1. Human in vivo assessment of the survival and germination of Heyndrickxia coagulans SNZ1969 spores delivered via gummy candies

Abstract: Confectionary products hold promise as unconventional food carriers for probiotic microorganisms. This study explored the delivery of Heyndrickxia coagulans SNZ1969, a spore-forming probiotic, using gummy candies. In this study, we prepared gummy candies containing bacterial spores with a viable count that remained stable during a 24-month shelf-life period, meeting the label claim of at least one billion CFUs per serving (24 g). Then, we carried out an intervention trial involving 24 healthy adults who consumed one serving per day for two weeks followed by an additional two weeks of follow-up. Fecal samples were collected and analyzed with a protocol that allowed the viable counts of SNZ1969, both in spore and vegetative forms. The obtained results revealed that bacterial spores germinated in all volunteers. SNZ1969 persistence in the gut was monitored for two weeks after the end of gummy candy consumption, indicating its potential for prolonged colonization. These findings highlight the potential of unconventional food carriers for probiotic delivery and suggest that spore-forming probiotics can be metabolically active in the human intestine. These findings provide information for the development of food products containing spore-forming probiotics and their potential benefits in promoting gastrointestinal health.

2. Collinsella aerofaciens as a predictive marker of response to probiotic treatment in non-constipated irritable bowel syndrome

Abstract: Probiotics are exploited for adjuvant treatment in IBS, but reliable guidance for selecting the appropriate probiotic to adopt for different forms of IBS is lacking. We aimed to identify markers for recognizing non-constipated (NC) IBS patients that may show significant clinical improvements upon treatment with the probiotic strain Lacticaseibacillus paracasei DG (LDG). To this purpose, we performed a post-hoc analysis of samples collected during a multicenter, double-blind, parallel-group, placebo-controlled trial in which NC-IBS patients were randomized to receive at least 24 billion CFU LDG or placebo capsules b.i.d. for 12 weeks. The primary clinical endpoint was the composite response based on improved abdominal pain and fecal type. The fecal microbiome and serum markers of intestinal (PV1 and zonulin), liver, and kidney functions were investigated. We found that responders (R) in the probiotic arm (25%) differed from non-responders (NR) based on the abundance of 18 bacterial taxa, including the families Coriobacteriaceae, Dorea spp. and Collinsella aerofaciens, which were overrepresented in R patients. These taxa also distinguished R (but not NR) patients from healthy controls. Probiotic intervention significantly reduced the abundance of these bacteria in R, but not in NR. Analogous results emerged for C. aerofaciens from the analysis of data from a previous trial on IBS with the same probiotic. Finally, C. aerofaciens was positively correlated with the plasmalemmal vesicle associated protein-1 (PV-1) and the markers of liver function. In conclusion, LDG is effective on NC-IBS patients with NC-IBS with a greater abundance of potential pathobionts. Among these, C. aerofaciens has emerged as a potential predictor of probiotic efficacy.

3. Lactic acid bacteria naturally associated with ready-to-eat rocket salad can survive the human gastrointestinal transit

Abstract: It was theorized that modernization and the decline in harmless microbial populations associated with food have altered the gut microbiota, impacting host metabolism and immunity. Western dietary patterns, characterized by processed foods and preservation methods, may significantly reduce the microbial population associated with food. To mitigate the consequences of bacterial deprivation, the integration of these diets with fermented foods is commonly proposed. Nonetheless, non-fermented food consumed raw may also be an important source of viable microbial cells for the human microbiome. This study investigates whether salad-associated LAB can survive the gastrointestinal transit (GIT) and contribute to the gut microbiota. LAB strains were quantified and isolated from rocket salad (Eruca vesicaria subsp. sativa), and their survival through GIT was assessed via intervention trials in healthy adults and in vitro. Moreover, bacterial communities in fecal samples were analyzed after three days of rocket salad consumption. Washing with a sodium hypochlorite solution drastically reduced total bacterial load and eliminated viable LAB. The quantity of LAB introduced through salads did not significantly alter the gut microbiota composition. Rocket salads harbored Weissella and Leuconostoc species. A significant increase in Weissella spp. but not in Leuconostoc spp. was observed after the consumption of rocket salad. Simulated GIT experiments suggested that the food matrix and the initial number of ingested viable bacteria may have been important in determining survival. These findings propose that plant products could serve as sources of live LAB for the human gut. Further research with diverse vegetables and longer interventions is needed, encouraging studies on raw, non-fermented foods and their impact on the human intestinal microbiome.

Abstract: The 15-year pulsar timing data set collected by the North American Nanohertz Observatory for Gravitational Waves (NANOGrav) shows positive evidence for the presence of a low-frequency gravitational-wave (GW) background. In this paper, we investigate potential cosmological interpretations of this signal, specifically cosmic inflation, scalar-induced GWs, first-order phase transitions, cosmic strings, and domain walls. We find that, with the exception of stable cosmic strings of field theory origin, all these models can reproduce the observed signal. When compared to the standard interpretation in terms of inspiraling supermassive black hole binaries (SMBHBs), many cosmological models seem to provide a better fit resulting in Bayes factors in the range from 10 to 100. However, these results strongly depend on modeling assumptions about the cosmic SMBHB population and, at this stage, should not be regarded as evidence for new physics. Furthermore, we identify excluded parameter regions where the predicted GW signal from cosmological sources significantly exceeds the NANOGrav signal. These parameter constraints are independent of the origin of the NANOGrav signal and illustrate how pulsar timing data provide a new way to constrain the parameter space of these models. Finally, we search for deterministic signals produced by models of ultralight dark matter (ULDM) and dark matter substructures in the Milky Way. We find no evidence for either of these signals and thus report updated constraints on these models. Abstract: Nearly a century after the beginning of the antibiotic era, which has been associated with unparalleled improvements in human health and reductions in mortality associated with infection, the dwindling pipeline for new antibiotic classes coupled with the inevitable spread of antimicrobial resistance (AMR) poses a major global challenge. Historically, surveillance of bacteria with AMR typically relied on phenotypic analysis of isolates taken from infected individuals, which provides only a low-resolution view of the epidemiology behind an individual infection or wider outbreak. Recent years have seen increasing adoption of powerful new genomic technologies with the potential to revolutionise AMR surveillance by providing a high-resolution picture of the AMR profile of the bacteria causing infections and providing real-time actionable information for treating and preventing infection. However, many barriers remain to be overcome before genomic technologies can be adopted as a standard part of routine AMR surveillance around the world. Accordingly, the Surveillance and Epidemiology of Drug-resistant Infections Consortium convened an expert working group to assess the benefits and challenges of using genomics for AMR surveillance. In this Series, we detail these discussions and provide recommendations from the working group that can help to realise the massive potential benefits for genomics in surveillance of AMR.

