Ter gelegenheid van het aantikken van de honderd columns en zijn afscheid van de Wageningse universiteit verscheen bij Uitgeverij Noordboek het boekje Recht voor natuur: beleid en recht als contrapunt. Hierin zijn 35 van de meest actuele columns gebundeld en door de auteur van een inleiding voorzien.

Continuous glucose monitoring (CGM) is a promising, minimally invasive alternative to plasma glucose measurements for calibrating physiology-based mathematical models of insulin-regulated glucose metabolism, reducing the reliance on in-clinic measurements. However, the use of CGM glucose, particularly in combination with insulin measurements, to develop personalized models of glucose regulation remains unexplored. Here, we simultaneously measured interstitial glucose concentrations using CGM as well as plasma glucose and insulin concentrations during an oral glucose tolerance test (OGTT) in individuals with overweight or obesity to calibrate personalized models of glucose-insulin dynamics. We compared the use of interstitial glucose with plasma glucose in model calibration, and evaluated the effects on model fit, identifiability, and model parameters’ association with clinically relevant metabolic indicators. Models calibrated on both plasma and interstitial glucose resulted in good model fit, and the parameter estimates associated with metabolic indicators such as insulin sensitivity measures in both cases. Moreover, practical identifiability of model parameters was improved in models estimated on CGM glucose compared to plasma glucose. Together these results suggest that CGM glucose may be considered as a minimally invasive alternative to plasma glucose measurements in model calibration to quantify the dynamics of glucose regulation.

Background: We previously showed that dietary intervention effects on cardiometabolic health were driven by tissue-specific insulin resistance (IR) phenotype: individuals with predominant muscle IR (MIR) benefited more from a low-fat, high-protein, and high-fiber (LFHP) diet, whereas individuals with predominant liver insulin resistance (LIR) benefited more from a high-monounsaturated fatty acid (HMUFA) diet. Objectives: To further characterize the effects of LFHP and HMUFA diets and their interaction with tissue-specific IR, we investigated dietary intervention effects on fasting and postprandial plasma metabolite profile. Methods: Adults with MIR or LIR (40–75 y, BMI 25–40 kg/m2) were randomly assigned to a 12-wk HMUFA or LFHP diet (n = 242). After the exclusion of statin use, 214 participants were included in this prespecified secondary analysis. Plasma samples were collected before (T = 0) and after (T = 30, 60, 120, and 240 min) a high-fat mixed meal for quantification of 247 metabolite measures using nuclear magnetic resonance spectroscopy. Results: A larger reduction in fasting VLDL-triacylglycerol (TAG) and VLDL particle size was observed in individuals with MIR following the LFHP diet and those with LIR following the HMUFA diet, although no longer statistically significant after false discovery rate (FDR) adjustment. No IR phenotype-by-diet interactions were found for postprandial plasma metabolites assessed as total area under the curve (tAUC). Irrespective of IR phenotype, the LFHP diet induced greater reductions in postprandial plasma tAUC of the larger VLDL particles and small HDL particles, and TAG content in most VLDL subclasses and the smaller LDL and HDL subclasses (for example, VLDL-TAG tAUC standardized mean change [95% CI] LFHP = −0.29 [−0.43, −0.16] compared with HMUFA = −0.04 [−0.16, 0.09]; FDR-adjusted P for diet × time = 0.041). Background Body composition and body fat distribution are important predictors of cardiometabolic diseases. The etiology of cardiometabolic diseases is heterogenous, and partly driven by inter-individual differences in tissue-specific insulin sensitivity.Objectives To investigate (1) the associations between body composition and whole-body, liver and muscle insulin sensitivity, and (2) changes in body composition and insulin sensitivity and their relationship after a 12-week isocaloric diet high in mono-unsaturated fatty acids (HMUFA) or a low-fat, high-protein, high-fiber (LFHP) diet.Methods This subcohort analysis of the PERSON study includes 93 individuals (53% women, BMI 25-40 kg/m2, 40-75 years) who participated in this randomized intervention study. At baseline and after 12 weeks of following the LFHP, or HMUFA diet, we performed a 7-point oral glucose tolerance test to assess whole-body, liver, and muscle insulin sensitivity, and whole-body magnetic resonance imaging to determine body composition and body fat distribution. Both diets are within the guidelines of healthy nutrition.Results At baseline, liver fat content was associated with worse liver insulin sensitivity (beta [95%CI]; 0.12 [0.01; 0.22]). Only in women, thigh muscle fat content was inversely related to muscle insulin sensitivity (-0.27 [-0.48; -0.05]). Visceral adipose tissue (VAT) was inversely associated with whole-body, liver, and muscle insulin sensitivity. Both diets decreased VAT, abdominal subcutaneous adipose tissue (aSAT), and liver fat, but not whole-body and tissue-specific insulin sensitivity with no differences between diets. Waist circumference, however, decreased more following the LFHP diet as compared to the HMUFA diet (-3.0 vs. -0.5 cm, respectively). Dit rapport bevat een reflectie en vijftien beleidsaanbevelingen naar aanleiding van het rapport ‘De Europees- en internationaalrechtelijke status van de Waddenzee’ (Waddenacademie, december 2023).

