1. A pump-probe experiment in cw-mode on ionization of Rydberg atoms

Abstract: Rydberg atoms are in the focus of intense research due to the peculiar properties which make them interesting candidates for quantum optics and quantum information applications. In this work we study the ionization of Rydberg atoms due to their interaction with a trapping laser field, and a reaction microscope is used to measure photoelectron angular and energy distributions. Reaction microscopes are excellent tools when brandished against atomic photoionization processes involving pulsed lasers; the timing tied to each pulse is crucial in solving the subsequent equations of motion for the atomic fragments in the spectrometer field. However, when used in pump-probe schemes, which rely on continuous wave probe lasers, vital information linked to the time of flight is lost. This study reports on a method in which the standard ReMi technique is extended in time through coincidence measurements. This is then applied to the photoionization of $^6$Li atoms initially prepared in optically pumped $2^{2}S_{1/2}$ and $2^{2}P_{3/2}$ states. Multi-photon excitation from a tunable femtosecond laser is exploited to produce Rydberg atoms inside an infrared optical dipole trap; the structure and dynamics of the subsequent cascade back towards ground is evaluated., 11 pages, 10 figures

2. BDNF rs10501087, rs1491850 and rs11030094 polymorphisms associated with delayed progression in early-stage Parkinson's disease

Abstract: Parkinson's disease (PD) is heterogenous in its presentation, progression and response to therapies. Genetic polymorphisms may account for some of this variability. Several single nucleotide polymorphisms (SNPs) in the brain-derived neurotrophic factor gene BDNF have been associated with differing clinical outcomes from different dopaminergic replacement strategies, and one of these, the rs6265 SNP, has been associated with a milder clinical phenotype in the unmedicated, early-stage of PD. We examined if other BDNF SNPs with potential pharmacogenetic effects also are associated with different rates of disease progression. The Deprenyl And Tocopherol Antioxidative Therapy Of Parkinsonism (DATATOP) study was analyzed retrospectively. DNA samples (n = 217) were genotyped for the BDNF rs908867, rs11030094, rs10501087, rs1157659, and rs1491850 SNPs, and the primary endpoint was time to initiate symptomatic pharmacotherapy. Genotypes were compared using the Cox proportional hazard ratio (HR) with baseline age, sex, site, time since PD diagnosis and rs6265 genotype as covariates. The primary endpoint was associated with a delay with three SNPs: rs10501087 [HR (95% Confidence Interval) = 28.3 (3.6–223.1, p = 0.002) and 7.6 (1.9–29.8, p = 0.004) for T/T and T/C subjects, respectively, vs. C/C subjects], rs1491850 [HR = 3.3 (1.3–8.4, p = 0.04) and 2.8 (1.3–6.4, p = 0.03) for T/T and T/C subjects, respectively, vs. C/C subjects] and rs11030094 [HR = 2.5 (1.1–5.6, p = 0.03) and 2.0 (1.3–6.4, p = 0.03) for A/A and A/G subjects, respectively, vs. G/G subjects]. From the primary endpoint, specific rs10501087, rs1491850, and rs11030094 SNP genotypes are associated with a slower rate of PD progression in the unmedicated state. A prospective clinical trial examining many BDNF SNPs is warranted.

Abstract: Artificial intelligence (AI)‐based diagnostic algorithms have achieved ambitious aims through automated image pattern recognition. For neurological disorders, this includes neurodegeneration and inflammation. Scalable imaging technology for big data in neurology is optical coherence tomography (OCT). We highlight that OCT changes observed in the retina, as a window to the brain, are small, requiring rigorous quality control pipelines. There are existing tools for this purpose. Firstly, there are human‐led validated consensus quality control criteria (OSCAR‐IB) for OCT. Secondly, these criteria are embedded into OCT reporting guidelines (APOSTEL). The use of the described annotation of failed OCT scans advances machine learning. This is illustrated through the present review of the advantages and disadvantages of AI‐based applications to OCT data. The neurological conditions reviewed here for the use of big data include Alzheimer disease, stroke, multiple sclerosis (MS), Parkinson disease, and epilepsy. It is noted that while big data is relevant for AI, ownership is complex. For this reason, we also reached out to involve representatives from patient organizations and the public domain in addition to clinical and research centers. The evidence reviewed can be grouped in a five‐point expansion of the OSCAR‐IB criteria to embrace AI (OSCAR‐AI). The review concludes by specific recommendations on how this can be achieved practically and in compliance with existing guidelines.

