When someone violates a social norm, others may think that some sanction would be appropriate. We examine how the experience of emotions like anger and disgust relate to the judged appropriateness of sanctions, in a pre-registered analysis of data from a large-scale study in 56 societies. Across the world, we find that individuals who experience anger and disgust over a norm violation are more likely to endorse confrontation, ostracism and, to a smaller extent, gossip. Moreover, we find that the experience of anger is consistently the strongest predictor of judgments of confrontation, compared to other emotions. Although the link between state-based emotions and judgments may seem universal, its strength varies across countries. Aligned with theoretical predictions, this link is stronger in societies, and among individuals, that place higher value on individual autonomy. Hereditary Hemorrhagic Telangiectasia (HHT) is an autosomal-dominant genetic disorder involving defects in two predominant genes known as endoglin (ENG; HHT-1) and activin receptor-like kinase 1 (ACVRL1/ALK1; HHT-2). It is characterized by mucocutaneous telangiectases that, due to their fragility, frequently break causing recurrent epistaxis and gastrointestinal bleeding. Because of the severity of hemorrhages, the study of the hemostasis involved in these vascular ruptures is critical to find therapies for this disease. Our results demonstrate that HHT patients with high bleeding, as determined by a high Epistaxis Severity Score (ESS), do not have prolonged clotting times or alterations in clotting factors. Considering that coagulation is only one of the processes involved in hemostasis, the main objective of this study was to investigate the overall mechanisms of hemostasis in HHT-1 (Eng+/−) and HHT-2 (Alk1+/−) mouse models, which do not show HHT vascular phenotypes in the meaning of spontaneous bleeding. In Eng+/− mice, the results of in vivo and in vitro assays suggest deficient platelet-endothelium interactions that impair a robust and stable thrombus formation. Consequently, the thrombus could be torn off and dragged by the mechanical force exerted by the bloodstream, leading to the reappearance of hemorrhages. In Alk1+/− mice, an overactivation of the fibrinolysis system was observed. These results support the idea that endoglin and Alk1 haploinsufficiency leads to a common phenotype of impaired hemostasis, but through different mechanisms. This contribution opens new therapeutic approaches to HHT patients' epistaxis.

Riverine and coastal wetlands are vital ecosystems for nature and people. They provide significant contributions to human health and well-being, biodiversity conservation, climate change mitigation and adaptation. To halt the decline of wetland ecosystems in Europe and to boost their resilience, there is an urgent need to scale up action. This paper discusses barriers and presents recommendations for the effective restoration of riverine and coastal wetlands in Europe, highlighting the importance of integrating initiatives across science, policy and practice.

The contribution of biotic interactions to shaping broad-scale species distributions is, however, still debated, partly due to the difficulty of detecting their effects. We aim to test whether spatial exclusion between potentially competing species can be detected at the species range scale, and whether this pattern relates to fine-scale mechanisms of coexistence. Location Western Palearctic. Methods We develop and evaluate a measure of geographic avoidance that uses outputs of species distribution models to quantify geographic exclusion patterns expected if interspecific competition affects broad-scale distributions. We apply the measure to 10 Palearctic bat species belonging to four morphologically similar cryptic groups in which competition is likely to occur. We compare outputs to null models based on pairs of virtual species and to expectations based on ecological similarity and fine-scale coexistence mechanisms. We project changes in range suitability under climate change taking into account effects of geographic avoidance. Results Values of geographic avoidance were above null expectations for two cryptic species pairs, suggesting that interspecific competition could have contributed to shaping their broad-scale distributions. These two pairs showed highest levels of ecological similarity and no trophic or habitat partitioning. Considering the role of competition modified predictions of future range suitability. Urban, industrial, and agricultural development in river basins has resulted in the pollution of estuarine and coastal ecosystems with a great amount of organic microcontaminants (OMCs) such as pesticides and contaminants of emerging concern (CECs). This study takes the Ebro Delta as a case study to assess the increase or reduction of 25 OMCs in rice fields and one 86 ha constructed wetland (CW). Bentazone and MCPA were the most abundant pesticides in the rice-field drainage water, with a peak concentration of 21,318 and 938 ng/L respectively, whereas the greatest CEC concentrations were found for caffeine, benzotriazoles, and bisphenol A (20–71 ng/L, on average) in the rice irrigation water. Pesticide concentration increased after the irrigation water passed through the rice fields (from 102 to 1973 ng/L, on average), but CECs present in the irrigation water decreased by 37% (from 14 to 10 ng/L, on average). A mass balance study showed that the CW was capable of reducing OMCs by 67%. Risk assessment analysis showed that the cumulative hazard quotient for Daphnia magna, green algae, and fish was greater than 1 during several sampling campaigns for the rice-field drainage water, but the CW was capable of reducing it by 60–63%, resulting in values below 1, which indicates that the risk was not significant. The results thus indicate that rice fields reduce CECs, but increase pesticides, whereas the use of CWs seems to be a feasible nature-based solution to reduce the discharge of OMCs into estuarine and coastal areas.

