Abstract: Background: Mortality among people with HIV declined with the introduction of combination antiretroviral therapy. We investigated trends over time in all-cause and cause-specific mortality in people with HIV from 1999-2020. Methods: Data were collected from the D:A:D cohort from 1999 through January 2015 and RESPOND from October 2017 through 2020. Age-standardized all-cause and cause-specific mortality rates, classified using Coding Causes of Death in HIV (CoDe), were calculated. Poisson regression models were used to assess mortality trends over time. Results: Among 55716 participants followed for a median of 6 years (IQR 3-11), 5263 participants died (crude mortality rate [MR] 13.7/1000 PYFU; 95%CI 13.4-14.1). Changing patterns of mortality were observed with AIDS as the most common cause of death between 1999- 2009 (n = 952, MR 4.2/1000 PYFU; 95%CI 4.0-4.5) and non-AIDS defining malignancy (NADM) from 2010 -2020 (n = 444, MR 2.8/1000 PYFU; 95%CI 2.5-3.1). In multivariable analysis, all-cause mortality declined over time (adjusted mortality rate ratio [aMRR] 0.97 per year; 95%CI 0.96, 0.98), mostly from 1999 through 2010 (aMRR 0.96 per year; 95%CI 0.95-0.97), and with no decline shown from 2011 through 2020 (aMRR 1·00 per year; 95%CI 0·96-1·05). Mortality due all known causes except NADM also declined over the entire follow-up period. Conclusion: Mortality among people with HIV in the D:A:D and/or RESPOND cohorts decreased between 1999 and 2009 and was stable over the period from 2010 through 2020. The decline in mortality rates was not fully explained by improvements in immunologic-virologic status or other risk factors.

2. Uncertainty quantification for iterative algorithms in linear models with application to early stopping

Abstract: This paper investigates the iterates $\hbb^1,\dots,\hbb^T$ obtained from iterative algorithms in high-dimensional linear regression problems, in the regime where the feature dimension $p$ is comparable with the sample size $n$, i.e., $p \asymp n$. The analysis and proposed estimators are applicable to Gradient Descent (GD), proximal GD and their accelerated variants such as Fast Iterative Soft-Thresholding (FISTA). The paper proposes novel estimators for the generalization error of the iterate $\hbb^t$ for any fixed iteration $t$ along the trajectory. These estimators are proved to be $\sqrt n$-consistent under Gaussian designs. Applications to early-stopping are provided: when the generalization error of the iterates is a U-shape function of the iteration $t$, the estimates allow to select from the data an iteration $\hat t$ that achieves the smallest generalization error along the trajectory. Additionally, we provide a technique for developing debiasing corrections and valid confidence intervals for the components of the true coefficient vector from the iterate $\hbb^t$ at any finite iteration $t$. Extensive simulations on synthetic data illustrate the theoretical results.

3. How do supernova remnants cool? -- I. Morphology, optical emission lines, and shocks

Abstract: Supernovae (SNe) inject $\sim 10^{51}$ erg in the interstellar medium, thereby shocking and heating the gas. A substantial fraction of this energy is later lost via radiative cooling. We present a post-processing module for the FLASH code to calculate the cooling radiation from shock-heated gas using collisional excitation data from MAPPINGS V. When applying this tool to a simulated SN remnant (SNR), we find that most energy is emitted in the EUV. However, optical emission lines ($[$O III$]$, $[$N II$]$, $[$S II$]$, H${\alpha}$, H${\beta}$) are usually best observable. Our shock detection scheme shows that [S II] and [N II] emissions arise from the thin shell surrounding the SNR, while [O III], H$\rm \alpha$, and H$\rm \beta$ originate from the volume-filling hot gas inside the SNR bubble. We find that the optical emission lines are affected by the SNR's complex structure and its projection onto the plane of the sky because the escaping line luminosity can be reduced by 10 -- 80\% due to absorption along the line-of-sight. Additionally, the subtraction of contaminating background radiation is required for the correct classification of an SNR on the oxygen or sulphur BPT diagrams. Abstract: Engineering matrices for cell therapy requires design criteria that include the ability of these materials to support, protect and enhance cellular behavior in vivo. The chemical and mechanical formulation of the biomaterials can influence not only target cell phenotype but also cellular differentiation. In this study, we have demonstrated the effect of a gelatin (Gtn)—hyaluronic acid (HA) hydrogel on human retinal progenitor cells (hRPCs) and show that by altering the mechanical properties of the materials, cellular behavior is altered as well. We have created an interpenetrating network polymer capable of encapsulating hRPCs. By manipulating the stiffness of the hydrogel, the differentiation potential of the hRPCs was controlled. Interpenetrating network 75 (IPN 75; 75% HA) allowed higher expression of rod photoreceptor markers, whereas cone photoreceptor marker expression was found to be higher in IPN 50. In vivo testing of these living matrices performed in Long–Evans rats showed higher levels of rod photoreceptor marker expression when IPN 75 was injected versus IPN 50. These biomaterials mimic biological cues that are required to simulate the dynamic complexity of natural retinal ECM. These hydrogels can be used as a vehicle for cell delivery in vivo as well as for expansion and differentiation in an in vitro 3D system in a highly reproducible manner.

