Long-term kleptoplasty, the capability to retain functional stolen chloroplasts (kleptoplasts) for several weeks to months, has been shown in a handful of Sacoglossa sea slugs. One of these sea slugs is Elysia timida, endemic to the Mediterranean, which retains functional chloroplasts of the macroalga Acetabularia acetabulum. To understand how light modulates the lipidome of E. timida, sea slug specimens were subjected to two different 4-week light treatments: regular light and quasi-dark conditions. Lipidomic analyses were performed by HILIC-HR-ESI-MS and MS/MS. Quasi-dark conditions caused a reduction in the amount of essential lipids for photosynthetic membranes, such as glycolipids, indicating high level of kleptoplast degradation under sub-optimal light conditions. However, maximum photosynthetic capacities (Fv/Fm) were identical in both light treatments (≈0.75), showing similar kleptoplast functionality and suggesting that older kleptoplasts were targeted for degradation. Although more stable, the phospholipidome showed differences between light treatments: the amount of certain lipid species of phosphatidylethanolamine (PE), phosphatidylinositol (PI), and phosphatidylglycerol (PG) decreased under quasi-dark conditions, while other lipid species of phosphatidylcholine (PC), PE and lyso-PE (LPE) increased. Quasi-dark conditions promoted a decrease in the relative abundance of polyunsaturated fatty acids. These results suggest a light-driven remodelling of the lipidome according to the functions of the different lipids and highlight the plasticity of polar lipids in the photosynthetic sea slug E. timida. Nudibranchs are soft bodied, shell-less marine gastropod molluscs. They are widespread over a wide variety of environments. Some of these species are known due to their stenophagous dietary habits and their ability to retain photosynthetic endosymbionts from their preys. One of such nudibranchs is the aelolid Berghia stephanieae, a stenophagous nudibranch that retains both the photosynthetic endosymbionts and the nematocysts of its prey, the glass anemone Exaiptasia diaphana. Berghia stephanieae is known to be one of the best models for ecological studies addressing the mollusc-photosynthetic dinoflagellate endosymbiosis. However, these studies are often made with the use of invasive techniques, that commonly harm the study subject, and may even lead to their sacrifice. The glass anemone, E. diaphana, is seen as a model species to the study of cnidarian-microbiome symbiosis. As this species establishes a symbiosis with photosynthetic dinoflagellates, it has been used to study bleaching, the phenomenon that results from the loss of photosynthetic endosymbionts or pigments from the animal host. The present work addresses the maintenance of the symbiosis between B. stephanieae and its photosynthetic dinoflagellate endosymbionts acquired through the predation of E. diaphana using a non-invasive methodology, the measurement of pulse amplitude modulated chlorophyll fluorescence. While this methodology is commonly employed on plants and algae, it has already been successfully used to study kleptoplasty in sacoglossan sea slugs, as well as the maintenance of photosynthetic efficiency of photosynthetic dinoflagellate endosymbionts in some nudibranchs. Our experimental trials made possible to detect a positive correlation between the abundance of photosynthetic dinoflagellate endosymbionts within B. stephanieae and the minimum fluorescence of dark-adapted animals (F₀). The abundance of photosynthetic dinoflagellate endosymbionts within B. stephanieae and their photosynthetic efficiency were studied in sea slugs reared under different light intensities (high light, 80 µmol m- ² s - ¹; low light, 10 µmol m- ² s - ¹) and access to food. Exposure to different light intensities had no significant impact in the maintenance of B. stephanieae’s photosynthetic dinoflagellate endosymbionts, nor on their photosynthetic efficiency. However, starvation was shown to have a significant impact by decreasing both the abundance of photosynthetic dinoflagellate endosymbionts within B. stephanieae and their photosynthetic efficiency. The effect of different trophic regimes in the abundance of photosynthetic dinoflagellate endosymbionts within B. stephanieae and their photosynthetic efficiency was also tested. It was possible to show that sea slugs fed with bleached anemones (anemones lacking photosynthetic dinoflagellate endosymbionts) were impacted in a similar way to starved