Gestation length (GL) can potentially affect health and performance of both the dam and the newborn calf, and it is controlled by two genetic components, direct and maternal. This means that both the calf (direct effect) and the cow (maternal effect) genotypes contribute to determine GL and its variability. The aims of the present study were to estimate direct and maternal variance components of GL, develop a routine genetic evaluation of GL in Italian Holstein and evaluate potential (un)favourable associations with traits for which selection is undertaken in this population. A multiple-trait repeatability linear animal model was employed for the estimation of variance components considering GL in first and later parities as different traits. The posterior mean (PM) of heritability of the direct effect was 0.43 for first parity and 0.35 for later parities. The PM of heritability of the maternal effect was lower, being 0.08 for primiparae and 0.06 for pluriparae. The posterior standard deviation (PSD) of the heritability estimates was small, ranging from 0.001 to 0.005. The relationship of direct and maternal effects with important traits such as milk yield and fertility indicated that selecting for extreme GL, longer or shorter, may have negative consequences on several traits, suggesting that GL has an intermediate optimum in dairy cattle. In conclusion, this study reveals that selecting an intermediate GL in the Italian Holstein population is advisable. Although scarcely variable compared to other conventional traits for which Italian Holstein is selected, GL is heritable and a deeper knowledge can be useful for decision-making at the farm level., (© 2023 The Authors. The present study aimed to investigate the association between stayability (STAY) traits, muscularity, and body condition score (BCS) in the Italian Simmental dual-purpose cows. Data were collected from 2,656 cows linearly scored in their first lactation from 2002 to 2020 and reared in 324 herds. The binary trait STAY, which is the ability of a cow to stay in the herd, was obtained for each cow-lactation available up to parity 5 (from STAY1-2 to STAY4-5). Analysis of STAY was carried out using logistic regression, considering the fixed effect of energy corrected milk, conception rate, somatic cell score, and muscularity or BCS predicted at different time points. The herd of linear classification and residual error were the random effects. Primiparous cows with a medium BCS and muscularity in early lactation presented a more favorable STAY across life compared to thinner ones ( P < 0.05). In fact, cows with an intermediate BCS/muscularity were more likely to stay in the herd after the third lactation (STAY3-4), compared to those presenting a lower BCS/muscularity ( P < 0.01). However, cows whose muscularity was high were generally less likely to start the third lactation compared to the others. A potential explanation for this could be the willing to market cows with good conformation for meat purpose. Simmental is in fact a dual-purpose breed known for the good carcass yield and meat quality. Background: Colostral concentration of immunoglobulins (Ig) is crucial for the passive transfer of antibodies from the cow to the new-born calf. Direct determination of Ig by the gold standard radial immunodiffusion method is demanding in terms of time and costs. For this reason, a refractometer is commonly used at the farm level for an indirect estimation of colostrum quality, which is given as the Ig concentration. In this study, colostrum samples were collected from 548 Italian Holstein cows within 6 h of calving. The refractive index (BRIX, %) of these samples was assessed using a portable optical refractometer, as well as the concentration of total protein, IgG, IgA, and IgM by radial immunodiffusion. A four-trait animal model was used to estimate genetic parameters for BRIX and the different immunoglobulin isotypes. A receiver operating characteristic analysis was carried out to evaluate the BRIX diagnostic accuracy., Results: Colostral BRIX was moderately heritable (0.26) and its genetic and phenotypic correlations with IgG (0.91, 0.78), IgA (0.57, 0.57), and IgM (0.71, 0.61) were all positive and of similar order, although the genetic correlations were generally higher than the phenotypic correlations. Low-quality colostrum samples, defined as those with an IgG concentration lower than 50 g/L, were accurately identified by the refractive index on the BRIX scale, with an area under the curve of 0.90., Conclusions: The use of a refractometer is recommended on dairy farms to produce a proxy for colostral Ig concentration. BRIX is a useful phenotyping tool that can be used in cattle to improve the quality of colostrum for first feeding of calves through both traditional genetic and genomic strategies. To date, especially in Europe, few studies have analysed the implications of long journeys on pig welfare and economic losses, expressed in terms of transport mortality. This study retrospectively analysed data collected from slaughtering registers and travel journals in a large Italian abattoir. We focused on pig transports coming from abroad and arriving at the slaughter plant after long journeys (a total of 59,982 pigs over 370 journeys). We explored the relationship between mortality and the following variables: country of origin, journey duration, astronomical season, stocking density on the truck, number of stops, and prolonged stops during the journey (lasting more than 60 min, likely due to traffic jams or truck problems). Overall, the low mortality rate observed (0.09%) was in line with European estimates. The factors with a significant or tendential effect on mortality during transport were the astronomical season ( p = 0.0472, with higher mortality in spring) and the presence of prolonged stops during the journey ( p = 0.069, tendential effect). Journey duration, stocking density, country of origin, and the number of stops were not statistically significant. In conclusion, based on this case study, using transport mortality combined with data collected during the common routine activity by the Public Veterinary Services in slaughterhouses could be a simple screening method for identifying problematic journeys or transport conditions.