5. Using homosoils for quantitative extrapolation of soil mapping models

Abstract: Since the early 2000s, digital soil maps have been successfully used for various applications, including precision agriculture, environmental assessments and land use management. Globally, however, there are large disparities in the availability of soil data on which digital soil mapping (DSM) models can be fitted. Several studies attempted to transfer a DSM model fitted from an area with a well-developed soil database to map the soil in areas with low sampling density. This usually is a challenging task because two areas have hardly ever the same soil-forming factors in two different regions of the world. In this study, we aim to determine whether finding homosoils (i.e., locations sharing similar soil-forming factors) can help transferring soil information by means of a DSM model extrapolation. We hypothesize that within areas in the world considered as homosoils, one can leverage on areas with high sampling density and fit a DSM model, which can then be extrapolated geographically to an area with little or no data. We collected publicly available soil data for clay, silt, sand, organic carbon (OC), pH and total nitrogen (N) within our study area in Mali, West Africa and its homosoils. We fitted a regression tree model between the soil properties and environmental covariates of the homosoils, and applied this model to our study area in Mali. Several calibration and validation strategies were explored. We also compared our approach with existing maps made at a global and a continental scale. We concluded that geographic model extrapolation within homosoils was possible, but that model accuracy dramatically improved when local data were included in the calibration dataset. The maps produced from models fitted with data from homosoils were more accurate than existing products for this study area, for three (silt, sand, pH) out of six soil properties. Abstract: Kidney Precision Medicine Project (KPMP) is building a spatially specified human kidney tissue atlas in health and disease with single-cell resolution. Here, we describe the construction of an integrated reference map of cells, pathways, and genes using unaffected regions of nephrectomy tissues and undiseased human biopsies from 56 adult subjects. We use single-cell/nucleus transcriptomics, subsegmental laser microdissection transcriptomics and proteomics, near-single-cell proteomics, 3D and CODEX imaging, and spatial metabolomics to hierarchically identify genes, pathways, and cells. Integrated data from these different technologies coherently identify cell types/subtypes within different nephron segments and the interstitium. These profiles describe cell-level functional organization of the kidney following its physiological functions and link cell subtypes to genes, proteins, metabolites, and pathways. They further show that messenger RNA levels along the nephron are congruent with the subsegmental physiological activity. This reference atlas provides a framework for the classification of kidney disease when multiple molecular mechanisms underlie convergent clinical phenotypes.

