Granulated bentonite mixtures (GBMs) have been regarded as effective buffer materials in the deep geological disposal of radioactive waste due to their operational advantages, such as ease of transportation and in-situ placement/backfilling. Many studies have been done to characterize the hydraulic and thermal properties of GBMs as well as their swelling properties. Only limited studies, however, have investigated their gas transport properties, even though these properties affect their compactness during in-situ placement/backfilling and subsequent gas diffusion and advection in the buffer zone. The aim of this study is to understand the gas transport parameters, i.e., air permeability (ka) and gas diffusivity (Dp/Do), of tested samples compacted at different dry densities (DDs) under air-dried conditions, linking them with the measured density distribution characteristics determined by microfocus X-ray computed tomography (MFXCT) analysis. Two types of GBMs were used in this study: 1) FE-GBM (prepared from National Standard® bentonite, Wyoming, USA): this material was used in the Full-scale Emplacement (FE) experiment at the Mont Terri rock laboratory, Switzerland) and 2) OK-GBM (prepared from a bentonite, originating from Japan, with the trade name of OK bentonite, Kunimine Industries). The tested samples were firstly packed in a 100-cm3 acrylic core with different DDs, ranging from loose to dense (1.09 to 1.75 g/cm3), and scanned by MFXCT. The weighting factors, wf (fine fraction; lower density) and wc (coarse fraction; higher density) (wf + wc = 1), were determined after the peak separation of the measured CT brightness histograms from the reconstructed three-dimensional multiplanar reconstruction (MPR) images of the MFXCT analysis. The measured ka and Dp/Do were highly dependent on the DDs, the ka (ɛ) values fitted well with a power law model, and the Dp/Do (ɛ) was predicted accurately by several previously proposed models. For both FE-GBM and OK-GBM, there were good linear relationships between the gas transport parameters and wc × DD, implying that the weight of the coarse fraction controlled ka and Dp/Do. Moreover, the Kozeny-Carman model, incorporating the measured volumetric surfaces from the MFXCT analysis, was able to predict the ka values well.

International audience; Groundwater is essential for the Earth biosphere but is often contaminated by harmful chemical compounds due to both anthropogenic and natural causes. A key factor controlling the fate of harmful chemicals in groundwater is the reduction/oxidation (redox) conditions. The formation factors for the groundwater redox conditions are insufficiently understood. In this study, long-term groundwater quality beneath one of the world megacities was monitored and evaluated. We measured and compared hydrogeochemical conditions including groundwater quality (35 chemical parameters) and redox conditions of five aquifers in the Arakawa Lowland and Musashino Upland, southern Kanto Plain of the Tokyo Metropolitan area, Japan. Monitoring results suggested the following: The main origin of groundwater is precipitation in both the Lowland and Upland areas. The three aquifers in the Arakawa Lowland are likely fully separated, with one unconfined and two confined aquifers under iron reducing and methanogenic conditions, respectively. Oppositely, in the Musashino Upland, the water masses in the two aquifers are likely partly connected, under aerobic conditions, and undergoing the same groundwater recharge and flow processes under similar hydrogeological conditions. The different groundwater redox conditions observed are likely caused by the very different groundwater residence times for the Arakawa Lowland and Musashino Upland.

Understanding gas transport in porous media and its mechanism has broad applications in various research areas, such as carbon sequestration in deep saline aquifers and gas explorations in reservoir rocks. Gas transport is mainly controlled by pore space geometrical and morphological characteristics. In this study, we apply a physically-based model developed using concepts from percolation theory (PT) and the effective-medium approximation (EMA) to better understand diffusion and permeability of gas in packings of angular and rounded sand grains as well as glass beads. Two average sizes of grain i.e., 0.3 and 0.5 mm were used to pack sands in a column of 6 cm height and 4.9 cm diameter so that the total porosity of all packs was near 0.4. Water content, gas-filled porosity (also known as gas content), gas diffusion, and gas permeability were measured at different capillary pressures. The X-ray computed tomography method and the 3DMA-Rock software package were applied to determine the average pore coordination number z. Results showed that both saturation-dependent diffusion and permeability of gas showed almost linear behavior at higher gas-filled porosities, while deviated substantially from linear scaling at lower gas saturations. Comparing the theory with the diffusion and permeability experiments showed that the determined value of z ranged between 2.8 and 5.3, not greatly different from X-ray computed tomography results. The obtained results clearly indicate that the effect of the pore-throat size distribution on gas diffusion and permeability was minimal in these sand and glass bead packs.