7. Ready-To-Eat Rocket Salads as Potential Reservoir of Bacteria for the Human Microbiome

Abstract: Western-type diets are composed of foods with a reduced amount of naturally occurring microorganisms. It was hypothesized that a microbe-depleted diet can favor the alteration of the human intestinal microbial ecosystem, therefore contributing to the onset of chronic metabolic and immune diseases currently recognized as the most significant causes of death in the developed world.Reportedly, Western-type diets may induce the loss of key microbial taxa within the gastrointestinal microbiota, promoting the onset of noncommunicable diseases. It was hypothesized that the consumption of raw vegetables could contribute to the maintenance of the intestinal microbial community structure. In this context, we explored bacteria associated with commercial rocket salads produced through different farming practices: traditional (conventional, organic, and integrated) and vertical farming. Viable counts of mesophilic bacteria and lactic acid bacteria (LAB) were performed on plate count agar (PCA) and de Man-Rogosa-Sharpe (MRS) agar at pH 5.7, whereas metataxonomics through 16S rRNA gene sequencing was used to profile total bacteria associated with rocket salads. We found that rocket salads from vertical farming had much fewer viable bacteria and had a bacterial community structure markedly different from that of rocket salads from traditional farming. Furthermore, although alpha- and beta-diversity analyses did not differentiate rocket samples according to farming techniques, several bacterial taxa distinguished organic and integrated from conventional farming salads, suggesting that farming practices could affect the taxonomic composition of rocket bacterial communities. LAB were isolated from only traditional farming samples and belonged to different species, which were variably distributed among samples and could be partly associated with farming practices. 8. Use of kefir-derived lactic acid bacteria for the preparation of a fermented soy drink with increased estrogenic activity

Abstract: Fermented foods are receiving growing attention for their health promoting properties. In particular, there is a growing demand for plant-based fermented foods as dairy alternatives. Considering that soy is a vegetal food rich in nutrients and a source of the phytoestrogen isoflavones, the aim of this study was to select safe food micro-organisms with the ability to ferment a soy drink resulting in a final product with an increased estrogenic activity and improved functional properties. We used milk kefir grains, a dairy source of microorganisms with proven health-promoting properties, as a starting inoculum for a soymilk. After 14 passages of daily inoculum in fresh soy drink, we isolated four lactic acid bacterial strains: Lactotoccus lactis subsp. lactis K03, Leuconostc pseudo-mesenteroides K05, Leuconostc mesenteroides K09 and Lentilactobacillus kefiri K10. Isolated strains were proven to be safe for human consumption according to the assessment of their antibiotic resistance profile and comparative genomics. Furthermore, functional characterization of the bacterial strains demonstrated their ability to ferment sugars naturally present in soybeans and produce a creamy texture. In addition, we demonstrated, by means of a yeast-based bioluminescence reporter system, that the two strains belonging to the genus Leuconostoc increased the estrogenic activity of the soybean drink. In conclusion, the proposed application of the bacterial strains characterized in this study meets the growing demand of consumers for health-promoting vegetal food alter-natives to dairy products.

Abstract: Recently we found compelling evidence for a gravitational wave background with Hellings and Downs (HD) correlations in our 15-year data set. These correlations describe gravitational waves as predicted by general relativity, which has two transverse polarization modes. However, more general metric theories of gravity can have additional polarization modes which produce different interpulsar correlations. In this work we search the NANOGrav 15-year data set for evidence of a gravitational wave background with quadrupolar Hellings and Downs (HD) and Scalar Transverse (ST) correlations. We find that HD correlations are the best fit to the data, and no significant evidence in favor of ST correlations. While Bayes factors show strong evidence for a correlated signal, the data does not strongly prefer either correlation signature, with Bayes factors $\sim 2$ when comparing HD to ST correlations, and $\sim 1$ for HD plus ST correlations to HD correlations alone. However, when modeled alongside HD correlations, the amplitude and spectral index posteriors for ST correlations are uninformative, with the HD process accounting for the vast majority of the total signal. Using the optimal statistic, a frequentist technique that focuses on the pulsar-pair cross-correlations, we find median signal-to-noise-ratios of 5.0 for HD and 4.6 for ST correlations when fit for separately, and median signal-to-noise-ratios of 3.5 for HD and 3.0 for ST correlations when fit for simultaneously. While the signal-to-noise-ratios for each of the correlations are comparable, the estimated amplitude and spectral index for HD are a significantly better fit to the total signal, in agreement with our Bayesian analysis., Comment: 11 pages, 5 figures

