This article explores the role “whiteness” takes on in Mexico, where colonial, religious, and social heritages elevate it as an aesthetic ideal, simultaneously denying its underlying racism. It argues that skin tone is one of many physical and nonphysical features that together shape the concept of whiteness in a context of fluid, relational, and intertwined categories of class and racial classifications. Women in particular are pressured to “whiten” their bodies in adherence to beauty standards that reflect the collective aspiration of the country’s ethnically mixed society. Using empirical evidence, the article outlines Mexicans’ aesthetic perceptions and explores their attempts to approach these through bodily presentations and adjustments. It then discusses how the local beauty industry acts as a practical tool and a discursive mediator toward racialized appearances. Possessing its own historical, political, and racial background deeply entangled with whiteness, this sector reinforces the subjective basis of discriminatory practices in Mexico.

Anesthesiology and Pain Medicine - Abstract
To assess whether dual therapy with erenumab and onabotulinumtoxinA (BoNTA) was more effective than erenumab alone in chronic migraine.Calcitonin gene-related peptide (CGRP) is crucial in migraine. Erenumab binds to the canonical CGRP receptor in Aδ-fibers, and BoNTA prevents the release of CGRP from meningeal and extracranial C-fibers. It is still unknown whether dual therapy is more effective. This was a retrospective study in a Headache Unit. There was a thorough revision of charts of patients receiving erenumab from December 2019 to March 2021. The cohort was divided into three groups according to BoNTA at the start of erenumab: (1) WBT: were on BoNTA and maintained it as dual therapy; (2) WoBT: were on BoNTA and discontinued; (3) NoBT: were not on BoNTA. Primary endpoint was reduction in monthly headache days (MHD) at 12 weeks. Secondary endpoints were percent improvement and ≥50% reduction in MHD.Of 237 charts reviewed, 187 met the inclusion criteria. Seventy-three (39%) were included in WBT, 44 (23.5%) in WoBT, and 70 (37.4%) in NoBT. The reduction in MHD was less with dual therapy [WBT 4.7 ± 7.68, WoBT 5.12 ± 7.98 (p = 0.80), NoBT 8.21 ± 7.84 p = 0.009]. The percentage of improvement was higher in the erenumab-alone group [NoBT 35%, WoBT 22.3% (p = 0.92), WBT 21.7% (p = 0.001)]. The probability of achieving a ≥ 50% reduction in MHD was lower in WBT than in WoBT (OR 0.66, p = 0.35) and in the NoBT group (OR 0.57, p = 0.14).Our findings suggest that dual therapy is less effective than erenumab alone. However, since the design has multiple limitations, further prospective studies are required to validate these data.

- Abstract

Inborn errors of immunity are a group of genetic disorders caused by mutations that affect the development and/or function of several compartments of the immune system, predisposing patients to infections, autoimmunity, allergy and malignancies. In this regard, mutations that affect proteins involved in trafficking, priming, docking, or membrane fusion will impair the exocytosis of lytic granules of effector NK and cytotoxic T lymphocytes. This may predispose patients to hemophagocytic lymphohistiocytosis, a life-threatening immune disorder characterized by systemic lymphocyte and macrophage activation, and increased levels of cytokines, which lead to an uncontrolled hyperinflammation state and progressive multiorgan damage. In this review, we will describe a clinical case and recent advances in inborn errors of immunity predisposing to hemophagocytic lymphohistiocytosis. Summary sentence: Review of recent advances in inborn errors of immunity predisposing to hemophagocytic lymphohistiocytosis.

General Engineering - Abstract

General Materials Science - Abstract

Strengthening food security, in places where land and natural resources are limited or no longer available, is challenging. This is especially true for the production of staple food carbohydrates. Unlike some alternative foods, such as cultured meats, producing food carbohydrates using conventional agri-food approaches requires many natural resources, which are not available in some regions such as Singapore. Therefore, we must develop new, sustainable methods to enhance the quantity and nutritional quality of foods rich in carbohydrates. In this article, we review current developments in food security in the city-state of Singapore and emphasize the essential role of food carbohydrates in the food security plan. We discuss technology developments (i.e., indoor vertical farming, urban farming) used to enhance crop quality and production. We also make a few recommendations such as exploring underutilized and unconventional crops that are resilient and nutrient-dense, identifying hidden resources in local ecosystems (i.e., revalorizing agri-food processing by-products), and producing alternative carbohydrates (i.e., microbial and synthetic carbohydrates). Experience and approaches developed in Singapore provide an example to other regions and may inspire creativity in securing food availability.

