The technical feasibility of gasoline and diesel range hydrocarbons production through oligomerisation of olefins, starting from biomass with the intermediary steps of gasfication, water-gas shift reaction and syngas-to-olefins synthesis was investigated, through mathematical modelling and simulation on Matlab. The model for the gasifier was based on minimisation of Gibbs free energy and its results showed that higher carbon efficiencies could be achieved at lower pressures and steam inlet, and more inlet energy, by pre-heating the gasifying agents. The water-gas shift reactor was used to increase the ratio of hydrogen to carbon monoxide from the gasifier, before entering the syngas-to-olefins process. A 1-D model was employed to determine the concentration, temperature, and pressure profiles in the reactor. High inlet pressure and temperature were shown to be beneficial, by requiring smaller reactors for the desired ratios to be reached. Experimental data from scientific literature was used for empirical modelling of the Fischer-Tropsch reactor. Partial pressure of CO and H2 amounting to 1 bar, high temperature and H2/CO showed better production of the low olefins. A reaction mechanism and accordingly, rate equations were developed and employed in a plug-flow type reactor model, calculating the concentration profile of the olefins up to C20. High pressures were favourable for the production of heavier fractions, while elevated temperatures showed to cause more cracking of heavy hydrocarbons and consequently, less conversion. Based on the results of individual reactors, an integrated process flow diagram was suggested and optimised for maximum production of low olefins to the oligomerisation reactor (C2-4). The optimisation showed overall carbon efficiency of the process to be around 20%. Plants and algae need light to drive the photosynthetic machinery. An excess of light will, however, result in damage to the photosynthetic machinery and to the rest of the organism. The surplus of energy, when exposed to light stress is converted into less harmful heat energy through a process called non-photochemical quenching. This quenching is partly dependent on the xanthophyll pool located inside the thylakoid membrane. During light stress the xanthophyll violaxanthin is converted to zeaxanthin by violaxanthin de-epoxidase (VDE), triggering the zeaxanthin dependent light quenching. VDE is located on the lumen side of the thylakoid and is activated by the reduction of pH caused by photosynthesis. The sequence of VDE has been divided into three domains, a conserved N-terminal domain rich in cysteines, a lipocalin-like domain expected to bind the substrates and a negatively charged C-terminal domain rich in glutamic acid. In this work we have constructed cysteine mutants that revealed the importance of 12 of the 13 cysteines of VDE to the activity. These 12 cysteines were found to form disulphides in a pattern giving two hairpin structures and also increased the thermal stability of VDE. The active site of VDE does not appear to be exclusively located in the cysteine rich N-terminal domain. The expression of the N-terminal domain without the rest of VDE did not show catalytic activity. The rest of VDE without the N-terminal domain was also not able to catalyse the reaction, but after mixing of these two constructs the activity returned. This shows that the N-terminal domain and the rest of VDE can fold independently and also indicates that the active site is localised in the interface between the N-terminal domain and the lipocalin-like domain. Crosslinking of monomeric VDE could localise the N-terminal domain near the opening of the lipocalin barrel, where violaxanthin is predicted to bind. Apoptotic stimuli activate and oligomerise the pro-apoptotic proteins Bak and Bax resulting in mitochondrial outer membrane permeabilisation and subsequent cell death. This thesis investigates structural transitions occurring to Bak during apoptosis. I present crystal structures of a Bak core/latch dimer and demonstrate the dissociation of the core and latch domains upon Bak activation. I provide the first high-resolution details for the core domain dimer, a subunit upon which the larger Bak oligomer builds. Cellular assays, guided by the presented crystal structures, confirm the physiological relevance of these key events in the intrinsic apoptotic pathway (Chapter 2). I also describe the first crystal structures of Bak in complex with the BH3-domain of Bim (Chapter 3). These studies complement previous work performed on Bax and support an analogous mechanism of activation and oligomerisation. Certain detergents have been reported to activate Bak in vitro. Here I demonstrate that some detergents can oligomerise Bak and/or promote hetero-complexes between Bak and the pro-survival protein Mcl-1. I describe the production of homo-oligomeric and hetero-oligomeric complexes of Bak, which may be amenable to structural studies (Chapter 4). The literature on apoptosis assumes that mouse and human Bak are analogous in structure and therefore function. Here I report structural differences between Bak homologs from these two species (Chapter 5). These differences exist at the site of ligand binding, yet it remains unclear whether they result in functional variations. These data may aid in the development of novel agonists and antagonists of Bak, and could prove fundamental to designing murine-based pre-clinical trials. The integration of transmembrane (TM)-spanning regions of many channels and ion transporters is potentially compromised by the presence of polar and charged residues required for biological function. Although the two TMs of the ATP-gated ion channel subunit P2X2 each contain charged/polar amino acids, we found that each TM is efficiently membrane inserted when it is analysed in isolation, and uncovered no evidence for cooperativity between these two TMs during P2X2 integration. However, using minimal N-glycosylation distance mapping, we find that the positioning of TM2 in newly synthesized P2X2 monomers is distinct from that seen in subunits of the high-resolution structures of assembled homologous trimers. We conclude that P2X2 monomers are initially synthesised at the endoplasmic reticulum in a distinct conformation, where the extent of the TM-spanning regions is primarily defined by the thermodynamic cost of their membrane integration at the Sec61 translocon. In this model, TM2 of P2X2 subsequently undergoes a process of positional editing within the membrane that correlates with trimerisation of the monomer, a process requiring specific polar/charged residues in both TM1 and TM2. We postulate that the assembly process offsets any energetic cost of relocating TM2, and find evidence that positional editing of TM2 in the acid-sensing ion channel (ASIC1a) is even more pronounced than that observed for P2X2. Taken together, these data further underline the potential complexities involved in accurately predicting TM domains. We propose that the orchestrated repositioning of TM segments during subunit oligomerisation plays an important role in generating the functional architecture of active ion channels, and suggest that the regulation of this underappreciated biosynthetic step may provide an elegant mechanism for maintaining ER homeostasis., AuthorCount:7