Abstract: Overcoming obstacles to commercialization of algal-based processes for biofuels and co-products requires not just piecemeal incremental improvements, but rather a comprehensive and fundamental re-consideration starting with the selected algae and its associated cultivation, harvesting, biomass conversion, and refinement. A novel two-stage process designed to address challenges of mass outdoor microalgal cultivation for biofuels and co-products was previously demonstrated using an oleaginous, haloalkaline-tolerant, and multi-trophic green Chlorella vulgaris. ALP2 from a soda lake. This involved cultivating the microalgae in a fermenter heterotrophically or photobioreactor mixotrophically (first-stage) to rapidly obtain high cell densities and inoculate an open-pond phototrophic culture (second-stage) featuring high levels of NaHCO3, pH, and salinity. An improved two-stage cultivation that instead sustainably used as more cheap and sustainable inputs the organic carbon, nitrogen, and phosphorous from fractionation of waste was here demonstrated in a small-scale biorefinery process. The first cultivation stage consisted of two simultaneous batch flask cultures featuring (1) mixotrophic cell productivity of 7.25 × 107 cells mL−1 day−1 on BG-110 medium supplemented with 1.587 g L−1 urea and an enzymatic hydrolysate of pre-treated (torrefaction + grinding + ozonolysis + soaking ammonia) wheat-straw that corresponded to 10 g L−1 glucose, and (2) mixotrophic cell productivity of 2.25 × 107 cells mL−1 day−1 on BG-110 medium supplemented with 1.587 g L−1 urea and a purified and de-toxified condensate of pre-treated (torrefaction + grinding) wheat straw that corresponded to 0.350 g L−1 of potassium acetate. 12. Controlling growth factor diffusion by modulating water content in injectable hydrogels

Abstract: Recent advancements in the delivery of therapeutics for retinal diseases include the development of injectable hydrogels, networks of one or more hydrophilic polymers that contain a high-volume fraction of water. These systems are of particular interest due to their biocompatibility, permeability to water-soluble metabolites, and function as minimally invasive injectable delivery vehicles. Recently, hydrogels for ophthalmic applications have been developed that display a controlled release of factors necessary for cellular survival and proliferation. Understanding the relationship between the volume water fraction and the physical, chemical, and diffusion properties of the hydrogel scaffold could aid in the improvement of existing drug delivery treatments for retinal regeneration. In this study, we compared the diffusion and release of human epidermal growth factor (hEGF) encapsulated in different injectable homogenous and heterogenous hydrogels, namely gelatin-hydroxyphenyl propionic acid (Gtn-HPA) and hyaluronic acid-tyramine (HA-Tyr)-based hydrogels. These experimental results were compared with the measured stiffness and water content of these hydrogels and applied to different diffusion theories of polymers to determine the model of best fit. We find that the normalized diffusion and release of hEGF increases with free water content in injectable hydrogels: ranging from 0.176 at 41% free water in HA-Tyr to 0.2 at 53% free water in Gtn-HPA, whereas it decreases with hydrogel stiffness: 600 Pa for Gtn-HPA and 1440 Pa for HA-Tyr. Further, we compared our experimental data with theoretical diffusion models. We found that homogeneous theoretical models, notably the hydrodynamic model (giving a normalized diffusion close to 0.2), provide the most suitable explanation for the measured solute diffusion coefficient. Abstract: AbstractBiomaterial-based cell replacement approaches to regenerative medicine are emerging as promising treatments for a wide array of profound clinical problems. Here we report an interpenetrating polymer network (IPN) composed of gelatin-hydroxyphenyl propionic acid and hyaluronic acid tyramine that is able to enhance intravitreal retinal cell therapy. By tuning our bioinspired hydrogel to mimic the vitreous chemical composition and mechanical characteristics we were able to improve in vitro and in vivo viability of human retinal ganglion cells (hRGC) incorporated into the IPN. In vivo vitreal injections of cell-bearing IPN in rats showed extensive attachment to the inner limiting membrane of the retina, improving with hydrogels stiffness. Engrafted hRGC displayed signs of regenerating processes along the optic nerve. Of note was the decrease in the immune cell response to hRGC delivered in the gel. The findings compel further translation of the gelatin-hyaluronic acid IPN for intravitreal cell therapy.

15. Impact of the Revisit of Thermal Infrared Remote Sensing Observations on Evapotranspiration Uncertainty—A Sensitivity Study Using AmeriFlux Data

