Abstract

Objective: To summarize proceedings of a workshop convened to discuss the current state of science in the disease of osteoarthritis (OA), identify the knowledge gaps, and examine the developmental and regulatory challenges in bringing these products to market. Design: Summary of the one-day workshop held virtually on June 22nd, 2021. Results: Speakers selected by the Planning Committee presented data on the current approach to assessment of OA therapies, biomarkers in OA drug development, and the assessment of disease progression and long-term benefit. Conclusions: Demonstrated by numerous failed clinical trials, OA is a challenging disease for which to develop therapeutics. The challenge is magnified by the slow time of onset of disease and the need for clinical trials of long duration and/or large sample size to demonstrate the effect of an intervention. The OA science community, including academia, pharmaceutical companies, regulatory agencies, and patient communities, must continue to develop and test better clinical endpoints that meaningfully reflect disease modification related to long-term patient benefit.

7. IKKα-mediated biogenesis of miR-196a through interaction with Drosha regulates the sensitivity of cancer cells to radiotherapy

Abstract

Radioresistance is a major obstacle in successful clinical cancer radiotherapy, and the underlying mechanisms are not clear. Here we show that IKKα-mediated miR-196a biogenesis via interaction with Drosha regulates the sensitivity of nasopharyngeal carcinoma (NPC) cells to radiotherapy. Phosphorylation of IKKα at T23 site (p-IKKαT23) promotes the binding of IKKα to Drosha that accelerates the processing of miR-196a primary transcripts, leading to increased expressions of both precursor and mature miR-196a. Dephosphorylation of p-IKKαT23 downregulates miR-196a expression and promotes the resistance of NPC cells to radiation treatment. The miR-196a mimic suppresses while its inhibitor promotes the resistance of NPC to radiation treatment. Importantly, the expression of p-IKKαT23 is positively related to the expression of miR-196a in human NPC tissues, and expression of p-IKKαT23 and miR-196a is inversely correlated with NPC clinical radioresistance. Thus, our studies establish a novel mechanistic link between the inactivation of IKKαT23-Drosha-miR-196a pathway and NPC radioresistance, and de-inactivation of IKKαT23-Drosha-miR-196a pathway would be an efficient way to restore the sensitivity of radioresistant NPC to radiotherapy.

Abstract

Aging is a crucial factor in pavement performance and being able to determine its effect on a mixture is necessary to link its initial properties to the properties over time in order to ensure the intended service life. This is becoming more important now that climate change leads to increased variation in weather conditions, while environmental considerations cause changes in the constituent materials that are used. As a result, past experience is becoming less reliable. In this paper, the USA and European approaches to aging are compared, showing that those contain the same test equipment and almost identical conditions for aging. This allows the exchange of data and experience. The current tests are suitable for binders and give an indication of the sensitivity to aging. For short term aging, Rolling Thin Film Oven Test (RTFOT) conditioning gives a reasonable indication of bitumen aging during asphalt concrete production and construction. This only holds for penetration grade binders during hot mix production and construction. For long term aging, because of the many variables involved, developing a single test method to characterize aging sensitivity seems impossible. However, using more elaborate protocols in existing, practical tests can provide more information and the necessary input for kinetic aging expressions. A pressure aging vessel (PAV) protocol for testing at two temperatures and time intervals, specifically at 90 and 100 degrees Celsius and for 20 and 40 hours, respectively, is suggested. Using the same conditioning in characterizing materials for pavement construction and research will facilitate the exchange of data and enable faster developments.

9. Effect of testosterone therapy on the female voice

Abstract

Objectives This prospective study was designed to investigate the effect of testosterone, delivered by subcutaneous implants, on the female voice. Methods Ten women who had opted for testosterone therapy were recruited for voice analysis. Voices were recorded prior to treatment and at 3 months, 6 months, and 12 months while on testosterone therapy. Acoustic samples were collected with subjects reading a sentence, reading a paragraph, and participating in a conversation. Significant changes in the voice over time were investigated using a repeated-measures analysis of variance with the fundamental frequency (F-0) as a response variable. Demographic variables associated with characteristics of the voice were assessed. Results There were no significant differences in average F-0 related to smoking history, menopausal status, weight, or body mass index. There was no difference in average fundamental speaking frequency (sentence, paragraph, conversation) between the pre-treatment group and any post-treatment group at 3 and 12 months. There was an increase in sentence speech F-0 at 6 months. Two of three patients with lower than expected F-0 at baseline improved on testosterone therapy. Conclusion Therapeutic levels of testosterone, delivered by subcutaneous implant, had no adverse affect on the female voice including lowering or deepening of the voice.

