Abstract

This chapter reviews the morphodynamics of open-ocean barrier systems, synthesizing classic studies, current scientific knowledge, and future research directions regarding 14 barrier provinces worldwide. Within a coastal tectonic framework, it addresses: (1) Amero-trailing-edge coasts (USA’s New England coast, Mid-Atlantic Bight coast, North Carolina Outer Banks, Georgia Bight coast, Florida Atlantic coast; Brazil’s Santa Catarina coast; Europe’s German Bight coast; Australia’s southern and western coasts); (2) marginal-sea coasts (USA’s Florida Gulf Coast; Gulf Coast of Louisiana, Mississippi, and Alabama; Texas Gulf Coast; Australia’s eastern coast); and (3) collision coasts (USA’s Alaskan Pacific coast, Japan’s and New Zealand’s coasts). This chapter also provides a glossary, important conclusions, specific recommendations regarding future coastal research, and a robust, current set of references.

Item does not contain fulltext, Siglecs are a family of sialic acid-binding receptors expressed by cells of the immune system and a few other cell types capable of modulating immune cell functions upon recognition of sialoglycan ligands. While human Siglecs primarily bind to sialic acid residues on diverse types of glycoproteins and glycolipids that constitute the sialome, their fine binding specificities for elaborated complex glycan structures and the contribution of the glycoconjugate and protein context for recognition of sialoglycans at the cell surface are not fully elucidated. Here, we generated a library of isogenic human HEK293 cells with combinatorial loss/gain of individual sialyltransferase genes and the introduction of sulfotransferases for display of the human sialome and to dissect Siglec interactions in the natural context of glycoconjugates at the cell surface. We found that Siglec-4/7/15 all have distinct binding preferences for sialylated GalNAc-type O-glycans but exhibit selectivity for patterns of O-glycans as presented on distinct protein sequences. We discovered that the sulfotransferase CHST1 drives sialoglycan binding of Siglec-3/8/7/15 and that sulfation can impact the preferences for binding to O-glycan patterns. In particular, the branched Neu5Acα2-3(6-O-sulfo)Galβ1-4GlcNAc (6'-Su-SLacNAc) epitope was discovered as the binding epitope for Siglec-3 (CD33) implicated in late-onset Alzheimer's disease. The cell-based display of the human sialome provides a versatile discovery platform that enables dissection of the genetic and biosynthetic basis for the Siglec glycan interactome and other sialic acid-binding proteins.

3. Championing survival: connecting the unknown network of responders to address out-of-hospital cardiac arrest

Abstract

Early intervention for out-of-hospital cardiac arrest (OHCA) presents a challenge for Emergency Medical Services (EMS) across Europe. Strategies designed to address this include education and training initiatives for citizens and building CPR skills capacity and awareness amongst health care professionals. However, there is a need to improve access to volunteer first responders who can commence CPR and defibrillate before the arrival of EMS. In the UK, initiatives such GoodSAM have integrated crowdsourcing technology with ambulance services to allow them autonomy in alerting responders to OHCAs which is parallel to an EMS dispatch. These services are building capacity to improve the initial 'call for help' and time to commence CPR and defibrillation if indicated. The next step is to identify and implement appropriate methods for public engagement, involvement and eventual networking of resources with statutory bodies such as local EMS. As crowdsourcing volunteer responders is at an early stage, there is a need to determine whether crowdsourcing is associated with patient outcomes, what its impact is on those responding to OHCA, whether it facilitates or impedes current services, and whether it is a safe and cost effective way to involve citizens to intervene in the community during cardiac arrest or other medical emergencies? Addressing such issues is likely to provide further insight into the role and effectiveness of new technologies and their potential impact on the wider community.

Abstract

Satellite telemetry is an increasingly utilized technology in wildlife research, and current devices can track individual animal movements at unprecedented spatial and temporal resolutions. However, as we enter the golden age of satellite telemetry, we need an in-depth understanding of the main technological, species-specific and environmental factors that determine the success and failure of satellite tracking devices across species and habitats. Here, we assess the relative influence of such factors on the ability of satellite telemetry units to provide the expected amount and quality of data by analyzing data from over 3,000 devices deployed on 62 terrestrial species in 167 projects worldwide. We evaluate the success rate in obtaining GPS fixes as well as in transferring these fixes to the user and we evaluate failure rates. Average fix success and data transfer rates were high and were generally better predicted by species and unit characteristics, while environmental characteristics influenced the variability of performance. However, 48% of the unit deployments ended prematurely, half of them due to technical failure. Nonetheless, this study shows that the performance of satellite telemetry applications has shown improvements over time, and based on our findings, we provide further recommendations for both users and manufacturers.