sea slugs, as they both exhibited a decrease in the abundance of photosynthetic dinoflagellate endosymbionts and a lower photosynthetic efficiency. This finding shows that the photosynthetic dinoflagellate endosymbionts present within B. stephanieae need to be replenished regularly by these nudibranchs by preying on symbiotic glass anemones (anemones that have photosynthetic dinoflagellate endosymbionts). Overall, the present study is the starting point to future works on which chlorophyll fluorescence parameter F₀ may be used to infer the abundance, maintenance and photosynthetic efficiency of endosymbionts in nudibranchs. This methodology may be a powerful tool to study the consequences of bleaching of marine organisms on their stenophagous predators, as these are widely unknown and need to be further investigated. Os nudibrânquios são moluscos gastrópodes marinhos de corpo mole, sem concha, que ocorrem numa grande variedade de ambientes. Algumas destas espécies são conhecidas pelos seus hábitos alimentares estenófagos e por reterem endossimbiontes fotossintéticos presentes nas presas das quais se alimentam. Uma dessas espécies é o aeolídeo Berghia stephanieae, um nudibrânquio estenófago que retém os dinoflagelados endossimbiontes fotossintéticos e os nematocistos da sua presa, a anémona de vidro Exaiptasia diaphana. Berghia stephanieae é considerada uma espécie modelo para estudos ecológicos referentes à endossimbiose entre moluscos e dinoflagelados fotossintéticos. No entanto, estes estudos são frequentemente realizados com recurso a metodologias invasivas, que invariavelmente causam danos físicos aos organismos em estudo, ou podem mesmo levar ao seu sacrifício. A anémona de vidro, E. diaphana, é considerada uma espécie modelo para o estudo da simbiose entre os cnidários e os seus microbiomas. Devido à sua simbiose com dinoflagelados fotossintéticos, esta espécie tem sido utilizada para estudar o fenómeno de branqueamento, fenómeno resultante da perda de simbiontes ou pigmentos fotossintéticos presentes num animal hospedeiro. O presente trabalho estuda a prevalência da associação entre B. stephanieae e os dinoflagelados endossimbiontes fotossintéticos obtidos troficamente pela ingestão de E. diaphana através da utilização de uma metodologia não invasiva, a medição da fluorescência de pulso modelado da clorofila. Esta metodologia é utilizada de forma rotineira em plantas e algas, tendo, no entanto, já sido utilizada com sucesso para o estudo da cleptoplastia em lesmas marinhas da superordem Sacoglossa. Esta metodologia foi também já utilizada para estudar a manutenção da eficiência fotossintética de dinoflagelados endossimbiontes fotossintéticos presentes nalgumas espécies de nudibrânquios. Através da sua utilização foi possível identificar uma correlação positiva entre a abundância de dinoflagelados endossimbiontes fotossintéticos presentes em B. stephanieae e a fluorescência mínima de animais adaptados ao escuro (F₀). A abundância de dinoflagelados endossimbiontes fotossintéticos presentes em B. stephanieae e a sua eficiência fotossintética foram estudadas em lesmas marinhas com e sem acesso a fonte de alimento e criadas a diferentes intensidades de luz (luz alta, 80 µmol m- ² s - ¹; luz baixa, 10 µmol m- ² s - ¹). A exposição a diferentes intensidades de luz não teve efeitos na manutenção da eficiência fotossintética ou na abundância dos endossimbiontes fotossintéticos presentes em B. stephanieae. No entanto, a exposição à inanição teve um impacto significativo, fazendo decrescer tanto a abundância dos dinoflagelados endossimbiontes fotossintéticos presentes em B. stephanieae, tal como a sua eficiência fotossintética. Foi também testado o efeito de diferentes regimes tróficos na abundância de dinoflagelados endossimbiontes fotossintéticos presentes em B. stephanieae e a sua eficiência fotossintética. Este estudo permitiu evidenciar que indivíduos alimentados com anémonas branqueadas (anémonas sem dinoflagelados endossimbiontes fotossintéticos) apresentavam um impacto semelhante ao verificado em conspecíficos sujeitos a inanição, ou seja, um decréscimo na abundância de endossimbiontes fotossintéticos, assim como da sua eficiência fotossintética. Estes resultados mostram que os dinoflagelados endossimbiontes fotossintéticos presentes em B. stephanieae necessitam de ser repostos regularmente por estes nudibrânquios através da predação de anémonas vidro que apresentem estes endossimbiontes fotossintéticos. Este estudo serve de ponto de partida para