Nowadays, ENMs/NPLs particles have not yet been extensively measured in the environment, but there is increased concern that this size fraction may be more widely distributed and hazardous than larger-sized particles. This study aimed to examine the bioaccumulation potential of engineered nanomaterials and nanoplastics (ENMs/NPLs) across marine food webs, focusing on plankton communities and commercial fish species (Engraulis encrasicolus and Scomber colias) from the Gulf of Naples. Laboratory experiments on plankton assemblages exposed to fluorescent polystyrene nanoplastics (PS-NPs, 100 nm) for 24h at concentrations ranging from 0.01 to 10 mg/L confirmed nanoplastic uptake in phytoplankton and zooplankton, indicating a dose-dependent internalization in plankton communities. Notably, in natural samples no particles were detected in fish muscle or liver tissues, suggesting limited translocation. Unexpectedly, titanium oxide particles (<1 μm) were found in natural phytoplankton, highlighting the potential presence of other nanoparticles in marine systems. As β-diversity can be seen as a proxy of ecological connections among species assemblages, modeling the decay of similarity in species composition at increasing distance may help elucidate spatial patterns of connectivity and local- to large-scale processes driving community assembly within a marine region. This, in turn, may provide invaluable information for setting ecologically coherent networks of marine protected areas (MPAs) in which protected communities are potentially interrelated and can mutually sustain against environmental perturbations. However, field studies investigating changes in β-diversity patterns at a range of spatial scales and in relation to disturbance are scant, limiting our understanding of how spatial ecological connections among marine communities may affect their recovery dynamics. We carried out a manipulative experiment simulating a strong physical disturbance on subtidal rocky reefs at several locations spanning >1000 km of coast in the Adriatic Sea (Mediterranean Sea) and compared β-diversity patterns and decay of similarity with distance and time by current transport between undisturbed and experimentally disturbed macrobenthic assemblages to shed light on connectivity processes and scales involved in recovery. In contrast to the expectation that very local-scale processes, such as vegetative regrowth and larval supply from neighboring undisturbed assemblages, might be the major determinants of recovery in disturbed patches, we found that connectivity mediated by currents at larger spatial scales strongly contributed to shape community reassembly after disturbance. Across our study sites in the Adriatic Sea, β-diversity patterns suggested that additional protected sites that matched hotspots of propagule exchange could increase the complementarity and strengthen the ecological connectivity throughout the MPA network. More generally, conditional to habitat distribution and selection of sites of high conservation priority (e.g., biodiversity hotspots), setting network internode distance within 100-150 km, along with sizing no-take zones to cover at least 5 km of coast, would help enhance the potential connectivity of Mediterranean subtidal rocky reef assemblages from local to large scale. Very little information is reported for parasites of cnidarians, therefore, the present work aimed to investigate parasitic infections in one of the most widespread jellyfish in the Mediterranean Sea, Rhizostoma pulmo. The goals were to determine prevalence and intensity of parasites in R. pulmo, identify the species involved through morphological and molecular analysis, test whether infection parameters differ in different body parts and in relation to jellyfish size. 58 individuals were collected, 100% of them infected with digenean metacercariae. Intensity varied between 18.7 ± 6.7 per individual in 0-2 cm diameter jellyfish up to 505 ± 50.6 in 14 cm ones. Morphological and molecular analyses suggest that the metacercariae belonged to the family Lepocreadiidae and could be possibly assigned to the genus Clavogalea. Prevalence values of 100% suggest that R. pulmo is an important intermediate host in the life cycle of lepocreadiids in the region. Our findings also support the hypothesis that R. pulmo is an important part in the diet of teleost fish, which are reported as definitive hosts of lepocreadiids, since trophic transmission is necessary for these parasites to complete their life cycles. The invasive macroalga Caulerpa cylindracea has spread widely in the Mediterranean Sea, becoming a favorite food item for native fish for reasons yet unknown. By using a combination of behavioral, morphological, and molecular approaches, herein we provide evidence that the bisindole alkaloid caulerpin, a major secondary metabolite of C. cylindracea , significantly increases food intake in the model fish Danio rerio , influencing the regulation of genes involved in the orexigenic pathway. In addition, we found that the compound improves fish reproductive performance by affecting the hypothalamus-pituitary-gonadal axis. The obtained results pave the way for the possible valorization of C. cylindracea as a sustainable source of a functional feed additive of interest to face critical challenges both in aquaculture and in human nutrition., Competing Interests: The authors declare no conflicts of interest.

The investigation of jellyfish gastrovascular systems mainly focused on stain injections and dissections, negatively affected by thickness and opacity of the mesoglea. Therefore, descriptions are incomplete and data about tridimensional structures are scarce. In this work, morphological and functional anatomy of the gastrovascular system of Rhizostoma pulmo (Macri 1778) was investigated in detail with innovative techniques: resin endocasts and 3D X-ray computed microtomography. The gastrovascular system consists of a series of branching canals ending with numerous openings within the frilled margins of the oral arms. Canals presented a peculiar double hemi-canal structure with a medial adhesion area which separates centrifugal and centripetal flows. The inward flow involves only the "mouth" openings on the internal wing of the oral arm and relative hemi-canals, while the outward flow involves only the two outermost wings' hemi-canals and relative "anal" openings on the external oral arm. The openings differentiation recalls the functional characteristics of a through-gut apparatus. We cannot define the gastrovascular system in Rhizostoma pulmo as a traditional through-gut, rather an example of adaptive convergence, that partially invalidates the paradigm of a single oral opening with both the uptake and excrete function., Competing Interests: The authors have declared that no competing interests exist.