Abstract: The radio galaxy 3C 66B has been hypothesized to host a supermassive black hole binary (SMBHB) at its center based on electromagnetic observations. Its apparent 1.05-year period and low redshift ($\sim0.02$) make it an interesting testbed to search for low-frequency gravitational waves (GWs) using Pulsar Timing Array (PTA) experiments. This source has been subjected to multiple searches for continuous GWs from a circular SMBHB, resulting in progressively more stringent constraints on its GW amplitude and chirp mass. In this paper, we develop a pipeline for performing Bayesian targeted searches for eccentric SMBHBs in PTA data sets, and test its efficacy by applying it on simulated data sets with varying injected signal strengths. We also search for a realistic eccentric SMBHB source in 3C 66B using the NANOGrav 12.5-year data set employing PTA signal models containing Earth term-only as well as Earth+Pulsar term contributions using this pipeline. Due to limitations in our PTA signal model, we get meaningful results only when the initial eccentricity $e_0<0.5$ and the symmetric mass ratio $\eta>0.1$. We find no evidence for an eccentric SMBHB signal in our data, and therefore place 95% upper limits on the PTA signal amplitude of $88.1\pm3.7$ ns for the Earth term-only and $81.74\pm0.86$ ns for the Earth+Pulsar term searches for $e_0<0.5$ and $\eta>0.1$. Similar 95% upper limits on the chirp mass are $(1.98 \pm 0.05) \times 10^9\,M_{\odot}$ and $(1.81 \pm 0.01) \times 10^9\,M_{\odot}$. Abstract: Analysis of pulsar timing data have provided evidence for a stochastic gravitational wave background in the nHz frequency band. The most plausible source of such a background is the superposition of signals from millions of supermassive black hole binaries. The standard statistical techniques used to search for such a background and assess its significance make several simplifying assumptions, namely: i) Gaussianity; ii) isotropy; and most often iii) a power-law spectrum. However, a stochastic background from a finite collection of binaries does not exactly satisfy any of these assumptions. To understand the effect of these assumptions, we test standard analysis techniques on a large collection of realistic simulated datasets. The dataset length, observing schedule, and noise levels were chosen to emulate the NANOGrav 15-year dataset. Simulated signals from millions of binaries drawn from models based on the Illustris cosmological hydrodynamical simulation were added to the data. We find that the standard statistical methods perform remarkably well on these simulated datasets, despite their fundamental assumptions not being strictly met. They are able to achieve a confident detection of the background. However, even for a fixed set of astrophysical parameters, different realizations of the universe result in a large variance in the significance and recovered parameters of the background. 12. Modeling Current and Future High-Cadence Surveys of Repeating FRB Populations

Abstract: In recent years, the CHIME (Canadian Hydrogen Intensity Mapping Experiment) interferometer has revealed a large number of Fast Radio Bursts (FRBs), including a sizable population that demonstrates repeating behavior. This transit facility, employing a real-time FRB search pipeline, continually scans the sky with declinations between $-10^{\circ}$ and $90^{\circ}$ for events with fluences $\gtrapprox 0.4$ Jy ms. We simulate a population of repeating FRBs by performing Monte Carlo simulations of underlying source populations processed through a mock CHIME/FRB observing pipeline. Assuming intrinsic repeater rates follow a Poisson distribution, we test assumptions about the burst populations of the repeater sample, and construct models of the FRB sample assuming various cosmological distributions. We infer the completeness of CHIME/FRB observations as a function of observing cadence and redshifts out to 0.5. We find that, if all simulated bursts have a fixed Poisson probability of repetition over their integrated time of observation, repeating burst detections across comoving volume should continue to grow near linearly on the order of decades. We predict that around 170 of the current CHIME/FRB one-off sources will ultimately repeat. We also make projections for FRB repeaters by future facilities and demonstrate that the number of repeaters they find could saturate on a $\sim$3 yr timescale., Comment: 10 pages, 7 figures

Abstract: We present the results of a Bayesian search for gravitational wave (GW) memory in the NANOGrav 12.5-yr data set. We find no convincing evidence for any gravitational wave memory signals in this data set (Bayes factor = 2.8). As such, we go on to place upper limits on the strain amplitude of GW memory events as a function of sky location and event epoch. These upper limits are computed using a signal model that assumes the existence of a common, spatially uncorrelated red noise in addition to a GW memory signal. The median strain upper limit as a function of sky position is approximately $3.3 \times 10^{-14}$. We also find that there are some differences in the upper limits as a function of sky position centered around PSR J0613$-$0200. This suggests that this pulsar has some excess noise which can be confounded with GW memory. Finally, the upper limits as a function of burst epoch continue to improve at later epochs. Abstract: Noise characterization for pulsar-timing applications accounts for interstellar dispersion by assuming a known frequency-dependence of the delay it introduces in the times of arrival (TOAs). However, calculations of this delay suffer from mis-estimations due to other chromatic effects in the observations. The precision in modeling dispersion is dependent on the observed bandwidth. In this work, we calculate the offsets in infinite-frequency TOAs due to mis-estimations in the modeling of dispersion when using varying bandwidths at the Green Bank Telescope. We use a set of broadband observations of PSR J1643-1224, a pulsar with an excess of chromatic noise in its timing residuals. We artificially restricted these observations to a narrowband frequency range, then used both data sets to calculate residuals with a timing model that does not include short-scale dispersion variations. By fitting the resulting residuals to a dispersion model, and comparing the ensuing fitted parameters, we quantify the dispersion mis-estimations. Moreover, by calculating the autocovariance function of the parameters we obtained a characteristic timescale over which the dispersion mis-estimations are correlated. For PSR J1643-1224, which has one of the highest dispersion measures (DM) in the NANOGrav pulsar timing array, we find that the infinite-frequency TOAs suffer from a systematic offset of ~22 microseconds due to DM mis-estimations, with correlations over ~1 month. For lower-DM pulsars, the offset is ~7 microseconds. This error quantification can be used to provide more robust noise modeling in NANOGrav's data, thereby increasing sensitivity and improving parameter estimation in gravitational wave searches.