Die Bildung infektiöser Viruspartikel des humanen Zytomegalievirus (HCMV) ist ein mehr-stufiger Prozess. Sie beginnt mit der Verpackung der DNA in die Kapside im Kern, gefolgt von weiterer Reifung während des Transports durch das Zytoplasma und der abschließenden Freisetzung aus der Zelle. Im Zuge dieser Arbeit wurden zwei Proteine, die Einfluss auf die ebengenannten Prozesse haben, analysiert. Der erste Teil der Arbeit befasst sich mit der funktionellen Charakterisierung des HCMV Pro-teins pUL77. Es ist bekannt, dass das homologe Protein pUL25 in alpha-Herpesvirinae essentiell für die DNA-Verpackung ist. Zunächst konnte das Protein als Kapsid-assoziiertes strukturelles Protein identifiziert werden. Es wurden Interaktionen von pUL77 mit DNA-Verpackungs- und Kapsidproteinen gezeigt. Weiterhin wurde die DNA-Bindungsfähigkeit von pUL77 in verschiedenen „in vitro“-Experimenten untersucht. Zusammengefasst weisen unsere Ergebnisse auf eine Funktion von HCMV pUL77 bei der DNA-Verpackung hin. Im zweiten Teil der Arbeit wurde das HCMV Protein pUL71 charakterisiert, das in allen Herpesviren konserviert vorkommt, dessen Funktion jedoch nicht charakterisiert ist. Zunächst wurde das Protein als strukturelles Tegumentprotein mit “earlylate“ Expressionskinetik klassifiziert. Weiterhin wurden die subzelluläre Lokalisation sowie virale und zelluläre Interaktionspartner untersucht. Die Ergebnisse weisen auf eine Funktion von HCMV pUL71 bei der Reifung und beim Transport der Virionen im Zytoplasma hin. „In silico“-Vorhersagen zeigten ein „Leuzin Zipper“-Motiv in pUL71, das als mögliche Oligomerisationsdomäne dienen könnte. Mutationen wurden in dieses Motiv eingebracht und die resultierenden Proteine auf ihre Oligomerisationsfähigkeit mit „in vitro“-Methoden und in rekombinanten Viren untersucht. Zusammenfassend konnten wir zeigen, dass das „Leuzin Zipper“-Motiv wichtig für die Funktion von pUL71 ist und diese mit einer unbeeinträchtigten Oligomerisation des Proteins zusammen hängt., The morphogenesis of Human cytomegalovirus (HCMV) virions starts with the capsid assem-bly and DNA insertion in the nucleus followed by maturation during transport through the cytoplasm prior to release of virus progeny. In this study we are functionally characterising two proteins that are involved in those steps. The function of essential HCMV protein pUL77 is characterised in the first part of the study. HCMV pUL77 was shown to be a structural protein associated with capsids. Furthermore, our experiments demonstrated that HCMV pUL77 interacts with DNA packaging motor compo-nents and capsid proteins. The ability of HCMV pUL77 to bind double-stranded DNA was studied in “in vitro” assays designed for this study. The homologue α-Herpesvirinae protein pUL25 is described to be involved in processes connected with DNA packaging. Data ob-tained in this study demonstrates that HCMV pUL77 might serve a similar function. In the second part of the study HCMV pUL71, conserved throughout the Herpesvirus family but to date unclassified, was functionally characterised. HCMV pUL71 was defined a struc-tural tegument protein with early-late expression kinetics. We studied the sub-cellular local-isation and interactions of pUL71 with a subset of cellular and viral proteins. Thereby we could show that HCMV pUL71 function might be connected with processes of viral egress. By in silico analyses we identified a leucine zipper motif in pUL71 that might serve as a puta-tive oligomerisation domain. In order to investigate the function of the leucine zipper motif, we performed in vitro assays and investigated the alterations of the motif in the viral context. Taken together we can conclude that (i) an intact leucine zipper motif is crucial for the func-tion of pUL71 and (ii) this function is dependent upon undisturbed oligomerisation of the pro-tein.

