods: In a retrospective study of four German skin cancer centers, all histopathological reports of cutaneous sarcomas (AFX, PDS, dermatofibrosarcoma protuberans, cutaneous leiomyosarcoma, angiosarcoma, and Kaposi sarcoma) confirmed by board-certified dermatopathologists were analyzed during a time-period of seven years (2013-2019). Additionally, utilization of immunohistochemical markers (including pan-cytokeratin, S100, desmin, CD34, CD10, procollagen-1, CD99, CD14, and CD68) as an adjunct to diagnose AFX/PDS was recorded., Results: Overall, 255 cutaneous sarcomas were included in the present study. The diagnosis of a cutaneous sarcoma has consequently risen from 2013 to 2019 (from 16 to 52 annual cases). The results of AFX/PDS revealed 4.6 times more diagnoses in 2019 than in 2013. Atypical fibroxanthoma represented the most common subtype, displaying 49.3 % of all diagnosed cutaneous sarcomas. Additionally, the increase of AFX/PDS was linked to the use of immunohistochemistry, with specific immunohistochemical markers used in 57.1 % of cases in 2013 compared to 100 % in 2019., Conclusions: This retrospective study of four German skin cancer centers demonstrates a substantial rise of AFX/PDS, possibly due to recently established diagnostic and terminology standards. This rise is probably linked to increased utilization of specific immunohistochemical markers. Hintergrund Und Ziele: In den letzten Jahren konnten umfassende Erkenntnisse über die Pathogenese, Diagnostik und Behandlung von kutanen Sarkomen, insbesondere des atypischen Fibroxanthoms (AFX) und pleomorphen dermalen Sarkoms (PDS) gesammelt werden. Beide Entitäten zeigten innerhalb der letzten Dekade steigende Inzidenzraten. Die vorliegende Studie diente der Untersuchung, welchen Einfluss die neuen Erkenntnisse auf die Fallzahlen von AFX/PDS im Vergleich zu anderen Sarkom-Entitäten haben., Patienten Und Methodik: Diese retrospektive Studie wurde an vier deutschen Hauttumorzentren durchgeführt und alle von zertifizierten Dermatopathologen bestätigten histopathologischen Befunde von kutanen Sarkomen (AFX, PDS, Dermatofibrosarcoma protuberans, kutanes Leiomyosarkom, Angiosarkom und Kaposi-Sarkom) in einem Zeitraum von sieben Jahren (2013-2019) evaluiert. Accurate classification of melanocytic tumors is important for prognostic evaluation, treatment and follow-up protocols of patients. The majority of melanocytic proliferations can be classified solely based on clinical and pathological criteria, however in select cases a definitive diagnostic assessment remains challenging and additional diagnostic biomarkers would be advantageous. We analyzed melanomas, nevi, Spitz nevi and atypical spitzoid tumors using parallel sequencing (exons of 611 genes and 507 gene translocation analysis) and methylation arrays (850k Illumina EPIC). By combining detailed genetic and epigenetic analysis with reference-based and reference-free DNA methylome deconvolution we compared Spitz nevi to nevi and melanoma and assessed the potential for these methods in classifying challenging spitzoid tumors. Results were correlated with clinical and histologic features. Spitz nevi were found to cluster independently of nevi and melanoma and demonstrated a different mutation profile. Multiple copy number alterations and TERT promoter mutations were identified only in melanomas. Genome-wide methylation in Spitz nevi was comparable to benign nevi while the Leukocytes UnMethylation for Purity (LUMP) algorithm in Spitz nevi was comparable to melanoma. Histologically difficult to classify Spitz tumor cases were assessed which, based on methylation arrays, clustered between Spitz nevi and melanoma and in terms of genetic profile or copy number variations demonstrated worrisome features suggesting a malignant neoplasm. Comprehensive sequencing and methylation analysis verify Spitz nevi as an independent melanocytic entity distinct from both nevi and melanoma. Biogeochemical cycling in permafrost-affected ecosystems remains associated with large uncertainties, which could impact the Earth's greenhouse gas budget and future climate policies. In particular, increased nutrient availability following permafrost thaw could perturb the greenhouse gas exchange in these systems, an effect largely unexplored until now. Here, we