SUMMARYCCR4-NOT is a versatile eukaryotic protein complex that controls multiple steps in gene expression regulation from synthesis to decay. In yeast, CCR4-NOT has been implicated in stress response regulation, though this function in other organisms remains unclear. In a genome-wide RNAi screen, we identified a subunit of the CCR4-NOT complex, ccf-1, as a requirement for the C. elegans transcriptional response to cadmium and acrylamide stress. Using whole-transcriptome RNA sequencing, we show that knockdown of ccf-1 attenuates the activation of a broad range of stress protective genes in response to cadmium and acrylamide, including those encoding heat shock proteins and glutathione s-transferases. Consistently, survival assays show that knockdown of ccf-1 decreases C. elegans stress resistance. A yeast-2-hybrid screen using a CCF-1 bait identified the homeobox transcription factor PAL-1 as a physical interactor. Knockdown of pal-1 inhibits the activation of ccf-1 dependent stress genes and reduces C. elegans stress resistance. Gene expression analysis reveals that knockdown of pal-1 down-regulates the mRNA levels of elt-2 and elt-3, which serves as the master transcriptional co-regulators of stress response in the C. elegans intestinal and epidermal tissues respectively. These results reveal a new role for CCR4-NOT in stress response regulation with PAL-1 through the transcriptional control of elt-2 and elt-3 in C. elegans.

The ability to measure and predict atmospheric turbulence affecting laser beam propagation is a major concern when considering military applications. Such a method using a telescope, high resolution television camera, video tape recorder, digital storage oscilloscope, and calculator system has been devised, tested and utilized. A laser beam signal is recorded on video tape for further processing. This signal is displayed, stored and digitized using a Tektronix 468 digital storage oscilloscope. The digitized signal is sent to a Hewlett-Packard 9825 computing system for Fourier transform analysis and„determination of the refractive index structure constant, C . Several trials were conducted using He-Ne and Ga-As lasers. A model for calculating the index of refraction of atomic species present in the cavity of a Xenon Fluoride Laser is applied to Neon and Neon*(3s) . The model considers the variation from unity to be a function of the absorption cross section. Below the ionization threshold, the cross section is a set of discrete transitions between the various energy levels of the specie of concern. Above the 5p level, the discrete transitions become indistinct and the continuum cross section function is extended into this region. The continuum cross section function is a polynomial function fitted to independent research results. The results for Neon I agree with previous theoretical and experimental results. The model is extended to Neon*(3s). Comparison with the results of independent research of the polarizability and cavity phase shift derived from the calculation indicate accuracy is within ten percent. An experiment was conducted to determine the effective excitation cross sections for forming the C 3 n 2nd positive state of N ? in the lowest vibrational energy level by bombarding gaseous" nitrogen with electrons in the energy range 80 to 900 eV. Approximately 900 unknown atomic energy levels were predicted by extrapolation along the helium through sodium isoelectronic sequences. The extrapolations, based on well known regularities in atomic spectra, extend beyond the range of known values providing predictions in highly ionized atoms. The predicted energy levels are presented, along with the known values, in tabular form. In addition, as an aid to spectroscopists, 116 transitions are listed with known and predicted wavelengths. An analysis is made of the 8115A light emitted from the striated positive column of an Argon glow discharge to determine whether or not it is polarized. The procedure consists of separating the perpendicular and parallel components of vibration with a Wallaston prism, measuring the intensity of each component with a photomultiplier tube and comparing the signal intensities on a dual trace oscilloscope. Within the limitations placed upon the measurements by photomultiplier noise and system frequency response, no polarization is observed. The maximum polarization that could escape detection is 3%.