trabalhos futuros nos quais o parâmetro de fluorescência da clorofila F₀ poderá ser usado para inferir a abundância, manutenção e eficiência fotossintética de endossimbiontes fotossintéticos em nudibrânquios, assim como a manutenção da sua simbiose. Esta metodologia pode ser uma ferramenta valiosa para o estudo das consequências do branqueamento de organismos marinhos nos seus predadores estenófagos, consequências essas que são pouco conhecidas e, como tal, carecem de investigação. Portugal has approximately 950 km of a coastline with a high diversity of macroalgal species, most of them edible and commercially valuable. Macroalgae are becoming reliable sources of nutritional supplements and bioactive compounds. Thus, an increase in demand for these organisms requires sustainable seaweed aquaculture alternatives to produce algal biomass rather than exploiting natural resources. The company ALGAplus was pioneer in taming Codium tomentosum. Currently, this macroalgae is produced in integrated multitrophic aquaculture earth pond systems designed by ALGAplus, and mainly commercialized for food and cosmetics. This macroalgae is a source of valuable bioactive compounds used for health care (antioxidant, antitumor, hypoglycemic) and has a high nutritional value (rich in polyunsaturated fatty acids and vitamin A) causing an increasing consumer demand, and therefore a necessity for the optimization of C. tomentosum production. In this project, C. tomentosum produced by ALGAplus, was studied to determine the best conditions for this algae cultivation, considering effects on growth and photobiology at a nursery scale production. In the first phase of this work, different temperatures (13 ºC and 17 ºC) and different photoperiods (long-day and short-day) were evaluated. The second part aimed at assessing the variations in growth and productivity, in photosynthetic activity and photosynthetic pigments concentrations, of exposing C. tomentosum cultures to different light qualities with selected wavelengths (White, Blue and Red). The literature for this species, regarding growth conditions and photobiology is scarce. Therefore, this work aims to fill the gaps of knowledge regarding C. tomentosum photophysiology optimizing culturing protocols. The macroalga C. tomentosum showed higher growth rates when exposed to the long photoperiod, and no significant differences between temperatures. However, the photosynthetic activity evaluated by steady-state light curves, estimated from variable chlorophyll a measurements, was higher at the lower temperature tested. The pigment profile did not show significant changes between treatments, however there was an increase of pigment concentrations in the long photoperiod treatment. Photoperiod had also a ratio change on the carotenes identified, the presence of ε-carotene under short photoperiod and β-carotene under long photoperiod. The macroalga C. tomentosum showed lower productivity under blue light, concomitant with a considerable growth of green microalgae. At the end of the experimental period, the photosynthetic performance was similar between the different spectra. The photosynthetic pigment profiles (chlorophyll a and b, siphonoxanthyn e siphonein) showed increased concentrations under blue light, possibly as a result of competition for the available light with microalgae. The remaining spectral conditions (Red and White light) presented similar pigment concentrations. In conclusion, C. tomentosum achieved higher growth rates at long-day photoperiod. The use of red or white light is advised to control the growth of microalgal contaminations and to optimize growth. Portugal tem aproximadamente 950 Km de costas com elevada diversidade de macroalgas, algumas delas comestíveis e com valor comercial. As macroalgas têm se tornado fontes confiáveis de suplementos alimentares e compostos bioativos. Por isso a crescente procura por estes organismos leva à necessidade da sua produção em sistemas sustentáveis de produção de macroalgas, em vez da colheita in situ dos recursos naturais. A empresa ALGAplus foi pioneira na domesticação da macroalga Codium tomentosum. Esta é atualmente cultivada num sistema de aquacultura multitrófica integrada em terra concebido pela ALGAplus, sendo comercializada principalmente para alimentação humana e cosmética. Esta macroalga é fonte de importantes compostos bioativos utilizados em cuidados de saúde (antioxidantes, antitumorais, hipoglicémicos) e tem um grande valor nutricional (ácidos gordos polinsaturados, vitamina