In 2016, the order Mononegavirales was emended through the addition of two new families (Mymonaviridae and Sunviridae), the elevation of the paramyxoviral subfamily Pneumovirinae to family status (Pneumoviridae), the addition of five free-floating genera (Anphevirus, Arlivirus, Chengtivirus, Crustavirus, and Wastrivirus), and several other changes at the genus and species levels. This article presents the updated taxonomy of the order Mononegavirales as now accepted by the International Committee on Taxonomy of Viruses (ICTV).

Symptoms of citrus leprosis were observed during 2004 - 2005 in the East plains of Colombia, Departments of Meta and Casanare. To identify the causal agent, CORPOICA collected leaves of Valencia sweet orange exhibiting typical lesions of leprosis from several locations in the Departments of Casanare (Yopal, Aguazul) and Meta (Guamal, Villavicencio and Cumaral). Virus identification was done by RT-PCR and transmission electron microscopy (TEM). Ten of the 21 samples examined by TEM resulted positive to the leprosis virus (Citrus Leprosis Virus, CiLV-C). Short bacilliform particles characteristic of CiLV-C were found in the endoplasmic reticulum, and irregularly shaped viroplasms were present in the cytoplasm. Fourteen of the 32 samples processed by RT-PCR were positive to CiLV-C. RT-PCR analyses amplified cDNA fragments of expected size for samples collected in Meta. One of the amplifications was sequenced (GenBank accession No. DQ272491) and found to have 98% nucleotide sequence identity to the Brazilian CiLV-C isolate (GenBank accession No. AY289190.1). Sequence and primers were provided to ICA for CiLV-C diagnosis in Colombia. Mites collected from affected plants were identified as Brevipalpus phoenicis (Geijskes), one of the most important vectors of CiLV-C., Durante los años 2004 y 2005 se observaron síntomas de la leprosis de los cítricos en los departamentos de Meta y Casanare. Viroids, non-protein-coding small (246-401 nt) circular single-stranded RNAs with autonomous replication, are currently classified into two families. Within the family Pospiviroidae, Citrus exocortis viroid (CEVd) belongs to the genus Pospiviroid while Hop stunt viroid (HSVd) is the single member of the genus Hostuviroid. These pathogens are distributed worldwide and infect a large number of hosts. In Brazil, isolates of CEVd and HSVd have been detected in both citrus and grapevine. To characterize and study the genetic variability of these viroids, total RNA from leaves of grapevine Vitis vinifera 'Cabernet Sauvignon' and V. labrusca 'Niagara Rosada' from Bento Gonçalves, RS, was used as a template for RT-PCR amplification with specific primers for the five viroids described infecting grapevines [HSVd, CEVd, Grapevine yellow speckle viroid 1 (GYSVd-1), Grapevine yellow speckle viroid 2 (GYSVd-2) and Australian grapevine viroid (AGVd)]. Leaf samples of Citrus medica infected with CEVd from São Paulo were also analyzed. The resulting products were separated by agarose gel electrophoresis and DNA fragments of the expected size were eluted, cloned and sequenced. The grapevine samples analyzed were doubly infected by CEVd and HSVd. A phylogenetic analysis showed that the Brazilian grapevine HSVd variants clustered with other grapevine HSVd variants, forming a specific group separated from citrus variants, whereas the Brazilian CEVd variants clustered with other citrus and grapevine variants.

We report here the finding of a new plant virus, Stilbocarpa mosaic bacilliform badnavirus (SMBV) in Stilbocarpa polaris (Homb. et Jacq.) Gray on subantarctic Macquarie Island. This is the first vascular plant virus reported from any of the remote subantarctic islands, and represents the southernmost plant virus found anywhere. The virus causes mild to severe yellow mosaic symptoms in the perennial megaherb S. polaris. Virus distribution appears most widespread in the northern third of Macquarie Island, although S. polaris occurs around the whole island. DNA sequencing of a conserved part of the virus genome showed it is most closely related to badnaviruses of temperate and tropical plants, which are mainly transmitted by mealybugs (Hemiptera: Coccoidea). No mealybugs have been found on Macquarie Island, where the vector may be an aphid (Hemiptera: Aphidoidea) or thrip (Thysanoptera). We discuss possible origins, dispersal and evolution of this plant virus on isolated Macquarie Island.

Three potyviruses were identified by gene sequencing and found to be widespread in species of Apiaceae in Australia. Only celery mosaic virus was found in celery crops and in one of 180 specimens of feral carrot (Daucus carota). Another related but distinct novel potyvirus, carrot virus Y, was the only virus found in carrot crops and all except one feral carrot. A more distantly related novel potyvirus, apium virus Y, was found in plants of sea celery (Apium prostratum), cultivated parsley (Petroselinum crispum) and the immigrant weed species poison hemlock (Conium maculatum). These three potyviruses, together with celery yellow mosaic virus of South America and the closely related carrot thin leaf virus and carrot virus B of North America, form a distinct subgenus of the Potyviridae most closely related to turnip mosaic virus and two potyviruses of yam; yam mosaic virus from the Ivory Coast and Japanese yam mosaic virus. Celery mosaic and carrot virus Y are probably recent migrants to Australia, but apium virus Y may have been endemic longer. In ELISA tests using polyclonal antibodies against virions of celery mosaic virus, some isolates of carrot virus Y were indistinguishable from celery mosaic virus, whereas others gave smaller absorbancy values, and those of apium virus Y did not react. This study shows the value of virus identification based on gene sequencing for planning control measures.
