Aim: Food literacy programs aim to improve food related skills and knowledge and are selected Governments as a strategy to address dietary intakes at a community level. The purpose of this research is to identify behaviour change techniques in a food literacy program, which were assessed by the achievement of participant goals related to food literacy and dietary behaviour changes. Methods: Consensus methods were applied to map behaviour change techniques to an adult food literacy program. A second phase investigation applied content analysis of participant process evaluation data (collected between 2016 and June 2021) to align target behaviours and behaviour change techniques. Chi-square and ANCOVA were used to assess the statistically significant demographic characteristics, food literacy, and dietary behaviour scores for participants who set goals and recorded changes. Results: A total of 4697 program participants provided evaluation data from the 4-week food literacy program. Participants who set goals and made changes were statistically more likely to have higher food literacy factor scores (p < 0.001) and fruit serves (p = 0.004). Statistical analysis showed that participants were more likely to have set goals and recorded changes if they were female (p < 0.001), older (p < 0.001), higher education level (p < 0.001), had a higher socioeconomic status (p = 0.049), lived with children (p = 0.014), were born in Australia (p = 0.019), or did not identify as Indigenous (p < 0.001). The behaviour change technique mapping process identified nine techniques used weekly and a total of 22 techniques used over the entire 4 week program curriculum. Conclusions: This is the first Australian study to link the contribution of behaviour change techniques to food literacy and dietary behaviour change in an established effective food literacy program. Knowing the behaviour change techniques associated with effective programs will facilitate replication of effective intervention

Despite viral suppression, people living with HIV (PLHIV) report lower quality of life (QoL) than the general population, negatively impacting treatment adherence and wellbeing. This qualitative study explored factors influencing QoL of PLHIV. Participants completed a QoL questionnaire, with cut-off scores used to allocate participants into Low-Moderate QoL (n = 11) or High-Very High QoL (n = 10) focus groups. Thematic analysis indicated convergence across factors perceived to impact QoL, with some notable discrepancies. Socioeconomic stability, social connection and support, maintaining good health, adaptive attitudes and reduced impact of stigma were perceived to improve QoL, while obstacles to connection, ageing and poor HIV literacy in the general population were perceived to worsen QoL in both groups. The Low-Moderate QoL group alone identified socioeconomic stressors and ongoing burden of negative life experiences worsened their QoL. Results are presented in the context of local and global HIV health strategies, with implications for clinical management noted.

Global declines in ecosystem extent and condition mean there is an increasing demand for recovery and conservation plans. Conservation plans for ecological communities require a management framework with measurable, time‐bound objectives. Efficient and structured processes that facilitate timely and comparable conservation plans are essential, especially where resources are constrained. We describe a process to streamline the development of conservation plans by combining functionally similar community sub‐types into a multi‐community State and Transition Model that can be used to guide conservation planning. We demonstrate this approach in a case study using eucalypt dominated woodlands of southern Australia—an ecosystem which occupies a vast geographical range across temperate Australia and includes many distinct vegetation communities, a growing number of which are endangered or threatened. Australian woodland ecologists (grouped according to their knowledge of three broad woodland sub‐types) were asked to develop causal chains to describe all factors associated with transitions among woodland condition states, and estimate the likelihoods associated with each transition at two time‐scales. The resultant State and Transition Model includes a set of eight general condition states that are common to eucalypt dominated woodlands, and 364 unique causal chains describing the drivers of all plausible transitions. We include an example of how the same information can be presented as a series of decision trees aimed at supporting on‐ground management decisions. The case study demonstrates that it is possible to construct a detailed State and Transition Model that synthesizes knowledge across multiple similar vegetation communities. We present a new determination of the evolving galaxy UV luminosity function (LF) over the redshift range $8.58.5)>0.05$) to undertake a statistical calculation of the evolving UV LF. Our new measurements span $\simeq4$ magnitudes in UV luminosity at $z=9-12.5$, placing new constraints on both the shape and evolution of the LF at early times. We fit our observational data-points with a double-power law (DPL) function and explore the evolution of the DPL parameters. Our UV LF measurements yield a new estimate of the early evolution of cosmic star-formation rate density ($\rho_{\rm{SFR}}$) which confirms the gradual, log-linear decline deduced from early JWST studies, at least out to $z \simeq 12$. Finally we show that the observed early evolution of the galaxy UV LF (and $\rho_{\rm{SFR}}$) can be reproduced in a ${\rm \Lambda}$CDM Universe, with no change in dust properties or star-formation efficiency required out to $z \simeq 12$. Instead, we show that a progressive trend towards younger stellar population ages can reproduce the observations, and we show that the typical ages required at $z \simeq$ 8, 9, 10, and 11 all converge on a time $\simeq 380-330$ Myr after the Big Bang, indicative of a rapid emergence of early galaxies at $z \simeq 12 - 13$. This is consistent with the first indications of a steeper drop-off in the galaxy population we find beyond $z \simeq 13$, possibly reflecting the rapid evolution of the halo mass function at earlier times., Comment: 15 pages, 9 figures, submitted to MNRAS

BOLT (Battery Operated Light Transport) Aerospace is a student design team developing an electric Vertical Takeoff and Landing (VTOL) aircraft to compete in the Vertical Flight Society’s Design-Build-Vertical Flight Competition. The competition consists of two phases, one piloted, and the other autonomous. In both phases, the aircraft must be fitted with a payload, take off vertically, complete a lap around the designated course, and land vertically. The participating teams will receive points for the number of laps completed in ten minutes, the weight of the payload, and the fastest lap time. The competition also includes a scored technical report and presentation. The aircraft designed by BOLT Aerospace has been designated CARLOS (Craft for Aerial Relocation of Light Objects and Supplies) and will be electrically powered, capable of VTOL, have a six-foot wingspan, and a gross takeoff weight of twenty pounds.

