The pharmacovigilance practices aim to improve the safety and care of patient in relation to the utilization of medications, as well as other medical and paramedical interventions. Due to emergence of outbreaks of infections, the sub-Saharan African countries have been widely utilizing the trial drugs, which have been increasing the events of Adverse Drug Reactions (ADRs) in the region. This systematic literature review aims to examine the characteristics, knowledge, attitude and pharmacovigilance practices performed by health care professionals in sub-Saharan Africa. This systematic review included N=7 studies, published between 2010 and 2020, and dealing with characteristics, knowledge, attitude and pharmacovigilance practices performed by health care professionals of sub-Saharan African region were extracted from CINAHL, Science Direct, and PubMed. Seven journal articles were selected through PRISMA research tool to identify the key factors which included inadequate training, under-reporting, knowledge assessment, practice, and good attitude. All selected studies were published in sub-Saharan Africa for this systematic review. The healthcare professionals working in the region of sub-Sahara Africa lack expertise in pharmacovigilance practices, due to lack of awareness, unavailability of support, and several other factors. Pharmacovigilance practices can be improved by arranging education, training, and role-play sessions for them. In insect communities, the outcome of intra- and inter-specific competitions for food utilisation depend primarily upon density and duration even inter-specific competitions can occur when they are not sharing the same feeding niche such as between foliar feeders and stemborers. Experimental manipulations of larval densities and the durations of common diet feeding of fall armyworm (FAW), S. frugiperda, and the African lepidopteran stemborers, Busseola fusca, Sesamia calamistis and Chilo partellus, were conducted to determine how the density and the duration of resource utilization affected larval survival and the relative growth rate (RGR) in intra- and inter-specific interactions. The results showed both intra- and interspecific competitions were observed among all the four species and interspecific competition was significantly stronger between the stemborers than between the FAW and the stemborers. The results showed that multiple infestations of cereal plants with low larval densities of each species at optimum conditions will very likely prolong the coexistence between FAW and stemborers. In addition, the time partitioning of the resource use significantly influenced this coexistence.

Exotic invasive insect herbivores have the potential to interfere with existing herbivore-natural enemy interactions in new environments. Fall armyworm (FAW), Spodoptera frugiperda J.E. Smith (Lepidoptera: Noctuidae), is a new invasive pest in maize fields in Africa. Understanding the acceptability and suitability of FAW to existing maize stemborer-parasitoid interactions is the first step in elucidating the impact that this exotic insect pest can have on the existing natural enemies used in biological control of maize stemborers in Kenya. The most commonly used larval parasitoids for biological control programs against maize stemborer communities in East Africa are Cotesia flavipes Cameron (Hymenoptera: Braconidae) and two populations of the native Cotesia sesamiae (Cs-Inland and Cs-Coast) Cameron (Hymenoptera: Braconidae). All these parasitoid species attacked FAW larvae but none yielded offspring, although they induced high non-reproductive host mortality when compared to natural mortality. Furthermore, the parasitoids that inserted their ovipositor into FAW larvae exhibited no significant preference between FAW larvae and their respective stemborer hosts under dual-choice bioassays. In olfactometer bioassays, the parasitoids were more attracted to plants infested by FAW than uninfested plants and even showed a marked preference for the odours of plants infested by FAW over those of plants infested by their natural host counterparts. This study illustrates that exotic pests, such as FAW, can impact existing stemborer-parasitoid interactions associated with maize, even if they cannot be used as hosts by parasitoids associated with these stemborers. Although additional studies are needed, FAW might therefore have a negative impact on stemborer biological control existing before its invasion.