Abstract: This paper presents rigorous tests of pulsar timing array methods and software, examining their consistency across a wide range of injected parameters and signal strength. We discuss updates to the 15-year isotropic gravitational-wave background analyses and their corresponding code representations. Descriptions of the internal structure of the flagship algorithms \texttt{Enterprise} and \texttt{PTMCMCSampler} are given to facilitate understanding of the PTA likelihood structure, how models are built, and what methods are currently used in sampling the high-dimensional PTA parameter space. We introduce a novel version of the PTA likelihood that uses a two-step marginalization procedure that performs much faster when the white noise parameters remain fixed. We perform stringent tests of consistency and correctness of the Bayesian and frequentist analysis software. For the Bayesian analysis, we test prior recovery, injection recovery, and Bayes factors. For the frequentist analysis, we test that the cross-correlation-based optimal statistic, when modified to account for a non-negligible gravitational-wave background, accurately recovers the amplitude of the background. We also summarize recent advances and tests performed on the optimal statistic in the literature from both GWB detection and parameter estimation perspectives. Abstract: The 15-year pulsar timing data set collected by the North American Nanohertz Observatory for Gravitational Waves (NANOGrav) shows positive evidence for the presence of a low-frequency gravitational-wave (GW) background. In this paper, we investigate potential cosmological interpretations of this signal, specifically cosmic inflation, scalar-induced GWs, first-order phase transitions, cosmic strings, and domain walls. We find that, with the exception of stable cosmic strings of field theory origin, all these models can reproduce the observed signal. When compared to the standard interpretation in terms of inspiraling supermassive black hole binaries (SMBHBs), many cosmological models seem to provide a better fit resulting in Bayes factors in the range from 10 to 100. However, these results strongly depend on modeling assumptions about the cosmic SMBHB population and, at this stage, should not be regarded as evidence for new physics. Furthermore, we identify excluded parameter regions where the predicted GW signal from cosmological sources significantly exceeds the NANOGrav signal. These parameter constraints are independent of the origin of the NANOGrav signal and illustrate how pulsar timing data provide a new way to constrain the parameter space of these models. Finally, we search for deterministic signals produced by models of ultralight dark matter (ULDM) and dark matter substructures in the Milky Way. We find no evidence for either of these signals and thus report updated constraints on these models. Abstract: Pulsar timing arrays (PTAs) are galactic-scale gravitational wave detectors. Each individual arm, composed of a millisecond pulsar, a radio telescope, and a kiloparsecs-long path, differs in its properties but, in aggregate, can be used to extract low-frequency gravitational wave (GW) signals. We present a noise and sensitivity analysis to accompany the NANOGrav 15-year data release and associated papers, along with an in-depth introduction to PTA noise models. As a first step in our analysis, we characterize each individual pulsar data set with three types of white noise parameters and two red noise parameters. These parameters, along with the timing model and, particularly, a piecewise-constant model for the time-variable dispersion measure, determine the sensitivity curve over the low-frequency GW band we are searching. We tabulate information for all of the pulsars in this data release and present some representative sensitivity curves. We then combine the individual pulsar sensitivities using a signal-to-noise-ratio statistic to calculate the global sensitivity of the PTA to a stochastic background of GWs, obtaining a minimum noise characteristic strain of $7\times 10^{-15}$ at 5 nHz. A power law-integrated analysis shows rough agreement with the amplitudes recovered in NANOGrav's 15-year GW background analysis. Abstract: The North American Nanohertz Observatory for Gravitational Waves (NANOGrav) has reported evidence for the presence of an isotropic nanohertz gravitational wave background (GWB) in its 15 yr dataset. However, if the GWB is produced by a population of inspiraling supermassive black hole binary (SMBHB) systems, then the background is predicted to be anisotropic, depending on the distribution of these systems in the local Universe and the statistical properties of the SMBHB population. In this work, we search for anisotropy in the GWB using multiple methods and bases to describe the distribution of the GWB power on the sky. We do not find significant evidence of anisotropy, and place a Bayesian $95\%$ upper limit on the level of broadband anisotropy such that $(C_{l>0} / C_{l=0}) < 20\%$. We also derive conservative estimates on the anisotropy expected from a random distribution of SMBHB systems using astrophysical simulations conditioned on the isotropic GWB inferred in the 15-yr dataset, and show that this dataset has sufficient sensitivity to probe a large fraction of the predicted level of anisotropy. Abstract: We report multiple lines of evidence for a stochastic signal that is correlated among 67 pulsars from the 15-year pulsar-timing data set collected by the North American Nanohertz Observatory for Gravitational Waves. The correlations follow the Hellings-Downs pattern expected for a stochastic gravitational-wave background. The presence of such a gravitational-wave background with a power-law-spectrum is favored over a model with only independent pulsar noises with a Bayes factor in excess of $10^{14}$, and this same model is favored over an uncorrelated common power-law-spectrum model with Bayes factors of 200-1000, depending on spectral modeling choices. We have built a statistical background distribution for these latter Bayes factors using a method that removes inter-pulsar correlations from our data set, finding $p = 10^{-3}$ (approx. $3\sigma$) for the observed Bayes factors in the null no-correlation scenario. A frequentist test statistic built directly as a weighted sum of inter-pulsar correlations yields $p = 5 \times 10^{-5} - 1.9 \times 10^{-4}$ (approx. $3.5 - 4\sigma$). Assuming a fiducial $f^{-2/3}$ characteristic-strain spectrum, as appropriate for an ensemble of binary supermassive black-hole inspirals, the strain amplitude is $2.4^{+0.7}_{-0.6} \times 10^{-15}$ (median + 90% credible interval) at a reference frequency of 1/(1 yr). The inferred gravitational-wave background amplitude and spectrum are consistent with astrophysical expectations for a signal from a population of supermassive black-hole binaries, although more exotic cosmological and astrophysical sources cannot be excluded. Abstract: Evidence for a low-frequency stochastic gravitational wave background has recently been reported based on analyses of pulsar timing array data. The most likely source of such a background is a population of supermassive black hole binaries, the loudest of which may be individually detected in these datasets. Here we present the search for individual supermassive black hole binaries in the NANOGrav 15-year dataset. We introduce several new techniques, which enhance the efficiency and modeling accuracy of the analysis. The search uncovered weak evidence for two candidate signals, one with a gravitational-wave frequency of $\sim$4 nHz, and another at $\sim$170 nHz. The significance of the low-frequency candidate was greatly diminished when Hellings-Downs correlations were included in the background model. The high-frequency candidate was discounted due to the lack of a plausible host galaxy, the unlikely astrophysical prior odds of finding such a source, and since most of its support comes from a single pulsar with a commensurate binary period. Finding no compelling evidence for signals from individual binary systems, we place upper limits on the strain amplitude of gravitational waves emitted by such systems., Comment: 23 pages, 13 figures, 2 tables. Abstract: We present observations and timing analyses of 68 millisecond pulsars (MSPs) comprising the 15-year data set of the North American Nanohertz Observatory for Gravitational Waves (NANOGrav). NANOGrav is a pulsar timing array (PTA) experiment that is sensitive to low-frequency gravitational waves. This is NANOGrav's fifth public data release, including both "narrowband" and "wideband" time-of-arrival (TOA) measurements and corresponding pulsar timing models. We have added 21 MSPs and extended our timing baselines by three years, now spanning nearly 16 years for some of our sources. The data were collected using the Arecibo Observatory, the Green Bank Telescope, and the Very Large Array between frequencies of 327 MHz and 3 GHz, with most sources observed approximately monthly. A number of notable methodological and procedural changes were made compared to our previous data sets. These improve the overall quality of the TOA data set and are part of the transition to new pulsar timing and PTA analysis software packages. For the first time, our data products are accompanied by a full suite of software to reproduce data reduction, analysis, and results. Our timing models include a variety of newly detected astrometric and binary pulsar parameters, including several significant improvements to pulsar mass constraints. We find that the time series of 23 pulsars contain detectable levels of red noise, 10 of which are new measurements. 22. Constraints on Undetected Long-Period Binaries in the Known Pulsar Population