Cells respond to environmental stress by regulating gene expression at the level of both transcription and translation. The ∼50 modified ribonucleotides of the human epitranscriptome contribute to the latter, with mounting evidence that dynamic regulation of transfer RNA (tRNA) wobble modifications leads to selective translation of stress response proteins from codon-biased genes. Here we show that the response of human hepatocellular carcinoma cells to arsenite exposure is regulated by the availability of queuine, a micronutrient and essential precursor to the wobble modification queuosine (Q) on tRNAs reading GUN codons. Among oxidizing and alkylating agents at equitoxic concentrations, arsenite exposure caused an oxidant-specific increase in Q that correlated with up-regulation of proteins from codon-biased genes involved in energy metabolism. Limiting queuine increased arsenite-induced cell death, altered translation, increased reactive oxygen species levels, and caused mitochondrial dysfunction. In addition to demonstrating an epitranscriptomic facet of arsenite toxicity and response, our results highlight the links between environmental exposures, stress tolerance, RNA modifications, and micronutrients.

The photochemical degradation pathways of 6PPD-quinone (6PPDQ, 6PPD-Q), a toxic transformation product of the tire antiozonant 6PPD, were determined under simulated sunlight conditions typical of high-latitude surface waters. Direct photochemical degradation resulted in 6PPDQ half-lives ranging from 17.5 h at 20 °C to no observable degradation over 48 h at 4 °C. Sensitization of excited triplet-state pathways using Cs+ and Ar purging demonstrated that 6PPDQ does not decompose significantly from a triplet state relative to a singlet state. However, assessment of processes involving reactive oxygen species (ROS) quenchers and sensitizers indicated that singlet oxygen and hydroxyl radical do significantly contribute to the degradation of 6PPDQ. Investigation of these processes in natural lake waters indicated no difference in attenuation rates for direct photochemical processes at 20 °C. This suggests that direct photochemical degradation will dominate in warm waters, while indirect photochemical pathways will dominate in cold waters, involving ROS mediated by chromophoric dissolved organic matter (CDOM). Overall, the aquatic photodegradation rate of 6PPDQ will be strongly influenced by the compounding effects of environmental factors such as light screening and temperature on both direct and indirect photochemical processes. Transformation products were identified via UHPLC-Orbitrap mass spectrometry, revealing four major processes: (1) oxidation and cleavage of the quinone ring in the presence of ROS, (2) dealkylation, (3) rearrangement, and (4) deamination. These data indicate that 6PPDQ can photodegrade in cool, sunlit waters under the appropriate conditions: t1/2 = 17.4 h tono observable decrease (direct); t1/2 = 5.2-11.2 h (indirect, CDOM).

Monitoring vegetation restoration is challenging because monitoring is costly, requires long-term funding, and involves monitoring multiple vegetation variables that are often not linked back to learning about progress toward objectives. There is a clear need for the development of targeted monitoring programs that focus on a reduced set of variables that are tied to specific restoration objectives. In this paper, we present a method to progress the development of a targeted monitoring program, using a pre-existing state-and-transition model. We (1) use field data to validate an expert-derived classification of woodland vegetation states; (2) use these data to identify which variable(s) help differentiate woodland states; and (3) identify the target threshold (for the variable) that signifies if the desired transition has been achieved. The measured vegetation variables from each site in this study were good predictors of the different states. We show that by measuring only a few of these variables, it is possible to assign the vegetation state for a collection of sites, and monitor if and when a transition to another state has occurred. For this ecosystem and state-and-transition models, out of nine vegetation variables considered, the density of immature trees and percentage of exotic understory vegetation cover were the variables most frequently specified as effective to define a threshold or transition. We synthesize findings by presenting a decision tree that provides practical guidance for the development of targeted monitoring strategies for woodland vegetation.