Aquaculture threatens natural resources by fishing down the sea to supply fishmeal. Alternative protein sources in aquafeeds can provide a solution, particularly those that are waste from other operations and thereby reduce feed production costs. Toward this goal, we examined the waste biomass of marine periphyton from biofilters of an integrated multi-trophic aquaculture (IMTA) system as a replacement for fishmeal in diets of gilthead seabream (Sparus aurata). Four isoproteic (41%) and isolipidic (16.7%) aquafeeds were formulated with increased content of periphyton and a corresponding decrease in fishmeal from 20 to 15, 10, or 0%. The growth and biochemical content of seabream fingerlings (initial body weight 10 g) were examined over 132 days. Replacing 50% of fishmeal by waste periphyton improved feed conversion ratio (1.2 vs. 1.35 in the control diet) without harming fish growth. The complete replacement of fishmeal with periphyton resulted in 15% slower growth but significantly higher protein content in the fish flesh (59 vs. 52% in the control diet). Halving fishmeal content reduced feed cost by US$ 0.13 kg -1 feed and saved 30% in the cost of conversion of feed to fish biomass (US$ 0.58 kg -1 produced fish vs. $0.83 in the control diet). Finally, the total replacement of fishmeal by waste periphyton in the diet reduced the fish in-fish out ratio to below 1 (0.5-0.9) as compared to 1.36 in the control diet. Replacing fishmeal with on-farm produced periphyton minimizes aquaculture footprint through the removal of excess nutrients in effluents and the use of waste biomass to reduce the 'fish in' content in aquafeeds and fish production costs. This study evaluated the chronic toxicity (30 days) of different sizes of polyethylene terephthalate (PET) microplastics (60-3000 μm) provided alone or in combination with acid rain, on garden cress (Lepidium sativum). Both biometrical and physiological traits have been evaluated: i) percentage inhibition of seed germination, plant height, leaf number and fresh biomass production; ii) oxidative stress responses (hydrogen peroxide; ascorbic acid and glutathione production); iii) impairment in photosynthetic machinery in term of pigments production; iv) aminolevulinic acid and proline production. Results highlighted that different sizes of PET, alone or in combination with acid rain, are able to negatively affect both biometrical and physiological plant traits. In particular, the lower size of microplastics is able to negatively affect growth and development, as well as to trigger the oxidative burst. Coastal oceans are particularly affected by rapid and extreme environmental changes with dramatic consequences for the entire ecosystem. Seagrasses are key ecosystem engineering or foundation species supporting diverse and productive ecosystems along the coastline that are particularly susceptible to fast environmental changes. In this context, the analysis of phenotypic plasticity could reveal important insights into seagrasses persistence, as it represents an individual property that allows species' phenotypes to accommodate and react to fast environmental changes and stress. Many studies have provided different definitions of plasticity and related processes (acclimation and adaptation) resulting in a variety of associated terminology. Here, we review different ways to define phenotypic plasticity with particular reference to seagrass responses to single and multiple stressors. We relate plasticity to the shape of reaction norms, resulting from genotype by environment interactions, and examine its role in the presence of environmental shifts. The potential role of genetic and epigenetic changes in underlying seagrasses plasticity in face of environmental changes is also discussed. Different approaches aimed to assess local acclimation and adaptation in seagrasses are explored, explaining strengths and weaknesses based on the main results obtained from the most recent literature. We conclude that the implemented experimental approaches, whether performed with controlled or field experiments, provide new insights to explore the basis of plasticity in seagrasses. However, an improvement of molecular analysis and the application of multi-factorial experiments are required to better explore genetic and epigenetic adjustments to rapid environmental shifts. Plastics enter in terrestrial natural system primarily by agricultural purposes, while acid rain is the result of anthropogenic activities. The synergistic effects of microplastics and acid rain on plant growth are not known. In this study, different sizes of polyethylene terephthalate (PET) and acid rain are tested on Lepidium sativum, in two separate experimental sets. In the first one we treated plants only with PET, in the second one we used PET and acid rain together. In both experimentations we analyzed: i) plant biometrical parameters (shoot height, leaf number, percentage inhibition of seed germination, fresh biomass), and ii) oxidative stress responses (hydrogen peroxide; ascorbic acid and glutathione). Results carried out from our experiments highlighted that different sizes of polyethylene terephthalate are able to affect plant growth and physiological responses, with or without acid rain supplied during acute toxicity (6 days). Global change is striking harder and faster in the Mediterranean Sea than elsewhere, where high levels of human pressure and proneness to climate change interact in modifying the structure and disrupting regulative mechanisms of marine ecosystems. Rocky reefs are particularly exposed to such environmental changes with ongoing trends of degradation being impressive. Due to the variety of habitat types and associated marine biodiversity, rocky reefs are critical for the functioning of marine ecosystems, and their decline could profoundly affect the provision of essential goods and services which human populations in coastal areas rely upon. Here, we provide an up-to-date overview of the status of rocky reefs, trends in human-driven changes undermining their integrity, and current and upcoming management and conservation strategies, attempting a projection on what could be the future of this essential component of Mediterranean marine ecosystems., (Copyright © 2021 Elsevier Ltd. This review critically analyses >200 papers collected by searching on Pubmed the word "nanoplastics", a group of emerging contaminants which are receiving growing attention. The present review intends to provide an overview of current knowledge on nanoplastic pollution starting with the theory of polymer degradation, passing to laboratory confirmation of nanoplastic formation and ending with the possible occurrence in sea water samples. Most of the observations proposed focus the attention on polystyrene (PS) because the majority of research knowledge is based on this polymer. Moreover, we thoroughly describe what effects have been observed on different organisms tested in controlled conditions. Nanoplastics formation, fate and toxicity seem to be a very dynamic phenomenon. We compiled an updated global catalogue of parasites in cephalopods. Data were used to assess changes in taxonomic distinctness of parasites over two centuries and across the world's oceans, to quantify turnover and nestedness components of parasite β-diversity, and to attempt estimating their γ-diversity at a global scale. A total of 309 parasites infecting 164 cephalopods were found. We hypothesize that this diversity counts for less than half the potential parasite richness in this molluscan taxon. Taxonomic breadth of parasites was significantly above expectations from null models for Mediterranean Sea and NE Atlantic Ocean, whereas the opposite occurred for NW Pacific Ocean, where a few closely related genera characterized the parasite pool. β-diversity of parasites was very high and dominated by turnover, except for the Atlantic Ocean where a nested pattern among sub-basins emerged. Taxonomic relatedness of parasite species remained substantially unchanged through time, but species replacements largely occurred over the last two centuries. Our findings highlighted potential hotspots of taxonomic distinctness in cephalopod parasites, geographic regions deserving future research, and the need for a deeper understanding of the magnitude of marine parasite diversity, their biogeography, and their role in marine ecosystems. Our global overview may represent a baseline step for future advances in this direction.