Typhoid fever is caused by the bacteria Salmonella enterica subspecies enterica serovar Typhi (S. Typhi) and remains a significant health problem in many developing countries. Lack of adequate diagnostic capabilities has contributed greatly in making typhoid fever endemic in these regions. Reliable and inexpensive diagnostic tests are needed to improve the management of this disease burden. We evaluated the ability of staA, viaB and sopE genes to detect and differentiate between the three most prevalent Salmonella spp. in Kenya (S. Typhi, S. Typhimurium and S. Enteritidis) using conventional polymerase chain reaction (PCR). The staA primers and viaB primers were found to be specific only for the different strains of S. Typhi, producing PCR products of 585 bp and 540 bp, respectively. The sopE primers was demonstrated to be specific for all Salmonella spp. producing a 465 bp PCR product with no amplification with E. coli and S. boydii bacterial strains.

Olfactory cues may influence host plant preferences for oviposition of female moths within a community of stemborers that utilise the same resource. This study aimed to evaluate plant preferences for oviposition of gravid females of noctuid stemborers, Busseola fusca and Sesamia calamistis, and the crambid Chilo partellus for uninfested maize plants and plants infested by conspecific or heterospecific larvae. The involvement of volatile organic compounds (VOCs) emitted by uninfested and maize plants infested by conspecific or heterospecific larvae on moth orientation was studied in Y-tube olfactometer assays and in the field. All gravid female moths significantly preferred VOCs emitted by plants infested by conspecific or heterospecific larvae over those from uninfested plants, and female moths did not systematically prefer VOCs emitted by plants infested by conspecifics. Field trials confirmed these results. Chemical analysis by coupled gas chromatography/mass spectrometry showed that VOCs emitted by larvae-infested plants, regardless of the stemborer species, were compositionally richer than those released by uninfested plants but their emission intensity varied with species involved in the infestation. Busseola fusca larvae induced a compositionally richer VOCs profile than S. calamistis and C. partellus larvae. Eight candidate attractants were associated with larvae-infested plants. These results open new avenues to develop attractants specific to trap female stemborer moths in the field.

This has often led to misdiagnosis of the disease, thereby delaying appropriate treatment and making typhoid fever widespread in most resource poor areas. This study examines the ability of three different genes to detect and differentiate between the three most prevalent Salmonella strains in Kenya using a readily available and widely used genetic test known as conventional polymerase chain reaction (PCR). This research found that staA and viaB genes were specific only for S. Typhi, while the sopE gene was specific for all Salmonella strains. Consequently, conventional PCR using these staA and viaB genes for detection of S. Typhi shows great potential to be used as a readily available diagnostic tool to detect the presence of the S. Typhi organism in individuals or foods sample in Kenya.

Intra- and interspecific interactions within communities of species that utilize the same resources are characterized by competition or facilitation. The noctuid stemborers, Busseola fusca and Sesamia calamistis, and the crambid stemborer, Chilo partellus were the most important pests of maize in sub-Saharan Africa before the recent &ldquo, invasion&rdquo, of fall armyworm (FAW), Spodoptera frugiperda, which currently seriously limits maize yields in Africa. This new pest is interacting with the stemborer community at the larval stage in the use of maize resources. From a work done by Ntiri et al. [13] on the influence of temperature on the larval intra- and interspecific resources utilization within the community of Lepidoptera stemborers involving B. fusca, S. calamistis, and C. partellus, there is a need to update this study by adding the new pest, S. frugiperda, in order to understand the effect of temperature on the larval interactions of all these four species under the context of climate change. The influence of temperature on intra- and interspecific larval interactions was studied in the same protocol of Ntiri et al. [13] using artificial stems kept at different constant temperatures (15 &deg, C, 20 &deg, C, 25 &deg, C, and 30 &deg, C) in an incubator and assessing survival and relative growth rates of each species in single and multi-species experiments. After the inclusion of FAW into the experiments, with regard to relative growth rates, both intra- and interspecific competition was observed among all four species. With regard to survival rates, cannibalism can also explain the intra- and interspecific interactions observed among all four species. Interspecific competition was stronger between the stemborers than between the FAW and the stemborers. Similar to lepidopteran stemborers, temperature affected both survival and relative growth rates of the FAW as well. Regardless of the temperature, C. partellus was superior in interspecific interactions shown by higher relative growth and survival rates. The results suggest that the FAW will co-exist with stemborer species along entire temperature gradient, though competition and/or cannibalism with them is weak. In addition, temperature increases caused by climate change is likely to confer an advantage to C. partellus over the fall armyworm and the other noctuids.

