Abstract

Fungi that damage documents in archives may harm workers' health, depending on which mold species are inhaled, the concentrations of fungal species inhaled, and individual factors. Our aim was to identify and quantify fungi in archives and to investigate possible links with the symptoms experienced by workers. Ten French archives were sampled using an air impactor and electrostatic dust collectors. Allergies and general symptoms felt by 144 workers were reported using a self-report questionnaire. Utilizing culture-based analysis methods along with qPCR, Penicillium chrysogenum, Cladosporium sphaerospermum, and Aspergillus versicolor were the three main fungi in air and dust in terms of quantity and frequency. Median fungal concentrations in storage areas, ranged from 30 to 465 CFU/m(3). People working in the most contaminated archives did not report more symptoms of allergy than others. However, workers in contact with moldy documents reported more headaches (odds ratio, 2.4; 95% confidence interval, 1.1-5.3), fatigue (OR, 2.9; 95% CI, 1.2-6.7), eye irritation (OR, 5.4; 95% CI, 1.9-14.9), throat irritation (OR, 2.4; 95% CI, 1.0-5.7), coughing (OR, 3.2; 95% CI, 1.2-8.4), and rhinorrhea (OR, 2.6; 95% CI, 1.0-6.4) than others. Other parameters such as dust levels and concentrations of metabolites and chemical substances should be considered as confounding factors in further investigations to isolate the role of molds.Most studies about fungi and archives deal with the conservation of manuscripts and documents, and few discuss workers' health problems. Our study shows that archives do not represent a highly contaminated environment. Symptoms felt by workers were more often linked to direct contact with moldy documents than to high concentrations of mold in the air of archive storage areas. This study provides data on concentration levels in archives that could be used to interpret microbiological investigations in this type of environment in the future.

Abstract

International audience; Mycotoxins are secondary metabolites of filamentous fungi which can cause a wide range of systemic effects. Humanhealth effects of inhaled mycotoxins remain poorly documented, despite the large amounts present, associated with airborneparticles.Among thesemycotoxins, sterigmatocystin is one of themost prevalent. Because its chemical structure isclose to that of the aflatoxins, we studied its metabolism and its cellular consequences when in contact with the airwayepithelium, using the mass spectral signature from the 10% 13C uniformly enriched sterigmatocystin. The metabolismwas studied in vitro, using recombinant cytochrome P450s enzymes, and in porcine tracheal epithelial cell (PTEC)primary cultures at an air‐liquid interface. Themetabolites were analyzed by high‐performance liquid chromatographycoupled with tandem mass spectrometry detection. Expressed enzymes and PTECs were exposed to uniformly 13Cenrichedsterigmatocystin to confirm the relationship between sterigmatocystin and its metabolites because this isotopiccluster shape is conserved for all metabolites and their product ions. Incubation of sterigmatocystin with recombinantcytochrome P450 1A1 led to the formation of three metabolites identified as monohydroxysterigmatocystin,dihydroxysterigmatocystin and one glutathione adduct, the latter after the formation of a transient intermediate. Inthe PTEC cultures, sterigmatocystin metabolism resulted in a glucuro‐conjugate. Two other products were detected, asulfo‐conjugate and a glucuro‐conjugate of hydroxysterigmatocystin upon cytochrome P450 1A1 induction. This is thefirst study to report sterigmatocystin metabolism in airway epithelium, and it suggests that, contrary to the aflatoxins,sterigmatocystin is mainly detoxified into its conjugates and is unable to produce significant amounts of reactivemetabolites in respiratory cells, at least in pigs.

Abstract

The precise location of a substrate in cytochrome P450 (CYP) governs the orientation of the oxidation position. Such information is generally obtained from biochemical data, but modeling approaches have also been used to explain these locations. We used X-ray data and modeling techniques to distinguish between the series of putative linear or curved channels which lead the substrate from the outer side of the protein to the inner, and then into the heme pocket; these techniques were also used to identify the largest such channels. Two new methods for precisely determining the 3-D structure of proteins using X-ray crystallography were proposed in order to identify these channels: first, the use of both straight and curved channels, and second, the sphere method. These data are compared with Poulos channels, and with Caver (or Mol on line) modeling methodologies. Our methods were developed from studies of the interaction between cytochrome P450CAM(CYP101) fromPseudomonas putida(as expressed inEscherichia coli) and the indolic base β-carboline. Apart from the identification of potential access channels leading to the heme-containing active site, a new explanation was advanced for the substrate's hydroxylation position. The sphere method seems to have potential to become a general and direct method for prediction of substrate access channels from reduced- or low-resolution crystallographic data.