Considerable resources are required to routinely measure detailed milk compositional traits. Hence, an insufficient volume of phenotypic data can hinder genetic progress in these traits within dairy cow breeding programmes. The objective of the present study was to quantify the opportunities for breeding for improved milk protein and free amino acid (FAA) composition by exploiting mid-infrared spectroscopy (MIRS) predictions routinely recorded from milk samples. Genetic parameters for protein fractions and FAA composition were estimated using 134,546 test-day records from 16,166 lactations on 9,572 cows using linear mixed models. Heritability of MIRS-predicted protein fractions ranged from 0.19 (α-lactalbumin) to 0.55 (β-lactoglobulin A), while heritability of MIRS-predicted FAA ranged from 0.08 for glycine to 0.29 for glutamic acid. Genetic correlations among the MIRS-predicted FAA were moderate to strong ranging from -0.44 (aspartic acid and lysine) to 0.97 (glutamic acid and total FAA). Adjustment of the genetic correlations for the genetic merit of 24-h milk yield did not greatly affect the correlations. Results from the current study highlight the presence of exploitable genetic variation for both protein fractions and FAA in dairy cow milk. Besides, the direction of genetic correlations reveals that breeding programmes directly selecting for greater milk protein concentration carry with them favourable improvement in casein and whey fractions., (© 2022 The Authors. Background: The quality of colostrum administered to calves is based on its concentration in immunoglobulins G (IgG, g/L). Immunoglobulins A (IgA) and M (IgM) are also present but at a lower level. The gold standard reference analysis for these traits, radial immunodiffusion, is time-consuming and expensive. In order to define breeding strategies that are aimed at improving colostrum quality in dairy cattle, a large amount of data is needed, and the use of indicator traits would be beneficial. In the study presented here, we explored the heritabilities of reference (radial immunodiffusion) and near infrared-predicted IgG, IgA, and IgM concentrations and estimated their genetic correlations. First, the colostrum of 765 Holstein cows from nine herds was sampled to perform a reference analysis and the near-infrared spectra (400-2500 nm) were stored. We used a calibration set (28% of the initial samples) that was representative of the herds and cow parity orders to develop prediction equations for IgG, IgA, and IgM concentrations. Finally, these traits were predicted in the validation set (72% of the initial samples) to estimate genetic parameters for the predictions. Genetic correlations between reference and predicted values of each trait were estimated through bivariate linear animal models., Results: The three near-infrared-predicted immunoglobulin fractions were genetically correlated with their reference value. In particular, the reference and predicted IgG concentrations were strongly correlated at both the genetic (0.854 ± 0.314) and phenotypic level (0.767 ± 0.019). Weaker associations were observed for IgA and IgM concentrations, which were predicted with lower accuracy compared to IgG. Simulation analyses suggested that improving colostrum quality by selective breeding in Holstein cattle based on near-infrared predicted colostrum immunoglobulins concentrations is feasible. In addition, less than 10 mL of colostrum are needed for spectra acquisition and thus implementation of such analyses is possible in the near future., Conclusions: The concentrations of colostrum immunoglobulins can be predicted from near-infrared spectra and the genetic correlation between the reference and the predicted traits is positive and favourable, in spite of the large standard errors of the estimates. Extensive summer grazing is a dairy herd management practice frequently adopted in mountainous areas. Nowadays, this activity is threatened by its high labour demand, but it is fundamental for environmental, touristic and economic implications, as well as for the preservation of social and cultural traditions. Scarce information on the effects of such low-input farming systems on cattle health is available. Therefore, the present case study aimed at investigating how grazing may affect the health status of dairy cows by using milk traits routinely available from the national milk recording scheme. The research involved a dairy herd of 52 Simmental and 19 Holstein × Simmental crossbred cows. The herd had access to the pasture according to a rotational grazing scheme from late spring up to the end of summer. A total of 616 test day records collected immediately before and during the grazing season were used. Individual milk yield was registered during the milking procedure. Milk samples were analysed for composition (fat, protein, casein and lactose contents) and health-related milk indicators (electrical conductivity, urea and β-hydroxybutyrate) using mid-infrared spectroscopy. Somatic cell count (SCC) and differential SCC were also determined. Data were analysed with a linear mixed model, which included the fixed effects of the period of sampling, cow breed, stage of lactation and parity, and the random effects of cow nested within breed and the residual. The transition from barn farming to pasture had a negative effect on milk yield, together with a small deterioration of fat and protein percentages. Health-related milk indicators showed a minor deterioration of the fat to protein ratio, differential SCC and electrical conductivity, particularly towards the end of the grazing season, whereas the somatic cell score and β-hydroxybutyrate were relatively constant. Overall, the study showed that, when properly managed, pasture grazing does not have detrimental effects on dairy cows in terms of udder health and efficiency. Therefore, the proper management of cows on pasture can be a valuable solution to preserve the economic, social and environmental sustainability of small dairy farms in the alpine regions, without impairing cows' health.