Thus, receptor expression patterns can define viral tissue tropism and might to some extent predict the susceptibility of a host to a particular virus. Previously, others and we have shown that respiratory pathogens of the genus Gammacoronavirus, including chicken infectious bronchitis virus (IBV), require specific alpha2,3-linked sialylated glycans for attachment and entry. Here, we studied determinants of binding of enterotropic avian gammacoronaviruses, including turkey coronavirus (TCoV), guineafowl coronavirus (GfCoV), and quail coronavirus (QCoV), which are evolutionarily distant from respiratory avian coronaviruses based on the viral attachment protein spike (S1). We profiled the binding of recombinantly expressed S1 proteins of TCoV, GfCoV, and QCoV to tissues of their respective hosts. Protein histochemistry showed that the tissue binding specificity of S1 proteins of turkey, quail, and guineafowl CoVs was limited to intestinal tissues of each particular host, in accordance with the reported pathogenicity of these viruses in vivo. Glycan array analyses revealed that, in contrast to the S1 protein of IBV, S1 proteins of enteric gammacoronaviruses recognize a unique set of nonsialylated type 2 poly-N-acetyl-lactosamines. Lectin histochemistry as well as tissue binding patterns of TCoV S1 further indicated that these complex N-glycans are prominently expressed on the intestinal tract of various avian species. In conclusion, our data demonstrate not only that enteric gammacoronaviruses recognize a novel glycan receptor but also that enterotropism may be correlated with the high specificity of spike proteins for such glycans expressed in the intestines of the avian host. Thus, receptor expression patterns can define viral tissue tropism and might to some extent predict the susceptibility of a host to a particular virus. Previously, others and we have shown that respiratory pathogens of the genus Gammacoronavirus, including chicken infectious bronchitis virus (IBV), require specific alpha2,3-linked sialylated glycans for attachment and entry. Here, we studied determinants of binding of enterotropic avian gammacoronaviruses, including turkey coronavirus (TCoV), guineafowl coronavirus (GfCoV), and quail coronavirus (QCoV), which are evolutionarily distant from respiratory avian coronaviruses based on the viral attachment protein spike (S1). We profiled the binding of recombinantly expressed S1 proteins of TCoV, GfCoV, and QCoV to tissues of their respective hosts. Protein histochemistry showed that the tissue binding specificity of S1 proteins of turkey, quail, and guineafowl CoVs was limited to intestinal tissues of each particular host, in accordance with the reported pathogenicity of these viruses in vivo. Glycan array analyses revealed that, in contrast to the S1 protein of IBV, S1 proteins of enteric gammacoronaviruses recognize a unique set of nonsialylated type 2 poly-N-acetyl-lactosamines. Lectin histochemistry as well as tissue binding patterns of TCoV S1 further indicated that these complex N-glycans are prominently expressed on the intestinal tract of various avian species. In conclusion, our data demonstrate not only that enteric gammacoronaviruses recognize a novel glycan receptor but also that enterotropism may be correlated with the high specificity of spike proteins for such glycans expressed in the intestines of the avian host. 12. On the analysis of Canadian Holstein dairy cow lactation curves using standard growth functions

Abstract

Six classical growth functions (monomolecular, Schumacher, Gompertz, logistic, Richards, and Morgan) were fitted to individual and average (by parity) cumulative milk production curves of Canadian Holstein dairy cows. The data analyzed consisted of approximately 91,000 daily milk yield records corresponding to 122 first, 99 second, and 92 third parity individual lactation curves. The functions were fitted using nonlinear regression procedures, and their performance was assessed using goodness-of-fit statistics (coefficient of determination, residual mean squares, Akaike information criterion, and the correlation and concordance coefficients between observed and adjusted milk yields at several days in milk). Overall, all the growth functions evaluated showed an acceptable fit to the cumulative milk production curves, with the Richards equation ranking first (smallest Akaike information criterion) followed by the Morgan equation. Differences among the functions in their goodness-of-fit were enlarged when fitted to average curves by parity, where the sigmoidal functions with a variable point of inflection (Richards and Morgan) outperformed the other 4 equations. All the functions provided satisfactory predictions of milk yield (calculated from the first derivative of the functions) at different lactation stages, from early to late lactation. The Richards and Morgan equations provided the most accurate estimates of peak yield and total milk production per 305-d lactation, whereas the least accurate estimates were obtained with the logistic equation. In conclusion, classical growth functions (especially sigmoidal functions with a variable point of inflection) proved to be feasible alternatives to fit cumulative milk production curves of dairy cows, resulting in suitable statistical performance and accurate estimates of lactation traits.

