Guinea pigs (Cavia porcellus) are popular pets and used as laboratory animals, however, their dietary preferences are under-studied. It is important for guinea pig owners and carers to increase the opportunity for good animal welfare by providing personalized and enriching dietary options. Thus, this study aimed to assess the use of paired-stimulus and multiple-stimulus-without-replacement preference assessments to determine the food preferences of seven male guinea pigs and test the validity of the methods by testing the demand for the most- and least preferred foods as reinforcers for climbing an elevated ramp. Generally, the preference assessments identified the same foods as the most preferred for each guinea pig, but not the least preferred foods. Guinea pigs climbed up to the steepest angle for both most and least preferred foods, but a non-linear least squares regression and demand analysis indicated faster climbing and inelastic demand for most preferred foods compared to slower climbing and elastic demand for least preferred foods. Both preference assessment methods were valid in identifying a preferred food, however, the multiple-stimulus-without-replacement assessment was more efficient to conduct. High-preference foods such as parsley and dandelion are recommended as future training reinforcers for guinea pigs and as foods to consider using as enrichment in captive guinea pig systems to encourage good welfare.&nbsp

Two of every three persons living with dementia reside in low- and middle-income countries (LMICs). The projected increase in global dementia rates is expected to affect LMICs disproportionately. However, the majority of global dementia care costs occur in high-income countries (HICs), with dementia research predominantly focusing on HICs. This imbalance necessitates LMIC-focused research to ensure that characterization of dementia accurately reflects the involvement and specificities of diverse populations. Development of effective preventive, diagnostic, and therapeutic approaches for dementia in LMICs requires targeted, personalized, and harmonized efforts. Our article represents timely discussions at the 2022 Symposium on Dementia and Brain Aging in LMICs that identified the foremost opportunities to advance dementia research, differential diagnosis, use of neuropsychometric tools, awareness, and treatment options. We highlight key topics discussed at the meeting and provide future recommendations to foster a more equitable landscape for dementia prevention, diagnosis, care, policy, and management in LMICs. HIGHLIGHTS: Two-thirds of persons with dementia live in LMICs, yet research and costs are skewed toward HICs. LMICs expect dementia prevalence to more than double, accompanied by socioeconomic disparities. The 2022 Symposium on Dementia in LMICs addressed advances in research, diagnosis, prevention, and policy. The Nairobi Declaration urges global action to enhance dementia outcomes in LMICs.

In regions characterized by a high concentration of small reservoirs, there is often public debate about the effectiveness of these structures in locally adapting to and mitigating drought impacts, bearing in mind their potential to modify or induce drought events in downstream areas. In this study, we investigated the influence of a Dense Network of Small Reservoirs (DNR) on the emergence and intensification of drought impacts at catchment scale, as well as their local social benefits. This analysis was based on the Socio-Hydrological-Agricultural-Reservoir (SHARE) model, specially developed for this purpose, with a medium-sized catchment in the semi-arid region of Brazil as a case study. We identified that, while a DNR can prolong the effects of a hydrological drought on storage in a large strategic reservoir at the catchment outlet by obstructing surface-runoff connectivity, it plays a crucial role in mitigating drought impacts at a local level. Specifically, the presence of small reservoirs has the potential to boost local agricultural production by up to 5 times compared to scenarios without these structures. In addition, our simulation results suggest there is a notable reduction in the need for emergency water distribution by water trucks in the presence of a DNR. This study highlights the need for a balanced approach to implementing public policies, weighing the local benefits of small reservoirs against the possible downstream impacts on large reservoirs.

The dynamin-related guanosine triphosphatase, Drp1 (encoded by Dnm1l), plays a central role in mitochondrial fission and is requisite for numerous cellular processes; however, its role in muscle metabolism remains unclear. Here, we show that, among human tissues, the highest number of gene correlations with DNM1L is in skeletal muscle. Knockdown of Drp1 (Drp1-KD) promoted mitochondrial hyperfusion in the muscle of male mice. Reduced fatty acid oxidation and impaired insulin action along with increased muscle succinate was observed in Drp1-KD muscle. Muscle Drp1-KD reduced complex II assembly and activity as a consequence of diminished mitochondrial translocation of succinate dehydrogenase assembly factor 2 (Sdhaf2). Restoration of Sdhaf2 normalized complex II activity, lipid oxidation, and insulin action in Drp1-KD myocytes. Drp1 is critical in maintaining mitochondrial complex II assembly, lipid oxidation, and insulin sensitivity, suggesting a mechanistic link between mitochondrial morphology and skeletal muscle metabolism, which is clinically relevant in combatting metabolic-related diseases.