Following arterial occlusion, blood vessels respond by forming a new network of functional capillaries (angiogenesis), by re-organizing pre-existing capillaries through the recruitment of smooth muscle cells to generate new arteries (arteriogenesis) and by growing and remodeling pre-existing collateral arterioles into physiologically relevant arteries (collateral development). All these processes result in the recovery of organ perfusion. The importance of endoglin in post-occlusion reperfusion is sustained by several observations: i) endoglin expression is increased in vessels showing active angiogenesis/remodeling; ii) genetic endoglin haploinsufficiency in humans causes deficient angiogenesis; and iii) the reduction of endoglin expression by gene disruption or the administration of endoglin-neutralizing antibodies reduces angiogenesis and revascularization. However, the precise role of endoglin in the several processes associated with revascularization has not been completely elucidated and, in some cases, the function ascribed to endoglin by different authors is controversial. The purpose of this review is to organize in a critical way the information available for the role of endoglin in several phenomena (angiogenesis, arteriogenesis, and collateral development) associated with post-ischemic revascularization.

The effectiveness of treatments in stages aimed at reducing the leaching of a solution rich in Zn and Mn obtained from a recycling process introduced into the wood by the Bethell method and a second stage in which borax was added by brushing and by the Bethell method, was evaluated through a field test. Pinus taeda wood was used according to EN 252 (1989). This paper presents an analysis of the evaluation and evolution of the process of deterioration over 5 years test. Scheffer climate index was used as a method of microbiological risk assessment at the site of the test and microscopic analysis of wood was carried out to verify the type of damage. Treatments 3% and 6% in Zn with borax added by the Bethell method were the top performers among the evaluated none of them performed better than the reference preserver. The presence of the three types of rot, white, brown and soft both on untreated controls and treated wood, in a site with intermediate conditions for microbial deterioration was observed., A través de un ensayo de campo implementado de acuerdo a la norma EN 252, se evaluó la efectividad de tratamientos en etapas, tendientes a disminuir la lixiviación de una solución rica en Zn y Mn obtenida a partir de un proceso de reciclado aplicada en la madera por el método Bethell. BACKGROUND: Unanticipated difficult intubation remains a challenge in anaesthesia. The Simplified Airway Risk Index (SARI) is a multivariable risk model consisting of seven independent risk factors for difficult intubation. Our aim was to compare preoperative airway assessment based on the SARI with usual airway assessment.METHODS: From 01.10.2012 to 31.12.2013, 28 departments were cluster-randomized to apply the SARI model or usual airway assessment. The SARI group implemented the SARI model. The Non-SARI group continued usual airway assessment, thus reflecting a group of anaesthetists' heterogeneous individual airway assessments. Preoperative prediction of difficult intubation and actual intubation difficulties were registered in the Danish Anaesthesia Database for both groups. Patients who were preoperatively scheduled for intubation by advanced techniques (e.g. video laryngoscopy; flexible optic scope) were excluded from the primary analysis. Primary outcomes were the proportions of unanticipated difficult and unanticipated easy intubation.RESULTS: A total of 26 departments (15 SARI and 11 Non-SARI) and 64 273 participants were included. In the primary analyses 29 209 SARI and 30 305 Non-SARI participants were included.In SARI departments 2.4% (696) of the participants had an unanticipated difficult intubation vs 2.4% (723) in Non-SARI departments. Odds ratio (OR) adjusted for design variables was 1.03 (95% CI: 0.77-1.38). The proportion of unanticipated easy intubation was 1.42% (415) in SARI departments vs 1.00% (302) in Non-SARI departments. Adjusted OR was 1.26 (0.68-2.34).CONCLUSIONS: Using the SARI compared with usual airway assessment we detected no statistical significant changes in unanticipated difficult- or easy intubations.CLINICAL TRIAL REGISTRATION: NCT01718561.