Abstract: Thermal infrared remote sensing observations have been widely used to provide useful information on surface energy and water stress for estimating evapotranspiration (ET). However, the revisit time of current high spatial resolution (<100 m) thermal infrared remote sensing systems, sixteen days for Landsat for example, can be insufficient to reliably derive ET information for water resources management. We used in situ ET measurements from multiple Ameriflux sites to (1) evaluate different scaling methods that are commonly used to derive daytime ET estimates from time-of-day observations; and (2) quantify the impact of different revisit times on ET estimates at monthly and seasonal time scales. The scaling method based on a constant evaporative ratio between ET and the top-of-atmosphere solar radiation provided slightly better results than methods using the available energy, the surface solar radiation or the potential ET as scaling reference fluxes. On average, revisit time periods of 2, 4, 8 and 16 days resulted in ET uncertainties of 0.37, 0.55, 0.73 and 0.90 mm per day in summer, which represented 13%, 19%, 23% and 31% of the monthly average ET calculated using the one-day revisit dataset. The capability of a system to capture rapid changes in ET was significantly reduced for return periods higher than eight days. The impact of the revisit on ET depended mainly on the land cover type and seasonal climate, and was higher over areas with high ET. We did not observe significant and systematic differences between the impacts of the revisit on monthly ET estimates that are based on morning or afternoon observations. We found that four-day revisit scenarios provided a significant improvement in temporal sampling to monitor surface ET reducing by around 40% the uncertainty of ET products derived from a 16-day revisit system, such as Landsat for instance.

Abstract: Breast cancer (BC) risk for BRCA1 and BRCA2 mutation carriers varies by genetic and familial factors. About 50 common variants have been shown to modify BC risk for mutation carriers. All but three, were identified in general population studies. Other mutation carrier-specific susceptibility variants may exist but studies of mutation carriers have so far been underpowered. We conduct a novel case-only genome-wide association study comparing genotype frequencies between 60,212 general population BC cases and 13,007 cases with BRCA1 or BRCA2 mutations. We identify robust novel associations for 2 variants with BC for BRCA1 and 3 for BRCA2 mutation carriers, P < 10−8, at 5 loci, which are not associated with risk in the general population. They include rs60882887 at 11p11.2 where MADD, SP11 and EIF1, genes previously implicated in BC biology, are predicted as potential targets. These findings will contribute towards customising BC polygenic risk scores for BRCA1 and BRCA2 mutation carriers.

Abstract: The "System of Rice Intensification"(SRI) represents a sustainable alternative to improve household yields and incomes. This study aimed to evaluate the yields and the profitability of the SRI and the Traditional Practices (TP) in Ziguinchor district, Senegal. A directed sampling based on criteria for selecting the system used and the cultivated varieties common to both systems was applied to collect the yield parameters and yields of paddy rice. Thus, 18 producers in the Badiate, Essyl, and Fanda sites were selected, nine per system and four 1 m2 yield squares were installed in each selected producer plot. A total of 72 yield squares, 36 per system, were installed, and an individual questionnaire was randomly administered to 55 producers using at least one of the systems to collect data on rice production and costs. The yield parameters including the number of fertile tillers per m2, the number of spikes, the weight of the 1000 grains are significantly higher (p<0.05) in the SRI including yield compared to the traditional system. Transplanting density and plant duration are higher in TP (26±5.6 plants/m2 and 26 days) than in SRI (16±0.4 plants/m2 and 16 days). The lower the transplanting density, the higher the yield parameters and the yield. The economic profitability, determined based on the benefit/cost ratio, is higher in SRI (1.5) than in TP (1.2). The SRI required a lot of technicality in its implementation and generated more cost of production. However, SRI was more productive and economically more profitable than the traditional system.

Abstract: Breast cancer (BC) risk for BRCA1 and BRCA2 mutation carriers varies by genetic and familial factors. About 50 common variants have been shown to modify BC risk for mutation carriers. All but three, were identified in general population studies. Other mutation carrier-specific susceptibility variants may exist but studies of mutation carriers have so far been underpowered. We conduct a novel case-only genome-wide association study comparing genotype frequencies between 60,212 general population BC cases and 13,007 cases with BRCA1 or BRCA2 mutations. We identify robust novel associations for 2 variants with BC for BRCA1 and 3 for BRCA2 mutation carriers, P < 10-8, at 5 loci, which are not associated with risk in the general population. They include rs60882887 at 11p11.2 where MADD, SP11 and EIF1, genes previously implicated in BC biology, are predicted as potential targets. These findings will contribute towards customising BC polygenic risk scores for BRCA1 and BRCA2 mutation carriers.Copyright © 2021, The Author(s).

