Laterality indices (LIs) quantify the left-right asymmetry of brain and behavioural variables and provide a measure that is statistically convenient and seemingly easy to interpret. Substantial variability in how structural and functional asymmetries are recorded, calculated, and reported, however, suggest little agreement on the conditions required for its valid assessment. The present study aimed for consensus on general aspects in this context of laterality research, and more specifically within a particular method or technique (i.e., dichotic listening, visual half-field technique, performance asymmetries, preference bias reports, electrophysiological recording, functional MRI, structural MRI, and functional transcranial Doppler sonography). Experts in laterality research were invited to participate in an online Delphi survey to evaluate consensus and stimulate discussion. In Round 0, 106 experts generated 453 statements on what they considered good practice in their field of expertise. Statements were organised into a 295-statement survey that the experts then were asked, in Round 1, to independently assess for importance and support, which further reduced the survey to 241 statements that were presented again to the experts in Round 2. Based on the Round 2 input, we present a set of critically reviewed key recommendations to record, assess, and report laterality research for various methods.

Proton-sensing ovarian cancer G-protein coupled receptor (OGR1) plays an important role in pH homeostasis. Acidosis occurs at sites of intestinal inflammation and can induce endoplasmic reticulum (ER) stress and the unfolded protein response (UPR), an evolutionary mechanism that enables cells to cope with stressful conditions. ER stress activates autophagy, and both play important roles in gut homeostasis and contribute to the pathogenesis of inflammatory bowel disease (IBD). Using a human intestinal epithelial cell model, we investigated whether our previously observed protective effects of OGR1 deficiency in experimental colitis are associated with a differential regulation of ER stress, the UPR and autophagy. Caco-2 cells stably overexpressing OGR1 were subjected to an acidic pH shift. pH-dependent OGR1-mediated signalling led to a significant upregulation in the ER stress markers, binding immunoglobulin protein (BiP) and phospho-inositol required 1 alpha (IRE1 alpha), which was reversed by a novel OGR1 inhibitor and a c-Jun N-terminal kinase (JNK) inhibitor. Proton-activated OGR1-mediated signalling failed to induce apoptosis, but triggered accumulation of total microtubule-associated protein 1A/1B-light chain 3, suggesting blockage of late stage autophagy. Our results show novel functions for OGR1 in the regulation of ER stress through the IRE1 alpha -JNK signalling pathway, as well as blockage of autophagosomal degradation. OGR1 inhibition might represent a novel therapeutic approach in IBD.

Gp96 is an endoplasmic reticulum chaperone for multiple protein substrates. Its lack in intestinal macrophages of Crohn's disease (CD) patients is correlated with loss of tolerance against the host gut flora. Gp96 has been stablished to be an essential chaperone for Toll-like receptors (TLRs). We studied the impact of gp96-knockdown on TLR-function in macrophages. TLR2 and TLR4 expression was only decreased but not abolished when gp96 was knocked-down in cell lines, whereas in a monocyte/macrophage specific knock-out mouse model (LysMCre) TLR4 was abolished, while TLR2 was still present. Lipopolysaccharide (LPS)-induced NF-kappa B activation was still observed in the absence of gp96, and gp96-deficient macrophages were able to up-regulate surface TLR4 upon LPS treatment, suggesting that there is another chaperone involved in the folding of TLR4 upon stress responses. Moreover, LPS-dependent pro-inflammatory cytokines were still expressed, although to a lesser extent in the absence of gp96, which reinforces the fact that gp96 is involved in regulating signaling cascades downstream of TLR4 are impaired upon loss of gp96. In addition, we have also found a reduced phosphorylation of ERK and p38 kinases and an impaired response upon CSF1R activation in gp96 deficient macrophages. Our findings indicate that the loss of gp96 not only impairs TLR4 signaling, but is also associated with a diminished phosphorylation of ERK and mitogen-activated stress kinases resulting in an impaired signalling through several receptors, including CSF1R.

