Objectives In this study, the antiproliferative activity of 3 phenyl 4-(2-oxo-3-alkylimidazolidin-1-yl)benzenesulfonates (PAIB-SOs) was assessed in a time-dependent manner together with their hepatic stability and metabolism using human, mouse and rat liver microsomes. Methods CEU-818, -820 and -913 were selected as promising hit compounds. Their antiproliferative activity on human breast carcinoma MCF-7 cells was evaluated using escalating concentrations of drugs at 24, 36 and 48 h and the sulforhodamine B assay. Their hepatic stability was evaluated by HPLC-UV of extracts obtained from human, mouse and rat liver microsomes. Key findings The antiproliferative activity of PAIB-SOs is concentration and time-dependent and requires between 24 and 36 h of contact with MCF-7 cells to detect a significant antiproliferative activity. PAIB-SOs stability in microsomes usually decreases following this order: human ≈ (rat > mouse). The CEU-913 exhibits the longest half-life in rat and human liver microsomes while the CEU-820 exhibits the longest half-life in mouse liver microsomes. Conclusions Our in vitro results suggest that PAIB-SOs should have a minimum contact time of 24 h with the tumour to trigger significant antitumoural activity. The activity of mouse liver microsomes towards PAIB-SOs is higher than rat microsomes and tends to be higher than human liver microsomes.

The dyslipidemic state of visceral obesity is characterized by increased plasma triglyceride (TG) levels, low HDL-cholesterol concentrations and alterations in LDL composition and concentration. A functional, non-coding −514C>T single nucleotide polymorphism (SNP) of the hepatic lipase gene (LIPC) has been related to variation in HDL-cholesterol concentrations. Objectives: To investigate the hypotheses that the LIPC −514C>T polymorphism may be associated with a deteriorated lipoprotein-lipid profile and that environmental factor, such as abdominal obesity, alters this association. Methods: A total of 235 French-Canadian men from the greater Quebec City area were assigned into three groups on the basis of their LIPC −514C>T SNP, including 149 CC homozygotes, 75 CT heterozygotes, and 11 TT homozygotes. Results: In the present study, the highest values of BMI, waist circumference, and accumulation of visceral adipose tissue (VAT) were observed among TT homozygotes (p<0.05). After adjustment for age and BMI, TT homozygotes still showed higher plasma apolipoprotein (apo) AI and HDL-TG concentrations than the two other groups (p<0.05). When the two genotype groups (CC vs CT/TT) were further divided on the basis of VAT accumulation using a cut-off point of 130 cm2 (high vs low) it appears that irrespective of the genotype subjects with low VAT had higher HDL2-cholesterol concentrations (p<0.0001). However, lean carriers of the T allele had higher plasma HDL2-cholesterol levels than lean CC homozygotes. The beneficial effect of the T allele on plasma HDL2-cholesterol levels was abolished in the presence of visceral obesity (VAT>130 cm2). Conclusion: In summary, the presence of visceral obesity attenuates the impact of the LIPC −514C>T polymorphism on plasma HDL2-cholesterol levels.

Hyperapobetalipoproteinemia is a common feature of the metabolic syndrome and could result from the interaction between genetic and dietary factors. The objective of this study was to verify whether dietary fat intake interacts with the T94A polymorphism of the liver fatty acid-binding protein (LFABP) gene to modulate plasma apolipoprotein (apo) B levels. Dietary fat and saturated fat intakes were obtained by a dietitian-administered food frequency questionnaire and the LFABP T94A genotype was determined by a PCR-RFLP based method in 623 French-Canadian men recruited through the Chicoutimi Lipid Clinic (279 T94/T94, 285 T94/A94, and 59 A94/A94). The LFABP T94A polymorphism was not associated with plasma apo B levels when fat intake was not taken into consideration. However, in a model including the polymorphism, fat intake expressed as a percentage of total energy intake, the interaction term and covariates, the variance in apo B concentrations was partly explained by the LFABP T94A polymorphism (5.24%, p=0.01) and by the LFABP T94A ∗ fat interaction (6.25%, p=0.005). Results were similar when saturated fat replaced fat intake in the model (4.49%, p=0.02 for LFABP T94A and 6.43%, p=0.004 for the interaction). Moreover, in men consuming more than 30% of energy from fat, the odds ratio for having plasma apo B levels above 1.04 g/L for A94 carriers was of 0.40 (p=0.02) compared to T94/T94 homozygotes. Results were similar for carriers of the A94 allele consuming more than 10% of energy from saturated fat (OR: 0.32, p=0.005). In conclusion, T94/T94 exhibit higher apo B levels whereas carriers of the A94 allele seem to be protected against high apo B levels when consuming a high fat and saturated fat diet. These findings reinforce the importance to take into account gene–diet interactions in the prevention and management of the metabolic syndrome.