Abstract: Breast cancer (BC) risk for BRCA1 and BRCA2 mutation carriers varies by genetic and familial factors. About 50 common variants have been shown to modify BC risk for mutation carriers. All but three, were identified in general population studies. Other mutation carrier-specific susceptibility variants may exist but studies of mutation carriers have so far been underpowered. We conduct a novel case-only genome-wide association study comparing genotype frequencies between 60,212 general population BC cases and 13,007 cases with BRCA1 or BRCA2 mutations. We identify robust novel associations for 2 variants with BC for BRCA1 and 3 for BRCA2 mutation carriers, P < 10-8, at 5 loci, which are not associated with risk in the general population. They include rs60882887 at 11p11.2 where MADD, SP11 and EIF1, genes previously implicated in BC biology, are predicted as potential targets. These findings will contribute towards customising BC polygenic risk scores for BRCA1 and BRCA2 mutation carriers.Copyright © 2021, The Author(s).

24. Politically Feasible Reforms of Nonlinear Tax Systems

Abstract: We study reforms of nonlinear income tax systems from a political economy perspective. We present a median voter theorem for monotonic tax reforms, reforms so that the change in the tax burden is a monotonic function of income. We also provide an empirical analysis of tax reforms, with a focus on the United States. We show that past reforms have, by and large, been monotonic. We also show that support by the median voter was aligned with majority support in the population. Finally, we develop sufficient statistics that enable to test whether a given tax system admits a politically feasible reform.

Abstract: Chronic kidney disease (CKD) and acute kidney injury (AKI) are common, heterogeneous, and morbid diseases. Mechanistic characterization of CKD and AKI in patients may facilitate a precision-medicine approach to prevention, diagnosis, and treatment. The Kidney Precision Medicine Project aims to ethically and safely obtain kidney biopsies from participants with CKD or AKI, create a reference kidney atlas, and characterize disease subgroups to stratify patients based on molecular features of disease, clinical characteristics, and associated outcomes. An additional aim is to identify critical cells, pathways, and targets for novel therapies and preventive strategies. This project is a multicenter prospective cohort study of adults with CKD or AKI who undergo a protocol kidney biopsy for research purposes. This investigation focuses on kidney diseases that are most prevalent and therefore substantially burden the public health, including CKD attributed to diabetes or hypertension and AKI attributed to ischemic and toxic injuries. Reference kidney tissues (for example, living-donor kidney biopsies) will also be evaluated. Traditional and digital pathology will be combined with transcriptomic, proteomic, and metabolomic analysis of the kidney tissue as well as deep clinical phenotyping for supervised and unsupervised subgroup analysis and systems biology analysis. Participants will be followed prospectively for 10 years to ascertain clinical outcomes. Cell types, locations, and functions will be characterized in health and disease in an open, searchable, online kidney tissue atlas. All data from the Kidney Precision Medicine Project will be made readily available for broad use by scientists, clinicians, and patients.

28. Politically Feasible Reforms of Nonlinear Tax Systems

Abstract: We study reforms of nonlinear income tax systems from a political economy perspective. We present a median voter theorem for monotonic tax reforms, reforms so that the change in the tax burden is a monotonic function of income. We also provide an empirical analysis of tax reforms, with a focus on the United States. We show that past reforms have, by and large, been monotonic. We also show that support by the median voter was aligned with majority support in the population. Finally, we develop sufficient statistics that enable to test whether a given tax system admits a politically feasible reform.

