Sensory neurons cooperate with barrier tissues and resident immune cells to form a significant aspect of defensive strategies in concert with the immune system. This assembly of neuroimmune cellular units is exemplified across evolution from early metazoans to mammalian life. As such, sensory neurons possess the capability to detect pathogenic infiltrates at barrier surfaces. This capacity relies on mechanisms that unleash specific cell signaling, trafficking and defensive reflexes. These pathways exploit mechanisms to amplify and enhance the alerting response should pathogenic infiltration seep into other tissue compartments and/or systemic circulation. Here we explore two hypotheses: 1) that sensory neurons' potential cellular signaling pathways require the interaction of pathogen recognition receptors and ion channels specific to sensory neurons and; 2) mechanisms which amplify these sensing pathways require activation of multiple sensory neuron sites. Where possible, we provide references to other apt reviews which provide the reader more detail on specific aspects of the perspectives provided here.

The transient receptor potential vanilloid (TRPV) ion channel family is involved in multiple sensory and physiological functions including thermosensing and temperature-dependent neuroendocrine regulation. The objective of the present study was to investigate the number, origin and evolution of TRPV genes in metazoans, with special focus on the impact of the vertebrate whole-genome duplications (WGD). Gene searches followed by phylogenetic and synteny analyses revealed multiple previously undescribed TRPV genes. The common ancestor of Cnidaria and Bilateria had three TRPV genes that became four in the deuterostome ancestor. Two of these were lost in the vertebrate ancestor. The remaining two genes gave rise to two TRPV subfamilies in vertebrates, consisting of subtypes 1, 2, 3, 4, 9 and 5, 6, 7, 8, respectively. This gene expansion resulted from the two basal vertebrate WGD events (1R and 2R) and three local duplications before the radiation of gnathostomes. TRPV1, 4 and 5 have been retained in all gnathostomes investigated, presumably reflecting important functions. TRPV7 and 8 have been lost independently in various lineages but are still retained in cyclostomes, actinistians (coelacanth), amphibians, prototherians and basal actinopterygians (Polypteridae). TRPV3 and 9 are present in extant elasmobranchs, while TRPV9 was lost in the osteichthyan ancestor and TRPV3 in the actinopterygian ancestor. The coelacanth has retained the ancestral osteichthyan repertoire of TRPV1, 3, 4, 5, 7 and 8. TRPV2 arose in the tetrapod ancestor. Duplications of TRPV5 occurred independently in various lineages, such as cyclostomes, chondrichthyans, anuran amphibians, sauropsids, mammals (where the duplicate is called TRPV6), and actinopterygians (Polypteridae and Esocidae). After the teleost-specific WGD (3R) only TRPV1 retained its duplicate, whereas TRPV4 and 5 remained as single genes. Objective: Members of the transient receptor potential (TRP) channels are involved in mediating the electrical excitability and stimulus-secretion coupling in the pancreatic beta-cells. The expression and the relative abundance of different TRP channels in the human beta-cells are unknown. The objective of this study was to examine the expression of the TRP channels and their relative abundance in the human beta-cell. Methods: RNA sequencing data obtained from human islets, fluorescence-activated cell sorting-purified human beta-cell and human pancreatic acinar cells were analyzed. Gene counts and fragments per kilobase per million mapped reads were obtained. Results: Among the TRPC family only the TRPC1 was expressed in the human beta-cell. TRPV1 channels were not expressed in the human beta-cells. Among the TRPM family, TRPM4, TRPM7, TRPM2, and TRPM3 were expressed in the human beta-cell. Of the remaining TRP channels, TRPP2, TRPML1, and TRPML3 were expressed in these cells. Conclusions: By analyzing the RNA sequencing data, we have detected for the first time the TRP channels that are expressed in the purified human beta-cells, in comparison to the other relevant pancreatic cell types. Our study provides an opportunity to focus on these TRP channels for a better understanding of the electrophysiology and stimulus-secretion coupling in these cells.

