ObjectiveTo investigate how tobacco and tea spread among virgin populations and how the first addictions have subsequently influenced the behavior of present-day populations.DesignRetrospective observational study using data from frozen burials and levels of theobromine, theophylline, caffeine, nicotine, and cotinine measured in hair samples from frozen bodies of autochthonous people. Along with the collection of cultural data associated with the bodies, potential comorbidities were investigated.Main outcome measureWe combined LC-HRMS and LC-MS/MS tools for toxicological investigations in hair and we assessed the association between xenobiotic concentrations and geography using several permutation-based methods to infer the economic circuits of tobacco and tea. Comparison of the results obtained with ethno-botanical analyses allowed to identify the products from which the metabolites were derived.ResultsHair levels of theobromine, theophylline and caffeine vary with the type of beverage consumed: green, black or local herbal teas. At the beginning of our study period, a few heavy consumers of tobacco were found among light or passive consumers. Tobacco-related co-morbidities began to be recorded one century after contact with Europeans. Heavy tea users were only found from the 19th century and the heaviest users of the two substances date from this century. After the first contact, teas were widely consumed as beverages and medicines but also for shamanic reasons. Economic factors, fashion and social and family contacts seem to have played a decisive role in tobacco consumption very early on.ConclusionEpidemiological characteristics of present-day Yakutia suggest that the high prevalence of smokers and tea consumers, the prevalence of female smokers and tobacco use in the north, find their origins in the diffusion phenomena of the 18th and 19th century. Behavioral evolution governed the process of substance integration and was determinant for the continuity of use of these substances over a long period of time.

La schistosomiase demeure un réel problème de santé publique dans les pays tropicaux. La pathologie à Schistosoma mansoni est principalement causée par les Tmufs qui, embolisés dans le foie et l’intestin, sont à l’origine d’une nécrose tissulaire, de la mise en place d’une réaction inflammatoire granulomateuse pouvant évoluer vers une fibrose et d’un dysfonctionnement tissulaire. L’utilisation intensive du praziquantel, unique médicament contre toutes les formes de schistosomiase, est parfois à l’origine d’échecs thérapeutiques et pourrait conduire à une résistance du schistosome. Cette étude a été entreprise pour évaluer par analyse histologique l’effet préventif de l’extrait aqueux des parties aériennes de Sida pilosa (SpAE) sur la mise en place de la pathologie à Schistosoma mansoni chez la souris. Après infestation des souris mâles, elles ont été traitées quotidiennement à SpAE du jour 1 au jour 28 post-infection (p.i.) aux doses de 100 et 200 mg/kg. Les souris ont été sacrifiées au jour 56 p.i.. Les effets de SpAE sur l’installation des pathologies hépatique, intestinale et splénique ont alors été évalués. Pour ce faire, le foie, l’intestin grêle et la rate ont été fixés dans le formol 10 % tamponné puis des coupes de 5 µm réalisées. Le foie et l’intestin ont été colorés à l’hématoxylineéosine afin d’évaluer les infiltrations leucocytaires et la réaction granulomateuse. La coloration au Picrosirius a été entreprise pour la mise en évidence du collagène et la coloration au Periodic Acid Schiff (PAS) pour visualiser les structures glycoprotéiques du parasite. La coloration de Perls a été effectuée sur des coupes de rate pour la mise en évidence d’hémorragies tissulaires à travers la présence de sidérophages. Le traitement à SpAE a prévenu l’infiltration des cellules inflammatoires dans le foie des souris à 56 jours p.i. Le nombre et la taille des granulomes au niveau des parenchymes hépatique et intestinal étaient ainsi considérablement réduits. L’effet de cet extrait de plante a été également marqué par la diminution du dépôt de collagène au niveau des foyers granulomateux. Une présence plus importante de sidérophages a été notée sur la rate des animaux infestés et non traités comparativement à celle des animaux infestés puis traités à SpAE. Cette étude confirme l’inefficacité du praziquantel sur les stades larvaires de Schistosoma mansoni et démontre que l’extrait aqueux des parties aériennes de Sida pilosa préviendrait considérablement l’installation de la pathologie à Schistosoma mansoni., Schistosomiasis remains a real public health problem in tropical countries. Schistosoma mansoni-induced pathology is mainly caused by eggs which, embolized in the liver and intestine, lead to tissue necrosis, establishment of granulomatous inflammatory reactions which could progress to fibrosis and tissue dysfunction. There is no available vaccine against schistosomiasis and the intensive use of