This dataset includes measurements collected during a greenhouse trial in 2009-2010 in Bleiswijk, The Netherlands, and results of simulations aimed at reproducing the greenhouse as a mathematical model. The greenhouse trial was conducted between October 2009 and June 2010 by WUR Greenhouse Horticulture and Flower Bulbs in Bleiswijk, the Netherlands. The experiment compared the growth of cocktail tomatoes (variety Sunstream) in greenhouse compartments with 4 different installations of artificial lighting: (1) full LED toplights, (2) full HPS (high-pressure sodium) toplights, (3) hybrid HPS toplights and LED interlights (lamps between the crop rows), (4) hybrid LED toplights and LED interlights. In all installations, lamps provided in total 170 µmol/m2/s PAR light. In the hybrid installations 50% of the artificial light came from the toplights and 50% from the interlights. This dataset includes only the data used in Katzin et al (2020): outdoor weather, indoor climate, and greenhouse climate control actions, for installations (1) and (2) only (full LED toplights and full HPS toplights), between 19 October 2009 at 15:15 and 8 February 2010 at 15:15. Model simulations were performed by GreenLight, an open-source greenhouse simulation model. Four simulations were performed: (1) A simulation of the indoor climate with HPS lamps; (2) A simulation of the indoor climate with LED lamps; (3) A simulation of the energy use with HPS lamps ; (4) A simulation of the energy use with LED lamps.

Background: Tissue-specific insulin resistance (IR) predominantly in muscle (muscle IR) or liver (liver IR) has previously been linked to distinct fasting metabolite profiles, but postprandial metabolite profiles have not been investigated in tissue-specific IR yet. Given the importance of postprandial metabolic impairments in the pathophysiology of cardiometabolic diseases, we compared postprandial plasma metabolite profiles in response to a high-fat mixed meal between individuals with predominant muscle IR or liver IR. Methods: This cross-sectional study included data from 214 women and men with BMI 25–40 kg/m2, aged 40–75 years, and with predominant muscle IR or liver IR. Tissue-specific IR was assessed using the muscle insulin sensitivity index (MISI) and hepatic insulin resistance index (HIRI), which were calculated from the glucose and insulin responses during a 7-point oral glucose tolerance test. Plasma samples were collected before (T = 0) and after (T = 30, 60, 120, 240 min) consumption of a high-fat mixed meal and 247 metabolite measures, including lipoproteins, cholesterol, triacylglycerol (TAG), ketone bodies, and amino acids, were quantified using nuclear magnetic resonance spectroscopy. Differences in postprandial plasma metabolite iAUCs between muscle and liver IR were tested using ANCOVA with adjustment for age, sex, center, BMI, and waist-to-hip ratio. P-values were adjusted for a false discovery rate (FDR) of 0.05 using the Benjamini–Hochberg method. Results: Sixty-eight postprandial metabolite iAUCs were significantly different between liver and muscle IR. Liver IR was characterized by greater plasma iAUCs of large VLDL (p = 0.004), very large VLDL (p = 0.002), and medium-sized LDL particles (p = 0.026), and by greater iAUCs of TAG in small VLDL (p = 0.025), large VLDL (p = 0.003), very large VLDL (p = 0.002), all LDL subclasses (all p < 0.05), and small HDL particles (p = 0.011), compared to muscle IR. In nauwe afstemming en samenwerking met de ministeries van LNV en IenW heeft Rijkswaterstaat de Waddenacademie gevraagd een onderzoek uit te voeren naar voor de Waddenzee geldende internationale verdragen en Europese richtlijnen op het gebied van natuur- en waterbescherming. Doelstelling van het onderzoek is om een gedegen overzicht te krijgen van de inhoud en concrete consequenties van de internationale en Europese beschermingsregimes voor de Waddenzee. Wat zijn de doelen die bereikt moeten worden, welke verboden en verplichtingen vloeien er uit verdragen en EU-richtlijnen voort en besteden de beschermingsregimes ook aandacht aan onderwerpen zoals klimaatverandering, cumulatieve effecten, actief natuurherstel en monitoring? Deze en andere vragen worden voor 13 regimes in afzonderlijke hoofdstukken door juridische experts beantwoord en de bevindingen zijn samengebracht in een uitvoerig samenvattend hoofdstuk.