Abstract

Technical specifications for the asphalt concrete properties are developed to be able to specify mixtures that will perform well in pavement applications. Being able to identify and determine properties related to pavement performance in practice is crucial for both road authorities and contractors, since it allows for design and risk management by determining design life times and reliability. However, the properties of Asphalt Concrete (AC) change over its lifetime and since most pavement layers last for a decade or more these changes are crucial in determining the performance in practice. For many of the standard materials the effect of aging is implicitly dealt with in the safety factors that also account for other effects such as the variation rest periods/healing and variations in traffic and weather in the design methods and specifications. Rapid changes in the materials used (increasing percentages reclaimed asphalt, bio-bitumen, rejuvenators, waste materials) and in the production of both bitumen (new refining methods resulting in different composition of bitumen) and asphalt concrete itself (warm mix asphalt, porous asphalt concrete, rubber asphalt mixtures) lead to increased uncertainty in the effects of aging. As a result, the uncertainties in pavement performance increase, which means the prediction of maintenance and the necessary budgets is getting more inaccurate. In order to maintain the ability to reliably design and maintain pavements and determine the most cost-effective solutions for a given situation, a better understanding of the aging processes and objective methods to take into account aging effects on material properties is needed. This need is widely recognized, in the USA the Mechanical Empirical Design Guide takes aging into account through aging tests on the bitumen used and in Europe CEN TC227 works on establishing a method to assess the aging sensitivity of asphalt mixtures. This symposium aimed at combining the existing information and insights from ongoing research into recommendations that will allow the development of methods to determine aging sensitivity and the impact on pavement performance, facilitate the exchange of obtained data and stimulate further developments The resulting recommendations are: •Do make long term aging sensitivity of binders part of the bitumen standards and take the results from the aging sensitivity of binders into consideration when assessing AC properties. Be aware that RTFOT testing only gives an indication of the sensitivity of a penetration grade binder to aging during hot mix production and construction, it doesn’t work for hard grades, PMB’s or warm mixes. •Because of the many variables involved, developing one test method to characterise aging sensitivity seems improbable. However, PAV aging is both practical and, if tests at various conditions are carried out, able to give kinematic properties. A PAV protocol for testing at two temperatures and time intervals could provide practical characterisation information for the short term and enable model development and validation on the long term. RCAT and other aging procedures could also be used in this sense, but considering the availability of equipment and the wide spread experience, PAV is the best candidate to allow the rapid development of international experience with the approach. Based on the current standards and the work presented during the symposium, PAV tests at 90 and 100 degrees Celsius and 20 and 40 hours, respectively, are suggested. The low values for temperature and duration are based on the current standards and fit both the USA and CEN procedure, while research shows that after 40 hours at 100 degrees the chemical (FTIR) and rheological (DSR) properties of laboratory aged and field samples were similar (Section 6.5 and 8.5). At 100oC the temperature is low enough so that the effect of secondary reactions is negligible. As such, these conditions are appropriate for kinetic expressions for in service pavement performance. For high temperature processes and possibly also for repeated recycling (very long term) more sophisticated methods are needed. •Set-up and maintain field monitoring of temperature and UV radiation in various climate zones, as well as regular sampling over time and height to keep checking the predicted changes (from both tests and models), versus the actual changes in properties order to ensure reliability of the data as well as the applicability for pavement performance prediction. In setting up field tests, it is important to get both the composition of the virgin bitumen and the composition after mixing, transport and placement in the pavement. These compositions provide the starting points from both the material and pavement structure point of view and can be used to assess the development of aging products over time. There is a lot of discussion about the impact of binder recovery methods on the observed composition, so until it is proven that this does not have an influence, for comparisons the same recovery method should be used. •Set up a coordination and support action on AC-Aging to continue to exchange information and experiences, both in research and in construction projects. •develop an IR testing protocol, to facilitate the exchange of results and information. •Compare the bitumen composition that is found through various recovery methods to establish if there is an effect and if so, develop a procedure to address this. •To further understanding of aging, a Round Robin test on the differences in test conditions between US and EU, allowing better access to each other’s data and knowledge would be useful. •When developing aging tests for AC, it would be useful to look at the US experience. As long as there is no fundamentally correct method for assessing the aging, it would be preferable to standardize it as much as possible in order to allow cooperation and exchange of data.