praziquantel, the only drug against all Schistosoma species, sometimes resulted to therapeutic failures and could lead to schistosome resistance. This study was undertaken to evaluate by histological analysis, the preventive effect of the aqueous extract of Sida pilosa aerial parts (SpAE) on the establishment of Schistosoma mansoni pathology in mice. After infection, male mice were treated daily with 100 or 200 mg/kg of SpAE from day 1 to day 28 post-infection (p.i) and sacrificed at day 56 p.i.. The effects of this treatment on hepatic, intestinal and spleen pathologies were evaluated. Liver, small intestine and spleen were removed and fixed in 10% buffered formalin and 5 µm sections realized. The liver and intestine were stained with hematoxylin-eosin to evaluate leukocyte infiltration and granulomatous reaction. Picrosirius staining was used for the detection of collagen, and Periodic Acid Schiff staining (PAS) to visualize the glycoprotein structures of the parasite. Perls staining was performed on the spleen section to highlight tissue bleeding through the presence of siderophages. SpAE treatment prevented infiltration of inflammatory cells into the liver of mice at 56th day p.i.. The number and size of hepatic and intestinal granulomas were considerably reduced. The antifibrotic effect of this extract was also marked by the decrease in collagen deposition in granulomatous foci. Siderophages were more present on the spleen of infected-untreated mice than the ones of infected and SpAE-treated mice. This study confirmed the inefficacy of praziquantel on Schistosoma mansoni larval stages and revealed that the aqueous extract of Sida pilosa aerial parts could considerably prevent the installation of Schistosoma mansoni pathology.

Laportea. aestuans (Urticacea) est une plante utilisée dans la région du Centre Cameroun pour le traitement des fractures. La présente étude a pour but d’évaluer par analyse histologique les effets ostéo-inducteurs de l’extrait aqueux des feuilles de L. aestuans chez la rate (Wistar). Une cavité a été créée dans la portion antérieure de la diaphyse du fémur. L’extrait aqueux de L. aestuans a été administré quotidienne par voie orale (100-400 mg/kg) pendant deux semaines. Au terme de la période expérimentale, les animaux ont été sacrifiés et les fémurs fixés dans du formol 10% tamponné puis transférés dans de l’éthanol à 70%. Ils ont ensuite été déshydratés et inclus en paraffine. Des coupes de 5µm ont été réalisées et les colorations suivantes faites : - H&E - Picrosirius et observation en lumière polarisée. Les animaux fracturés ne recevant aucun traitement ont présenté une structure 150 Revue Française d’Histotechnologie 2018 - Vol. 30 - n°1 osseuse désorganisée avec la présence d’ostéoblastes et d’ostéocytes dans leurs lacunes. En lumière polarisée ces animaux ont présenté un aspect d’«os tissé» se traduisant par une trame collagénique peu organisée et une faible minéralisation. Les animaux traités à l’extrait de plante ont présenté une structure du cal osseux de plus en plus dense avec plus ostéoblastes et de plus en plus d’ostéocytes dans les lacunes à toutes les doses. En lumière polarisée et à la dose de 400 mg/kg d’extrait de plante, les animaux ont montré un début d’organisation lamellaire parallèle au sein de l’os tissé. L’extrait induirait une activation ostéoblastique, conduisant à une réparation osseuse plus rapide, ce qui justifierait son utilisation empirique dans le traitement des fractures., Laportea aestuans (Urticaceae) is a plant used in the Cameroon centre region for fracture healing. The purpose of this study is to evaluate by histological analysis, the osteoinductive effect of the aqueous leaves plant extract of L. aestuans in female Wistar rats. A drill hole injury was created in the diaphysis of the femur. The aqueous extract of L. aestuans was administered orally (100-400 mg/kg) for two weeks. At the end of the experimental period, the animals were sacrificed and the femurs fixed in 10% buffered formalin and then transferred into 70% ethanol. They were then dehydrated and embedded in paraffin. Sections of 5µm were made and the following colorations realized : H & E, Picrosirius staining and observation in polarized light. Fractured animals receiving no treatment exhibited a disorganized bone structure with the presence of osteoblasts and osteocytes in their lacuna. In polarized light, these animals presented an appearance of «woven bone» resulting in a poorly organized collagen structure and low mineralization. Animals treated with plant extract exhibited an increasingly dense callus structure with more osteoblasts and more and more osteocytes in the lacuna at all doses. In polarized light and at the dose of 400 mg/kg, the animals showed a parallel lamellar organization within the woven bone. Thus, the extract would induce osteoblastic activation, leading to faster bone repair, justifying its empirical use in the treatment of bone fractures.

