The ability to track the structural condition of existing structures is one of the main concerns of bridge owners and operators. In the context of bridge maintenance programs, visual inspection predominates nowadays as the primary source of information. Yet, visual inspections alone are insufficient to satisfy the current needs for safety assessment. From this perspective, extensive research on structural health monitoring has been developed in recent decades. However, the transfer rate from laboratory experiments to real-case applications is still unsatisfactory. This paper addresses the main limitations that slow the deployment and the acceptance of real-size structural health monitoring systems (SHM) and presents a novel real-time analysis algorithm based on random variable correlation for condition monitoring. The proposed algorithm was designed to respond automatically to detect unexpected events, such as local structural failure, within a multitude of random dynamic loads. The results are part of a project on SHM, where a high sensor-count monitoring system based on long-gauge fiber Bragg grating sensors (LGFBG) was installed on a prestressed concrete bridge in Neckarsulm, Germany. The authors also present the data management system developed to handle a large amount of data, and demonstrate the results from one of the implemented post-processing methods, the principal component analysis (PCA). The results showed that the deployed SHM system successfully translates the massive raw data into meaningful information. The proposed real-time analysis algorithm delivers a reliable notification system that allows bridge managers to track unexpected events as a basis for decision-making.

Das Institut für den Schutz maritimer Infrastrukturen des Deutschen Zentrums für Luft- und Raumfahrt e.V. in Bremerhaven befasst sich mit der Entwicklung von automatisierten Technologien und echtzeitnahen Verarbeitungsmethoden zur Erstellung neuartiger Lagebilder für den maritimen Bereich. Über- und Unterwasser-Drohnen (UAV, AUV, ROV) werden mit neuartigen optischen Kamera- und Sonarsystemen eingesetzt, um dreidimensionale Lagebilddaten aufzunehmen. Spektrale Informationen und dreidimensionale Punktwolken werden zu Lagedarstellungen kombiniert, die die Lösung von Sicherheitsfragestellungen vereinfachen und beschleunigen (Suche von Lecks, Schäden an Anlagen, Erfassung von Fahrzeugen und Personen auf dem Hafengelände). Die Technologie leistet einen wichtigen Beitrag zum Überblick über die maritimen Infrastrukturen. Durch die gute Verfügbarkeit und die Leistungsfähigkeit der Systeme sowie Möglichkeiten zum automatisierten Betrieb lässt sich der zeitliche Aufwand der Modellierung auf ein Minimum reduzieren.

The determination of the moisture content in building materials is a contro versially discussed subject. The only scientifically accepted reference is the gravimetric method, i.e. the moisture assessment of a sample of building material by drying. Yet, even here, disagreements arise from the question of the “correct” drying temperature, which results from the multiple definitions in use for the dry material state. The significance of moisture for a virtually inconceivable number of matters in civil engineering reflects in the nonetheless numerous approaches for its measurement. Some of them are successfully established in practice, despite of the debates on their accuracy and reproducibility. The reason lies in the urgent need for suitable moisture measurement methods. Their use is not totally condemnable, but requires a profound understanding of the underlying physical concepts and the knowledge of each method’s application, possibilities and limitations. The measurement of a material’s electrical impedance for the determination of its moisture content are often viewed especially critical. This objection is justified primarily by the strong influence of salts and other conductive inclusions, such as clay or metals. However, the method has some unique features, which, if applied right, can give valuable insight into processes within the considered material or investigated structure. In contrast to many other methods, it is possible to realize an instrumented monitoring of an object outside of laboratory conditions. The measurement location remains fixed wherever chosen and therefore allows the comprehension of gradual processes, such as drying or accumulation of salts, while offering the possibility to continuously take the ever-changing ambient conditions into account. The focus of this research work lies on putting impedance spectroscopy into practical application. The method itself has long been studied, but only the latest developments of fully integrated impedance converters have made it possible to incorporate impedance functionality into structural health monitoring systems. Their