- Abstract
BACKGROUND: Hypertrophic cardiomyopathy (HCM) is caused by sarcomere gene mutations (genotype-positive HCM) in ≈50% of patients and occurs in the absence of mutations (genotype-negative HCM) in the other half of patients. We explored how alterations in the metabolomic and lipidomic landscape are involved in cardiac remodeling in both patient groups. METHODS: We performed proteomics, metabolomics, and lipidomics on myectomy samples (genotype-positive N=19; genotype-negative N=22; and genotype unknown N=6) from clinically well-phenotyped patients with HCM and on cardiac tissue samples from sex- and age-matched and body mass index-matched nonfailing donors (N=20). These data sets were integrated to comprehensively map changes in lipid-handling and energy metabolism pathways. By linking metabolomic and lipidomic data to variability in clinical data, we explored patient group-specific associations between cardiac and metabolic remodeling. RESULTS:HCM myectomy samples exhibited (1) increased glucose and glycogen metabolism, (2) downregulation of fatty acid oxidation, and (3) reduced ceramide formation and lipid storage. In genotype-negative patients, septal hypertrophy and diastolic dysfunction correlated with lowering of acylcarnitines, redox metabolites, amino acids, pentose phosphate pathway intermediates, purines, and pyrimidines. In contrast, redox metabolites, amino acids, pentose phosphate pathway intermediates, purines, and pyrimidines were positively associated with septal hypertrophy and diastolic impairment in genotype-positive patients. CONCLUSIONS: We provide novel insights into both general and genotype-specific metabolic changes in HCM. Distinct metabolic alterations underlie cardiac disease progression in genotype-negative and genotype-positive patients with HCM.

- Abstract
Aims: Genetic hypertrophic cardiomyopathy (HCM) is caused by mutations in sarcomere protein-encoding genes (i.e. genotype-positive HCM). In an increasing number of patients, HCM occurs in the absence of a mutation (i.e. genotype-negative HCM). Mitochondrial dysfunction is thought to be a key driver of pathological remodelling in HCM. Reports of mitochondrial respiratory function and specific disease-modifying treatment options in patients with HCM are scarce. Methods and results: Respirometry was performed on septal myectomy tissue from patients with HCM (n = 59) to evaluate oxidative phosphorylation and fatty acid oxidation. Mitochondrial dysfunction was most notably reflected by impaired NADH-linked respiration. In genotype-negative patients, but not genotype-positive patients, NADH-linked respiration was markedly depressed in patients with an indexed septal thickness ≥10 compared with <10. Mitochondrial dysfunction was not explained by reduced abundance or fragmentation of mitochondria, as evaluated by transmission electron microscopy. Rather, improper organization of mitochondria relative to myofibrils (expressed as a percentage of disorganized mitochondria) was strongly associated with mitochondrial dysfunction. Pre-incubation with the cardiolipin-stabilizing drug elamipretide and raising mitochondrial NAD+ levels both boosted NADH-linked respiration. Conclusion: Mitochondrial dysfunction is explained by cardiomyocyte architecture disruption and is linked to septal hypertrophy in genotype-negative HCM. Despite severe myocardial remodelling mitochondria were responsive to treatments aimed at restoring respiratory function, eliciting the mitochondria as a drug target to prevent and ameliorate cardiac disease in HCM. Mitochondria-targeting therapy may particularly benefit genotype-negative patients with HCM, given the tight link between mitochondrial impairment and septal thickening in this subpopulation.