The potential to improve drought management is high but uncertain due to rare analyses of impacts datasets, predominantly because there are few impacts monitoring programmes to generate the datasets. Drought impacts monitoring is conducted on the ground in much of Brazil by local observers at monthly and municipality scale to support the Brazilian Drought Monitor. In Ceará state, within drought-prone semiarid northeast Brazil, over 3600 drought impacts reports were completed by agricultural extension officers from 2019 to 2022. We investigated, through manual coding and observer interviews, the reported drought impacts and impact drivers. Analysis provided a catalogue of the experienced impacts and showed that impacts still occur, and are often normalised, during non-drought periods, sometimes as lingering effects of previous droughts. The impact drivers were predominantly non-extreme hydrometeorological conditions or a result of socio-technical vulnerabilities such as insufficient water infrastructure. The normalisation of “impacts” included, in particular: a generally accepted high level of crop losses and consistently low reservoir levels around which the agricultural and domestic systems are adapted. Conventional drought indices often did not align with experienced impact severity, highlighting the limitations of relying solely on these indices for emergency response. Continual impacts monitoring could be extremely valuable anywhere in the world for identifying vulnerabilities and informing proactive measures to reduce drought and other hazard risk, in addition to guiding targeted mitigation efforts.

Flash droughts, characterized by rapid onset and intensification, are increasingly occurring as a consequence of climate change and rising temperatures. However, existing hydrometeorological definitions fail to encompass the full range of flash droughts, many of which have distinct local physical attributes. Consequently, these events often go undetected or unforecast in generic global flash drought assessments and are underrepresented in research. To address this gap, we conducted a comprehensive survey to gather information on local nomenclature, characteristics, and impacts of flash droughts worldwide. The survey revealed the widespread occurrence of these phenomena, highlighting their underre-searched nature. By analyzing case studies, through literature review often in local languages to unearth elusive studies, we identified five different types of flash droughts based on their specific characteristics. Our study aims to increase awareness about the complexity and diverse impacts of flash droughts, emphasizing the importance of considering regional contexts and the vulnerability of affected populations. The reported impacts underscore the need for better integration of all flash drought types in drought research, monitoring, and management. Monitoring a combination of indicators is crucial for timely detection and response to this emerging and escalating threat.

The major concern of applying citizen science in water resources is the quality of the data. However, there are limited scientific studies addressing this concern and showing the data value. In this study, we established a citizen science program in the Akaki catchment which hosts Addis Ababa, Ethiopia. Citizen scientists monitored river stage at multiple gauging sites for multiple years. We evaluated the quality of citizen science data through a systematic quality control. Reference data was obtained from neighboring stations of the citizen science program and professionals while the evaluation involved graphical inspections and statistical methods. The quality-controlled data were applied to evaluate the spatial and temporal variation of rainfall-runoff relationships. Initially, large numbers of suspicious data were detected using single station data but that was significantly reduced when the data of multiple sites were compared. Further comparison against professional data revealed excellent agreement with high correlation coefficient (r >0.95), and low centered root mean square error (RMSE) <0.03–0.08 mm. The citizen science data indicated a large difference in rainfall-runoff relationship over the dominantly urban and rural sub-catchments. The citizen science data allowed comparison of runoff coefficient and base flow index for recent and historical periods where recent streamflow data is unavailable from a formal data source. This study illustrates the immense value of (i) multiple data quality assessment steps for building confidence on the quality of citizen science data, and (ii) citizen science for enhancing our understanding of rainfall-runoff relationships and change in a rapidly urbanizing catchment.