Fenofibrate, a peroxisome proliferated activated receptor alpha (PPARa) agonist, has been shown to decrease plasma triglyceride (TG) and increase plasma highdensity lipoprotein (HDL) cholesterol levels despite a large interindividual variation in the response. Fenofibrate-activated PPARa binds to a DNA sequence element termed PPAR response element (PPRE) present in regulatory regions of target genes. A PPRE has been identified in the proximal 5¢ flanking region of the gene encoding the liver fatty acid binding protein (LFABP). LFABP is a small cytosolic protein of 14 kDa present in the liver and the intestine and is a member of the superfamily of the fatty acid binding proteins (FABPs). FABPs play a role in the solubilization of long-chain fatty acids (LCFAs) and their CoA-ester to various intracellular organelles. FABPs serves as intracellular acceptors of LCFAs, and they may also have an impact in ligand-dependent transactivation of PPARs in trafficking LCFAs to the nucleus. Since PPARs are known to regulate the transcription of many genes involved in lipid metabolism, the importance of LFABP in fatty acid uptake has to be considered. The aim of this study was to verify whether genetic variations in the LFABP gene may impact on plasma lipoprotein/lipid levels in the fasting state as well as on the response to a lipid-lowering therapy with fenofibrate on plasma lipids and obesity variables. We also wanted to verify whether the presence of the PPARa L162V mutation interacts with genetic variants in LFABP gene. To achieve this goal, we first determined the genomic structure of the human LFABP gene and then designed intronic primers to sequence the coding regions, all exon-intron splicing boundaries, and the promoter region of the gene in 24 patients showing divergent plasma lipoprotein/lipid response to fenofibrate. L’effet protecteur du jeûne a été observé dans plusieurs domaines de la santé. Pour l’étudier, un modèle de foie de rat perfusé ex vivo a ici été utilisé. Dans ce modèle, nous n’avons pas observé, pour un jeûne de 18h, l’effet protecteur du jeûne rapporté pour le foie in situ. Nous avons cependant souligné l’importance du coût énergétique du mécanisme protecteur du foie isolé soumis au stress expérimental de l’ischémie/reperfusion et corrélé la protection à un taux plus élevé de glycogène et une charge énergétique nécessitant le dépassement d’un seuil en deçà duquel la protection s’estompait. L’administration de substrats énergétiques (lactate et alanine) nous a permis de confirmer le besoin énergétique de la protection du foie isolé. Ensuite, l’accroissement de la durée du jeûne du rat donneur de 18 à 24 heures s’est avérée hépatoprotectrice et, plus globalement, a montré que la capacité de mobilisation énergétique et, à ce titre, l’autophagie contribuaient à l’hépatoprotection, le coût énergétique bien connu de l’autophagie étant en adéquation avec les précédents travaux du doctorat. Trois voies de signalisation candidates pour l’activation de l’autophagie, impliquant AMPK, HMGB1 et ADP, ont été étudiées. La phosphorylation de l’AMPK était augmentée dans le foie de rat à jeun 24h vs 18h. Toutefois, l’ajout d’AICAR, un activateur de l’AMPK, bien qu’augmentant sa phosphorylation dans le foie isolé de rat à jeun 18h, n’a pas induit de protection. L’accumulation cytoplasmique d’HMGB1, connue pour induire l’autophagie, n’a montré aucune corrélation avec les marqueurs de la cytolyse hépatique (LDH) et de l’autophagie (rapport LC3II/Actine). L’ADP, dans ses rapports ADP/(AMP+ADP+ATP) et ADP/(AMP+ATP), était plus élevée pour les foies de rat à jeun 24h et a été corrélée à l’hépatoprotection. Des études ont rapporté des différences entre les sexes dans le développement et la fonctionnalité des systèmes de détoxification dans des tissus non-reproductifs, tels que le foie. L’objectif principal de mon projet était de mieux comprendre ce dimorphisme sexuel impliqué dans le métabolisme et la communication cellulaire par les jonctions lacunaires dans le foie. Nos données ont indiqué que les rats mâles et femelles démontrent une réponse différente suite à une exposition à l’hexachlorobenzène. Les femelles exprimaient des niveaux plus élevés de cytochrome p450 1A1 et des niveaux plus faibles de l’uroporphyrinogène décarboxylase, comparé aux mâles. De plus, nos résultats ont démontré un dimorphisme sexuel dans l’expression de la Cx32 et la Cx43, dans le foie, au cours du développement. Des cellules souches hépatiques purifiées ont été utilisées afin de développer un modèle in vitro de culture tridimensionnelle afin d’étudier les jonctions lacunaires. Les protéines des jonctions lacunaires (Cx26, Cx32 et Cx43) et la protéine des jonctions adhérentes (E-cadhérine) ont été détectées par immunofluorescence dans les organoïdes en prolifération et les organoïdes différenciés. Des niveaux plus élevés de la Cx26, Cx32 et d’E-cadhérine ont été observés dans les organoïdes différenciés comparés aux non différenciés. Les niveaux d’ARNm de la Cx43 étaient plus élevés dans les organoïdes non différenciés que les organoïdes différenciés. À une dose de 100 nM d’estradiol, les niveaux d’ARNm de la Cx26 étaient significativement augmentés. En conclusion, nos données indiquent que les jonctions lacunaires sont partiellement régulées par les hormones stéroïdiennes dans le foie. The detoxification enzyme UDP-glucuronosyltransferase UGT2B10 is specialized in the N-linked glucuronidation of many drugs and xenobiotics. Preferred substrates possess tertiary aliphatic amines and heterocyclic amines such as tobacco carcinogens and several anti-depressants and anti-psychotics. We hypothesized that alternative splicing (AS) constitutes a mean to regulate steady state levels of UGT2B10 and enzyme activity. We established the transcriptome of UGT2B10 in normal and tumoral tissues of multiple individuals. Highest expression was in the liver, where ten AS transcripts represented 50% of the UGT2B10 transcriptome in 50 normal livers and 44 hepatocellular carcinomas. One abundant class of transcripts involves a novel exonic sequence and leads to two alternative (alt.) variants with novel in-frame C-termini of 10 or 65 amino acids. Their hepatic expression was highly variable among individuals, correlated with canonical transcript levels, and was 3.5 fold higher in tumors. Evidence for their translation in liver tissues was acquired by mass spectrometry. In cell models, they co-localized with the enzyme and influenced the conjugation of amitriptyline and levomedetomidine by repressing or activating the enzyme (40-70%; P<0.01), in a cell context-specific manner. A high turnover rate for the alt. proteins, regulated by the proteasome, was observed in contrast to the more stable UGT2B10 enzyme. Moreover, a drug-induced remodelling of UGT2B10 splicing was demonstrated in the HepaRG hepatic cell model, which favored alt. variants expression over the canonical transcript. Our findings support a significant contribution of AS in the regulation of UGT2B10 expression in the liver with an impact on enzyme activity.

