Luminescent bacteria-based biosensors are widely used for fast and sensitive monitoring of food safety, water quality, and other environmental pollutions. Recent advancements in biomedical engineering technology have led to improved portability, integration, and intelligence of these biotoxicity assays. Moreover, genetic engineering has played a significant role in the development of recombinant luminescent bacterial biosensors, enhancing both detection accuracy and sensitivity. This review provides an overview of recent advances in the development and applications of novel luminescent bacteria-based biosensors, and future perspectives and challenges in the cutting-edge research, market translation, and practical applications of luminescent bacterial biosensing are discussed., (© 2024 John Wiley & Sons Ltd.)

At the beginning of the paper, the level of necessary phenomenology of complex models is discussed. When working with complex systems, which of course include living organisms and ecological systems, it is necessary to use a phenomenological description. An illustration of the phenomenological approach is given, which captures the most significant general principles or patterns of interactions; the specific values of the parameters cannot be calculated from the first principles, but are determined empirically. An appropriate interpretation is also chosen empirically and pragmatically. However, in order to simulate a wider range of situations, it becomes necessary to lower the level of phenomenology, switch to a more detailed description of the system, introducing interaction between selected elements of the system. The requirements for a system model combining ecological, metabolic and genetic levels of cell culture description are formulated. A mathematical model of quorum sensing dynamics during the growth of batch culture of luminescent bacteria at different concentrations of the nutrient substrate has been developed. The model contains four blocks describing ecological, energy, quorum and luminescent aspects of bacterial culture growth. The model demonstrated good agreement with the experimental data obtained. When analyzing the model, three oddities in the behavior of the culture were noted, which presumably can change the idea of some processes taking place during the development of a culture of luminescent bacteria. The results obtained suggest the presence of some additional control system for the luminescent reaction via the synthesis pathways of FMN · Н2 or aliphatic aldehyde. In this case, the generalized description of the contribution of energy metabolism to luminescence only through ATP is too strong a simplification. As a result of comparing the model dynamics with the experiment, a discrepancy arose between the concentration of the substrate (peptone) measured in the experiment and its effective influence on the bacterial population growth. This discrepancy seems to indicate peptone is not the leading substrate, and growth is limited by nutrients contained in the yeast extract, the concentration of which did not change in these experiments. The light emitted by a luminescent bacterium serves as a unique native channel of information regarding the intracellular processes within the individual cell. In the presence of highly sensitive equipment, it is possible to obtain the distribution of bacterial culture cells by the intensity of light emission, which correlates with the amount of luciferase in the cells. When growing on rich media, the luminescence intensity of individual cells of brightly luminous strains of the luminescent bacteria Photobacterium leiognathi and Ph. phosporeum reaches 104-105 quanta/s. The signal of such intensity can be registered using sensitive photometric equipment. All experiments were carried out with bacterial clones (genetically homogeneous populations). A typical dynamics of luminous bacterial cells distributions with respect to intensity of light emission at various stages of batch culture growth in a liquid medium was obtained. To describe experimental distributions, a phenomenological model that links the light of a bacterial cell with the history of events at the molecular level was constructed. The proposed phenomenological model with a minimum number of fitting parameters (1.5) provides a satisfactory description of the complex process of formation of cell distributions by luminescence intensity at different stages of bacterial culture growth. This may be an indication that the structure of the model describes some essential processes of the real system. In this paper, the releasing characteristics and biological toxicity of Tl, Hg, As and Sb in waste of Lanmuchang mercury-thallium mine were studied. The results indicated that strong acidity can significantly promote the release of Tl from waste. With the increase of pH, the release of Sb grew steadily, while Hg and As showed a trend of first increasing and then decreasing. Fe 2 (SO 4 ) 3 contributed less to the release of As and Sb than to that of Hg and Tl. FeCl 3 significantly inhibited the release of As, Sb and Tl. In the leaching experiments of litter and root exudates, the lixiviums appeared neutral, and the litter and root exudates solution significantly reduced the release of Tl, and showed less toxicity to luminescent bacteria. Predicting the crucial effect of single metal pollutants against the aquatic ecosystem has been highly debatable for decades. However, dealing with complex metal mixtures management in toxicological studies creates a challenge, as heavy metals may evoke greater toxicity on interactions with other constituents rather than individually low acting concentrations. Moreover, the toxicity mechanisms are different between short term and long term exposure of the metal toxicant. In this study, acute and chronic toxicity based on luminescence inhibition assay using newly isolated Photobacterium sp.NAA-MIE as the indicator are presented. Photobacterium sp.NAA-MIE was exposed to the mixture at a predetermined ratio of 1:1. TU (Toxicity Unit) and MTI (Mixture Toxic Index) approach presented the mixture toxicity of Hg 2+ + Ag + , Hg 2+ + Cu 2+ , Ag + + Cu 2+ , Hg 2+ + Ag + + Cu 2+ , and Cd 2+ + Cu 2+ showed antagonistic effect over acute and chronic test. Binary mixture of Cu 2+ + Zn 2+ was observed to show additive effect at acute test and antagonistic effect at chronic test while mixture of Ni 2+ + Zn 2+ showing antagonistic effect during acute test and synergistic effect during chronic test. Thus, the strain is suitable and their use as bioassay to predict the risk assessment of heavy metal under acute toxicity without abandoning the advantage of chronic toxicity extrapolation.