scope regarding accuracy or frequency range is certainly reduced compared to laboratory devices. However, this evolution provides the opportunity for a permanent instrumentation of structures outside of laboratory conditions to assess moisture developments therein. Special attention was set in the research to the development and optimization of suitable impedance electrodes, which show both sufficient durability for instrumented monitoring and minimize the influence of the electric double layer. It was found that nickel-graphite silicon electrodes are well suited. To be capable of understanding impedance data, which is influenced under natural conditions by many factors that do not or only in other ways occur in a protected laboratory environment, it is necessary to study these effects on the impedance. The two main sources of material moisture are the humidity from the air and capillary water. The present thesis regards impedance measurements for qualitative moisture monitoring of porous building materials, particularly sandstone. Long-term measurements were carried out to research the gradually adapting material moisture of different sandstones caused by a change in the ambient relative humidity between 54 and 100 %. The considered sandstone varieties were selected from southern Germany. In detail Abbacher green sandstone, Burgpreppacher sandstone, Main sandstone white-grey, Sander reed sandstone and Trebgaster new red sandstone. Further measurements were performed to study the detectability of water fronts during the capillary rise of water and the subsequent drying process, as well as the influence of temperature changes. To begin with, the scientific motivation for the present work is presented in chapter 1. Chapter 2 introduces the basic concepts of porosity, water storage and transport therein, and the effects of soluble salts in this water. Chapter 3 gives an overview of electrical impedance monitoring with special respect to measurements on porous materials. Possibilities for the temperature compensation of impedance data are presented. The developed device for impedance monitoring is described in chapter 4. Its conception in the global context of other moisture measurement options and with regard to the specific field of monitoring of protected historic structures is examined. The optimization of the impedance electrodes and the choice of a suitable measurement frequency are discussed. Chapter 5 presents the impedance monitoring of sandstone in the hygroscopic and over-hygroscopic moisture range. A look at the time-dependent data collected for different sandstone types during a long-term experiment with stepwise variation of the ambient relative humidity is followed by an analysis of the impedance regarding material moisture and its distribution within the porous network. The measurement of capillary rise of water and a reflection on possible electrode configurations along with their consequences for the detection of capillary water fronts are regarded thereafter. The chapter closes with a description of the procedure for temperature compensation of impedance data. An example of applied impedance monitoring is presented in the concluding chapter 6, which regards both the hygroscopic and over-hygroscopic moisture range. Two more application examples follow, which show a perspective for other applications. The first pictures the monitoring of hydrating screed, the second illustrates the use of impedance monitoring for the assessment of self-healing concrete., Die Bestimmung der Feuchte von Baumaterialen ist eine kontrovers diskutierte Thematik. Die einzig wissenschaftlich anerkannte Referenzmethode ist die gravimetrische Messung, d.h. die Erfassung der Feuchte eines Stücks des Baumaterials durch Trocknen. Jedoch besteht selbst hier Uneinigkeit über die „richtige“ Temperatur, mit welcher der trockene Materialzustand entsprechend der verwendeten Definition erreicht werden soll. Die Bestimmung der Feuchte ist für eine nahezu unüberschaubare Bandbreite an Fragestellungen im Bauwesen relevant, die Ansätze für die Feuchtemessung sind entsprechend vielseitig. Einige davon haben sich in der Praxis erfolgreich etabliert, ungeachtet der Debatten zur Genauigkeit und Wiederholbarkeit der Messergebnisse. Der Grund dafür liegt in dem dringenden Bedarf an geeigneten Feuchtemessmethoden. Ihre Verwendung ist dennoch nicht völlig verwerflich, sie setzt aber ein fundiertes Verständnis der zugrundeliegenden physikalischen Konzepte und das Wissen um die Anwendung, die Möglichkeiten und Grenzen der Methoden voraus. Die Messung der elektischen Impedanz eines Materials zur Bestimmung von dessen Feuchtegehalt wird häufig besonders kritisch gesehen. Dieser Vorbehalt liegt in erster Linie im starken Einfluss von Salzen und anderen leitfähigen Einschlüssen, wie Lehm oder Metallen, begründet. Dennoch