Hypoxia regulates autophagy and nucleotide-binding oligomerization domain receptor, pyrin domain containing (NLRP)3, two innate immune mechanisms linked by mutual regulation and associated to IBD. Here we show that hypoxia ameliorates inflammation during the development of colitis by modulating autophagy and mammalian target of rapamycin (mTOR)/NLRP3 pathway. Hypoxia significantly reduces tumor necrosis factor a, interleukin (IL)-6 and NLRP3 expression, and increases the turnover of the autophagy protein p62 in colon biopsies of Crohn's disease patients, and in samples from dextran sulfate sodium-treated mice and Il-10(-/-) mice. In vitro, NF-.kappa signaling and NLRP3 expression are reduced through hypoxia-induced autophagy. We also identify NLRP3 as a novel binding partner of mTOR. Dimethyloxalylglycine-mediated hydroxylase inhibition ameliorates colitis in mice, downregulates NLRP3 and promotes autophagy. We suggest that hypoxia counteracts inflammation through the downregulation of the binding of mTOR and NLRP3 and activation of autophagy.

Background and Aims: Transketolase-like (TKTL) 1 is one of the key enzymes for anaerobic sugar degradation even in the presence of oxygen (aerobic glycolysis). Transketolase-dependent reactions supply malignant tumors with ribose and NADPH. Therefore, TKTL1 activity could be crucial for tumor proliferation and survival. The aim of the study was to evaluate the expression of TKTL1 in colorectal cancer (CRC) and its regulation under hypoxic conditions. Methods: We studied TKTL1 mRNA and protein expression in CRC cell lines and human CRC biopsies by quantitative real-time PCR, Western blotting and immunohistochemistry. Regulation of TKTL1 under oxygen depletion was analyzed by cultivating cells either in a three-dimensional spheroid model or in a hypoxia incubator chamber. Results: TKTL1 mRNA was heterogeneously expressed in monolayers of cells with high levels in HT-29 and SW480. TKTL1 protein was also clearly detectable in HT-29 and SW480. Hypoxia-inducible factor (HIF)-1α protein expression correlated with TKTL1 protein expression in SW480 spheroids over time. On the one hand, induction of hypoxia in T84 spheroids did not induce TKTL1; on the other hand, hypoxia by incubation at 1% O2 in a hypoxia incubator chamber clearly showed an upregulation of TKTL1. In 50% of CRC patients, TKTL1 protein expression was upregulated in tumor compared to non-tumor tissue. The immunohistochemical staining of TKTL1 in CRC patient samples resulted in 14 positive and 30 negative samples. Conclusions: TKTL1 expression correlated with HIF-1α protein expression and was induced upon hypoxic conditions which could facilitate energy supply to tumors under these circumstances.

Biological sex and sex hormones are known to affect functional cerebral asymmetries (FCAs). Men are generally more lateralized than women. The effect size of this sex difference is small but robust. Some of the inconsistencies in the literature may be explained by sex-related hormonal differences. Most studies focusing on neuromodulatory properties of sex hormones on FCAs have investigated women during the menstrual cycle. Although contradictions exist, these studies have typically shown that levels of estradiol and/or progesterone correlate with the degree of FCAs, suggesting that sex differences in FCAs partially depend on hormonal state and day of testing. The results indicate that FCAs are not fixed but are hormone dependent, and as such they can dynamically change within relatively short periods throughout life. Many issues raised in this Mini-Review refer not only to FCAs but also to other aspects of functional brain organization, such as functional connectivity within and between the cerebral hemispheres. Our understanding of sex differences in brain and behavior as well as their clinical relevance will improve significantly if more studies routinely take sex and sex hormones into account.

Apoptosis controls the adaptive immune system through regulation of central and peripheral lymphocyte deletion. Therefore, substances that selectively interact with the intrinsic apoptosis pathway in lymphocytes offer unexplored opportunities to pharmacologically modulate the immune response. Here, we present evidence that the BH3-mimetic ABT-737 suppresses allogeneic immune responses. In vitro, ABT-737 prevented allogeneic T-cell activation, proliferation, and cytotoxicity by apoptosis induction, but without impairing the physiological functions of remaining viable T cells. In vivo, ABT-737 was highly selective for lymphoid cells and inhibited allogeneic T- and B-cell responses after skin transplantation. The immunosuppressive effect of ABT-737 was markedly increased in combination with low-dose cyclosporine A, as shown by the induction of long-term skin graft survival without significant inflammatory infiltrates in 50% of the recipients in an MHC class I single antigen mismatched model. Thus, pharmacological targeting of Bcl-2 proteins represents a novel immunosuppressive approach to prevent rejection of solid organ allografts.