Although luminescent bacteria-based bioluminescence inhibition assay has been widely used in the toxicity assessment of environmental pollutants, the response of a luminescent bacterium usually lacks specificity to a target analyte. Recently, some specific analyte inductive promoters were fused to the lux genes for the purpose of selective bioluminescent sensing, and suits of specific promoters were fused to lux genes to compose a bioluminescent array sensor for simultaneous identification of multiple analytes. However, specific promoter-based methods still suffer from drawbacks including limited selectivity, slow responding time, expensive to construct different promoters involved plasmids, and laborious to find new promoters. Herein, we proposed a novel strategy to construct a lux reporter array sensor by directly transforming the natural lux genes in different bacterial hosts without the involvement of any specific promoters. Due to the distinct pathways of signal production, the responding time of the current different bacterial host (DBH)-based lux reporter array has nearly an order of magnitude faster than with specific promoter-based methods. The DBH-based lux reporter array was successfully used for simultaneous identification, quantification, and toxicity/bioactivity assessment of multiple metal ions. Obviously, all the chemical synthetic material-based metal ion sensing methods cannot simultaneously achieve analysis and toxicity evaluation. This approach possessed additional advantages of facile construction, easy operation, high selectivity, fast response, and strong adaptability to other analytes. A different bacterial host-based lux reporter array was established for simultaneous analysis and toxicity assessment of multiple metal ions.

Developments in industrial applications inevitably accelerate the discharge of enormous substances into the environment, whereas multi-component mixtures commonly cause joint toxicity which is distinct from the simple sum of independent effect. Thus, ecotoxicological assessment, by luminescent bioassays has recently brought increasing attention to overcome the environmental risks. Based on the above viewpoint, this review included a brief introduction to the occurrence and characteristics of toxic bioassay based on the luminescent bacteria. In order to assess the environmental risk of mixtures, a series of models for the prediction of the joint effect of multi-component mixtures have been summarized and discussed in-depth. Among them, Quantitative Structure-Activity Relationship (QSAR) method which was widely applied in silico has been described in detail. Furthermore, the reported potential mechanisms of joint toxicity on the luminescent bacteria were also overviewed, including the Trojan-horse type mechanism, funnel hypothesis, and fishing hypothesis. It has been proven that the raw wastewater, secondary effluent and even reclaimed water may have toxic effects on aquatic organisms. In the present study, fractionation procedures combined with bioassays using luminescent bacteria were conducted to identify the fractions that contributed to the acute and long-term toxicity of municipal wastewater. Solid phase extraction was used to divide dissolved organic matter from the wastewater into three fractions, including non-polar, medium-polar and polar fraction. Among these fractions, although the acute toxicity of municipal wastewater was mainly caused by polar and medium-polar chemicals, the acute toxicity induced by the unit mass of the medium-polar fraction was the greatest. Using three kinds of resins, the organic substances in municipal wastewater were classified into six fractions, and the long-term toxicity of these fractions was further identified. The long-term toxicity of the hydrophobic neutrals, which were the primary toxic substances in raw wastewater, decreased after the conventional secondary biological treatment. Hydrophilic neutrals, which accounted for the majority of organic substances in the secondary effluent, were the main substances with long-term toxicity in the secondary effluent. The identification of fractions with acute and long-term toxicity in municipal wastewater is beneficial for further treatment to attenuate the ecotoxicity of wastewater before discharge into the aquatic environment.