We introduce a new jet clustering algorithm named SIFT (Scale-Invariant Filtered Tree) that maintains the resolution of substructure for collimated decay products at large boosts. The scale-invariant measure combines properties of kT and anti-kT by preferring early association of soft radiation with a resilient hard axis, while avoiding the specification of a fixed cone size. Integrated filtering and variable-radius isolation criteria block assimilation of soft wide-angle radiation and provide a halting condition. Mutually hard structures are preserved to the end of clustering, automatically generating a tree of subjet axis candidates. Excellent object identification and kinematic reconstruction for multi-pronged resonances are realized across more than an order of magnitude in transverse energy. The clustering measure history facilitates high-performance substructure tagging, which we quantify with the aid of supervised machine learning. RNA editing, the endogenous modification of nucleic acids, is known to be altered in genes with important neurological function in schizophrenia (SCZ). However, the global profile and molecular functions of disease-associated RNA editing remain unclear. Here, we analyzed RNA editing in postmortem brains of four SCZ cohorts and uncovered a significant and reproducible trend of hypoediting in patients of European descent. We report a set of SCZ-associated editing sites via WGCNA analysis, shared across cohorts. Using massively parallel reporter assays and bioinformatic analyses, we observed that differential 3' untranslated region (3'UTR) editing sites affecting host gene expression were enriched for mitochondrial processes. Furthermore, we characterized the impact of two recoding sites in the mitofusin 1 (MFN1) gene and showed their functional relevance to mitochondrial fusion and cellular apoptosis. Our study reveals a global reduction of editing in SCZ and a compelling link between editing and mitochondrial function in the disease.

Magnetic skyrmions are topological quasiparticles whose non-volatility, detectability, and mobility make them exciting candidates for low-energy computing. Previous works have demonstrated the feasibility and efficiency of current-driven skyrmions in cascaded logic structures inspired by reversible computing. As skyrmions can be propelled through the voltage-controlled magnetic anisotropy (VCMA) effect with much greater efficiency, this work proposes a VCMA-based skyrmion propagation mechanism that drastically reduces energy dissipation. Additionally, we demonstrate the functionality of skyrmion logic gates enabled by our novel voltage-based propagation and estimate its energy efficiency relative to other logic schemes. The minimum dissipation of this VCMA-driven magnetic skyrmion logic at 0 K is found to be $\sim$6$\times$ the room-temperature Landauer limit, indicating the potential for sub-Landauer dissipation through further engineering., Comment: 4 pages, 6 figures

Whether the human fetus and the prenatal intrauterine environment (amniotic fluid and placenta) are stably colonized by microbial communities in a healthy pregnancy remains a subject of debate. Here we evaluate recent studies that characterized microbial populations in human fetuses from the perspectives of reproductive biology, microbial ecology, bioinformatics, immunology, clinical microbiology and gnotobiology, and assess possible mechanisms by which the fetus might interact with microorganisms. Our analysis indicates that the detected microbial signals are likely the result of contamination during the clinical procedures to obtain fetal samples or during DNA extraction and DNA sequencing. Furthermore, the existence of live and replicating microbial populations in healthy fetal tissues is not compatible with fundamental concepts of immunology, clinical microbiology and the derivation of germ-free mammals. These conclusions are important to our understanding of human immune development and illustrate common pitfalls in the microbial analyses of many other low-biomass environments. The pursuit of a fetal microbiome serves as a cautionary example of the challenges of sequence-based microbiome studies when biomass is low or absent, and emphasizes the need for a trans-disciplinary approach that goes beyond contamination controls by also incorporating biological, ecological and mechanistic concepts.

Particularly in arid and semiarid areas, more and more populations rely almost entirely on imported water. However, the extent to which intentional discharge into transiting river systems and unintentional leakage may be augmenting water resources for communities along and down gradient of the water transfer scheme has not previously been subject to research. The objective of this study was to assess both the potential of a large-scale water transfer (WT) scheme to increase groundwater availability by channel transmission losses in a large dryland aquifer system (2,166 km²) in Brazil, and the capability of the receiving streams to transport water downstream under a prolonged drought. An integrated surface-water/groundwater model was developed to improve the estimation of the groundwater resources, considering the spatio-temporal variability of infiltrated rainfall for aquifer recharge. Aquifer recharge from the WT scheme was simulated under prolonged drought conditions, applying an uncertainty analysis of the most influential fluxes and parameters. The annual recharge (66 mm/year) was approximately twice the amount of water abstracted (1990–2016); however, the annual recharge dropped to 13.9 mm/year from 2012 to 2016, a drought period. Under similar drought conditions, the additional recharge (6.89 × 106 m³/year) from the WT scheme did not compensate for the decrease in groundwater head in areas that do not surround the receiving streams. Actually, the additional recharge is counteracted by a decrease of 25% of natural groundwater recharge or an increase of 50% in pumping rate; therefore, WT transmission losses alone would not solve the issue of the unsustainable management of groundwater resources.