Checkpoint inhibitors represent an effective treatment approach for a variety of cancers through their inhibition of immune regulatory pathways within the tumor microenvironment (TME). Unfortunately only a minority of patients with cancer achieve clinical benefit from immunotherapy, with the TME emerging as an important predictor of outcomes and sensitivity to therapy. The extent and pattern of T-cell infiltration can vary prominently within/across tumors and represents a biological continuum. Three immune profiles have been identified along this continuum: 'immune-desert' or 'T-cell cold' phenotype, 'immune-active', 'inflamed', or 'T-cell hot' phenotype, and 'immune excluded' phenotype. Of the three profiles, immune excluded remains the most ill-defined with no clear, universally accepted definition even though it is commonly associated with lack of response to immune checkpoint inhibitors and poor clinical outcomes. To address this, 16 multidisciplinary cancer experts from around the world were invited to participate in a symposium using a three-round modified Delphi approach. The first round was an open-ended questionnaire distributed via email and the second was an in-person discussion of the first round results that allowed for statements to be revised as necessary to achieve a maximum consensus (75% agreement) among the rating committee (RC). The final round questionnaire was distributed to the RC via email and had a 100% completion rate. The Delphi process resulted in moving us closer to a consensus definition for immune exclusion that is practical, clinically pertinent, and applicable across a wide range of cancer histologies. A general consensus of the role of immune exclusion in resistance to checkpoint therapy and five research priorities emerged from this process. Together, these tools could help efforts designed to address the underlying mechanisms of immune exclusion that span cancer types and, ultimately, aid in the development of treatments to target

Times of transition are laden with ambiguity, and the move from high school to college has an additional component of role changes to add to this uncertainty. In the spring and summer of 2020, this transition was disrupted by the changes brought on by the pandemic, affecting social norms, routines, and overall mental health outcomes. For professionals such as counselors and advisors to be able to provide specialized support, it is important to understand emerging adults' experiences at this time and to identify factors that helped them cope with this transition. Understanding the development of coping strategies has direct implications for both therapy and clinical practice which can work together to provide a higher quality of care for the people affected by the stress of major life transitions. In this study, I used a mixed-methods design to understand the experience of emerging adults who graduated high school during the pandemic, and the relation between tolerance for ambiguity and coping strategies, resilience, and psychological impact.

Purpose: Multiple sclerosis (MS) is a chronic inflammatory disease of the central nervous system that causes debilitating symptoms. Currently, there is insufficient evidence to recommend a special diet for people with MS to slow disease progression and reduce symptoms. Little is known about the dietary choices made by people with MS. This study aimed to explore the interpretations of healthy eating in people recently diagnosed with MS. Objectives were to investigate the types of changes in food choices and to describe the impact of making these changes. Design/methodology/approach: A social constructionist approach applying qualitative secondary analysis of semi-structured interviews was conducted (n = 11). Interviews were transcribed, coded and analysed using a deductive approach. Findings: Participants were mostly female (82%), mean age 47 years and mean time since diagnosis eight months. Four themes emerged from the data: (1) moving in the direction of the dietary guidelines, (2) modifying intake of dietary fat, (3) requiring mental effort and (4) needing input from a dietitian. Practical implications: The directions of food choices and the absence of dietetic input highlighted in this study suggest the need for evidence-based nutrition education that enables people with MS to tailor dietary guidelines according to their preferences. Originality/value: How people interpret healthy eating advice and the impact on making food choice changes is useful for explaining dietary changes in MS. Special diets promoted for MS provide conflicting advice, and the lack of access to dietitians means that additional mental effort is required when interpreting healthy eating messages and diets.

We present a study of the connection between the escape fraction of Lyman-alpha (Ly$\alpha$) and Lyman-continuum (LyC) photons within a sample of N=152 star-forming galaxies selected from the VANDELS survey at $3.85

We present a new determination of the evolving galaxy UV luminosity function (LF) over the redshift range $9.5250$ arcmin$^2$) data set of JWST NIRCam near-infrared imaging assembled from thirteen public JWST surveys. Our relatively large-area search allows us to uncover a sample of 61 robust $z>9.5$ candidates detected at $\geq 8\sigma$, and hence place new constraints on the intermediate-to-bright end of the UV LF. When combined with our previous JWST+UltraVISTA results, this allows us to measure the form of the LF over a luminosity range corresponding to four magnitudes ($M_{1500}$). At these early times we find that the galaxy UV LF is best described by a double power-law function, consistent with results obtained from recent ground-based and early JWST studies at similar redshifts. Our measurements provide further evidence for a relative lack of evolution at the bright-end of the UV LF at $z=9-11$, but do favour a steep faint-end slope ($\alpha\leq-2$). The luminosity-weighted integral of our evolving UV LF provides further evidence for a gradual, smooth (exponential) decline in co-moving star-formation rate density ($\rho_{\mathrm{SFR}}$) at least out to $z\simeq12$, with our determination of $\rho_{\mathrm{SFR}}(z=11)$ lying significantly above the predictions of many theoretical models of galaxy evolution., Comment: 20 pages, 9 figures, accepted for publication in MNRAS

We present a spectro-photometric study of a mass-complete sample of quiescent galaxies at $1.0 < z < 1.3$ with $\mathrm{log_{10}}(M_{\star}/\mathrm{M_{\odot}}) \geq 10.3$ drawn from the VANDELS survey, exploring the relationship between stellar mass, age and star-formation history. Within our sample of 114 galaxies, we derive a stellar-mass vs stellar-age relation with a slope of $1.20^{+0.28}_{-0.27}$ Gyr per decade in stellar mass. When combined with recent literature results, we find evidence that the slope of this relation remains consistent over the redshift interval $0