open, Studi presenti in letteratura hanno dimostrato che nel lievito S.cerevisiae lo stress generato dallo shock ipotonico induce un aumento transiente della [Ca2+]i. Dai nostri studi risulta che lo shock ipotonico è mediato, in cellule cresciute in terreno ricco, da due componenti cineticamente distinte, una molto rapida (circa 5 secondi), leggermente inibita da calcio e dipendente dal calcio extracellulare, e l’altra più lenta (10-20 sec.), fortemente sensibile alla presenza di calcio nell’ambiente esterno, al punto da non essere rilevabile anche a concentrazioni extracellulari di 1 µM. Mentre la prima risposta sembra essere mediata da un trasportatore localizzato sulla membrana plasmaticala seconda è chiaramente dipendente da un rilascio dagli store intracellulari. Alcuni dati presenti in letteratura sui geni regolati dal calcio ci hanno suggerito di studiare il ruolo nella risposta a shock ipotonico di due geni omologhi, di cui il primo è stata proposto come codificante per un putativo canale per ioni simile ai canali TRP di mammifero: YOR365C e FLC2. Flc2 è localizzata sulle membrane plasmatica, di Golgi ed ER ed è stato proposto come putativo trasportatore di flavine assieme ai suoi omologhi Flc1 e Flc3. Tuttavia Flc2 era già stato precedentemente identificato come un omologo del canale del calcio pkd2 di S. pombe. Effettivamente in base a nostri allineamenti è risultato evidente come Flc2 appartenga a una sottofamiglia di canali TRP-like tipica dei funghi, a cui appartengono anche le mucolipine umane e le proteine spray fungine, ma non le proteine Pkd2. Phosphoinositides serve as signature motifs for different cellular membranes and often are required for the function of membrane proteins. Here, we summarize clear evidence supporting the concept that many ion channels are regulated by membrane phosphoinositides. We describe tools used to test their dependence on phosphoinositides, especially phosphatidylinositol 4,5-bisphosphate, and consider mechanisms and biological meanings of phosphoinositide regulation of ion channels. This lipid regulation can underlie changes of channel activity and electrical excitability in response to receptors. Since different intracellular membranes have different lipid compositions, the activity of ion channels still in transit towards their final destination membrane may be suppressed until they reach an optimal lipid environment. The heart as a contractile hollow organ finely tunes mechanical parameters such as stroke volume, stroke pressure and cardiac output according to filling volumes, filling pressures via intrinsic and neuronal routes. At the cellular level, cardiomyocytes in beating hearts are exposed to large mechanical stress during successive heart beats. Although the mechanisms of excitation–contraction coupling are well established in mammalian heart cells, the putative contribution of mechanosensitive channels to Ca2+ homeostasis, Ca2+ signaling and force generation has been primarily investigated in relation to heart disease states. For instance, transient receptor potential channels (TRPs) are up-regulated in animal models of congestive heart failure or hypertension models and seem to play a vital role in pathological Ca2+ overload to cardiomyocytes, thus aggravating the pathology of disease at the cellular level. Apart from that, the contribution of mechanosensitive channels (MsC) in the normal beating heart to the downstream force activation cascade has not been addressed. We present an overview of the current literature and concepts of mechanosensitive channel involvement in failing hearts and cardiomyopathies and novel data showing a likely contribution of Ca2+ influx via mechanosensitive channels in beating normal cardiomyocytes during systolic shortening.

The hereditary cerebellar ataxias are a diverse group of neurodegenerative disorders primarily characterised by loss of balance and coordination due to dysfunction of the cerebellum and its associated pathways. Although many genetic mutations causing inherited cerebellar ataxia have been identified, a significant percentage of patients remain whose cause is unknown. The transient receptor potential (TRP) family member TRPC3 is a non-selective cation channel linked to key signalling pathways that are affected in cerebellar ataxia. Furthermore, genetic mouse models of TRPC3 dysfunction display cerebellar ataxia, making the TRPC3 gene an excellent candidate for screening ataxic patients with unknown genetic aetiology. Here, we report a genetic screen for TRPC3 mutations in a cohort of 98 patients with genetically undefined late-onset cerebellar ataxia and further ten patients with undefined episodic ataxia. We identified a number of variants but no causative mutations in TRPC3. Our findings suggest that mutations in TRPC3 do not significantly contribute to the cause of late-onset and episodic human cerebellar ataxias.