L'origine intra-tissulaire des cellules épithéliales non-parenchymateuses émergeant des cultures de cellules hépatiques demeure inconnue. Des cultures d'hépatocytes et de cellules non-parenchymateuses de foie de rat obtenues par des méthodes sélectives de dispersion du tissu ont été analysées à l'aide d'anticorps monoclonaux réagissant avec les cytokératines et la vimentine de foie de rat, d'anticorps polyvalents dirgés contre les cytokératines de sabots de vache et la vimentine de lentilles de porc, et d'anticorps monoclonaux dirigés contre des antigènes exposés en surface des cellules ovales et des canaux biliaires ou des hépatocytes. L'analyse par immunotransfert de protéines de cellules épithéliales non-parenchymateuses de foie de rat en culture révèle que l'anticorps anti-cytokératines d'hépatocytes de rat reconnaît une cytokératine de masse relative (Mr) 55 000, l'anticorps anti-cytokératines de sabots de vache réagit avec une cytokératine de Mr 52 000 et que les anticorps anti-vimentine réagissent avec la vimentine. Des analyses similaires avec les protéines de fibroblastes de rat en culture confirment que les anticorps anti-vimentine reconnaissent la vimentine. La coloration de coupes de foie congelé par double marquage en immunofluorescence utilisant les anticorps anti-cytokératines et anti-vimentine rélève un patron de réactivité unique pour chacune des différentes structures cellulaires du foie. Les hépatocytes sont colorés seulement par l'anticorps anti-cytokératine de Mr 55 000 tandis que les cellules sinusoidales réagissent seulement avec l'anti-vimentine. Cultures of rat hepatocytes and several types of nonparenchymal cells obtained by selective tissue dispersion procedures were typed with monoclonal antibodies to rat liver cytokeratin and vimentin, polyvalent antibodies to cow hoof cytokeratins and porcine lens vimentin, and monoclonal antibodies to surface membrane components of ductular oval cells and hepatocytes. Immunoblot analysis revealed that, in cultured rat liver nonparenchymal epithelial cells, the anti-rat hepatocyte cytokeratin antibody recognized a cytokeratin of relative mass (M,) 55 000 and the anti-cow hoof cytokeratin antibody reacted with a cytokeratin of M, 52 000, while the anti-vimentin antibodies detected vimentin in both cultured rat fibroblasts and nonparenchymal epithelial cells. Analyses on the specificity of anti-cytokeratin and anti-vimentin antibodies toward the various cellular structures of liver by double immunofluorescence staining of frozen tissue sections revealed unique reactivity patterns. For example, hepatocytes were only stained with anti-Mr 55 000 cytokeratin antibody, while the sinusoidal cells reacted only with the anti-vimentin antibodies. In contrast, epithelial cells of the bile ductular structures and mesothelial cells of the Glisson capsula reacted with all the anti-cytokeratin and anti-vimentin antibodies. It should be stressed, however, that the reaction of the anti-vimentin antibodies on bile ductular cells was weak. The same analysis on tissue sections using the anti-ductular oval cell antibody revealed that it reacted with bile duct structures but not with the Glisson capsula. Oval cells emerging in rat liver at the early period of 3-methyl-4-dimethylaminoazobenzene treatment constitute a mixed epithelial cell compartment with respect to alpha-fetoprotein (AFP) and cytokeratin differential expression, and include a subpopulation which exhibits a phenotype intermediate between ductular cells and hepatocytes (Germain et al., Cancer Res., 45:673-681, 1985). In the present study we have examined the developmental potential of ductular oval cells in primary culture and after in vivo transfer. The use of monoclonal and polyclonal antibodies directed against cytokeratins of Mr 39,000 (CK39), 52,000 (CK52), and 55,000 (CK55) and vimentin, and also monoclonal antibodies against exposed surface components of oval cells (BDS7) and normal hepatocytes (HES6) allowed us to establish the ductular phenotype of the oval cells. A highly enriched preparation of oval cells was obtained by perfusion/digestion of the liver with collagenase, treatment of the cell suspension with trypsin and DNase, selective removal of hepatocytes by panning using the anti-HES6 antibody, and cell separation by isopyknic centrifugation in a Percoll gradient. The procedure yielded about 8 x 10(7) cells, of which 95% expressed CK39, CK52, and BDS7, 84% gamma-glutamyl transpeptidase, and 5% albumin and AFP. The primary response of cultured oval cells to various combinations of growth and differentiation promoting factors was evaluated with respect to their capacity to initiate DNA synthesis as measured by [3H]thymidine labeling from day 1 to 3, and/or to produce albumin and AFP and express tyrosine aminotransferase. Culture in the presence of either serum or clot blood extract resulted in a low proliferative activity with less than 5% of the nuclei being labeled. Chez la vache laitière à haut rendement, la période de transition entourant la parturition est généralement accompagnée d’un déficit énergétique, d’un métabolisme des lipides perturbé et d’une augmentation de l’inflammation et du stress oxydatif. Afin d’améliorer la balance énergétique et de diminuer certains effets négatifs associés à la période de