Abstract: Although neutron star-black hole binaries have been identified through mergers detected in gravitational waves, a pulsar-black hole binary has yet to be detected. While short-period binaries are detectable due to a clear signal in the pulsar's timing residuals, effects from a long-period binary could be masked by other timing effects, allowing them to go undetected. In particular, a long-period binary measured over a small subset of its orbital period could manifest via time derivatives of the spin-frequency incompatible with isolated pulsar properties. We assess the possibility of pulsars having unknown companions in long-period binaries and put constraints on the range of binary properties that may remain undetected in current data, but that may be detectable with further observations. We find that for 35% of canonical pulsars with published higher order derivatives, the precision of measurements is not enough to confidently reject binarity (period greater than ~2 kyr), and that a black-hole binary companion could not be ruled out for a sample of pulsars without published constraints if the period is greater than 1 kyr. While we find no convincing cases in the literature, we put more stringent limits on orbital period and longitude of periastron for the few pulsars with published higher-order frequency derivatives (n greater than 3). Abstract: Pulsar timing array collaborations, such as the North American Nanohertz Observatory for Gravitational Waves (NANOGrav), are seeking to detect nanohertz gravitational waves emitted by supermassive black hole binaries formed in the aftermath of galaxy mergers. We have searched for continuous waves from individual circular supermassive black hole binaries using the NANOGrav's recent 12.5-year data set. We created new methods to accurately model the uncertainties on pulsar distances in our analysis, and we implemented new techniques to account for a common red noise process in pulsar timing array data sets while searching for deterministic gravitational wave signals, including continuous waves. As we found no evidence for continuous waves in our data, we placed 95\% upper limits on the strain amplitude of continuous waves emitted by these sources. At our most sensitive frequency of 7.65 nanohertz, we placed a sky-averaged limit of $h_0 < $ $(6.82 \pm 0.35) \times 10^{-15}$, and $h_0 <$ $(2.66 \pm 0.15) \times 10^{-15}$ in our most sensitive sky location. 24. Towards modelling of corrugation ridges at ice-sheet grounding lines

Abstract: Improvements in the resolution of sea-floor mapping techniques have revealed extremely regular, sub-metre-scale ridge landforms produced by the tidal flexure of ice-shelf grounding lines as they retreated very rapidly (i.e. at rates of several kilometres per year). Guided by such novel sea-floor observations from Thwaites Glacier, West Antarctica, we present three mathematical models for the formation of these corrugation ridges at a tidally migrating grounding line (that is retreating at a constant rate), where each ridge is formed by either constant till flux to the grounding line, till extrusion from the grounding line, or the resuspension and transport of grains from the grounding-zone bed. We find that both till extrusion (squeezing out till like toothpaste as the ice sheet re-settles on the sea floor) and resuspension and transport of material can qualitatively reproduce regular, delicate ridges at a retreating grounding line, as described by sea-floor observations. By considering the known properties of subglacial sediments, we agree with existing schematic models that the most likely mechanism for ridge formation is till extrusion at each low-tide position, essentially preserving an imprint of the ice-sheet grounding line as it retreated. However, when realistic (shallow) bed slopes are used in the simulations, ridges start to overprint one another, suggesting that, to preserve the regular ridges that have been observed, grounding line retreat rates (driven by dynamic thinning?) may be even higher than previously thought.

Abstract: X-ray diffraction (XRD) is an essential technique to determine a material's crystal structure in high-throughput experimentation, and has recently been incorporated in artificially intelligent agents in autonomous scientific discovery processes. However, rapid, automated and reliable analysis method of XRD data matching the incoming data rate remains a major challenge. To address these issues, we present CrystalShift, an efficient algorithm for probabilistic XRD phase labeling that employs symmetry-constrained pseudo-refinement optimization, best-first tree search, and Bayesian model comparison to estimate probabilities for phase combinations without requiring phase space information or training. We demonstrate that CrystalShift provides robust probability estimates, outperforming existing methods on synthetic and experimental datasets, and can be readily integrated into high-throughput experimental workflows. In addition to efficient phase-mapping, CrystalShift offers quantitative insights into materials' structural parameters, which facilitate both expert evaluation and AI-based modeling of the phase space, ultimately accelerating materials identification and discovery., Comment: 13 pages, 6 figures

Abstract: We report on the latest results of a Parkes multibeam survey for pulsars and dispersed radio bursts in the Large Magellanic Cloud (LMC). We conducted both periodicity and single-pulse searches at a much larger range of trial dispersion measures (DMs) than previously searched. We detected 229 single pulses with signal-to-noise ratio ($\rm{S/N}) > 7$ that were classified by the deep learning network FETCH as being real (with $> 90$% likelihood), of which 9 were from the known giant-pulse-emitting pulsar PSR B0540$-$69. Two possibly repeating sources were detected with DMs suggesting that they lie within the LMC, but these require confirmation. Only 3 of the 220 unknown pulses had S/N greater than 8, and the DM distribution for these detected pulses follows an exponential falloff with increasing DM and does not show any excess of signals at DM values expected for the LMC. These features suggest that the detected pulses are not likely to be real, although they are visually compelling. We also report the discovery of a new pulsar (PSR J0556$-$67) in our periodicity search. This pulsar has a spin period of 791 ms, a DM of 71 cm$^{-3}$ pc, an estimated 1400 MHz flux density of $\sim 0.12$ mJy, and shows no evidence of binary motion. Future observations may be able to confirm whether any of the weak but promising candidates in our single pulse and periodicity searches, including our two possible repeaters, are real or not., Comment: 17 pages, 11 figures

