Publisher Copyright: © 2022 American Chemical Society We study the fusion of homogeneous cell aggregates and of hybrid aggregates combining cells and microparticles. In all cases, we find that the contact area does not vary linearly over time, as observed for liquid drops, but rather it follows a power law in t2/3. This result is interpreted by generalizing the fusion model of soft viscoelastic solid balls to viscoelastic liquid balls, akin to jelly pearls. We also explore the asymmetric fusion between a homogeneous aggregate and a hybrid aggregate. This latter experiment allows the determination of the self-diffusion coefficient of the cells in a tissue by following the spatial distribution of internalized particles in the cells.

Abstract

Summary: Both cross-presentation of antigens by dendritic cells, a key pathway triggering T cell immunity and immune tolerance, and survival of several pathogens residing in intracellular vacuoles are intimately linked to delayed maturation of vesicles containing internalized antigens and microbes. However, how early endosome or phagosome identity is maintained is incompletely understood. We show that Toll-like receptor 4 (TLR4) and Fc receptor ligation induces interaction of the GTPase Rab14 with the kinesin KIF16b mediating plus-end-directed microtubule transport of endosomes. As a result, Rab14 recruitment to phagosomes delays their maturation and killing of an internalized pathogen. Enhancing anterograde transport by overexpressing Rab14, promoting the GTP-bound Rab14 state, or inhibiting retrograde transport upregulates cross-presentation. Conversely, reducing Rab14 expression, destabilizing Rab14 endosomes, and inhibiting anterograde microtubule transport by Kif16b knockdown compromise cross-presentation. Therefore, regulation of early endosome trafficking by innate immune signals is a critical parameter in cross-presentation by dendritic cells. : Weimershaus et al. identify a molecular complex that controls the intracellular trafficking along microtubules of antigens internalized by dendritic cells. They show that this trafficking is regulated by innate immune signals and regulates presentation of internalized antigens to T lymphocytes. Abstract

Transendothelial cell macroaperture (TEM) tunnels control endothelium barrier function and are triggered by several toxins from pathogenic bacteria that provoke vascular leakage. Cellular dewetting theory predicted that a line tension of uncharacterized origin works at TEM boundaries to limit their widening. Here, by conducting high-resolution microscopy approaches we unveil the presence of an actomyosin cable encircling TEMs. We develop a theoretical cellular dewetting framework to interpret TEM physical parameters that are quantitatively determined by laser ablation experiments. This establishes the critical role of ezrin and non-muscle myosin II (NMII) in the progressive implementation of line tension. Mechanistically, fluorescence-recovery-after-photobleaching experiments point for the upstream role of ezrin in stabilizing actin filaments at the edges of TEMs, thereby favouring their crosslinking by NMIIa. Collectively, our findings ascribe to ezrin and NMIIa a critical function of enhancing line tension at the cell boundary surrounding the TEMs by promoting the formation of an actomyosin ring., Holes in endothelial barriers, called transendothelial cell macroapertures (TEMs), are predicted to be limited by line tension of unknown origin. Here the authors identify an actomyosin cable encircling TEMs and establish a role for ezrin in stabilising F-actin bundles, allowing their crosslinking by non-muscle myosin IIa.

Abstract

We describe the preparation of 1,4′-bipyrazolyls and 4-pyrazolylpyrimidines by the reaction of 2-pyrazolyl-3-dimethylamino acrylate and acrylonitrile with double nucleophilic reagents such as hydrazines, urea and guanidine. Reactions were performed under microwave irradiation in 5-60 min. This is a useful procedure for the preparation of valuable compounds with applications in medicinal and coordination chemistry. © 2006 Elsevier Ltd. All rights reserved.

6. Cyclopentadithiophene-functionalized Ru(II)-bipyridine sensitizers for dye-sensitized solar cells

Abstract

Two ruthenium sensitizers TT-230 and TT-232 based on cyclopenta[2,1-b:3,4-b']dithiophene (CDT) were tested in dye-sensitized solar cells (DSSCs) using,a tri-iodide/iodide redox couple, and their performances compared to the benchmark Z907. Both dyes of general formula Ru(LL')(NCS)(2) feature the same 4.4'-dicarboxylic acid 2,2'-bipyridine moiety acting as the anchoring group (L) to attach the dye on the TiO2 surface, and two CDT subunits as ancillary groups (L' either directly-linked (11-230) or vinylene-linked (TT-232) to the bipyridine moiety. In comparison with TT-230, the it-extended conjugation in TT-232 caused significant redshift and broadening of the absorption bands. However, despite better light-harvesting properties, the DSSC device sensitized with TT-232 strikingly underperformed compared to TT-230 under same conditions (PCE = 3.2% and 6.1%, respectively). The use of co-adsorbent (CHENO) or surfactants did not help to improve the photovoltaic properties of TT-230, and resulted in a degradation of the overall efficiency of the cell. Abstract

The detailed synthesis and characterization of four ruthenium(II) complexes [RuLL(NCS)(2)] is reported, in which L represents a 2,2-bipyridine ligand functionalized at the 4,4 positions with benzo[1,2-b:4,5-b]dithiophene derivatives (BDT) and L is 2,2-bipyridine-4,4-dicarboxylic acid unit (dcbpy) (NCS=isothiocyanate). The reaction conditions were adapted and optimized for the preparation of these amphiphilic complexes with a strong lipophilic character. The photovoltaic performances of these complexes were tested in TiO2 dye-sensitized solar cell (DSSC) achieving efficiencies in the range of 3-4.5% under simulated one sun illumination (AM1.5G).

8. Adapting Ruthenium Sensitizers to Cobalt Electrolyte Systems

Abstract

In this work, we report the use of bulky substitutions in a new heteroleptic ruthenium(II) bipyridine complex, Ru(NCS)(2)LL', coded TT-230 to obtain high open-circuit potential in a dye-sensitized solar cell (where L is a bipyridine ligand appended with two cyclopenta(2,1-b;3,4-bA)dithiophene moieties, and L' = 4,4,'-dicarboxylic acid 2,2'-bipyridine). The electrolytes based on cobalt complexes have shown significant advantages in terms of attainable open-circuit potential compared to the standard iodide/tri-iodide redox mediators. These merits of the cobalt complexes were previously realized with a porphyrin sensitizer, achieving a V-OC greater than 1 V in DSC. However, with conventional Ru(II)-polypyridyl complexes such as the C101 dye, similar increase in the V-OC could not be attained due to the enhanced recombination. In this work, we have shown that the use of bulky substituents can prevent the back reaction of photogenerated electron and subsequently increase the open-circuit potential of the device. The recombination processes were investigated by transient photovoltage decay measurements.

Abstract

Experimental studies of sediment transport rates due to near shore waves are often conducted in oscillating water tunnels (OWTs). In an OWT, the oscillatory motion produced by the piston propagates almost instantaneously along the entire tunnel. Consequently, unlike the wave motion in the sea or in a wave flume, flow in an OWT is uniform along the tunnel, and second - order wave propagation effects (such as Longuet - Higgins's streaming) are absent. The effect of these hydrodynamic differences between OWT and sea waves on sediment transport rates has generally been neglected. In this paper we present a simple, practical formulation to evaluate bed shear stresses and bed load transport rates due to asymmetric and skewed waves plus a current in an OWT, based on fitting the exact results of a rigorous, analytical model of the OWT wave - current boundary layer. By then accounting for real wave effects we find that wave propagation significantly affects the predicted period - averaged net sediment transport rates.