The extremely rapid assembly of the earliest galaxies during the first billion years of cosmic history is a major challenge for our understanding of galaxy formation physics. The advent of JWST has exacerbated this issue by confirming the existence of galaxies in significant numbers as early as the first few hundred million years. Perhaps even more surprisingly, in some galaxies, this initial highly efficient star formation rapidly shuts down, or quenches, giving rise to massive quiescent galaxies as little as 1.5 billion years after the Big Bang. However, due to their faintness and red colour, it has proven extremely challenging to learn about these extreme quiescent galaxies, or to confirm whether any exist at earlier times. Here we report the spectroscopic confirmation of a massive quiescent galaxy, GS-9209, at redshift $z=4.658$, just 1.25 billion years after the Big Bang, using JWST NIRSpec. From these data we infer a stellar mass of $M_* = 3.8\pm0.2\times10^{10}\ M_\odot$, which formed over a $\simeq200$ Myr period before this galaxy quenched its star formation activity at $z=6.5^{+0.2}_{-0.5}$, when the Universe was $\simeq800$ million years old. Based on the presence of broad H$\alpha$ in the spectrum and a high narrow-line [NII]/H$\alpha$ ratio, we infer the presence of an accreting supermassive black hole, with a mass of $M_\bullet = 5\pm1\times10^{8}\ M_\odot$. This large black hole mass relative to the stellar mass suggests that active galactic nucleus (AGN) feedback may have been responsible for quenching this galaxy. GS-9209 is also extremely compact, with an effective radius, $r_e=215\pm20$ parsecs. This galaxy is both a likely descendent of the highest-redshift submillimetre galaxies and quasars, and a likely progenitor for the dense, ancient cores of the most massive local galaxies., Comment: 23 pages, 6 figures, 2 tables, accepted for publication in Nature

Issue Addressed: Lifelong eating behaviours are established in childhood. Improving parents' food literacy skills is essential, as parents play a fundamental role in estab lishing their children's healthy eating behaviours and preferences for nutritious food. This paper describes the development and evaluation of an innovative program that combines food literacy with positive parent feeding practices, targeting parents in disadvantaged areas of Western Australia. Methods: The Food Sensations® for Parents five-week program was delivered to par ticipants from community-based parenting organisations during 2020 and 2021. For mative research and a pre-post evaluation design were adopted. Results: Pre- and post-evaluation data were collected from 224 participants (96% female). There was a statistically significant improvement in the mean score for 13 food literacy behaviours, 10 positive parenting feeding practices and a mean increase in parents' daily vegetable intake of 1/3 serve. Participants reported signifi cantly greater net improvements in food literacy behaviours than feeding practices, the largest being the Use a nutrition information panel to make food choices (33.1%). Multivariate logistic regression analyses found English as a first language, being older than 35, and from a higher Socio-Economic Index for Areas resulted in a higher likeli hood of positive changes in behaviours and practices. Conclusions: The findings indicate that the program is effective in improving the fre quency of use of food literacy behaviours, positive parenting feeding practices and increasing vegetable consumption. So What?: Analysing improvements in food literacy behaviours and feeding practices provides clarity on what change can be expected with a five-week parent program.

Many cancers hijack translation to increase the synthesis of tumor-driving proteins, the messenger mRNAs of which have specific codon usage patterns. Termed 'codon-biased translation' and originally identified in stress response regulation, this mechanism is supported by diverse studies demonstrating how the 50 RNA modifications of the epitranscriptome, specific tRNAs, and codon-biased mRNAs are used by oncogenic programs to promote proliferation and chemoresistance. The epitranscriptome writers METTL1-WDR4, Elongator complex protein (ELP)1-6, CTU1-2, and ALKBH8-TRM112 illustrate the principal mechanism of codon-biased translation, with gene amplifications, increased RNA modifications, and enhanced tRNA stability promoting cancer proliferation. Furthermore, systems-level analyses of 34 tRNA writers and 493 tRNA genes highlight the theme of tRNA epitranscriptome dysregulation in many cancers and identify candidate tRNA writers, tRNA modifications, and tRNA molecules as drivers of pathological codon-biased translation.

We reduce and analyse the available JWST ERO and ERS NIRCam imaging (SMACS0723, GLASS, CEERS) in combination with the latest deep ground-based near-infrared imaging in the COSMOS field (provided by UltraVISTA DR5) to produce a new measurement of the evolving galaxy UV luminosity function (LF) over the redshift range z = 8 - 15. This yields a new estimate of the evolution of UV luminosity density (rho(UV)), and hence cosmic star formation rate density (rho(SFR)) out to within = 12, one of which appears to set a new redshift record as an apparently robust galaxy candidate at z similar or equal to 16.4, the properties of which we therefore consider in detail. The advances presented here emphasize the importance of achieving high dynamic range in studies of early galaxy evolution, and re-affirm the enormous potential of forthcoming larger JWST programmes to transform our understanding of the young Universe.