- Abstract
Hypertrophic cardiomyopathy (HCM) is a relatively common genetic heart disease characterised by myocardial hypertrophy. HCM can cause outflow tract obstruction, sudden cardiac death and heart failure, but severity is highly variable. In this exploratory cross-sectional study, circulating acylcarnitines were assessed as potential biomarkers in 124 MYBPC3 founder variant carriers (59 with severe HCM, 26 with mild HCM and 39 phenotype-negative [G + P-]). Elastic net logistic regression identified eight acylcarnitines associated with HCM severity. C3, C4, C6-DC, C8:1, C16, C18 and C18:2 were significantly increased in severe HCM compared to G + P-, and C3, C6-DC, C8:1 and C18 in mild HCM compared to G + P-. In multivariable linear regression, C6-DC and C8:1 correlated to log-transformed maximum wall thickness (coefficient 5.01, p = 0.005 and coefficient 0.803, p = 0.007, respectively), and C6-DC to log-transformed ejection fraction (coefficient -2.50, p = 0.004). Acylcarnitines seem promising biomarkers for HCM severity, however prospective studies are required to determine their prognostic value. Graphical abstract: [Figure not available: see fulltext.].

- Abstract
Improving the performance of solar modules requires the implementation of both spectral and directional irradiance optimization. The performance of bifacial four-terminal tandem minimodules with a 100 cm2 area is reported, both indoor and outdoor measurements. We demonstrate a 24.5 mW/cm2 (bifacial irradiance 200 W/m2) bifacial tandem power density, measured according to a tandem adapted IEC60904-1-2 protocol, which constitutes a 3 mW/cm2 gain with respect to monofacial use. In addition, we show that in outdoor measurements bifacial four terminal tandems outperform identical monofacial four terminal tandems by 26% on average, depending on incident angle, injection level and temperature, in a 10% albedo environment. The average gain is higher for outdoor performance than for indoor performance, due to variations in relative rear irradiance.

OBJECTIVES: We sought to determine the phenotype and prognosis of MYH7-related DCM. We also evaluated the influence of variant location on phenotypic expression. METHODS: We studied clinical data from 147 individuals with DCM-causing MYH7 variants (47.6% female; 35.6 ± 19.2 years) recruited from 29 international centers. RESULTS: At initial evaluation, 106 (72.1%) patients had DCM (left ventricular ejection fraction: 34.5% ± 11.7%). Median follow-up was 4.5 years (IQR: 1.7-8.0 years), and 23.7% of carriers who were initially phenotype-negative developed DCM. Phenotypic expression by 40 and 60 years was 46% and 88%, respectively, with 18 patients (16%) first diagnosed at <18 years of age. Thirty-six percent of patients with DCM met imaging criteria for LV noncompaction. During follow-up, 28% showed left ventricular reverse remodeling. Incidence of adverse cardiac events among patients with DCM at 5 years was 11.6%, with 5 (4.6%) deaths caused by end-stage heart failure (ESHF) and 5 patients (4.6%) requiring heart transplantation. The major ventricular arrhythmia rate was low (1.0% and 2.1% at 5 years in patients with DCM and in those with LVEF of ≤35%, respectively). ESHF and major ventricular arrhythmia were significantly lower compared with LMNA-related DCM and similar to DCM caused by TTN truncating variants. CONCLUSIONS: MYH7-related DCM is characterized by early age of onset, high phenotypic expression, low left ventricular reverse remodeling, and frequent progression to ESHF. Heart failure complications predominate over ventricular arrhythmias, which are rare.

- Abstract
Improving the performance of solar modules requires the implementation of both spectral and directional irradiance optimization. The performance of bifacial four-terminal tandem minimodules with a 100 cm2 area is reported, both indoor and outdoor measurements. We demonstrate a 24.5 mW/cm2 (bifacial irradiance 200 W/m2) bifacial tandem power density, measured according to a tandem adapted IEC60904-1-2 protocol, which constitutes a 3 mW/cm2 gain with respect to monofacial use. In addition, we show that in outdoor measurements bifacial four terminal tandems outperform identical monofacial four terminal tandems by 26% on average, depending on incident angle, injection level and temperature, in a 10% albedo environment. The average gain is higher for outdoor performance than for indoor performance, due to variations in relative rear irradiance.