enhance the terrestrial ecosystem model QUINCY (QUantifying Interactions between terrestrial Nutrient CYcles and the climate system), which simulates fully coupled carbon (C), nitrogen (N) and phosphorus (P) cycles in vegetation and soil, with processes relevant in high latitudes (e.g., soil freezing and snow dynamics). In combination with site-level and satellite-based observations, we use the model to investigate impacts of increased nutrient availability from permafrost thawing in comparison to other climate-induced effects and CO 2 fertilization over 1960 to 2018 across the high Arctic. Our simulations show that enhanced availability of nutrients following permafrost thaw account for less than 15% of the total Gross primary productivity increase over the time period, despite simulated N limitation over the high Arctic scale. As an explanation for this weak fertilization effect, observational and model data indicate a mismatch between the timing of peak vegetative growth (week 26-27 of the year, corresponding to the beginning of July) and peak thaw depth (week 32-35, mid-to-late August), resulting in incomplete plant use of nutrients near the permafrost table. The resulting increasing N availability approaching the permafrost table enhances N loss pathways, which leads to rising nitrous oxide (N 2 O) emissions in our model. Site-level emission trends of 2 mg N m -2  year -1 on average over the historical time period could therefore predict an emerging increasing source of N 2 O emissions following future permafrost thaw in the high Arctic., (© 2022 The Authors. Rice cultivation requires high amounts of phosphorus (P). However, significant amounts of P fertilizer additions may be retained by iron (Fe) oxides and are thus unavailable for plants. At the same time, rice cultivation has a high demand for silicic acid (Si), reducing Si availability after short duration of rice cultivation. By studying a paddy chronosequence with rice cultivation up to 2000 years, we show that Si limitation, observed as early as a few decades of rice cultivation, is limiting P availability along the paddy soils chronosequence. Using near edge X-ray absorption fine structure spectroscopy (NEXAFS) in a scanning transmission (soft) X-ray microscope (STXM) we show release of available P was linked to a Si-induced change in speciation of Fe-phases in soil particles and competition of Si with P for binding sites. Hence, low Si availability is limiting P availability in paddy soils. We propose that proper management of Si availability is a promising tool to improve the P supply of paddy plants., (© 2022. Background: Onychomycosis numbers among the most common fungal infections in humans affecting finger- or toenails. Histology remains a frequently applied screening technique to diagnose onychomycosis. Screening slides for fungal elements can be time-consuming for pathologists, and sensitivity in cases with low amounts of fungi remains a concern. Convolutional neural networks (CNNs) have revolutionized image classification in recent years. The goal of our project was to evaluate if a U-NET-based segmentation approach as a subcategory of CNNs can be applied to detect fungal elements on digitized histologic sections of human nail specimens and to compare it with the performance of 11 board-certified dermatopathologists., Methods: In total, 664 corresponding H&E- and PAS-stained histologic whole-slide images (WSIs) of human nail plates from four different laboratories were digitized. Histologic structures were manually annotated. A U-NET image segmentation model was trained for binary segmentation on the dataset generated by annotated slides., Results: The U-NET algorithm detected 90.5% of WSIs with fungi, demonstrating a comparable sensitivity with that of the 11 board-certified dermatopathologists (sensitivity of 89.2%)., Conclusions: Our results demonstrate that machine-learning-based algorithms applied to real-world clinical cases can produce comparable sensitivities to human pathologists. Our established U-NET may be used as a supportive diagnostic tool to preselect possible slides with fungal elements. Slides where fungal elements are indicated by our U-NET should be reevaluated by the pathologist to confirm or refute the diagnosis of onychomycosis.