transition, il est fréquent que les producteurs administrent des diètes riches en lipides (ex. Mégalac®, riche en acide palmitique) aux vaches laitières à haut rendement. Par exemple, l’ajout de lipides saturés à la ration améliore la balance énergétique et diminue l’accumulation des triglycérides dans le foie. Cependant, plusieurs troubles métaboliques demeurent, tels que l’inflammation et le stress oxydatif, pouvant être observés entre autres au niveau du foie. Comme alternative aux lipides saturés, l’ajout d’acides gras polyinsaturés (AGPIs) pourrait contribuer à améliorer la balance énergétique tout en diminuant certains effets négatifs associés à la période de transition. Toutefois, les AGPIs sont sujets aux phénomènes de peroxydation ce qui pourrait augmenter le stress oxydatif chez l’animal. Pour limiter ces dommages oxydatifs, l’utilisation de la graine de lin entière, riche en acides gras oméga-3 (n-3) et en lignans (antioxydants), en supplément alimentaire apparaît comme une avenue prometteuse. Des travaux antérieurs rapportent qu’une supplémentation en différents dérivés de la graine de lin (ex. AGPIs, lignans) module l’expression de certains gènes impliqués dans la réponse au stress oxydatif et le métabolisme des lipides dans la glande mammaire de la vache laitière. Cependant, il n’existe aucune étude rapportant l’effet de divers composés de la graine de lin sur le métabolisme lipidique et certains marqueurs de stress oxydatif dans le foie de la vache laitière. Cet article discute des nouveaux facteurs de bon pronostic identifiés dans l’hépatite alcoolique. Outre la prolifération des hépatocytes - phénomène évident mais pouvant être compromis en cas d’atteinte sévère du foie -, la prolifération des cellules progénitrices est désormais associée à un meilleur pronostic. Le rôle des macrophages hépatiques et de facteurs de croissance comme l'HGF, ou de prolifération comme le SPINK1 ou le TWEAK, est également discuté.

Le carcinome hépatocellulaire figure parmi les cancers les plus meurtriers et, encore en 2015, les infections aux virus de l'hépatite B et C sont en tête de lice des principales causes d'apparition de cancer du foie. En effet, 80% des cas de carcinomes hépatocellulaires sont attribuables à une infection virale par un de ces deux agents pathogènes. D'un point de vue épidémiologique, le carcinome hépatocellulaire n'est pas celui qui frappe le plus d'individus en Amérique du Nord. En effet, seulement une personne sur 264 sera affectée par ce cancer. Toutefois, il est important de l'étudier en raison de son faible taux de survie, ce qui le classe en cinquième place des cancers les plus meurtriers. Dernièrement, les recherches font mention des modifications dans l'épissage alternatif comme mécanisme d’apparition de plusieurs types de cancer, notamment les cancers du sein, de l'ovaire et de la prostate. C'est pour cette raison que nous avons investigué l'implication des virus de l'hépatite B et C dans l'épissage alternatif de gènes cellulaires, afin de mettre à jour un nouveau mécanisme de carcinogénèse utilisé par les oncovirus pour transformer leurs cellules hôtes. L'étude présentée dans ce mémoire rapporte des changements dans l'épissage alternatif de plusieurs gènes cellulaires, lorsque l’on évalue les patrons d’épissage alternatif des transcrits provenant d’analyse de séquençage d’ARN à haut débit de tumeurs du foie comparativement à des tissus hépatiques sains. Ces analyses ont permis de déterminer certains changements attribuables à la présence de chacun des deux virus hépatotropiques au niveau de l’épissage alternatif, et d’identifier certains changements qui pourraient impliquer la transformation du tissu sain en tissu cancéreux. Malgré de nombreuses améliorations dans la prise en charge de la polykystose rénale autosomique dominante (PKRAD) au cours des 20 dernières années, l’infection bactérienne du rein ou du foie, avec, au premier chef, l’infection intrakystique, reste une complication sérieuse et potentiellement sévère. L’infection d’un kyste rénal est la plus fréquente. Le diagnostic différentiel avec une hémorragie intrakystique est essentiel : il se base sur la présentation clinique (en particulier la sévérité et la durée de la fièvre), le taux de protéine C-réactive (CRP) et la leucocytose, ainsi que la densité tomodensitométrique du kyste suspect. L’infection d’un kyste hépatique survient surtout chez les patients ayant une hépatomégalie kystique, est parfois redoutable et a une propension à la récidive. Dans les deux localisations, l’examen d’imagerie le plus performant est la tomographie par émission de positons couplée à la tomodensitométrie utilisant comme marqueur le fluor 18 incorporé dans une molécule de glucose formant le [18F]-fluorodésoxyglucose ([18F]-FDG-TEP/TDM) et le traitement de première ligne est une antibiothérapie de 6 semaines par une fluoroquinolone. La ponction-aspiration du ou des kyste(s) infecté(s) est indiquée dans le traitement initial des kystes très volumineux et à titre diagnostique dans les kystes résistant à un traitement antibiotique bien conduit. Chez les patients candidats à la transplantation rénale, un antécédent d’infection kystique rénale récidivante justifie l’exérèse préalable du ou des rein(s) infecté(s), tandis qu’un antécédent d’infection kystique hépatique