While human exposure to perfluorooctanoic acid (PFOA) can lead to ulcerative colitis, the molecular mechanisms responsible for PFOA-induced intestinal toxicity are unclear. Herein, we examined the toxicity of PFOA toward human colorectal cancer cells (HCT116) from three dimensions: the cytotoxic phenotype, cell respiration, and transcription levels of metabolism-related genes. Formazan was used to assess how PFOA exposure affects HCT116-cell relative viability, after which the mitochondrial respiratory activities of these cells were determined by analyzing extracellular flux. The quantitative real-time polymerase chain reaction (qPCR) method was used to detect metabolism-related gene expression levels. The cytotoxicity assay revealed that the HCT116 showed significantly inhibited relative activities compared to those of the control when exposed to 300 μmol/L PFOA for 48 h ( p <0.01), with most cells retained at the G0/G1 stage. In contrast, the mitochondrial respiratory activities of the HCT116 were promoted by concentrations of PFOA as low as 50 μmol/L. Two genes related to cellular metabolism (dipeptidase 1 ( DPEP1 ) and sphingosine kinase 1 ( SPHK1 )) were found to be related to the PFOA-promoted formation of ulcerative colitis using our self-developed Metabolic Gene and Pathway Query software and Comparative Toxicogenomics Database (CTD). The qPCR studies revealed that DPEP1 and SPHK1 expression levels were enhanced by 8-10 times in HCT116 exposed to 300 μmol/L PFOA relative to the control, whereas this trend was not observed for HCT116 exposed to 50 μmol/L PFOA. Collectively, these results suggest that the respiratory activity of cellular mitochondria may serve as an index for determining the interference effects associated with PFOA and that metabolic pathways mediated by DPEP1 and SPHK1 may be involved in the development of PFOA-induced ulcerative colitis. Future studies should investigate the relationships between changes in metabolism-related genes ( DPEP1 and SPHK1 ) and the mitochondrial respiratory activities of intestinal cells, and verify the roles played by the DPEP1 and SPHK1 genes in PFOA-induced intestinal inflammation using in-vivo models.

In the present study, two most commonly used Perfluoroalkyl substances (PFASs), namely perfluorooctanoic acid (PFOA) and perfluorooctanesulfonate (PFOS), were determined in 45 tap water samples from the city of Isfahan (Iran) by dispersive liquid-liquid extraction (DLLME) and liquid chromatography-mass spectrophotometry (LC-MS) analysis. Risk assessment was also performed to determine the risk to human health. The mean concentration of PFOA was 38.1 ± 26.4ng/L (min = 5.1 and max = 1056ng/L). The mean concentration of PFOS was 33.7 ± 25.09ng/L (min = 4.3 and max = 99.2ng/L). The combined concentrations of PFOA and PFOS were above the US-EPA advisory levels (70ng/L) in 48.8% of the samples. The distance between the sampling locations and the water treatment plant showed no significant correlation ( p  > 0.05). The results of the risk assessment showed that all calculated hazard quotients (HQ) and hazard indices (HI) are below 1, indicating that the risk to human health from exposure to PFOA and PFOS via drinking water in the city of Isfahan was not high for adults and children. These results indicate a significant contamination of Isfahan tap water by PFOA and PFOS of unknown origin. To date, only four studies directly measured dermal absorption kinetics of perfluorooctanoic acid (PFOA) in human skin. Reported kinetic parameters spanned two to five orders of magnitude, demonstrating the need to determine the causes of variability and identify the most appropriate dermal absorption factors for use in exposure assessments. We evaluated the reliability and physiological relevance of studies that measured PFOA fractional absorption, steady-state flux (J ss ), and dermal permeability coefficient (K p ). We verified whether the reported kinetic parameters were measured under appropriate conditions (i.e., fractional absorption under finite dose conditions, and J ss and K p under infinite dose conditions). We recommend the following values measured at the approximate pH of the skin, and in aqueous solvents or relevant consumer product matrices, for use as provisional values in PFOA exposure assessments: 1.6% fractional absorption under finite dose conditions, and 0.132 μg/cm 2 -hr and 0.000044 cm/h for J ss and K p , respectively, under infinite dose conditions. Using the recommended absorption factors, we estimated PFOA exposures in children from soil and water via dermal and ingestion routes. In November 2023, the International Agency for Research on Cancer (IARC) classified PFOA as "carcinogenic to humans" (Group 1) and PFOS as "possibly carcinogenic to humans" (Group 2B). We evaluated these classifications, considering the epidemiology, experimental animal, and mechanistic evidence. It is our opinion that the IARC Working Group overstated the available evidence for the carcinogenicity of PFOA and PFOS. Epidemiology studies have shown weak and inconsistent associations across studies. Studies reporting increased incidences of tumors in experimental animals exposed to PFOA or PFOS had statistically significant results that were driven by the presence of benign adenomas. The IARC Working Group used the key characteristics of carcinogens (KCCs, which comprise 10 chemical and/or biological properties of known human carcinogens) approach to upgrade the carcinogenicity classifications for PFOA and PFOS from initially lower classifications that were based on the strength of the epidemiology and experimental animal evidence. However, this is not a robust assessment of mechanistic evidence, as it fails to consider the quality, external validity, and relevance of the evidence. Perfluorinated alkyl substances (PFAS), such as perfluorooctanesulfonic acid (PFOS) and perfluorooctanoic acid (PFOA), are pervasive organic contaminants that are widespread in aquatic environments, posing significant health risks to humans and wildlife. Due to their persistent nature, urgent removal is necessary. Conventional adsorbents are inefficient at removing PFOS and PFOA, highlighting the need for alternative materials. Herein, we present a synthetic method for quaternary ammonium cation-doped carbon nanoparticles (QACNs) using a solution plasma process for the efficient removal of PFOS and PFOA. QACN is formed simultaneously through a one-step discharge of nonequilibrium plasma at the interface of benzene and pyridinium chloride. The resulting material exhibited a high surface electrical charge and enhanced hydrophilicity as well as an amorphous structure of a nonporous nature, involving nanoparticles with an undefined shape. The obtained adsorbent demonstrated high adsorption efficiency and stability, adsorbing 998.45 and 889.37 mg g -1 of PFOS and PFOA, respectively, exceeding the efficiencies of conventional carbon-based adsorbents (80.89-313.15 mg g -1 ). The adsorption performance was dependent on the adsorbent dosage, pH of the solution, and the coexisting ionic species. Adsorption studies, including adsorption kinetics, isotherms, and thermodynamics, revealed that PFOS and PFOA were chemisorbed to the QACN surface, forming multilayers endothermically and spontaneously. Experimental and computational analyses revealed that adsorption primarily occurs via electronic interactions between the PFAS active sites and the quaternary ammonium group in the carbon framework. The slightly lower adsorption potential of the PFOS and PFOA fluorocarbon chains on the adsorbent was elucidated. Furthermore, the dispersibility of the adsorbent in solution significantly affected the adsorption performance. These findings highlight the potential of the novel synthetic method proposed in this study, offering a pathway for the development of highly effective carbon adsorbents for environmental remediation.

The disruption of per- and polyfluoroalkyl substances (PFASs) on bile acid (BA) homeostasis has raised public concerns, making the evaluation of their effects and underlying mechanisms a high priority. Although the use of perfluorooctanoic acid (PFOA) has been restricted, it remains a widespread legacy PFAS in the environment. Concurrently, the use of its prevalent short-chain alternative, perfluorobutanoic acid (PFBA), is increasing, yet the toxicity assessment of PFBA remains inadequate. In this study, C57BL/6N mice were exposed to PFOA and PFBA (0.4 or 10 mg/kg body weight) by gavage for 28 days. The results showed that both PFOA and PFBA significantly increased hepatic weight, although PFBA exhibited lower bioaccumulation than PFOA in the liver. Targeted metabolomics revealed that PFOA significantly decreased total BA levels and altered their composition. Conversely, PFBA, without significantly altering total BA levels, notably changed their composition, such as increasing the proportion of cholic acid. Further investigations using in vivo and in vitro assays suggested that PFOA inhibited the expression of Cyp7A1, a key BA synthetase, potentially via PPARα activation, thereby reducing BA levels. In contrast, PFBA enhanced Cyp7A1 expression, associated with the inhibition of intestinal Farnesoid X receptor-fibroblast growth factor 15 (FXR-FGF15) pathway. In this work, a novel optical-chemical sensor for the detection of per- and polyfluorinated substances (PFASs) in a real scenario is presented. The proposed sensing approach exploits the multimode characteristics of plastic optical fibers (POFs) to achieve unconventional sensors via surface plasmon resonance (SPR) phenomena. The sensor is realized by the coupling of an SPR-POF platform with a novel chemical chip based on different polymeric nanolayers over the core of a D-shaped POF, one made up of an optical adhesive and one of a molecularly imprinted polymer (MIP) for PFAS. The chemical chip is used to launch the light into the SPR D-shaped POF platform, so the interaction between the analyte and the MIP's sites can be used to modulate the propagated light in the POFs and the SPR phenomena. Selectivity tests and dose-response curves by standard PFOA water solutions were carried out to characterize the detection range sensor response, obtaining a wide PFAS response range, from 1 ppt to 1000 ppt. Then, tests performed on river water samples collected from the Bormida river paved the way for the applicability of the proposed approach to a real scenario.