Abstract: Microbiomes are all around us in natural and cultivated ecosystems, for example, soils, plants, animals and our own body. Microbiomes are essential players of biotechnological applications, and their functions drive human, animal, plant and environmental health. The rapidly developing microbiome research landscape was studied by a global mapping excercise and bibliometric analysis. Although microbiome research is performed in many different science fields, using similar concepts within and across fields, microbiomes are mostly investigated one ecosystem at-a-time. In order to fully understand microbiome impacts and leverage microbial functions, research needs to adopt a systems approach connecting microbiomes and research initiatives in divergent fields to create understanding on how microbiomes can be modulated for desirable functions as a basis of sustainable, circular bioeconomy., This work was supported by the European Union’s Horizon 2020 R&I programme under grant agreement No. 818116 (MicrobiomeSupport).

8. Drug delivery of 6-bromoindirubin-3’-glycerol-oxime ether employing poly(d,l-lactide-co-glycolide)-based nanoencapsulation techniques with sustainable solvents

Abstract: Background: Insufficient solubility and stability of bioactive small molecules as well as poor biocompatibility may cause low bioavailability and are common obstacles in drug development. One example of such problematic molecules is 6-bromoindirubin-3'-glycerol-oxime ether (6BIGOE), a hydrophobic indirubin derivative. 6BIGOE potently modulates the release of inflammatory cytokines and lipid mediators from isolated human monocytes through inhibition of glycogen synthase kinase-3 in a favorable fashion. However, 6BIGOE suffers from poor solubility and short half-lives in biological aqueous environment and exerts cytotoxic effects in various mammalian cells. In order to overcome the poor water solubility, instability and cytotoxicity of 6BIGOE, we applied encapsulation into poly(d,l-lactide-co-glycolide) (PLGA)-based nanoparticles by employing formulation methods using the sustainable solvents Cyrene™ or 400 g/mol poly(ethylene glycol) as suitable technology for efficient drug delivery of 6BIGOE. Results: For all preparation techniques the physicochemical characterization of 6BIGOE-loaded nanoparticles revealed comparable crystallinity, sizes of about 230 nm with low polydispersity, negative zeta potentials around − 15 to − 25 mV, and biphasic release profiles over up to 24 h. Nanoparticles with improved cellular uptake and the ability to mask cytotoxic effects of 6BIGOE were obtained as shown in human monocytes over 48 h as well as in a shell-less hen’s egg model. Intriguingly, encapsulation into these nanoparticles fully retains the anti-inflammatory properties of 6BIGOE, that is, favorable modulation of the release of inflammation-relevant cytokines and lipid mediators from human monocytes. Conclusions: Our formulation method of PLGA-based nanoparticles by applying sustainable, non-toxic solvents is a feasible nanotechnology that circumvents the poor bioavailability and biocompatibility of the cargo 6BIGOE. This technology yields favorable drug delivery systems for efficient interference with inflammatory processes, with improved pharmacotherapeutic potential. Abstract: We evaluated the accuracy of biology informed genomic prediction for dry matter intake in 2,162 Dutch Holstein cows. Sequence variants were selected from meta-analyses including GWAS summary statistics for QTL and metabolomic QTL in several dairy and crossbred beef populations. Selected variants were prioritized in GBLUP models in a five-fold cross- validation. The accuracies were compared to genomic prediction based on routine 50k genotype data. The average accuracy for the 50k scenario was 0.683. Adding selected sequence variants in the GBLUP model did not improve the accuracies for dry matter intake. Next steps will include testing Bayesian variable selection methods to prioritize variants in genomic prediction for dry matter intake.