Schottky Mass Spectrometry (SMS) and Isochronous Mass Spectrometry (IMS) have been successfully applied for direct mass measurements of exotic nuclides at FRS-ESR facility at GSI. Both experimental methods are sensitive to single stored ions and can cover very efficiently a large number of nuclides in one run. Presently, more than 1100 masses of different nuclides have been covered by SMS and IMS in several FRS-ESR experiments whereby about 350 of them have been determined for the first time. In this paper, we present the status of our experimental program and use the experimental mass values to test the predictive power of modern mass models.

GSI Scientific Report 2014 - GSI Report 2015-1

A $^{238}$U projectile beam was used to create cadmium isotopes via abrasion-fission at 410 MeV/u in a beryllium target at the entrance of the in-flight separator FRS at GSI. The fission fragments were separated with the FRS and injected into the isochronous storage ring ESR for mass measurements. The Isochronous Mass Spectrometry (IMS) was performed under two different experimental conditions, with and without B$\rho$-tagging at the dispersive central focal plane of the FRS. In the experiment with B$\rho$-tagging the magnetic rigidity of the injected fragments was determined by an accuracy of $2\times 10^{-4}$. A new method of data analysis, using a correlation matrix for the combined data set from both experiments, has provided mass values for 25 different isotopes for the first time. The high selectivity and sensitivity of the experiment and analysis has given access even to rare isotopes detected with a few atoms per week. In this letter we present for the $^{129,130,131}$Cd isotopes mass values directly measured for the first time. The Cd results clearly show a very pronounced shell effect at $N=82$ which is in agreement with the conclusion from $\gamma$-ray spectroscopy of $^{130}$Cd and confirms the assumptions of modern shell-model calculations., Comment: 7 pages, 5 figures, 3 tables

Granulomatose Hautveranderungen nach einer Herpes-zoster-Erkrankung sind selten und ihre Pathogenese ist unklar. Anhand eines immunsupprimierten Patienten mit postherpetischen Granulomen und Nachweis von Varizella-zoster-Virus-DNA im Granulomgewebe werden neuere pathogenetische Vorstellungen zur Entstehung dieser Hautveranderungen besprochen.

It has been shown for several DNA probes that the recently introduced Fast-FISH (fluorescence in situ hybridization) technique is well suited for quantitative microscopy. For highly repetitive DNA probes the hybridization (renaturation) time and the number of subsequent washing steps were reduced considerably by omitting denaturing chemical agents (e.g., formamide). The appropriate hybridization temperature and time allow a clear discrimination between major and minor binding sites by quantitative fluorescence microscopy. The well-defined physical conditions for hybridization permit automatization of the procedure, e.g., by a programmable thermal cycler. Here, we present optimized conditions for a commercially available X-specific a-satellite probe. Highly fluorescent major binding sites were obtained for 74oC hybridization temperature and 60 min hybridization time. They were clearly discriminated from some low fluorescent minor binding sites on metaphase chromosomes as well as in interphase cell nuclei. On average, a total of 3.43 ± 1.59 binding sites were measured in metaphase spreads, and 2.69 ± 1.00 in interphase nuclei. Microwave activation for denaturation and hybridization was tested to accelerate the procedure. The slides with the target material and the hybridization buffer were placed in a standard microwave oven. After denaturation for 20 s at 900 W, hybridization was performed for 4 min at 90 W. The suitability of a microwave oven for Fast-FISH was confirmed by the application to a chromosome 1-specific a-satellite probe. In this case, denaturation was performed at 630 W for 60 s and hybridization at 90 W for 5 min. In all cases, the results were analyzed quantitatively and compared to the results obtained by Fast-FISH. The major binding sites were clearly discriminated by their brightness

Research on oxytocin (OT) indicates that it has stress reducing effects. This leads to opposing predictions of decreased and increased aggression which we examine in this study. Following completion of a state anxiety measure and administration of OT or a placebo, female participants took part in a competitive aggression game (PSAP) for a monetary prize which, if won, would be paid to a loved one. In the game, three options were available: participants could earn points; attack their opponent by deducting points; and defend themselves against point deduction by their opponent. There was no main effect of OT on these responses, however there was an interaction with state anxiety. In the placebo condition, women higher in state anxiety showed a significantly higher ratio of Attack-to-Earn responses than low anxiety women. Under oxytocin, there was a significant reduction in their Attack:Earn ratio resulting in no significant difference between high and low state anxiety groups. There was a similar trend for the Defend:Earn ratio. The reduction of reactive aggression in state anxious women supports the view that OT may decrease negative behavior and increase constructive behavior even under conditions of provocation.