Dans l’infection à Schistosoma mansoni, la majorité des Tmufs pondus par les vers femelles sont embolisés dans l’intestin puis le foie où ils déterminent la formation de granulomes évoluant vers le stade fibrotique. L’utilisation intensive du praziquantel, médicament de choix contre la schistosomiase pourrait conduire à une résistance des souches de schistosomes, d’où l’importance de développer une chimiothérapie alternative ou complémentaire. Cette étude histologique a été conduite pour vérifier le potentiel antifibrotique de la fraction à l’acétate d’éthyle de l’extrait méthanolique des racines de Ozoroa pulcherrima (EAOp) chez des souris infestées par S. mansoni. Un traitement quotidien à EAOp (100, 200 ou 400 mg/kg) a été réalisé pendant 28 jours à partir du 36ème jour post-infestation. Les souris ont été sacrifiées au 65ème jour post-infection, puis des paramètres parasitologiques et histologiques évalués dans le foie. Des coupes de foie de 5 µm ont été colorées à l’hématoxyline-éosine afin d’évaluer les infiltrations leucocytaires et la réaction granulomateuse. La coloration au picrosirius a été également entreprise pour la mise en évidence du collagène, ainsi que la coloration au Giemsa pour visualiser les Tmufs du parasite et la larve qu’ils renferment. The intensive use of praziquantel for the treatment of schistosomiasis has raised concerns about the possible emergence of drug-resistant schistosomes. The search for alternative drugs has become a priority. The aim of this study was to histologically assess the antifibrotic potential of the ethyl acetate fraction from Ozoroa pulcherrima (EAOp) roots methanolic extract on S. mansoni-infected mice. The fraction at 100, 200 and 400 mg/kg was administered orally and daily to infected mice for 28 days, starting from 36th day post-infection. All mice were sacrificed at 65th day post-infection. Parasitological burden and histology of the liver were conducted. Hematoxylin and eosin (H&E) staining was performed to evaluate the inflammatory infiltration and the granulomatous reaction. Liver sections were also stained by picrosirius method for collagen evaluation. Giemsa staining method was also performed to visualize parasite eggs and the larvae inside it. L’altération d’un os en période post-mortem, est à l’origine d’une détérioration identifiable en histologie. La dégradation de l’os se caractérise notamment par une altération collagénique observable précocement. Dans le but de pouvoir caractériser cette altération, deux côtes ont été prélevées sur 6 corps humains et étudiées prospectivement pendant les 2 ans suivant le décès. Chaque os a fait l’objet d’une coupe histologique étudiée après coloration au rouge Sirius afin de mettre en évidence la dégradation du collagène en fonction du temps. Une altération osseuse temporelle a pu être mise en évidence par une dégradation architecturale de l’os lamellaire, en l’absence de toute attaque microbienne sur cette période post-mortem. La coloration pratiquée à l’aide du rouge Sirius a permis d’attribuer cette altération à une dégradation collagénique de type hydrolyse chimique en raison de l’affinité de ce colorant pour les acides aminés Lysine, Hydroxylysine et Arginine. Notre travail a permis d’affirmer que des échantillons osseux humains étudiés en environnement contrôlé, et analysés pendant 24 mois, subissent une trajectoire diagénétique dont l’élément principal est l’hydrolyse collagénique., The diagenesis of a bone in the post-mortem period causes an identifiable deterioration in histology. More particularly, the degradation of the bone is characterized by a collagenous alteration which is observable very early. In order to be able to characterize this alteration, two ribs collected from 6 human bodies were studied prospectively during 2 years. Each bone was studied after staining with red Sirius in order to demonstrate the degradation of collagen as a function of time. A temporal bone alteration could be demonstrated by an architectural degradation of the lamellar bone in the absence of any microbial attack on this post-mortem period. The coloring carried out using red Sirius correlated this alteration to a collagenic degradation by chemical hydrolysis because the affinity of this colorant to the amino acids Lysine, Hydroxylysine and Arginine. Our work asserts that human bone samples studied in controlled environment, and analyzed for 24 months, undergo a diagenetic trajectory whose main element is collagen hydrolysis.