Assessment of eco-toxicant using bioluminescent bacterial assay is a widely used and globally accepted method. In this work, a new luminescent bacterium was isolated from squid (Loligo duvauceli) and identified as Photobacterium leiognathi strain AK-MIE using 16S rRNA, phylogeny analysis. The predicted optimum conditions by RSM were 2.76% (w/v) NaCl, 2.28% (w/v) peptone, 0.34% (w/v) yeast extract, and pH 6.83 with 541,211.80 RLU of luminescent production whereas the predicted optimum conditions by ANN were 2.21% (w/v) NaCl, 2.27% (w/v) peptone, 0.39% (w/v) yeast extract, and pH 6.94 which produced 541,986.20 RLU. The validation analysis of both RSM and ANN show 0.60% and 0.69% deviation from the predicted results indicating that both models provided good quality predictions with ANN showing a superior data fitting capability for non-linear regression analysis. Toxicity tests show strain AK-MIE was sensitive to mercury (concentration causing 50% inhibition or IC 50 of 0.00978 mgL -1 ), followed by cadmium (IC 50 of 0.5288 mgL -1 ), copper IC 50 of (0.8117 mgL -1 ), silver (IC 50 of 1.109 mgL -1 ), and lead (IC 50 of 10.71 mgL -1 ) which are more sensitive than previously isolated luminescent bacteria, suggesting that strain AK-MIE has the potential to be used in toxicity assessment of heavy metals in the environment. Based on the field trial results, several sediment samples from industrial areas in Bangi, Selangor managed to inhibit the bioluminescence of strain AK-MIE. In the bioethanol industry, per liter of the produced alcohol 9 to 14 liters of vinasse are obtained as a byproduct. If the vinasse is directly shed into bodies of water without an adequate treatment, it may have negative effects on the existing biota and human health due to its high turbidity and color, low pH and high content of organic material. The purpose of this study was to assess the acute toxicity of vinasse by means of a rapid test with Aliivibrio fischeri and compare it with a standard immobilization assay with Daphnia magna . The standard assay of D. magna by means of its EC 50 of 4.7% showed that organism was more sensitive to the contaminant, in comparison with the 69.6% obtained with the A. fischeri which suggests that it should be continuesly used as one of the organisms of first choice for the evaluation of the acute toxicity of this effluent., (Copyright © 2019 SETOX & Institute of Experimental Pharmacology and Toxicology, SASc.)