Production cross sections for neutron-rich nuclei from the fragmentation of a 82Se beam at 139 MeV/u were measured. The longitudinal momentum distributions of 126 neutron-rich isotopes of elements 11, 13 pages, 11 figures, accepted to Phys.Rev.C

Beta-delayed proton emission from nuclides in the neighborhood of 100Sn was studied at the National Superconducting Cyclotron Laboratory. The nuclei were produced by fragmentation of a 120 MeV/nucleon 112Sn primary beam on a Be target. Beam purification was provided by the A1900 Fragment Separator and the Radio Frequency Fragment Separator. The fragments of interest were identified and their decay was studied with the NSCL Beta Counting System (BCS) in conjunction with the Segmented Germanium Array (SeGA). The nuclei 96Cd, 98Ing, 98Inm and 99In were identified as beta-delayed proton emitters, with branching ratios bp = 5.5(40)%, 5.5+3 -2%, 19(2)% and 0.9(4)%, respectively. The bp for 89Ru, 91,92Rh, 93Pd and 95Ag were deduced for the first time with bp = 3+1.9 -1.7%, 1.3(5)%, 1.9(1)%, 7.5(5)% and 2.5(3)%, respectively. The bp = 22(1)% for 101Sn was deduced with higher precision than previously reported. The project Services@MediGRID consortium established a tool set of grid-based biomedical services since 2008. The services are related to genetic analysis, genome data visualization, and pharmacokinetic modeling. Furthermore, business concepts for these services have been examined which are supported by an accounting and billing service. While the tools cover a whole service chain for biomedicine, the business concepts are rather heterogeneous. However, the overall addressed target market areas show promising potential. In addition, a structured coaching process reduces friction in the technology transfer from grid computing to biomedicine. This should be considered for similar future endeavors.

We propose to install a storage ring at an ISOL-type radioactive beam facility for the first time. Specifically, we intend to setup the heavy-ion, low-energy ring TSR at the HIE-ISOLDE facility in CERN, Geneva. Such a facility will provide a capability for experiments with stored secondary beams that is unique in the world. The envisaged physics programme is rich and varied, spanning from investigations of nuclear ground-state properties and reaction studies of astrophysical relevance, to investigations with highly-charged ions and pure isomeric beams. The TSR might also be employed for removal of isobaric contaminants from stored ion beams and for systematic studies within the neutrino beam programme. In addition to experiments performed using beams recirculating within the ring, cooled beams can also be extracted and exploited by external spectrometers for high-precision measurements. The existing TSR, which is presently in operation at the Max-Planck Institute for Nuclear Physics in Heidelberg, is well-suited and can be employed for this purpose. The physics cases as well as technical details of the existing ring facility and of the beam and infrastructure requirements at HIE-ISOLDE are discussed in the present technical design report.

The isomeric and {\beta} decays of the N = Z +2 nucleus 96Ag were investigated at NSCL. A cascade of {\gamma}-ray transitions originating from the de-excitation of a {\mu}s isomer was observed for the first time and was found in coincidence with two previously-known transitions with energies of 470 and 667 keV. The isomeric half-life was determined as 1.45(7) {\mu}s, more precise than previously reported. The existence of a second, longer-lived {\mu}s isomer, associated with a 743-keV transition, is also proposed here. Shell model results within the (p_{3/2}p_{1/2}f_{5/2}g_{9/2}) model space, using the jj44b interaction, reproduced level energies and isomeric decay half-lives reasonably well.