Pure Magnéli-phase Ti 4 O 7 were prepared by means of a Plasma Torch (PT) coating method and integrated into an advanced electro-catalytic oxidation (AEO) process in order to degrade perfluorooctanoic acid (PFOA) and perfluorooctanesulfonic acid (PFOS) persistent pollutants present in waters. The X-ray diffraction analysis confirmed the polycrystalline nature of the pure Magnéli phase PT-Ti 4 O 7 coatings (∼100 μm thick)). The Raman spectra of the PT-Ti 4 O 7 coatings also exhibited the two characteristic peaks (at 138 and 183 cm -1 ) of the PT-Ti 4 O 7 Magnéli phase. Scanning electron microscopy revealed the nanostructured hierarchical morphology of the PT-Ti 4 O 7 thus conferring them high surface area. The PT-Ti 4 O 7 anodes are shown to achieve higher degradation efficiencies towards PFOA and PFOS in comparison with the conventional boron-doped diamond anodes. By investigating several AEO parameters (including current density, treatment time, nature of the anode material), we were able to optimise the AEO process. Thus, for both PFOA and PFOS (at an initial concentration of 500 ppb in synthetic wastewaters), degradation efficiencies as high as 96.6% and 99.7% were achieved, respectively, with a current density of 20 mA/cm 2 , a treatment time of 120 min and PT-Ti 4 O 7 mesh-type anodes. PFOA and PFOS can be degraded by both direct anodic electrochemical oxidation ( • OH radicals) and indirect electrochemical oxidation via mediators, such as persulphate acid (H 2 S 2 O 8 ) generated by sulphate anodic oxidation. The degradation of both compounds followed pseudo-first-order kinetics. The reaction rate constant (k) for PFOS removal was 4.63 × 10 -2 min -1 , whereas 2.76 × 10 -2 min -1 was recorded for PFOA removal. Subsequently, we have used the above optimal AEO operating conditions to treat real wastewater effluents (containing 17 types of PFAS molecules with a total content of 8500 ppb) and achieved a degradation rate of 39.1%-87.4% for eight of the 17 PFAS compounds. Perfluoroalkyl and polyfluoroalkyl substances (PFASs) are persistent compounds characterized by stable C-F bonds giving them high thermal and chemical stability. Numerous studies have highlighted the presence of PFASs in the environment, surface waters and animals and humans. Exposure to these chemicals has been found to cause various health effects and has necessitated the need to develop methods to remove them from the environment. To date, the use of photocatalytic degradation and membrane separation to remove PFASs from water has been widely studied; however, these methods have drawbacks hindering them from being applied at full scale, including the recovery of the photocatalyst, uneven light distribution and membrane fouling. Therefore, to overcome some of these challenges, there has been research involving the coupling of photocatalysis and membrane separation to form photocatalytic membrane reactors which facilitate in the recovery of the photocatalyst, ensuring even light distribution and mitigating fouling. This review not only highlights recent advancements in the removal of PFASs using photocatalysis and membrane separation but also provides comprehensive information on the integration of photocatalysis and membrane separation to form photocatalytic membrane reactors. It emphasizes the performance of immobilized and slurry systems in PFAS removal while also addressing the associated challenges and offering recommendations for improvement. Factors influencing the performance of these methods will be comprehensively discussed, as well as the nanomaterials used for each technology. Additionally, knowledge gaps regarding the removal of PFASs using integrated photocatalytic membrane systems will be addressed, along with a comprehensive discussion on how these technologies can be applied in real-world applications.

Per- and polyfluoroalkyl substances (PFASs) have been widely detected in various food, which has attracted worldwide concern. However, the factors influencing the transfer and bio-accumulation of PFASs from soils to wheat in normal farmland, is still ambiguous. We investigated the PFASs accumulation in agricultural soils and grains from 10 cites, China, and evaluated the health risks of PFASs via wheat consumption. Our results show that ∑PFASs in soils range from 0.34 μg/kg to 1.59 μg/kg with PFOA and PFOS dominating, whilst ∑PFASs in wheats range from 2.74 to 6.01 μg/kg with PFOA, PFBA and PFHxS dominating. The lower pH conditions and high total organic carbon (TOC) could result in the higher accumulation of PFASs in soils and subsequently in wheat grains, whilst the bioaccumulation factors of PFASs increase with increasing pH conditions but not with TOC. The estimated daily intake (EDI) values of PFBA, PFOA, and PFHxS are relatively high, but data supports that ingesting wheat grains does not result in any potential risk to the human beings. Given environmental persistence, potential for bioaccumulation, and toxicity of Perfluorooctanoic acid (PFOA) and perfluorooctane sulfonate (PFOS), the scientific community has increasingly focused on researching their toxicology and degradation methods. This paper presents a survey of recent research advances in the toxicological effects and degradation methods of PFOA and PFOS. Their adverse effects on the liver, nervous system, male reproductive system, genetics, and development are detailed. Additionally, the degradation techniques of PFOA and PFOS, including photochemical, photocatalytic, and electrochemical methods, are analyzed and compared, highlighted the potential of these technologies for environmental remediation. The biotransformation pathways and mechanisms of PFOA and PFOS involving microorganisms, plants, and enzymes are also presented. As the primary green degradation pathway for PFOA and PFOS, Biodegradation uses specific microorganisms, plants or enzymes to remove PFOA and PFOS from the environment through redox reactions, enzyme catalysis and other pathways. Currently, there has been a paucity of research conducted on the biodegradation of PFOA and PFOS. However, this degradation technology is promising owing to its specificity, cost-effectiveness, and ease of implementation. Furthermore, novel materials/methods for PFOA and PFOS degradation are presented in this paper. These novel materials/methods effectively improve the degradation efficiency of PFOA and PFOS and provide new ideas and tools for the degradation of PFOA and PFOS. This information can assist researchers in identifying flaws and gaps in the field, which can facilitate the formulation of innovative research ideas., (© 2024. Cosmetics make up one of the consumer product categories most widely known to contain perfluoroalkyl and polyfluoroalkyl substances (PFASs), including precursors to perfluorooctanoic acid (PFOA) and other perfluoroalkyl acids (PFAAs). Because of the way cosmetics are used, most of the PFASs present in these products are likely to reach wastewater treatment plants (WWTPs), which suggests that cosmetics may contribute significantly to the load of PFOA and other PFASs at WWTPs. However, the majority of PFASs present as intentional ingredients in cosmetics cannot be quantified with the available analytical methods. To address this issue, we developed a methodology to estimate the total PFAS mass in cosmetics as well as the corresponding mass of total organic fluorine and of fluorinated side chains associated with PFAA precursors, using various ingredient databases and ingredient concentrations reported by manufacturers. Our results indicate that the cosmetics sold in California during a one-year period cumulatively contain 650-56 000 kg of total PFASs, 370-37 000 kg of organic fluorine, and 330-20 000 kg of fluorinated side chains associated with PFAA precursors. Among the 16 product subcategories considered, >90% of the PFAS mass came from shaving creams and gels, hair care products, facial cleansers, sun care products, and lotions and moisturizers, while the sum of all nine makeup subcategories accounted for <3%. Comparing our estimates to available WWTP influent data from the San Francisco Bay Area suggests that cosmetics may account for at least 4% of the precursor-derived PFAAs measured in wastewater. As the first study ever to estimate the total mass of PFASs contained in cosmetics sold in California, our results shed light on the significance of certain cosmetics as a source of PFASs to WWTPs and can inform effective source reduction efforts.