A capillary barrier cover system (CBCS) is a low-cost solution for limiting water infiltration into solid waste landfills. Moreover, hydrophobized sublayer sand grains enhance the impermeable properties of the CBCS. In this study, we have assessed water repellency characteristics for oleic acid (OA) mixed sands representing four different particle size fractions ranging from 0.105 to 0.84 mm. Initial soil-water contact angle (αi), and the time dependence of contact angle were measured by the sessile drop method. Results showed that the αi value for fine and middle sand fractions increased rapidly from 0o to ~100o with increasing OA content from 0 to around 3.0 g OA kg-1 sand. The hydrophobized fine and middle sand fractions are suitable for sublayer in the CBCS. A recently proposed equation, α (t) = αi exp (-At), where t is time and A is the coefficient of coefficient of temporal change in contact angle (α), well captured the time dependence of α.

Modelling water distribution and flow in partially saturated soils requires knowledge of the soil-water characteristic (SWC). However, measurement of the SWC is challenging and time-consuming, and in some cases not feasible. This study introduces two predictive models (Xw-model and Xw*-model) for the SWC, derived from readily available soil properties such as texture and bulk density. A total of 46 soils from different horizons at 15 locations across Denmark were used for models evaluation. The Xw-model predicts the volumetric water content as a function of volumetric fines content (organic matter and clay). It performed reasonably well for the dry-end (above a pF value of 2.0; pF = log(|Ψ|), where Ψ is the matric potential in cm), but did not do as well closer to saturated conditions. The Xw*-model gives the volumetric water content as a function of volumetric content of particle size fractions (organic matter, clay, silt, fine and coarse sand), variably included in the model depending on the pF value. The volumetric content of a particular soil particle size fraction was included in the model if it was assumed to contribute to the pore size fraction still occupied with water at the given pF value. Hereby, the Xw*-model implicitly assumes that a given particle size fraction creates an analogue pore size fraction and, also, is based on the validity of the well-known capillary law equation relating equivalent drained pore size to the soil-water matric potential. The Xw*-model was found to be quite robust, and it performed exceptionally well for all tested pF values ranging from 0.4 to 4.2. For prediction of the continuous SWC, it is recommended that the van Genuchten model is parameterized based on the SWC data points predicted by the Xw*-model.

Secreted frizzled-related protein-5 (sFRP-5) has been identified as one of the secreted antagonists that bind Wnt protein. However, the functional significance of sFRP-5 in renal cell cancer (RCC) has not been reported. We hypothesized that sFRP-5 may be epigenetically downregulated through DNA methylation and histone modification and function as a tumor suppressor gene in RCC. Using tissue microarray and real-time RT-PCR, we found that sFRP-5 was significantly downregulated in kidney cancer tissues and cell lines, respectively. DNA bisulfite sequencing of the sFRP-5 promoter region in RCC cell lines showed it to be densely methylated whereas there was few promoter methylation in normal kidney. The sFRP-5 expression was restored and the acetylation of H3 and H4 histones associated with the sFRP-5 promoter region were significantly increased after treatment with demethylation agent (5-Aza-dc) and histone deacetylase inhibitor (TSA). When RCC cells were transfected with the sFRP-5 gene, significant inhibition of anchorage independent colony formation and cell invasion were observed compared to controls. The sFRP-5 transfection also significantly induced apoptosis in RCC cells. In conclusion, this is the first report documenting that the sFRP-5 is downregulated by promoter methylation and histone acetylation and functions as a tumor suppressor gene by inducing apoptosis in RCC cells.