METHODS: A multicentre cross-sectional study was performed in 170 patients who were treated nonoperatively for an acetabular fracture in three level 1 trauma centres. Using the post-injury diagnostic CT scan, the maximum gap and step-off values in the weightbearing dome were digitally measured by two trauma surgeons. Native hip survival was reported using Kaplan-Meier curves. Predictors for conversion to THA were determined using Cox regression analysis. RESULTS: Of 170 patients, 22 (13%) subsequently received a THA. Native hip survival in patients with a step-off ≤ 2 mm, > 2 to 4 mm, or > 4 mm differed at five-year follow-up (respectively: 94% vs 70% vs 74%). Native hip survival in patients with a gap ≤ 2 mm, > 2 to 4 mm, or > 4 mm differed at five-year follow-up (respectively: 100% vs 84% vs 78%). Step-off displacement > 2 mm (> 2 to 4 mm hazard ratio (HR) 4.9, > 4 mm HR 5.6) and age > 60 years (HR 2.9) were independent predictors for conversion to THA at follow-up. CONCLUSION: Patients with minimally displaced acetabular fractures who opt for nonoperative fracture treatment may be informed that fracture displacement (e.g. gap and step-off) up to 2 mm, as measured on CT images, results in limited risk on conversion to THA. Step-off ≥ 2 mm and age > 60 years are predictors for conversion to THA and can be helpful in the shared decision-making process.

Objective: A proinflammatory adipose tissue (AT) microenvironment and systemic low-grade inflammation may differentially affect tissue-specific insulin sensitivity. This study investigated the relationships of abdominal subcutaneous AT (aSAT) and circulating immune cells, aSAT gene expression, and circulating inflammatory markers with liver and skeletal muscle insulin sensitivity in people with overweight and obesity. Methods: Individuals with overweight and obesity from the PERSonalized Glucose Optimization Through Nutritional Intervention (PERSON) Study (n = 219) and the Maastricht Study (replication cohort; n = 1256) underwent a seven-point oral glucose tolerance test to assess liver and muscle insulin sensitivity, and circulating inflammatory markers were determined. In subgroups, flow cytometry was performed to identify circulating and aSAT immune cells, and aSAT gene expression was evaluated. Results: The relative abundances of circulating T cells, nonclassical monocytes, and CD56dim CD16+ natural killer cells were inversely associated with liver, but not muscle, insulin sensitivity in the PERSON Study. The inverse association between circulating (classical) monocytes and liver insulin sensitivity was confirmed in the Maastricht Study. In aSAT, immune cell populations were not related to insulin sensitivity. Furthermore, aSAT gene expression of interleukin 6 and CD14 was positively associated with muscle, but not liver, insulin sensitivity. Conclusions: The present findings demonstrate that circulating immune cell populations and inflammatory gene expression in aSAT show distinct associations with liver and muscle insulin sensitivity.

In their letter of 26 June 2023 Copa*Cogeca asked the members of the European Parliament’s Committee on Environment to reject the Nature Restoration Law proposal. They based their arguments for this rejection on what they considered as ‘red line’ issues, which makes the law proposal unacceptable to them. As Legal Working Group (LWG) from SERE , we analysed these ‘red line’ issues and we concluded that their red line objections against the Nature Restoration Law proposal cannot withstand legal scrutiny. Many of their concerns are already present in the Commission proposal and are further addressed in the Swedish presidency compromise proposal. Further weakening of the Nature Restoration Law is against existing EU legislation.