27. On the radio spectra of Galactic millisecond pulsars

Abstract: With recent advances in the sensitivity of radio surveys of the Galactic disk, the number of millisecond pulsars (MSPs) has increased substantially in recent years such that it is now possible to study their demographic properties in more detail than in the past. We investigate what can be learned about the radio spectra of the MSP population. Using a sample of 179 MSPs detected in eleven surveys carried out at radio frequencies in the range 0.135-6.6 GHz, we carry out detailed modeling of MSP radio spectral behaviour in this range. Employing Markov Chain Monte Carlo simulations to explore a multi-dimensional parameter space, and accurately accounting for observational selection effects, we find strong evidence in favour of the MSP population having a two-component power-law spectral model scaling with frequency, $\nu$. Specifically, we find that MSP flux density spectra are approximately independent of frequency below 320 MHz, and proportional to $\nu^{-1.5}$ at higher frequencies. This parameterization performs significantly better than single power-law models which over predict the number of MSPs seen in low-frequency (100-200 MHz) surveys. We compared our results with earlier work, and current understanding of the normal pulsar population, and use our model to make predictions for MSP yields in upcoming surveys. Abstract: The millisecond pulsar J1713+0747 underwent a sudden and significant pulse shape change between April 16 and 17, 2021 (MJDs 59320 and 59321). Subsequently, the pulse shape gradually recovered over the course of several months. We report the results of continued multi-frequency radio observations of the pulsar made using the Canadian Hydrogen Intensity Mapping Experiment (CHIME) and the 100-meter Green Bank Telescope (GBT) in a three-year period encompassing the shape change event, between February 2020 and February 2023. As of February 2023, the pulse shape had returned to a state similar to that seen before the event, but with measurable changes remaining. The amplitude of the shape change and the accompanying TOA residuals display a strong non-monotonic dependence on radio frequency, demonstrating that the event is neither a glitch (the effects of which should be independent of radio frequency, $\nu$) nor a change in dispersion measure (DM) alone (which would produce a delay proportional to $\nu^{-2}$). However, it does bear some resemblance to the two previous "chromatic timing events" observed in J1713+0747 (Demorest et al. 2013; Lam et al. 2016), as well as to a similar event observed in PSR J1643-1224 in 2015 (Shannon et al. 2016)., Comment: 18 pages, 8 figures. 31. Millisecond Cadence Radio Frequency Interference Filters

Abstract: Radio Frequency Interference (RFI) greatly reduces sensitivity of radio observations to astrophysical signals and creates false positive candidates in searches for radio transients. Real signals are missed while considerable computational and human resources are needed to remove RFI candidates. In the context of transient astrophysics, this makes effective RFI removal vital to effective searches for fast radio bursts and pulsars. Radio telescopes typically sample at rates that are high enough for there to be tens to hundreds of samples along the transient's pulse. Mitigation techniques should excise RFI on this timescale to account for a changing radio frequency environment. We evaluate the effectiveness of three filters, as well as a composite of the three, that excises RFI at the cadence that the data are recorded. Each of these filters operates in a different domain and thus excises as a different RFI morphology. We analyze the performance of these four filters in three different situations: (I) synthetic pulses in Gaussian noise; (II) synthetic pulses injected into real data; (III) four pulsar observations. From these tests, we gain insight into how the filters affect both the pulse and the noise level. This allows use to outline which and how the filters should be used based on the RFI present and the characteristics of the source signal. Abstract: We report the discovery of a highly circularly polarized, variable, steep-spectrum pulsar in the Australian Square Kilometre Array Pathfinder (ASKAP) Variables and Slow Transients (VAST) survey. The pulsar is located about $1^\circ$ from the center of the Large Magellanic Cloud, and has a significant fractional circular polarization of $\sim$20%. We discovered pulsations with a period of 322.5 ms, dispersion measure (DM) of 157.5 pc cm$^{-3}$, and rotation measure (RM) of $+456$ rad m$^{-2}$ using observations from the MeerKAT and the Parkes telescopes. This DM firmly places the source, PSR J0523$-$7125, in the Large Magellanic Cloud (LMC). This RM is extreme compared to other pulsars in the LMC (more than twice that of the largest previously reported one). The average flux density of $\sim$1 mJy at 1400 MHz and $\sim$25 mJy at 400 MHz places it among the most luminous radio pulsars known. It likely evaded previous discovery because of its very steep radio spectrum (spectral index $\alpha \approx -3$, where $S_\nu \propto \nu^\alpha$) and broad pulse profile (duty cycle $\gtrsim35$%). We discuss implications for searches for unusual radio sources in continuum images, as well as extragalactic pulsars in the Magellanic Clouds and beyond. Our result highlighted the possibility of identifying pulsars, especially extreme pulsars, from radio continuum images. 34. On the radio spectra of Galactic millisecond pulsars

