1. Efektivitas Pemberian NAA, BAP, 2iP, dan TDZ terhadap Pertumbuhan In Vitro Krisan (Chrysanthemum morifolium)

This study formulate efficient protocol in tissue of culture Chrysanthemum as an early stages for development of Chrysanthemum plant cell engineering. Cell suspension culture experiments were arranged in a completely randomized design (CDR) using two factors. The first factor is type and concentration of auxin, (NAA 0.1 mg L-1; 1.5 mg L-1; 1.0 mg L-1; 1.5 mg L-1; 2.0 mg L-1, 3 mg L-1 and without auxin). The second factor is concentration of BAP (0.5 mg L-1; 1.0 mg L-1; dan 2.0 mg L-1; and without BAP). In plantlet regeneration experiment we used a completely randomized design (CRD) in factorial of two factors. The first factor is type and concentration of NAA and 2iP (each 0 mg L-1; 0.1 mg L-1; 0.5 mg L-1; 1.0 mg L-1). The second factor is concentration of BAP (0 mg L-1; 0.5 mg L-1; 1.0 mg L-1; dan 2.0 mg L-1). Separateexperiment on regeneration was also tested using of TDZ (0 mg L-1; 0.5 mg L-1; 1.0 mg L-1; 1.5 mg L-1; 2.0 mg L-1). Callus used for regeneration was 100 mg. Each combination of treatments in the experiments were repeated 4 times. Analysis of variance show largest callus biomass weight gain after 30 days suspension in combination treatment of NAA 1.0 mg L-1 + BAP 0.5 mg L-1 (1.55 g). Addition of NAA 0.5 mg L-1 and BAP 0.5 mg l-1 on the regeneration medium showed the best response to the growing rate of shoots (30 days), number of shoots (5 shoots) and shoots length (2.88 cm). The addition of 2iP 0.5 mg L-1 and BAP 2.0 mg L-1 in regeneration media gives the best results on the rate of growth of shoot (30 days) and plant height (1.88 cm). Highest number of shoots obtained on treatment of the addition of BAP 0.5 mg L-1 without 2iP (3 shoots). Callus grown on media with TDZ (0.5-2.0 mg L-1) changes color from green to brown. It seems that the addition Thidiazuron on regeneration medium inhibit the formulation of shoot.