A novel, water-stable, perylene diimide (PDI) based metal-organic framework (MOF), namely, U-1 , has been synthesized for selective and sensitive detection of perfluorooctanoic acid (PFOA) in mixed aqueous solutions. The MOF shows highly selective fluorescence turn-on detection via the formation of a PFOA-MOF complex. This PFOA-MOF complex formation was confirmed by various spectroscopic techniques. The detection limit of the MOF for PFOA was found to be 1.68 μM in an aqueous suspension. Upon coating onto cellulose paper, the MOF demonstrated a significantly lower detection limit, down to 3.1 nM, which is mainly due to the concentrative effect of solid phase extraction (SPE). This detection limit is lower than the fluorescence sensors based on MOFs previously reported for PFAS detection. The MOF sensor is regenerable and capable of detecting PFOA in drinking and tap water samples. The PDI-MOF-based sensor reported herein represents a novel approach, relying on fluorescence turn-on response, that has not yet been thoroughly investigated for detecting per- and polyfluoroalkyl substances (PFAS) until now.

Background: In the past, perfluorooctanoic acid (PFOA) was produced and applied as an emulsifier in a fluoropolymer production plant in the Altötting district, southern Bavaria (Germany). This chemical was released directly into the environment, resulting in the contamination of the local drinking water. During a human biomonitoring (HBM) survey in 2018, increased median PFOA blood serum levels, compared to a normally exposed control group with no known source of PFOA exposure from Munich, Germany, were detected in the resident population (23.18 μg/l in the general population, 20.71 μg/l in the children's group). The follow-up study aimed to investigate whether purification of the drinking water as the main PFOA exposure source has been successful in reducing internal PFOA exposure and to estimate the association of internal PFOA exposure with possible influencing factors., Methods: Only individuals who had already participated in the HBM study in 2018 were included. For the determination of the PFOA serum concentration, 5 ml of blood was drawn from each participating person. Blood samples were collected in the period from June to August 2022. Furthermore, information on sociodemographic characteristics, health status, dietary behaviour and other lifestyle factors were collected by means of a self-administered questionnaire. To examine the association of PFOA blood serum levels with possible influencing factors, such as age, gender and consumption of fish and game meat, a logistic regression model with a PFOA value > 10 μg/l as outcome was used., Results: A total of 764 individuals participated in the follow-up study in 2022. Analyses were performed separately for the general population (n = 559), women of reproductive age (15-49 years old) (n = 120), and children under 12 years old (n = 30). Median PFOA blood levels have decreased by 56.9% in the general population, by 59.8% in the group of women of reproductive age and by 73.4% in the group of children under 12 years old. In the general population, a higher probability of a PFOA value > 10 μg/l was found for those aged 40-59 years (Odds ratio (OR) = 2.33 (95%CI: 1.23 to 4.43, p = 0.01) and those aged 60 years and older (OR = 5.32, 95%CI: 2.78 to 10.19, p < 0.001)., Conclusions: In all study groups, the median PFOA serum levels decreased as expected after a half-life of four years, which confirms that contamination via drinking water has ceased. Furthermore, our study identified age as a significant predictor of internal PFOA exposure, while no influence was found for the consumption of fish and game meat. Perfluorooctanoic acid (PFOA) is a persistent contaminant with detrimental effects on the natural environment. This persistence leads to potential enrichment and osmotic transfer, which can affect normal circulation in the environment. PFOA poses significant threats to both the natural environment and human health. Therefore, the development of cost-effective, highly efficient, and environment-friendly PFOA adsorbents is a crucial endeavor. This paper presents the catalyst-free one-pot synthesis of fluorinated nitrogen-rich porous organic polymers (POP-3F) via a Schiff-base condensation reaction. The reaction between the nitrogen-rich compound 1,4-bis(2,4-diamino-1,3,5-triazine)benzene and p -trifluoromethylbenzaldehyde yielded POP-3F. The introduction of fluorine atoms into the nitrogen-rich porous organic polymer enhanced its hydrophobicity, thereby facilitating favorable fluoro-fluorine interactions with PFOA and, thus, improving the efficacy of the adsorbent. Scanning electron microscopy (SEM), Fourier transform infrared (FT-IR) spectroscopy, X-ray diffraction (XRD), solid-state nuclear magnetic resonance (ssNMR) spectroscopy, X-ray photoelectron spectroscopy (XPS), nitrogen adsorption-desorption analysis, and thermogravimetric analysis (TGA) were used to confirm the successful synthesis and characterization of POP-3F. Liquid chromatography-tandem mass spectrometry (LC-MS/MS) was conducted in negative electrospray ionization (ESI) mode coupled with multi-reaction monitoring mode (MRM). The instrument was equipped with an Atlantis T3 column (100 mm×2.1 mm, 3 μm), and analysis was conducted using an external standard method. The influences of various factors on PFOA adsorption by POP-3F, including pH, salt concentration, and humic acid presence, were investigated. The highest PFOA removal rate (98.6%) was achieved at a pH of 2, indicating the applicability of POP-3F for the effective removal of PFOA from acidic industrial wastewater. The removal rate of PFOA was unaffected by increases in NaCl concentration. This phenomenon can be attributed to electrostatic interactions between the protonated secondary amines in POP-3F and deprotonated PFOA. Upon the addition of NaCl, a double electric layer is formed on the POP-3F surface, with Cl - ions in the outer layer and Na + ions in the inner layer, which weakened these interactions. Humic acid is competitively adsorbed with PFOA. However, POP-3F demonstrated good removal rates even in the presence of high humic acid concentrations in water. Adsorption isotherm and kinetics experiments were conducted at the optimal pH to explore the relevant adsorption mechanism. The results showed a rapid initial adsorption rate, with 95.4% PFOA removal within 5 min. Optimal adsorption equilibrium was achieved within 6 h, and the removal rate decreased by only 0.3% after 24 h. This finding indicates that POP-3F exhibits sustained efficacy for PFOA removal. Langmuir fitting analysis revealed a theoretical maximum adsorption capacity of 191 mg/g for POP-3F; this value surpasses those of activated carbon materials and most other adsorbents, highlighting the superior PFOA-adsorption performance of POP-3F. Additionally, matrix effects minimally affected the removal of PFOA by POP-3F, with only a slight reduction (0.1%) observed in simulated natural water. The recyclability of POP-3F was assessed over five adsorption-desorption cycles. The removal efficenecy exhibited a minor decrease of only 0.67% after five cycles. These results demonstrate the recyclability of the proposed adsorbent, which translates into cost reduction through reusability. This characteristic renders POP-3F a promising candidate for the economical and efficient removal of PFOA from wastewater in practical applications.

Objective: To establish a method for the determination of two perfluorinated compounds in urine by liquid chromatography-tandem mass spectrometry. Methods: In November 2022, urine samples were extracted by acidic methanol, purified by WAX solid phase extraction column, and eluted with methanol water, then Waters ACQUITY UPLC BEH C18 column (100 mm×2.1 mm, 1.7 μm) was used with 1.0 mmol/L ammonium acetate solution and methanol as mobile phase. The gradient elution was carried out, the detection was carried out by electrospray negative ion multiple response monitoring (MRM) mode, and the quantitative method was internal standard method. Results: Perfluorooctanoic acid and perfluorooctane sulfonic acid had a good linear relationship in the concentration range of 0.5-50.0 μg/L, and the correlation coefficient was >0.999. The limit of detection was 0.017 μg/L, and the limit of quantitation was 0.005 μg/L. The average recoveries were 96.3% and 101.8%, respectively. Days of precision were 3.5%-6.2% and 3.1%-7.4%, respectively, daytime precision were 4.3%-6.8% and 4.7%-8.1%, respectively. Conclusion: The established method of liquid chromatography-tandem mass spectrometry is high sensitivity and accuracy, and is suitable for the determination of perfluorooctanoic acid and perfluorooctane sulfonic acid in human urine.

