3. Identification of a novel B. gibsoni 27-kDa protein as a serodiagnostic antigen

application/pdf, A novel gene encoding 27-kDa protein was identified by the screening of Babesia gibsoni cDNA library with acutely infected dog serum. The BgP27 is a single copy gene with a predicted open reading frame of 762 bp and 254 amino acids. The phylogenic analysis of the deduced amino acid of BgP27 demonstrated considerable identities with members of Plasmodium berghei circumsporozoite protein family that ranged between 18.4% and 22.8%. The BgP27 was expressed as a glutathione S-transferase fusion protein in Escherichia coli. The serum raised in mice against the recombinant protein specifically reacted with a 27-kDa protein in the extracts of B. gibsoni parasites. Confocal laser scanning microscopic observation showed high fluorescent reactivity with both extracellular and intracellular merozoite. Furthermore, recombinant BgP27 was used as an antigen in an enzyme-linked immunosorbent assay (ELISA). Thus, the kinetics of the anti-BgP27 antibody was detected in serial serum samples collected from a B. gibsoni-infected dog. IgG levels were high throughout the course of infection. In addition, the ELISA was able to differentiate between B. gibsoni-infected dog serum and B. canis subspecies-infected dog serum or normal dog serum. The diagnostic performance of BgP27-ELISA revealed the potential use of the antigen for detection of infection in dogs.

International audience; Des structures cryogéniques organisées en réseaux polygonaux de 20 à 30 m ont été mises en évidence dans les sols de la plaine de Pierrelaye-Bessancourt. L'affaissement/déformation des horizons E et BT dans les structures cryogéniques témoigne d'une pédogenèse ancienne intervenue avant la mise en place des cryoturbations lors de la dernière période périglaciaire. Par ailleurs, des revêtements minces d'argile limpides dans la partie supérieure du BT ainsi que des accumulations calcaires disposées en larges bandes horizontales, donc non affectées par la cryoturbation, témoignent eux d'une pédogénèse plus récente, post-cryogénies, durant l'Holocène. Sous épandages d'eaux usées, les sols présentent des traits pédologiques spécifiques qui n'apparaissent jamais dans les sols hors du périmètre : la dégradation des revêtements d'argile, la formation de ferranes et manganes, et le blanchiment de l'horizon E. Ces traits pédologiques témoignent d'une évolution pédologique récente, liée aux grands volumes d'eaux usées (≈ 2000 mm par an), qui ont traversés ces sols durant le 20ième siècle. La configuration des structures cryogéniques conduit à des conditions hydrodynamiques contrastées à l'échelle décamétrique. L'impact sur le transfert de métaux vers le substratum est attesté par des teneurs plus élevées en métaux (Zn et Cu) dans la fraction réactive fine des substrats calcaires à l'écart des structures invaginées.

- Abstract
In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. A key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process vs. those that measure flux through the autophagy pathway (i.e., the complete process); thus, a block in macroautophagy that results in autophagosome accumulation needs to be differentiated from stimuli that result in increased autophagic activity, defined as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (in most higher eukaryotes and some protists such as Dictyostelium) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the field understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular autophagy assays, we hope to encourage technical innovation in the field. © 2012 Landes Bioscience.

- Abstract
The autophagic process is one of the best examples of a conserved mechanism of survival in eukaryotes. At the molecular level there are impressive similarities between unicellular and multicellular organisms, but there is increasing evidence that the same process may be used for different ends, i.e., survival or death, at least at the cellular level. Arthropods encompass a wide variety of invertebrates such as insects, crustaceans and spiders, and thus represent the taxon in which most of the investigations on autophagy in nonmammalian models are performed. The present review is focused on the genetic basis and the physiological meaning of the autophagic process in key models of arthropods. The involvement of autophagy in programmed cell death, especially during oogenesis and development, is also discussed. © 2010 Landes Bioscience.

Experiment A. Chronic toxicity. 140 one day-old male broiler chicks divided in four groups. Every other day: intraperitoneal (ip) L-NAME (LN, 10 mg/kg BW) ; L-arginine (LA, 100 mg/kg BW), L-arginine and a L-NAME combination (100 and 10 mg/kg BW, respectively), and physiological saline (0.90% w/v ; 0.5 mL/kg BW). Seven birds were euthanized weekly during their first five weeks of life. Experiment B. Subchronic toxicity. Equal doses of the same substances: ip every eight hours into 38 day- old birds over 36 hours. Experiment C. Acute toxicity. Thirty day-old birds: one ip dose of various concentrations of L-NAME (50, 100 and 150 mg/kg BW) ; L-arginine (100 mg/kg BW) and saline (0.5 ml/kg BW). All the birds were euthanized after six hours. The end of experiment A: the gross finding of ascites- pulmonary hypertension syndrome (PHS) confirmed vasoconstrictory effect of L-NAME in five birds (LN). Toward the end of experiment A ; in experiment C: histopathological findings (myocardial/pulmonary oedema /LN/ ; congestion/ haemorrhages /LA/) the most prominent (higher LN-doses /100 and 150 mg/kg BW/). Irreversible myocardiolysis and hepatocellulolysis: confirmed in all three experiments (LN). Focal myocardial degeneration solely: in the L- NAME/L-arginine simultaneously treated group. Haematological and blood chemistry values ; stress index value ; and relative organ weights agreed with well-known literature data on PHS. Experiments A and C. The severity of LN- provoked hypoxic changes and plentiful haemorrhages (LA) - dose and time dependent. Mild changes in L-NAME/L-arginine group confirmed a protective role of LA.