We present a first-look analysis of the JWST ERO data in the SMACS J0723.3-7327 cluster field. We begin by reporting 10 new spectroscopic redshifts from lambda(obs) = 1.8-5.2 mu m NIRSpec medium-resolution (R = lambda/Delta lambda = 1000) data. These are determined via multiple high-SNR emission line detections with five objects at 1 < z < 3 displaying multiple rest-frame near-infrared Hydrogen Paschen lines, and five objects at 5 < z < 9 displaying rest-frame optical Oxygen and Hydrogen Balmer lines. For the five higher-redshift galaxies we extract fluxes in six NIRCam bands spanning lambda(obs) = 0.8-5 mu m and perform spectral energy distribution fitting in combination with existing HST photometry. The 7 < z < 9 objects exhibit a U-shaped pattern across the F277W, F356 W, and F444 W bands, indicating a Balmer break seen in emission (Balmer jump) and high-equivalent-width [O III] emission. This indicates an extremely young stellar population with the bulk of the current mass having formed within the past 10 Myr. We report robust stellar masses and mean stellar ages from our spectral fitting with the four z > 6 galaxies exhibiting low-stellar masses from log (10) (M-*/M-circle dot) = 7.1-8.2, and correspondingly young mean stellar ages of only a few Myr. This work highlights the critical importance of combining large upcoming NIRCam surveys with NIRSpec follow-up to measure the spectroscopic redshifts necessary to robustly constrain physical parameters.

In order to measure the bravery level of the crowd, a survey is given to analyze any correlations made between the data received. Embry-Riddle Aeronautical University students were given the opportunity to work in partnership with BRAVE Enterprises to analyze a dataset of over 900 participants. The goal of the BRAVE Research Project is to find a quantified value based on the participants answers that will provide BRAVE Enterprises with information that will aid in increasing bravery levels in people. This analysis would provide numerical data which depicts the similarities found in the participants bravery levels. Excel, Python, and Tableu will be used to make the analysis of the dataset.

Current next-generation RNA-sequencing (RNA-seq) methods do not provide accurate quantification of small RNAs within a sample, due to sequence-dependent biases in capture, ligation and amplification during library preparation. We present a method, absolute quantification RNA-sequencing (AQRNA-seq), that minimizes biases and provides a direct, linear correlation between sequencing read count and copy number for all small RNAs in a sample. Library preparation and data processing were optimized and validated using a 963-member microRNA reference library, oligonucleotide standards of varying length, and RNA blots. Application of AQRNA-seq to a panel of human cancer cells revealed >800 detectable miRNAs that varied during cancer progression, while application to bacterial transfer RNA pools, with the challenges of secondary structure and abundant modifications, revealed 80-fold variation in tRNA isoacceptor levels, stress-induced site-specific tRNA fragmentation, quantitative modification maps, and evidence for stress-induced, tRNA-driven, codon-biased translation. AQRNA-seq thus provides a versatile means to quantitatively map the small RNA landscape in cells.

Abstract Queuosine (Q) is a conserved hypermodification of the wobble base of tRNA containing GUN anticodons but the physiological consequences of Q deficiency are poorly understood in bacteria. This work combines transcriptomic, proteomic and physiological studies to characterize a Q-deficient Escherichia coli K12 MG1655 mutant. The absence of Q led to an increased resistance to nickel and cobalt, and to an increased sensitivity to cadmium, compared to the wild-type (WT) strain. Transcriptomic analysis of the WT and Q-deficient strains, grown in the presence and absence of nickel, revealed that the nickel transporter genes (nikABCDE) are downregulated in the Q– mutant, even when nickel is not added. This mutant is therefore primed to resist to high nickel levels. Downstream analysis of the transcriptomic data suggested that the absence of Q triggers an atypical oxidative stress response, confirmed by the detection of slightly elevated reactive oxygen species (ROS) levels in the mutant, increased sensitivity to hydrogen peroxide and paraquat, and a subtle growth phenotype in a strain prone to accumulation of ROS.

We present a first-look analysis of the JWST ERO data in the SMACS J0723.3-7327 cluster field. We begin by reporting 10 new spectroscopic redshifts from lambda(obs) = 1.8-5.2 mu m NIRSpec medium-resolution (R = lambda/Delta lambda = 1000) data. These are determined via multiple high-SNR emission line detections with five objects at 1 < z < 3 displaying multiple rest-frame near-infrared Hydrogen Paschen lines, and five objects at 5 < z < 9 displaying rest-frame optical Oxygen and Hydrogen Balmer lines. For the five higher-redshift galaxies we extract fluxes in six NIRCam bands spanning lambda(obs) = 0.8-5 mu m and perform spectral energy distribution fitting in combination with existing HST photometry. The 7 < z < 9 objects exhibit a U-shaped pattern across the F277W, F356 W, and F444 W bands, indicating a Balmer break seen in emission (Balmer jump) and high-equivalent-width [O III] emission. This indicates an extremely young stellar population with the bulk of the current mass having formed within the past 10 Myr. We report robust stellar masses and mean stellar ages from our spectral fitting with the four z > 6 galaxies exhibiting low-stellar masses from log (10) (M-*/M-circle dot) = 7.1-8.2, and correspondingly young mean stellar ages of only a few Myr. This work highlights the critical importance of combining large upcoming NIRCam surveys with NIRSpec follow-up to measure the spectroscopic redshifts necessary to robustly constrain physical parameters.