- Abstract
This paper presents the results from an extensive interlaboratory comparison of angular-dependent measurements on encapsulated photovoltaic (PV) cells. Twelve international laboratories measure the incident angle modifier of two unique PV devices. The absolute measurement agreement is ±2.0% to the weighted mean for angles of incidence (AOI) ≤ 65°, but from 70°–85° the range of measurement deviations increases rapidly from 2.5%–23%. The proficiency of the measurements is analyzed using the expanded uncertainties published by seven of the laboratories, and it is found that most of the angular-dependent measurements are reproducible for AOI ≤ 80°. However, at 85° one laboratory’s measurement do not agree to the weighted mean within the stated uncertainty, and measurement uncertainty as high as 16% is needed for the laboratories without uncertainty to be comparable. The poor agreement obtained at 85° indicates that the PV community should place minimal reliance on angular-dependent measurements made at this extreme angle until improvements can be demonstrated. The cloud-based Daidalos ray tracing model is used to simulate the angular-dependent losses of the mono-Si device and it is found that the simulation agrees to the median measurement within 0.6% for AOI ≤ 70° and within 1.4% for AOI ≤ 80°. Finally, the impact that the angular-dependent measurement deviations have on climate specific energy rating (CSER) is evaluated for the six climates described in the IEC 61853-4 standard. When one outlier measurement is excluded, the angular-dependent measurements reported in this work cause a 1.0%–1.8% range in CSER and a 1.0%–1.5% range in annual energy yield, depending on the climate.

Hypertrophic Cardiomyopathy (HCM) is a common inherited heart disease with poor risk prediction due to incomplete penetrance and a lack of clear genotype–phenotype correlations. Advanced imaging techniques have shown altered myocardial energetics already in preclinical gene variant carriers. To determine whether disturbed myocardial energetics with the potential to serve as biomarkers are also reflected in the serum metabolome, we analyzed the serum metabolome of asymptomatic carriers in comparison to healthy controls and obstructive HCM patients (HOCM). We performed non-quantitative direct-infusion high-resolution mass spectrometry-based untargeted metabolomics on serum from fasted asymptomatic gene variant carriers, symptomatic HOCM patients and healthy controls (n = 31, 14 and 9, respectively). Biomarker panels that discriminated the groups were identified by performing multivariate modeling with gradient-boosting classifiers. For all three group-wise comparisons we identified a panel of 30 serum metabolites that best discriminated the groups. These metabolite panels performed equally well as advanced cardiac imaging modalities in distinguishing the groups. Seven metabolites were found to be predictive in two different comparisons and may play an important role in defining the disease stage. This study reveals unique metabolic signatures in serum of preclinical carriers and HOCM patients that may potentially be used for HCM risk stratification and precision therapeutics.

- Abstract
Introduction Despite major advances in our understanding of genetic cardiomyopathies, they remain the leading cause of premature sudden cardiac death and end-stage heart failure in persons under the age of 60 years. Integrated research databases based on a large number of patients may provide a scaffold for future research. Using routine electronic health records and standardised biobanking, big data analysis on a larger number of patients and investigations are possible. In this article, we describe the UNRAVEL research data platform embedded in routine practice to facilitate research in genetic cardiomyopathies. Design Eligible participants with proven or suspected cardiac disease and their relatives are asked for permission to use their data and to draw blood for biobanking. Routinely collected clinical data are included in a research database by weekly extraction. A text-mining tool has been developed to enrich UNRAVEL with unstructured data in clinical notes. Preliminary results Thus far, 828 individuals with a median age of 57 years have been included, 58% of whom are male. All data are captured in a temporal sequence amounting to a total of 18,565 electrocardiograms, 3619 echocardiograms, data from over 20,000 radiological examinations and 650,000 individual laboratory measurements. Conclusion Integration of routine electronic health care in a research data platform allows efficient data collection, including all investigations in chronological sequence. Trials embedded in the electronic health record are now possible, providing cost-effective ways to answer clinical questions. - Abstract
The aim of this work is to compare angle of incidence (AOI) measurement setups for solar cells between laboratories with such capability. For the first time, we compare relative light transmission measurements among eight laboratories, whose measurement techniques include indoor and outdoor methods. We present the relative transmission measurements on three 156 mm x 156 mm crystalline-Si (c-Si) samples with different surface textures. The measurements are compared using the expanded uncertainties provided by each laboratory. Five of the eight labs showed an agreement better than ±2% to the weighted mean between AOIs from -75° to 70°. At AOIs of ±80° and ±85°, the same five labs showed a worst case deviation to the weighted mean of -3% to 5% and 0% to 18%, respectively. When measurement uncertainty is considered, the results show that measurements at the highest incidence angle of ±85° are problematic, as measurements from four out of the six labs reporting uncertainty were found non-comparable within their stated uncertainties. At 85° AOI a high to low range of up to 75% was observed between all eight laboratories.