Abstract: Background Rare Eye Diseases (RED) are the leading cause of visual impairment and blindness for children and young adults in Europe. This heterogeneous group of conditions includes over 900 disorders ranging from relatively prevalent disorders such as retinitis pigmentosa to very rare entities such as developmental eye anomalies. A significant number of patients with RED have an underlying genetic etiology. One of the aims of the European Reference Network for Rare Eye Diseases (ERN–EYE) is to facilitate improvement in diagnosis of RED in European member states. Main body Technological advances have allowed genetic and genomic testing for RED. The outcome of genetic testing allows better understanding of the condition and allows reproductive and therapeutic options. The increase of the number of clinical trials for RED has provided urgency for genetic testing in RED. A survey of countries participating in ERN-EYE demonstrated that the majority are able to access some forms of genomic testing. However, there is significant variability, particularly regarding testing as part of clinical service. Some countries have a well-delineated rare disease pathway and have a national plan for rare diseases combined or not with a national plan for genomics in medicine. In other countries, there is a well-established organization of genetic centres that offer reimbursed genomic testing of RED and other rare diseases. Clinicians often rely upon research-funded laboratories or private companies. Notably, some member states rely on cross-border testing by way of an academic research project. Consequently, many clinicians are either unable to access testing or are confronted with long turnaround times. Overall, while the cost of sequencing has dropped, the cumulative cost of a genomic testing service for populations remains considerable. Short conclusion Despite technological advances, critical gaps in genomic testing remain in Europe, especially in smaller countries where no formal genomic testing pathways exist. Even within larger countries, the existing arrangements are insufficient to meet the demand and to ensure access. ERN-EYE promotes access to genetic testing in RED and emphasizes the clinical need and relevance of genetic testing in RED.

Abstract: There is no "Planet B". We are living in a climate crisis and the responsibility to address it lies with us all. As astronomers, we have a unique perspective on Earth and climate change: our knowledge of distant worlds means that we appreciate the fragility of Earth and understand the insurmountable difficulties of finding an alternative home. Astronomers for Planet Earth (A4E) is an international group of 800+ past and present astronomy students, educators, and professional scientists united to share their astronomical perspective about the Earth and climate change with the public. The primary goals of the organization are 1) to provide astronomers interested in addressing the climate crisis with a community to discuss ideas and share resources, 2) to provide the public with information about the climate crisis, and 3) to provide the climate movement with an astronomical perspective. A4E is a fast growing network of astronomers from over 40 countries. Since our inception in 2019, we have produced numerous outreach videos, have been involved in organising sustainability sessions at international conferences, have created several calls to action, and our members have authored several articles in Nature Astronomy addressing the intersection of astronomy and climate change....

Abstract: Microbiomes are all around us in natural and cultivated ecosystems, for example, soils, plants, animals and our own body. Microbiomes are essential players of biotechnological applications, and their functions drive human, animal, plant and environmental health. The rapidly developing microbiome research landscape was studied by a global mapping excercise and bibliometric analysis. Although microbiome research is performed in many different science fields, using similar concepts within and across fields, microbiomes are mostly investigated one ecosystem at-a-time. In order to fully understand microbiome impacts and leverage microbial functions, research needs to adopt a systems approach connecting microbiomes and research initiatives in divergent fields to create understanding on how microbiomes can be modulated for desirable functions as a basis of sustainable, circular bioeconomy. Abstract: The articles included in the present issue of the Journal of the LUCAS Graduate Conference, titled Animals (Un)tamed. Human–Animal Encounters in Science, Art, and Literature, are the result of the diverse and interdisciplinary research on our multifaceted relationship with animals which is currently taking place. The fifth biennial LUCAS Graduate Conference, held in April 2019, shared this same theme. This conference on “Animals: Theory, Practice, and Representation” gave PhD and Master students the opportunity to present, exchange and discuss their research relating to animal studies with participants from diverse backgrounds. The resulting exchange focused on a re-examination of the relationship between humans and animals, and the definitions involved. Many of the articles in the present issue are result of the lively debate which took place during this conference. The contributions to this issue of JLGC reflect the wide variety of approaches in animal studies.

19. Impact of emerging technologies on the electricity load profile of residential areas

Abstract: Technological progress and the ambition to de-carbonise the energy sector are changing the way electricity is generated and used. In the residential sector, more efficient electric devices, electrification of mobility and heating systems, as well as on-site PV and battery systems are the main technologies driving this change. These changes will impact the characteristics of the electric demand. A comprehensive understanding of these changes is essential to efficiently design the energy system of tomorrow. The target of this research is to explore the possible changes of the electric load profiles with the current technological development, both at an individual household level and at an aggregate level. An inner-city area of 1550 houses is simulated with a time resolution of 1 minute using a stochastic bottom-up approach. Three scenarios considering refurbishment of buildings, efficiency increase in devices and emerging technologies are simulated. Differences in the load profiles are then analysed. Results show that future load profiles will change significantly. A strong seasonality will appear due to the increased penetration of PV, m-CHP and heat pumps. Intra-day fluctuations will also increase, mainly due to the introduction of electric vehicles and PV. Annual peak load will rise significantly mainly due to the electric back-up heaters of air-source heat pumps. Increased electrification increases annual electricity demand, but results show that this will be partly compensated by better insulated buildings, more energy efficient household devices, and on-site production from PV-Battery systems and m-CHP.