We present an analysis and numerical study of an optimal control problem for the Landau-de Gennes (LdG) model of nematic liquid crystals (LCs), which is a crucial component in modern technology. They exhibit long range orientational order in their nematic phase, which is represented by a tensor-valued (spatial) order parameter $Q = Q(x)$. Equilibrium LC states correspond to $Q$ functions that (locally) minimize an LdG energy functional. Thus, we consider an $L^2$-gradient flow of the LdG energy that allows for finding local minimizers and leads to a semi-linear parabolic PDE, for which we develop an optimal control framework. We then derive several a priori estimates for the forward problem, including continuity in space-time, that allow us to prove existence of optimal boundary and external ``force'' controls and to derive optimality conditions through the use of an adjoint equation. Next, we present a simple finite element scheme for the LdG model and a straightforward optimization algorithm. We illustrate optimization of LC states through numerical experiments in two and three dimensions that seek to place LC defects (where $Q(x) = 0$) in desired locations, which is desirable in applications., Comment: 26 pages, 9 figures

Human actions induce and modify droughts. However, scientific gaps remain with respect to how hydrological processes, anthropogenic dynamics, and individuals' perceptions of impacts are intrinsically entangled in drought occurrence and evolution. This adds complexity to drought assessment studies that cannot be addressed by the natural and environmental sciences alone. Furthermore, it poses a challenge with respect to developing ways to evaluate human behaviour and its pattern of co-evolution with the hydrological cycle-mainly related to water use and landscape modifications. During fieldwork in Brazil, we observed how drought impacts were experienced by people who were exposed to a multi-year drought. Evaluating our data, it appeared that prospect theory, a behavioural economic theory that is usually applied to explain decision-making processes under uncertainty, has explanatory power regarding what we observed in the field. Therefore, we propose an interdisciplinary approach to improve the understanding of drought impact emergence using this theory. When employing prospect theory in this context, drought impacts are considered failed welfare expectations ("prospects") due to water shortage. A shifting baseline after prolonged exposure to drought can therefore mitigate experienced drought impacts. We demonstrate that this theory can also contribute to explaining socio-hydrological phenomena, such as reservoir effects. This new approach can help bridge natural science and social science perspectives, resulting in integrated drought management that considers the local context.

The metabolome is the biochemical basis of plant form and function, but we know little about its macroecological variation across the plant kingdom. Here, we used the plant functional trait concept to interpret leaf metabolome variation among 457 tropical and 339 temperate plant species. Distilling metabolite chemistry into five metabolic functional traits reveals that plants vary on two major axes of leaf metabolic specialization—a leaf chemical defense spectrum and an expression of leaf longevity. Axes are similar for tropical and temperate species, with many trait combinations being viable. However, metabolic traits vary orthogonally to life-history strategies described by widely used functional traits. The metabolome thus expands the functional trait concept by providing additional axes of metabolic specialization for examining plant form and function.