Background: Some of the most common cutaneous neoplasms are Bowen’s disease and seborrheic keratosis, a malignant and a benign proliferation, respectively. These entities represent a significant fraction of a dermatopathologists’ workload, and in some cases, histological differentiation may be challenging. The potential of deep learning networks to distinguish these diseases is assessed. Methods: In total, 1935 whole-slide images from three institutions were scanned on two different slide scanners. A U-Net-based segmentation deep learning algorithm was trained on data from one of the centers to differentiate Bowen’s disease, seborrheic keratosis, and normal tissue, learning from annotations performed by dermatopathologists. Optimal thresholds for the class distinction of diagnoses were extracted and assessed on a test set with data from all three institutions. Results: We aimed to diagnose Bowen’s diseases with the highest sensitivity. A good performance was observed across all three centers, underlining the model’s robustness. In one of the centers, the distinction between Bowen’s disease and all other diagnoses was achieved with an AUC of 0.9858 and a sensitivity of 0.9511. Seborrheic keratosis was detected with an AUC of 0.9764 and a sensitivity of 0.9394. Nevertheless, distinguishing irritated seborrheic keratosis from Bowen’s disease remained challenging. Conclusions: Bowen’s disease and seborrheic keratosis could be correctly identified by the evaluated deep learning model on test sets from three different centers, two of which were not involved in training, and AUC scores > 0.97 were obtained. The method proved robust to changes in the staining solution and scanner model. We believe this demonstrates that deep learning algorithms can aid in clinical routine; however, the results should be confirmed by qualified histopathologists.

There is increased interest in the terrestrial silicon cycle in the last decades as its different compounds and species have large implications for ecosystem performance in terms of soil nutrient and water availability, ecosystem productivity as well as ecological aspects such as plant-microbe and plant-animal feedbacks. The currently existing analytical methods are limited. Fourier-transform infrared spectroscopy (FTIR) analysis is suggested being a promising tool to differentiate between the different Si species. We report here on the differentiation of varying Si-species/Si-binding (in synthetic material) using FTIR-analyses. Therefore, we collected FTIR-spectra of five different amorphous silica, Ca-silicate, sodium silicate (all particulate), a water-soluble fraction of amorphous silica and soil affected by volcanic activity and compared their spectra with existing data. A decrease of the internal order of the materials analyzed was indicated by peak broadening of the Si-O-Si absorption band. Peak shifts at this absorption band were induced by larger ions incorporated in the Si-O-Si network. Additionally, short-range ordered aluminosilicates (SROAS) have specific IR absorption bands such as the Si-O-Al band. High concentrations of arsenic (As) in groundwater threaten the environment and public health. Geogenically, groundwater As contamination predominantly occurs via its mobilization from underground As-rich sediments. In an aquatic ecosystem, As is typically driven by several underlying processes, such as redox transitions, microbially driven reduction of iron (Fe) oxide minerals, and release of associated As. Notably, dissolved As mobilized from soils and sediments exhibits high affinity for dissolved organic matter (DOM). Thus, high DOM concentrations can increase As mobility. Therefore, it is crucial to understand the complex interactions and biogeochemical cycling of As, DOM, and Fe oxides. This review collates knowledge regarding the fate of As in multicomponent As-DOM-Fe systems, including ternary complexes involving both Fe and DOM. Additionally, the release mechanisms of As from sediments into groundwater in the presence of both Fe and DOM have been discussed. The mechanisms of As mobilization/sorption at the solid-water interface can be affected by negatively charged DOM competing for sorption sites with As on Fe (oxy)(hydr)oxides and may be further modified by other anionic ubiquitous species such as phosphate, silicic acid, or sulfur. This review emphasizes the need for a comprehensive understanding of the impact of DOM, Fe oxides, and related biogeochemical processes on As mobilization to aquifers. The review identifies important knowledge gaps that may aid in developing applicable practices for preventing the spread of As contamination in aquatic resources and traditional soil management practices., (Copyright © 2021 Elsevier Ltd. Drought and the availability of mineable phosphorus minerals used for fertilization are two of the important issues agriculture is facing in the future. High phosphorus availability in soils is necessary to maintain high agricultural yields. Drought is one of the major threats for terrestrial ecosystem performance and crop production