Syftet med studien var att undersöka hur tarmfloran hos kalvar varierar med ålder och segment i magtarmkanalen samt att undersöka förekomsten av Faecalibacterium prausnitzii, Lactobacillus spp. och segmenterade filamentösa bakterier (SFB). Tidigare studier har visat att dessa bakterier har viktiga funktioner för utvecklingen av immunförsvaret i tarmen hos vissa däggdjur samt för försvaret mot kolonisation av patogener. Bakteriefloran i magtarmkanalen hos unga kalvar är hittills väldigt lite studerad. Fem kalvar avlivades och provtogs vid en ålder av 2 dagar och fyra kalvar vid 7 dagars ålder. Från dessa kalvar samlades prov från slemhinnan/epitelet i abomasum, rumen, duodenum, ileum, cecum och colon samt träck. Ytterligare träckprover togs från andra kalvar vid en ålder av 2, 7, 14 och 28 dagar samt vid avvänjning och vid 6 månaders ålder. Bakteriefloran karakteriserades med hjälp av T-RFLP-analys (terminal restriction fragment lenght polymporhism) av genen som kodar för 16S ribosomalt RNA, där T-RFLP användes för att få en profil över bakteriefloran. Dessutom användes PCR för att specifikt detektera F. prausnitzii, Lactobacillus spp. och SFB. Resultaten visar att bakteriefloran varierar med åldern och att floran i prover från samma segment av magtarmkanalen hos olika individer liknade varandra mer än prover från olika segment inom samma kalv. Lactobacillus spp. och F. prausnitzii förekom hos samtliga kalvar från och med 7 dagars ålder. SFB påvisades i träckprover från flera kalvar från 28 dagars ålder och uppåt men inte i några vävnadsprover. From these calves scrapings from the mucosa of the abomasum, rumen, duodenum, ileum, cecum and colon as well as feces were collected. Additional fecal samples were collected from other calves at the age of 2, 7, 14 and 28 days, at weaning and at 6 month of age. The intestinal flora was characterized by T-RFLP (terminal restriction fragment lenght polymorhism) analysis of the gene that encodes for 16S ribosomal RNA and T-RFLP was used to generate a profile of the gut flora. PCR was used to specifically detect F. prausnitzii, Lactobacillus spp., and SFB. The results show that the intestinal flora varies with age and that the flora in the same segment of the gastrointestinal tract have higher similarity to the same segment in other calves compared to different segments within one calf. Lactobacillus spp. and F. prausnitzii where found in all calves 7 days and older while SFB was found in several fecal samples from calves aged 28 days and older but in no tissue samples. The intestinal flora of calves develops with age, varies with different segments i the gastrointestinal tract and contains bacteria that can be important for calf health.

The human small intestine is a key site for interactions between the intestinal microbiota and the mucosal immune system. Here we investigated the immunomodulatory properties of representative species of commonly dominant small-intestinal microbial communities, including six streptococcal strains (four Streptococcus salivarius, one S. equinus, one S. parasanguinis) one Veillonella parvula strain, one Enterococcus gallinarum strain, and Lactobacillus plantarum WCFS1 as a bench mark strain on human monocyte-derived dendritic cells. The different streptococci induced varying levels of the cytokines IL-8, TNF-a, and IL-12p70, while the V. parvula strain showed a strong capacity to induce IL-6. E. gallinarum strain was a potent inducer of cytokines and TLR2/6 signalling. As Streptococcus and Veillonella can potentially interact metabolically and frequently co-occur in ecosystems, immunomodulation by pair-wise combinations of strains were also tested for their combined immunomodulatory properties. Strain combinations induced cytokine responses in dendritic cells that differed from what might be expected on the basis of the results obtained with the individual strains. A combination of (some) streptococci with Veillonella appeared to negate IL-12p70 production, while augmenting IL-8, IL-6, IL-10, and TNF-a responses. This suggests that immunomodulation data obtained in vitro with individual strains are unlikely to adequately represent immune responses to mixtures of gut microbiota communities in vivo. Nevertheless, analysing the immune responses of strains representing the dominant species in the intestine may help to identify immunomodulatory mechanisms that influence immune homeostasis.