récidivante ou d’angiocholite récidivante fait envisager une transplantation hépatique. Parmi les manifestations extrarénales et extrahépatiques de la PKRAD, la diverticulose colique est la seule qui pourrait être associée à un risque plus élevé d’infection et ce, chez les patients dialysés et transplantés ; une observation qui mériterait d’être confirmée., Despite advances in the management of autosomal dominant polycystic kidney disease over the past two decades, infection of liver and kidney cysts remains a serious and potentially threatening complication. Kidney cyst infection is the most frequent complication. It is differentiated from hemorrhage by the clinical presentation (mainly the severity and duration of fever), C-reactive protein (CRP) and white blood cells levels, and the density of the suspected cyst on computed tomography. Liver cyst infection occurs more frequently in patients with large cysts volumes. It can be life threatening and has a tendency to recur. In both infections, the best radiological imaging technique is positron emission tomography after intravenous injection of [18F]-fluorodeoxyglucose combined with computed tomography. Treatment with a fluoroquinolone should be continued for 6 weeks. Cyst aspiration is necessary only when cysts are very large and/or when infection is resistant to antibiotic treatment. In patients who are candidates to kidney transplantation, a history of recurrent kidney cyst infection justifies pre-transplant nephrectomy, while a past history of recurrent liver cyst infection or angiocholitis leads to consider liver transplantation. Among extrarenal and extrahepatic complications of polycystic disease, colic diverticulosis is reported to be associated with increased risk of infection in patients on hemodialysis and after kidney transplantation. However, this observation needs to be confirmed.

L'enjeu est particulièrement important pour des filières à forte valeur ajoutée, comme le canard gras. Dans le cadre d'essais sur l'amélioration génétique du canard (2), des méthodes rapides – spectroscopie proche infrarouge (SPIR) – de prédiction de la qualité ont été testées pour le foie gras et pour le filet (magret). Il s'agissait de permettre la mesure sur un grand nombre d'échantillons de paramètres dont la méthode de référence est particulièrement longue (matières grasses [MG] par extraction Folch, rendement technologique, entre autres) et d'étudier la possibilité de les appliquer ensuite dans l'industrie. Les spectres des échantillons ont été acquis sur un spectromètre ASD Labspec, directement sur le produit entier (foie ou magret), sans préparation. L'étalonnage a été réalisé avec des mesures de composition chimique (matière sèche [MS], minéraux, protéines, MG) ou technologique (par exemple taux de fonte pour le foie, pertes à la cuisson pour le filet) (1). La précision des étalonnages a varié selon les paramètres (tableau I). Background: "Foie gras" is produced predominantly in France and about 90% of the commercialized product is obtained from male mule ducks. The melting rate (percentage of fat released during cooking) is the main criterion used to determine the quality of "foie gras". However, up to now the melting rate could not be predicted without causing liver damage, which means that selection programs could not use this criterion. Methods: Fatty liver phenotypes were obtained for a population of over 1400 overfed male mule ducks. The phenotypes were based on two types of near-infrared spectra (on the liver surface and on ground liver) in order to predict the melting rate and liver composition (ash, dry matter, lipid and protein contents). Genetic parameters were computed in multiple traits with a "sire-dam" model and using a Gibbs sampling approach. Results: The estimates for the genetic parameters show that the measured melting rate and the predicted melting rate obtained with two near-infrared spectrometer devices are genetically the same trait: genetic correlations are very high (ranging from +0.89 to +0.97 depending on the mule duck parental line and the spectrometer) and heritabilities are comparable. The predictions based on the spectra of ground liver samples using a laboratory spectrometer correlate with those based on the surface spectra using a portable spectrometer (from +0.83 to +0.95 for dry matter, lipid and protein content) and are particularly high for the melting rate (higher than +0.95). Although less accurate than the predictions obtained using the spectra of ground liver samples, the phenotypic prediction of the melting rate based on surface spectra is sufficiently accurate to be used by "foie gras" processors. Les anguilles d'Amérique (Anguilla rostrata) et d'Europe (Anguilla anguilla) ont vu leurs populations décliner de façon très importante depuis les années 1980. Comme pour d'autres facteurs (surpêche, obstacles à la migration, changements du climat et des courants marins), la contamination semble jouer un rôle dans les déclins observés. En particulier, des effets sur l'activité d'enzymes liées aux différentes capacités métaboliques peuvent ainsi se traduire par une influence sur la croissance, la capacité de nage, les capacités respiratoires, et l'accumulation de réserves énergétiques, et possiblement contribuer à compromettre leur migration et/ou leur