High-spin states in 73Kr were studied at the XTU tandem accelerator of the Laboratori Nazionali di Legnaro using the reaction 40Ca(40 Ca,2pn) at a beam energy of 185 MeV. Gamma rays were detected with the EUROBALL spectrometer. Particle detection enabled us to identify the reaction channel leading to 73Kr. The yrast bands of positive and negative parity were established up to probable spins of 61/2+ and 63/2- , respectively. The energies of the two identified bands agree well with the predictions of the configuration-dependent Cranked Nilsson-Strutinsky approximation and indicate that the nucleus remains collective up to the maximum spins of the respective high-spin configurations. The positive-parity band represents one of the rare cases where band non-termination at the highest spin of the configuration could be established experimentally.

Graft-versus-Host Disease (GvHD) is the most serious complication of allogeneic stem cell transplantation (SCT) and results from an activation of donor lymphocytes by recipient antigen-presenting cells (APCs). For a long time, it has been postulated that the intestinal microflora and endotoxin exert a crucial step in this APC activation, as there is early and severe gastrointestinal damage induced by pretransplant conditioning. With the detailed description of pathogen-associated molecular patterns and pathogen recognition receptors single nucleotide polymorphisms of TLRs and especially NOD2 have been identified as potential risk factors of GvHD and transplant related complications thus further supporting the crucial role of innate immunity in SCT, related complications. Gastrointestinal decontamination and neutralization of endotoxin have been used to interfere with this early axis of activation with some success but more specific approaches of modulation of innate immunity are needed for further improvement of clinical outcome.

The enhanced cross sections of several new nuclei relative to a simple thermal evaporation framework, previously shown to describe similar production cross sections, indicates that nuclei in the region around 62-Ti might be more stable than predicted by current mass models and could be an indication of a new island of inversion similar to that centered on 31-Na., Comment: 4 pages, 3 figures, to be published in Physical Review Letters, 2009

Production cross sections for neutron-rich nuclei from the fragmentation of a 76Ge beam at 132 MeV/u were measured. The longitudinal momentum distributions of 34 neutron-rich isotopes of elements 13, accepted for publication in Phys.Rev.C

High-spin states in 83Rb were populated in the reaction 11B + 76Ge at beam energies of 45 and 50 MeV. Gamma rays were detected with the spectrometer GASP. The level scheme of 83Rb was extended up to 13.9 MeV. Mean lifetimes of 23 levels were determined using the Doppler-shift-attenuation method. Among the bands newly established is a sequence including intense M1 transitions and cross-over E2 transitions. This sequence turns out to be irregular and thus shows that magnetic rotation observed in the neighboring odd-odd isotopes cannot be sustained in this odd-even nuclide. Excited states in 83Rb were interpreted in terms of the shell model using the model space pi (0f5/2, 1p3/2, 1p1/2, 0g9/2) nu (1p1/2, 0g9/2). The configuration predicted for the M1 sequence is analogous to the one predicted for the M1 band in the neighbor 84Rb and reproduces the transition strengths at high spin.

It is argued that orbital electron-capture decays of neutral $^{142}$Pm atoms implanted into the lattice of a solid (LBNL experiment) do not fulfil the constraints of true two-body beta decays, since momentum as well as energy of the final state are distributed among three objects, namely the electron neutrino, the recoiling daughter atom and the lattice phonons. To our understanding, this could be a reason for the non-observation of a periodic time modulation in the number of electron-capture decays of implanted neutral $^{142}$Pm atoms., 2 p. comment on arXiv:0807.0649v1

The results of measurements of the production of neutron-rich nuclei by the fragmentation of a 48Ca beam at 142 MeV/u are presented. Evidence was found for the production of a new isotope that is the most neutron-rich silicon nuclide, 44Si, in a net neutron pick-up process. A simple systematic framework was found to describe the production cross sections based on thermal evaporation from excited prefragments that allows extrapolation to other weak reaction products., 12 pages, 4 figures, 1 table; corrected figures 1, 3 & 4, corrected typos, updated bibliography

Transition rate measurements are reported for the first and the second 2+ states in N=Z 64Ge. The experimental results are in excellent agreement with large-scale Shell Model calculations applying the recently developed GXPF1A interactions. Theoretical analysis suggests that 64Ge is a collective gamma-soft anharmonic vibrator. The measurement was done using the Recoil Distance Method (RDM) and a unique combination of state-of-the-art instruments at the National Superconducting Cyclotron Laboratory (NSCL). States of interest were populated via an intermediate-energy single-neutron knock-out reaction. RDM studies of knock-out and fragmentation reaction products hold the promise of reaching far from stability and providing lifetime information for excited states in a wide range of nuclei.