11. Testosterone and breast cancer prevention

Abstract

Testosterone (T) is the most abundant biologically active hormone in women. Androgen receptors (AR) are located throughout the body including the breast where T decreases tissue proliferation. However, T can be aromatized to estradiol (E2), which increases proliferation and hence, breast cancer (BCA) risk. Increased aromatase expression and an imbalance in the ratio of stimulatory estrogens to protective androgens impacts breast homeostasis. Recent clinical data supports a role for T in BCA prevention. Women with symptoms of hormone deficiency treated with pharmacological doses of T alone or in combination with anastrozole (A), delivered by subcutaneous implants, had a reduced incidence of BCA. In addition, T combined with A effectively treated symptoms of hormone deficiency in BCA survivors and was not associated with recurrent disease. Most notably, T+A implants placed in breast tissue surrounding malignant tumors significantly reduced BCA tumor size, further supporting T direct antiproliferative, protective and therapeutic effect. 16. Testosterone therapy in women: Myths and misconceptions

Abstract

Although testosterone therapy is being increasingly prescribed for men, there remain many questions and concerns about testosterone (T) and in particular, T therapy in women. A literature search was performed to elucidate the origin of, and scientific basis behind many of the concerns and assumptions about T and T therapy in women. This paper refutes 10 common myths and misconceptions, and provides evidence to support what is physiologically plausible and scientifically evident: T is the most abundant biologically active female hormone, T is essential for physical and mental health in women, T is not masculinizing, T does not cause hoarseness, T increases scalp hair growth, T is cardiac protective, parenteral T does not adversely affect the liver or increase clotting factors, T is mood stabilizing and does not increase aggression, T is breast protective, and the safety of T therapy in women is under research and being established. Abandoning myths, misconceptions and unfounded concerns about T and T therapy in women will enable physicians to provide evidenced based recommendations and appropriate therapy. © 2012 Elsevier Ireland Ltd.

17. Improvement in scalp hair growth in androgen-deficient women treated with testosterone: a questionnaire study

Abstract

Background Androgens are thought to have an adverse effect on female scalp hair growth. However, our clinical experience of androgen replacement therapy in women with androgen deficiency, in which hair loss was seldom reported, led us to question this concept. Objectives To evaluate the effect of subcutaneous testosterone therapy on scalp hair growth in female patients. Methods A total of 285 women, treated for a minimum of 1 year with subcutaneous testosterone implants for symptoms of androgen deficiency, were asked to complete a survey that included questions on scalp and facial hair. Age, body mass index (BMI) and serum testosterone levels were examined. Results Out of the 285 patients, 76 (27%) reported hair thinning prior to treatment; 48 of these patients (63%) reported hair regrowth on testosterone therapy (responders). Nonresponders (i.e. no reported hair regrowth on therapy) had significantly higher BMIs than responders (P = 0.05). Baseline serum testosterone levels were significantly lower in women reporting hair loss prior to therapy than in those who did not (P = 0.0001). There was no significant difference in serum testosterone levels, measured 4 weeks after testosterone implantation, between responders and nonresponders. No patient in this cohort reported scalp hair loss on testosterone therapy. A total of 262 women (92%) reported some increase in facial hair growth. Conclusions Subcutaneous testosterone therapy was found to have a beneficial effect on scalp hair growth in female patients treated for symptoms of androgen deficiency. We propose this is due to an anabolic effect of testosterone on hair growth. The fact that no subject complained of hair loss as a result of treatment casts doubt on the presumed role of testosterone in driving female scalp hair loss. These results need to be confirmed by formal measurements of hair growth.