Leptospirosis is a zoonotic disease with a high burden in Latin America, including northeastern Argentina, where flooding events linked to El Niño are associated with leptospirosis outbreaks. The aim of this study was to evaluate the value of using hydrometeorological indicators to predict leptospirosis outbreaks in this region. We quantified the effects of El Niño, precipitation, and river height on leptospirosis risk in Santa Fe and Entre Ríos provinces between 2009 and 2020, using a Bayesian modelling framework. Based on several goodness of fit statistics, we selected candidate models using a long-lead El Niño 3.4 index and shorter lead local climate variables. We then tested predictive performance to detect leptospirosis outbreaks using a two-stage early warning approach. Three-month lagged Niño 3.4 index and one-month lagged precipitation and river height were positively associated with an increase in leptospirosis cases in both provinces. El Niño models correctly detected 89% of outbreaks, while short-lead local models gave similar detection rates with a lower number of false positives. Our results show that climatic events are strong drivers of leptospirosis incidence in northeastern Argentina. Therefore, a leptospirosis outbreak prediction tool driven by hydrometeorological indicators could form part of an early warning and response system in the region.

Prickly pears (Opuntia ficus-indica) are potential sources of functional ingredients because they are rich in betalains and phenolic compounds. However, mentioned bioactives may degrade during storage when exposed to air, light, and heat which could limit their application. To increase the stability and bioaccessibility of prickly pear extracts, we compared the ultrasound-assisted freeze-dried microencapsulation of seven excipient mixtures. The physical and physico-chemical properties (humidity, hygroscopicity, thermal analysis and morphology) and the qualitative and quantitative analysis of betalains and phenolic compounds (measured by high performance liquid chromatography) were analysed in each microparticle formulation. Stability-improving factors such as low humidity and hygroscopicity were observed in all microparticles. However, microparticle morphology was influenced by the excipient formulation. Encapsulation efficiency was higher than 60% for betalains and phenolic acids, however, flavonoids encapsulation efficiency was 14–35%. Based on the previous, the three best microparticles were selected: 100% maltodextrin (E2); 50% maltodextrin, 25% microcrystalline cellulose, 15% hydroxyl‑propyl-methyl cellulose, and 10% xanthan gum (E5); and 100% β-cyclodextrin (E7). A static in vitro gastrointestinal digestion (INFOGEST method) was performed with these microparticles where the quantitative analysis of the bioactive compounds (HPLC) and their bioaccessibility was assessed. The bioaccessibility of bioactive compounds in encapsulated prickly pear extracts was improved when compared to the control. Microparticles containing maltodextrin and microcrystalline cellulose (E2) had the highest bioaccessibility and showed potential for the future formulation of functional foods., This work was supported by the Spanish Ministry of Science and Innovation [RTA2015-00044-C02-02 and PID2020-118300RB-C21] research projects.

Purpose: We evaluated the potential associations between the consumption of ultra-processed food (UPF) and the incidence of obesity among Uruguayan and Brazilian preschoolers. Design and methods:We conducted a longitudinal analysis using data from preschool children fromUruguay and Brazil. The “Health, child development and nutritional survey” (ENDIS) was conducted in Uruguay in 2013–2014 and 2015–2016. The Brazilian survey (Pelotas 2015 Birth Cohort) has measures from 2017 and 2019. The main outcome measure was obesity defined as body mass index (BMI) for age and sex ≥ +3 z-scores. The score of UPF consumption was the main exposure measured. Multilevel crude and adjusted Poisson regressions were performed to estimate risk ratios and the respective 95% Confidence Intervals (95% CI). Results: The overall incidence of obesity in this group of young Latin-American children with a mean age of 48 months was 4.1%. We observed a relationship between UPF and obesity with statistical significance (RR: 1.10 (95% CI, 1.02–1.18). Adjustment for weight at birth, age, sex, breastfeeding, country, and time between waves resulted in a similar relationship but lack of statistical significance. Conclusions: Whilst in this study we did not find strong evidence of an association between the incidence of obesity and the intake at baseline and currently of UPF, results suggest that higher UPF consumption is more favorable than reduced consumption for the development of obesity. Practice implications: The present study reinforces the importance of nutrition education and more effective public policies for promoting healthier food choices in early childhood. - Abstract