Precision nutrition based on metabolic phenotype may increase the effectiveness of interventions. In this proof-of-concept study, we investigated the effect of modulating dietary macronutrient composition according to muscle insulin-resistant (MIR) or liver insulin-resistant (LIR) phenotypes on cardiometabolic health. Women and men with MIR or LIR (n = 242, body mass index [BMI] 25–40 kg/m2, 40–75 years) were randomized to phenotype diet (PhenoDiet) group A or B and followed a 12-week high-monounsaturated fatty acid (HMUFA) diet or low-fat, high-protein, and high-fiber diet (LFHP) (PhenoDiet group A, MIR/HMUFA and LIR/LFHP; PhenoDiet group B, MIR/LFHP and LIR/HMUFA). PhenoDiet group B showed no significant improvements in the primary outcome disposition index, but greater improvements in insulin sensitivity, glucose homeostasis, serum triacylglycerol, and C-reactive protein compared with PhenoDiet group A were observed. We demonstrate that modulating macronutrient composition within the dietary guidelines based on tissue-specific insulin resistance (IR) phenotype enhances cardiometabolic health improvements. This assessment is based on the Commission proposal for a Nature Restoration Law (hereafter referred to as NRL) from 22 June 2022.2 The Legal Working Group is aware that the proposal is currently being discussed by the Council and Parliament. It is therefore not an article-by-article assessment, but a more general assessment of several legal aspects of the law we consider to be particularly important. Our choice of the discussed aspects was based on ongoing political discussions on the law (e.g. in the EU parliament and Council). The note gives legal arguments why certain articles should remain in the law or should be amended or added to the law. Legal arguments include legal certainty for stakeholders, coherence with other EU legislation, legitimate expectations, accepted legal principles, etc. Where relevant, we include concrete suggestions for amending and improving the law proposal.

Voor een duurzame teelt van potplanten onder glas moet het energieverbruik en remmiddelengebruik worden verminderd. Plantvorm bij bloeiende potplanten zoals kalanchoë is een belangrijk kwaliteitscriterium. Internodiën en bloemstelen mogen niet te gestrekt zijn en het blad mag niet te grof zijn. Met name in de winter onder lage lichtintensiteit kunnen internodiën en bloemstelen overmatig strekken. Vele factoren kunnen deze compactheid beïnvloeden, waaronder veredeling, remmiddelen, nutriëntengift, potmaat, temperatuur-strategie en belichting. Enkele remmiddelen zijn inmiddels verboden en het valt te verwachten dat steeds minder middelen zullen worden toegelaten vanuit de wetgeving. Belichting kan grote invloed uitoefenen op de plantvorm en bloei. Naast daglengte en lichtintensiteit speelt het spectrum daarbij een rol. Terwijl onder traditionele SON-T belichting het spectrum vast staat, kan er met energiezuinige LED-belichting gestuurd worden met de samenstelling van het lichtspectrum. LED biedt dus kansen om plantvorm en bloei te sturen en het gebruik van energie en remmiddelen mogelijk terug te dringen. Restoring functional ecosystems in Europe inter alia requires restoring large mammal (megafauna) diversity and densities to levels well above the current downgraded state, which results from a human-driven wave of megafauna extinctions and extirpations in the Late Pleistocene and Holocene. Combining law and natural science, we address the question to what degrees international legal instruments support or require such megafauna rewilding efforts. We provide an overview of Europe's current megafauna plus those species that would likely have been present without human interference. We categorize these species and restoration scenarios in ways that make sense from a legal perspective and identify and interpret relevant legal instruments. Our analysis indicates that Article 8(f) of the Convention on Biological Diversity requires restoring the diversity and densities of Europe's megafauna as far as possible-which is indeed quite far. Depending on the circumstances, megafauna rewilding measures can also be required or supported by various other legal instruments at global, pan-European, European Union and other levels.