Sources of infestation are the key elements to be considered in the development of habitat management techniques for the control of maize stemborers. Several wild plants, grasses mostly, have been identified that serve as hosts for stemborers and their parasitoids during the off-season when maize is not present in the field. However, their abundance is much lower in wild plants compared to cultivated fields. Thus, the role of wild plants as a reservoir for cereal stemborers and their parasitoids is still controversial, particularly in agro-ecosystems with reduced wild habitat. We studied the occurrence of different maize stemborers and associated parasitoids in maize stem residues and wild grasses during non-cropping seasons as potential carry-over populations to subsequent early season maize plants. Surveys were conducted in the central region of Kenya during long and short dry seasons in maize residues and wild grasses as well as during the two rainy seasons in maize plants at earlier and late whorl stages during the years of 2017 and 2018. Wild habitat had a higher species diversity than maize residues habitat, but maize residues had a higher abundance of maize stemborer species, such as Busseola fusca, Sesamia calamistis, and Chilo partellus, and of associated parasitoid species (i.e., Cotesia flavipes and Cotesia sesamiae) than wild plants. Our surveys, complemented by field parasitoid releases of C. flavipes and C. sesamiae, indicated that maize residues constitute a better refugia reservoir not only of the maize stemborers but also of C. flavipes and C. sesamiae during non-cropping seasons as compared to wild plants and, thus, might constitute in this region the main source of both stemborers and C. flavipes/C. sesamiae carry-over in maize plants during the subsequent cropping season. Thus, systematic destruction of maize residues would not help the biological control of lepidopteran stemborers. This is particularly true in areas with reduced wild habitat.

The stem borer Busseola fusca (Fuller) (Lepidoptera: Noctuidae) is an important pest of maize and sorghum in sub-Saharan Africa. This insect has oligophagous feeding habits, feeding mostly on maize and sorghum with a narrow range of wild Poaceous plant species. We hypothesised that first instar B. fusca larvae, the critical stage for successful establishment on a host plant, can establish and then grow on a particular plant as a result of induction of a complement of digestive enzymes that mediates host acceptance at first instars. A fast semi-quantitative analysis of potentially digestive enzymatic activities present in the first larvae previously fed for 4 days on leaves of host and non-host plants was performed using the API-ZYM kit system able to detect a multiplex of enzyme activities. Regardless of the plant species, the larvae exhibited higher activities of the carbohydrate metabolising enzymes than of aminopeptidases and proteases. In addition, highest activities of carbohydrates degrading enzymes were exhibited by larvae that consumed leaves of the most preferred plant species of B. fusca. Conversely, esterases were only detected in neonate larvae that consumed leaves of the less preferred and non-host plants. No alkaline phosphatase and lipase activities were detected. The significance of these results was discussed in terms of food requirements of first instar larvae when settling on a plant.

Foraging insect parasitoids use specific chemical cues to discriminate between host and non-host species. Several compounds have been identified in “host location and acceptance.” However, nothing is known about the molecular variations in these compounds that could account for host-range differences between parasitoid species. In a previous study, it was shown that during the host-finding process, contact between the braconid Cotesia flavipes and its host is crucial, and that α-amylase of oral secretions from the host plays a key role for host acceptance and oviposition by the parasitoid. The present study sought to establish whether the variations in this enzyme could explain specific host recognition in different host-parasitoid associations. Different species and populations of the C. flavipes complex specialized on graminaceous lepidopteran stemborers were used. Electrophoresis of α-amylase revealed different isoforms that mediate the parasitoid's oviposition acceptance and preference for a specific host. This discovery opens up new avenues for investigating the evolutionary processes at play in chemically-mediated host specialization in the species-rich Cotesia genus.