hat die Methode einige einzigartige Eigenschaften, die, bei korrekter Anwendung, wertvolle Einblicke in die Prozesse im betrachteten Material oder in der untersuchten Struktur geben können. Im Gegensatz zu vielen anderen Verfahren kann hiermit ein instrumentieres Monitoring eines Objekts außerhalb einer Laborumgebung verwirklicht werden. Der Messpunkt bleibt konstant an einer Stelle und ermöglicht damit das Erfassen langsamer Vorgänge, wie der Trocknung oder der Anreicherung mit Salzen, unter kontinuierlicher Berücksichtigung der sich ständig ändernden Umgebungsbedingungen. Der Schwerpunkt der Forschungsarbeit liegt in der Aufgabe, die Impedanzspektroskopie in die praktische Anwendung zu bringen. Die Methode selbst wurde lange erforscht, aber erst die neuesten Entwicklungen für integrierte Schaltungen von Impedanzkonvertern haben es ermöglicht, elektrische Impedanzmessungen in Geräte für die Bauwerksüberwachung zu integrieren. Im Vergleich zu Laborgeräten ist ihr Messumfang hinsichtlich Genauigkeit oder Frequenzbereich sicherlich eingeschränkt. Jedoch bietet diese Entwicklung die Möglichkeit, dauerhaft Strukturen außerhalb der Laborumgebung zu instrumentieren, um die Feuchteänderung darin zu erfassen. Besonderere Aufmerksamkeit wurde dabei der Forschung an geeigneten Impedanzelektroden gewidmet, die sowohl für eine Dauerüberwachung geeignet sind als auch den Einfluss der elektrischen Doppelschicht minimieren. Es wurde festgestellt, dass Nickel-Graphit-Silikonelektroden gut geeignet sind. Impedanzdaten werden unter natürlichen Umgebungsbedingungen durch vielfältige Faktoren beeinflusst, welche in einer geschützen Laborumgebung nicht oder nur in anderer Weise auftreten. Um die Messdaten interpretieren zu können, ist es notwendig, diese Einflüsse auf die Impedanz zu untersuchen. Die Materialfeuchte ist hauptsächlich von der relativen Feuchte der Luft sowie vom Kapillarwasser abhängig. In der vorliegenden Arbeit werden Impedanzmessungen für die qualitative Dauerüberwachung der Feuchte poröser Baumaterialien, insbesondere Sandstein, betrachtet. Es wurden Langzeitmessungen durchgeführt, um die sich langsam anpassenden Materialfeuchten verschiedener Sandsteine zu untersuchen. Diese werden im Experiment durch änderungen der relativen Luftfeuchte im Bereich zwischen 54 und 100 % hervorgerufen. Die betrachteten Sandsteinvarietäten wurden aus Süddeutschland gewählt. Im Einzelnen waren dies Abbacher Grünsandstein, Burgpreppacher Sandstein, Mainsandstein weiß-grau, Sander Schilfsandstein und Trebgaster Buntsandstein. Weiter wurden Impedanzmessungen durchgeführt, um die Erkennbarkeit von Wasserfronten bei kapillarem Wassertransport und den nachfolgenden Trocknungsprozess sowie den Einfluss von Temperaturänderungen zu untersuchen. Einleitend wird in Kapitel 1 die wissenschaftliche Motivation für die vorliegende Arbeit dargelegt. In Kapitel 2 werden die grundlegenden Konzepte der Porosität, von Wasserspeicherung und -transport darin und die Effekte von im Porenwasser gelösten Salzen vorgestellt. Kapitel 3 gibt einen überblick zur Dauerüberwachung mit elektrischen Impedanzmessungen unter besonderer Berücksichtigung der Messung an porösen Materialien. Es werden Möglichkeiten für die Temperaturkompensation von Impedanzdaten vorgestellt. Das entwickelte Messsystem für Impedanzmonitoring wird in Kapitel 4 beschrieben. Seine Konzeption wird im übergreifenden Kontext weiterer Feuchtemessmethoden betrachtet, auch im Hinblick auf den speziellen Anwendungsfall des Monitorings denkmalgeschützter, historischer Bauten. Die Optimierung der Impedanzelektroden sowie die Wahl einer geeigneten Messfrequenz werden diskutiert. In Kapitel 5 wird das Impedanzmonitoring von Sandstein im hygroskopischen und überhygroskopischen Feuchtebereich dargestellt. Auf die Betrachtung der zeitabhängigen Daten, die für verschiedene Sandsteintypen während eines Langzeitversuchs unter stufenweiser änderung der Umgebungsfeuchte gesammelt wurden, folgt eine Analyse der Impedanz hinsichtlich Materialfeuchte und dessen Verteilung im Porensystem. Im Anschluss werden Messungen des kapillaren Aufstiegs von Wasser und überlegungen zu möglichen Elektrodenkonfigurationen sowie deren Auswirkungen auf die Erkennung von kapillaren Wasserfronten betrachtet. Das Kapitel schließt mit einer Diskussion über die Temperaturkompensation von Impedanzdaten. Ein Beispiel für praktisch angewendetes Impedanzmonitoring wird im abschließenden Kapitel 6 vorgestellt. Hierbei werden sowohl der hygroskopische, als auch der überhygroskopische Feuchtebereich betrachtet. Es folgen zwei weitere Beispiele, mit denen eine Perspektive auf weitere Anwendungsgebiete eröffnet wird. Das erste schildert die überwachung der Hydratation von Estrich, das zweite veranschaulicht die Nutzung von Impedanzmonitoring für die Bewertung von selbstheilendem Beton.