Abstract: With recent advances in the sensitivity of radio surveys of the Galactic disk, the number of millisecond pulsars (MSPs) has increased substantially in recent years such that it is now possible to study their demographic properties in more detail than in the past. We investigate what can be learned about the radio spectra of the MSP population. Using a sample of 179 MSPs detected in eleven surveys carried out at radio frequencies in the range 0.135-6.6 GHz, we carry out detailed modeling of MSP radio spectral behaviour in this range. Employing Markov Chain Monte Carlo simulations to explore a multi-dimensional parameter space, and accurately accounting for observational selection effects, we find strong evidence in favour of the MSP population having a two-component power-law spectral model scaling with frequency, $\nu$. Specifically, we find that MSP flux density spectra are approximately independent of frequency below 320 MHz, and proportional to $\nu^{-1.5}$ at higher frequencies. This parameterization performs significantly better than single power-law models which over predict the number of MSPs seen in low-frequency (100-200 MHz) surveys. We compared our results with earlier work, and current understanding of the normal pulsar population, and use our model to make predictions for MSP yields in upcoming surveys. Abstract: We report on the latest results of a Parkes multibeam survey for pulsars and dispersed radio bursts in the Large Magellanic Cloud (LMC). We conducted both periodicity and single-pulse searches at a much larger range of trial dispersion measures (DMs) than previously searched. We detected 229 single pulses with signal-to-noise ratio ($\rm{S/N}) > 7$ that were classified by the deep learning network FETCH as being real (with $> 90$% likelihood), of which 9 were from the known giant-pulse-emitting pulsar PSR B0540$-$69. Two possibly repeating sources were detected with DMs suggesting that they lie within the LMC, but these require confirmation. Only 3 of the 220 unknown pulses had S/N greater than 8, and the DM distribution for these detected pulses follows an exponential falloff with increasing DM and does not show any excess of signals at DM values expected for the LMC. These features suggest that the detected pulses are not likely to be real, although they are visually compelling. We also report the discovery of a new pulsar (PSR J0556$-$67) in our periodicity search. This pulsar has a spin period of 791 ms, a DM of 71 cm$^{-3}$ pc, an estimated 1400 MHz flux density of $\sim 0.12$ mJy, and shows no evidence of binary motion. Future observations may be able to confirm whether any of the weak but promising candidates in our single pulse and periodicity searches, including our two possible repeaters, are real or not., Comment: 17 pages, 11 figures

Abstract: Using Bayesian analyses we study the solar electron density with the NANOGrav 11-year pulsar timing array (PTA) dataset. Our model of the solar wind is incorporated into a global fit starting from pulse times-of-arrival. We introduce new tools developed for this global fit, including analytic expressions for solar electron column densities and open source models for the solar wind that port into existing PTA software. We perform an ab initio recovery of various solar wind model parameters. We then demonstrate the richness of information about the solar electron density, $n_E$, that can be gleaned from PTA data, including higher order corrections to the simple $1/r^2$ model associated with a free-streaming wind (which are informative probes of coronal acceleration physics), quarterly binned measurements of $n_E$ and a continuous time-varying model for $n_E$ spanning approximately one solar cycle period. Abstract: We search NANOGrav's 12.5-year data set for evidence of a gravitational wave background (GWB) with all the spatial correlations allowed by general metric theories of gravity. We find no substantial evidence in favor of the existence of such correlations in our data. We find that scalar-transverse (ST) correlations yield signal-to-noise ratios and Bayes factors that are higher than quadrupolar (tensor transverse, TT) correlations. Specifically, we find ST correlations with a signal-to-noise ratio of 2.8 that are preferred over TT correlations (Hellings and Downs correlations) with Bayesian odds of about 20:1. However, the significance of ST correlations is reduced dramatically when we include modeling of the Solar System ephemeris systematics and/or remove pulsar J0030$+$0451 entirely from consideration. Even taking the nominal signal-to-noise ratios at face value, analyses of simulated data sets show that such values are not extremely unlikely to be observed in cases where only the usual TT modes are present in the GWB. In the absence of a detection of any polarization mode of gravity, we place upper limits on their amplitudes for a spectral index of $\gamma = 5$ and a reference frequency of $f_\text{yr} = 1 \text{yr}^{-1}$. Among the upper limits for eight general families of metric theories of gravity, we find the values of $A^{95\%}_{TT} = (9.7 \pm 0.4)\times 10^{-16}$ and $A^{95\%}_{ST} = (1.4 \pm 0.03)\times 10^{-15}$ for the family of metric spacetime theories that contain both TT and ST modes., Comment: 24 pages, 18 figures, 3 appendices. 38. An Eclipsing Black Widow Pulsar in NGC 6712

Abstract: We report the discovery of the first radio pulsar associated with NGC 6712, an eclipsing black widow (BW) pulsar, J1853$-$0842A, found by high-sensitivity searches using the Five-hundred-meter Aperture Spherical radio Telescope. This 2.15 ms pulsar is in a 3.56 hr compact circular orbit with a very low mass companion likely of mass 0.018 to 0.036 $M_{\rm \odot}$ and exhibits eclipsing of the pulsar signal. Though the distance to PSR J1853$-$0842A predicted from its dispersion measure ($155.125 \pm 0.004$ cm$^{-3}$ pc) and Galactic free electron density models are about 30\% smaller than that of NGC 6712 obtained from interstellar reddening measurements, this is likely due to limited knowledge about the spiral arms and Scutum stellar cloud in this direction. Follow-up timing observations spanning 445 days allow us to localize the pulsar's position to be 0.14 core radii from the center of NGC 6712 and measure a negative spin-down rate for this pulsar of $-2.39(2)\times10^{-21}\rm s s^{-1}$. The latter cannot be explained without the acceleration of the GC and decisively supports the association between PSR J1853--0842A and NGC 6712. Considering the maximum GC acceleration, Galactic acceleration, and Shklovskii effect, we place an upper limit on the intrinsic spin-down rate to be $1.11\times10^{-20}\rm~s~s^{-1}$. From an analysis of the eclipsing observations, we estimate the electron density of the eclipse region to be about $1.88\times10^6\rm cm^{-3}$. 39. What it Takes to Measure Reionization with Fast Radio Bursts

Abstract: Fast Radio Bursts (FRBs) are recently-discovered extra-galactic radio transients which exhibit a distance-dependent dispersion of their signal, and thus can be used as cosmological probes. In this article we, for the first time, apply a model-independent approach to measure reionization from synthetic FRB data assuming these signals are detected beyond redshift 5. This method allows us to constrain the full shape of the reionization history as well as the CMB optical depth $\tau$ while avoiding the pitfalls of commonly used model-based techniques. Already 100 localized FRBs, originating from redshifts z>5, could constrain (at 68% confidence level) the CMB optical depth to within 11%, and the midpoint of reionization to 4%, surpassing current state-of-the-art CMB bounds and quasar limits. Owing to the higher numbers of expected FRBs at lower redshifts, the $\tau$ constraints are asymmetric (+14%, -7%) providing a much stronger lower limit. 40. Comprehensive analysis of a dense sample of FRB 121102 bursts