The complex [Cp*Rh(μ‐SPh)3RhCp*]Cl was used as a starting material to synthesize various oligomeric materials of the general formula [Cp*Rh(μ‐SPh)x(μ‐Cl)3–x{Rh(μ‐SPh)3}nRhCp*] (x = 1 to 3; n = 1 to 4), which are formally formed by insertion of nRh(SPh)3 units into one μ‐Rh–SPh bond. The insertion of Ir(SPh)3 was also observed to generate theheterotrimetallic species. All complexes were observed by using HRMS‐TOF and [Cp*Rh(μ‐SPh)3Rh(μ‐SPh)3RhCp*]Cl, [3+]Cl, was characterized by using X‐ray crystallography.

In this thesis the applicability of this method to count the number of spins is tested. Furthermore, this work explored the limits, up to which PELDOR data obtained on copper(II)-nitroxide complexes can be quantitatively interpreted. Spin counting provides access to oligomerization studies – monitoring the assembly of homo- or hetero-oligomers from singly labeled compounds. The experimental calibration was performed using model systems, which contain one to four nitroxide radicals. The results show that monomers, dimers, trimers, and tetramers can be distinguished within an error of 5% in the number of spins. Moreover, a detailed analysis of the distance distributions in model complexes revealed that more than one distance can be extracted from complexes bearing several spins, as for example three different distances were resolved in a model tetramer – the other three possible distances being symmetry related. Furthermore, systems exhibiting mixtures of oligomeric states complicate the analysis of the data, because the average number of spin centers contributes nonlinearly to the signal and different relaxation behavior of the oligomers has to be treated explicitly. Experiments solving these problems are proposed in the thesis. Thus, for the first time spin counting has been experimentally calibrated using fully characterized test systems bearing up to four spins. Moreover, the behavior of mixtures was quantitatively interpreted. In addition, it has been shown that several spin-spin distances within a molecule can be extracted from a single dataset. Der Glycinrezeptor (GlyR) ist ein inhibitorischer Chloridkanal, der u.a. im Rückenmark, im Hirnstamm und in der Retina exprimiert wird und dort maßgeblich an der Prozessierung von motorischen und/oder sensorischen Signalen beteiligt ist. Als Mitglied der Superfamilie der nikotinischen Acetylcholinrezeptoren ist der GlyR ein Pentamer, dessen Untereinheiten jeweils aus einer extrazellulären N-terminalen Domäne, vier Transmembranregionen und einer großen intrazellulären Schleife bestehen. Bislang wurden vier ligandenbindende alpha-Untereinheiten (alpha 1-4) und eine für die synaptische Verankerung verantwortliche beta-Untereinheit identifiziert. Die beta-Untereinheit bildet im Gegensatz zu den alpha-Untereinheiten keine Homooligomere; sie ist nur in heterooligomeren GlyRs mit einer bisher angenommenen Stöchiometrie von 3alpha:2beta zu finden. Wie bei allen Mitgliedern der Superfamilie erfolgt die Ligandenbindung innerhalb der extrazellulären Domänen an der Kontaktstelle zwischen zwei benachbarten Untereinheiten. Um den Mechanismus der Ligandenbindung am homo- und heterooligomeren GlyR in dieser Arbeit aufzuklären, wurden Modelle der N-terminalen Domänen der alpha1- und beta-Untereinheiten basierend auf der Kristallstruktur des Acetylcholin-Bindeproteins generiert. Zur Verifizierung der Rolle aller im Modell in der Bindungstasche lokalisierten Aminosäureseitenketten wurden diese durch zielgerichtete PCR-Mutagenese substituiert, anschließend die mutierten Untereinheiten in Xenopus laevis Oozyten exprimiert und elektrophysiologisch charakterisiert. So konnten die wichtigsten Seitenketten der alpha1-Ligandenbindungstasche identifiziert werden, die an der Bindung der Agonisten Glycin und Taurin, des Antagonisten Strychnin und des Modulators 3alpha-(3’-Methoxybenzoyloxy)nortropan beteiligt sind. Die anschließende Untersuchung des heterooligomeren alpha1beta-GlyRs konnte zum ersten Mal eine direkte Beteiligung der beta-Untereinheit an der Ligandenbindung aufzeigen, wobei die