SMAR 2017, the fourth International Conference on Smart Monitoring, Assessment and Rehabilitation of Civil Structures, ZURICH, SUISSE, 13-/09/2017 - 15/09/2017; One of the aims of Infravation FASSTbridge (Fast and effective Solution for the Strengthening of existing steel bridges) project is to develop a specific reinforcement system dedicated to the preventive fatigue reinforcement of steel bridges. The proposed system relies on the use of an adhesively bonded composite plate. The adhesive has been specifically formulated within the project to respect the required fulfillments of the studied application. The communication will first give insight of the required properties of the adhesive, the composite plate, and the overall system. Based on these requirements and existing literature and standards on the topic, the chosen experimental plan will be described. It will concern investigations on the adhesive before curing, the adhesive after curing, the composite plate and the whole reinforcing system. Both short term and durability investigations are being considered. This experimental plan is currently carried out by the different partners implied within the projects and first results will be presented, especially regarding the properties of the adhesive and the composite plate and first results obtained on assembly.

To study mechanisms of T cell-mediated rejection of B cell lymphomas, we developed a murine lymphoma model wherein three potential rejection antigens, human c-MYC, chicken ovalbumin (OVA), and GFP are expressed. After transfer into wild-type mice 60-70% of systemically growing lymphomas expressing all three antigens were rejected; lymphomas expressing only human c-MYC protein were not rejected. OVA expressing lymphomas were infiltrated by T cells, showed MHC class I and II upregulation, and lost antigen expression, indicating immune escape. In contrast to wild-type recipients, 80-100% of STAT1-, IFN-{gamma}-, or IFN-{gamma} receptor-deficient recipients died of lymphoma, indicating that host IFN-{gamma} signaling is critical for rejection. Lymphomas arising in IFN-{gamma}- and IFN-{gamma}-receptor-deficient mice had invariably lost antigen expression, suggesting that poor overall survival of these recipients was due to inefficient elimination of antigen-negative lymphoma variants. Antigen-dependent eradication of lymphoma cells in wild-type animals was dependent on cross-presentation of antigen by cells of the tumor stroma. These findings provide first evidence for an important role of the tumor stroma in T cell-mediated control of hematologic neoplasias and highlight the importance of incorporating stroma-targeting strategies into future immunotherapeutic approaches.

