Object counting is a challenging task with broad application prospects in security surveillance, traffic management, and disease diagnosis. Existing object counting methods face a tri-fold challenge: achieving superior performance, maintaining high generalizability, and minimizing annotation costs. We develop a novel training-free class-agnostic object counter, TFCounter, which is prompt-context-aware via the cascade of the essential elements in large-scale foundation models. This approach employs an iterative counting framework with a dual prompt system to recognize a broader spectrum of objects varying in shape, appearance, and size. Besides, it introduces an innovative context-aware similarity module incorporating background context to enhance accuracy within messy scenes. To demonstrate cross-domain generalizability, we collect a novel counting dataset named BIKE-1000, including exclusive 1000 images of shared bicycles from Meituan. The aim of this article is to identify potential diagnostic and prognostic biomarkers of sepsis to improve the survival of patients with sepsis.Methods: We downloaded 7 datasets from Gene Expression Omnibus database and screened the immune-related differential genes (IRDEGs). The related functions of IRDEGs were analyzed through Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG). CIBERSORT was used to evaluate the infiltration of the immune cells, and Pearson algorithm of R software was used to calculate the correlation between the immune cell content and gene expression to screen the genes most related to immune cells in sepsis group, which were intersected with IRDEGs to obtain common genes. Key genes were identified from common genes based on the area under the receiver operating characteristic curve (AUC) greater than 0.8 in the 6 datasets. We then analyzed the predictive value of key genes in sepsis survival. Finally, we verified the expression of key genes in patients with sepsis by PCR analysis.Results: A total of 164 IRDEGs were obtained, which were associated mainly with inflammatory and immunometabolic responses. Background and aims The structured, simplified modified Rankin scale questionnaire (smRSq) may increase reliability over the interrogative approach to scoring the modified Rankin scale (mRS) in acute stroke research and practice. During the conduct of the alteplase-dose arm of the international ENhanced Control of Hypertension ANd Thrombolysis StrokE stuDy (ENCHANTED), we had an opportunity to compare each of these approaches to outcome measurement. Methods Baseline demographic data were recorded together with the National Institutes of Health Stroke Scale (NIHSS). Follow-up measures obtained at 90 days included mRS, smRSq, and the 5-Dimension European Quality of life scale (EQ-5D). Agreements between smRSq and mRS were assessed with the Kappa statistic. Multiple logistic regression was used to identify baseline predictors of Day 90 smRSq and mRS scores. Treatment effects, based on Day 90 smRSq/mRS scores, were tested in logistic and ordinal logistic regression models. Results SmRSq and mRS scores had good agreement (weighted Kappa 0.79, 95% confidence interval (CI) 0.78–0.81), while variables of age, atrial fibrillation, diabetes mellitus, pre-morbid mRS (1 vs. 0), baseline NIHSS scores, and imaging signs of cerebral ischemia, similarly predicted their scores. Odds ratios for death or disability, and ordinal shift, 90-day mRS scores using smRSq were 1.05 (95% CI 0.91–1.20; one-sided P = 0.23 for non-inferiority) and 0.98 (95% CI 0.87–1.11; P = 0.02 for non-inferiority), similar to those using mRS. Conclusions This study demonstrates the utility of the smRSq in a large, ethnically diverse clinical trial population. Scoring of the smRSq shows adequate agreement with the standard mRS, thus confirming it is a reliable, valid, and useful alternative measure of functional status after acute ischemic stroke. Logistic regression models were used to determine associations of lacunar AIS with 90-day outcomes (primary, modified Rankin Scale [mRS] scores 2–6; secondary, other mRS scores, intracerebral hemorrhage [ICH], and early neurologic deterioration or death) and treatment effects of low- vs standard-dose alteplase across lacunar and nonlacunar AIS with adjustment for baseline covariables.ResultsOf 2,588 participants with available imaging and clinical data, we classified cases as definite/probable lacunar (n = 490) or nonlacunar AIS (n = 2,098) for primary analyses. Regardless of alteplase dose received, lacunar AIS participants had favorable functional (mRS 2–6, adjusted odds ratio [95% confidence interval] 0.60 [0.47–0.77]) and other clinical or safety outcomes compared to participants with nonlacunar