reproduction dans la mer des Sargasses. Dans le cadre du projet IMMORTEEL, la présente étude avait pour but de déterminer si les contaminants inorganiques accumulés dans le foie et le muscle (Cd, Cu, Zn, Ag, As, Pb, Cr, Ni, Se et Hg) pouvaient affecter les capacités métaboliques des deux espèces d'anguilles au stade jaune. Les mesures d'activités d'enzymes du foie et du muscle associées aux capacités aérobies (citrate synthase, CS; cytochrome C oxydase, CCO), aux capacités anaérobies (lactate déshydrogénase, LDH), aux capacités glycolytiques (pyruvate kinase, PK) et au métabolisme des lipides (glucose-6-phosphate déshydrogénase, G6PDH) ont alors été faites. Dans le foie d'A. rostrata, les plus fortes corrélations obtenues indiquent que la PK et la CS sont liées positivement au Zn et au Pb respectivement, tandis que la G6PDH et la LDH sont principalement affectées négativement par l'Ag et le Ni respectivement. Au niveau du muscle, la PK, la CS et la CCO sont toutes affectées particulièrement par le Hg, même si seule la PK montre une corrélation positive. Du côté d'A. anguilla, la PK et la CS du foie sont toutes deux affectées par le Pb, mais de manière positive et négative respectivement, puis la LDH est liée négativement au Zn. La prévalence de l’obésité et des problèmes qui y sont associés (hypertension artérielle, dyslipidémie, hyperglycémie) inclus dans une entité clinique appelée syndrome métabolique, est croissante dans nos populations et devient un problème de santé mondial majeur. Un état inflammatoire chronique de bas grade a été démontré comme ayant un rôle dans le développement de l’insulinorésistance, un facteur clef du syndrome métabolique. Le tissu adipeux est un acteur important de ce processus. En effet, en cas d’obésité, le tissu adipeux est infiltré par des macrophages présentant un phénotype inflammatoire, dont les médiateurs influencent l’insulinosensibilité tant localement qu’à distance, dans les muscles et le foie. La résistance à l’insuline au niveau du foie résulte d’un défaut de suppression de production de glucose par les hépatocytes en réponse au stimulus insulinique. Ceci peut mener à une hyperglycémie et au diabète. Nous avons démontré le rôle central du foie dans l’initiation de l’insulinorésistance en réponse à l’alimentation riche en graisse chez la souris. En effet, les souris nourries avec une alimentation riche en graisse durant 3 jours présentent un gain pondéral significatif, une stéatose hépatique et une insulinorésistance spécifiquement hépatique, prouvée in vivo par la technique du clamp hyperinsulinémique-euglycémique et confirmée par une altération des voies de signalisation de l’insuline. Par contre, la captation périphérique de glucose de même que les voies de signalisation de l’insuline au niveau du muscle demeurent inaltérées. Contrastant avec l’inflammation décrite dans l’obésité, aucun changement n’a été mis en évidence au niveau des macrophages du tissu adipeux en termes de nombre ou de morphologie dans ce modèle à court terme. Characterization of Montreal municipal wastewater effluent (MWWE) has identified the presence of pharmaceuticals and persona} care products (PPCP), alkyphenols (AP), polychlminated biphenyls (PCB), polyaromatic hydrocarbons (PAH), and heavy metals. MWWE contains mixtures of xenobiotics that have been shown to disrupt normal metabolic and cellular functions. Previous studies have shown that fish exposed to MWWE have resulted in altered cellular signaling in the testis. These processes are facilitated by transmembrane proteins called connexins (Cx) which form gap junctions (GJ). GJ allow the passage of small molecules between neighbouring cells, called gap junction intercellular signaling (GJIC). Brook trout exposed to MWWE for 4 and 12 weeks showed altered testicular Cx levels at both ti me points. At 4 weeks there was a significant increase in both Cx43 and Cx31. At 12 weeks a decrease in Cx31 and an increase in Cx43 and Cx43.4 was observed. The presence of xenobiotics has been shown to target and disrupt Cx mRNA levels resulting in altered cellular functions. Similar results were reported in Spottail shiners living within the effluent plume of MWWE. Shiners displayed altered testicular Cx levels resulting in delayed spermatogenesis, increased vitellogenin levels, intersex characteristics and decreased sperm motility. MWWE has been shown to target Cxs in the testis causing an alteration in key signaling pathways. lnterestingly, few studies have examined the effects of MWWE on cellular signaling in the liver considering the important role it plays in xenobiotic metabolism. Xenobiotics can accumulate in the li ver if the complex signaling required to carry out the detoxification process is disrupted. GJIC plays an important role in the regulation of normal homeostatic processes in the liver including xenobiotic metabolism, bile secretion and glycogenolysis. La fibrose hépatique est le stigmate lésionnel de la progression de la maladie vers la cirrhose comme dans toutes les hépatopathies chroniques. Certains facteurs de risque cliniques environnementaux ont été identifiés. Toutefois, ils n’expliquent pas l’extrême variabilité individuelle de la progression de la fibrose. L’influence ethnique à développer une maladie plus sévère ainsi que les études de concordance des