Im ersten Teil dieser Arbeit konnten durch eine einfache Methode die verschieden substituierten 1,4-Dihydropyrano[2,3-c]pyrazole hergestellt werden. In einer Dreikomponenten-Reaktion von Malondinitril, 1H-Pyrazol-5(4H)-onen und verschiedenen, kommerziell erhältlichen Aldehyden wurden die entsprechenden Zielmoleküle synthetisiert. Vor allem die einfache Durchführung dieser Reaktion soll hervorgehoben werden, denn die Synthese wurde in Ethanol bei Raumtemperatur durchgeführt und nahezu alle der 68 erhaltenen 1,4- Dihydropyrano[2,3-c]pyrazole kristallisierten aus. Die erwünschte Diversität konnte durch die Variation drei verschiedener Alkyl- oder aromatischer Reste erreicht werden. Ebenso konnten verschiedene Derivatisierungsmöglichkeiten, wie die Verseifung der Nitrilgruppe, die Acetylierung der 6-Aminogruppe, der Einsatz von Ketonen anstatt der Aldehyde und die Darstellung eines Pyrano[2,3-c]pyrazol-6(1H)ons, aufgezeigt werden. Die Aktivitäten dieser Verbindungen gegen p38 MAPK waren sehr gering. Die besten Substanzen zeigten eine Hemmung zwischen 42 und 47% bei einer Konzentration von 10 µM. Es konnten daher auch keinerlei Struktur-Wirkungsbeziehungen aus diesen Testergebnissen abgeleitet werden. Der Bindemodus dieser Substanzen wurde basierend auf Modelling und Literaturrecherche validiert und ergab, dass weitreichende Änderungen des Grundgerüstes nötig wären, um die erwünschte Potenz zu erreichen. Der auf Liganden-basierte Wirkstoffdesign aufbauende, zweite Teil der Arbeit, befasste sich mit der Modifikation der Scios-Leitstruktur. Hierbei konnte das Harnstoff-Derivat anhand der Methode von Knölker synthetisiert werden. Der Ersatz der Amidgruppe durch den Harnstoff geht aufgrund eingeschränkter Rotationsmöglichkeiten mit dem Verlust der Aktivität einher. Um eine CH2-Gruppe zwischen Harnstoff und Indol anzubringen, was die Rotation gewährleisten würde, konnte in analoger Syntheseroute ausgehend von kommerziell erhältlichem 5-(Aminomethyl)-1H-indol nur die Harnstoff-Verbindung mit einer Boc-Schutzgruppe am Stickstoffatom des Indols hergestellt werden. Die Abspaltung dieser Schutzgruppe führte jedoch ebenso zur Spaltung an der benzylischen Position und es konnte nur 4-(4-Fluorbenzyl)piperazin-1-carboxamid isoliert werden. Um bereits das N-Methylierte Indolderivat zur Bildung des Harnstoffes einzusetzen, sollte dies ausgehend von 5-Brom-1H-indol hergestellt werden. Der letzte Schritt, die Reduktion von 1-Methyl-1H-indol-5-carbonitril zum entsprechenden (1-Methyl-1H-indol-5-yl)methanamin, konnte nicht erfolgreich synthetisiert werden. Um die Keton-Derivate der Azaindole herzustellen, wurde grundsätzlich von zwei Varianten ausgegangen. Zum einen kann zuerst das Keton gebildet werden und danach der Ring zum Azaindol geschlossen werden, oder am Azaindol-Gerüst soll die Ketonbindung gebildet werden. Dabei kamen bei der ersten Variante folgende Methoden leider erfolglos zum Einsatz: - Friedel-Crafts-Acylierung - Umsatz einer Grignard-Verbindung mit einem Säurechlorid - Palladium-katalysierte Reaktion einer Boronsäure mit einem Carbonsäureester - ortho-Lithiierung und Umsatz mit einem Säurechlorid - ortho-Lithiierung und Umsatz mit einem Weinreb-Amid - ortho-Lithiierung und Umsatz mit einem Aldehyd (mit anschließender Oxidation zum Keton) Bei der zweiten Variante blieben folgende Synthesemethoden ebenso ohne Ketonbildung: - Einsatz von LDA (bei -78 °C) zur ortho-Lithiierung und Umsatz mit einem Weinreb-Amid - Einsatz von s-BuLi (bei -78 °C) zur ortho-Lithiierung und Umsatz mit einem Weinreb-Amid - Einsatz von s-BuLi (bei -40 °C) zur ortho-Lithiierung und Umsatz mit einem Imidazolid - Einsatz von s-BuLi (bei -40 °C) zur ortho-Lithiierung, TEMEDA zur Komplexierung und Umsatz mit einem Imidazolid - Einsatz von s-BuLi (bei -78 °C) zur ortho-Lithiierung, PMDTA zur Komplexierung und Umsatz mit einem Imidazolid Des Weiteren wurden verschiedene heterozyklische 2-Carbonsäuren (Benzimidazol, Benzofuran und Imidazo[1,2-a]pyridin) erfolgreich hergestellt und zu den entsprechenden Amiden