This study aims to evaluate the expression of biomarkers of oxidative stress (LPO, GPx, AtCh, SOD) in mussels (Mytilus galloprovincialis) following the exposure to suspensions of microparticles irregular shaped fibres of Polyethylene terephthalate of different sizes (small 5-60 μm, S-PET; medium 61-499 μm, M-PET; large 500-3000 μm, L-PET) at a single dose of 0.1 g/L. Mussels were tested under two different starting pH conditions of marine water: standard (8.0) and acidified (7.5). The results obtained from this study show that: i) PET microplastics are able to induce biochemical stress in mussels; ii) among the biomarkers tested, LPO and GPx were more effective in detecting the stress induced by microplastic in both initial pH conditions; iii) the expression of biomarkers was influenced by the size of the microparticle. This study aims to evaluate effects induced by the exposure of key marine species to leachates and suspensions of different particle-size of PET microparticles, a plastic polymer that is actually considered safe for the environment. Leachates and suspensions of small (5-60 μm); medium (61-499 μm) and large (500-3000 μm) PET were tested on bacteria (V. fischeri; UNI EN ISO 11348-3:2009), algae (P. tricornutum; UNI EN ISO 10253:2016E), and echinoderms (P. lividus; EPA 600/R-95-136/Section 15) species both under standard (8.0) and acidified (7.5) pH conditions. Results obtained show that: i) conversely to larval stage of P. lividus, bacterial and algal tested species are not sensitive to PET pollution under all tested conditions; ii) different tested particle-sizes of PET produce effects that are not always related to their particle-size; iii) differences comparing acidified and standard pH conditions were recorded; iv) concerning echinoderms, food availability produce significant differences compared to fasting conditions; v) special attention on the possible interactions between MPs and other stressors (e.g., food and pH) is needed in order to give a better picture of natural occurring dynamics on marine ecosystems especially in the future frame of global changes., Competing Interests: Declaration of competing interest The authors declare that they have no known competing financial interests or personal relationships that could have appeared to influence the work reported in this paper., (Copyright © 2020. Extensive loss of macroalgal forests advocates for large-scale restoration interventions, to compensate habitat degradation and recover the associated ecological functions and services. Yet, restoration attempts have generally been limited to small spatial extensions, with the principal aim of developing efficient restoration techniques. Here, the success of outplanting Cystoseira amentacea v. stricta germlings cultured in aquaria was experimentally explored at a scale of tens of kms, by means of a multifactorial experimental design. In the intertidal rocky shores of SE Italy, locations with a continuous distribution for hundreds of meters or with few thalli forming patches of few centimeters of C. amentacea canopy were selected. In each location, the effects of adult conspecifics and the exclusion of macrograzers (salema fish and sea urchins) on the survival of germlings were tested. We evaluated the most critical determinants of mortality for germlings, including the overlooked pressure of mesograzers (e.g. amphipods, small mollusks, polychaetes). Despite the high mortality observed during outplanting and early settlement stages, survival of C. amentacea germlings was consistently favored by the exclusion of macrograzers, while the presence of adult conspecifics had no effects. In addition, the cost analysis of the interventions showed the feasibility of the ex-situ method, representing an essential tool for preserving Cystoseira forests. Although the chemical warfare between invasive and native species has become a central problem in invasion biology, the molecular mechanisms by which bioactive metabolites from invasive pests influence local communities remain poorly characterized. This study demonstrates that the alkaloid caulerpin (CAU)-a bioactive component of the green alga Caulerpa cylindracea that has invaded the entire Mediterranean basin-is an agonist of peroxisome proliferator-activated receptors (PPARs). Our interdisciplinary study started with the in silico prediction of the ligand-protein interaction, which was then validated by in vivo, ex vivo and in vitro assays. On the basis of these results, we candidate CAU as a causal factor of the metabolic and behavioural disorders observed in Diplodus sargus , a native edible fish of high ecological and commercial relevance, feeding on C. cylindracea . Moreover, given the considerable interest in PPAR activators for the treatment of relevant human diseases, our findings are also discussed in terms of a possible nutraceutical/pharmacological valorisation of the invasive algal biomasses, supporting an innovative strategy for conserving biodiversity as an alternative to unrealistic campaigns for the eradication of invasive pests.

The biological invasion of the green algae Caulerpa cylindracea represents a serious scientific and public issue in the Mediterranean Sea, essentially due to strong modifications both to habitat structure and native benthic communities. Although alterations in health status and changes in flesh quality of some marine species (dietary exposed to C. cylindracea ) have been observed, no studies on cause-effect relationships have been carried out. Here, for the first time, through a controlled feeding experiment followed by ¹H NMR Spectroscopy and multivariate analysis (PCA, OPLS-DA), we showed that caulerpin taken with diet is directly responsible of changes observed in metabolic profile of fish flesh, including alteration of lipid metabolism, in particular with a reduction of ω3 PUFA content. The potential of caulerpin to directly modulate lipid metabolism opens up new questions about causal mechanism triggered by algal metabolite also in view of a possible exploitation in the nutraceutical/medical field.

