There is a plethora of information about metastasis through the circulatory system, however not much information is available about the tumor cells dissemination through the lymphatic system or the lymphatic microenvironment that aids in this process in breast cancer metastasis.Purpose: The study designed to examine the tumor-derived secretome in lymph before reaching the draining lymph nodes.Methods: Using a microsurgical technique, we have collected the lymph in transit from the primary tumor en route to the regional lymph node in animals with metastatic and non-metastatic mammary carcinoma and healthy controls. The lymph samples were subjected to LC-MS/MS analysis, bioinformatics, and pathway analysis.Results: The metastatic tumor-draining lymph before its entry into the closest regional lymph node contain 26 proteins with > 175-folds in abundance compared to lymph from non-metastatic tumor-bearing animals. Among these proteins were biliverdin reductase B, heat shock protein, coagulation factor XIII, lymphocytes cytosol protein 1, and aldose reductase. These proteins were not identified in the lymph from healthy animals. Pathways analysis revealed that cadherin-mediated endocytosis, acute phase response, junction signaling, gap junction, VEGF singling, and PI3K/AKT singling pathways are overrepresented in the lymph from metastatic tumor-bearing compared to the lymph from non-metastatic tumor-bearing animals. Among the significantly up-regulated proteins in the lymph from metastatic tumor-bearing animals were proteins that identified in exosomes include heat shock protein, enolase 1 alpha, S100, and biliverdin reductase B. One of the proteins significantly down-regulated in lymph from animals with metastasis is Kininogen, a known metastasis inhibitor protein.Conclusion: Proteins and exosomal proteins in lymph draining a metastatic tumor are different from those in lymph draining non-metastatic tumors, and these proteins involved in pathways that regulate tumor cells migration and invasion.Keywords: lymph, lymphatic vessels, cancer, breast, lymph node, metastasis, exosome, tumor-derived factors

Abstract

In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. A key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process vs. those that measure flux through the autophagy pathway (i.e., the complete process); thus, a block in macroautophagy that results in autophagosome accumulation needs to be differentiated from stimuli that result in increased autophagic activity, defined as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (in most higher eukaryotes and some protists such as Dictyostelium) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the field understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular autophagy assays, we hope to encourage technical innovation in the field. © 2012 Landes Bioscience.

Abstract

This thesis focuses on milk proteins fractionation by dynamic filtration. A two-stage process has been proposed in order to separate milk proteins into three main fractions: casein micelles, α-Lactalbumin and β-Lactoglobulin. The first stage of the process consists in microfiltration of skim milk for separating casein micelles from whey proteins. The performances of two dynamic filtration modules: the Multi Shaft Disk (MSD) module and the rotating disk module have been compared. The results have shown that the MSD module presents high permeate flux, good whey proteins transmission and high casein micelles rejection. The second stage of the process is the separation of α-Lactalbumin from β-Lactoglobulin by ultrafiltration. The rotating disk module equipped with a disk with vanes and rotating at 2000 rpm, was used. The protein transmission and selectivity were constant during whey concentration. This is an advantage of dynamic filtration because in crossflow filtration protein transmission and selectivity decreased during whey concentration. The selectivities obtained with dynamic filtration, without changes in pH and ionic strength of the solution, were in the same range as those obtained with crossflow filtration after pH and ionic strength optimization., Ce mémoire de thèse est consacré au fractionnement des protéines du lait par filtration dynamique. Un procédé en cascade a été proposé pour séparer les protéines du lait en trois fractions principales : les caséines, l'α-Lactalbumine et la β-Lactoglobuline. La première étape du procédé consiste en une microfiltration du lait afin de séparer les caséines des protéines du lactosérum. Une comparaison entre les performances des deux modules de filtration dynamique : le module Multi Shaft Disk (MSD) et le module à disque rotatif, a été effectuée. Nous avons obtenu les meilleurs résultats avec le prototype MSD, fournissant des flux de perméat élevés, ainsi que une très bonne transmission de protéines solubles et une rétention élevée des micelles de caséines. La deuxième étape du procédé est la séparation d'α-Lactalbumine de la β-Lactoglobuline, par ultrafiltration. Le prototype à disque rotatif muni d'un disque à ailettes tournant à 2000 tr.min-1 a été utilisé. Nous avons obtenu des transmissions de protéines et des sélectivités constantes lors de la concentration du lactosérum. Ceci constitue un avantage des systèmes dynamiques, car en filtration tangentielle les transmissions de protéines et les sélectivités diminuent avec la concentration. Les sélectivités obtenues en filtration dynamique, sans modification de pH ni de la force ionique, sont de l'ordre de grandeur de celles obtenues en filtration tangentielle après optimisation du pH et de la force ionique.