El tenofovir (TDF), es el único inhibidor de la transcriptasa inversa análogo nucleótido para el tratamiento de la infección por virus de la inmunodeficiencia humana (VIH). Ocasionalmente, puede producir insuficiencia renal aguda y síndrome de Fanconi. Presentamos el caso de un varón de 64 años con infección por VIH conocida desde hace 22 años, en tratamiento con tenofovir. En las revisiones ambulatorias refería un cuadro progresivo de astenia y dolores óseos difusos. En varias determinaciones se había observado una elevación de la fosfatasa alcalina y la paratohormona (PTH). Durante el último mes empeoró su estado, por lo que fue ingresado en el hospital. Entre los datos analíticos destacaban: glucosuria marcada, hipofosfatemia, hiperfosfaturia e hipouricemia. Todas las alteraciones se resolvieron tras suspender el TDF, lo que ilustra la importancia de que los clínicos incluyan la posibilidad de tubulopatía proximal por TDF en pacientes con dolores óseos, síndrome general o alteraciones del metabolismo mineral., Tenofovir (TDF), is the only nucleotide analogue reverse transcriptase inhibitor for treating human immunodeficiency virus (HIV). Occasionally, it may cause acute renal failure and Fanconi syndrome. We report the case of a 64-year-old male diagnosed with HIV infection 22 years previous and treated with tenofovir. In outpatient follow-up, the patient complained of progressive fatigue and diffuse aching bones.In several check-ups, increased alkaline phosphatase and parathyroid hormone (PTH) were observed. Over the past month, his condition worsened and he was admitted to hospital. Analytical data included marked glycosuria, hypophosphatemia, hyperphosphaturia and hypouricemia. All changes were resolved when TDF was discontinued.This illustrates the importance of clinical evaluations that include possible TDF-induced proximal tubulopathy in patients with general bone pain syndrome or mineral metabolism disturbances.

Objectives: After the last pandemic the knowledge regarding influenza A infection has improved however, the outcomes of influenza B infection remain poorly studied. The aim of this study was to compare the features of influenza B versus influenza A(H1N1)pdm09 infections during the 2010-2011 epidemic-season., Methods: A prospective, observational-cohort of adults with laboratory-confirmed influenza infection during the 2010-2011 epidemic-season was studied., Results: Fifty cases of influenza B and 80 of influenza A(H1N1)pdm09 infection were enrolled. Among patients with influenza B, the median age was 34 years-old (23-64), 30% pregnant, 24% obese, 34% transplant recipients and 14% with bacterial co-infection. Twenty-eight percent of patients had pneumonia with alveolar localized pattern and five (10%) died. Pneumonia was associated with delayed antiviral therapy, older age, higher Charlson score, invasive mechanical ventilation and bacterial co-infection. Obesity and pregnancy were not associated with complicated influenza B infection. The proportion of pneumonia, admission to the ICU and mortality did not differ between cases of influenza A(H1N1)pdm09 and influenza B infection., Conclusions: Influenza B infection causes severe infection and it is associated with pneumonia or death, similar to influenza A(H1N1)pdm09 infection. Rapid diagnosis and early antiviral therapy are necessary for managing influenza pneumonia during epidemic periods. © 2012 The British Infection Association.

Many members of the benthic fauna of the Antarctic continental shelf share close phylogenetic relationships to the deep-sea fauna adjacent to Antarctica and in other ocean basins. It has been suggested that connections between the Southern Ocean and the deep sea have been facilitated by the presence of a deep Antarctic continental shelf coupled with submerging Antarctic bottom water and emerging circumpolar deep water. These conditions may have allowed ‘polar submergence’, whereby shallow Southern Ocean fauna have colonised the deep sea and ‘polar emergence’, whereby deep-sea fauna colonised the shallow Southern Ocean. A recent molecular study showed that a lineage of deep-sea and Southern Ocean octopuses with a uniserial sucker arrangement on their arms appear to have arisen via polar submergence. A distantly related clade of octopuses with a biserial sucker arrangement on their arms (historically placed in the genus Benthoctopus) is also present in the deep-sea basins of the world and the Southern Ocean. To date their evolutionary history has not been examined. The present study investigated the origins of this group using 3133 base pairs (bp) of nucleotide data from five mitochondrial genes (12S rRNA, 16S rRNA, cytochrome c oxidase subunit I, cytochrome c oxidase subunit III, cytochrome b) and the nuclear gene rhodopsin from at least 18 species (and 7 outgroup taxa). Bayesian relaxed clock analyses showed that Benthoctopus species with a high-latitude distribution in the Southern Hemisphere represent a paraphyletic group comprised of three independent clades. The results suggest that the Benthoctopus clade originated in relatively shallow Northern Hemisphere waters. Benthoctopus species distributed in the Southern Ocean are representative of polar emergence and occur at shallower depths than non-polar Benthoctopus species.