jumeaux mono- et di-zygotiques sur la prévalence de la cirrhose alcoolique ont suggéré l’existence de facteurs génétiques associés. Les nombreuses études de gènes candidats réalisées n’ont identifié que très peu de variants associés de manière reproductible. Pour l’HCC par exemple, le score de risque de cirrhose ou « CRS » a montré sa capacité à prédire une fibrose avancée dans diverses cohortes caucasiennes. Récemment, une étude d’association pangénomique (GWAS) dans la stéatohépatopathie non-alcoolique (NAFLD) a mis en évidence un singleton (single nucleotide polymorphism [SNP]) particulier (rs738409 C>G) dans le gène PNPLA3. Ce dernier s’est révélé être, dans diverses études, le SNP ayant l’impact le plus robuste et le plus reproductible dans cette maladie. Liver tissue predominantly consists of hepatocytes, which carry out metabolic functions and are responsible for bile production, and biliary cells or cholangiocytes, which line biliary ducts through which bile flows to the digestive tract. These two cell populations originate from common precursors called hepatoblasts. In mice, hepatocyte and biliary lineages segregate at embryonic stage E13. This process is controlled by a network of transcription factors. Biliary cells differentiate around branches of the portal vein and constitute a ring of cells, the ductal plate, where biliary duct formation is initiated. In order to characterize the transcriptional control network involved in biliary duct development, we have first identified new markers of biliary development, namely SOX9 and osteopontin. Using these new markers, we were able to uncover that biliary duct development occurs via an undescribed mode of tubulogenesis, duringwhich transient, primitive ductal structures are formed from the ductal plate. These transient structures are asymmetrical, that is they are lined by biliary cells on their portal side while they are lined by hepatoblasts on their parenchymal side. Maturation of these structures takes place by differentiation of hepatoblasts toward biliary cells, resulting in a radial symmetry of the biliary ducts which become entirely lined by cholangiocytes. In addition, we have revealed an early expression of two members of the transcription factor family Sry-related HMG box (SOX), namely SOX4 and SOX9, in biliary cells. The role of both genes was investigated by means of an in vivo knockout approach. This showed that SOX4 and SOX9 stimulate biliary duct morphogenesis and differentiation. Chez la souris, la ségrégation des lignées hépatocytaire et biliaire s’opère vers le jour embryonnaire E13 et est contrôlée par un réseau de facteurs de transcription. Les cellules biliaires se différencient autour des branches de la veine porte et constituent un anneau de cellules, appelé plaque ductale, à partir duquel se forment les canaux biliaires. En vue de caractériser le réseau de régulation transcriptionnelle opérant au cours du développement des canaux biliaires, nous avons dans un premier temps identifié de nouveaux marqueurs de développement biliaire, à savoir SOX9 et l'ostéopontine. A l'aide de ces marqueurs, nous proposons un nouveau modèle de tubulogenèse des canaux biliaires qui est caractérisée par la formation transitoire de structures ductales primitives à partir de la plaque ductale. Ces structures transitoires sont asymétriques, c'est-à-dire délimitées du côté portal par des cellules biliaires et du côté parenchymateux par des hépatoblastes. Leur maturation se fait par différenciation des hépatoblastes en cellules biliaires, conférant ainsi une symétrie radiaire aux canaux biliaires qui, à l'état mature, sont entièrement délimités par des cholangiocytes. Nous avons également découvert que deux membres de la famille des facteurs de transcription Sry-related HMG box (SOX), SOX4 et SOX9 sont exprimés dans les cellules biliaires dès le début du développement biliaire. Fenofibrate, a peroxisome proliferated activated receptor alpha (PPARa) agonist, has been shown to decrease plasma triglyceride (TG) and increase plasma highdensity lipoprotein (HDL) cholesterol levels despite a large interindividual variation in the response. Fenofibrate-activated PPARa binds to a DNA sequence element termed PPAR response element (PPRE) present in regulatory regions of target genes. A PPRE has been identified in the proximal 5¢ flanking region of the gene encoding the liver fatty acid binding protein (LFABP). LFABP is a small cytosolic protein of 14 kDa present in the liver and the intestine and is a member of the superfamily of the fatty acid binding proteins (FABPs). FABPs play a role in the solubilization of long-chain fatty acids (LCFAs) and their CoA-ester to various intracellular organelles. FABPs serves as intracellular acceptors of LCFAs, and they may also have an impact in ligand-dependent transactivation of PPARs in trafficking LCFAs to the nucleus. Since PPARs are known to regulate the transcription of many genes involved in lipid metabolism, the importance of LFABP in fatty acid uptake has to be considered. The aim of this study was to verify whether genetic variations in the LFABP gene may impact on plasma lipoprotein/lipid levels in the fasting state as well as on the response to a lipid-lowering therapy with fenofibrate on plasma lipids and obesity variables. Hyperapobetalipoproteinemia