A), aumentando a procura e, com isto, a necessidade de otimizar a sua produção. Neste trabalho, a macroalga C. tomentosum produzida pela ALGAplus, foi objecto de estudo para determinar as melhores condições de cultivo, tendo em conta o seu crescimento e fotobiologia numa escala de produção de maternidade. Numa primeira fase, foram avaliadas diferentes temperaturas (13 ºC e 17 ºC) e diferentes fotoperíodos (dia longo e dia curto). Numa segunda parte, foram avaliadas variações no crescimento e produtividade, na atividade fotossintética e nas concentrações de pigmentos fotossintéticos, em resposta à exposição a diferentes qualidades de luz de crescimento com comprimentos selecionados (branco, vermelho e azul), em condições de fotoperíodo longo. A bibliografia para esta espécie no que diz respeito às condições de crescimento e fotobiologia é ainda escassa. Assim, este trabalho pretende preencher falhas no conhecimento da fotofisiologia de C. tomentosum, de forma a optimizar protocolos de cultivo. A macroalga C. tomentosum apresentou maior crescimento sob fotoperíodo longo, não se tendo registado efeitos significativos relativamente à temperatura. No entanto, a atividade fotossintética, avaliada através de curvas de luz derivadas da variação de fluorescência da clorofila a, foi superior na temperatura mais baixa. O perfil de pigmentos não expressou diferenças muito acentuadas entre tratamentos. Verificou-se um aumento da concentração de pigmentos sob fotoperíodo longo. O fotoperíodo alterou os ratios de carotenos presentes, tendo o ε-caroteno sido identificado em fotoperíodo curto e o β-caroteno em fotoperíodo longo. A macroalga C. tomentosum apresentou menor produtividade sob luz azul, concomitante com um elevado crescimento de microalgas verdes. No final do período da experiência o desempenho fotossintético foi semelhante entre as condições de espectro de luz. Os perfis de pigmentos fotorreceptores (clorofila a e b, sifonaxantina e sifoneína) apresentaram aumentos de concentração nas culturas do espectro azul, talvez desencadeados por competição pela luz disponível com as microalgas. Nos restantes espectros de luz (vermelho e branco), as culturas apresentaram concentrações de pigmentos semelhantes. Concluímos que a macroalga C. tomentosum alcança melhores taxas de crescimento em fotoperíodo longo. É aconselhada a utilização de luz de crescimento branca ou vermelha por forma a controlar a contaminação por microalgas verdes e otimizar o crescimento. Mestrado em Biologia Marinha This work aims to investigate the role of food and light availability in the photosynthesis and maternal investment of the sea slug Elysia viridis. Two experiments were conducted to test the importance of food – Codium tomentosum – and to test the importance of the autotrophic component with three different food and light regimes, respectively. The results suggest that maternal investment is greater when food exists in abundance and less when animals are starving. A positive relationship was also observed between the abundance of food and the length of the animal, the egg mass area and egg volume. The study investigating the maternal investment in three different light regimes had some experimental limitations; however, it was observed that sea slugs exposed to an average light intensity had a greater investment in reproduction. It can be concluded that both heterotrophic and autotrophic components influence the investment of these sea slugs in their reproduction. Este trabalho pretende investigar o papel das componentes heterotrófica e autotrófica no investimento maternal da lesma do mar Elysia viridis. Foram efetuadas duas experiências para testar a importância da disponibilidade de alimento – Codium tomentosum – e para testar a importância da componente autotrófica, com três regimes diferentes de comida e de luz. Os resultados sugerem que o investimento maternal é maior quando a comida existe em abundância e menor quando os animais não têm acesso ao alimento; foi notado também uma relação positiva entre a abundância de alimento e o comprimento do animal, o tamanho das suas posturas e dos seus ovos. O estudo que investigou o investimento maternal em três diferentes regimes de luz teve algumas limitações experimentais, no entanto, pode-se observar que as lesmas do mar expostas a uma intensidade média de luz têm um maior investimento na reprodução. Logo concluímos que ambas as componentes heterotrófica e autotrófica influenciam o investimento destes animais na sua reprodução.