Abstract

INTRODUCTION: Opportunistic dimorphic yeast C. albicans (CA) is part of normal harmful commensal human flora but small changes in local or systemic immune system, or other injuries could promote invasiveness of CA. CA may cause a broad spectrum of diseases ranging from mucocutaneous to systemic infection. Except host factors (immune status, protective barriers), CA secreted hydrolytic enzymes: proteases, phospholipases, lipases and hemolysins, and these enzymes contribute pathogenicity. The aim of this research was to isolate and detect lipophilic low-molecular-weight (LMW) metabolites of CA with possible toxicity. MATERIAL AND METHODS: CA (N=60) were collected from patient with vaginitis, urinary-tract infection, and from blood (fungaemia). Non-pathogenic CA (N=30) was isolated from mouth and stool specimens. Production of LMW metabolites were performed using method described by Shah and Larsen (1) in Minimal essential medium (MEM), and influence of dimorphism (monocell yeasts or (pseudo)hyphae formation under influence of CO2 and foetal calf serum) were also investigate. Using HPLC/DAD system (2, 3), LMW metabolites were isolated and separated. Chemical structure of isolated compound(s) was determinate using spectral data, retention time, retention index and mass-spectrometry. Concentration was calculated with HPLC using calibration curve with known concentration of standard vs. peak area. RESULTS: All investigated CA strains produce metabolite which UV spectra have maximum at 224 and 279 nm and minimum at 243 nm. Using library of mycotoxins, this metabolite has similar UV spectra and retention time as indol-compounds. This metabolite was identified as 3-indoleethanol (tryptophol) after comparison of negative mode electron spray (EI) MS spectral data of a reference compound. This metabolite displays a base peak at m/z 160 and MS2 were observed at m/z 130 (100%), 116 (33%), and 142. Concentration of 3-indoleethanol ranged from 8.3 μ g/mL to 124.6 μ g/mL, and its production is similar when CA is in yeast (monocell) shape or (pseudo)hyphae. There were no differences between clinical and commensal CA isolates. Gliotoxin was not detected. CONCLUSION: 3-indoleethanol was confirmed as major LMW metabolite of CA. Abstract

Some ATP-binding-cassette (ABC) transporters confer multidrug resistance (MDR) to pathogenic microorganisms and human tumor cells by mediating the extrusion of a variety of chemotherapeutic drugs out of the cell. One major resistance to chemotherapy has been correlated with the presence of these ABC transporters, acting as molecular " pumps " that actively transport drugs out of the cell, the most prevalent of these being P-glycoprotein (P-gp). Although the X-ray structures of two bacterial ABC proteins have been determined, and the first structures of mouse Pgp released recently in the Protein Data Bank (PDB), establishing molecular structures of human P-gp is still of particular interest because of its clinical relevance. Using homology modeling tools, we rebuilt three structures of human P-gp in different conformations: one nucleotide-bound and two in the absence of nucleotide. Nucleotide binding swaps the accessibility of the transporter from cytoplasmic (inward) facing to extracellular (outward) facing. We placed the three modeled conformational states in a native-like membrane environment, in order to locate several known mutations that alter specificity of drug binding, in particular relatively to the membrane surroundings. We then rebuilt two modeled structures of hamster P-gp in inward and outward conformations, and conducted docking studies of three molecules without any structural similarity (verapamil, vinblastine and tentoxin), but having competitive, non competitive and partial overlapping binding modes. The best poses obtained are consistent with the existence of two different pharmacophores for the recognition of the drug transported by P-glycoprotein., La P-glycoprotéine (P-gp) appartient à la famille des transporteurs ABC qui confèrent, aux microorganismes pathogènes et cellules tumorales humaines, par transport actif à travers la membrane plasmique, une résistance à de multiples molécules antibiotiques et anticancéreuses sans parenté structurale. Malgré les quelques structures de transporteurs ABC bactériens, la caractérisation par microscopie électronique de la P-gp, et la récente structure de P-gp de souris déposée dans la Protein Data Bank (PDB), obtenir une structure pour la P-gp humaine est d'un intérêt particulier à cause de son importance clinique. Actuellement, il n'existe pas de modèle structural d'interaction P-gp/substrat permettant d'expliquer sa multispécificité. Par modélisation par homologie, nous avons reconstruit trois structures de la Pgp humaine: une en présence et deux autres en absence de nucléotide. La liaison du nucléotide change l'accessibilité du transporteur de la face cytoplasmique vers la face extracellulaire. Ces trois états conformationnels ont été placés dans un environnement membranaire afin de révéler la localisation des mutations spécifiques altérant la liaison de la P-gp à certains médicaments. Enfin, nous avons réalisé une étude de docking sur deux structures modélisées de P-gp de Hamster dans les deux orientations. Ce docking a concerné trois molécules sans aucune ressemblance structurale (vérapamil, vinblastine et tentoxine) mais dont les fixations sont mutuellement soit compétitives, soit non-compétitives. Les meilleures poses obtenues sont compatibles avec l'existence de deux pharmacophores distincts pour la reconnaissance des drogues transportées par la P-gp.