Objectives:\ud \ud HLA-B27 is a common genetic risk factor for the development of Spondyloarthritides (SpA). HLA-B27 can misfold to form cell-surface heavy chain homodimers (B272) and induce pro-inflammatory responses that may lead to SpA pathogenesis. The presence of B272 can be detected on leukocytes of HLA-B27+ Ankylosing spondylitis (AS) patients and HLA-B27 transgenic rats. We characterized a novel B272–specific monoclonal antibody to study its therapeutic use in HLA-B27 associated disorders.\ud \ud Methods:\ud \ud The monoclonal HD5 antibody was selected from a phage library to target cell-surface B272 homodimers and characterized for affinity, specificity and ligand binding. The immune modulating effect of HD5 was tested in HLA-B27 transgenic rats. Onset and progression of disease profiles were monitored during therapy. Cell-surface B272 and expansion of pro-inflammatory cells from blood, spleen and draining lymph nodes were assessed by flow cytometry.\ud \ud Results:\ud \ud HD5 bound B272 with high specificity and affinity (Kd = 0.32 nM). HD5 blocked cell-surface interaction of B272 with immune regulatory receptors KIR3DL2, LILRB2 and Pirb. In addition, HD5 modulated the production of TNF from CD4+ T-cells by limiting B272 interactions in vitro. In an HLA-B27 transgenic rat model repetitive dosing of HD5 reduced the expansion of pro-inflammatory CD4+ T-cells, and decreased the levels of soluble TNF and number of cell-surface B272 molecules.\ud \ud Conclusion:\ud \ud HD5 predominantly inhibits early TNF production and expansion of pro-inflammatory CD4+ T-cells in HLA-B27 transgenic rats. Monoclonal antibodies targeting cell-surface B272 propose a new concept for the modulation of inflammatory responses in HLA-B27 related disorders.

About 50 new states with excitation energies up to 7 MeV and angular momentum I ≤ 18(+) were observed and for many of them, spin and parity could be firmly assigned. The nucleus 70Se was studied using the 40Ca(40Ca, 2α) reaction at a beam energy of 185 MeV. Gamma rays were measured with the EUROBALL III spectrometer. The known positive-parity bands have been extended and one new band of positive parity and two of negative parity have been identified. These bands are interpreted in terms of the cranked Nilsson-Strutinsky approach. Calculations suggest that the two negative-parity bands, which have the same signature, are both based on a configuration with two protons and three neutrons lifted from the fp shell to the g9/2 orbital, but at different nuclear shapes. This represents a shape coexistence at high spin.

High-spin states in 82Rb and 84Rb were populated in the reaction 11B + 76Ge at beam energies of 45 and 50 MeV. Gamma rays were detected with the spectrometer GASP. The level schemes of 82Rb and 84Rb were extended up to 6.0 and 7.4 MeV, respectively. Mean lifetimes of five levels in 82Rb and eleven levels in 84Rb were determined using the Doppler-shift-attenuation method. Regular magnetic dipole bands including strong M1 and weak E2 transitions observed in both nuclei show the characteristic features of magnetic rotation. These bands have been successfully described in the tilted-axis cranking model on the basis of the four-quasiparticle configuration pi(fp) pi(g_{9/2}^2) \nu(g_{9/2}). The calculations reproduce the band-like properties as well as absolute B(M1) and B(E2) transition strengths in both nuclei, which supports the concept of magnetic rotation. Excited states in 84Rb were also interpreted in terms of the shell model using the model space pi(0f_{5/2},1p_{3/2},1p_{1/2},0g_{9/2}) nu(1p_{1/2},0g_{9/2}). These calculations reproduce the low-spin states. Moreover, calculated states with the main configuration pi(0f_{5/2}^{-2}1p_{3/2}^{-1} 0g_{9/2}^2) nu(0g_{9/2}^{-3}) can be combined to M1 sequences that reproduce roughly the experimental transition strengths, but do not describe the features of magnetic rotation, as regular level spacings and B(M1) values decreasing with increasing rotational frequency.
