Background: Young children with autism spectrum disorder (ASD) are at risk for challenges with their emotion regulation abilities, and often have dysregulating behaviors. For young children who exhibit dysregulation, this might also impact their ability to jointly engage with others. This study aims to examine the relationship between dysregulation (challenges with ER) and joint engagement within the context of a free play interaction between toddlers with ASD and their caregivers.Methods: Participants include 149 children with ASD (M age 21 months) and their caregivers prior to intervention (baseline). Bivariate correlations and regression analyses were conducted to explore the relations between dysregulation, joint engagement, and caregiver strategies. Results: Of the 149 children included in this study, 122 (82%) demonstrated at least one episode of dysregulation (lasting over three seconds) within the caregiver interaction, and 50% of those behaviors were deemed intense. Child dysregulation is significantly, and negatively, related to time spent jointly engaged with their caregiver (r(148)=-.260, p=.001). Caregivers increased use of active strategies (e.g., redirection) and avoidant strategies are related to increased child dysregulation (active strategies: r(147)=.260, p=.001; avoidant strategies: r(147)=.196, p=.017). In contrast, caregivers’ use of developmentally appropriate engagement and regulation strategies (JASPER Caregiver Strategies) is predictive of less time dysregulated (F(4,143)=7.618, p<.001, R2=.146). Higher JASPER caregiver strategies are associated with less time unengaged (r(148)=-.246, p=.003). A significant moderation effect of JASPER caregiver strategies on the relation between dysregulation and time unengaged was found, F(5,114)=9.712, p=<.001, R2=.299. Hepatocellular carcinoma (HCC) ranks among the five most common cancer entities worldwide and leads to hundred-thousands of deaths every year. Despite some groundbreaking therapeutical revelations during the last years, the overall prognosis remains poor. Although the immune system fights malignant transformations with a robust anti-tumor response, certain immune mediators have also been shown to promote cancer development. For example, interleukin (IL)-22 has been associated with HCC progression and worsened prognosis in multiple studies. However, the underlying mechanisms of the pathological role of IL-22-signaling as well as the role of its natural antagonist IL-22 binding protein (IL-22BP) in HCC remain elusive. Here, we corroborate the pathogenic role of IL-22 in HCC by taking advantage of two mouse models. Moreover, we observed a protective role of IL-22BP during liver carcinogenesis. While IL-22 was mainly produced by CD4+ T cells in HCC, IL-22BP was abundantly expressed by neutrophils during liver carcinogenesis. Hepatocytes could be identified as a major target of this pathological IL-22-signaling. Moreover, abrogation of IL-22 signaling in hepatocytes in IL22ra1flox/flox × AlbCre+ mice reduced STEAP4 expression-a known oncogene-in HCC in vivo. Likewise, STEAP4 expression correlated with IL22 levels in human HCC samples, but not in healthy liver specimens. Fragile X syndrome (FXS) is associated with deficits in various types of learning, including those that require the hippocampus. Relatedly, hippocampal long-term potentiation (LTP) is impaired in the Fmr1 knockout (KO) mouse model of FXS. Prior research found that infusion of brain-derived neurotrophic factor (BDNF) rescues LTP in the KOs. Here, we tested if, in Fmr1 KO mice, up-regulating BDNF production or treatment with an agonist for BDNF's TrkB receptor restores synaptic plasticity and improves learning. In hippocampal slices, bath infusion of the TrkB agonist 7,8-dihydroxyflavone (7,8-DHF) completely restored otherwise impaired hippocampal field CA1 LTP of Fmr1 KOs without effect in wild types (WTs). Similarly, acute, semi-chronic, or chronic treatments with 7,8-DHF