Although, the volatiles play a relevant role in the localization by parasitoids of their hosts feeding on plants, the host identification process for acceptance occurs mainly during contact between the parasitoid and its host where host products related to feeding activities, fecal pellets and oral secretions, play a crucial role. The purpose of this study was to identify the nature of the contact kairomone(s) that mediate the acceptance for oviposition of the parasitoid Cotesia flavipes Cameron (Hymenoptera, Braconidae), which was released in Kenya in 1993 to control the invasive crambid Chilo partellus (Swinhoe). Using host and non-hosts of C. flavipes, we showed that it is mainly the oral secretions of the larvae that harbour the active compound(s) that mediate host acceptance for oviposition by C. flavipes. Using an integration of behavioral observations and biochemical approaches, the active compound of the oral secretions was identified as an alpha-amylase. Using synthetized alpha-amylases from Drosophila melanogaster (an insect model for which syntheses of active and inactive alpha-amylases are available), we observed that the conformation of the enzyme rather than its catalytic site as well as its substrate and its degradation product is responsible for host acceptance and oviposition mediation of C. flavipes females. The results suggest that the alpha-amylase from oral secretions of the caterpillar host is a good candidate for an evolutionary solution to host acceptance for oviposition in C. flavipes.

The noctuid stem borer Busseola fusca is an important pest of maize and sorghum in Sub-Saharan Africa. The presence of this species occurred mostly on cultivated than on wild habitats. Busseola fusca is oligophagous having a narrow range of a wild grass species. This might be due, in part, to differences in silicon (Si) content in plant tissues between cultivated and wild grasses. In the present study, we have tested this hypothesis by studying the survival and the relative growth rate (RGR) expressed as daily weight gains of B. fusca larvae on maize and six wild host plants, mostly present in the natural habitat where B. fusca occurred, and correlated with their Si contents. Survival and RGR of B. fusca larvae were considerably higher on maize and wild sorghum than on the other grass species, and they were negatively related to plant Si content. This was corroborated with results on RGR from artificial diets amended with increasing levels of Si. In addition, if Si was added to maize growing substrate B. fusca larval growth was significantly reduced confirming the involvement of Si in B. fusca larvae – Poaceae interactions. The results provide insight into the possible mechanisms of oligophagy of B. fusca and provide a correlative support for a physical role of plant endogenous Si in impeding feeding of B. fusca larvae.

Foraging parasitoids use chemical signals in host recognition and selection processes. Thereby, chemicals from the herbivore hosts play a crucial role. When different herbivores are present in the same plant or field, the perception of specific volatiles and contact compounds emitted from the host itself enable the parasitoids both to differentiate between hosts and non-hosts and to estimate the health status of its host. During the host feeding process, contact between the parasitoid and its host is very crucial, and oral secretions from the host play a key role during the first contact for such evaluation by the parasitoid. Using an integration of behavioral observations, biochemical and sensory physiological approaches we demonstrate that female parasitoids of Cotesia flavipes recognize their host and oviposit in reaction to an α-amylase, which is present in the oral secretions of the larvae of their host, Chilo partellus. This activity was also mediated by a purified α-amylase synthetized from Drosophila melanogaster. Using this synthetized enzyme, we further demonstrate that the conformation of the enzyme rather than its catalytic site is responsible for this activity. This enzyme is activating gustatory neurons of the terminal antennal sensilla chaetica of C. flavipes females. α-amylases are therefore good candidates for an evolutionary solution to host selection in parasitoids, thus opening new avenues for investigations in hosts-parasitoids interactions.