Perfluorooctanoic acid (PFOA), commonly found in drinking water, leads to widespread exposure through skin contact, inhalation, and ingestion, resulting in detectable levels of PFOA in the bloodstream. In this study, we found that exposure to PFOA disrupts cardiac function in human induced pluripotent stem cell-derived cardiomyocytes (hiPSC-CMs). We observed reductions in field and action potentials in hiPSC-CMs exposed to PFOA. Furthermore, PFOA demonstrated a dose-dependent inhibitory effect on various ion channels, including the calcium, sodium, and potassium channels. Additionally, we noted dose-dependent inhibition of the expression of these ion channels in hiPSC-CMs following exposure to PFOA. These findings suggest that PFOA exposure can impair cardiac ion channel function and decrease the transcription of genes associated with these channels, potentially contributing to cardiac dysfunction such as arrhythmias. Per- and polyfluoroalkyl substances (PFAS) occur widely in drinking water, and consumption of contaminated drinking water is an important human exposure route. Granular activated carbon (GAC) adsorption can effectively remove PFAS from water. To support the design of GAC treatment systems, a rapid bench-scale testing procedure and scale-up approach are needed to assess the effects of GAC type, background water matrix, and empty bed contact time (EBCT) on GAC use rates. The overarching goal of this study was to predict PFAS breakthrough curves obtained at the pilot-scale from rapid small-scale column test (RSSCT) data. The scale-up protocol was developed for pilot data obtained with coagulated/settled surface water (TOC = 2.3 mg/L), three GACs, and two EBCTs. Between 7 and 11 PFAS breakthrough curves were available for each pilot column. RSSCT designs were investigated that assumed intraparticle diffusivity is independent of GAC particle size (i.e., constant diffusivity [CD]) or linearly dependent on GAC particle size (i.e., proportional diffusivity [PD]). CD-RSSCTs effectively predicted the bed volumes of water that could be treated at the pilot-scale to reach 50% breakthrough ( BV 50 % ) of individual PFAS. In contrast, PD-RSSCTs overpredicted BV 50 % obtained at the pilot-scale by a factor of ~2-3. The shape of PFAS breakthrough curves obtained with CD-RSSCTs deviated from those obtained at the pilot-scale, indicating that intraparticle diffusivity was dependent on GAC particle diameter ( d p ) . Using the pore surface diffusion model (PSDM), intraparticle diffusivity was found to be proportional to ( d p ) 0.25 when considering data up to about 70% PFAS breakthrough. This proportionality factor can be used to design RSSCTs or scale up existing CD-RSSCT data using the PSDM. Using pilot-scale data obtained with groundwater and wastewater-impacted groundwater as well as with additional GACs, the developed RSSCT scale-up approach was validated for PFAS breakthrough percentages up to 70%. Lithium Bis(trifluoromethanesulfonyl)imide (LiTFSI ie. HQ-115), a polymer electrolyte used in energy applications, has been detected in the environment, yet its health risks and environmental epigenetic effects remain unknown. This study aims to unravel the potential health risks associated with LiTFSI, investigate the role of DNA methylation-induced toxic mechanisms in its effects, and compare its hepatotoxic impact with the well-studied Perfluorooctanoic Acid (PFOA). Using a murine model, six-week-old male CD1 mice were exposed to 10 and 20 mg/kg/day of each chemical for 14 days as 14-day exposure and 1 and 5 mg/kg/day for 30 days as 30-day exposure. Results indicate that PFOA exposure induced significant hepatotoxicity, characterized by liver enlargement, and elevated serum biomarkers. In contrast, LiTFSI exposure showed lower hepatotoxicity, accompanied by mild liver injuries. Despite higher bioaccumulation of PFOA in serum, LiTFSI exhibited a similar range of liver concentrations compared to PFOA. Reduced Representative Bisulfite Sequencing (RRBS) analysis revealed distinct DNA methylation patterns between 14-day and 30-day exposure for the two compounds. Both LiTFSI and PFOA implicated liver inflammatory pathways and lipid metabolism. Transcriptional results showed that differentially methylated regions in both exposures are enriched with cancer/disease-related motifs. Furthermore, Peroxisome proliferator-activated receptor alpha (PPARα), a regulator of lipid metabolism, was upregulated in both exposures, with downstream genes indicating potential oxidative damages. The ubiquity of dissolved organic matter (DOM) makes it encounter the released perfluorooctanoic acid (PFOA) in subsurface environment. However, the effect of DOM (e.g., humic acid, HA) on PFOA transport in soils and the critical influencing factors and mechanisms remain obscure. Column experiments were conducted to explore PFOA transport with the presence of different concentrations of HA in three types of soils and two types of Al oxide coated sand. Results revealed soil properties significantly regulate the effects of HA on PFOA transport, for which pore size distribution, minerals content (e.g., Al oxide) and pH were critical influencing soil-properties. For soil with large mesopore volume, pore blockage caused by HA controlled the effect of HA on PFOA transport. Large mesopore volume significantly alleviated pore blockage of HA, and led to insignificant effects of HA on PFOA transport. For soil exhibited minimum mesopore volume, Al oxide content and pH dominated the effect of HA on PFOA transport. Results from Al oxide coated sand (low mesopore volume) columns further proved that higher Al oxide content and lower pH caused more significant facilitating effect of HA on PFOA transport via site competition. Perfluorooctanoic acid (PFOA) is an emerging pollutant that is non-biodegradable and presents severe environmental and human health risks. In this study, we present an effective and mild approach for PFOA degradation that involves the use of nitrogen-doped carbon foam anchored with nanoscale zero-valent iron (nZVI@NCF) to activate low concentration peroxymonosulfate (PMS) for the treatment. The nZVI@NCF/PMS system efficiently removed 84.4% of PFOA (2.4 μM). The active sites of nZVI@NCF including Fe 0 (110) and graphitic nitrogen played crucial roles in the degradation. Electrochemical analyses and density functional theory calculations revealed that nZVI@NCF acted as an electronic donor, transferring electrons to both PMS and PFOA during the reaction. By further analyzing the electron paramagnetic resonance and byproducts, it was determined that electron transfer and singlet oxygen were responsible for PFOA degradation. Three degradation pathways involving decarboxylation and surface reduction of PFOA in the nZVI@NCF/PMS system were determined. Seven-day sublethal toxicity tests were performed with the freshwater invertebrates Ceriodaphnia dubia, Hyalella azteca, and Chironomus dilutus to determine the effects of per- or polyfluorinated alkyl substances (PFAS) of varying chain length within four classes: perfluoroalkyl carboxylic acids (PFCAs), perfluoroalkyl sulfonic acids (PFSAs), perfluoroalkane sulfonamides, and fluorotelomer sulfonic acids. In general, toxicity increased with increasing chain length, but the slopes of these relationships varied markedly by species and chemical class. The toxicity of individual PFCAs was similar among species. The toxicity of PFSAs was similar to PFCAs for C. dubia and H. azteca, whereas PFSAs were much more toxic to C. dilutus, with median effect concentrations (EC50s) as low as 0.022 mg perfluorooctane sulfonate (PFOS)/L and 0.012 mg perfluorononane sulfonate (PFNS)/L. Despite the high sensitivity to PFOS and PFNS, C. dilutus was not very sensitive to structurally similar fluorotelomer sulfonates (6:2 and 8:2). Perfluoroalkane sulfonamides were the most toxic class tested among all species (e.g., EC50s of 0.011 and 0.017 mg perfluorooctane sulfonamide/L for C. dilutus and H. azteca, respectively). The efficient transfer of H 2 plays a critical role in catalytic hydrogenation, particularly for the removal of recalcitrant contaminants from water. One of the most persistent contaminants, perfluorooctanoic acid (PFOA), was used to investigate how the method of H 2 transfer affected the catalytic hydrodefluorination ability of elemental palladium nanoparticles (Pd 0 NPs). Pd 0 NPs were synthesized through an in situ autocatalytic reduction of Pd 2+ driven by H 2 from the membrane. The Pd 0 nanoparticles were directly deposited onto the membrane fibers to form the catalyst film. Direct delivery of H 2 to Pd 0 NPs through the walls of nonporous gas transfer membranes enhanced the hydrodefluorination of PFOA, compared to delivering H 2 through the headspace. A higher H 2 lumen pressure (20 vs 5 psig) also significantly increased the defluorination rate, although 5 psig H 2 flux was sufficient for full reductive defluorination of PFOA. Calculations made using density functional theory (DFT) suggest that subsurface hydrogen delivered directly from the membrane increases and accelerates hydrodefluorination by creating a higher coverage of reactive hydrogen species on the Pd 0 NP catalyst compared to H 2 delivery through the headspace. This study documents the crucial role of the H 2 transfer method in the catalytic hydrogenation of PFOA and provides mechanistic insights into how membrane delivery accelerates hydrodefluorination.