rescued a simple hippocampus-dependent form of spatial learning (object location memory: OLM) in Fmr1 KOs without effect in WTs. The agonist also restored object recognition memory, which depends on cortical regions. Semi-chronic, but not acute, treatment with the ampakine CX929, which up-regulates BDNF expression, lowered the training threshold for OLM in WT mice and rescued learning in the KOs. Positive results were also obtained in a test for social recognition. An mGluR5 antagonist did not improve learning. Quantification of synaptic immunolabeling demonstrated that 7,8-DHF and CX929 increase levels of activated TrkB at excitatory synapses. Moreover, CX929 induced a robust synaptic activation of the TrkB effector ERK1/2. This study utilized an age- and IQ-matched sample of school-aged youth (n = 228) diagnosed with ASD to assess sex differences on the ADOS and ADI-R, parent-reported autistic traits, and adaptive skills. Although females and males were rated similarly on gold-standard diagnostic measures overall, females with higher IQs were less likely to meet criteria on the ADI-R. The BTBR T(+) Itpr3(tf)/J mouse strain exhibits behaviors that align with the major diagnostic criteria of autism. To further evaluate the BTBR strains cognitive impairments, we quantified hippocampus-dependent object location memory (OLM) and found that one-third of the BTBR mice exhibited robust memory, whereas the remainder did not. Fluorescence deconvolution tomography was used to test whether synaptic levels of activated extracellular signal-regulated kinase 1/2 (ERK1/2), a protein that contributes importantly to plasticity, correlate with OLM scores in individual mice. In hippocampal field CA1, the BTBRs had fewer post-synaptic densities associated with high levels of phosphorylated (p-) ERK1/2 as compared with C57BL/6 mice. Although counts of p-ERK1/2 immunoreactive synapses did not correlate with OLM performance, the intensity of synaptic p-ERK1/2 immunolabeling was negatively correlated with OLM scores across BTBRs. Metabotropic glutamate receptor (mGluR) 5 signaling activates ERK1/2. Therefore, we tested whether treatment with the mGluR5 antagonist MPEP normalizes synaptic and learning measures in BTBR mice: MPEP facilitated OLM and decreased synaptic p-ERK1/2 immunolabeling intensity without affecting numbers of p-ERK1/2+ synapses. In contrast, semi-chronic ampakine treatment, which facilitates memory in other models of cognitive impairment, had no effect on OLM in BTBRs. Recent studies have shown that short, spaced trains of afferent stimulation produce much greater long-term potentiation (LTP) than that obtained with a single, prolonged stimulation episode. The present studies demonstrate that spaced training regimens, based on these LTP timing rules, facilitate learning in wild-type (WT) mice and can offset learning and synaptic signaling impairments in the fragile X mental retardation 1 (Fmr1) knockout (KO) model of fragile X syndrome. We determined that 5 min of continuous training supports object location memory (OLM) in WT but not Fmr1 KO mice. However, the same amount of training distributed across three short trials, spaced by one hour, produced robust long-term memory in the KOs. At least three training trials were needed to realize the benefit of spacing, and intertrial intervals shorter or longer than 60 min were ineffective. Multiple short training trials also rescued novel object recognition in Fmr1 KOs. The spacing effect was surprisingly potent: just 1 min of OLM training, distributed across three trials, supported robust memory in both genotypes. Spacing also rescued training-induced activation of synaptic ERK1/2 in dorsal hippocampus of Fmr1 KO mice. These results show that a spaced training regimen designed to maximize synaptic potentiation facilitates recognition memory in WT mice and can offset synaptic signaling and memory impairments in a model of congenital intellectual disability. more...