Background and aims: Ectopic lipid storage is implicated in type 2 diabetes pathogenesis; hence, exercise to deplete stores (i.e., at the intensity that allows for maximal rate of lipid oxidation; MLO) might be optimal for restoring metabolic health. This intensity (“Fatmax”) is estimated during incremental exercise (“Fatmax test”). However, in “the field” general recommendations exist regarding a range of percentages of maximal heart rate (HR) to elicit MLO. The degree to which this range is aligned with measured Fatmax has not been investigated. We compared measured HR at Fatmax, with maximal HR percentages within the typically recommended range in a sample of 26 individuals (Female: n = 11, European ancestry: n = 17). Methods and results: Subjects completed a modified Fatmax test with a 5-min warmup, followed by incremental stages starting at 15 W with work rate increased by 15 W every 5 min until termination criteria were reached. Pulmonary gas exchange was recorded and average values for V˙ o2 and V˙ co2 for the final minute of each stage were used to estimate substrate-oxidation rates. We modeled lipid-oxidation kinetics using a sinusoidal model and expressed MLO relative to peak V˙ o2 and HR. Bland-Altman analysis demonstrated lack of concordance between HR at Fatmax and at 50%, 70%, and 80% of age-predicted maximum with a mean difference of 23 b·min−1. Conclusion: Our results indicate that estimated “fat-burning” heart rate zones are inappropriate for prescribing exercise to elicit MLO and we recommend direct individual exercise lipid oxidation measurements to elicit these values.

Purpose of Review: There is a growing interest in understanding the health effects of exposure to per- and polyfluoroalkyl substances (PFAS) through the study of the human metabolome. In this systematic review, we aimed to identify consistent findings between PFAS and metabolomic signatures. We conducted a search matching specific keywords that was independently reviewed by two authors on two databases (EMBASE and PubMed) from their inception through July 19, 2022 following PRISMA guidelines. Recent Findings: We identified a total of 28 eligible observational studies that evaluated the associations between 31 different PFAS exposures and metabolomics in humans. The most common exposure evaluated was legacy long-chain PFAS. Population sample sizes ranged from 40 to 1,105 participants at different stages across the lifespan. A total of 19 studies used a non-targeted metabolomics approach, 7 used targeted approaches, and 2 included both. The majority of studies were cross-sectional (n = 25), including four with prospective analyses of PFAS measured prior to metabolomics. Summary: Most frequently reported associations across studies were observed between PFAS and amino acids, fatty acids, glycerophospholipids, glycerolipids, phosphosphingolipids, bile acids, ceramides, purines, and acylcarnitines. Corresponding metabolic pathways were also altered, including lipid, amino acid, carbohydrate, nucleotide, energy metabolism, glycan biosynthesis and metabolism, and metabolism of cofactors and vitamins. We found consistent evidence across studies indicating PFAS-induced alterations in lipid and amino acid metabolites, which may be involved in energy and cell membrane disruption.

Coherent elastic neutrino-nucleus scattering (CE$\nu$NS) is a process in which neutrinos scatter on a nucleus which acts as a single particle. Though the total cross section is large by neutrino standards, CE$\nu$NS has long proven difficult to detect, since the deposited energy into the nucleus is $\sim$ keV. In 2017, the COHERENT collaboration announced the detection of CE$\nu$NS using a stopped-pion source with CsI detectors, followed up the detection of CE$\nu$NS using an Ar target. The detection of CE$\nu$NS has spawned a flurry of activities in high-energy physics, inspiring new constraints on beyond the Standard Model (BSM) physics, and new experimental methods. The CE$\nu$NS process has important implications for not only high-energy physics, but also astrophysics, nuclear physics, and beyond. This whitepaper discusses the scientific importance of CE$\nu$NS, highlighting how present experiments such as COHERENT are informing theory, and also how future experiments will provide a wealth of information across the aforementioned fields of physics., Comment: contribution to Snowmasss 2021. The significance of personal tutoring continues to increase as a result of contextual developments and the outcomes of key research on student retention and success, and yet these developments simultaneously create significant challenges in delivery within the pastoral model of personal tutoring. In addition, it remains an under-developed and under-researched area. Personal tutors’ needs and concerns have been established, and assessment of an intervention to address them has been recommended. This study examines the impact of the intervention of tailored professional development materials for tutoring within a pastoral model created in response to these issues. It reveals the usefulness of this developmental support and the need for such guidance for this work. It is argued that there are implications in terms of approaches to tutoring within this pastoral model, developmental support provision and a need for consistency of standards in personal tutoring across the sector.

