Organic photosensitizers (PSs) with aggregation-induced emission (AIE) characteristics have shown several advantages in photodynamic therapy (PDT). However, how to design PSs with the required optical and biological properties is a formidable challenge. In this work, a series of AIE active pyridine- and pyridinium-functionalized carbazole with different substituents are designed and synthesized. Compared to other analogs, BK-Pys-BP containing benzophenone moiety on the pyridinium unit produces reactive oxygen species (ROS) efficiently in the presence of light due to its large spin-orbit coupling constant to promote efficient intersystem crossing. Based on its structure, three molecules with enhanced donor-acceptor strength and conjugation are further investigated to realize emission covering the whole visible region. The cationic nature and excellent ROS generation ability of benzophenone-containing AIE photosensitizers allows for specific imaging of mitochondria, killing cancer cells, and as a singlet oxygen donor to catalyze the formation of endoperoxides. Thus, the present work presents an effective strategy for creating AIE photosensitizers with efficient ROS generation for wide biological applications. A molecular design concept to enhance reactive oxygen species (ROS) generation is presented, and full-wavelength visible light materials are synthesized and applied to photodynamic therapy and catalysis of endoperoxide formation. This work provides insight into improving the ROS efficiency of photosensitizers and controlling the release of 1O2 to achieve afterglow therapy.image

Light-mediated therapies such as photodynamic therapy (PDT) are considered emerging cancer treatment strategies. However, there are still lots of defect with common photosensitizers (PSs), such as short emission wavelength, weak photostability, poor cell permeability, and low PDT efficiency. Therefore, it is very important to develop high-performance PSs. Recently, luminogens with aggregation-induced emission (AIE) characteristics and red/near-infrared (NIR) emissive have been reported as promising PSs for image-guided cancer therapy, due to them being able to prevent autofluorescence in physiological environments, their enhanced fluorescence in the aggregated state, and generation of reactive oxygen species (ROS). Herein, we developed PSs named TBTCPM and MTBTCPM with donor–acceptor (D–A) structures, strong red/NIR, excellent targeting specificities to good cell permeability, and high photostability. Interestingly, both of them can efficiently generate ROS under white light irradiation and possess excellent killing effect on cancer cells. This study, thus, not only demonstrates applications in cell image-guided PDT cancer therapy performances but also provides strategy for construction of AIEgens with long emission wavelengths. © 2023 John Wiley & Sons, Ltd.

Two novel highly selective quinoline-based fluorescent probes (1 and 2) with an aggregation induced emission (AIE) feature have been designed and synthesized for the rapid analysis of Cu2+ ions in aqueous media and on paper strips with a fluorescence quenching mechanism. Moreover, probes 1 and 2 exhibit excellent sensitivity and anti-interference for Cu2+ detection, and the detection limits are as low as 1.3 x 10(-8) M and 8.5 x 10(-8) M, respectively, which are much lower than the allowable standard of Cu2+ (approximate to 20 M) in drinking water (EPA). More importantly, these two probes were successfully applied for the determination of Cu2+ in real aqueous samples and fabrication of simple device test strips for rapid and on-site detection of Cu2+ ions. Interestingly, they can also be regenerated by adding an excess of S2-. Additionally, the crystallographic structure of probe 1 was confirmed through a single crystal X-ray study. Job's plot analysis and ESI-MS spectroscopic studies reflect the 1:1 complexation of the 1-Cu2+ and 2-Cu2+ complexes. Furthermore, DFT/TDDFT calculations were performed in order to help in understanding the electronic properties of the complexes and the chelation-induced quenching mechanism.