in future. Among the measures proposed to cope with the upcoming challenges of intensifying drought stress and to decrease the need for phosphorus fertilizer application is the fertilization with silica (Si). Here we tested the importance of soil Si fertilization on wheat phosphorus concentration as well as wheat performance during drought at the field scale. Our data clearly showed a higher soil moisture for the Si fertilized plots. This higher soil moisture contributes to a better plant performance in terms of higher photosynthetic activity and later senescence as well as faster stomata responses ensuring higher productivity during drought periods. The plant phosphorus concentration was also higher in Si fertilized compared to control plots. Overall, Si fertilization or management of the soil Si pools seem to be a promising tool to maintain crop production under predicted longer and more serve droughts in the future and reduces phosphorus fertilizer requirements., (© 2021. Various studies showed a decrease of drought stress specific parameters of plants after silicon (Si) fertilization. But all studies differed in soil Si concentration between the control and Si treatments. As amorphous silica (ASi) was recently found to cause a strong increase of water holding capacity and plant available water in soils, a combined effect of soil moisture and plant response due to Si addition was assumed. In this study, the influence of the soil Si content was excluded by using the same Si enriched soil for treatments of two rice lines, lsi1 mutant defective in Si uptake and its wild-type rice. Most plant parameters, such as nutrient contents, biomass, specific leaf area, specific root length, leaf water content and C allocation did not differ significantly between the genotypes neither under flooded conditions, nor under drought conditions. Only photosynthesis and stomatal conductance were slightly higher for the wild type in both drought and flooded treatments. Overall, our data showed that Si accumulation within the plant tissues has only a minor effect on plant performance under drought stress. Silicon (Si) accumulation is known to alleviate various biotic and abiotic stressors in plants with potential ecological consequences. However, for dicotyledonous plants our understanding of Si variation remains limited. We conducted a comparative experimental study to investigate (1) interspecific variation of foliar Si concentrations across 37 dicotyledonous forbs of temperate grasslands, (2) intraspecific variation in foliar Si concentration in response to soil Si availability, the influence of (3) phylogenetic relatedness, and (4) habitat association to moisture. Foliar Si differed markedly (approx. 70-fold) across the investigated forbs, with some species exhibiting Si accumulation similar to grasses. Foliar Si increased with soil Si availability, but the response varied across species: species with higher Si accumulation capacity showed a stronger response, indicating that they did not actively upregulate Si uptake under low soil Si availability. Foliar Si showed a pronounced phylogenetic signal, i.e., closely related species exhibited more similar foliar Si concentrations than distantly related species. Significant differences in foliar Si concentration within closely related species pairs nevertheless support that active Si uptake and associated high Si concentrations has evolved multiple times in forbs. Foliar Si was not higher in species associated with drier habitats, implying that in dicotyledonous forbs of temperate grasslands high foliar Si is not an adaptive trait to withstand drought. Our results demonstrated considerable inter- and intraspecific variation in foliar Si concentration in temperate forbs. Cutaneous vascular tumors consist of a heterogeneous group of benign proliferations, including a range of hemangiomas and vascular malformations, as well as heterogeneous groups of both borderline and malignant neoplasms such as Kaposi's sarcoma and angiosarcomas. The genetics of these tumors have been assessed independently in smaller individual cohorts making comparisons difficult. In our study, we analyzed a representative cohort of benign vascular proliferations observed in a clinical routine setting as well as a selection of malignant vascular proliferations. Our cohort of 104 vascular proliferations including hemangiomas, malformations, angiosarcomas and Kaposi's sarcoma were screened by targeted next-generation sequencing for activating genetic mutations known or assumed to be potentially relevant in vascular proliferations. An association analysis was performed for mutation status and clinico-pathological parameters. Frequent activating hotspot mutations in GNA genes, including GNA14 Q205, GNA11 and GNAQ Q209 were identified in 16 of 64 benign vascular tumors (25%). GNA gene mutations were particularly frequent (52%) in cherry (senile) hemangiomas (13 of 25). In angiosarcomas, activating RAS mutations ( HRAS and NRAS ) were identified in three samples (16%). No activating GNA or RAS gene mutations were identified