Crassulacean acid metabolism (CAM) is a photosynthetic pathway that temporally separates the nocturnal CO2 uptake, via phosphoenolpyruvate carboxylase (PEPC, C4 carboxylation), from the diurnal refixation by Rubisco (C3 carboxylation). At the end of the day (CAM-Phase IV), when nocturnally stored CO2 has depleted, stomata reopen and allow additional CO2 uptake, which can be fixed by Rubisco or by PEPC. This work examined the CO2 uptake via C3 and C4 carboxylation in phase IV in the CAM species Phalaenopsis “Sacramento” and Kalanchoe blossfeldiana “Saja.” Short blackout periods during phase IV caused a sharp drop in CO2 uptake in K. blossfeldiana but not in Phalaenopsis, indicating strong Rubisco activity only in K. blossfeldiana. Chlorophyll fluorescence revealed a progressive decrease in ΦPSII in Phalaenopsis, implying decreasing Rubisco activity, while ΦPSII remained constant in phase IV in K. blossfeldiana. However, short switching to 2% O2 indicated the presence of photorespiration and thus Rubisco activity in both species throughout phase IV. Lastly, in Phalaenopsis, accumulation of starch in phase IV occurred. These results indicate that in Phalaenopsis, PEPC was the main carboxylase in phase IV, although Rubisco remained active throughout the whole phase. This will lead to double carboxylation (futile cycling) but may help to avoid photoinhibition.

A low-calorie diet (LCD) is an effective strategy to lose weight and improve cardiometabolic risk factors, however, sexual dimorphism may be present. This study aims to investigate sexual dimorphism in cardiometabolic risk factors following weight loss and after weight maintenance. 782 overweight/obese participants (65% women) of the DiOGenes trial followed an 8-week LCD (~800 kcal/day), with a 6-months follow-up weight maintenance period on ad libitum diets varying in protein content and glycemic index. Men lost more body weight during the LCD period (−12.8 ± 3.9 vs. −10.1 ± 2.8 kg, respectively, p < 0.001), but regained more weight during the follow-up period than women (1.5 ± 5.4 vs. −0.5 ± 5.5 kg, respectively, p < 0.001). Even though beneficial LCD-induced changes in cardiometabolic risk factors were found for both sexes, improvements in HOMA-IR, muscle and hepatic insulin sensitivity, triacylglycerol, HDL−, LDL− and total cholesterol, diastolic blood pressure, cholesterol esters, sphingomyelins and adiponectin were more pronounced in men than women (std. ß range: 0.073–0.144, all q < 0.05), after adjustment for weight change. During follow-up, women demonstrated a lower rebound in HDL-cholesterol, triacylglycerol and diacylglycerol (std. ß range: 0.114–0.164, all q < 0.05), independent of changes in body weight. Overall, we demonstrated sexual dimorphism in LCD-induced changes in body weight and cardiometabolic risk profile, which may be attributed to differences in body fat distribution and metabolic status.