Background Salmonella has significant public health implications causing food borne and zoonotic diseases in humans. Treatment of infections due to Salmonella is becoming difficult due to emergence of drug resistant strains. There is therefore need to characterize the circulating non-typhoidal Salmonella (NTS) serovars in domestic animals and animal products in Kenya as well as determine their antibiotic resistance profiles. Methods A total of 740 fecal samples were collected from cows (n = 150), pigs (n = 182), chicken (n = 191) and chicken eggs (n = 217) from various markets and abattoirs in Nairobi. The prevalence of NTS serovars using culture techniques and biochemical tests, antimicrobial sensitivity testing using disc diffusion method of the commonly prescribed antibiotics and phylogenetic relationships using 16S rRNA were determined. Results The results showed that the overall prevalence of Salmonella was 3.8, 3.6, 5.9 and 2.6% for pigs, chicken, eggs and cows respectively. Two serovars were isolated S. Typhimurium (85%) and S. Enteritidis (15%) and these two serovars formed distinct clades on the phylogenetic tree. Forty percent of the isolates were resistant to one or more antibiotics. Conclusion The isolation of Salmonella Typhimurium and Salmonella Enteritidis that are resistant to commonly used antibiotics from seemingly healthy animals and animal products poses a significant public health threat. This points to the need for regular surveillance to be carried out and the chain of transmission should be viewed to ascertain sources of contamination.

The stem borer Busseola fusca (Fuller) (Lepidoptera: Noctuidae) is a major pest of maize, Zea mays L., and sorghum, Sorghum bicolor (L.) Moench (both Poaceae), in sub-Saharan Africa. Like in many other lepidopteran insects, the success of B. fusca in recognizing and colonizing a limited variety of plants is based on the interaction between its sensory systems and the physicochemical characteristics of its immediate environment. The sensilla on the maxillary galeae of B. fusca larvae are typical of Lepidoptera and comprise two uniporous styloconic sensilla, which are contact chemoreceptors, three basiconic sensilla, and two aporous sensilla chaetica. The maxillary palp is two-segmented and has eight small basiconic sensilla at the tip, which were also found to be gustatory. The antennae of B. fusca larvae are short and simple. The sensilla of the antenna are composed of two aporous sensilla chaetica, three multiporous cone-shaped basiconic sensilla, three small basiconic sensilla, and one aporous styloconic sensillum. The basiconic sensillum located on the third antennal segment displayed a contact chemoreception response. The other basiconic sensilla did not show any action potential activity in tip-recording tests. The significant and positive dose-response curve obtained for the antennal basiconic sensillum with sucrose indicated for the first time the presence of gustatory chemoreceptors on the antennae of a lepidopteran larva.

International audience; The chemical composition of plant surfaces plays a role in selection of host plants by herbivorous insects. Once the insect reaches the plant, these cues determine host acceptance. Laboratory studies have shown that the stem borer Busseola fusca (Lepidoptera: Noctuidae), an important pest of sorghum and maize in sub-Saharan Africa, is able to differentiate between host and non-host plant species. However, no information is available on the cues used by this insect to seek and accept the host plant. Thus, the role of surface phytochemical stimuli on host selection and oviposition by B. fusca was studied in the laboratory using two host plants, sorghum, Sorghum bicolor, and maize, Zea mays, and one non-host plant, Napier grass, Pennisetum purpureum. The numbers of eggs and egg masses deposited on the three plant species were compared first under no-choice and choice conditions. In both cases, more eggs and egg masses were laid on maize and sorghum than on the non-host. Artificial surrogate stems treated with a water or chloroform surface extract of each plant were then compared with surrogate stems treated with, respectively, water or chloroform as controls, under similar conditions. Surrogate stems treated with plant water extracts did not show an increase in oviposition when compared to controls, indicating that the major compounds in these extracts, i.e., simple sugars and free amino acids, are not significantly responsible for the oviposition preference. By contrast, a chloroform extract of sorghum enhanced oviposition on the surrogate stems compared to the control, while those of maize and Napier grass showed no significant effects. Analysis of the chloroform extract of sorghum showed higher amounts of alpha-amyrin, -amyrin, and n-nonacosane compared to those of maize and Napier grass. A blend of the three chemicals significantly increased oviposition compared to the chloroform-treated control, indicating that these compounds are part of the surface chemical signature of the plant responsible for host recognition and oviposition by B. fusca.