in Kaposi's sarcomas. Our study identifies GNA14 Q205, GNA11 and GNAQ Q209 mutations as being the most common and mutually exclusive mutations in benign hemangiomas. Accurate and fast assessment of resection margins is an essential part of a dermatopathologist's clinical routine. In this work, we successfully develop a deep learning method to assist the dermatopathologists by marking critical regions that have a high probability of exhibiting pathological features in whole slide images (WSI). We focus on detecting basal cell carcinoma (BCC) through semantic segmentation using several models based on the UNet architecture. The study includes 650 WSI with 3443 tissue sections in total. Two clinical dermatopathologists annotated the data, marking tumor tissues' exact location on 100 WSI. The rest of the data, with ground-truth sectionwise labels, are used to further validate and test the models. We analyze two different encoders for the first part of the UNet network and two additional training strategies: (a) deep supervision, (b) linear combination of decoder outputs, and obtain some interpretations about what the network's decoder does in each case. The best model achieves over 96%, accuracy, sensitivity, and specificity on the Test set.

Background: Tinea capitis is a highly contagious infectious disease caused by dermatophytes. In Central Europe, it is mainly caused by zoophilic dermatophytes, as, for example Microsporum (M) canis or Trichophyton (T) mentagrophytes and increasingly by anthropophilic fungi. T tonsurans was commonly related to the Tinea gladiatorum, where transmission occurred between infected persons or via contaminated floors., Objective: Reporting the transmission of this highly contagious dermatophyte for the first time via beard shaving and hairdressing in barber shops in Germany., Patients and Methods: 18 young male patients developed tinea capitis and/or barbae shortly after shavings of the beard and/or hair in a barber shop. Native, cultural and molecular diagnostics as well as tissue biopsies and resistance tests were performed of skin and hair samples., Results: In all samples, T tonsurans could be identified. The medical history and the clinical picture suggest a transmission through contaminated hairdressing tools. The patients were treated with terbinafine or itraconazole in combination with or exclusively with topical antimycotics., Conclusion: The transmission and a resulting increase in the incidence of infections with T tonsurans may be due to shavings with direct skin contact of insufficiently disinfected hairdressing tools. This path of infection has already been observed in Africa and is now being described for the first time in Germany. Plants' ability to take up silicon from the soil, accumulate it within their tissues and then reincorporate it into the soil through litter creates an intricate network of feedback mechanisms in ecosystems. Here, we provide a concise review of silicon's roles in soil chemistry and physics and in plant physiology and ecology, focusing on the processes that form these feedback mechanisms. Through this review and analysis, we demonstrate how this feedback network drives ecosystem processes and affects ecosystem functioning. Consequently, we show that Si uptake and accumulation by plants is involved in several ecosystem services like soil appropriation, biomass supply, and carbon sequestration. Considering the demand for food of an increasing global population and the challenges of climate change, a detailed understanding of the underlying processes of these ecosystem services is of prime importance. Silicon and its role in ecosystem functioning and services thus should be the main focus of future research.

While Si influences nutrient stoichiometry and decomposition of graminoid litter, it is still unclear how Si influences anoxic litter decomposition and CH 4 formation in graminoid dominated fen peatlands. First, Eriophorum vaginatum plants were grown under different Si and P availabilities, then shoots and roots were characterized regarding their proportions of C, Si, N and P and regarding C quality. Subsequently the Eriophorum shoots were subjected to anoxic decomposition. We hypothesized; that (I) litter grown under high Si availability would show a higher Si but lower nutrient mass fractions and a lower share of recalcitrant carbon moieties; (II) high-Si litter would show higher CH 4 and CO 2 production rates during anoxic decomposition; (III) methanogenesis would occur earlier in less recalcitrant high-Si litter, compared to low-Si litter. We found a higher Si mass fraction that coincides with a general decrease in C and N mass fractions and decreased share of recalcitrant organic moieties. For high-Si litter, the CH 4 production rate was higher, but there was no long-term influence on the CO 2 production rate. More labile high-Si litter and a differential response in nutrient stoichiometry led to faster onset of methanogenesis. This may have important implications for our understanding of anaerobic carbon turnover in graminoid-rich fens.