Raphidascarididae are among the most abundant and widespread parasitic nematodes in the marine environment. The life-cycle of most raphidascaridid species is poorly known and information about their distribution and host range is lacking in many geographical areas, as is the taxonomy of several species. A study of larval and adult stages of Hysterothylacium fabri (Rudolphi, 1819) Deardorff and Overstreet, 1980 (Nematoda: Raphidascarididae) infecting the striped goatfish Mullus surmuletus Linnaeus, 1758 (Mullidae) and the Mediterranean stargazer Uranoscopus scaber Linnaeus, 1759 (Uranoscopidae) from the Ionian Sea (central Mediterranean) has been carried out by combining light and scanning electron microscopy observations and molecular analyses through polymerase chain reaction-restriction fragment length polymorphism and sequencing of the ITS rDNA gene. Results indicate that U. scaber and M. surmuletus represent suitable definitive and intermediate/paratenic hosts of H. fabri, respectively, in the Mediterranean and highlight the importance of combining genetic and morphological data to study the taxonomy and epidemiology of parasites widely distributed in different fish species and aquatic ecosystems.

Most of accumulation curves tend to underestimate species richness, as they do not consider spatial heterogeneity in species distribution, or are structured to provide lower bound estimates and limited extrapolations. The total-species (T-S) curve allows extrapolations over large areas while taking into account spatial heterogeneity, making this estimator more prone to attempt upper bound estimates of regional species richness. However, the T-S curve may overestimate species richness due to (1) the mismatch among the spatial units used in the accumulation model and the actual units of variation in β-diversity across the region, (2) small-scale patchiness, and/or (3) patterns of rarity of species. We propose a new framework allowing the T-S curve to limit overestimation and give an application to a large dataset of marine mollusks spanning over 11 km 2 of subtidal bottom (W Mediterranean). As accumulation patterns are closely related across the taxonomic hierarchy up to family level, improvements of the T-S curve leading to more realistic estimates of family richness, that is, not exceeding the maximum number of known families potentially present in the area, can be considered as conducive to more realistic estimates of species richness. Results on real data showed that improvements of the T-S curve to accounts for true variations in β-diversity within the sampled areas, small-scale patchiness, and rarity of families led to the most plausible richness when all aspects were considered in the model. Data on simulated communities indicated that in the presence of high heterogeneity, and when the proportion of rare species was not excessive (>2/3), the procedure led to almost unbiased estimates. Our findings highlighted the central role of variations in β-diversity within the region when attempting to estimate species richness, providing a general framework exploiting the properties of the T-S curve and known family richness to estimate plausible upper bounds in γ-diversity.

Coccidian parasites of the genus Aggregata are known to parasitize cephalopods as definitive hosts, however one of the genus members, A. octopiana, has shown an unresolved phylogeny within the same definitive host, the common octopus (Octopus vulgaris). Our study represents a large-scale investigation aimed at characterizing morphological traits and phylogeny of A. octopiana isolated from O. vulgaris inhabiting three distinct geographic areas of the central Mediterranean: The Adriatic, Ionian and Tyrrhenian Seas. The morphology of sporogonic stages of the parasite in octopus tissues was assessed by light and electron microscopy; molecular characterization has been carried out using the 18S rRNA locus. Our results support the hypothesis that two morphologically and genetically different A. octopiana infect O. vulgaris in the investigated areas of the Mediterranean Sea. The invasive green alga Caulerpa cylindracea has become an important component of the diet of the Mediterranean white seabream Diplodus sargus. As a consequence of this "exotic diet", the algal bisindolic alkaloid caulerpin accumulates in the fish tissues. Although the compound shows structural similarity to endogenous indolamines that modulate animal behaviour, the potential impact of caulerpin on fish behaviour still remains unexplored. In this report, behavioural experiments both on groups and on single fish responding towards a mirror were performed under different doses of dietary caulerpin. Differences between treated and control groups for each behaviour and for the overall aggressive pattern during the different experimental phases showed that the aggressiveness of D. sargus decreased with the administration of caulerpin. These results call the attention to a still unexplored potential ability of bioactive metabolites from marine invasive species, to alter the behaviour on native species, with putative negative effects on patterns of fish growth and population dynamics.

The red pigment caulerpin, a secondary metabolite from the marine invasive green algae Caulerpa cylindracea can be accumulated and transferred along the trophic chain, with detrimental consequences on biodiversity and ecosystem functioning. Despite increasing research efforts to understand how caulerpin modifies fish physiology, little is known on the effects of algal metabolites on mammalian cells. Here we report for the first time the mitochondrial targeting activity of both caulerpin, and its closely related derivative caulerpinic acid, by using as experimental model rat liver mitochondria, a system in which bioenergetics mechanisms are not altered. Mitochondrial function was tested by polarographic and spectrophotometric methods. Both compounds were found to selectively inhibit respiratory complex II activity, while complexes I, III, and IV remained functional. These results led us to hypothesize that both algal metabolites could be used as antitumor agents in cell lines with defects in mitochondrial complex I. Ovarian cancer cisplatin-resistant cells are a good example of cell lines with a defective complex I function on which these molecules seem to have a toxic effect on proliferation. Assessing the distribution and intensity of human threats to biodiversity is a prerequisite for effective spatial planning, harmonizing conservation purposes with sustainable development. In the Mediterranean Sea, the management of Marine Protected Areas (MPAs) is rarely based on explicit consideration of the distribution of multiple stressors, with direct assessment of their effects on ecosystems. This gap limits the effectiveness of protection and is conducive to conflicts among stakeholders. Here, a fine scale assessment of the potential effects of different combinations of stressors (both land- and marine-based) on vulnerable rocky habitats (i.e. lower midlittoral and shallow infralittoral) along 40 km of coast in the western Mediterranean (Ionian Sea) has been carried out. The study area is a paradigmatic example of socio-ecological interactions, where several human uses and conservation measures collide. Significant differences in the structure of assemblages according to different combinations of threats were observed, indicating distinct responses of marine habitats to different sets of human pressures. A more complex three-dimensional structure, higher taxon richness and β-diversity characterized assemblages subject to low versus high levels of human pressure, consistently across habitats. In addition, the main drivers of change were: closeness to the harbour, water quality, and the relative extension of beaches. Our findings suggest that, although efforts to assess cumulative impacts at large scale may help in individuating priority areas for conservation purposes, the fact that such evaluations are often based on expert opinions and not on actual studies limits their ability to represent real environmental conditions at local scale. Systematic evaluations of local scale effects of anthropogenic drivers of change on biological communities should complement broad scale management strategies to achieve effective sustainability of human exploitation of marine resources.