Abstract: Fluorescence lifetime imaging microscopy (FLIM) systems are limited by their slow processing speed, low signal-to-noise ratio (SNR), and expensive and challenging hardware setups. In this work, we demonstrate applying a denoising convolutional network to improve FLIM SNR. The network will be integrated with an instant FLIM system with fast data acquisition based on analog signal processing, high SNR using high-efficiency pulse-modulation, and cost-effective implementation utilizing off-the-shelf radio-frequency components. Our instant FLIM system simultaneously provides the intensity, lifetime, and phasor plots \textit{in vivo} and \textit{ex vivo}. By integrating image denoising using the trained deep learning model on the FLIM data, provide accurate FLIM phasor measurements are obtained. The enhanced phasor is then passed through the K-means clustering segmentation method, an unbiased and unsupervised machine learning technique to separate different fluorophores accurately. Our experimental \textit{in vivo} mouse kidney results indicate that introducing the deep learning image denoising model before the segmentation effectively removes the noise in the phasor compared to existing methods and provides clearer segments. Hence, the proposed deep learning-based workflow provides fast and accurate automatic segmentation of fluorescence images using instant FLIM. The denoising operation is effective for the segmentation if the FLIM measurements are noisy. Abstract: Recent years have seen a decline in herbage production and tiller populations in New Zealand's perennial ryegrass (Lolium perenne) dairy pastures. One hypothesis is that modern genotypes are less suited to the warmer, drier weather experienced under changing climate patterns. In this study, a combination of long‐term trial data (2011–2017) and a process‐based pasture model (BASGRA) was used to explore the causes and possible mitigation of the observed production and population loss at three sites (dryland sites in Northland and Waikato and an irrigated site in Canterbury). Bayesian calibration was used to identify the model parameter sets that were consistent with the trial data and to identify differences in plant morphology and responses between sites. The model successfully simulated the observed differences in tiller numbers between the dryland sites, where populations and production declined rapidly after the second year and the irrigated site where populations and production were maintained. Analysis of the model calibrations along with preliminary scenario simulations suggests that increased tiller mortality associated with drought was the main cause of persistence failure at the dryland sites and that decreasing grazing pressure or breeding for tolerance to higher temperatures may not be successful in preventing this.

37. Low-oxygen and knock-out serum maintain stemness in human retinal progenitor cells

Abstract: Using stem and progenitor cells to treat retinal disorders holds great promise. Using defined culture conditions to maintain the desires phenotype is of utmost clinical importance. We cultured human retinal progenitor cells (hRPCs) in different conditions: such as normoxia (20% oxygen), and hypoxia (5% oxygen) with and without knock-out serum replacement (KOSR) to evaluate its effect on these cells. KOSR is known nutrient supplement often used to replace bovine serum for culturing embryonic or pluripotent stem cells, especially those destined for clinical applications. The purpose of this study was to identify the impact of different environmental and chemical cues to determine if this alters the fate of these cells. Our results indicate that cells cultured with or without KOSR do not show significant differences in viability, but that the oxygen tension can significantly change their viability (higher in hypoxia than normoxia). However, cells with KOSR in hypoxia condition expressed significantly higher stemness markers such as C-myc and Oct4 (31.20% and 13.44% respectively) in comparison to hRPCs cultured in KOSR at normoxia (12.07% and 4.05%). Furthermore, levels of markers for retinal commitment such as rhodopsin were significantly lower in the KOSR supplemented cells in hypoxia culture compared to normoxia. KOSR is known to improve proliferation and maintain stemness of embryonic cells and our experiments suggest that hRPCs maintain their proliferation and stemness characteristics in hypoxia with KOSR supplement. Normoxia, however, results in mature cell marker expression, suggesting a profound effect of oxygen tension on these cells.

Abstract: High serum concentrations of kidney-derived protein uromodulin [Tamm-Horsfall protein (THP)] have recently been shown to be independently associated with low mortality in both older adults and cardiac patients, but the underlying mechanism remains unclear. Here, we show that THP inhibits the generation of reactive oxygen species (ROS) both in the kidney and systemically. Consistent with this experimental data, the concentration of circulating THP in patients with surgery-induced acute kidney injury (AKI) correlated with systemic oxidative damage. THP in the serum dropped after AKI and was associated with an increase in systemic ROS. The increase in oxidant injury correlated with postsurgical mortality and need for dialysis. Mechanistically, THP inhibited the activation of the transient receptor potential cation channel, subfamily M, member 2 (TRPM2) channel. Furthermore, inhibition of TRPM2 in vivo in a mouse model mitigated the systemic increase in ROS during AKI and THP deficiency. Our results suggest that THP is a key regulator of systemic oxidative stress by suppressing TRPM2 activity, and our findings might help explain how circulating THP deficiency is linked with poor outcomes and increased mortality.