Perfluorooctanoic acid (PFOA) and perfluorooctanesulfonic acid (PFOS) are polyfluoroalkyl substances (PFAS) used as surface coatings in manufacturing. Exposure to PFAS was shown to be correlated with infertility, low birth weight, and delayed aspects of pubertal development in mammals. Despite many correlational studies, there have been few direct investigations examining the link between PFAS exposure and early animal development. The aim of this study was to (1) examine the effects of PFOA on development and reproduction using the roundworm Caenorhabditis elegans , a model with a high predictive value for human reproductive toxicity and (2) compare observations to exposure to PFOS. PFAS exposure did not markedly alter egg hatching but delayed population growth, in part due to slower larval development. PFAS-exposed worms took longer to progress through larval stages to reach reproductive maturity, and this was not attributed to PFOA-induced toxicity to their food. Our results provide a robust benchmark for testing developmental and reproductive toxicity for other PFAS and PFAS-alternatives which continue to be used in manufacturing and released into the environment.

In this study, PFOA removal and defluorination were examined during vacuum ultraviolet (VUV) photolysis in the presence of sulfite and sulfite/iodide conditions. PFOA (24 μM) degradation rate constant (k obs ) and defluorination amount in VUV photolysis, and VUV/sulfite, and VUV/sulfite/iodide reactions under nitrogen-purging condition were 5.50 × 10 -3 , 7.26 × 10 -2 , 1.60 × 10 -1 min -1 , and 34.6, 72.7, 73.9% in 6 h, respectively. When tert-butanol (t-BuOH), NO 2 - , and NO 3 - ions were added as radical scavengers, hydrated electrons (e aq - ) was confirmed as the main species responsible for degrading PFOA and mediating defluorination in VUV-based reactions. While, during VUV photolysis, short-chain perfluoroalkyl carboxylic acids (PFCAs), such as PFHpA, PFHxA, PFPeA, and PFBA, were mainly produced as transformation products (TPs) by the chain-shortening mechanism, additional 14 and 15 TPs were identified in the VUV/sulfite and VUV/sulfite/iodide reactions by LC-QTOF/MS, respectively. The main degradation mechanisms in these reactions are H-F exchange (e.g., TP395 (m/z = 394.9739) and TP377 (m/z = 376.9838)), •SO 3 - -F exchange (TP474, m/z = 474.9323), carbon double bond formation by defluorination (e.g., TP392 (m/z = 392.9455), TP410 (m/z = 410.9355), and TP436 (m/z = 436.9347)), and H-F exchange followed by hydration reaction (TP393, m/z = 392.9773), respectively. PFOA degradation pathways were proposed for these VUV-based reactions based on the identified TPs, their time profiles, and the density functional theory (DFT). Due to wastes from production of fluorinated materials and use of aqueous fire-fighting foams (AFFF), soils contaminated with perfluorooctanoic acid (PFOA) is of concern. However, current PFOA-contaminated soil disposal techniques have relatively low degradation efficiencies and are not suitable for on-site remediation. In this study, an electrical resistance heating (ERH) device and a box experimental device were used to study whether ERH induced persulfate activation (ERH/PS) could degrade PFOA in the soil. The results indicated that single ERH and single PS addition could not effectively degrade PFOA (with approximately 0.3 % and 3.9 % degradation after 9 h, respectively), while the degradation efficiency of PFOA with coupled ERH/PS could reach 87.3 % after 9 h of reaction. Moreover, effects of PS content, heating temperature, and soil organic matter on the degradation of PFOA were explored. During the ERH/PS process, PFOA was gradually transformed into short chain perfluorinated compounds and finally mineralized to fluoride ions. Finally, using a box experimental device, PS was effectively transported to the target contaminated area through electrokinetic (EK)-assisted delivery. After activating PS through ERH, the degradation rate of PFOA could reach 95.5 %. Perfluorooctanoic acid (PFOA) and perfluorooctane sulfonate (PFOS) are among the most frequently detected chemicals among the per- and polyfluoroalkyl substances in aquatic environments. Because of their high detection frequency, persistence, and potential toxicity, interest in both PFOA and PFOS has increased in recent years. However, a substantial number of PFOA and PFOS toxicity tests only report nominal, or unmeasured, treatment concentrations, which may complicate the determination of protective values. In addition, previous literature has indicated that differences between nominal and measured concentrations of both PFOA and PFOS could be linked to experimental conditions (e.g., feeding regimes for test organisms, test vessel material [glass or plastic], use of solvent, and the presence of substrate). Therefore, this critical review examined whether nominal and measured concentrations were in close agreement with each other among the current PFOA and PFOS aquatic toxicity literature and if experimental conditions were associated with any observed differences. Nominal and measured concentrations in the current PFOA and PFOS aquatic toxicity literature generally displayed a high degree of linear correlation and relatively low median percent differences. Correlations between measured and nominal concentrations were >0.98 for PFOA and >0.95 for PFOS in freshwater tests across experimental conditions. For saltwater tests, correlations of >0.84 were observed for PFOA and PFOS (separate and combined) across experimental conditions. While measured PFOA and PFOS toxicity tests are generally preferred, the present meta-analysis demonstrated that experimental conditions had little influence on observed discrepancies between nominal and measured concentrations, with the exception of PFOS saltwater tests and PFOA and PFOS freshwater studies that contained substrate. Unmeasured tests with these conditions should be considered carefully based on project needs, with the caveat that the data sets for these two experimental conditions were limited. Cellulose-based materials are a sustainable alternative to polymers derived from petroleum. Cellulose nanocrystal (CNC) is a biopolymer belonging to this family; it is commonly known for its important physical and chemical properties and ability to form a film. Modifying CNC via electrostatic interaction provided by cationic polymers is a facile and promising technique to enlarge the application of CNC. Herein, we report the preparation of films, from blends of negatively charged CNC and positively charged poly (trimethyl aminoethyl methacrylate) (PTMAEMA). The interaction between CNC and PTMAEMA was verified by using a quartz crystal microbalance with dissipation monitoring (QCM-D), as well as by measuring the particle size and ζ-potential of the casting mixture. Per- and polyfluoroalkyl substances (PFAS) are man-made long-lasting chemical compounds that are found in everyday household items. Today they occur in the environment as a major group of pollutants. These compounds are broadly used in commercial product preparation such as, for food packaging, nonstick coatings, and firefighting foam. In humans, PFAS can cause immune disorders, impaired fetal development, abnormal skeletal tissue development, osteoarthritis, thyroid dysfunctions, cholesterol changes, affect insulin regulation and lipid metabolism, and are also involved in the development of fatty liver disease. In the current study, we investigated the effect of low, but physiologically relevant, concentrations of perfluorooctanoic acid (PFOA), heptafluorobutyric acid (HFBA), and perfluorotetradecanoic acid (PFTA) on gene expression markers of an inflammatory response (tnfa, il-1b, il-6, rplp0, edem1, and dnajc3a), unfolded protein response (UPR) (bip, atf4a, atf6, xbp1, and ddit3), senescence (p21, pai1, smp30, mdm2, and baxa), lipogenesis (scd1, acc, srebp1, pparγ, and fasn) and autophagy (p62, atg3, atg7, rab7, lc3b, and becn1) in AB wild-type (+/+), spns1-wt sibling (+/+), (+/-) and spns1 homozygous mutant (-/-) zebrafish embryos. Exposure to PFOA and HFBA (50 and 100 nM) specifically modulated inflammatory, UPR, senescence, lipogenic, and autophagy signaling in spns1-wt (+/+), (+/-), and spns1-mutant (-/-) zebrafish embryos. Furthermore, PFOA, but not HFBA, upregulated lipogenic-related gene expression and enhanced hepatic steatosis in spns1-wt (+/+), (+/-) zebrafish embryos. Combined exposure to PFOA, HFBA, and PFTA differentially expressed inflammatory, senescence, lipogenic, and autophagy-associated gene expression in spns1-mutant (-/-) zebrafish embryos compared with spns1-wt (+/+), (+/-) and AB-wt (+/+) zebrafish embryos. In addition, chronic exposure (∼2 months) to PFOA (120-600 nM) upregulated the expression of hepatic lipogenic and steatosis biomarkers in AB-wt (+/+) zebrafish. The present study investigates the impact of two endocrine disruptors, namely Bisphenols (BPs) and Perfluoroalkyls (PFs), on human stem cells. These chemicals leach from plastic, and when ingested through contaminated food and water, they interfere with endogenous hormone signaling, causing various diseases. While the ability of BPs and PFs to cross the placental barrier and accumulate in fetal serum has been documented, the exact consequences for human development require further elucidation. The present research work explored the effects of combined exposure to BPs (BPA or BPS) and PFs (PFOS and PFOA) on human placenta (fetal membrane mesenchymal stromal cells, hFM-MSCs) and amniotic fluid (hAFSCs)-derived stem cells. The effects of the xenobiotics were assessed by analyzing cell proliferation, mitochondrial functionality, and the expression of genes involved in pluripotency and epigenetic regulation, which are crucial for early human development. Our findings demonstrate that antenatal exposure to BPs and/or PFs may alter the biological characteristics of perinatal stem cells and fetal epigenome, with potential implications for health outcomes at birth and in adulthood. Further research is necessary to comprehend the full extent of these effects and their long-term consequences.