Abstract

El Síndrome Premenstrual (SPM) afecta a gran cantidad de mujeres en edad reproductiva y se caracteriza por una repetición cíclica de síntomas físicos y psicológicos que, en algunos casos, pueden llegar a ser lo suficientemente severos como para interferir en los patrones de vida. El objetivo de esta revisión bibliográfica es conocer cómo afecta el SPM el rendimiento laboral de las mujeres. A pesar de las significativas repercusiones personales y económicas, existen pocas investigaciones que estudien la relación entre SPM y rendimiento laboral. Sin embargo, la literatura es concluyente en señalar que la productividad laboral es un importante dominio de la vida de la mujer adversamente afectado por el SPM, manifestado en un aumento de la tasa de ausentismo laboral y reducción de la productividad. Se concluye, según la literatura revisada, que el SPM afecta el rendimiento laboral, lo que sugiere a los profesionales de la salud tomar un rol activo en la derivación y tratamiento oportuno, para de esta forma lograr un aumento en la productividad laboral de las mujeres The premenstrual syndrome (PMS) affects a large quantity of women of reproductive age, and it characterizes because of a cyclic recurrence of physical and psychological simptoms that, in some cases, might become severe enough as to interfere with a persons life pattern. The objective of this paper is to gather a bibliographic revision that will lead us to know how PMS affects the working performance of women. Although the repercussions in this matter are very significant, there are only a few studies about the relation and interdependence between this syndrome and the efficiency at work. However, some literature establishes that the PMS largely affects the working productivity of women, which is manifested in reduction of productivity and larger rates of working absenteeism. The conclusion we support after having checked papers, reports and literature written about PMS is that this syndrome does affect the working performance of those how suffer it, by enhancing the rates of labor absenteeism and reducing productivity at work. This suggests that health professionals should take a more active role in the treatment of this disease, in order to achieve the goal of increasing the productivity at work for women

Abstract

CD49d, the α-chain of the integrin heterodimer α4β1 (CD49d/CD29) is expressed on cell surface of ~40% of chronic lymphocytic leukemia (CLL) cases. It mediates both cell–cell and cell–matrix adhesion through binding to the vascular cell adhesion molecule-1 (VCAM-1) and fibronectin (FN), respectively. These interactions regulate CLL cell recirculation from the blood stream to tissue sites, as well as the transmission to CLL cells of signals of pro-survival and resistance to drug-induced apoptosis.1 Consistent with this functional role in CLL, a recent worldwide meta-analysis carried out on a large patient cohort, definitely demonstrated the independent prognostic relevance of CD49d in this disease

Abstract

Investigation of cell signaling pathways in 16 clear cell renal cell carcinomas to identify groups based on commonly shared phosphorylation-driven signaling networks. Using laser capture microdissection and reverse-phase protein arrays, we profiled 75 key nodes spanning signaling pathways important in tumorigenesis. Analysis revealed significantly different (P < 0.05) signaling levels for 27 nodes between two groups of samples, designated A (4 samples; high EGFR, RET, and RASGFR1 levels, converging to activate AKT/mTOR) and B (12 samples; high ERK1/2 and STAT phosphorylation). Group B was further partitioned into groups C (7 samples; elevated expression of LC3B) and D (5 samples; activation of Src and STAT). Network analysis indicated that group A was characterized by signaling pathways related to cell cycle and proliferation, and group B by pathways related to cell death and survival. Homogeneous clear cell renal cell carcinomas could be stratified into at least two major functional groups.

Abstract

Thyroid carcinoma is the most common endocrine malignancy and the first cause of death among endocrine cancers. We show that the tumorigenic capacity in thyroid cancer is confined in a small subpopulation of stem-like cells with high aldehyde dehydrogenase (ALDHhigh) activity and unlimited replication potential. ALDHhigh cells can be expanded indefinitely in vitro as tumor spheres, which retain the tumorigenic potential upon delivery in immunocompromised mice. Orthotopic injection of minute numbers of thyroid cancer stem cells recapitulates the behavior of the parental tumor, including the aggressive metastatic features of undifferentiated thyroid carcinomas, which are sustained by constitutive activation of cMet and Akt in thyroid cancer stem cells. The identification of tumorigenic and metastagenic thyroid cancer cells may provide unprecedented preclinical tools for development and preclinical validation of novel targeted therapies.