The phenolic compounds have posed public concern for potential threats to human health and ecosystem. Tert-butylphenols (TBPs), as one group of emerging contaminants, showed potential endocrine disrupting effects and aquatic toxicities. In the present study, we detected concentrations of 2,4-DTBP ranging from <0.001 to 0.057 μg/L (detection limit: 0.001 μg/L) in drinking water source from the Qiantang River in East China in April 2016. The endocrine disrupting effects of 2-TBP, 2,4-DTBP and 2,6-DTBP toward human estrogen receptor α (ERα), androgen receptor (AR) and thyroid hormone receptor β (TRβ) were evaluated using human recombinant two-hybrid yeast bioassay. Their aquatic toxicities were investigated with indicator organisms including Photobacterium phosphoreum, Vibrio fischeri and freshwater green alga Chlamydomonas reinhardtii. 2-TBP and 2,4-DTBP exhibited moderate antagonistic effects toward human ERα and AR in a concentration-dependent manner. 2-TBP significantly inhibited the light emission of P. phosphoreum. 2-TBP, 2,4-DTBP and 2,6-DTBP significantly inhibited the growth of C. reinhardtii and reduced the chlorophyll content. Our results suggest the potential adverse effects of TBPs on human health and aquatic organisms. A novel tool, the DAMTA analyzer (Device for Analytical Monitoring and Toxicity Assessment), designed for fully automated toxicity measurements based on luminescent bacteria as well as for concomitant determination of chemical parameters, was developed and field-tested. The instrument is a robotic water analyzer equipped with a luminometer and a spectrophotometer, integrated on a thermostated reaction plate which contains a movable carousel with 80 cuvettes. Acute toxicity is measured on-line using a wild type Photobacterium phosphoreum strain with measurable bioluminescence and unaltered sensitivity to toxicants lasting up to ten days. The EC50 values of reference compounds tested were consistent with A. fischeri and P. phosphoreum international standards and comparable to previously published data. Concurrently, a laboratory trial demonstrated the feasibility of use of the analyzer for the determination of nutrients and metals in parallel to the toxicity measurements. In a prolonged test, the system was installed only in toxicity mode at the premises of the World Fair "Expo Milano-2015″, a high security site to ensure the quality of the supplied drinking water. The monitoring program lasted for six months during which ca. 2400 toxicity tests were carried out; the results indicated a mean non-toxic outcome of -5.5 ± 6.2%. In order to warrant the system's robustness in detecting toxic substances, Zn was measured daily with highly reproducible inhibition results, 70.8 ± 13.6%. These results assure that this novel toxicity monitor can be used as an early warning system for protection of drinking water sources from emergencies involving low probability/high impact contamination events in source water or treated water., (Copyright © 2018 Elsevier Inc. Ultraviolet (UV) photodegradation is increasingly applied to control volatile organic compounds (VOCs) due to its degradation capabilities for recalcitrant compounds. However, sometimes the UV photodegradation products are also toxic and can affect human health. Here, 10 VOCs at 150~200 ppm in air were treated using a laboratory-scale UV reactor with 185/254 nm irradiation, and the biotoxicity of their off-gas was studied by investigating their off-gas absorption solutions. The CO₂ increase and VOC decrease were 39~128 ppm and 0~42 ppm, respectively, indicating that the VOCs and their products were mineralized in off-gas absorption solutions. The total organic carbon (TOC) of the absorption solutions are 4~20 mg∙L -1 . Luminescent bacteria and Daphnia magna were used to detect the acute toxicity, and an umu assay was used to determine the genotoxic potential. Trichloroethylene showed a highest toxicity to luminescent bacteria, while chlorobenzene had the lowest toxicity. Water-soluble UV photodegradation products for styrene are very toxic to Daphnia magna . In the umu assay, the genotoxicities of off-gas absorption solutions of trichloroethylene, methylbenzene, ethyl acetate, butyl alcohol, and styrene were 51.26, 77.80, 86.89, 97.20, and 273.62 mg (4-NQO)·L -1 respectively. In addition, the analysis of the genotoxicity/TOC and intermediates products indicated that the off-gas absorption solutions of styrene, trichloroethylene, and butyl alcohol contain many highly toxic substances.

In order to accurately evaluate the comprehensive ecotoxicity of domestic wastewater and the toxicity reduction efficiency of different treatment processes, standardized assays focusing on Vibrio fischeri , SOS/umu test and yeast estrogen screen were applied. Simultaneously, male zebrafish was used to address the mode of action of endocrine disrupting effects of wastewater and reclaim water on aquatic life. Results of this study indicated that the influent was severely polluted:highly acute toxicity, genotoxicity and estrogenicity were observed. After the secondary biological treatment, the above toxicities were significantly reduced, making the water quality improved. However, the estrogencity of secondary effluent was detected at 1.89 ng·L -1 , which still posed potential risk on the aquatic life in receiving water. In addition, the up-regulation of estrogen receptor ( esr 1 ) and vitellogenin ( vtg 1 ) in the livers of zebrafish was observed after exposure to 2.5 times concentrated wastewaters, which demonstrated that wastewater modulates hormonal activities of aquatic life by disturbing target genes expression. Moreover, the slight down-regulation of esr 1 was observed in the gonads. These observations reflect that the evaluation of biological toxic effect should be analyzed in different tissues or organs, so that more comprehensive information about the adverse effects of wastewaters on aquatic life can be obtained.