Der Glycinrezeptor (GlyR) ist ein inhibitorischer Chloridkanal, der u.a. im Rückenmark, im Hirnstamm und in der Retina exprimiert wird und dort maßgeblich an der Prozessierung von motorischen und/oder sensorischen Signalen beteiligt ist. Als Mitglied der Superfamilie der nikotinischen Acetylcholinrezeptoren ist der GlyR ein Pentamer, dessen Untereinheiten jeweils aus einer extrazellulären N-terminalen Domäne, vier Transmembranregionen und einer großen intrazellulären Schleife bestehen. Bislang wurden vier ligandenbindende alpha-Untereinheiten (alpha 1-4) und eine für die synaptische Verankerung verantwortliche beta-Untereinheit identifiziert. Die beta-Untereinheit bildet im Gegensatz zu den alpha-Untereinheiten keine Homooligomere; sie ist nur in heterooligomeren GlyRs mit einer bisher angenommenen Stöchiometrie von 3alpha:2beta zu finden. Wie bei allen Mitgliedern der Superfamilie erfolgt die Ligandenbindung innerhalb der extrazellulären Domänen an der Kontaktstelle zwischen zwei benachbarten Untereinheiten. Um den Mechanismus der Ligandenbindung am homo- und heterooligomeren GlyR in dieser Arbeit aufzuklären, wurden Modelle der N-terminalen Domänen der alpha1- und beta-Untereinheiten basierend auf der Kristallstruktur des Acetylcholin-Bindeproteins generiert. Zur Verifizierung der Rolle aller im Modell in der Bindungstasche lokalisierten Aminosäureseitenketten wurden diese durch zielgerichtete PCR-Mutagenese substituiert, anschließend die mutierten Untereinheiten in Xenopus laevis Oozyten exprimiert und elektrophysiologisch charakterisiert. So konnten die wichtigsten Seitenketten der alpha1-Ligandenbindungstasche identifiziert werden, die an der Bindung der Agonisten Glycin und Taurin, des Antagonisten Strychnin und des Modulators 3alpha-(3’-Methoxybenzoyloxy)nortropan beteiligt sind. Die anschließende Untersuchung des heterooligomeren alpha1beta-GlyRs konnte zum ersten Mal eine direkte Beteiligung der beta-Untereinheit an der Ligandenbindung aufzeigen, wobei die

The new efficient and environmentally safe methods for preparation of various classes of organic and organoelement compounds, including organonickel sigma-complexes and organophosphorus compounds bearing P-C bonds have been created using the electrochemical methods. The synthetic application of the elaborated techniques towards the process of formation of new carbon-carbon, carbon-metal and carbon-phosphorus bonds are discussed. The mechanisms of the proposed processes and the nature of the formed in the overall electrochemical process intermediates are disclosed. The elaborated methods operated in the principals of "green chemistry" can be considered as an efficient alternative to some classical methods for preparation of active catalysts, biologically active molecules and new polynuclear complexes displaying practically useful properties.

This multidrug efflux capability, together with a broad tissue distribution in the body, means that ABCG2 exerts a range of effects on normal physiology such as kidney urate transport, as well as contributing towards the pharmacokinetic profiles of many exogenous drugs. The primary sequence of ABCG2 contains only half the number of domains required for a functioning ABC transporter and so it must oligomerise in order to function, yet its oligomeric state in intact cell membranes remains uncharacterized. We have analysed ABCG2 in living cell membranes using a combination of fluorescence correlation spectroscopy, photon counting histogram analysis, and stepwise photobleaching to demonstrate a predominantly tetrameric structure for ABCG2 in the presence or absence of transport substrates. These results provide the essential basis for exploring pharmacological manipulation of oligomeric state as a strategy to modulate ABCG2 activity in future selective therapeutics.

We have analysed ABCG2 in living cell membranes using a combination of fluorescence correlation spectroscopy, photon counting histogram analysis, and stepwise photobleaching to demonstrate a predominantly tetrameric structure for ABCG2 in the presence or absence of transport substrates. These results provide the essential basis for exploring pharmacological manipulation of oligomeric state as a strategy to modulate ABCG2 activity in future selective therapeutics.