Silicon (Si) speciation and availability in soils is highly important for ecosystem functioning, because Si is a beneficial element for plant growth. Si chemistry is highly complex compared to other elements in soils, because Si reaction rates are relatively slow and dependent on Si species. Consequently, we review the occurrence of different Si species in soil solution and their changes by polymerization, depolymerization, and condensation in relation to important soil processes. We show that an argumentation based on thermodynamic endmembers of Si dependent processes, as currently done, is often difficult, because some reactions such as mineral crystallization require months to years (sometimes even centuries or millennia). Furthermore, we give an overview of Si reactions in soil solution and the predominance of certain solid compounds, which is a neglected but important parameter controlling the availability, reactivity, and function of Si in soils. We further discuss the drivers of soil Si cycling and how humans interfere with these processes. The soil Si cycle is of major importance for ecosystem functioning; therefore, a deeper understanding of drivers of Si cycling (e.g., predominant speciation), human disturbances and the implication for important soil properties (water storage, nutrient availability, and micro aggregate stability) is of fundamental relevance.

Chronic recalcitrant dermatophytoses, due to Trichophyton ( T .) mentagrophytes Type VIII are on the rise in India and are noteworthy for their predominance. It would not be wrong to assume that travel and migration would be responsible for the spread of T . mentagrophytes Type VIII from India, with many strains resistant to terbinafine, to other parts of the world. From September 2016 until March 2020, a total of 29 strains of T . mentagrophytes Type VIII (India) were isolated. All patients were residents of Germany: 12 females, 15 males and the gender of the remaining two was not assignable. Patients originated from India (11), Pakistan (two), Bangladesh (one), Iraq (two), Bahrain (one), Libya (one) and other unspecified countries (10). At least two patients were German-born residents. Most samples (21) were collected in 2019 and 2020. All 29 T . mentagrophytes isolates were sequenced (internal transcribed spacer (ITS) and translation elongation factor 1-α gene (TEF1-α)). All were identified as genotype VIII (India) of T . mentagrophytes . In vitro resistance testing revealed 13/29 strains (45%) to be terbinafine-resistant with minimum inhibitory concentration (MIC) breakpoints ≥0.2 µg/mL. The remaining 16 strains (55%) were terbinafine-sensitive. Point mutation analysis revealed that 10/13 resistant strains exhibited Phe 397 Leu amino acid substitution of squalene epoxidase (SQLE), indicative for in vitro resistance to terbinafine. Two resistant strains showed combined Phe 397 Leu and Ala 448 Thr amino acid substitutions, and one strain a single Leu 393 Phe amino acid substitution. Out of 16 terbinafine-sensitive strains, in eight Ala 448 Thr, and in one Ala 448 Thr +, new Val 444 Ile amino acid substitutions were detected. Resistance to both itraconazole and voriconazole was observed in three out of 13 analyzed strains. Treatment included topical ciclopirox olamine plus topical miconazole or sertaconazole. Oral itraconazole 200 mg twice daily for four to eight weeks was found to be adequate. Terbinafine-resistant T . mentagrophytes Type VIII are being increasingly isolated. In Germany, transmission of T . mentagrophytes Type VIII from the Indian subcontinent to Europe should be viewed as a significant public health issue.