20. The indirubin derivative 6-bromoindirubin-3′-glycerol-oxime ether (6BIGOE) potently modulates inflammatory cytokine and prostaglandin release from human monocytes through GSK-3 interference

Abstract: Indirubin is a natural bis-indole alkaloid contained as active ingredient in the traditional Chinese remedy Danggui Longhui Wan. Indirubin and its 3′-oxime derivatives exhibit anti-cancer and anti-inflammatory properties and they inhibit glycogen synthase kinase (GSK)-3 in cell-free assays where 6-bromoindirubin-3′-oxime (6BIO) is among the most potent analogs. Here, we reveal 6-bromoindirubin-3′-glycerol-oxime ether (6BIGOE) as highly potent derivative able to inhibit pro-inflammatory cytokine, chemokine and prostaglandin (PG) release in human primary monocytes while increasing anti-inflammatory interleukin (IL)-10 levels. 6BIGOE suppressed lipopolysaccharide (LPS)-induced IL-1β and PGE2 release with IC50 of 0.008 and 0.02 µM, respectively, being ≥ 12-fold more potent than 6BIO. The effects of 6BIGOE are mediated via intracellular inhibition of GSK-3, where 6BIGOE again surpassed the effectiveness of 6BIO despite the higher potency of the latter in cell-free GSK-3 activity assays. Side-by-side comparison of 6BIGOE (0.1 µM) with the selective GSK-3 inhibitor SB216763 (5 µM) revealed congruent properties such as enrichment of β-catenin and suppression of cyclooxygenase (COX)-2 protein levels due to GSK–3 inhibition. Metabololipidomics using ultra-performance liquid chromatography-tandem mass spectrometry showed that 6BIGOE selectively decreases pro-inflammatory COX-derived product formation without marked modulation of other lipid mediators. In summary, 6BIGOE is a highly potent indirubin derivative in the cellular context that favorably modulates pro- and anti-inflammatory cytokines as well as COX-2-derived PG via interference with GSK-3. © 2020 Elsevier Inc.

Abstract: This paper presents a stochastic bottom-up model for flexibility in the residential electric load profile. The model accounts for user-behaviour and all relevant technologies, characterising their diversity in sizing and controller settings. The flexibility of technology is mapped into a linear electrical storage equivalent model, which is parameterised on-the-fly during simulation of the load profiles. Using a linear storage representation makes it suitable for optimisation methods that are commonly used in optimal controls. The model is used to study the flexibility of individual technologies as well as of a residential area in Germany taking into account technology penetration rates. The results indicate that EV and PV battery systems offer the largest values of switchable power and storage capacity. However when technology penetration rates are taken into account, heat pumps emerge as a major provider of flexibility.