The green alga Caulerpa cylindracea is a non-autochthonous and invasive species that is severely affecting the native communities in the Mediterranean Sea. Recent researches show that the native edible fish Diplodus sargus actively feeds on this alga and cellular and physiological alterations have been related to the novel alimentary habits. The complex effects of such a trophic exposure to the invasive pest are still poorly understood. Here we report on the metabolic profiles of plasma from D. sargus individuals exposed to C. cylindracea along the southern Italian coast, using 1H NMR spectroscopy and multivariate analysis (Principal Component Analysis, PCA, Orthogonal Partial Least Square, PLS, and Orthogonal Partial Least Square Discriminant Analysis, OPLS-DA). Fish were sampled in two seasonal periods from three different locations, each characterized by a different degree of algal abundance. The levels of the algal bisindole alkaloid caulerpin, which is accumulated in the fish tissues, was used as an indicator of the trophic exposure to the seaweed and related to the plasma metabolic profiles. The profiles appeared clearly influenced by the sampling period beside the content of caulerpin, while the analyses also supported a moderate alteration of lipid and choline metabolism related to the Caulerpa-based diet.

Hydrozoa of the Mediterranean Sea are well known and a recent monograph covers 457 species. Mediterranean non-Siphonophoran Hydrozoa comprises 398 species, an increasing number due to continuous updates, representing about 10 % of the 3,702 currently valid species reported in a recent world assessment of hydrozoan diversity. Many new records are non indigenous species, previously described species that occurred elsewhere and whose arrival was presumably caused by human activities. However, many species reported in the past are not recorded in recent times. Realistic assessments of species pools require addition of new species, but also subtraction of species not found since a certain period. With the confidence of extinction index, cases of putative extinction can be raised. Out of the 398 known species, only 162 (41 %) have been reported in the last decade, while 53 (13 %) are not recorded in the literature since at least 41 years. According to the confidence of extinction index, 60 % of the 53 missing species are extinct, and 11 % are putatively extinct from the basin. From a biogeographical point of view, the missing species are: 34 % endemic, 19 % boreal, 15 % Mediterranean-Atlantic, 11 % Indo-Pacific, 11 % circumtropical, 4 % cosmopolitan, 2 % tropical-Atlantic, 4 % non-classifiable. Fluctuations in species composition into a certain area cause heavy variability in the expression of both structural and functional biodiversity. As consequence, the regional biodiversity should be analyzed through its temporal evolution, to detect changes and their possible causes. This approach has profound consequences on biodiversity assessments and also on the compilation of red lists.

Rare evidences support that Marine Protected Areas (MPAs) enhance the stability of marine habitats and assemblages. Based on nine years of observation (2001-2009) inside and outside a well managed MPA, we assessed the potential of conservation and management actions to modify patterns of spatial and/or temporal variability of Posidonia oceanica meadows, the lower midlittoral and the shallow infralittoral rock assemblages. Significant differences in both temporal variations and spatial patterns were observed between protected and unprotected locations. A lower temporal variability in the protected vs. unprotected assemblages was found in the shallow infralittoral, demonstrating that, at least at local scale, protection can enhance community stability. Macrobenthos with long-lived and relatively slow-growing invertebrates and structurally complex algal forms were homogeneously distributed in space and went through little fluctuations in time. In contrast, a mosaic of disturbed patches featured unprotected locations, with small-scale shifts from macroalgal stands to barrens, and harsh temporal variations between the two states. Opposite patterns of spatial and temporal variability were found for the midlittoral assemblages. Despite an overall clear pattern of seagrass regression through time, protected meadows showed a significantly higher shoot density than unprotected ones, suggesting a higher resistance to local human activities. Our results support the assumption that the exclusion/management of human activities within MPAs enhance the stability of the structural components of protected marine systems, reverting or arresting threat-induced trajectories of change.

The available taxonomic expertise and knowledge of species is still inadequate to cope with the urgent need for cost-effective methods to quantifying community response to natural and anthropogenic drivers of change. So far, the mainstream approach to overcome these impediments has focused on using higher taxa as surrogates for species. However, the use of such taxonomic surrogates often limits inferences about the causality of community patterns, which in turn is essential for effective environmental management strategies. Here, we propose an alternative approach to species surrogacy, the "Best Practicable Aggregation of Species" (BestAgg), in which surrogates exulate from fixed taxonomic schemes. The approach uses null models from random aggregations of species to minimizing the number of surrogates without causing significant losses of information on community patterns. Surrogate types are then selected in order to maximize ecological information. We applied the approach to real case studies on natural and human-driven gradients from marine benthic communities. Outcomes from BestAgg were also compared with those obtained using classic taxonomic surrogates. Results showed that BestAgg surrogates are effective in detecting community changes. In contrast to classic taxonomic surrogates, BestAgg surrogates allow retaining significantly higher information on species-level community patterns than what is expected to occur by chance and a potential time saving during sample processing up to 25% higher. Our findings showed that BestAgg surrogates from a pilot study could be used successfully in similar environmental investigations in the same area, or for subsequent long-term monitoring programs. BestAgg is virtually applicable to any environmental context, allowing exploiting multiple surrogacy schemes beyond stagnant perspectives strictly relying on taxonomic relatedness among species. This prerogative is crucial to extend the concept of species surrogacy to ecological traits of species, thus leading to ecologically meaningful surrogates that, while cost effective in reflecting community patterns, may also contribute to unveil underlying processes. A specific R code for BestAgg is provided.