Afgelopen jaar heeft er bij het Delphy Improvement Centre een proef gelopen waarbij een heel nieuw teeltconcept voor aardbei werd beproefd: ‘Aardbei in balans met verse doordragers'. Background: It is well-established that the etiology of type 2 diabetes differs between individuals. Insulin resistance (IR) may develop in different tissues, but the severity of IR may differ in key metabolic organs such as the liver and skeletal muscle. Recent evidence suggests that these distinct tissue-specific IR phenotypes may also respond differentially to dietary macronutrient composition with respect to improvements in glucose metabolism. Objective: The main objective of the PERSON study is to investigate the effects of an optimal vs. suboptimal dietary macronutrient intervention according to tissue-specific IR phenotype on glucose metabolism and other health outcomes. Methods: In total, 240 overweight/obese (BMI 25 – 40 kg/m2) men and women (age 40 – 75 years) with either skeletal muscle insulin resistance (MIR) or liver insulin resistance (LIR) will participate in a two-center, randomized, double-blind, parallel, 12-week dietary intervention study. At screening, participants undergo a 7-point oral glucose tolerance test (OGTT) to determine the hepatic insulin resistance index (HIRI) and muscle insulin sensitivity index (MISI), classifying each participant as either “No MIR/LIR,” “MIR,” “LIR,” or “combined MIR/LIR.” Individuals with MIR or LIR are randomized to follow one of two isocaloric diets varying in macronutrient content and quality, that is hypothesized to be either an optimal or suboptimal diet, depending on their tissue-specific IR phenotype (MIR/LIR). Extensive measurements in a controlled laboratory setting as well as phenotyping in daily life are performed before and after the intervention. The primary study outcome is the difference in change in disposition index, which is the product of insulin sensitivity and first-phase insulin secretion, between participants who received their hypothesized optimal or suboptimal diet. A low-calorie diet (LCD) is an effective strategy to lose weight and improve cardiometabolic risk factors, however, sexual dimorphism may be present. This study aims to investigate sexual dimorphism in cardiometabolic risk factors following weight loss and after weight maintenance. 782 overweight/obese participants (65% women) of the DiOGenes trial followed an 8-week LCD (~800 kcal/day), with a 6-months follow-up weight maintenance period on ad libitum diets varying in protein content and glycemic index. Men lost more body weight during the LCD period (−12.8 ± 3.9 vs. −10.1 ± 2.8 kg, respectively, p < 0.001), but regained more weight during the follow-up period than women (1.5 ± 5.4 vs. −0.5 ± 5.5 kg, respectively, p < 0.001). Even though beneficial LCD-induced changes in cardiometabolic risk factors were found for both sexes, improvements in HOMA-IR, muscle and hepatic insulin sensitivity, triacylglycerol, HDL−, LDL− and total cholesterol, diastolic blood pressure, cholesterol esters, sphingomyelins and adiponectin were more pronounced in men than women (std. ß range: 0.073–0.144, all q < 0.05), after adjustment for weight change. During follow-up, women demonstrated a lower rebound in HDL-cholesterol, triacylglycerol and diacylglycerol (std. ß range: 0.114–0.164, all q < 0.05), independent of changes in body weight. Overall, we demonstrated sexual dimorphism in LCD-induced changes in body weight and cardiometabolic risk profile, which may be attributed to differences in body fat distribution and metabolic status.

Climate change will increasingly impact species and habitat composition of protected areas, even if precise impacts are difficult to predict, especially in smaller areas. This raises questions for management authorities, not only regarding the ecological integrity of protected areas but also regarding wildlife that ‘escape’ and cause damage. The protected area is traditionally the primary responsibility of the management authority, but the introduction of charismatic and potentially damage-causing wildlife touches on the overlapping and shared commercial interests of the tourism industry and the neighbouring rural communities. As climate change manifests, the complex relationship between these three stakeholders is likely to become strained by the increased frequency of damage caused by wildlife as they attempt to move out of or expand their home ranges beyond the boundaries of the protected area. It is concluded that a laissez-faire approach to climate change by conservation authorities or protected area managers is likely to be problematic – particularly with respect to relationships with neighbouring rural communities. A greater awareness of climate change impacts among all stakeholders is required, including conservation agencies, the tourism industry and neighbouring rural communities and managing escaped wildlife should become a joint responsibility which is founded on a contractual agreement between these stakeholders.

The recolonization of wolves in European human-dominated landscapes poses a conservation challenge to protect this species and manage conflicts. The question of how humans can co-exist with large carnivores often triggers strong emotions. Here we provide an objective, science-based discussion on possible management approaches. Using existing knowledge on large carnivore management from Europe and other parts of the globe, we develop four potential wolf management scenarios; 1) population control, 2) protection and compensation, 3) fencing, 4) managing behaviour of wolf and man. For each scenario, we discuss its impact on wolf ecology, its prospects of reducing wolf-human conflicts and how it relates to current European legislation. Population control and fencing of local wolf populations are problematic because of their ecological impacts and conflicts with European legislation. In contrast, a no-interference approach does not have these problems but will likely increase human-wolf conflicts. Despite the large challenges in European, human-dominated landscapes, we argue that wolf management must focus on strengthening the separation between humans and wolves by influencing behaviour of wolves and humans on a fine spatio-temporal scale to prevent and reduce conflicts. As separation also demands a sufficiently large wild prey base, we urge restoring natural ungulate populations, to reduce human-wolf conflicts. Mutual avoidance provides the key to success, and is critical to avoid creating the conditions for reinstating wolf persecution as the default policy in Europe.