Drinking water treatment residue (DWTR) seems to be very promising for controlling lake sediment pollution. Logically, acquisition of the potential toxicity of DWTR will be beneficial for its applications. In this study, the toxicity of DWTR and sediments amended with DWTR to Aliivibrio fischeri was evaluated based on the Microtox(®) solid and leachate phase assays, in combination with flow cytometry analyses and the kinetic luminescent bacteria test. The results showed that both solid particles and aqueous/organic extracts of DWTR exhibited no toxicity to the bacterial luminescence and growth. The solid particles of DWTR even promoted bacterial luminescence, possibly because DWTR particles could act as a microbial carrier and provide nutrients for bacteria growth. Bacterial toxicity (either luminescence or growth) was observed from the solid phase and aqueous/organic extracts of sediments with or without DWTR addition. Further analysis showed that the solid phase toxicity was determined to be related mainly to the fixation of bacteria to fine particles and/or organic matter, and all of the observed inhibition resulting from aqueous/organic extracts was identified as non-significant. Moreover, DWTR addition not only had no adverse effect on the aqueous/organic extract toxicity of the sediment but also reduced the solid phase toxicity of the sediment. Overall, in practical application, the solid particles, the water-soluble substances transferred to surface water or the organic substances in DWTR had no toxicity or any delayed effect on bacteria in lakes, and DWTR can therefore be considered as a non-hazardous material., (Copyright © 2016 Elsevier Ltd. Background/purpose: To investigate the effect of nanosilver particles in solution stabilized in a matrix of sodium alginate on the growth and development of pathogenic bacteria such as Staphylococcus aureus , Enterococcus faecalis , Escherichia coli , Proteus vulgaris , Enterobacter cloacae , the antibiotic-resistant strain of Pseudomonas aeruginosa , the yeast-like fungus Candida albicans , and the luminescent bacteria Photobacterium leiognathi Sh1., Methods: Isolates of pathogenic bacteria obtained from bronchoalveolar and peritoneal lavage samples from Wistar rats with experimental pneumonia and peritonitis were tested for their susceptibility to silver nanoparticles in solution with an alginate stabilizer. The antifungal activity of silver nanoparticles in sodium alginate was studied for C. albicans (strain CCM885) using the Sabouraud agar method. The biocidal impact of silver nanoparticles in solution with a sodium alginate matrix on the luminescent bacteria P. leiognathi Sh1 was investigated using a BLM 8801 luminometer., Results: It was observed that a 0.02-0.05% nanosilver solution with an alginate stabilizer limits the growth and development of pathogenic bacteria within the first 24 hours of exposure. If the concentration of nanosilver solution is 0.0005-0.05%, it inhibits the viability of the fungus C. albicans . A nanosilver solution at a concentration of 0.05-0.2 μg/mL represses bioluminescence in the bacteria P. leiognathi Sh1. From these results, it appears that the biocidal effect of nanosilver is related either to the presence of ions that are formed during dissolution, or to the availability of nanoparticles that interrupt the membrane permeability of bacterial cells., Conclusion: Silver nanoparticles stabilized in a solution of sodium alginate possess significant in vitro antimicrobial activity, which is manifested by inhibition of the bioluminescence of P. leiognathi Sh1, and inhibition of the growth and development of the pathogenic bacteria S. aureus , E. faecalis , E. coli , P. vulgaris, E. cloacae , the antibiotic-resistant strain of P. aeruginosa , and the fungus C. albicans .

Non-invasive in vivo imaging represents a powerful tool to monitor cellular and molecular processes in the living animal. In the special case of bacteria-mediated cancer therapy using bioluminescent bacteria, it opens up the possibility to follow the course of the microorganisms into the tumor via the circulation. The mechanism by which bacteria elicit their anti-tumor potential is not completely understood. However, this knowledge is crucial to improve bacteria as an anti-cancer tool that can be introduced into the clinic. For the study of these aspects, in vivo imaging can be considered a key technology.

We assessed the applicability of multi-strain bacterial bioreporter bioassays to drug screening. To this end, we investigated the reactions of a panel of 15 luminescent recombinant Escherichia coli bacterial bioreporters to a library of 420 pharmaceuticals. The panel included bacterial bioreporters associated with oxidative stress, DNA damage, heat shock, and efflux of excess metals. Eighty nine drugs elicited a response from at least one of the panel members and formed distinctive clusters, some of which contained closely related drugs. In addition, we tested a group of selected nine drugs against a collection of about 2000 different fluorescent transcriptional reporters that covers the great majority of gene promoters in E. coli. The sets of induced genes were in accord with the in vitro toxicity of the tested drugs, as reflected by the response patterns of the 15-member panel, and provided more insights into their toxicity mechanisms. Facilitated by microplates and robotic systems, all assays were conducted in high-throughput. Our results thus suggest that multi-strain assemblages of bacterial bioreporters have the potential for playing a significant role in drug development alongside current in vitro toxicity tests., (Copyright © 2015 The Authors.