- Abstract
Communicating statistics in health risk communication is a fundamental part of managing public health emergencies. Effective communication requires careful planning and the anticipation of possible information demands from the population. The information should be clear, relevant, easy to understand, timely, accurate and precise, allowing the public to make informed decisions about protective behaviours. COVID-19, being a new disease, with little known about its characteristics and effects, has challenged governments and healthcare systems in all countries. This article discusses the statistical issues involved, and the experiences of risk communication in four countries – Canada, Chile, Ecuador and England. These countries have communicated risks differently, partly because of their different healthcare systems, as well as socioeconomic, cultural and political realities. During a pandemic, health authorities and governments must step up to the challenge of communicating statistical information under pressure and with urgency, when little is known about the disease, the situation is dynamic and evolving, and the general public is gripped with fear and anxiety. This is in addition to the existing challenges relating to the generation of data of different quality by diverse sources, and a public with varying levels of statistical literacy. From a statistical perspective, communiqués about risks and numbers should convey the uncertainty there is about the information, the inherent variabilities in the system, the precision and accuracy of estimates and the assumptions behind projections. Complex technical concepts, such as ‘flattening the curve’, ‘range in risk estimates’ and ‘projected trends,’ should be explained.

Pressure injury (PI) corresponds to a skin damage of ischemic aetiology that affects the integrity of the skin and is produced by prolonged pressure or friction between a hard internal and external surface. Treatment can be challenging when there is no resolution with usual care. The use of autologous platelet-rich plasma (APRP) gel arises as a therapeutic possibility in the presence of chronic pressure injuries. The case of a patient with chronic PI who has been treated with APRP is presented, achieving resolution of the lesion.

- Abstract
Nothofagus antartica (NA) is a native tree of the Patagonian Andes forests. Although its leaves have been ancestrally used in infusions for medical and food purposes, there are no deep insights on its toxicity. The aim of the paper was to assess the safety and antioxidant activity of infusions of NA leaves. The determination of acute and subacute oral toxicity was assessed using Swiss albino mice. A single limit dose (2000 mg/kg) was administered for acute toxicity assays, and a repeated dose (800 mg/kg), for subacute tests. For the measurement of antioxidant activity, the ABTS scavenging activity was determined. The effects of NA infusion were evaluated on the animal weight and on the functionality and histology of key organs (e.g., liver, kidney, intestine). The antioxidant activity of NA infusion was determined in vitro and also in feces and serum of treated animals. After the acute and subacute toxicity tests, no death or signs of toxicity were observed. In addition, no significant differences in biochemical parameters or tissue structure were registered between control and treated mice. On the other hand, the antioxidant activity of the NA infusion was greater than that observed for the standard of ascorbic acid. Moreover, the periodic administration of NA infusion could increase the internal antioxidant capacity in mice at intestinal level. Results support the safe of consuming NA leaves infusions by humans and suggest their contribution for modulating the oxidative stress at the gut level.

Abstract The adaptation of specialized care for newborns with Down syndrome (DS) requires recognition of its characteristics at the local level. Goal: to characterize newborns diagnosed with DS and their mothers in maternal and child care units in the city of Medellín. Also, explore the factors associated with timely prenatal diagnosis. Methods: observational, cross-sectional study, based on medical records of newborns diagnosed with DS in seven institutions in Medellín, Antioquia between January 2015 and December 2019, taking into account both maternal and neonatal variables. Initially, a univariate analysis was performed using absolute and relative frequencies, then a bivariate analysis was performed taking into account neonatal outcomes according to access to prenatal care and the time at which the diagnosis was made (prenatal or neonatal), and finally a multivariate analysis for late neonatal diagnosis. Results: 50.2% of the mothers at the time of delivery were 35 years old or younger, of these 59.9% belonged to the contributory regime and 83.4% had 4 prenatal check-ups or more, despite this, it was found that only 33.7% had a prenatal diagnosis of DS. The majority (91.4%) of the neonates had a diagnosis of congenital heart disease and the prolonged hospital stay was related to related morbidity. Conclusion: DS is one of the most common chromosomal abnormalities in our environment, which can be diagnosed early. However, in our study it was possible to show that despite the high coverage of prenatal controls, the frequency of antenatal diagnosis is lower, which requires that these controls be carried out by medical personnel trained in patients with this type of pathology. This will finally be reflected in a better acceptance by the family towards the arrival of a child with this condition and, of course, better access to specialized health services.