A large number of bacteria, including agents responsible for diseases, characterise sewage-polluted seawaters. Apart from standards for bathing waters and bivalve aquaculture waters, there are no general microbiological standards applicable to seawaters to help decide if bacterial pollution is within acceptable ranges. This study represents an attempt towards the issue of comparing the susceptibility of different marine invertebrates subjected to polluted seawater with a high microbial contamination. We explored the survival rates and the microbiological accumulation of mollusc bivalves, echinoderms and crustaceans species exposed to sewage-polluted seawaters. Microbiological analyses were performed on the polluted seawater and on the homogenates of exposed and unexposed specimens. Culturable bacteria (22 °C and 37 °C) and microbial pollution indicators (total coliforms, Escherichia coli and intestinal enterococci) were measured. When exposed to the sewage-polluted seawater, the examined invertebrates showed different survival rates. In the filter feeders, bacterial densities at 22 °C and 37 °C rose after 96 h of exposure to sewage. The highest concentrations of total coliforms and intestinal enterococci were found in exposed bivalve Mytilus galloprovincialis. The concentrations of bacteria growing at 37 °C were lower in the exposed deposit feeders compared to the polluted seawater. Some yeasts were absent in several exposed species although these yeasts were present in the polluted seawater. Our data suggest that the examined filter feeders, given their capability to survive and accumulate bacteria, may counteract the effects of sewage and restore seawater quality.

Background: Betanodaviruses are the causative agents of Viral Encephalopathy and Retinopathy (VER). To date, more than 50 species have proved to be susceptible and among them, those found in genus Epinephelus are highly represented. Clinical disease outbreaks are generally characterized by typical nervous signs and significant mortalities mainly associated with aquaculture activities, although some concerns for the impact of this infection in wild fish have been raised. In this study, the authors present the first documented report describing an outbreak of VER in wild species in the Mediterranean basin., Case Presentation: In late summer--early winter 2011 (September-December), significant mortalities affecting wild Dusky grouper (Epinephelus marginatus), Golden grouper (Epinephelus costae) and European sea bass (Dicentrarchus labrax) were reported in the municipality of Santa Maria di Leuca (Northern Ionian Sea, Italy). The affected fish showed an abnormal swimming behavior and swollen abdomens. During this epizootic, five moribund fish showing clear neurological signs were captured and underwent laboratory investigations. Analytical results confirmed the diagnosis of VER in all the specimens. Genetic characterization classified all betanodavirus isolates as belonging to the RGNNV genotype, revealing a close genetic relationship with viral sequences obtained from diseased farmed fish reared in the same area in previous years., Conclusion: The close relationship of the viral sequences between the isolates collected in wild affected fish and those isolated during clinical disease outbreaks in farmed fish in the same area in previous years suggests a persistent circulation of betanodaviruses and transmission between wild and farmed stocks. Further investigations are necessary to assess the risk of viral transmission between wild and farmed fish populations, particularly in marine protected areas where endangered species are present.

The effectiveness of secondary vs primary treatments of wastewaters in mitigating the effects of sewage discharge on the multivariate structure and diversity of rocky invertebrate assemblages was assessed over a nine-year period through a beyond-BACI experimental design. Assemblages from different tidal levels (i.e. mid-shore, low-shore, 3 m and 8 m subtidal) were sampled at the impact location (I) and three control locations (Cs) at a hierarchy of spatial scales. The improvement in water treatment significantly changed the structure, diversity and cover of low intertidal assemblages. Faunal cover at 8 m subtidal increased significantly after the treatment upgrade at I. The secondary treatment also affected patterns of spatial heterogeneity between I and Cs for mid-shore and 3 m subtidal assemblages. This study demonstrates that powerful experimental designs combined with univariate and multivariate analytical approaches are fundamental in distinguishing the subtle effects of human impact from those of natural processes., (Copyright © 2012 Elsevier Ltd. The spatial distributions of marine fauna and of pollution are both highly structured, and thus the resulting high levels of autocorrelation may invalidate conclusions based on classical statistical approaches. Here we analyse the close correlation observed between proxies for the disturbance associated with gas extraction activities and amphipod distribution patterns around four hydrocarbon platforms. We quantified the amount of variation independently accounted for by natural environmental variables, proxies for the disturbance caused by platforms, and spatial autocorrelation. This allowed us to demonstrate how each of these three factors significantly affects the community structure of amphipods. Sophisticated statistical techniques are required when taking into account spatial autocorrelation; nevertheless our data demonstrate that this approach not only enables the formulation of robust statistical inferences but also provides a much deeper understanding of the subtle interactions between human disturbance and natural factors affecting the structure of marine invertebrates communities., (Copyright © 2012 Elsevier Ltd. Taxonomic distinctness has been applied successfully for the exploration of biodiversity patterns, yet its relevance in environmental impact assessment is far from being unquestioned. In this study, we assessed the potential of taxonomic distinctness to discern perturbed and unperturbed sites by analysing Mediterranean nematode assemblages. Geographic and habitat-related effects on the performance of the index were also explored. Above all, our findings do not corroborate the conjecture that taxonomic distinctness could be largely unaffected by natural variability, habitat features, and biogeographic context, casting doubts on potential generalization concerning its application as an indicator of environmental stress. Taxonomic distinctness represents an excellent metric to identifying taxonomic properties of ecological systems but, as for other ecological indices, it should be viewed as a complementary tool in environmental impact assessment, due to its sensitiveness to specific environmental features of systems being investigated., (Copyright © 2012 Elsevier Ltd. The green alga Caulerpa racemosa var. cylindracea has invaded Mediterranean seabed including marine reserves, modifying the structure of habitats and altering the distributional patterns of associated organisms. However, the understanding of how such invasion can potentially affect functional properties of Mediterranean subtidal systems is yet to be determined. In this study, we show that C. racemosa changes foraging habit of the native white seabream, Diplodus sargus. In invaded areas, we found a high frequency of occurrence of C. racemosa in the stomach contents of this omnivorous fish (72.7 and 85.7%), while the alga was not detected in fish from a control area. We also found a significant accumulation of caulerpin, one of the main secondary metabolites of C. racemosa, in fish tissues. The level of caulerpin in fish tissues was used here as an indicator of the trophic exposure to the invasive pest and related with observed cellular and physiological alterations. Such effects included activation of some enzymatic pathways (catalase, glutathione peroxidases, glutathione S-transferases, total glutathione and the total oxyradical scavenging capacity, 7-ethoxy resorufin O-deethylase), the inhibition of others (acetylcholinesterase and acylCoA oxidase), an increase of hepatosomatic index and decrease of gonadosomatic index. The observed alterations might lead to a detrimental health status and altered behaviours, potentially preventing the reproductive success of fish populations. Results of this study revealed that the entering of alien species in subtidal systems can alter trophic webs and can represent an important, indirect mechanism which might contribute to influence fluctuations of fish stocks and, also, the effectiveness of protection regimes.