We present an analysis of the UV continuum slopes for a sample of $176$ galaxy candidates at $8 < z_{\mathrm{phot}} < 16$. Focusing primarily on a new sample of $125$ galaxies at $\langle z \rangle \simeq 11$ selected from $\simeq 320$ arcmin$^2$ of public JWST imaging data across $15$ independent datasets, we investigate the evolution of $\beta$ in the galaxy population at $z > 8$. In the redshift range $8 < z < 10$, we find evidence for a relationship between $\beta$ and $M_{\rm UV}$, such that galaxies with brighter UV luminosities display redder UV slopes, with $\rm{d}\beta/ \rm{d} M_{\rm UV} = -0.17 \pm 0.03$. A comparison with literature studies down to $z\simeq2$ suggests that a $\beta-M_{\rm UV}$ relation has been in place from at least $z\simeq10$, with a slope that does not evolve strongly with redshift, but with an evolving normalisation such that galaxies at higher redshifts become bluer at fixed $M_{\rm UV}$. We find a significant trend between $\beta$ and redshift, with the inverse-variance weighted mean value evolving from $\langle \beta \rangle = -2.17 \pm 0.05$ at $z = 9.5$ to $\langle \beta \rangle = -2.56 \pm 0.05$ at $z = 11.5$. Based on a comparison with stellar population models, we find that at $z>10.5$ the average UV continuum slope is consistent with the intrinsic blue limit of `dust-free' stellar populations $(\beta_{\mathrm{int}} \simeq -2.6)$. These results suggest that the moderately dust-reddened galaxy population at $z < 10$ was essentially dust free at $z \simeq 11$. The extremely blue galaxies being uncovered at $z>10$ place important constraints on the dust content of early galaxies, and imply that the already observed galaxy population is likely supplying an ionizing photon budget capable of maintaining ionized IGM fractions of $\gtrsim 5$ per cent at $z\simeq11$., Comment: 16 pages, 12 figures, submitted to MNRAS

This has been due mainly to the rise of effective machine learning and deep learning algorithms, together with increases in data collection and processing power, which have made rapid progress in many areas. However, use of this technology has brought with it philosophical issues and practical problems, in particular, epistemic and ethical. In this paper the authors, with backgrounds in philosophy, maternity care practice and clinical research, draw upon and extend a recent framework for shared decision-making (SDM) that identified a duty of care to the client's knowledge as a necessary condition for SDM. This duty entails the responsibility to acknowledge and overcome epistemic defeaters. This framework is applied to the use of AI in maternity care, in particular, the use of machine learning and deep learning technology to attempt to enhance electronic fetal monitoring (EFM). In doing so, various sub-kinds of epistemic defeater, namely, transparent, opaque, underdetermined, and inherited defeaters are taxonomized and discussed. The authors argue that, although effective current or future AI-enhanced EFM may impose an epistemic obligation on the part of clinicians to rely on such systems' predictions or diagnoses as input to SDM, such obligations may be overridden by inherited defeaters, caused by a form of algorithmic bias. The existence of inherited defeaters implies that the duty of care to the client's knowledge extends to any situation in which a clinician (or anyone else) is involved in producing training data for a system that will be used in SDM. Any future AI must be capable of assessing women individually, taking into account a wide range of factors including women's preferences, to provide a holistic range of evidence for clinical decision-making.

Rationale: Chronic obstructive pulmonary disease (COPD) is variable in its development. Lung microbiota and metabolites collectively may impact COPD pathophysiology, but relationships to clinical outcomes in milder disease are unclear. Objectives: Identify components of the lung microbiome and metabolome collectively associated with clinical markers in milder stage COPD. Methods: We analyzed paired microbiome and metabolomic data previously characterized from bronchoalveolar lavage fluid in 137 participants in the SPIROMICS (Subpopulations and Intermediate Outcome Measures in COPD Study), or (GOLD [Global Initiative for Chronic Obstructive Lung Disease Stage 0-2). Datasets used included 1) bacterial 16S rRNA gene sequencing; 2) untargeted metabolomics of the hydrophobic fraction, largely comprising lipids; and 3) targeted metabolomics for a panel of hydrophilic compounds previously implicated in mucoinflammation. We applied an integrative approach to select features and model 14 individual clinical variables representative of known associations with COPD trajectory (lung function, symptoms, and exacerbations). Measurements and Main Results: The majority of clinical measures associated with the lung microbiome and metabolome collectively in overall models (classification accuracies, >50%, P < 0.05 vs. chance). Lower lung function, COPD diagnosis, and greater symptoms associated positively with Streptococcus, Neisseria, and Veillonella, together with compounds from several classes (glycosphingolipids, glycerophospholipids, polyamines and xanthine, an adenosine metabolite). In contrast, several Prevotella members, together with adenosine, 5-methylthioadenosine, sialic acid, tyrosine, and glutathione, associated with better lung function, absence of COPD, or less symptoms. Significant correlations were observed between specific metabolites and bacteria (Padj < 0.05). Conclusions: Components of the lung microbiome and metabolome in combination relate to outcome