Abstract: The land is of particular importance to the human being, not only because it is our, as well as terrestrial biomes’, habitat, but the land surface also plays a unique role in the Earth system. For example, it regulates the climate through exchange of matter, energy, and momentum with the atmosphere [1]. Data on the status and dynamics of the land surface variables are essential for understanding the land surface processes and entangling interactions between the land and other Earth system components. Since the emergence of remote sensing, the land surface is among its key study domains. The quantitative remote sensing system does not directly measure land surface parameters of interest. Instead, the signature remote sensors receive is electromagnetic radiation reflected, scattered, and emitted from both the surface and the atmosphere. The inversion algorithm is needed to obtain land surface parameters from remotely sensed data. It is not a trivia task to reliably retrieve land surface parameters since the remote sensing signature is a function of not only the variable of interest but also many other atmosphere and surface characteristics. Multifaceted aspects of the remote sensing data, such as the temporal, spectral, spatial, polarized information, as well as ancillary and prior knowledge, are typically used in a synthetic way to improve the quality of land parameter retrievals [2]. Numerous parameters regarding the land surface properties can now be estimated with the help of remote sensing, to name a few, surface cover type, snow cover and amount, surface altitude, surface radiative fluxes, biophysical parameters, biochemical variables, vegetation structure, and many other variables. Abstract: Marine transform faults and associated fracture zones (MTFFZs) cover vast stretches of the ocean floor, where they play a key role in plate tectonics, accommodating the lateral movement of tectonic plates and allowing connections between ridges and trenches. Together with the continental counterparts of MTFFZs, these structures also pose a risk to human societies as they can generate high magnitude earthquakes and trigger tsunamis. Historical examples are the Sumatra-Wharton Basin Earthquake in 2012 (M8.6) and the Atlantic Gloria Fault Earthquake in 1941 (M8.4). Earthquakes at MTFFZs furthermore open and sustain pathways for fluid flow triggering reactions with the host rocks that may permanently change the rheological properties of the oceanic lithosphere. In fact, they may act as conduits mediating vertical fluid flow and leading to elemental exchanges between Earth’s mantle and overlying sediments. Chemicals transported upward in MTFFZs include energy substrates, such as H2 and volatile hydrocarbons, which then sustain chemosynthetic, microbial ecosystems at and below the seafloor. Moreover, up- or downwelling of fluids within the complex system of fractures and seismogenic faults along MTFFZs could modify earthquake cycles and/or serve as “detectors” for changes in the stress state during interseismic phases. Despite their likely global importance, the large areas where transform faults and fracture zones occur are still underexplored, as are the coupling mechanisms between seismic activity, fluid flow, and life. This manuscript provides an interdisciplinary review and synthesis of scientific progress at or related to MTFFZs and specifies approaches and strategies to deepen the understanding of processes that trigger, maintain, and control fluid flow at MTFFZs

Abstract: Agricultural intensification and efficient use and targeting of fertilizer inputs on smallholder farms is key to sustainably improve food security. The objective of this paper is to demonstrate how high-resolution satellite and unmanned aerial vehicle (UAV) images can be used to assess the spatial variability of yield, and yield response to fertilizer. The study included 48 and 50 smallholder fields monitored during the 2014 and 2015 cropping seasons south-east of Koutiala (Mali), cropped with the five major crops grown in the area (cotton, maize, sorghum, millet and peanuts). Each field included up to five plots with different fertilizer applications and one plot with farmer practice. Fortnightly, in-situ in each field data were collected synchronous with UAV imaging using a Canon S110 NIR camera. A concurrent series of very high-resolution satellite images was procured and these images were used to mask out trees. For each plot, we calculated vegetation index means, medians and coefficients of variation. Cross-validated general linear models were used to assess the predictability of relative differences in crop yield and yield response to fertilizer, explicitly accounting for the effects of fertility treatments, between-field and within-field variabilities. Differences between fields accounted for a much larger component of variation than differences between fertilization treatments. Vegetation indices from UAV images strongly related to ground cover (R2=0.85), light interception (R2=0.79) and vegetation indices derived from satellite images (R2 values of about 0.8). Within-plot distributions of UAV-derived vegetation index values were negatively skewed, and within-plot variability of vegetation index values was negatively correlated with yield. Abstract: In Mali's cotton belt, spatial variability in management practices, soil fertility and rainfall strongly impact crop productivity and the livelihoods of smallholder farmers. To identify crop growth conditions and hence improve food security, accurate assessment of local crop production is key. However, production estimates in heterogeneous smallholder farming systems often rely on labor-intensive surveys that are not easily scalable, nor exhaustive. Recent advances in high-resolution earth observation (EO) open up new possibilities to work in heterogeneous smallholder systems. This paper develops a method to estimate individual crop production at farm-to-community scales using high-resolution Sentinel-2 time series and ground data in the commune of Koningue, Mali. Our estimation of agricultural production relies on (i) a supervised, pixel-based crop type classification inside an existing cropland mask, (ii) a comparison of yield estimators based on spectral indices and derived leaf area index (LAI), and (iii) a Monte Carlo approach combining the resulting unbiased crop area estimate and the uncertainty on the associated yield estimate. Results show that crop types can be mapped from Sentinel-2 data with 80% overall accuracy (OA), with best performances observed for cotton (Fscore 94%), maize (88%) and millet (83%), while peanut (71%) and sorghum (46%) achieve less. Incorporation of parcel limits extracted from very high-resolution imagery is shown to increase OA to 85%. Obtained through inverse radiative transfer modeling, Sen2-Agri estimates of LAI achieve better prediction of final grain yield than various vegetation indices, reaching R2 of 0.68, 0.62, 0.8 and 0.48 for cotton, maize, millet and sorghum respectively. The uncertainty of Monte Carlo production estimates does not exceed 0.3% of the total production for each crop type.