Taxonomic Sufficiency (TS) has been proposed as a short-cut method to quantify changes of biological assemblages in environmental monitoring. However, issues about the efficacy of taxonomic surrogates in depicting long-term temporal patterns of marine assemblages are still scant. Here we report on the adoption of TS combined with data transformations to describe patterns of North Adriatic polychaete assemblages through 20 years. Univariate and multivariate analyses revealed large spatial-temporal variation characterizing the assemblages. The efficiency to discriminate between the two research periods (1990-1994 vs. 2004-2008) was reduced when data were analyzed at family and order level leading to misinterpreting the sources of assemblage variation. Further information was lost with data transforms. Families may represent appropriate assemblages' descriptors in long-term monitoring, but using TS coupled with data transformations could lead to hazardous loss of information. We suggest that periodical analysis at fine taxonomic level should be routinely alternated to long-term monitoring based on TS in order to check its effectiveness., (Copyright © 2010 Elsevier Ltd. Posidonia oceanica meadows can be severely damaged by dredge-fill operations. We report on the construction of gas pipelines that occurred between 1981 and 1993 in SW Sicily, Italy. A large portion of the meadow was mechanically removed, and the excavated trench was filled with a mosaic of substrates, ranging from sand to consolidated rock debris. Meadow loss and recovery were quantified over 7 years after the end of operations. We recorded an overall loss of 81.20 ha of meadow. Substrate strongly affected recovery as the percent cover by P. oceanica consistently increased on calcareous rubble, reaching values of 44.37 ± 3.05% in shallow sites after 7 years, whereas no significant increase occurred on other substrates. As in the Mediterranean Sea exploitation of coastal areas continues to grow with consequent impacts on P. oceanica meadows, this case study illustrates how artificial rubble-like materials could be employed to support the restoration of damaged meadows., (Copyright © 2010 Elsevier Ltd. Taxonomic Sufficiency (TS) is a promising analysis technique, particularly in light of the current need for rapid and reliable procedures in marine impact assessment and monitoring. However, generalizations are still difficult and there are few studies comparing the effectiveness of TS under different environmental settings. The present study investigates whether reduced taxonomy can be used to detect natural and human-driven patterns of variation in mollusk and polychaete assemblages from subtidal soft and hard bottoms in the Mediterranean. Results showed that, unlike in polychaetes, mollusk families represent effective taxonomic surrogates across a range of environmental contexts. These findings suggest that the mechanisms behind TS in mollusks could act homogeneously across habitats and environmental conditions. In contrast, multiple factors could interact to determine the robustness of polychaetes to taxonomic aggregation. This study highlights the need to go beyond the current pragmatism in this field of work and focus on the reasons underlying TS effectiveness in order to provide a general framework on the application of taxonomic surrogates in marine systems.

Substratum type and topographic complexity influence the settlement and persistence of benthic organisms. However, the combined effect of these two factors in affecting colonization patterns at different scales has rarely been investigated. A manipulative experiment was conducted to test the interplay of rock type and roughness in affecting the pattern of subtidal assemblages and to provide tests for the generality of effects across a range of spatial scales (centimetres to hundreds of metres). Replicate tiles of four different rock types, with two levels of surface roughness were deployed in rocky subtidal habitats (5 m depth) at two sites (separated by hundreds of metres) at each of three locations (separated by tens of kilometres). Spatial and temporal variation in the colonization patterns over 9 months differed among rock types. However, large-scale processes appeared to be far more important than substratum type or roughness in determining assemblage structure. Predicting the consequences of the introduction of artificial structures into the coastal marine environment is critical as increasingly parts of coastlines are being modified within the Mediterranean and other regions. The results suggest that further investment is needed to manage and mitigate the effects of the deployment of artificial structures in coastal areas.

Imposex, i.e. the development of additional male sex organs (penis and/or vas deferens), in females of gonochorist marine and freshwater gastropods, is known to be caused by tributyltin (TBT), and it has been widely used as a biomonitoring tool in environmental surveys for TBT pollution assessment. In this study, we experimentally tested the potential to induce imposex by another endocrine disruptor (polychlorinated biphenyls [PCBs] mixture--Aroclor 1260). Adults of Hexaplex trunculus with low imposex level, coming from an Italian Marine Protected Area, were injected separately with different doses of tributyltin chloride (TBTCl) and Aroclor 1260. The compounds were dissolved in ethanol and the organisms were narcotised by immersion in MgCl(2) solution before injection. Before and after the experiment, butyltin compounds (BuTs) and PCB tissue concentrations were determined. A significant increase in imposex with respect to non-treated organisms was observed in all treatments, including artefact controls. No clear correlation was observed between BuTs and PCB tissue concentrations and indices of imposex incidence. Based on these results, no assumption can be formulated about PCB effect on imposex development. Nevertheless, they suggest that the imposex level increase, at least in H. trunculus, in laboratory conditions might not be caused by TBT only, but it would rather be a non-specific response to different stress stimuli.