Queuosine (Q) is a conserved hypermodification of the wobble base of tRNA containing GUN anticodons but the physiological consequences of Q deficiency are poorly understood in bacteria. This work combines transcriptomic, proteomic and physiological studies to characterize a Q-deficient Escherichia coli K12 MG1655 mutant. The absence of Q led to an increased resistance to nickel and cobalt, and to an increased sensitivity to cadmium, compared to the wild-type (WT) strain. Transcriptomic analysis of the WT and Q-deficient strains, grown in the presence and absence of nickel, revealed that the nickel transporter genes (nikABCDE) are downregulated in the Q- mutant, even when nickel is not added. This mutant is therefore primed to resist to high nickel levels. Downstream analysis of the transcriptomic data suggested that the absence of Q triggers an atypical oxidative stress response, confirmed by the detection of slightly elevated reactive oxygen species (ROS) levels in the mutant, increased sensitivity to hydrogen peroxide and paraquat, and a subtle growth phenotype in a strain prone to accumulation of ROS.

We present a study designed to measure the average LyC escape fraction ($\langle f_{\rm esc}\rangle$) of star-forming galaxies at z=3.5. We assemble a sample of 148 galaxies from the VANDELS survey at $3.35\leq z_{\rm spec}\leq3.95$, selected to minimize line-of-sight contamination of their photometry. For this sample, we use ultra-deep, ground-based, $U-$band imaging and HST $V-$band imaging to robustly measure the distribution of $\mathcal{R_{\rm obs}}$ $=(L_{\rm LyC}/L_{\rm UV})_{\rm obs}$. We then model the distribution as a function of $\langle f_{\rm esc}\rangle$, carefully accounting for attenuation by dust, and the IGM (and CGM). A maximum likelihood fit to the $\mathcal{R_{\rm obs}}$ distribution returns a best-fitting value of $\langle f_{\rm esc}\rangle =0.07\pm0.02$, a result confirmed using an alternative Bayesian inference technique (both exclude $\langle f_{\rm esc}\rangle=0.0$ at $> 3\sigma$). By splitting our sample in two, we find evidence that $\langle f_{\rm esc}\rangle$ is positively correlated with Ly$\alpha$ equivalent width, with high and low sub-samples returning best fits of $\langle f_{\rm esc}\rangle=0.12^{+0.06}_{-0.04}$ and $\langle f_{\rm esc} \rangle=0.02^{+0.02}_{-0.01}$, respectively. In contrast, we find evidence that $\langle f_{\rm esc}\rangle$ is anti-correlated with intrinsic UV luminosity and UV dust attenuation; with low UV luminosity and dust attenuation sub-samples returning best fits in the range $0.10 \leq \langle f_{\rm esc}\rangle \leq 0.22$. We do not find evidence for a clear correlation between $f_{\rm esc}$ and galaxy stellar mass, suggesting it is not a primary indicator of leakage. Although larger samples are needed to further explore these trends, they suggest that it is entirely plausible that the low dust and metallicity galaxies found at z > 6 will display the $\langle f_{\rm esc}\rangle\geq0.1$ required to drive reionization., Comment: 15 pages (+ appendix), 12 figures, 1 table, Accepted for publication in Monthly Notices of the Royal Astronomical Society

The mammalian circadian clock exerts control of daily gene expression through cycles of DNA binding. Here, we develop a quantitative model of how a finite pool of BMAL1 protein can regulate thousands of target sites over daily time scales. We used quantitative imaging to track dynamic changes in endogenous labelled proteins across peripheral tissues and the SCN. We determine the contribution of multiple rhythmic processes coordinating BMAL1 DNA binding, including cycling molecular abundance, binding affinities, and repression. We find nuclear BMAL1 concentration determines corresponding CLOCK through heterodimerisation and define a DNA residence time of this complex. Repression of CLOCK:BMAL1 is achieved through rhythmic changes to BMAL1:CRY1 association and high-affinity interactions between PER2:CRY1 which mediates CLOCK:BMAL1 displacement from DNA. Finally, stochastic modelling reveals a dual role for PER:CRY complexes in which increasing concentrations of PER2:CRY1 promotes removal of BMAL1:CLOCK from genes consequently enhancing ability to move to new target sites.

Objective: To evaluate if a birthing room designed with person-centred considerations improves labour and birth outcomes for nulliparous women when compared to regular birthing rooms. Methods: A randomised controlled trial was conducted at a Swedish labour ward between January 2019 and October 2020. Nulliparous women in spontaneous labour were randomised either to a birthing room designed with person-centred considerations (New room) or a Regular room. The primary outcome was a composite of four variables: vaginal non-instrumental birth; no oxytocin augmentation; postpartum blood loss < 1000 ml; and a positive childbirth experience. To detect a difference of 8% between the groups, 1274 study participants were needed, but the trial was terminated early due to consequences of the Covid-19 pandemic. Results: A total of 406 women were randomised; 204 to the New room and 202 to the Regular room. There was no significant difference in the primary outcome between the groups (42.2% versus 35.1%; odds ratio: 1.35, 95% Confidence Interval 0.90-2.01; p = 0.18). Participants in the New room used epidural analgesia to a lower extent (54.4% versus 65.3%, relative risk: 0.83, 95% Confidence Interval 0.71-0.98; p = 0.03) and reported to a higher degree that the room contributed to a sense of safety, control, and integrity (p=< 0.001). Conclusions: The hypothesis that the New room would improve the primary outcome could not be verified. Considering the early discontinuation of the study, results should be interpreted with caution. Nevertheless, analyses of our secondary outcomes emphasise the experiential value of the built birth environment in improving care for labouring women.