30. Assessing day-to-day regularity of sleep-wake patterns: theoretical and practical implications of available metrics

Abstract: Background. Day-to-day changes in sleep-wake patterns are important to quantify because they can result in circadian disruption, a risk factor for health outcomes. Traditionally, sleep regularity has been assessed by comparing each day to the average sleep-wake pattern, using metrics such as standard deviation (StDev) and Interdaily Stability (IS). Recently, metrics have been proposed to instead capture variability between consecutive days: the Sleep Regularity Index (SRI) and the Composite Phase Deviation (CPD). Here, we systematically compared these metrics across a range of sources of day-to-day variability, including naps, awakenings, and missing data. Methods. Sleep-wake patterns were synthetically generated over 2-28 days with a weekdayweekend structure. Daily sleep variability was introduced by randomly drawing daily midsleeps and/or sleep durations from a normal distribution with standard deviation ranging from 0- 120min. Average estimates and 95% confidence intervals (CIs) were calculated for each metric under the following scenarios: (1) ‘scrambling’ the order of days, (2) fragmented sleep (i.e. naps, wake after sleep onset (WASO), and all-nighters), (3) varying number of days, and (4) randomly vs. non-randomly (i.e. very early/late sleep more likely to be missing) missing data. Results. (1) Scrambling did not affect IS and StDev values but did affect SRI and CPD values, showing that the metrics measure sleep regularity on different time scales: global vs. circadian. (2) SRI and IS behaved similarly for naps and WASO but differed for all-nighters: SRI values increased (more regular) when all-nighters exceeded 50% of nights, whereas IS yielded monotonically lower (less regular) scores. (3) When based on £ 7 days, StDev and IS overestimated how regular patterns were by up to 40% whereas SRI and CPD were more stable, yet with wider CIs requiring up to 40% larger samples. (4) All metrics were highly sensitive to non-randomly missing data but remarkably stable for up to 50% randomly missing data. Conclusions. All examined metrics have been used for quantifying sleep regularity, yet they measure different aspects and should be seen as complementary rather than redundant. Studies should consider including more than one metric and examining mechanistic links between circadian disruption and sleep regularity.

31. Art Between Species: Two Case Studies of Animal's Agency in Interspecies Art

Abstract: By examining two exemplary cases, this paper addresses the contemporary phenomenon of artistic collaborations between human and non-human animals, which is referred to as interspecies art. Interspecies art has become increasingly significant since the beginning of the twenty-first century and excels at challenging binary oppositions by crediting animals’ creative abilities. Located within the field of human–animal studies, this article combines art historical methods with agency concepts derived from praxeology and action theory. The innovative approach of connecting these ideas of animal agency with interspecies art provides the framework to analyse Aaron Angell’s Gallery Peacetime inhabited by axolotls and CMUK, an interspecies collective consisting of humans and parrots. In order to make the animals’ participation visible as well as to provide a deeper understanding of interspecies art, these specific human–animal relations are examined using Lisa Jevbratt’s and Jessica Ullrich’s criteria for interspecies art and Mieke Roscher’s concepts of entangled and relational agency. This analysis is complemented by a field study and proves to be fertile for revealing the animals’ strong involvement in the artworks as well as beyond the art context.

The Magnetospheric Multiscale (MMS) mission provides a unique opportunity for the long-term monitoring the Earth's magnetotail current sheet prior to and during reconnection activity. Using MMS observations of a number of reconnection events in the magnetotail we investigate how the spatial current density profile across the current sheet varies in time before reconnection is initiated. In addition, we study waves in the current sheet on different scales. These waves may help us understand the role of MHD and kinetic instabilities during current sheet breaking.

Abstract: We report global observations of high‐m poloidal waves during the recovery phase of the 22 June 2015 magnetic storm from a constellation of widely spaced satellites of five missions including Magnetospheric Multiscale (MMS), Van Allen Probes, Time History of Events and Macroscale Interactions during Substorm (THEMIS), Cluster, and Geostationary Operational Environmental Satellites (GOES). The combined observations demonstrate the global spatial extent of storm time poloidal waves. MMS observations confirm high azimuthal wave numbers (m ~ 100). Mode identification indicates the waves are associated with the second harmonic of field line resonances. The wave frequencies exhibit a decreasing trend as L increases, distinguishing them from the single‐frequency global poloidal modes normally observed during quiet times. Detailed examination of the instantaneous frequency reveals discrete spatial structures with step‐like frequency changes along L. Each discrete L shell has a steady wave frequency and spans about 1 R E, suggesting that there exist a discrete number of drift‐bounce resonance regions across L shells during storm times., Key Points Observed long‐lasting high‐m poloidal waves associated with second harmonics of field line resonances during a major magnetic stormDemonstrated global spatial extent of storm time poloidal FLR region using observations from a constellation of widely spaced satellitesRevealed discrete spatial structures of resonant L shells with step‐like frequency changes