Exposure to persistent organic pollutants during the perinatal period is of particular concern because of the potential increased risk of neurological disorders in adulthood. Here we questioned whether exposure to perfluorooctanoic acid (PFOA) and perfluorooctane sulfonate (PFOS) could alter myelin formation and regeneration. First, we show that PFOS, and to a lesser extent PFOA, accumulated into the myelin sheath of postnatal day 21 (p21) mice, whose mothers were exposed to either PFOA or PFOS (20 mg/L) via drinking water during late gestation and lactation, suggesting that accumulation of PFOS into the myelin could interfere with myelin formation and function. In fact, PFOS, but not PFOA, disrupted the generation of oligodendrocytes, the myelin-forming cells of the central nervous system, derived from neural stem cells localised in the subventricular zone of p21 exposed animals. Then, cerebellar slices were transiently demyelinated using lysophosphatidylcholine and remyelination was quantified in the presence of either PFOA or PFOS. Only PFOS impaired remyelination, a deleterious effect rescued by adding thyroid hormone (TH). Similarly to our observation in the mouse, we also showed that PFOS altered remyelination in Xenopus laevis using the Tg(Mbp:GFP-ntr) model of conditional demyelination and measuring, then, the number of oligodendrocytes. The functional consequences of PFOS-impaired remyelination were shown by its effects using a battery of behavioural tests. In sum, our data demonstrate that perinatal PFOS exposure disrupts oligodendrogenesis and myelin function through modulation of TH action. Per- and polyfluoroalkyl substances (PFAS) are an emerging class of compounds that cause health and environmental problems worldwide. In aquatic environments, PFAS may bioaccumulate in sediment organisms, which can affect the health of organisms and ecosystems. As such, it is important to develop tools to understand their bioaccumulation potential. In the present study, the uptake of perfluorooctanoic acid (PFOA) and perfluorobutane sulfonic acid (PFBS) from sediments and water was assessed using a modified polar organic chemical integrative sampler (POCIS) as a passive sampler. While POCIS has previously been used to measure time-weighted concentrations of PFAS and other compounds in water, in our study, the design was adapted for analyzing contaminant uptake and porewater concentrations in sediments. The samplers were deployed into seven different tanks containing PFAS-spiked conditions and monitored over 28 days. One tank contained only water with PFOA and PFBS, three tanks contained soil with 4% organic matter, and three tanks contained soil combusted at 550 °C to minimize the influence of labile organic carbon. The uptake of PFAS from the water was consistent with previous research using a sampling rate model or simple linear uptake. For the samplers placed in the sediment, the uptake process was explained well using a mass transport based on the external resistance from the sediment layer. Uptake of PFOS in the samplers occurred faster than that of PFOA and was more rapid in the tanks containing the combusted soil. A small degree of competition was observed between the two compounds for the resin, although these effects are unlikely to be significant at environmentally relevant concentrations. The external mass transport model provides a mechanism to extend the POCIS design for measuring porewater concentrations and sampling releases from sediments. Background: Perfluorooctanoic acid (PFOA) is detected in the blood of virtually all biomonitoring study participants. Assessing health risks associated with blood PFOA levels is challenging because exposure guidance values (EGVs) are typically expressed in terms of external dose. Biomonitoring equivalents (BEs) consistent with EGVs could facilitate health-based interpretations., Objective: To i) derive BEs for serum/plasma PFOA corresponding to non-cancer EGVs of the U.S. Environmental Protection Agency (U.S. EPA), the Agency for Toxic Substances and Disease Registry (ATSDR) and Health Canada, and ii) compare with PFOA concentrations from national biomonitoring surveys., Methods: Starting from EGV points of departure, we employed pharmacokinetic data/models and uncertainty factors. Points of departure in pregnant rodents (U.S. EPA 2016, ATSDR) were converted into fetus and pup serum concentrations using an animal gestation/lactation pharmacokinetic model, and equivalent human fetus and child concentrations were converted into BEs in maternal serum using a human gestation/lactation model. The point of departure in adult rodents (Health Canada) was converted into a BE using experimental data. For epidemiology-based EGVs (U.S. EPA 2023, draft), BEs were directly based on epidemiological data or derived using a human gestation/lactation pharmacokinetic model. BEs were compared with Canadian/U.S. biomonitoring data., Results: Non-cancer BEs (ng/mL) were 684 (Health Canada, 2018) or ranged from 15 to 29 (U.S. EPA, 2016), 6-10 (ATSDR, 2021) and 0.2-0.8 (U.S. EPA, 2023, draft). Ninety-fifth percentiles of serum levels from the 2018-2019 Canadian Health Measures Survey (CHMS) and the 2017-2018 National Health and Nutrition Examination Survey (NHANES) were slightly below the BE for ATSDR, and geometric means were above the non-cancer BEs for the U.S. EPA (2023, draft)., Conclusion: Non-cancer BEs spanned three orders of magnitude. Perfluorooctanoic acid (PFOA) is the most prominent member of a widely utilized family of compounds named Perfluoroalkyl substances (PFASs). Initially produced for use in both industrial and consumer applications, it has since been recognized that PFASs are extremely persistent in the environment where they have been characterized as persistent organic pollutants (POPs). While previous studies have demonstrated that PFOA may induce disorders of lipid and carbohydrate metabolism, the precise mechanisms by which PFOA produces this phenotype and the involvement of downstream AMPK/mTOR pathways remains unclear. In this study, male rats were exposed to 1.25, 5 and 20 mg PFOA/kg body weight/day for 28 days by oral gavage. After 28 days, blood was collected and tested for serum biochemical indicators and livers were removed and weighed. To investigate aberrant metabolism in rats exposed to PFOA, livers were analyzed by performing LC-MS/MS untargeted metabolomics, quantitative real-time PCR, western blotting, immunohistochemical staining was also performed on exposed tissues. Our results showed that exposure to PFOA induced liver damage, increased the expression of glucose and lipid related biochemical indexes in liver and serum, and altered the expression levels of AMPK/mTOR pathway related genes and proteins. Perfluorooctanoic acid (PFOA) is an environmentally ubiquitous synthetic chemical highly persistent in organisms. PFOA exposure is pernicious to reproductive health as indicated by reports of male infertility. However, the PFOA toxicity mechanism to Leydig cells remains poorly understood. Therefore, this study aimed to investigate the toxicological events occurring in TM3 Leydig cells treated with PFOA (250, 500, 750 µM) for 24 h. PFOA was shown to significantly decrease cell viability resulting from inhibition of proliferation and elevation of apoptotic ratio in a dose dependent manner. Upregulation of pro-apoptotic gene expressions such as Bax, Bad, and p53, was observed in combination with an increase in the apoptosis-related protein levels of Bax, cleaved caspase-3, cleaved caspase-8, and phosphorylated p53. Furthermore, exposure of PFOA lead to mitochondrial damage involving mitochondrial membrane permeabilization. A release of cytochrome c and collapse of the mitochondrial membrane potential (∆Ψm) were observed compared to the untreated control. Additionally, PFOA stimulated unfolded protein response (UPR) upregulating ER stress marker, Bip/GRP78, and upregulated protein levels of UPR signal molecules IRE1, p-JNK, p-ERK1/2, p-p53, CHOP, and ERO1. Background: Per- and polyfluoroalkyl substances (PFASs) including perfluorooctanoic acid (PFOA) are ubiquitous environmental contaminants. Prior analysis in the large "C8 Health Project" population defined abnormal alanine aminotransferase (ALT) with statistically derived cutoffs (>45 IU/L in men, >34 IU/L in women)., Objective: To explore the degree to which PFOA was associated with modern, clinically predictive ALT biomarker cutoffs in obese and nonobese participants, excluding those with diagnosed liver disease., Methods: We reevaluated the relationship of serum PFOA to abnormal ALT using predictive cutoff recommendations including those of the American College of Gastroenterology (ACG). Evaluations modeled lifetime cumulative exposure and measured internal PFOA exposure., Results: ACG cutoff values (≥34 IU/L for males, ≥25 IU/L for females) classified 30% of males (3815/12,672) and 21% of females (3359/15,788) above ALT cutoff values. Odds ratios (OR) for above cutoff values were consistently associated with modeled cumulative and measured serum PFOA. Linear trends were highly significant. ORs by quintile showed near monotonic increases. Trends were stronger for the overweight and obese. However, all weight classes were affected., Conclusion: Predictive cutoffs increase the OR for abnormal ALT results. Obesity increases ORs, yet association with abnormal ALT pertains to all weight classes. The results are discussed in context of current knowledge about the health implications of PFOA hepatotoxicity.