Aims The aim of this study was to assess medium-term improvements following total hip arthro-plasty (THA), and to evaluate what effect different preoperative Oxford Hip Score (OHS) thresholds for treatment may have on patients’ access to THA and outcomes. Methods Patients undergoing primary THA at our institution with an OHS both preoperatively and at least four years postoperatively were included. Rationing thresholds were explored to identify possible deprivation of OHS improvement. Results Overall, 2,341 patients were included. Mean OHS was 19.7 (SD 8.2) preoperatively and 39.7 (SD 9.8) at latest follow-up. An improvement of at least eight-points, the minimally impor-tant change (MIC), was seen in 2,072 patients (88.5%). The mean improvement was 20.0 points (SD 10.5). If a rationing threshold of OHS of 20 points had been enforced, 90.8% of those treated would have achieved the MIC, but only 54.3% of our cohort would have had access to surgery; increasing this threshold to 32 would have enabled 89.5% of those treated to achieve the MIC while only depriving 6.5% of our cohort. The ‘rationed’ group of OHS > 20 had significantly better OHS at latest follow-up (42.6 vs 37.3; p < 0.001), while extending the rationing threshold above 32 showed postoperative scores were more significantly af-fected by the ceiling effect of the OHS. Conclusion The OHS was not designed as a tool to ration healthcare, but if it had been used at our institution for this cohort, applying an OHS threshold of 20 to routine THA access would have excluded nearly half of patients from having a THA; a group in which over 85% had a significant improvement in OHS. Where its use for rationing is deemed necessary, use of a higher threshold may be more appropriate to ensure a better balance between patient access to treatment and chances of achieving good to excellent outcomes.

Climate change and other global change drivers threaten plant diversity in mountains worldwide. A widely documented response to such environmental modifications is for plant species to change their elevational ranges. Range shifts are often idiosyncratic and difficult to generalize, partly due to variation in sampling methods. There is thus a need for a standardized monitoring strategy that can be applied across mountain regions to assess distribution changes and community turnover of native and non-native plant species over space and time. Here, we present a conceptually intuitive and standardized protocol developed by the Mountain Invasion Research Network (MIREN) to systematically quantify global patterns of native and non-native species distributions along elevation gradients and shifts arising from interactive effects of climate change and human disturbance. Usually repeated every five years, surveys consist of 20 sample sites located at equal elevation increments along three replicate roads per sampling region. At each site, three plots extend from the side of a mountain road into surrounding natural vegetation. The protocol has been successfully used in 18 regions worldwide from 2007 to present. Analyses of one point in time already generated some salient results, and revealed region-specific elevational patterns of native plant species richness, but a globally consistent elevational decline in non-native species richness. Non-native plants were also more abundant directly adjacent to road edges, suggesting that disturbed roadsides serve as a vector for invasions into mountains. From the upcoming analyses of time series, even more exciting results can be expected, especially about range shifts. Implementing the protocol in more mountain regions globally would help to generate a more complete picture of how global change alters species distributions. Though the total cross section is large by neutrino standards, CE$\nu$NS has long proven difficult to detect, since the deposited energy into the nucleus is $\sim$ keV. In 2017, the COHERENT collaboration announced the detection of CE$\nu$NS using a stopped-pion source with CsI detectors, followed up the detection of CE$\nu$NS using an Ar target. The detection of CE$\nu$NS has spawned a flurry of activities in high-energy physics, inspiring new constraints on beyond the Standard Model (BSM) physics, and new experimental methods. The CE$\nu$NS process has important implications for not only high-energy physics, but also astrophysics, nuclear physics, and beyond. Comparison of SOC stock changes following medium-term (5 and 10 years) and long-term (>50 years) warming revealed that all SOC stock reduction occurred within the first 5 years of warming, after which continued warming no longer reduced SOC stocks. This rapid equilibration of SOC observed in Andosol suggests a critical role for ecosystem adaptations to warming and could imply short-lived soil carbon-climate feedbacks. Our data further revealed that the soil C loss occurred in all aggregate size fractions and that SOC stock reduction was only visible in topsoil (0-10cm). SOC stocks in subsoil (10-30cm), where plant roots were absent, showed apparent conservation after >50 years of warming. Tundra ecosystems hold large stocks of soil organic matter (SOM), likely due to low temperatures limiting rates of microbial SOM decomposition more than those of SOM accumulation from plant primary productivity and microbial necromass inputs. Here we test the hypotheses that distinct tundra vegetation types and their carbon supply to characteristic rhizosphere microbes determine SOM cycling independent of temperature. In the subarctic Scandes, we used a three-way factorial design with paired heath and meadow vegetation at each of two elevations, and with each combination of vegetation type and elevation subjected during one growing season to either ambient light (i.e., ambient plant productivity), or 95% shading (i.e., reduced plant productivity). We assessed potential above- and belowground ecosystem linkages by uni- and multivariate analyses of variance, and structural equation modelling. We observed direct coupling between tundra vegetation type and microbial community composition and function, which underpinned the ecosystem’s potential for SOM storage. Greater primary productivity at low elevation and ambient light supported higher microbial biomass and nitrogen immobilisation, with lower microbial mass-specific enzymatic activity and SOM humification. Congruently, larger SOM at lower elevation and in heath sustained fungal-dominated microbial communities, which were less substrate-limited, and invested less into enzymatic SOM mineralisation, owing to a greater carbon-use efficiency (CUE). Our results highlight the importance of tundra plant community characteristics (i.e., productivity and vegetation type), via their effects on soil microbial community size, structure and physiology, as essential drivers of SOM turnover. Little is known about the bacteria that reside in the human gallbladder and the mechanisms that allow them to survive within this harsh environment. Here we describe interactions between two strains from a human bile sample, one Ruminococcus gauvreauii (IPLA60001), belonging to the Lachnospiraceae family, and the other, designated as Ruminococcoides bili (IPLA60002; DSM 110008) most closely related to Ruminococcus bromii within the family Ruminococcaceae. We provide evidence for bile salt resistance and sporulation for these new strains. Both differed markedly in their carbohydrate metabolism. The R. bili strain mainly metabolized resistant starches to form formate, lactate and acetate. R. gauvreauii mainly metabolized sugar alcohols, including inositol and also utilized formate to generate acetate employing the Wood Ljungdahl pathway. Amino acid and vitamin biosynthesis genomic profiles also differed markedly between the two isolates, likely contributing to their synergistic interactions, as revealed by transcriptomic analysis of cocultures. Transcriptome analysis also revealed that R. gauvreauii IPLA60001 is able to grow using the end-products of starch metabolism formed by the R. bili strain such as formate, and potentially other compounds (such as ethanolamine and inositol) possibly provided by the autolytic behavior of R. bili.