Pharmacists in the community and the essential requirement to safeguard their own health have become fundamental since the spread of the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). The aims of this paper were (I) to analyze the directives provided to pharmacists in 2020 regarding preventative safety measures to be adopted; (II) to determine the number of pharmacists who came into contact with SARS-CoV-2 in North-West Italy and relate this to the adopted preventative measures. The first aim was pursued by conducting a bibliographic research, consulting the principal regulatory sources. The second one was achieved with an observational study by administering a questionnaire and performing a serological test. The various protection measures imposed by national and regional legislation were analyzed. Two hundred and eighty-six pharmacists (about 8% of the invited ones) responded to the survey. Ten pharmacists reported a positive result to the serological test. Of the subjects who presented a positive result, three declared that they had not used a hand sanitizer, while two stated that they had not scheduled the cleaning and decontamination of surfaces. Two interviewees had not set up a system of quota restrictions on admissions. In four cases, a certified cleaning company had decontaminated the premises. The results of our study show that during the coronavirus disease 2019 (COVID-19) pandemic, the most pressing challenge for community pharmacists has been the protection of staff and clients inside the pharmacy; the challenge to be faced in the near future will probably be the management of new responsibilities.

Food antioxidants enhance products shelf life and stability during technological treatments through the maintenance of their physical and chemical properties. Moreover, they are endowed with several positive effects on human health, including cell membranes preservation, enzyme functionality, and DNA integrity. Milk has been described in relation to a wide array of fat soluble and water-soluble antioxidant compounds, in particular vitamin A, C, and E, lactoferrin and peptides derived from casein and whey proteins. The total antioxidant activity (TAA) of milk is a novel and scarcely explored trait, defined as the sum of antioxidant contributions of the aforementioned compounds. On this background, the aims of the present study were to investigate the variability of milk TAA on a large scale exploiting predictions obtained through mid-infrared (MIR) spectroscopy and to estimate genetic parameters of this trait in Holstein cows. Individual milk samples were collected between January 2011 and December 2018 during the routine milk recording procedure. Samples were analysed for gross composition through MIR spectroscopy and MIR spectra were stored. Milk TAA was then predicted (pTAA) from the stored milk MIR spectra (111,653 test-day records of 9519 cows in 344 herds) using the previously developed prediction model; considering the prediction accuracy, pTAA might be considered a proxy of the TAA determined through the reference method. Overall, pTAA averaged 7.16 mmoL/L of Trolox equivalents, showed a nadir around 40 days after calving and increased thereafter, following a linear trend up to the end of lactation. The lowest pTAA was observed in milk sampled from June to September. Milk pTAA was heritable (0.401 ± 0.015) and genetically associated to fat yield (0.366 ± 0.049), crude protein (CP) yield (0.238 ± 0.052), fat percentage (0.616 ± 0.022) and CP percentage (0.754 ± 0.015). The official selection index of Italian Holstein put the 49% of the emphasis on fat and protein yield and percentage; therefore, it derives that an indirect favourable selection for milk pTAA should be already in progress in Italian Holstein population.