Abstract: We present an analysis of a densely repeating sample of bursts from the first repeating fast radio burst, FRB 121102. We reanalysed the data used by Gourdji et al. (2019) and detected 93 additional bursts using our single-pulse search pipeline. In total, we detected 133 bursts in three hours of data at a center frequency of 1.4 GHz using the Arecibo telescope, and develop robust modeling strategies to constrain the spectro-temporal properties of all the bursts in the sample. Most of the burst profiles show a scattering tail, and burst spectra are well modeled by a Gaussian with a median width of 230 MHz. We find a lack of emission below 1300 MHz, consistent with previous studies of FRB 121102. We also find that the peak of the log-normal distribution of wait times decreases from 207 s to 75 s using our larger sample of bursts, as compared to that of Gourdji et al. (2019). Our observations do not favor either Poissonian or Weibull distributions for the burst rate distribution. We searched for periodicity in the bursts using multiple techniques but did not detect any significant period. The cumulative burst energy distribution exhibits a broken power-law shape, with the lower and higher-energy slopes of $-0.4\pm0.1$ and $-1.8\pm0.2$, with the break at $(2.3\pm0.2)\times 10^{37}$ ergs. We provide our burst fitting routines as a python package BURSTFIT that can be used to model the spectrogram of any complex FRB or pulsar pulse using robust fitting techniques. Abstract: We search for a first-order phase transition gravitational wave signal in 45 pulsars from the NANOGrav 12.5 year dataset. We find that the data can be modeled in terms of a strong first order phase transition taking place at temperatures below the electroweak scale. However, we do not observe any strong preference for a phase-transition interpretation of the signal over the standard astrophysical interpretation in terms of supermassive black holes mergers; but we expect to gain additional discriminating power with future datasets, improving the signal to noise ratio and extending the sensitivity window to lower frequencies. Abstract: Background Vascular services is changing rapidly, having emerged as a new specialty with its own training and specialised techniques. This has resulted in the need for reconfiguration of services to provide adequate specialist provision and accessible and equitable services. Objectives To identify the effects of service configuration on practice, resource use and outcomes. To model potential changes in configuration. To identify and/or develop electronic data collection tools for collecting patient-reported outcome measures and other clinical information. To evaluate patient preferences for aspects of services other than health-related quality of life. Design This was a multiple methods study comprising multiple systematic literature reviews; the development of a new outcome measure for users of vascular services (the electronic Personal Assessment Questionnaire – Vascular) based on the reviews, qualitative studies and psychometric evaluation; a trade-off exercise to measure process utilities; Hospital Episode Statistics analysis; and the development of individual disease models and a metamodel of service configuration. Setting Specialist vascular inpatient services in England. Data sources Modelling and Hospital Episode Statistics analysis for all vascular inpatients in England from 2006 to 2018. Qualitative studies and electronic Personal Assessment Questionnaire – Vascular evaluation with vascular patients from the Sheffield area. Abstract: We use femtosecond extreme ultraviolet pulses derived from a free electron laser to excite and probe surface acoustic waves (SAWs) on the (001) surface of single crystal SrTiO3. SAWs are generated by a pair of 39.9 nm pulses crossed at the sample with the crossing angle defining the SAW wavelength at 84 nm. Detection of SAWs is performed via diffraction of a time-delayed 13.3 nm probe pulse by SAW-induced surface ripples. Despite the low reflectivity of the sample in the extreme ultraviolet range, the reflection mode detection is found to be efficient because of an increase in the diffraction efficiency for shorter wavelengths. We describe a methodology for extracting the SAW attenuation in the presence of a thermal grating, which is based on measuring the decay of oscillations at twice the SAW frequency. The proposed approach can be used to study ultrahigh frequency SAWs in a broad range of materials and will bridge the wave vector gap in surface phonon spectroscopy between Brillouin scattering and He atom scattering.

47. Unequal but essential:How subsistence consumer–entrepreneurs negotiate unprecedented shock with extraordinary resilience during COVID-19

Abstract: We use qualitative interviews to study subsistence consumers confronting the global, pervasive and extended challenges of COVID-19, encompassing literally all realms of daily life. For subsistence consumers whose circumstances are filled with day-to-day uncertainty and a small margin of error to begin with, the pandemic has led to manifold uncertainties and a disappearing margin of error, with potentially lethal consequences. Their constraints to thinking and lack of self-confidence arising from both low income and low literacy are magnified in the face of the complex, invisible pandemic and the fear and panic it has caused. Characteristic relational strengths are weakened with social distancing and fear of infection. Yet, subsistence consumers display humanity in catastrophe, and confront the uncontrollable by reiterating a higher power. Consumption is reduced to the very bare essentials and income generation involves staying the course versus finding any viable alternative. We derive implications for consumer affairs. Abstract: We search for a first-order phase transition gravitational wave signal in 45 pulsars from the NANOGrav 12.5 year dataset. We find that the data can be modeled in terms of a strong first order phase transition taking place at temperatures below the electroweak scale. However, we do not observe any strong preference for a phase-transition interpretation of the signal over the standard astrophysical interpretation in terms of supermassive black holes mergers; but we expect to gain additional discriminating power with future datasets, improving the signal to noise ratio and extending the sensitivity window to lower frequencies. Abstract: As SARS-CoV-2 continues to spread and evolve, detecting emerging variants early is critical for public health interventions. Inferring lineage prevalence by clinical testing is infeasible at scale, especially in areas with limited resources, participation, or testing/sequencing capacity, which can also introduce biases. SARS-CoV-2 RNA concentration in wastewater successfully tracks regional infection dynamics and provides less biased abundance estimates than clinical testing. Tracking virus genomic sequences in wastewater would improve community prevalence estimates and detect emerging variants. However, two factors limit wastewater-based genomic surveillance: low-quality sequence data and inability to estimate relative lineage abundance in mixed samples. Here, we resolve these critical issues to perform a high-resolution, 295-day wastewater and clinical sequencing effort, in the controlled environment of a large university campus and the broader context of the surrounding county. We develop and deploy improved virus concentration protocols and deconvolution software that fully resolve multiple virus strains from wastewater. We detect emerging variants of concern up to 14 days earlier in wastewater samples, and identify multiple instances of virus spread not captured by clinical genomic surveillance. Our study provides a scalable solution for wastewater genomic surveillance that allows early detection of SARS-CoV-2 variants and identification of cryptic transmission.