Poor reactivity of extraordinarily strong per- and polyfluoroalkyl substances (PFAS) with TiO 2 makes it necessary to advance photocatalytic materials. In this present work, lead (Pb)-doped TiO 2 coated with reduced graphene oxide (TiO 2 -Pb/rGO) was prepared using hydrothermal method, and then its photocatalytic activity with various PFAS in water, especially perfluorooctanoic acid (PFOA), was investigated. PFAS decomposition kinetics by TiO 2 -Pb/rGO was compared with neat TiO 2 , Pb-doped TiO 2 (TiO 2 -Pb), and rGO-coated TiO 2 (TiO 2 /rGO). TiO 2 -Pb/rGO (0.33 g/L) under ultraviolet (UV) showed superior removal of PFOA (10 mg/L) at 98% after 24 h, following TiO 2 -Pb/UV at 80%, TiO 2 /rGO/UV at 70%, TiO 2 /UV at <10%, and UV at <10%. Doping of TiO 2 with Pb and introduction of rGO to TiO 2 greatly changed the physicochemical properties of TiO 2 and the subsequent charge transfer mechanism. Radical scavenger experiments indicated that holes, superoxide radical anion, and singlet oxygen were responsible for the observed PFOA decomposition. Decomposition of PFOA by TiO 2 -Pb/rGO under UV led to formation of short-chain perfluorocarboxylic acids (PFCAs) as reaction intermediates through step-by-step removal of CF 2 units. Polyfluoroalkyl substance (6:2 fluorotelomer sulfonate [6:2FTS]) and long-chain PFCAs such as PFOA were significantly removed and defluorinated by TiO 2 -Pb/rGO, whereas it was ineffective toward perfluorosulfonic acids and short-chain PFCAs. Removal kinetics decreased in the order of 6:2FTS > PFOA >> PFOS > PFHpA ≈ PFHxS ≈ PFBA ≈ PFBS. Pb doping to TiO 2 /rGO showed better performance than Fe doping. Overall, this study implied that proper designing of TiO 2 photocatalytic materials enables to expedite the decomposition of persistent organic pollutants in water, in particular highly challenging fluorinated chemicals. PRACTITIONER POINTS: Photocatalytic decomposition of various PFAS using TiO 2 -Pb/rGO was studied. TiO 2 -Pb/rGO shows better photoactivity towards PFAS than TiO 2 -Pb and TiO2/rGO system. Scavenger test indicated that h+, • O2-, and iO 2 are responsible for PFOA removal. Using TiO 2 -Pb/rGO, PFOA removal was comparable under UVA, UVB, and UVC, and it can be explained by spanning UV absorption to 415 nm. Liquid-liquid extraction (LLE) using ionic liquids (ILs)-based methods to remove perfluoroalkyl chemicals (PFACs), such as perfluorooctanoic acid (PFOA) and perfluorooctane sulfonic acid (PFOS), from wastewater, is an important strategy. However, the lack of physicochemical and LLE data limits the selection of the most suitable ILs for the extraction of PFACs. In this work, 1763 ILs for PFACs extraction from water were systematically screened using COSMOtherm to estimate the infinite dilution activity coefficient (lnγ ∞ )of PFOA and PFOS in water and ILs. To evaluate the accuracy of COSMOtherm, 8 ILs with various lnγ ∞ values were selected, and their extraction efficiency (E) and distribution coefficient (D exp ) were measured experimentally. The results showed that the predicted lnγ ∞ decreased as the increase of experimental extraction efficiency of PFOA or PFOS, while the tendency of predicted distribution coefficient (D pre ) was consistent with the experimental (D exp ) results. Perfluorooctanoic acid (PFOA), a class of permanent organic pollutants, is frequently detected in surface and ground water, with the latter made up primarily of porous media (such as soils, sediments, and aquifers) that harbor microbial communities. Therefore, we investigated the effects of PFOA on water ecosystems and found that, under stimulation by 2.4 μM PFOA, denitrifiers were significantly enriched due to their hosting antibiotic resistant genes (ARGs), which were 1.45 times more abundant than the control. Furthermore, denitrifying metabolism was enhanced by Fe(II) electron donation. Specifically, 2.4 μM PFOA significantly enhanced the removal of total inorganic nitrogen by 178.6%. The microbial community became predominated by denitrifying bacteria (67.8% abundance). Notably, the nitrate-reduction ferrous-oxidizing (NRFO) bacteria Dechloromonas, Acidovorax, Bradyrhizorium, etc. were significantly enriched. The selective pressures of PFOA driving the enrichment of denitrifiers were twofold. First, the toxic PFOA induced denitrifying bacteria to produce ARGs, mainly including the efflux (occupying 55.4%) and antibiotic inactivation (occupying 41.2%) types, which improved microbial tolerance to PFOA. The risk of horizontal ARGs transmission was elevated as the overall number of horizontally transmissible ARGs increased by 47.1%. Second, Fe(II) electrons were transported via the porin-cytochrome c extracellular electrons transfer system (EET), promoting the expression of nitrate reductases, which in turn further enhanced denitrification. Understanding the fate and transport of perfluorooctanoic acid (PFOA) in soil and groundwater is essential to reliable assessments of its risks. This study investigated the impacts of Gram-positive Bacillus subtilis (BS), Gram-negative Pseudomonas aeruginosa (PA) and wild microbiota (WM) biofilm on the transport of PFOA in saturated sand columns at two ionic strengths (i.e., 1.0 and 20.0 mM NaCl). The retention of PFOA in biofilm-coated sand columns was higher than that in uncoated sand columns, due to biofilm-induced reinforced hydrophobic interactions and surface roughness, and decreased zeta potential. However, the retention effects varied among biofilm bacterial species with PFOA retardation factors in PA, WM and BS columns of 1.29-1.38, 1.21-1.29 and 1.11-1.15, respectively. Notably, PA biofilm had the most pronounced effect on PFOA retention. While increasing ionic strength promoted the retention of PFOA in BS biofilm-coated sand, it had no significant impact on PFOA transport in PA and WM biofilm-coated sand. This could be attributed to the differences in biofilm composition, deviating the ionic strengths effects on electrostatic double layer compression. The advection dispersion equation coupled with two-site kinetic retention model well described the transport of PFOA in all saturated columns. Owing to its difficulty in degrading and ease of accumulation in the body, perfluorooctanoic acid (PFOA) has a detrimental effect on reproduction. This study aimed to examine the effect of PFOA concentration in follicular fluid during ovulation stimulation on embryo quality and the impact of PFOA exposure on the metabolic components of follicular fluid. This was a single-center prospective study that included 25 patients with diminished ovarian reserve (DOR), 25 with normal ovarian reserve (NOR), and 25 with polycystic ovary syndrome (PCOS). Follicular fluid samples were analyzed using ultra-high performance liquid chromatography-tandem mass spectrometry. We demonstrated that the PFOA levels of follicular fluid in the DOR group were higher than those in the NOR group and PCOS group (P < 0.05). PFOA concentration in the PCOS group was negatively correlated with high-quality embryos (P < 0.05). To gain more insight into the impact of PFOA on the metabolic composition of follicular fluid, we classified the DOR group based on the PFOA concentration, for which metabolomic analysis was performed. In the high-concentration PFOA group, there was an increase and a decrease in three and nine metabolites, respectively, compared to that in the low-concentration group. Perfluorooctanoic acid (PFOA) has been largely used in the manufacturing industry but a few years ago it turned out to be a dangerous pollutant which is now of concern for terrestrial and aquatic environments. Here, we investigated the bioaccumulation of PFOA in the sea urchin Paracentrotus lividus after exposure to different concentrations of the pollutant for 28 days. We observed rapid uptake of PFOA in the coelomic fluid collected weekly during the exposure period and high bioaccumulation in gonads at the end of the experiment. Interestingly, animals were also able to fast depurate when relocated to a clean environment. In addition, to assess the effect of PFOA on sea urchins' physiological