Abstract

We report unambiguous proof of the stability of a carbinol intermediate in the case of P450 metabolism of an N-methylated natural cyclo-peptide, namely tentoxin. Under mild acidic or neutral conditions, the lifetime of carbinol-amide is long enough to be fully characterized. This metabolite has been characterized using specifically labeled (14) C-methyl tentoxin isotopomers, HPLC, HPLC-MS, MS-MS and NMR. Under stronger acidic conditions, the stability of this metabolite vanishes through deformylation. A theoretical mechanistic investigation reveals that the stability is governed by the accessibility of the nitrogen lone pair and its protonation state. For carbinol-amines, even in neutral conditions, the energy barrier for deformylation is low enough to allow rapid deformylation. Carbinol-amide behaves differently. Under neutral conditions, delocalization of the nitrogen lone pair increases the energy barrier of deformylation that is a slow process under such conditions. After protonation, we were able to optimize a deformylation transition that is lower in energy and thus accounts for the lower stability of carbinol-amides observed experimentally in acidic conditions. Finally, by considering the protocol usually used for extraction and analysis of this type of metabolite, carbinol-amide may thus be frequently ignored in drug metabolism pathways.

Abstract

Mycotoxins are secondary metabolites of filamentous fungi which can cause a wide range of systemic effects. Humanhealth effects of inhaled mycotoxins remain poorly documented, despite the large amounts present, associated with airborneparticles.Among thesemycotoxins, sterigmatocystin is one of themost prevalent. Because its chemical structure isclose to that of the aflatoxins, we studied its metabolism and its cellular consequences when in contact with the airwayepithelium, using the mass spectral signature from the 10% 13C uniformly enriched sterigmatocystin. The metabolismwas studied in vitro, using recombinant cytochrome P450s enzymes, and in porcine tracheal epithelial cell (PTEC)primary cultures at an air‐liquid interface. Themetabolites were analyzed by high‐performance liquid chromatographycoupled with tandem mass spectrometry detection. Expressed enzymes and PTECs were exposed to uniformly 13Cenrichedsterigmatocystin to confirm the relationship between sterigmatocystin and its metabolites because this isotopiccluster shape is conserved for all metabolites and their product ions. Incubation of sterigmatocystin with recombinantcytochrome P450 1A1 led to the formation of three metabolites identified as monohydroxysterigmatocystin,dihydroxysterigmatocystin and one glutathione adduct, the latter after the formation of a transient intermediate. Inthe PTEC cultures, sterigmatocystin metabolism resulted in a glucuro‐conjugate. Two other products were detected, asulfo‐conjugate and a glucuro‐conjugate of hydroxysterigmatocystin upon cytochrome P450 1A1 induction. This is thefirst study to report sterigmatocystin metabolism in airway epithelium, and it suggests that, contrary to the aflatoxins,sterigmatocystin is mainly detoxified into its conjugates and is unable to produce significant amounts of reactivemetabolites in respiratory cells, at least in pigs.