18. Beneficial effects of testosterone therapy in women measured by the validated Menopause Rating Scale (MRS)

Abstract

Objectives: This study was designed to measure the beneficial effects of continuous testosterone therapy, delivered by subcutaneous implant, in the relief of somatic, psychological and urogenital symptoms in both pre- and post-menopausal patients, utilizing the validated Health Related Quality of Life (HRQOL), Menopause Rating Scale (MRS). Study design: 300 pre- and post-menopausal women with symptoms of relative androgen deficiency, were asked to self-administer the 11-item MRS, at baseline and 3 months after their first insertion of the subcutaneous testosterone implant. Baseline hormone measurements, menopausal status and BMI, were assessed to determine correlation with symptoms and clinical outcome. Main outcome measurements: Changes related to therapy were determined. Total MRS scores as well as psychological, somatic and urogenital subscale scores were compared prior to therapy and following testosterone implant therapy. Results: Pre-menopausal and post-menopausal females reported similar hormone deficiency symptoms. Both groups demonstrated similar improvement in total score, as well as psychological, somatic and urogenital subscale scores with testosterone therapy. Better effect was noted in women with more severe complaints. Higher doses of testosterone correlated with greater improvement in symptoms. Conclusion: Continuous testosterone alone, delivered by subcutaneous implant, was effective for the relief of hormone deficiency symptoms in both pre- and post-menopausal patients. The validated, HRQOL questionnaire, Menopause Rating Scale (MRS), proved a valuable tool in the measurement of the beneficial effects of testosterone therapy in both cohorts. © 2010 Elsevier Ireland Ltd. Extremhochwasser und ihre Auswirkungen haben in den letzten Jahren im Kontext der rezenten Klimaveränderungen in Zentraleuropa zunehmende Aufmerksamkeit erhalten. Die Betrachtung historischer Hochwasserkatastrophen sowie ihrer regionalen Auswirkungen im Rahmen langzeitlicher Klimavariabilität liefern ein wertvolles Verständnis zu vergangenen, gegenwärtigen sowie zukünftigen Beziehungen zwischen Klima und extremen Hochwassern (BARDOSSY & FILIZ 2005). Daher erhalten historische Hochwasser ein immer größeres Interesse in der Hochwasserforschung und im angewandten Hochwasserschutz (BENITO et al. 2005, GLASER & STANGL 2004).

Abstract

A sample of events with two distinct jets, in addition to the proton remnant, has been identified in deep inelastic, neutral current ep interactions recorded at HERA by the ZEUS experiment. For these events, the mass of the hadronic system ranges from 40 to 260 GeV. The salient features of the observed jet production agree with the predictions of higher order QCD.

Abstract

A search for any resonant state coupled to an electron and a proton constituent has been performed using collisions of electron and proton beams at HERA. In a sample with integrated luminosity of 26 nb-1, no evidence has been found for production of leptoquarks with decays to e- + jet or v + jet. Limits on the coupling strength of scalar leptoquarks to electron and quark have been determined for masses above 25 GeV. For example, scalar isosinglet leptoquarks (S0) with electroweak coupling strength to (e- u) states are ruled out at the 95% confidence level for masses below 168 GeV for left-handed couplings and below 176 GeV for right-handed couplings.

Abstract

In deep inelastic, neutral current scattering of electrons and protons at square-root s = 296 GeV, we observe in the ZEUS detector events with a large rapidity gap in the hadronic final state. They occur in the region of small Bjorken x and are observed up to Q2 of 100 GeV2. They account for about 5% of the events with Q2 greater-than-or-equal-to 10 GeV2. Their general properties are inconsistent with the dominant mechanism of deep inelastic scattering, where color is transferred between the scattered quark and the proton remnant, and suggest that the underlying production mechanism is the diffractive dissociation of the virtual photon.