AIS. In order to attract a broader audience and encourage the user group to buy artworks after visiting a gallery, this project aims to create a new gallery concept combining the physical and the virtual dimension. The main objective of this master thesis is to use virtual reality to enhance the current art experience and encourage their purchases, designing intuitive interactions between the viewer and the virtual art gallery. The project started with researching from three aspects, including human, technology and business, to explore the current situation of the galleries and customers, as well as the possible design opportunities. The result showed there were mainly two reasons why more galleries closed because of few customers in recent years. On the one hand, current galleries often couldn’t attract customers to view around for a long time. On the other hand, novice customers don’t have an easy and friendly way to purchase artworks. Therefore, it requires the design to find a proper way to help the small art businesses adopt a more modern way of selling artworks, and attract younger customers. The desired user experience focusing on three key elements (approachable, emotions, purchase) is determined after analyzing. During the conceptualization, three concepts focusing on three key elements were developed by conducting co-creation sessions. User evaluation was conducted using paper prototyping to test certain features and people’s likelihood of use. Six key elements that should be included in the final concept was concluded from the evaluation. By conducting the prototyping process, four steps of building a virtual gallery were found, and the process is able to be used as a guideline for researchers or designers who want to develop a virtual gallery. In order to find a relatively better interaction for certain functions, two gallery concepts were created to compare the interactions. Blueberries (Vaccinium spp.) are a favourite fruit and they are produced worldwide. In New Zealand, blueberries are the main export berry fruit and contribute greatly to export income. More than 2,800 tonnes of blueberries were produced in the 2017/2018 harvesting season and earned $34.8 million export income in 2018. Currently, 740 ha of the blueberry plantations can be found in both the South Island and North Island. Otautau is the main growing region in the South Island while Waihopo, the Waikato regions of Ngatea and Ohaupo, the Bay of Plenty and Hastings are the regions in the North Island, producing most of the fresh blueberries in New Zealand. However, blueberry leaf rust has become a prevalent disease in blueberry production and a concerning issue for blueberry growers. In Hastings production sites, serious infections have been found in recent years. Although fungicides were applied to control blueberry leaf rust, this form of control is incomplete and unsustainable for blueberry production. The deployment of varieties that are naturally resistant would be a better option for managing blueberry leaf rust disease. Currently, few cultivars are available for this purpose, but breeding for rust resistance can address this demand. The main issues preventing the production of resistant varieties are insufficient knowledge about this rust pathogen in New Zealand, and the lack of resistant germplasm sources and efficient resistance screening procedures. In order to attract a broader audience and encourage the user group to buy artworks after visiting a gallery, this project aims to create a new gallery concept combining the physical and the virtual dimension. The main objective of this master thesis is to use virtual reality to enhance the current art experience and encourage their purchases, designing intuitive interactions between the viewer and the virtual art gallery. The project started with researching from three aspects, including human, technology and business, to explore the current situation of the galleries and customers, as well as the possible design opportunities. The result showed there were mainly two reasons why more galleries closed because of few customers in recent years. On the one hand, current galleries often couldn’t attract customers to view around for a long time. On the other hand, novice customers don’t have an easy and friendly way to purchase artworks. Therefore, it requires the design to find a proper way to help the small art businesses adopt a more modern way of selling artworks, and attract younger customers. The desired user experience focusing on three key elements (approachable, emotions, purchase) is determined after analyzing. During the conceptualization, three concepts focusing on three key elements were developed by conducting