Scientific research on developing and characterizing eco-friendly metal nanoparticles (NPs) is an active area experiencing currently a systematic and continuous growth. A variety of physical, chemical and more recently biological methods can be used for the synthesis of metal nanoparticles. Among them, reports supporting the potential use of algae in the NPs green synthesis, contribute with only a minor proportion, although seaweed was demonstrated to perform as a successful reducing and stabilizing agent. Thus, the first part of the present review depicts the up-to-date information on the use of algae extracts for the synthesis of metal nanoparticles, including a deep discussion of the certain advantages as well as some limitations of this synthesis route. In the second part, the available characterization techniques to unravel their inherent properties such as specific size, shape, composition, morphology and dispersibility are comprehensively described, to finally focus on the factors affecting their applications, bioactivity, potential toxic impact on living organisms and incorporation into food matrices or food packaging, as well as future prospects. The present article identifies the key knowledge gap in a systematic way highlighting the critical next steps in the green synthesis of metal NPs mediated by algae.

Development of multicellular organisms is a complex process involving precise coordination of growth among individual cells. Understanding organogenesis requires measurements of cellular behaviors over space and time. In plants, such a quantitative approach has been successfully used to dissect organ development in both leaves and external floral organs, such as sepals. However, the observation of floral reproductive organs is hampered as they develop inside tightly closed floral buds, and are therefore difficult to access for imaging. We developed a confocal time-lapse imaging method, applied here to Arabidopsis (Arabidopsis thaliana), which allows full quantitative characterization of the development of stamens, the male reproductive organs. Our lineage tracing reveals the early specification of the filament and the anther. Formation of the anther lobes is associated with a temporal increase of growth at the lobe surface that correlates with intensive growth of the developing locule. Filament development is very dynamic and passes through three distinct phases: (1) initial intense, anisotropic growth, and high cell proliferation; (2) restriction of growth and proliferation to the filament proximal region; and (3) resumption of intense and anisotropic growth, displaced to the distal portion of the filament, without cell proliferation. This quantitative atlas of cellular growth dynamics provides a solid framework for future studies into stamen development.

Lipopolysaccharide (LPS) induces the activation of dendritic cells (DCs) throughout the engagement of toll-like receptor 4. LPS-activated DCs show increased capacity to process and present pathogen-derived antigens to activate naïve T cells. DCs-based vaccines have been successfully used to treat some cancer types, and lately transferred to the field of infectious diseases, in particular against HIV. However, there is no vaccine or DC therapy for any parasitic disease that is currently available. The immune response against Trypanosoma cruzi substantially relies on T cells, and both CD4+ and CD8+ T lymphocytes are required to control parasite growth. Here, we develop a vaccination strategy based on DCs derived from bone marrow, activated with LPS and loaded with TsKb20, an immunodominant epitope of the trans-sialidase family of proteins. We extensively characterized the CD8+ T cell response generated after immunization and compared three different readouts: a tetramer staining, ELISpot and Activation-Induced Marker (AIM) assays. To our knowledge, this work shows for the first time a proper set of T cell markers to evaluate specific CD8+ T cell responses in mice. We also show that our immunization scheme confers protection against T. cruzi, augmenting survival and reducing parasite burden in female but not male mice. We conclude that the immunization with LPS-activated DCs has the potential to prime significant CD8+ T cell responses in C57BL/6 mice independently of the sex, but this response will only be effective in female, possibly due to mice sexual dimorphisms in the response generated against T. cruzi.

- Abstract

In the pathogen Typanosoma cruzi, the calcium ion (Ca2+) regulates key processes for parasite survival. However, the mechanisms decoding Ca2+ signals are not fully identified or understood. Here, we investigate the role of a hypothetical Ca2+-binding protein named TcCAL1 in the in vitro life cycle of T. cruzi. Results showed that the overexpression of TcCAL1 fused to a 6X histidine tag (TcCAL1-6xHis) impaired the differentiation of epimastigotes into metacyclic trypomastigotes, significantly decreasing metacyclogenesis rates. When the virulence of transgenic metacyclic trypomastigotes was explored in mammalian cell invasion assays, we found that the percentage of infection was significantly higher in Vero cells incubated with TcCAL1-6xHis-overexpressing parasites than in controls, as well as the number of intracellular amastigotes. Additionally, the percentage of Vero cells with adhered metacyclic trypomastigotes significantly increased in samples incubated with TcCAL1-6xHis-overexpressing parasites compared with controls. In contrast, the differentiation rates from metacyclic trypomastigotes to axenic amastigotes or the epimastigote proliferation in the exponential phase of growth have not been affected by TcCAL1-6xHis overexpression. Based on our findings, we speculate that TcCAL1 exerts its function by sequestering intracellular Ca2+ by its EF-hand motifs (impairing metacyclogenesis) and/or due to an unknown activity which could be amplified by the ion binding (promoting cell invasion). This work underpins the importance of studying the kinetoplastid-specific proteins with unknown functions in pathogen parasites.