The neurotrophin gene family, including nerve growth factor (NGF), brain-derived neurotrophic factor (BDNF), neurotrophin-3 (NT-3), and NT-4/NT-5, supports the survival of distinct peripheral neurons, however, actions upon central neurons are relatively undefined. In this study we have compared different neurotrophins in the regulation of neuronal survival and function using dissociated embryonic cell cultures from two brain regions, the basal forebrain (BF) and locus coeruleus (LC). In the BF, NGF increased choline acetyl transferase (ChAT) activity, but did not influence cholinergic cell survival. In contrast to NGF, BDNF, NT-3, and the novel neurotrophin, NT-4, all increased ChAT activity and cholinergic cell survival. We also examined embryonic LC neurons in culture. LC neurons are unresponsive to NGF. In contrast, NT-3 and NT-4 elicited significant increases in survival of noradrenergic LC neurons, the first demonstration of trophic effects in this critical brain region. Identification of factors supporting coeruleal and basal forebrain neuronal survival may provide insight into mechanisms mediating degeneration of these disparate structures in clinical disorders.

Neurotrophin-4 (NT-4), a recently discovered novel member of the family of neurotrophic factors structurally related to nerve growth factor (NGF), is abundantly expressed in the Xenopus laevis ovary. In this study we have localized NT-4 mRNA expressing cells in the Xenopus ovary by in situ hybridization and have used this technique together with Northern blot analyses to quantify NT-4 mRNA expression during oogenesis in Xenopus. In situ hybridization of sections through the Xenopus ovary using an alpha-[35S]-dATP labeled Xenopus NT-4 mRNA specific probe showed an intense labeling over the cytoplasm of oocytes with a diameter of 50-200 microns corresponding to stage I according to Dumont (1972). Labeling was also seen over the cytoplasm of stages II to IV although with a lower intensity than over stage I oocytes. No labeling was seen over more mature oocytes of stages V and VI. NT-4 mRNA could not be detected in the early embryo from the onset of cleavage division to the neurula stage suggesting that the NT-4 gene is not expressed during Xenopus early embryogenesis. The confinement of NT-4 mRNA in the Xenopus ovary to immature oocytes suggests that NT-4 mRNA expression is strictly regulated during oogenesis and that the NT-4 protein could play a role as a maturation factor for immature oocytes.

Evolutionary conservation of members of the NGF family in vertebrates was studied by DNA sequence analysis of PCR fragments for NGF, BDNF, and NT-3 from human, rat, chicken, viper, Xenopus, salmon, and ray. The results showed that the three factors are highly conserved from fishes to mammals. Phylogenetic trees reflecting the evolution and speciation of the members of the NGF family were constructed. In addition, the gene for a fourth member of the family, neurotrophin-4 (NT-4), was isolated from Xenopus and viper. The NT-4 gene encodes a precursor protein of 236 amino acids, which is processed into a 123 amino acid mature NT-4 protein with 50%-60% amino acid identity to NGF, BDNF, and NT-3. The NT-4 protein was shown to interact with the low affinity NGF receptor and elicited neurite outgrowth from explanted dorsal root ganglia with no and lower activity in sympathetic and nodose ganglia, respectively. Northern blot analysis of different tissues from Xenopus showed NT-4 mRNA only in ovary, where it was present at levels over 100-fold higher than those of NGF mRNA in heart.

Biological and immunological properties of recombinant human, rat, and chicken nerve growth factors (NGFs) were studied and compared. Recombinant NGF proteins were produced in a transient expression system using COS cells and levels of secreted NGF protein were assessed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis of conditioned media from in vivo [35S]cysteine-labeled cell cultures. Antigenic differences among the three NGFs were studied by immunoblotting and immunoprecipitation of secreted cell products using a rabbit polyclonal antiserum against purified mouse NGF, and by a two-site enzyme immunoassay (EIA) with a monoclonal antibody against mouse NGF. Although all three NGFs were recognized equally well in the immunoblotting, only one-third of the chicken NGF protein could be detected by immunoprecipitation or by the EIA as compared to the rat and human NGFs. Thus, changes in the three-dimensional structure of the NGF molecule are most likely responsible for the antigenic differences between avian and mammalian NGFs. The three NGF proteins were also compared in their ability to displace 125I-mouse NGF from low-affinity NGF receptors on rat pheochromocytoma PC12 cells. Similar displacement curves and values were obtained for each NGF protein, indicating that structural differences among these molecules do not affect low-affinity binding to NGF receptors. Biological activities were studied by the ability of the conditioned media to promote neurite outgrowth from explants of E9 chick sympathetic ganglia and from PC12 cells. Although the rat system showed a slight preference for the homologous molecule, the morphological changes, dose-response curves, and maximal stimulation values obtained with the different NGFs were practically indistinguishable in the chicken bioassay.(ABSTRACT TRUNCATED AT 250 WORDS)