In East Africa, lepidopteran stemborers such as Chilo partellus and Busseola fusca are major constraints to production of maize, which is the main staple food crop in the region. Cereals depend on silicon (Si)-based defences to fight off herbivores. Using altitudinal ranges in the East African highlands as ecological surrogates for inferring climate change, it was shown that Si concentrations in soil and maize decreased with altitude. This was attributed, in part, to low temperatures at high altitudes, which negatively affected Si assimilation by maize. Experiments showed that B. fusca was more susceptible to Si than C. partellus . Hence the predominance of B. fusca in the highlands and of C. partellus in the lowlands could be partly explained by altitudinal differences in Si concentrations in maize plants. Therefore, a rise in temperature due to climate change should enhance the plants’ Si assimilation and as a result C. partellus might move into the higher altitudes and increasingly displace B. fusca .

The rapid increase and subsequent management of Typhoid fever in Kenya has been complicated by the emergence and rise of antimicrobial resistance to the Salmonella Typhi pathogen. This situation is yet to be addressed due to the paucity of information regarding the circulating strains and their antimicrobial resistance profiles. This study was hence designed to genetically characterize and evaluate the antimicrobial resistance patterns of human Salmonella Typhi isolates circulating among patients in Kenyatta National Hospital the largest referral health facility in Kenya. Fifty (n=50) Salmonella Typhi archived sample isolates were obtained from stool samples of patients suffering from typhoid fever who attended the facility from 2010 to 2015. The isolates were evaluated for phylogenetic relationships targeting 16s rRNA gene sequences and antimicrobial susceptibilities against a panel of 12 drugs by disk diffusion method. Phylogenetically, the isolates clustered close to Salmonella Typhi strain CT18 and its plasmid pHCM1 and plasmid IncHI1 of Salmonella Typhi strain R27 both associated with drug resistance. Antimicrobial susceptibility tests showed that 73% of the isolates were resistant to the Aminopenicillin, Sulfonamide, Phenicol and Aminoglycoside classes of drugs. A resistance of between 7% to 19% was observed for the Beta-lactamase, Fluoroquinolone and Cephalosporin classes of drugs. Only one of the isolates was fully susceptible to all the antimicrobial drugs used. A total of 23% of the isolates were intermediately resistant to all the 12 drugs used. The high resistance patterns of Salmonella Typhi evidenced necessitates the re-evaluation of the currently used antibiotic regimen and also underscores the importance of continued drug resistance monitoring for this pathogen. Plant sugars are often considered as primary feeding stimuli, conditioning host plant acceptance by herbivorous insects. Of the nine sugars identified from methanolic extracts of seven grass species, only turanose, a sucrose isomer, was negatively correlated with the survival and growth of the noctuid larva of cereal stemborer, Busseola fusca. Sucrose was the most abundant sugar, although it did not vary significantly in concentration among the plant species studied. Using Styrofoam™ cylinders impregnated with increasing concentrations of turanose or sucrose, the two sugars had opposing effects: turanose appeared phagodeterrent while sucrose was phagostimulatory. Electrophysiological studies indicated that B. fusca larvae were able to detect both sugars via their styloconic sensilla located on the mouthparts. The findings indicate that, whereas sucrose is a feeding stimulant and positively influences food choice by B. fusca larvae, turanose negatively contributes to larval food choice. The balance in concentrations of both sugars, however, somehow influences the overall host plant choice made by the larvae. This can partly explain host plant suitability and choice by this caterpillar pest in the field.

International audience; The present study was aimed at characterizing differences in mate acceptance and host plant recognition between Busseola fusca (Lep.: Noctuidae) reared for several generations under laboratory conditions and wild conspecifics, directly collected from maize stems in the field. The mating success was significantly higher in laboratory reared when compared with the wild B. fusca population. Oviposition on artificial stems was significantly higher for laboratory-reared insects than for the wild ones. Moreover, unlike adults of the wild strain, laboratory-reared B. fusca showed no preference to oviposit on surrogate stems impregnated with maize extracts. Long-range attraction to the host plant was significantly lower for laboratory-reared insects. Furthermore, the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) analysis of mitochondrial DNA showed that the wild type and laboratory populations belonged to the same genetic strain, indicating that these differences between the populations were mainly phenotypic.