PURPOSE: The international perinatal literature focuses on depression in the postpartum period. Prevalence and pathways of depression, anxiety and stress from pregnancy through the first postpartum year are seldom investigated. METHODS: MAMMI is a prospective cohort study of 3009 first-time mothers recruited in pregnancy. Depressive, anxiety and stress symptoms measured using the Depression, Anxiety and Stress Scale (DASS 21) in pregnancy and at 3-, 6-, 9- and/or 12-months postpartum. RESULTS: Prevalence of depressive and stress symptoms was lowest in pregnancy, increasing to 12-months postpartum. Anxiety symptoms remained relatively stable over time. In the first year after having their first baby, one in ten women reported moderate/severe anxiety symptoms (9.5%), 14.2% reported depression symptoms, and one in five stress symptoms (19.2%). Sociodemographic factors associated with increased odds of postpartum depression, anxiety and stress symptoms were younger age and being born in a non-EU country; socioeconomic factors were not living with a partner, not having postgraduate education and being unemployed during pregnancy. Retrospective reporting of poor mental health in the year prior to pregnancy and symptoms during pregnancy were strongly associated with poor postpartum mental health. CONCLUSIONS: The current findings suggest that the current model of 6-week postpartum care in Ireland is insufficient to detect and provide adequate support for women's mental health needs, with long-term implications for women and children.

The mechanical response of ceramic matrix composites depends critically on slip along the matrix-fiber interface, which is usually achieved with thin coatings on the fibers. Environmental attack of such coatings (enabled by ingress of reactants through matrix cracks) often leads to significant degradation, through removal of the coating via volatilization and oxidation of exposed SiC surfaces. The extent of the volatilization region extending from the matrix crack plane (i.e. recession length) is strongly coupled to the formation of oxide, which ultimately fills open gaps and arrests further reactions. This dissertation presents models to quantify these effects over a broad range of environmental conditions, coating thickness and matrix crack opening. Analytical solutions are presented for the time to close recession gaps via oxidation, and the associated terminal recession lengths obtained near free surfaces. A broad parameter study illustrates that recession behaviors are controlled by a competition between volatilization and oxidation rates. As such, the extent of recession is highly sensitive to water vapor and temperature, providing an explanation for disparate observations of recession under seemingly similar conditions. The extent of recession in the interior of composites is also illustrated, using a straightforward reaction-transport model. Recession lengths decay rapidly away from the free surface, with the extent of recession penetration scaling with maximum recession at the free surface.Models are also presented to quantify the impact of BN coating recession on SiC/SiC ceramic matrix composite strength as a function of environmental conditions and crack spacing. Under the assumption of uniform coating recession, an analytical expression for BN recession length is combined with composite fragmentation models to calculate retained strength. To detect a difference of 8% between the groups, 1274 study participants were needed, but the trial was terminated early due to consequences of the Covid-19 pandemic. Results: A total of 406 women were randomised; 204 to the New room and 202 to the Regular room. There was no significant difference in the primary outcome between the groups (42.2% versus 35.1%; odds ratio: 1.35, 95% Confidence Interval 0.90-2.01; p = 0.18). Participants in the New room used epidural analgesia to a lower extent (54.4% versus 65.3%, relative risk: 0.83, 95% Confidence Interval 0.71-0.98; p = 0.03) and reported to a higher degree that the room contributed to a sense of safety, control, and integrity (p=< 0.001). Conclusions: The hypothesis that the New room would improve the primary outcome could not be verified. Considering the early discontinuation of the study, results should be interpreted with caution. Nevertheless, analyses of our secondary outcomes emphasise the experiential value of the built birth environment in improving care for labouring women.

Objective: To evaluate if a birthing room designed with person-centred considerations improves labour and birth outcomes for nulliparous women when compared to regular birthing rooms. Nulliparous women in spontaneous labour were randomised either to a birthing room designed with person-centred considerations (New room) or a Regular room. For centuries, scientists have explored the limits of biological jump height1,2, and for decades, engineers have designed jumping machines3-18 that often mimicked or took inspiration from biological jumpers. Despite these efforts, general analyses are missing that compare the energetics of biological and engineered jumpers across scale. Here we show how biological and engineered jumpers have key differences in their jump energetics. The jump height of a biological jumper is limited by the work its linear motor (muscle) can produce in a single stroke. By contrast, the jump height of an engineered device can be far greater because its ratcheted or rotary motor can 'multiply work' during repeated strokes or rotations. As a consequence of these differences in energy production, biological and engineered jumpers should have divergent designs for maximizing jump height. Following these insights, we created a device that can jump over 30 metres high, to our knowledge far higher than previous engineered jumpers and over an order of magnitude higher than the best biological jumpers. Our work advances the understanding of jumping, shows a new level of performance, and underscores the importance of considering the differences between engineered and biological systems.