co-creation sessions. User evaluation was conducted using paper prototyping to test certain features and people’s likelihood of use. Six key elements that should be included in the final concept was concluded from the evaluation. By conducting the prototyping process, four steps of building a virtual gallery were found, and the process is able to be used as a guideline for researchers or designers who want to develop a virtual gallery. In order to find a relatively better interaction for certain functions, two gallery concepts were created to compare the interactions. This study investigated the total costs caused by COPD and the influencing factors for the high economic burden in urban areas of China.Patients and methods: A cross-sectional study was carried out among 678 COPD patients in four cities in China in 2011. The average annual direct medical costs (DMCs), direct nonmedical costs (DNMCs), and indirect costs (ICs) on COPD were measured by median and mean (± standard deviation). Logistic regression model was used to explore factors related to high total costs on COPD.Results: The median annual DMCs, DNMCs, and ICs per COPD patient were RMB 5565 Yuan (US$ 862), 0 Yuan (US$ 0), and 0 Yuan (US$ 0), respectively, and the mean annual DMCs, DNMCs, and ICs per COPD patient were RMB 11968 (±22422) Yuan [US$ 1853 (±3472)], 539 (±2092) Yuan [US$ 83 (±324)], and 2087 (±8110) Yuan [US$ 323 (±1256)], respectively. The annual DMCs, DNMCs, and ICs for diagnosed COPD patients were RMB 195.70 billion Yuan (US$ 30.30 billion), 8.78 billion Yuan (US$ 1.36 billion), and 34.10 billion Yuan (US$ 5.28 billion), respectively, in China. Hospitalization accounted for 56.7% of the total costs. High economic burden was significantly related to age, acute exacerbations, and disease severity in COPD patients.Conclusion: COPD posed a heavy economic burden in China. Maize plants were grown in pots filled with a mixture of sandy and black soil for 5 weeks, and then half of the plants were exposed to low temperature for 1 week while the rest of the plants were grown under ambient temperature and severed as control. Different AMF resulted in different root colonization and low temperature significantly decreased AM colonization. Low temperature remarkably decreased plant height and total dry weight but increased root dry weight and root-shoot ratio. The AM plants had higher proline content compared with the non-AM plants. The maize plants inoculated with Glomus etunicatum and G. intraradices had higher malondialdehyde and soluble sugar contents under low temperature condition. The effect of four different arbuscular mycorrhizal fungi (AMF) on the growth and lipid peroxidation, soluble sugar, proline contents, and antioxidant enzymes activities of Zea mays L. was studied in pot culture subjected to two temperature regimes. Maize plants were grown in pots filled with a mixture of sandy and black soil for 5 weeks, and then half of the plants were exposed to low temperature for 1 week while the rest of the plants were grown under ambient temperature and severed as control. Different AMF resulted in different root colonization and low temperature significantly decreased AM colonization. Low temperature remarkably decreased plant height and total dry weight but increased root dry weight and root-shoot ratio. The AM plants had higher proline content compared with the non-AM plants. The maize plants inoculated with Glomus etunicatum and G. intraradices had higher malondialdehyde and soluble sugar contents under low temperature condition. The activities of catalase (CAT) and peroxidase of AM inoculated maize were higher than those of non-AM ones. Low temperature noticeably decreased the activities of CAT. The tumor suppressor p53 plays pivotal roles in maintaining the integrity of the genome and regulates cell cycle arrest and programmed cell death (apoptosis). Tumors without a functioning p53 are more likely to have a very aggressive clinical course. We proposed to use an RNA-based combinatorial approach to select for single-stranded RNA molecules that will bind to a mutant p53 protein, correct its conformation, and restore its sequence-specific DNA-binding activity. We have successfully accomplished the tasks we proposed for the first year of funding, including (1) expression and purification of human wild-type p53 protein in insect cells, (2) in vitro selection of RNA aptamers from a pool of RNA molecules that bind to p53 protein, and (3) sequence-specific DNA binding assays using the EMSA technique. Sequence and structural analysis of the selected RNAs revealed that the ability of RNA molecules to interfere with sequence-specific DNA binding of p53 may depend on their tertiary structures.