is a common feature of the metabolic syndrome and could result from the interaction between genetic and dietary factors. The objective of this study was to verify whether dietary fat intake interacts with the T94A polymorphism of the liver fatty acid-binding protein (LFABP) gene to modulate plasma apolipoprotein (apo) B levels. Dietary fat and saturated fat intakes were obtained by a dietitian-administered food frequency questionnaire and the LFABP T94A genotype was determined by a PCR-RFLP based method in 623 French-Canadian men recruited through the Chicoutimi Lipid Clinic (279 T94/T94, 285 T94/A94, and 59 A94/A94). The LFABP T94A polymorphism was not associated with plasma apo B levels when fat intake was not taken into consideration. However, in a model including the polymorphism, fat intake expressed as a percentage of total energy intake, the interaction term and covariates, the variance in apo B concentrations was partly explained by the LFABP T94A polymorphism (5.24%, p=0.01) and by the LFABP T94A ∗ fat interaction (6.25%, p=0.005). Results were similar when saturated fat replaced fat intake in the model (4.49%, p=0.02 for LFABP T94A and 6.43%, p=0.004 for the interaction). Moreover, in men consuming more than 30% of energy from fat, the odds ratio for having plasma apo B levels above 1.04 g/L for A94 carriers was of 0.40 (p=0.02) compared to T94/T94 homozygotes. Results were similar for carriers of the A94 allele consuming more than 10% of energy from saturated fat (OR: 0.32, p=0.005). In conclusion, T94/T94 exhibit higher apo B levels whereas carriers of the A94 allele seem to be protected against high apo B levels when consuming a high fat and saturated fat diet. These findings reinforce the importance to take into account gene–diet interactions in the prevention and management of the metabolic syndrome.

The dyslipidemic state of visceral obesity is characterized by increased plasma triglyceride (TG) levels, low HDL-cholesterol concentrations and alterations in LDL composition and concentration. A functional, non-coding −514C>T single nucleotide polymorphism (SNP) of the hepatic lipase gene (LIPC) has been related to variation in HDL-cholesterol concentrations. Objectives: To investigate the hypotheses that the LIPC −514C>T polymorphism may be associated with a deteriorated lipoprotein-lipid profile and that environmental factor, such as abdominal obesity, alters this association. Methods: A total of 235 French-Canadian men from the greater Quebec City area were assigned into three groups on the basis of their LIPC −514C>T SNP, including 149 CC homozygotes, 75 CT heterozygotes, and 11 TT homozygotes. Results: In the present study, the highest values of BMI, waist circumference, and accumulation of visceral adipose tissue (VAT) were observed among TT homozygotes (p < 0:05). After adjustment for age and BMI, TT homozygotes still showed higher plasma apolipoprotein (apo) AI and HDL-TG concentrations than the two other groups (p < 0:05). When the two genotype groups (CC vs CT/TT) were further divided on the basis of VAT accumulation using a cut-off point of 130 cm2 (high vs low) it appears that irrespective of the genotype subjects with low VAT had higher HDL2-cholesterol concentrations (p < 0:0001). However, lean carriers of the T allele had higher plasma HDL2-cholesterol levels than lean CC homozygotes. The beneficial effect of the T allele on plasma HDL2-cholesterol levels was abolished in the presence of visceral obesity (VAT > 130 cm2). Conclusion: In summary, the presence of visceral obesity attenuates the impact of the LIPC −514C>T polymorphism on plasma HDL2-cholesterol levels.

[Multi detector CT and MRI of benign liver tumors] With multidetector helical CT, the entire liver can be imaged rapidly with thin slices. High temporal and spatial resolution is important to improve multiphase examinations of the liver. The need to perform multiphase examinations to diagnose benign liver tumors should, however, be assessed by keeping in mind the risk of radiation and contrast-induced adverse events. MRI offers several advantages relative to CT. Excellent contrast resolution is obtained. Multiphase examinations can be performed in all patients. Reticuloendothelial contrast agents may be useful to characterize benign tumors. Several benign liver tumors, particularly hemangiomas and lesions of focal nodular hyperplasia, can be characterized with CT and MRI. Histologic examination is still needed to characterize some benign liver tumors.

Les résultats obtenus ont montré que les phosphatidylcholines et les phosphatidyléthanolamines sont les phospholipides majoritaires avec plus de 63 p. 100 des phospholipides totaux ; elles ont un taux plus élevé que chez d'autres espèces. Les phosphatidylinositols représentent le plus faible taux avec 5 p. 100 environ, alors que les sphingmyélines, qui semblent être à l'état de trace chez le rat, représentent plus 14 p. 100 chez le chamelon. Au niveau mitochondrial, les acides palmatique, linoléique, stéarique et oléique constituent la majorité des acides gras mitochondriaux. Contrairement à d'autres espèces, l'effet du sexe, de la masse corporelle et du régime alimentaire de l'animal n'influencent pas la composition en acides gras, notamment les longueurs de chaînes et insaturations.