The exploitation of fossil fuels in the Mediterranean Sea will likely lead to an increase in the number of offshore platforms, a recognized threat for marine biodiversity. To date, in this basin, few attempts have been made to assess the impact of offshore gas and oil platforms on the biodiversity of benthic assemblages. Here, we adopted a structured experimental design coupled with high taxonomic resolution to outline putative effects of gas platforms on soft-bottom macrofauna assemblages in the North Ionian Sea. The analysis was based on a total of 20,295 specimens of 405 taxa, almost entirely identified at species level. Multivariate and univariate analyses showed idiosyncratic patterns of assemblage change with increasing distance from the platforms. Potential reasons underlying such inconsistency are analyzed and the view that structured experimental monitoring is a crucial tool to quantify the extent and magnitude of potential threats and to provide sound baseline information on biodiversity patterns is supported.

The present study investigates the effect of the filtering activity of Sabella spallanzanii on the culturable heterotrophic bacterial community through the comparison of the bacterial diversity in transplanted polychaetes and the surrounding seawater. For isolation of culturable heterotrophic bacteria, seawater samples as well as polychaete homogenates were plated in triplicates onto Bacto Marine Agar 2216 (Difco). All the colonies grown were isolated, subcultured and identified by several morphological, biochemical and cultural methods. Some bacterial genera showed higher average abundances in polychaetes than in seawater (i.e. Lucibacterium and Photobacterium). Aeromonas represented a conspicuous component of the bacterial community both in S. spallanzanii and seawater. The presence of Cytophaga and Pseudomonas was also relevant in the examined seawater samples. The selective concentrations of some bacterial genera inside S. spallanzanii either by grazing on bacteria or their capability as bacterial reservoirs, provides evidence for the role of macrobenthic invertebrates as key determinants for microbial diversity.

Understanding whether Marine Protected Areas (MPAs) can be considered as a suitable tool for restoring the structure and function of populations and assemblages is urgently needed to achieve an effective policy of mitigation of human impact in coastal management. However, to date, the role played by MPAs in enhancing ecosystems resilience has been more advocated than unambiguously documented. 2. This study was designed to test whether full protection in marine reserves facilitates recovery of benthos impacted by the date mussel Lithophaga lithophaga fishery, one of the most harmful human activities affecting subtidal rocky habitats in the Mediterranean Sea. 3. The effects of this destructive fishery were reproduced at one fully protected location (P) and at two unprotected control locations (Cs) in the SW Mediterranean Sea. At each location, three plots (4 m2) of rocky surface at 4-6 m depth were disturbed experimentally, while another three plots served as reference. In each plot, the species composition and relative cover of the sessile benthic assemblages were sampled photographically on each of five occasions during a period of 20 months. 4. Over and above variation in habitat features among locations, multivariate and univariate analyses revealed significant differences between P-vs.-Cs in patterns of assemblage recovery and showed that, at the fully protected location, recovery was faster than at the unprotected control locations. 5. Our results suggest that MPAs have the potential to change the trajectories of recovery of disturbed assemblages by accelerating the processes of recolonization and call for further investigation to identify the specific mechanisms underlying increased resilience.

Marine protected areas (MPAs) are aimed at managing and protecting marine environments. Their design, however, often disregards both a thorough knowledge of the distribution of habitats and assemblages and the use of proper experimental evaluations of the efficacy of MPAs by comparing protected vs. unprotected zones. About 200 MPAs have been recently instituted in the Mediterranean area, but the evidence of their efficacy is scant. The MPA of Torre Guaceto (Southern Adriatic Sea, Italy) is one of the rare cases of effective protection enforcement. The reserve was instituted more than 10 years ago, a period currently considered as sufficient to show responses by organisms to protection. The MPA is divided into a C zone, the general reserve, where many activities are permitted, a B zone, the partial reserve where restrictions increase and two A zones, the integral reserve where access is prohibited. The goals of the paper were to map the distribution of benthic assemblages to assess if they were properly represented in the differently protected zones, and to test the efficacy of protection by quantifying possible differences between the assemblages in two control areas and in the two A zones, where human impact is completely excluded. The analysis of habitat and assemblage distribution within the MPA showed that the zones with total protection do not include most valuable environmental types. Most of the considered variables (i.e. cover of substratum, number of taxa, and average abundance of the most common taxa) were not significantly different in and out of the A zones, at each time of sampling. Results, however, suggested a possible effect of protection in modifying patterns of abundance of sponges under Cystoseira canopy (more abundant in the fully protected zone). In the subtidal habitat, differences were found in the structure of the whole assemblage and in the abundance of encrusting coralline red algae (more abundant outside the fully protected area). Notwithstanding the correct general methodology employed in the study, a lack of statistical power could have a role in preventing the detection of ecologically relevant effects of protection. In some instances, data pooling allowed a discrimination between cases where there was clearly no effect of protection and cases where there might be. On this basis, the optimization of this experimental design should be considered in further studies. In any case, if the goals of MPAs have not been clearly stated, efficacy of protection might prove very difficult to test even with the use of sound experimental designs.

This study examined the defensive ability of human dental pulp against H2O2 in healthy and reversible and irreversible pulpitis tissues through determination of catalase activity by spectrophotometric methods. Thirty-five systemically healthy patients were donors of the pulp tissue, and pulp conditions were assessed using clinical and X-ray evaluations. Catalase activity was 1.61 +/- 0.23 U mg(-1) protein in the healthy tissues, 2.99 +/- 0.45 U mg(-1) protein in the reversible pulpitis tissues, and 2.44 +/- 467 mU mg(-1) protein in the irreversible pulpitis tissues. All differences between the groups were statistically significant. These results point to a role for catalase during dental pulp inflammation in humans, and therefore demonstrate an inherent biological defense system against reactive oxidants in human dental pulp.