Abstract

Pandemic influenza A H1N1 (pH1N1) virus emerged in 2009. In the subsequent 4 years, it acquired several genetic changes in its hemagglutinin (HA). Mutations may be expected while virus is adapting to the human host or upon evasion from adaptive immune responses. However, pH1N1 has not displayed any major antigenic changes so far. We examined the effect of the amino acid substitutions found to be most frequently occurring in the pH1N1 HA protein before 1 April 2012 on the receptor-binding properties of the virus by using recombinant soluble HA trimers. Two changes (S186P and S188T) were shown to increase the receptor-binding avidity of HA, whereas two others (A137T and A200T) decreased binding avidity. Construction of an HA protein tree revealed the worldwide emergence of several HA variants during the past few influenza seasons. Strikingly, two major variants harbor combinations of substitutions (S186P/A137T and S188T/A200T, respectively) with opposite individual effects on binding. Stepwise reconstruction of the HA proteins of these variants demonstrated that the mutations that increase receptor-binding avidity are compensated for by the acquisition of subsequent mutations. The combination of these substitutions restored the receptor-binding properties (avidity and specificity) of these HA variants to those of the parental virus. The results strongly suggest that the HA of pH1N1 was already optimally adapted to the human host upon its emergence in April 2009. Moreover, these results are in agreement with a recent model for antigenic drift, in which influenza A virus mutants with high and low receptor-binding avidity alternate.

Abstract

Robotics is a valuable tool for engaging students in the hands-on application of science, technology, engineering, and mathematics (STEM) concepts. Robotics competitions such as FIRST LEGO League (FLL) can increase students’ interest in the STEM subjects and can foster their problem solving and teamwork skills. This paper reports on a study investigating students’ perceptions on the influence of participating in a FLL competition on their learning. The students completed questionnaires regarding their perceptions of their learning during the FLL challenge and were also interviewed to gain a deeper understanding of their questionnaire responses. The results show that the students were engaged with the FLL challenge and held positive views regarding their experience. The results also suggest that students involved with the FLL challenge improved their learning about real-world applications, problem solving, engagement, communication, and the application of the technology/engineering cycle.

The case of the Australian Southern Rock Lobster Association describes real issues faced by the Market Development Manager of a collective agri-food organization (SRL) representing all the southern rock lobster fishermen in Victoria, South Australia and Tasmania. The case deals with recent globalization issues faced by the Australian rock lobster industry: the rise of China as a vital and risky market of high-end food products; the financial crisis that affected the US starting from 2006 and the rise of sustainability issues that constrained the rock lobster supply. This case is designed for advanced BSc and MSc in agribusiness and international business. It is relevant for both for strategic management and supply chain management courses. In this case, managers, policy-makers and academics find challenging questions that are still open and can be similarly posed to other agribusiness industries worldwide attempting to compete collectively in international markets.

Commanders are often concerned they will not receive sufficient mission funding or they will not be able to adequately control their funds. Virtually every commander surveyed expressed the feeling that his knowledge and experience in the area of financial management is often limited and insufficient to meet the responsibilities of command. 20. Extracting Embedded Generalized Networks from Linear Programming Problems.

Abstract

If a linear program (LP) possesses a large generalized network (GN) submatrix, this structure can be exploited to decrease solution time. The problems of definding maximum sets of GN constraints and finding maximum embedded GN submatrices are shown to be NP-complete indicating that reliable, efficient solution of these problems is difficult. Therefore, efficient heuristic algorithms are developed for identifying such structure and are tested on a selection of twenty-three real-world problems. The best of four algorithms for identifying GN constraint sets finds a set which is maximum in twelve cases and average 99.1% of maximum. On average, the GN constraints identified comprise more than 62.3% of the total constraints in these problems. The algorithm for identifying embedded GN submatrices finds submatrices whose sizes, rows plus columns, average 96.8% of an LP upper bound. Over 91.3% of the total constraint matrix was identified as a GN submatrix in these problems, on average. Abstract

A complete, unified description is given of the design, implementation and use of a family of very fast and efficient large scale minimum-cost (primal simplex) network programs. The class of capacitated generalized transshipment problems solved includes the capacitated and uncapacitated generalized transportation problems and the continuous generalized assignment problem, as well as the pure network flow models which are specializations of these problems. These formulations are used for a large number of diverse applications to determine how (or at what rate) flows through the arcs of a network can minimize total shipment costs. A generalized network problem can also be viewed as a linear program with at most two non-zero entries in each column of the constraint matrix; this property is exploited in the mathematical presentation with special emphasis on data structures for basis representation, basis manipulation, and pricing mechanisms. A literature review accompanies computational testing of a promising ideas, and extensive experimentation is reported which has produced GENNET, an extremely efficient family of generalized network systems. A unifying concept for large-scale linear programming is developed. This approach called 'factorization' allows one to isolate the effect of different types of constraints and variables in the algebraic representation of the tableau. Two different factorizations based on a double representation of the basic tableau are presented. The use of factorization to obtain efficient algorithms for generalized upper bounding and block diagonal constraints as well as the general network problem is discussed. Abstract

A document from an extensive collection spanning four generations of the Woodward family that operated merchant pig iron companies in West Virginia and Alabama. The collection begins with Stimpson Harvey Woodward (S. H. Woodward), a native of Massachusetts, who moved from Pittsburgh to Wheeling, West Virginia in 1852. He had interests in an iron company as early as 1852 in West Virginia and began Alabama operations in 1869. The family business continued in Alabama until the death of S. H. Woodward's great-grandson in 1965.