Multiple lines of evidence suggest that disturbances in excitatory transmission contribute to depression. Whether these defects involve the number, size, or composition of glutamatergic contacts is unclear. This study used recently introduced procedures for fluorescence deconvolution tomography in a well-studied rat model of congenital depression to characterize excitatory synapses in layer I of infralimbic cortex, a region involved in mood disorders, and of primary somatosensory cortex. Three groups were studied: (1) rats bred for learned helplessness (cLH); (2) rats resistant to learned helplessness (cNLH); and (3) control Sprague Dawley rats. In fields within infralimbic cortex, cLH rats had the same numerical density of synapses, immunolabeled for either the postsynaptic density (PSD) marker PSD95 or the presynaptic protein synaptophysin, as controls. However, PSD95 immunolabeling intensities were substantially lower in cLH rats, as were numerical densities of synapse-sized clusters of the AMPA receptor subunit GluA1. Similar but less pronounced differences (comparable numerical densities but reduced immunolabeling intensity for PSD95) were found in the somatosensory cortex. In contrast, non-helpless rats had 25% more PSDs than either cLH or control rats without any increase in synaptophysin-labeled terminal frequency. Compared with controls, both cLH and cNLH rats had fewer GABAergic contacts. Stabilization of long-term potentiation (LTP) depends on reorganization of the dendritic spine actin cytoskeleton. The present study tested whether this involves activity-driven effects on the actin-regulatory protein cortactin, and whether such effects are disturbed in the Fmr1 knock-out (KO) model of fragile X syndrome, in which stabilization of both actin filaments and LTP is impaired. LTP induced by theta burst stimulation (TBS) in hippocampal slices from wild-type mice was associated with rapid, broadly distributed, and NMDA receptor-dependent decreases in synapse-associated cortactin. The reduction in cortactin content was blocked by blebbistatin, while basal levels were reduced by nocodazole, indicating that cortactin's movements into and away from synapses are regulated by microtubule and actomyosin motors, respectively. These results further suggest that synapse-specific LTP influences cytoskeletal elements at distant connections. The rapid effects of TBS on synaptic cortactin content were absent in Fmr1 KOs as was evidence for activity-driven phosphorylation of the protein or its upstream kinase, ERK1/2. Phosphorylation regulates cortactin's interactions with actin, and coprecipitation of the two proteins was reduced in the KOs. We propose that, in the KOs, excessive basal phosphorylation of ERK1/2 disrupts its interactions with cortactin, thereby blocking the latter protein's use of actomyosin transport systems. These impairments are predicted to compromise the response of the subsynaptic cytoskeleton to learning-related afferent activity, both locally and at distant sites. more...

Due to the stringent population bottleneck that occurs during sexual HIV-1 transmission, systemic infection is typically established by a limited number of founder viruses. Elucidation of the precise forces influencing the selection of founder viruses may reveal key vulnerabilities that could aid in the development of a vaccine or other clinical interventions. Here, we utilize deep sequencing data and apply a genetic distance-based method to investigate whether the mode of sexual transmission shapes the nascent founder viral genome. Analysis of 74 acute and early HIV-1 infected subjects revealed that 83% of men who have sex with men (MSM) exhibit a single founder virus, levels similar to those previously observed in heterosexual (HSX) transmission. In a metadata analysis of a total of 354 subjects, including HSX, MSM and injecting drug users (IDU), we also observed no significant differences in the frequency of single founder virus infections between HSX and MSM transmissions. However, comparison of HIV-1 envelope sequences revealed that HSX founder viruses exhibited a greater number of codon sites under positive selection, as well as stronger transmission indices possibly reflective of higher fitness variants. Moreover, specific genetic "signatures" within MSM and HSX founder viruses were identified, with single polymorphisms within gp41 enriched among HSX viruses while more complex patterns, including clustered polymorphisms surrounding the CD4 binding site, were enriched in MSM viruses. While our findings do not support an influence of the mode of sexual transmission on the number of founder viruses, they do demonstrate that there are marked differences in the selection bottleneck that can significantly shape their genetic composition. Syngap1 haploinsufficiency is a common cause of sporadic intellectual disability. Syngap1 mutations disrupt developing pyramidal neurons, although it remains unclear if this process contributes to cognitive abnormalities. Here, we found that haploinsufficiency restricted to forebrain glutamatergic neurons was sufficient to disrupt cognition and removing mutations from this population prevented cognitive abnormalities. In contrast, manipulating Syngap1 function in GABAergic neurons had no effect on cognition, excitability, or neurotransmission, highlighting the specificity of Syngap1 mutations within forebrain excitatory neurons. Interestingly, cognitive abnormalities were reliably predicted by the emergence of enhanced excitatory synaptic function in mature superficial cortical pyramidal cells, which was a neurophysiological disruption caused by Syngap1 dysfunction in developing, but not adult, forebrain neurons. We conclude that reduced cognition in Syngap1 mutants is caused by isolated damage to developing forebrain glutamatergic neurons. Purpose: T-cell-located CD4 antigen represents one of the therapeutic targets in rheumatoid arthritis (RA). However, up to now there is no established imaging tool to visualize this target in vivo. The aim of our study was to assess the safety and tolerability of a technetium-99 m labelled murine anti-human CD4 IgG1-Fab fragment ([99mTc]-anti-CD4-Fab, [99mTc]-EP1645) in patients with active synovitis due to RA, and to evaluate its potential as a marker of disease activity. Methods: In the present phase I proof of principle study five patients with RA were examined. Planar scans of the whole body, hands, and feet were taken 30 min up to 24 h after application of 550 ± 150 MBq [99mTc]-anti-CD4-Fab, followed by visual analyses, comparison with clinical data in 68 joints per patient and semiquantitative analysis of hand and wrist joints. Results: Neither infusion related adverse events nor adverse events during follow up were observed. No increase in human anti-murine antibody titres was seen. All patients had positive scans in almost 70% of clinically affected joints. Positive scans were also found in 8% of joints without evidence of swelling or tenderness. Conclusion: Scintigraphy with [99mTc]-anti-CD4-Fab is a promising technique for evaluation of inflammatory activity in patients with RA, pre-therapeutical evaluation of CD4 status and therapy control. Tracer uptake in clinically inconspicuous joints strongly indicates diagnostic potential of [99mTc]-anti-CD4-Fab. Whether this technique is eligible as a prognostic factor in RA needs to be analysed in further studies as well as the pathophysiological background of clinically affected joints lacking tracer uptake. more...