Abstract: Background: This retrospective study aimed to investigate a correlation between computed tomography (CT) imaging obtained opportunistically during screening for lumbar spine fractures and dual-energy x-ray absorptiometry (DEXA) scans. Methods: A total of 159 patients over a 5-year study period who had undergone CT scan to investigate for a lumbar spine fracture and a DEXA scan within 12-months of each other were included in the study. Measurements of the region of interest (ROI) through the first 4 lumbar vertebrae were performed to establish Hounsfield unit (HU) values. Measurements for each level were made at three separate locations: mid-vertebral body, and just above and below the inferior and superior endplates, respectively. The HU values were correlated with T-scores obtained from DEXA scans and were further analyzed according to age and gender. Results: There was a statistically significant correlation between HU values with T-score. Using the Pearson correlation coefficients, a moderate correlation of HU value to hip DEXA for T-score. There was a moderate-strong correlation between DEXA lumbar spine with mid-vertebral body HU, with L3 having the strongest correlation (r2=0.7269). The normal group had a mean HU value of 139.3 (95% CI 119–159.2), the osteopenic group had a mean of 105.9 (95% CI 90.4–123.4), and the osteoporotic group had a mean of 72.4 (95% CI 60.1–81.7). Conclusion: This current study demonstrated that opportunistic CT imaging can be utilized to infer bone quality and provide information about the presence of osteoporosis and subsequently fracture risk without the need for additional imaging, radiation exposure, cost, or patient time.

40. Long-term electricity load forecasting: Current and future trends

Abstract: Long-term power-system planning and operation, build on expectations concerning future electricity demand and future transmission/generation capacities. This paper reviews current methodologies for forecasting long-term hourly electricity demand on an aggregate scale (regional or nationally), for 10-50 years ahead. We discuss the challenges of these methodologies in a future energy system featuring more renewable energy sources and tighter coupling between the power sector and the building and transport sectors. Finally, we conclude with some recommendations on aspects to be taken into account regarding long-term load forecasts in a changing power system.

Abstract: Einleitung: Schläfrigkeit am Arbeitsplatz ist speziell in der Luftfahrt von besonderem Interesse für die Sicherheit von Personal und Passagieren. Sehr frühe und späte Dienstzeiten sowie Nachtarbeit gehen mit erhöhter Schläfrigkeit einher, nicht zuletzt durch einen damit verbundenen Schlafmangel. Adenosin ist eine neurochemische Substanz im Gehirn, die sich während des Wachseins ansammelt und während des Schlafes wieder abgebaut wird. Erhöhte Adenosinkonzentrationen können sich negativ auf das Leistungsvermögen und somit die Sicherheit im Luftverkehr auswirken. Ein validiertes bio-mathematisches Modell baut auf diesem Adenosin-System auf, um akkurate Vorhersagen zu Schläfrigkeit und Leistungsvermögen in verschiedenen Arbeitsmodellen (z.B. Anzahl und Verteilung von Früh-/Nachtdiensten sowie freier Tage) zu treffen. Fragestellung: Vergleich der Auswirkung verschiedener (Schicht-)Arbeitspläne der kommerziellen Luftfahrt auf Schlaf und Schläfrigkeit unter Anwendung eines bio-mathematischen Adenosin-Modells. Methodik: Ein validiertes mathematisches Modell des Adenosin-Systems im menschlichen Gehirn wird angewandt zur Vorhersage von Schlaf (Dauer, Zeitpunkt) und Schläfrigkeit. Schläfrigkeit wird hierbei quantifiziert als das Ausbleiben einer Reaktion im sog. Psychomotorischen Vigilanztest (PVT), einem validierten Instrument zur Erfassung der Schläfrigkeit. Reale Dienstpläne von Piloten und Kabinenpersonal werden als Modellinput herangezogen und deren Auswirkungen auf Schlaf und Schläfrigkeit verglichen. Ergebnisse: Dienstpläne von Piloten und Kabinenpersonal werden derzeit über Auskünfte von Airlines sowie in der Literatur veröffentlichte Daten gesammelt und anschließend in das vorhandene Modell eingespeist. Schlussfolgerungen: Ein validiertes Modell zur Vorhersage der Schläfrigkeit von Piloten und Kabinenpersonal kann eingesetzt werden, um Dienstpläne hinsichtlich Schlaf und Aufmerksamkeit zu optimieren und mit Schläfrigkeit verbundene Sicherheitsrisiken zu minimieren.