Abstract

Results are presented on neutral current, deep inelastic scattering measured in collisions of 26.7 GeV electrons and 820 GeV protons. The events typically populate a range in Q2 from 10 to 100 GeV2. The values of x extend down to x is similar to 10(-4) which is two orders of magnitude lower than previously measured at such Q2 values in fixed target experiments. The measured cross sections are in accord with the extrapolations of current parametrisations of parton distributions.

30. Chronic stress and age-related increases in the proinflammatory cytokine IL-6

Abstract

Overproduction of IL-6, a proinflammatory cytokine, is associated with a spectrum of age-related conditions including cardiovascular disease, osteoporosis, arthritis, type 2 diabetes, certain cancers, periodontal disease, frailty, and functional decline. To describe the pattern of change in IL-6 over 6 years among older adults undergoing a chronic stressor, this longitudinal community study assessed the relationship between chronic stress and IL-6 production in 119 men and women who were caregiving for a spouse with dementia and 106 noncaregivers, with a mean age at study entry of 70.58 (SD = 8.03) for the full sample. On entry into this portion of the longitudinal study, 28 of the caregivers' spouses had already died, and an additional 50 of the 119 spouses died during the 6 years of this study. Levels of IL-6 and health behaviors associated with IL-6 were measured across 6 years. Caregivers' average rate of increase in IL-6 was about four times as large as that of noncaregivers. Moreover, the mean annual changes in IL-6 among former caregivers did not differ from that of current caregivers even several years after the death of the impaired spouse. There were no systematic group differences in chronic health problems, medications, or health-relevant behaviors that might have accounted for caregivers' steeper IL-6 slope. These data provide evidence of a key mechanism through which chronic stressors may accelerate risk of a host of age-related diseases by prematurely aging the immune response.

The total photoproduction cross section is determined from a measurement of electroproduction with the ZEUS detector at HERA. The Q^2 values of the virtual photons are in the range 10^-7 < Q^2 < 2 10^-2 GeV. The gamma-p cross section in the gamma-p centre of mass energy (186-223) GeV is 154 +-16(stat.) +-32 (syst.) mub

Abstract

The purpose of this article is to summarize research findings pertaining to the effects of functional electrical stimulation (FES) lower limb exercise training on health, fitness, and function in individuals with spinal cord injury. This lays the foundation for defining the potential clinical efficacy of FES exercise for reducing the risk of secondary medical complications. This article also reviews the effects of FES exercise on the muscular, skeletal, integumentary, immune, and cardiopulmonary systems, as well as on functional performance.

Abstract

Using the ARGUS detector at DORIS II, we have analyzed tau decays into π − π − π + π 0 ν τ . The branching ratio was determined to be (4.2±0.5±0.9)%. From a study of the three pion sub-system, we find evidence for the decay τ − → ωπ − ν τ with a branching ratio of (1.5±0.3±0.3)%. The ωπ − system is found to be predominantly in a J P =1 − state. There is no evidence for second-class axial-vector currents. The spectral function for the ωπ − final state is in agreement with a prediction based on CVC and σ e + e −→ ωπ 0 .

Abstract

Sera from healthy individuals and patients with infectious mononucleosis, Burkitt lymphoma, nasopharyngeal carcinoma, or other malignancies were examined for their capacity to neutralize Epstein-Barr virus (EBV)-induced DNase activity. Sera were found that neutralized the EBV DNase but not herpes simplex virus type 1 or type 2 DNases, and vice versa. Sera from 46 of the 49 patients with nasopharyngeal carcinoma examined (94%) neutralized > 6 units of EBV DNase per ml of serum. In contrast, only 19% of 47 patients with Burkitt lymphoma, 12% of 183 patient with other malignancies, 4% of 58 patients with infectious mononucleosis, and none of 101 healthy individuals had such levels of neutralizing activity. The neutralizing factor was found in the IgG fraction derived from nasopharyngeal carcinoma sera. There was no correlation between the concentration of these antibodie and the titers of IgG ad IgA antibodies to the EBV capsid antigen, the early antigen complex, or the EBV-associated nuclear antigen.