pathways, we analysed the expression profile of some marker genes both in the gonads and in the embryos obtained from parents exposed to PFOA. Our results suggest that PFOA is a persistent, bioaccumulative compound that adversely affects the health of the exposed organisms and their offspring by causing significant changes in the expression of some key target genes and the occurrence of developmental anomalies in the embryos., (© 2022. Carbonaceous materials have emerged as a method of persulfate activation for remediation. In this study, persulfate activation using powdered activated carbon (PAC) was demonstrated at temperatures relevant to groundwater (5-25 °C). At room temperature, increasing doses of PAC (1-20 g L -1 ) led to increased persulfate activation (3.06 × 10 -6 s -1 to 2.10 × 10 -4 with 1 and 20 g L -1 PAC). Activation slowed at lower temperatures (5 and 11 °C); however, substantial (>70 %) persulfate activation was achieved. PAC characterization showed that persulfate is activated at the surface of the PAC, as indicated by an increase in the PAC C:O ratio. Similarly, electron paramagnetic resonance (EPR) spectroscopy studies with a spin trapping agents (5,5-dimethyl-1-pyrroline N-oxide (DMPO)) and 2,2,6,6-tetramethylpiperidine (TEMP) revealed that singlet oxygen was not the main oxidizing species in the reaction. DMPO was oxidized to form 5,5-dimethylpyrrolidone-2(2)-oxyl-(1) (DMPOX), which forms in the presence of strong oxidizers, such as sulfate radicals. The persulfate/PAC system is demonstrated to simultaneously degrade both perfluorooctanoic acid (PFOA) and 1,4-dioxane at room temperature and 11 °C. With a 20 g L -1 PAC and 75 mM persulfate, 80 % and 70 % of the PFOA and 1,4-dioxane, respectively, degraded within 6 h at room temperature. At 11 °C, the same PAC and persulfate doses led to 57% dioxane degradation and 54 % PFOA degradation within 6 h. The present study investigated the roles of peroxydisulfate (PDS) radicals and sulfate radicals (SO 4 •- ) during electrochemical oxidation of perfluorooctanoic acid (PFOA). The effect of operating parameters such as different types of electrolytes (NaCl, NaClO 4 2- ) during electrochemical oxidation of perfluorooctanoic acid (PFOA). The effect of operating parameters such as different types of electrolytes (NaCl, NaClO 4 ), initial pH, current density, dose of electrolyte, and initial concentration of PFOA using electrochemical oxidation for perfluorooctanoic acid (PFOA) decomposition study was investigated. A difference in the removal efficiency with different electrolytes (i.e., Cl 2 SO 4 ), initial pH, current density, dose of electrolyte, and initial concentration of PFOA using electrochemical oxidation for perfluorooctanoic acid (PFOA) decomposition study was investigated. A difference in the removal efficiency with different electrolytes (i.e., Cl - , ClO 4 - , and SO 4 2- ) illustrated an increasing effect of electrooxidation of PFOA in the order of ClO 4 -  < Cl -  < SO 4 2- , which suggested that • OH induced oxidation and direct e - transfer reaction continued to play a crucial role in oxidation of PFOA. At the optimum treatment condition of j = 225.2 Am -2 , Na 2 SO 4 concentration = 1.5 gL -1 , [PFOA] o  = 50 mgL -1 and initial pH = 3.8 maximum PFOA removal of 92% and TOC removal of 80% was investigated at 240 min. The formation of three shorter-chain perfluorocarboxylates (i.e., perfluoroheptanoic acid (PFHpA), perfluorohexanoic acid (PFHxA), and perfluoropentanoic acid (PFPeA) and formate (HCOO - ) ions were detected as by-products of PFOA electro-oxidation, showing that the C-C bond first broken in C 7 F 15 and then mineralized into CO 2 , and fluoride ion (F - ). The fluorine recovery as F - ions and the organic fluorine as the shorter-chain by-products were also obtained. Per- and polyfluoroalkyl substances (PFAS) are a complex group of man-made chemicals with high stability and mobility leading to ubiquitous environmental contamination and accumulation especially of some long-chain perfluoroalkyl acids (PFAA) in humans. While dietary intake is the main route of exposure, transdermal uptake from cosmetic products usually is considered negligible. However, PFAS are present in a part of these products, and recent epidemiological studies have provided evidence for relevant uptake via this route. The crucial question is whether PFAA in cosmetic products can cross the human skin barrier. A defined amount (110 µg) of 13 C 4 -perfluorooctanoic acid ( 13 C 4 -PFOA) was mixed into a sunscreen (30 g) which was applied on the whole skin of a volunteer. The plasma concentrations of 13 C 4 -PFOA were determined in serial blood samples taken over 115 days using UHPLC-MS/MS and 13 C 2 -PFOA as internal standard. After application, 13 C 4 -PFOA plasma levels increased continuously, reaching levels of 3, 56 and 118 ng/L after 6 h, 3 days and 10 days, respectively. A maximum level of 132 ng/L was measured 22 days after application, representing 9.4 % of the PFOA level resulting from the volunteer's background exposure (1400 ng/L, equivalent to 1.4 ng/mL). In the following weeks, the levels slightly decreased with an estimated half-life of 1.8 years. Perfluorooctanoic acid (PFOA), a "forever chemical", is continuously discharged and mitigated in the environment despite its production and use being severely restricted globally. Due to the transformation, attachment, and adsorption of PFOA in aquatic environments, PFOA accumulates in the porous media of sediments, soils, and vadose regions. However, the impact of trace PFOA in the porous media on interstitial water and water safety is not clear. In this work, we simulated a porous media layer using a sand column and explored the effects of µg-level PFOA migration on microbial community alternation, microbial function regulation, and the generation and spread of microbial risks. After 60 days of PFOA stimulation, Proteobacteria became the dominant phylum with an abundance of 91.8%, since it carried 71% of the antibiotic resistance genes (ARGs). Meanwhile, the halogen-related Dechloromonas abundance increased from 0.4% to 10.6%. In addition, PFOA significantly stimulated protein (more than 1288%) and polysaccharides (more than 4417%) production by up-regulating amino acid metabolism (p< 0.001) and membrane transport (p < 0.001) to accelerate the microbial aggregation. More importantly, the rapidly forming biofilm immobilized and blocked PFOA. The more active antioxidant system repaired the damaged cell membrane by significantly up-regulating glycerophospholipid metabolism and peptidoglycan biosynthesis. It is worth noting that PFOA increased the abundance of antibiotic resistance genes (ARGs) and human bacterial pathogens (HBPs) in porous media by 30% and 106%. PFOA increased the proportion of vertical transmission ARGs (vARGs), and co-occurrence network analysis (r ≥ 0.8, p ≤ 0.01) verified that vARGs were mainly mediated by HBPs. Perfluorooctanoic acid (PFOA) can rapidly activate signaling pathways independent of nuclear hormone receptors through membrane receptor regulation, which leads to endocrine disrupting effects. In the present work, the molecular initiating event (MIE) and the key events (KEs) which cause the endocrine disrupting effects of PFOA have been explored and determined based on molecular dynamics simulation (MD), fluorescence analysis, transcriptomics, and proteomics. MD modeling and fluorescence analysis proved that, on binding to the G protein-coupled estrogen receptor-1 (GPER), PFOA could induce a conformational change in the receptor, turning it into an active state. The results also indicated that the binding to GPER was the MIE that led to the adverse outcome (AO) of PFOA. In addition, the downstream signal transduction pathways of GPER, as regulated by PFOA, were further investigated through genomics and proteomics to identify the KEs leading to thr endocrine disrupting effects. Two pathways (Endocrine resistance, ERP and Estrogen signaling pathway, ESP) containing GPER were regulated by different concentration of PFOA and identified as the KEs.