43. Assessing day-to-day regularity of sleep: theoretical and practical implications of available metrics

Abstract: Objectives Sleep regularity has emerged as an important factor for health in recent years. Metrics differ in their approach to quantifying sleep regularity: Interdaily Stability (IS), Social Jet Lag (SJL), and Standard Deviation (SD) assess variability relative to the mean, whereas the more recent metrics Composite Phase Deviation (CPD) and Sleep Regularity Index (SRI) capture variability on a circadian timescale (i.e., between consecutive days). We systematically assessed and compared these metrics using a range of simulations. Methods Daily sleep patterns were generated for 8 weeks, with later and longer sleep on weekends (0:00-9:00) than weekdays (23:00-6:00). Random variation in sleep timing was systematically increased from 0 min to 360 min in 30-min steps. Mean values and 95% confidence intervals (CIs) were calculated across 10,000 iterations for IS, SJL, SD, CPD, and SRI. Missing data were generated by removing 24h entries, either randomly or non-randomly (e.g., 50% earliest/latest sleep onsets). Results With increasing variation in sleep onset time, all metrics reflected higher irregularity, except SJL, which is sensitive to weekly but not daily changes in sleep timing. As expected, 95% CIs were generally wider for consecutive metrics CPD and SRI than for overall metrics IS and SD. Over the first 14 days, average estimates of IS changed as much as 50% while CPD and SRI remained stable, indicating that IS tends to overestimate how regular sleep patterns are when based on relatively few days. For missing data, 95% CIs were generally wider for consecutive than overall metrics, while their average estimates were more stable, especially for 50% of missing data. The amount of tolerable missing data (e.g., not affecting mean estimates) decreased substantially with increasing non-randomly missing data or variation in sleep timing. Conclusions Overall metrics require relatively many days for an accurate estimate, whereas consecutive metrics such as CPD and SRI are sensitive to daily changes and can better reflect the regularity of patterns that are based on only a few sleep episodes. The right choice of metric may depend on study length, anticipated regularity of the study population, likelihood and distribution of missing data as well as whether the outcome of interest is local vs. global (i.e., accident vs. chronic illness). Future work will examine the metrics’ sensitivity to shift work, nights with no sleep, naps, and fragmented sleep patterns.

Abstract: AIMS Our aim was to report on a survey initiated by the European Association of Percutaneous Cardiovascular Interventions (EAPCI) concerning opinion on the evidence relating to dual antiplatelet therapy (DAPT) duration after coronary stenting. METHODS AND RESULTS Results from three randomised clinical trials were scheduled to be presented at the American Heart Association Scientific Sessions 2014 (AHA 2014). A web-based survey was distributed to all individuals registered in the EuroIntervention mailing list (n=15,200) both before and after AHA 2014. A total of 1,134 physicians responded to the first (i.e., before AHA 2014) and 542 to the second (i.e., after AHA 2014) survey. The majority of respondents interpreted trial results consistent with a substantial equipoise regarding the benefits and risks of an extended versus a standard DAPT strategy. Two respondents out of ten believed extended DAPT should be implemented in selected patients. After AHA 2014, 46.1% of participants expressed uncertainty about the available evidence on DAPT duration, and 40.0% the need for clinical guidance. CONCLUSIONS This EAPCI survey highlights considerable uncertainty within the medical community with regard to the optimal duration of DAPT after coronary stenting in the light of recent reported trial results. Updated recommendations for practising physicians to guide treatment decisions in routine clinical practice should be provided by international societies.

Abstract: The Fluxgate-Searchcoil Merged (FSM) data product for the Magnetospheric Multiscale (MMS) mission is created by combining the level-2 science quality data from the fluxgate and searchcoil magnetometers into a single level-3 data product. The merging method involves noise floor and calibration parameters determined both on the pre- and post-flight. Here, we describe the statistical inter-calibration process as well as the merging filter.

Abstract: International audience; In July 2017, the MMS constellation was evolving in the magnetotail with an apogee of 25 Earth radii and an average inter-satellite distance of 10 km (i.e. at electron scales). On 23rd of July around 16:19 UT, MMS was located at the edge of the current sheet which was in a quasi-static state. Then, MMS suddenly entered in the central plasma sheet and detected the local onset of a small substorm as indicated by the AE index (∼400 nT). Fast earthward plasma flows were measured during about 1 hour starting with a period of quasi-steady flow and followed by a saw-tooth like series of plasma jets. This plasma transport sequence ended up by a flow reversal still occurring close to the magnetic equator. Thanks to the unprecedented MMS measurement capability, these different phases are analyzed in terms of wave activity, current signatures, particle acceleration and heating. The origin of these two phases of plasma transport is discussed.