The complex effects of stress on learning and memory are mediated, in part, by stress-induced changes in the composition and structure of excitatory synapses. In the hippocampus, the effects of stress involve several factors including glucocorticoids and the stress-released neuropeptide corticotropin-releasing hormone (CRH), which influence the integrity of dendritic spines and the structure and function of the excitatory synapses they carry. CRH, at nanomolar, presumed-stress levels, rapidly abolishes short-term synaptic plasticity and destroys dendritic spines, yet the mechanisms for these effects are not fully understood. Here we tested the hypothesis that glutamate receptor-mediated processes, which shape synaptic structure and function, are engaged by CRH and contribute to spine destabilization. In cultured rat hippocampal neurons, CRH application reduced dendritic spine density in a time- and dose-dependent manner, and this action depended on the CRH receptor type 1. CRH-mediated spine loss required network activity and the activation of NMDA, but not of AMPA receptors; indeed GluR1-containing dendritic spines were resistant to CRH. Downstream of NMDA receptors, the calcium-dependent enzyme, calpain, was recruited, resulting in the breakdown of spine actin-interacting proteins including spectrin. Pharmacological approaches demonstrated that calpain recruitment contributed critically to CRH-induced spine loss. In conclusion, the stress hormone CRH co-opts mechanisms that contribute to the plasticity and integrity of excitatory synapses, leading to selective loss of dendritic spines. Purpose [1134 C]DASB is currently the most frequently applied highly selective radiotracer for visualisation and quantification of central SERT. Its use, however, is hampered by the short half life of carbon-11, the moderate cortical test retest reliability, and the lack for quantifying endogenous serotonin. Labelling with fluorine-18 allows in principle for longer acquisition times for kinetic analysis in brain tissue and may provide higher sensitivity. The aim of our study was to firstly apply in human the new highly SERT-selective fluorine-18 labelled fluoromethyl analogue of (+)-McN5652 ((+)-[1840 F]FMe-McN5652) and to evaluate its potential for SERT quantification. Methods The PET data of five healthy volunteers (3 male, 2 female, age 39 ± 10 years) coregistered with individual MRI were semi-quantitatively assessed by volume-of-interest analysis using the software package PMOD. Rate constants and distribution volume ratios (DVR) were calculated using a 2-tissue compartment model and arterial input function measurement were corrected for metabolites/plasma data. Standardized uptake region-to-cerebellum ratios as measure of specific radiotracer accumulation were compared with those of a [1146 C]DASB-PET data set of healthy subjects (10 male, 11 female, 38 ± 8 years). Results The 2-tissue compartment model provided adequate fits to the data. Estimates of total distribution volume (VT) demonstrated good identifiability based on coefficient of variation (COV) for the volumes of interest (VOIs) in SERT-rich and also for cortical areas (COV VT < 10%). Compared with [11C]DASB-PET, there was a tendency to lower mean values in (+)-[1851 F]FMe-McN5652 PET, however, the standard deviation was also somewhat lower. Altogether, cerebral (+)-[1853 