Abstract: OBJECTIVES: Premenstrual syndrome and premenstrual dysphoric disorder (PMS/PMDD) bother a substantial number of women. Homeopathy seems a promising treatment, but it needs investigation using reliable study designs. The feasibility of organizing an international randomized pragmatic trial on a homeopathic add-on treatment (usual care [UC] + HT) compared with UC alone was evaluated. DESIGN: A multicenter, randomized, controlled pragmatic trial with parallel groups. SETTINGS/LOCATION: The study was organized in general and private homeopathic practices in the Netherlands and Sweden and in an outpatient university clinic in Germany. SUBJECTS: Women diagnosed as having PMS/PMDD, based on prospective daily rating by the daily record of severity of problems (DRSP) during a period of 2 months, were included and randomized. INTERVENTIONS: Women were to receive UC + HT or UC for 4 months. Homeopathic medicine selection was according to a previously tested prognostic questionnaire and electronic algorithm. Usual care was as provided by the women's general practitioner according to their preferences. OUTCOME MEASURES: Before and after treatment, the women completed diaries (DRSP), the measure yourself concerns and well-being, and other questionnaires. Intention-to-treat (ITT) and per protocol (PP) analyses were performed. RESULTS: In Germany, the study could not proceed because of legal limitations. In Sweden, recruitment proved extremely difficult. In the Netherlands and Sweden, 60 women were randomized (UC + HT: 28; UC: 32), data of 47/46 women were analyzed (ITT/PP). After 4 months, relative mean change of DRSP scores in the UC + HT group was significantly better than in the UC group (p = 0.03). CONCLUSIONS: With respect to recruitment and different legal status, it does not seem feasible to perform a larger, international, pragmatic randomized trial on (semi-)individualized homeopathy for PMS/PMDD. Abstract: Ice-sheet and sea-level fluctuations during the Early and Middle Pleistocene are as yet poorly understood. A stalactite from a karst cave in North West Sicily (Italy) provides the first evidence of four marine inundations that correspond to relative sea-level highstands at the time of the Middle Pleistocene Transition. The speleothem is located ∼97 m above mean sea level as result of Quaternary uplift. Its section reveals three marine hiatuses and a coral overgrowth that fixes the age of final marine ingression at 1.124 ± 0.2, thus making this speleothem the oldest stalactite with marine hiatuses ever studied to date. Scleractinian coral species witness light-limited conditions and water depth of 20–50 m. Integrating the coral-constrained depth with the geologically constrained uplift rate and an ensemble of RSL scenarios, we find that the age of the last marine ingression most likely coincides with Marine Isotope Stage 35 on the basis of a probabilistic assessment. Our findings are consistent with a significant Antarctic ice-sheet retreat.