In flooded paddy soils, inorganic and methylated thioarsenates contribute substantially to arsenic speciation besides the much-better-investigated oxyarsenic species, and thioarsenate uptake into rice plants has recently been shown. To better understand their fate when soil redox conditions change, that is, from flooding to drainage to reflooding, batch incubations and unplanted microcosm experiments were conducted with two paddy soils covering redox potentials from E H -260 to +200 mV. Further, occurrence of thioarsenates in the oxygenated rice rhizosphere was investigated using planted rhizobox experiments. Soil flooding resulted in rapid formation of inorganic thioarsenates with a dominance of trithioarsenate. Maximum thiolation of inorganic oxyarsenic species was 57% at E H -130 mV and oxidation caused nearly complete dethiolation. Only monothioarsenate formed again upon reflooding and was the major inorganic thioarsenate detected in the rhizosphere. Maximum thiolation of mono- and dimethylated oxyarsenates was about 70% and 100%, respectively, below E H 0 mV. Dithiolated species dominated over monothiolated species below E H -100 mV. Among all thioarsenates, dimethylated monothioarsenate showed the least transformation upon prolonged oxidation. It also was the major thiolated arsenic species in the rhizosphere with concentrations comparable to its precursor dimethylated oxyarsenate, which is especially critical since dimethylated monothioarsenate is highly carcinogenic.

More frequent and longer drought periods are predicted threatening agricultural yield. The capacity of soils to hold water is a highly important factor controlling drought stress intensity for plants. Biogenic amorphous silica (bASi) pools in soils are in the range of 0-6% and are suggested to help plants to resist drought. In agricultural soils, bASi pools declined to values of ~1% or lower) due to yearly crop harvest, decreasing water holding capacity of the soils. Here, we assessed the contribution of bASi to water holding capacity (WHC) of soil. Consequently, ASi was mixed at different rates (0, 1, 5 or 15%) with different soils. Afterwards, the retention curve of the soils was determined via Hyprop method. Here we show that bASi increases the soil water holding capacity substantially, by forming silica gels with a water content at saturation higher than 700%. An increase of bASi by 1% or 5% (weight) increased the water content at any water potential and plant available water increased by up to > 40% or > 60%, respectively. Our results suggest that soil management should be modified to increase bASi content, enhancing available water in soils and potentially decreasing drought stress for plants in terrestrial ecosystems.

Despite available vaccination, measles is one of the leading causes of death among young children in developing countries. In clinical practice, the spectrum of differential diagnoses of morbilliform exanthemas associated with fever is wide, and it can be hard to differentiate from other infectious eruptions, especially in adults or in atypical courses in immunocompromised patients. The goal of our study was to identify characteristic histomorphological and immunohistochemical patterns of measles exanthema through the study of 13 skin biopsy specimens obtained from 13 patients with this disease and a review of cases in the literature. Histopathological features of measles exanthema are quite distinctive and characterized by a combination of multinucleated keratinocytes, and individual and clustered necrotic keratinocytes in the epidermis with pronounced folliculosebaceous as well as acrosyringeal involvement. Immunohistochemical staining of skin biopsies with anti-measles virus (MeV) nucleoprotein and anti-MeV phosphoprotein can be of great value in confirming the diagnosis of measles. Both methods can serve as quick additional diagnostic tools for prompt implementation of quarantine measures and for providing medical assistance, even in patients in whom the clinician did not consider measles as a differential diagnosis of the rash due to the rarity of the disease in a putatively vaccinated community.

An otherwise healthy 50-year-old woman was evaluated for the presence of 2 erythematous, and slightly pruritic plaques, involving both cheeks for 30 years. Left-side skin biopsy showed a diffuse proliferation of ductal structures horizontally arranged and involving the reticular dermis that resembled tubular adenoma embedded in a sclerotic stroma and surrounded by a peculiar periductal desmoplasia. Nuclear atypia or mitosis was not found. Contralateral biopsy showed identical findings. Differential diagnosis included microcystic adnexal carcinoma (MAC) and plaque-like syringoma and a peculiarly horizontally arranged tubular adenoma. We ruled out MAC as the lesions were long-standing, without infundibular cysts, solid strands, or perineural infiltration. Our case closely resembled those previously described as sweat duct proliferation associated with aggregates of elastic tissue and atrophoderma vermiculatum, although striking differences were observed, as our case did not present aggregates of elastic tissue, did not involve the papillary and superficial reticular dermis, and presented evidences of decapitation secretion as a sign of apocrine differentiation. We consider our case as a MAC simulator and we propose the descriptive name of bilateral facial apocrine fibrosing hamartoma.