F]FMe-McN5652 uptake corresponds well with the known SERT distribution also in humans. The aim of our study was to first apply in human the new highly SERT‐selective fluorine‐18 labelled fluoromethyl analogue of (+)‐McN5652 ([18F]FMe‐McN). Methods: The synthesis of [18F]FMe‐McN was performed according to Zessin et al. with some modifications. Briefly, the demethylated (+)‐McN5652 was reacted with bromo‐[18F]fluoromethane to yield [18F]FMe‐McN, which was purified by reversed‐phase HPLC. For in vivo human studies, five healthy volunteers (2 female, age 39±10 years) underwent dynamic PET over 120 minutes after intravenuous injection of a 90 s bolus of 298±57 MBq [18F]FMe‐McN and a static acquisition over 30 minutes 3 h p.i.. PET data were coregistered with individual MRI data set using PMOD and VOI analysis was performed. Target‐to‐background‐ratios (TB‐R, cerebellum as background structure) were compared with those of a reference data set assessed by [11C]DASB‐PET in 21 healthy subjects (11 female, 38±8 years). Results: TB‐R ([18F]FMe‐McN) displays no hemispheric differences. The values are for the frontal cortex (FC) 1.02±0.04 (right‐hand side) and 1.01±0.03 (left), for the head of caudate region (caud) 1.46±0.16 (right) and 1.50±0.15 (left) and for the raphé region 2.04±0.11. Corresponding TB‐R ([11C]DASB) are 1.10±0.07 (FC right, ANOVA p=0.05), 1.08±0.78 (FC left, 0.06), 2.14±0.21 (caud right, 0.02), 2.06±0.19 (caud left, 0.04) und 2.23±0.39 (raphé, 0.04). Visually, image quality of [11C]DASB‐PET is superior to [18F]FMe‐McN . Conclusion: Cerebral radiotracer uptake fits well with the known SERT distribution also in humans. Ziel/Aim: Die zentralen SERT lassen sich mittels SPECT-Radiotracern in SERT-reichen Gehirnarealen und mit PET-Markern wie [11C]DASB hochselektiv im gesamten Gehirn darstellen. Die kortikale Test-Retest-Reliabilität von [11C]DASB PET ist lediglich moderat, eine Quantifizierung endogenen Serotonins gelingt mit dieser Methode nicht. Möglicherweise können Tracer mit längerlebigen Nukliden diese Mängel beseitigen helfen. Ziel unserer Studie war die erstmalige Applikation eines Fluor-18-markierten SERT-Radiotracers im Menschen. Methodik/Methods: In Anlehnung an (1) wurde die Synthese von [18F]FMe-McN 5652 unter GMP-Bedingungen in einem modifizierten Synthesemodul "TRACERlab FxF-N” als 2-Stufen-2-Topf-Reaktion adaptiert: Stufe 1: Synthese von [18F]Fluormethylbromid durch nukleophile [18F]Fluorierung von Dibrommethan und nachfolgende Reinigung mittels Silicagel-Kartuschen (2). Stufe 2: [18F]Fluormethylierung eines entsprechenden Thiolat-Präkursors. Da [18F]FMe-McN 5652 in wässriger Lösung nicht stabil ist, wurde eine HPLC-Reinigung unter Verwendung eines ethanolischen Eluenten entwickelt, in deren Produktfraktion der Tracer stabil ist. Die PET-Datensätze, erhoben bei 5 gesunden Probanden (2 weiblich, Alter 39±10 Jahre) als dynamische Akquisition über 120 Min. nach i.v. 90-Sek.-Bolusinjektion von 298±57 MBq [18F]FMe-McN 5652 sowie statische Aufnahmen über 30 Min, wurden mit dem individuellen MRT koregistriert (PMOD) und mittels VOI analysiert (Target-/Background-Ratios, TB-R, Background=Zerebellum). Ergebnisse/Results: Die TB-R für den frontalen Kortex (FC) betragen 1,02±0,04 für rechts und 1,01±0,03 für links, für die Kaudatuskopfregion (Kaud) 1,46±0,16 (rechts) und 1,50±0,15 (links) und für die RaphÈ-Region 2,04±0,11.