Selected amino acid residues in chicken nerve growth factor (NGF) were replaced by site-directed mutagenesis. Mutated NGF sequences were transiently expressed in COS cells and the yield of NGF protein in conditioned medium was quantified by Western blotting. Binding of each mutant to NGF receptors on PC12 cells was evaluated in a competition assay. The biological activity was determined by measuring stimulation of neurite outgrowth from chick sympathetic ganglia. The residues homologous to the proposed receptor binding site of insulin (Ser18, Met19, Val21, Asp23) were substituted by Ala. Replacement of Ser18, Met19 and Asp23 did not affect NGF activity. Modification of Val21 notably reduced both receptor binding and biological activity, suggesting that this residue is important to retain a fully active NGF. The highly conserved Tyr51 and Arg99 were converted into Phe and Lys respectively, without changing the biological properties of the molecule. However, binding and biological activity were greatly impaired after the simultaneous replacement of both Arg99 and Arg102 by Gly. The three conserved Trp residues at positions 20, 75 and 98 were substituted by Phe. The Trp mutated proteins retained 15-60% of receptor binding and 40-80% of biological activity, indicating that the Trp residues are not essential for NGF activity. However, replacement of Trp20 significantly reduced the amount of NGF in the medium, suggesting that this residue may be important for protein stability.

Nerve growth factor (NGF) supports sympathetic and sensory neurons in the peripheral nervous system and also functions in the development and maintenance of cholinergic neurons in the basal forebrain. NGF distribution can be studied in the brain of the rat and mouse with the use of a sensitive two-site enzyme immunoassay (EIA) for mouse NGF. It would be of interest to measure the NGF protein also in the human brain, especially against the background that the cholinergic neurons are severely deteriorated in senile dementia of the Alzheimer type. The limited immunological cross-reactivity between NGFs from different species has previously hampered attempts to determine levels of the human NGF. We have now examined the biological activity and immunological properties of human recombinant NGF protein in medium conditioned by COS cells transfected with the human NGF gene. The human NGF behaved similar to mouse NGF in a sympathetic ganglion bioassay. The monoclonal antibody 27/21 to mouse NGF was shown to effectively block the activity of both the human recombinant NGF and mouse native NGF. A two-site EIA using monoclonal antibody 27/21 was optimized. Under the conditions used, the EIA detected the human recombinant NGF with the same sensitivity (1 pg/ml) as shown for the mouse NGF. It should now be possible to test this EIA also on homogenized tissue to examine human NGF in brain samples from Alzheimer patients and age-matched controls.

Within the context of global climate change and overfishing of fish stocks, there is some evidence that cephalopod populations are benefiting from this changing setting. These invertebrates show enhanced phenotypic flexibility and are found from polar regions to the tropics. Yet, the global patterns of species richness in coastal cephalopods are not known. Here, among the 370 identified-species, 164 are octopuses, 96 are cuttlefishes, 54 are bobtails and bottletails, 48 are inshore squids and 8 are pygmy squids. The most diverse ocean is the Pacific (with 213 cephalopod species), followed by the Indian (146 species) and Atlantic (95 species). The least diverse are the Southern (15 species) and the Arctic (12 species) Oceans. Endemism is higher in the Southern Ocean (87%) and lower in the Arctic (25%), which reflects the younger age and the "Atlantification" of the latter. The former is associated with an old lineage of octopuses that diverged around 33 Ma. Within the 232 ecoregions considered, the highest values of octopus and cuttlefish richness are observed in the Central Kuroshio Current ecoregion (with a total of 64 species), followed by the East China Sea (59 species). This pattern suggests dispersal in the Central Indo-Pacific (CIP) associated with the highly productive Oyashio/Kuroshio current system. In contrast, inshore squid hotspots are found within the CIP, namely in the Sunda Shelf province, which may be linked to the occurrence of an ancient intermittent biogeographic barrier: a land bridge formed during the Pleistocene which severely restricted water flow between the Pacific and Indian Oceans, thereby facilitating squid fauna differentiation.