Humans modify ecosystems and biodiversity worldwide, with negative consequences for ecosystem functioning. Promoting plant diversity is increasingly suggested as a mitigation strategy. However, our mechanistic understanding of how plant diversity affects the diversity of heterotrophic consumer communities remains limited. Here, we disentangle the relative importance of key components of plant diversity as drivers of herbivore, predator, and parasitoid species richness in experimental forests and grasslands. We find that plant species richness effects on consumer species richness are consistently positive and mediated by elevated structural and functional diversity of the plant communities. The importance of these diversity components differs across trophic levels and ecosystems, cautioning against ignoring the fundamental ecological complexity of biodiversity effects. Importantly, plant diversity effects on higher trophic-level species richness are in many cases mediated by modifications of consumer abundances. In light of recently reported drastic declines in insect abundances, our study identifies important pathways connecting plant diversity and consumer diversity across ecosystems.

Phosphorus availability in soils is an important parameter influencing primary production in terrestrial ecosystems. Phosphorus limitation exists in many soils since a high proportion of soil phosphorus is stored in unavailable forms for plants, such as bound to iron minerals or stabilized organic matter. This is in spite of soils having a high amount of total soil phosphorus. The feasibility of silicon to mobilize phosphorus from strong binding sites of iron minerals has been shown for marine sediments but is less well studied in soils. Here we tested the effect of silicon on phosphorus mobilization for 143 Artic soils (representing contrasting soil characteristics), which have not been affected by agriculture or other anthropogenic management practices. In agreement with marine studies, silicon availabilities were significantly positive correlated to phosphorus mobilization in these soils. Laboratory experiments confirmed that silicon addition significantly increases phosphorus mobilization, by mobilizing Fe(II)-P phases from mineral surfaces. Silicon addition increased also soil respiration in phosphorus deficient soils. We conclude that silicon is a key component regulating mobilization of phosphorous in Arctic soils, suggesting that this may also be important for sustainable management of phosphorus availability in soils in general.

Rice straw increasingly remains on the fields for nutrient supply to the next generation of crop plants. It can be applied either fresh or after burning to black carbon or ash. A central concern during rice cultivation is accumulation of carcinogenic arsenic and the question arises how much rice straw application contributes to nutrient versus arsenic supply in paddy fields. Laboratory incubation experiments were performed to assess the effect of rice straw, black carbon and ash on element mobilization. Our experiments showed initially higher silicon and phosphorus release from black carbon compared to fresh straw amendments. However, more re-sorption to soil lead to finally slightly lower pore water concentrations for black carbon versus fresh straw amendments. Highest arsenic, iron, manganese and dissolved organic carbon concentrations were observed after fresh rice straw application. Black carbon and ash application lead to only minor increases of arsenic compared to controls without amendments. Overall, for silicon and phosphorus the soil acts as sink while for iron and arsenic it was the main source. In summary, burning of rice straw to black carbon prior to application seems to yield a high increase in desired nutrient and a decrease in undesired arsenic mobilization in paddy soils.

Background: Understanding the long-term effect of alemtuzumab on the immune system of multiple sclerosis (MS) patients is crucial., Objective: To report a case of acute sarcoidosis (Löfgren's syndrome) in a relapsing-remitting MS patient, 1.5 years after the second course of alemtuzumab treatment., Case Report: Sarcoidosis was confirmed dermatohistologically, radiologically, and serologically. Analysis of the lymphocyte subpopulations showed a persistent effect of alemtuzumab treatment (CD4/CD8 ratio increased, absolute lymphocyte count of CD19-positive cells increased while CD3/4/8-positive cells were decreased)., Conclusion: Our case highlights the profound effect of alemtuzumab on the immune system and its possible risk for autoimmune complications.