Metabolites produced by microbial fermentation in the human intestine, especially short-chain fatty acids (SCFAs), are known to play important roles in colonic and systemic health. Our aim here was to advance our understanding of how and why their concentrations and proportions vary between individuals. We have analysed faecal concentrations of microbial fermentation acids from 10 human volunteer studies, involving 163 subjects, conducted at the Rowett Institute, Aberdeen, UK over a 7-year period. In baseline samples, the % butyrate was significantly higher, whilst % iso-butyrate and % iso-valerate were significantly lower, with increasing total SCFA concentration. The decreasing proportions of iso-butyrate and iso-valerate, derived from amino acid fermentation, suggest that fibre intake was mainly responsible for increased SCFA concentrations. We propose that the increase in % butyrate among faecal SCFA is largely driven by a decrease in colonic pH resulting from higher SCFA concentrations. Consistent with this, both total SCFA and % butyrate increased significantly with decreasing pH across five studies for which faecal pH measurements were available. Colonic pH influences butyrate production through altering the stoichiometry of butyrate formation by butyrate-producing species, resulting in increased acetate uptake and butyrate formation, and facilitating increased relative abundance of butyrate-producing species (notably Roseburia and Eubacterium rectale).

The significance of personal tutoring continues to increase as a result of contextual developments and the outcomes of key research on student retention and success, and yet these developments simultaneously create significant challenges in delivery within the pastoral model of personal tutoring. In addition, it remains an under-developed and under-researched area. Personal tutors’ needs and concerns have been established, and assessment of an intervention to address them has been recommended. This study examines the impact of the intervention of tailored professional development materials for tutoring within a pastoral model created in response to these issues. It reveals the usefulness of this developmental support and the need for such guidance for this work. It is argued that there are implications in terms of approaches to tutoring within this pastoral model, developmental support provision and a need for consistency of standards in personal tutoring across the sector.