- Abstract
Open loop, experimental force and power measurements of a radial, redundant-axis, magnetic bearing at temperatures to 1000 degrees F (538 degrees C) and rotor speeds to 15,000 RPM along with theoretical temperature and force models are presented in this paper. The experimentally measured force produced by a single C-core using 22A was 600 pounds (2.67 kN) at room temperature and 380 pounds (1.69 kN) at 1000 degrees F (538 degrees C). These values were compared with force predictions based on a 1D magnetic circuit analysis and a thermal analysis of gap growth as a function of temperature. Tests under rotating conditions showed that rotor speed has a negligible effect on the bearing's load capacity. One C-core required approximately 340 watts of power to generate 190 pounds (8.45 kN) of magnetic force at 1000 degrees F (538 degrees C). However, the magnetic air gap was much larger than at room temperature. The data presented is after the bearing had already operated 6 thermal cycles and 11 total (not consecutive) hours at 1000 degrees F (538 degrees C). Motors, magnetic bearings, and other electromagnetic actuators that can operate at 1000 degrees F (540 degrees C) hold great promise for providing increased efficiency in machinery for many applications ranging from pebble-bed nuclear reactors and chemical processing to aircraft and unmanned aerial combat vehicle (UCAV) propulsion systems. This report discusses in detail the design and fabrication of a high-temperature, heteropolar, radial magnetic bearing that was operated at 1000 degrees F (540 degrees C). The development of high-temperature wire and a coil fabrication process are two significant technical barriers overcome by the Army Research Laboratory (ARL), the NASA Glenn Research Center, and the Texas A&M University (TAMU) team. This is ARL/NASA/TAMU's third-generation, high-temperature magnetic bearing. The motivation for this research came from the pursuit of a more electric gas turbine engine and a high-temperature, large-diameter, 4-million-DN (diam in millimeters times rotor speed in revolutions per minute) rotor support system. - Abstract
Open loop, experimental force and power measurements of a three-axis, radial, heteropolar magnetic bearing at room temperature for rotor speeds up to 20,000 RPM are presented in this paper. The bearing, NASA Glenn Research Center's and Texas A&M's third generation high temperature magnetic bearing, was designed to operate in a 1000 deg. F (540 deg. C) environment and was primarily optimized for maximum load capacity. The experimentally measured force produced by one C-core of this bearing was 630 lb. (2.8 kN) at 16 A, while a load of 650 lbs (2.89 kN) was predicted at 16 A using 1D circuit analysis. The maximum predicted radial load for one of the three axes is 1,440 lbs (6.41 kN) at room temperature. The maximum measured load of an axis was 1050 lbs. (4.73 kN). Results of test under rotating conditions showed that rotor speed has a negligible effect on the bearing's load capacity. A single C-core required approximately 70 W of power to generate 300 lb (1.34 kN) of magnetic force. The room temperature data presented was measured after three thermal cycles up to 1000 deg. F (540 deg.