Water buffaloes ( Bubalus bubalis ) are very sensitive to environmental stimulus before and during milking, and this explains why disrupted milk ejections due to blood oxytocin level instability are frequent in this species. According to the literature, the manual stimulation (MS) of teats before milking promotes oxytocin release and allows milk ejection to start within 2-3 min. However, the pre-milking stimulation of teats is not always part of the milking routine in Italian buffalo farms; moreover, buffaloes with unstable milk let-down are sometimes treated with exogenous oxytocin (OX). Different types of pre-milking stimulation can impact differently on the mammary gland epithelium and structures and, therefore, on milk yield. In this study, we observed the changes in teat morphological traits before and after the application of three types of stimuli, i.e., no manual stimulation (NS), MS, and OX, in buffaloes reared in an Italian dairy farm. In particular, measurements were available for 23 and 21 buffaloes for front and rear teats, respectively. Subsequently, the effect of the pre-milking stimulation type was estimated on teat morphological characteristics and on milk traits recorded after the application of stimuli. The results showed that the teat canal length was shorter ( P < 0.05) after stimulation in the case of MS and OX compared to NS. Cistern diameter was overall greater for MS and lower for OX. On the contrary, teat wall thickness was greater and lower for OX and MS, respectively. Milk yield and quality (fat, protein, and somatic cell score) were similar across the three types of pre-milking stimulation. The aim of the present study was to investigate the ability of a handheld near-infrared spectrometer to predict total and gelatinized starch, insoluble fibrous fractions, and mineral content in extruded dry dog food. Intact and ground samples were compared to determine if the homogenization could improve the prediction performance of the instrument. Reference analyses were performed on 81 samples for starch and 99 for neutral detergent fiber (NDF), acid detergent fiber (ADF), acid detergent lignin (ADL), and minerals, and reflectance infrared spectra (740 to 1070 nm) were recorded with a SCiO™ near-infrared (NIR) spectrometer. Prediction models were developed using modified partial least squares regression and both internal (leave-one-out cross-validation) and external validation. The best prediction models in cross-validation using ground samples were obtained for gelatinized starch (residual predictive deviation, RPD = 2.54) and total starch (RPD = 2.33), and S (RPD = 1.92), while the best using intact samples were obtained for gelatinized starch (RPD = 2.45), total starch (RPD = 2.08), and K (RPD = 1.98). Through external validation, the best statistics were obtained for gelatinized starch, with an RPD of 2.55 and 2.03 in ground and intact samples, respectively. Overall, there was no difference in prediction models accuracy using ground or intact samples. In conclusion, the miniaturized NIR instrument offers the potential for screening purposes only for total and gelatinized starch, S, and K, whereas the results do not support its applicability for the other traits.

The aim of this study was to estimate genetic associations between alternative somatic cell count (SCC) traits and milk yield, composition and udder type traits in Italian Jersey cows. Alternative SCC traits were test-day (TD) somatic cell score (SCS) averaged over early lactation (SCS&#95;150), standard deviation of SCS of the entire lactation (SCS&#95;SD), a binary trait indicating absence or presence of at least one TD SCC >400,000 cells/ml in the lactation (Infection) and the ratio of the number of TD SCC >400,000 cells/ml to total number of TD in the lactation (Severity). Heritabilities of SCC traits, including lactation-mean SCS (SCS&#95;LM), ranged from 0.038 to 0.136. Genetic correlations between SCC traits were moderate to strong, with very few exceptions. Unfavourable genetic associations between milk yield and SCS&#95;SD and Infection indicated that high-producing cows were more susceptible to variation in SCC than low-producing animals. Cows with deep udders, loose attachments, weak ligaments and long teats were more susceptible to an increase of SCC in milk. This study aimed to investigate factors affecting protein fractions, namely α-casein (α-CN), β-casein (β-CN), κ-casein (κ-CN), β-lactoglobulin (β-LG) and α-lactalbumin (α-LA) predicted from milk infrared spectra in milk of dairy and dual-purpose cattle breeds. The dataset comprised 735,328 observations from 49,049 cows in 1782 herds. Results highlighted significant differences of protein fractions in milk of the studied breeds. Significant variations of protein fractions were found also through parities and lactation, with the latter thoroughly influencing protein fractions percentage. Interesting correlations ( r ) were estimated between β-CN, κ-CN and β-LG, expressed as percentage of crude protein, and milk urea nitrogen ( r = 0.31, -0.20 and -0.26, respectively) and between α-LA and fat percentage ( r = 0.41). The present study paves the way for future studies on the associations between protein fractions and milk technological properties, and for the estimation of genetic parameters of predicted protein composition.

This study estimated the effect of Holstein-Friesian, Brown Swiss, Jersey, Simmental and Alpine Grey cattle breeds on milk mineral contents (Ca, Mg, P, K, and Na) in multibreed herds using data predicted with mid-infrared spectroscopy. The dataset included 139,821 observations from 16,566 cows and 977 herds. Fixed effects considered in the mixed model were breed, parity, stage of lactation and first-order interactions, and random effects were cow, herd-test-date, and the residual. Multiple comparisons of least squares means were performed for the main effect of breed, parity, and stage of lactation using Bonferroni adjustment. Holstein-Friesian yielded milk with the lowest fat, protein, and casein concentration, and Ca, Mg, and P contents, whereas Jersey cows produced milk with the greatest fat, protein, and casein concentration, and Ca and Mg contents. Results of this study suggest that mixing milk from different breeds could enhance milk composition and technological ability, and therefore contribute to improve dairy industry efficiency., (© 2018 Japanese Society of Animal Science.)