Study region: The Akaki catchment is found in the Upper Awash River Basin in Ethiopia. Study focus: Understanding the accuracy of rainfall forecasts in the data-scarce urban catchment has a multitude of benefits given the increased urban flood risk caused by climate change and urbanization. In this study, accuracy of the weather research and forecasting (WRF) model rainfall forecast was evaluated using citizen science data. Categorical and continuous accuracy evaluation metrics were used beside gauge representativeness effect. New hydrological insights for the region: The rainfall forecasts performance accuracy is high for 1–3-days lead-time but deteriorates for 4–5-days lead-time. The WRF model captured the temporal dynamics and the rainfall amount according to the estimated KGE values. The model has relatively higher detection performance for no rain and light rain events (< 6 mm/day), but it has lower performance for moderate and heavy rain events (> 6 mm/day). Use of data from a single rain gauge misrepresents the accuracy level of the rainfall forecast in the study area. The gauge representativeness error contributed a variance of 28.08–83.33 % to the variance of WRF-gauge rainfall difference. Thus, the use of citizen science rainfall monitoring program is an essential alternative source of information where in-situ rainfall monitoring is limited that can be used to understand the “true” accuracy of WRF rainfall forecasts.

Drought monitoring and early warning systems (DEWSs) are seen as helpful tools to tackle drought at an early stage and reduce the possibility of harm or loss. They usually include indices attributed to meteorological, agricultural and/or hydrological drought: physically based drought drivers. These indices are used to determine the onset, end and severity of a drought event. Drought impacts, like water and food securities, are less monitored or even not included in DEWSs. Therefore, the likelihood of experiencing these impacts is often simply linearly linked to drivers of drought. The aim of this study is to evaluate the validity of the assumed direct linkage between drivers of drought and water and food insecurity impacts of drought. We reviewed scientific literature on both drivers and impacts of drought. We conducted a bibliometric analysis based on 5000+ scientific studies in which selected drought indices (drivers) and drought-related water and food insecurities (impacts) were mentioned in relation to a geographic area. Our review shows that there is a tendency in scientific literature to focus on drivers of drought, with the preferred use of meteorological and remotely sensed drought indices. Studies reporting drought impacts are more localised, with relatively many studies focusing on sub-Saharan Africa and Australasia for impacts with regard to food security and water security, respectively. Our review further suggests that studies of food and water insecurity impacts related to drought are dependent on both the physical and human processes occurring in the geographic area, i.e. the local context. With the aim of increasing the relevance and utility of the information provided by DEWSs, we argue in favour of additional consideration of drought impact indices oriented towards sustainable development and human welfare.

Drought management is currently informed by a variety of approaches, mostly responding to drought crisis when it happens. Toward more effective and integrated drought management, we introduce a conceptual drought diagnosis framework inspired by diagnostic concepts from the field of medicine. This framework comprises five steps: 1. Initial diagnostic assessment; 2. Diagnostic testing; 3. Consultation; 4. Communication of the diagnosis; and 5. Treatment and prognosis. To illustrate the need for the proposed approach, four case studies of recently drought-affected regions were selected: the city of Cape Town, the state of California, the Northeast region of Brazil, and the Horn of Africa. Contrasting elements for these cases include the geographic extent and political boundaries, climate, socio-economics, and the relevance of different water resources (e.g., rainfall, reservoirs, and aquifers). For each case, we identified documented practices and policies and reflected on them in terms of drought misdiagnosis or incomplete diagnosis that have aggravated socio-economic and environmental drought impacts. A common example is the preference for technical solutions (e.g., installing infrastructure to augment water supply), rather than measures that reduce vulnerability. Analysis of these four drought case studies confirmed the anticipated need for a comprehensive approach to drought diagnosis for more successful treatment and prevention of drought. Using an analogy with medical science can be helpful toward comprehensively diagnosing droughts for a variety of contexts and assessing the effectiveness of proposed interventions. This framework can help drought managers to be more proactive in enabling drought-affected regions to become more drought resilient in the future.