umgesetzt. Die Synthese von 4-Brom-1H-indol-2-carbonsäure, die zur Anbringung einer Oxalamidkette in die 4-Position benötigt wird, gelang durch die Hemetsberger-Knittel Indolsynthese. Diese Synthesemethode wurde erfolgreich in der Mikrowelle nach Validierung der Methode durchgeführt. Die Oxalamidkette konnte jedoch durch Brom/Lithium- Austausch und Umsatz mit dem entsprechenden Weinreb-Amid nicht angebracht werden. Die Friedel-Crafts-Acylierung von Indol-2-carbonsäureester in Dichlormethan lieferte das 3-, 5- und 7-Acylderivat im Verhältnis von 2:1:1. Aus den Testdaten können folgende Struktur-Wirkungsbeziehungen abgeleitet werden. Der p-Fluor-Substituent an der Benzylgruppe verbesserte die Aktivität geringfügig und die Piperidin-Amide waren potenter als die entsprechenden Piperazin-Amide. Eine weitere Verbesserung der Hemmwirkung konnte durch N-Methylierung der Indole erreicht werden. Allgemein brachten Substituenten in 3-Position keine Verbesserung der Hemmwirkung im Gegensatz zu den unsubstituierten. Die Acetylgruppe am Indol-Grundgerüst wurde am Besten in Position 7 toleriert. Das Ziel der Arbeit, die Entwicklung neuer Leitstrukturen mithilfe von Struktur- bzw. Liganden-basiertem Wirkstoffdesign, konnte zum Teil erreicht werden. Zudem liefert sie neu gewonnene Erkenntnisse sowohl im synthetischen als auch im Bereich der Struktur-Wirkungsbeziehungen, die als neue Ausgangspunkte für weiterführende Forschungsarbeiten genutzt werden können, um in naher Zukunft diverse potente Inhibitoren zu entwickeln. Structure- and ligand-based drug design was applied to find new lead structures for p38 MAP kinase. In the first part of this thesis different substituted 1,4-dihydropyrano[2,3-c]pyrazoles were synthesized. In the second part of this work the modification of the Scios compound was studied.

The aim of the present work was to contribute useful information towards the development of phytoremediation as an ecological compatible and cost effective clean up strategy for contaminated soils. A key role in this context is played by the mycorrhizal symbiosis of soil-fungi and plant-roots, which stimulates the biodegradation of contaminants in their surrounding (mycorrhizosphere). For the experiments sand was spiked with a predefined PHC test-solution and Pinus sylvestris was selected as host plant. According to an initial batch test Suillus bovinus showed the highest PHC tolerance, while Rhizopogon roseolus was able to utilise PHC as a carbon source. The tolerant symbionts needed to demonstrate their ability to form a steady mycorrhiza under PHC conditions in Petri dish microcosms, and their sustainability in subsequent pot experiments. The results of these experiments revealed that extensive colonisation, which led to the most effective PHC remediation, could be achieved by systematic in-vitro mycorrhization. Rhizopogon roseolus vigorously enhanced plant growth under PHC stress and Suillus bovinus prevents the movement of contaminants. Mycorrhizal pine seedlings accelerated PHC removal by stimulating an effective PHC degrading community of microorganisms. Furthermore, among other effects, mycorrhizal plants improved control of the soil water balance by increasing transpiration and the oxygen supply of the soil. To upgrade the comparability of the results to field conditions the investigations were completed using large-scale lysimeter tests for the observation of important natural attenuation processes like volatilisation, translocation, sorption, degradation and leaching of contaminants as well as plant and fungal development, soil respiration, soil water balance, and further physical and chemical soil parameters. The results made obvious, that the application of appropriate plant-fungus-associations can critically affect the success of phytoremediation.

2-Punkt Transportmessungen, die in der Vergangenheit an ZnSe-basierenden DMS/NMS/DMS Multischichtsystemem durchgeführt wurden, zeigten eine 25-prozentige Erhöhung des Widerstandes beim Übergang vom unpolarisierten in den polarisierten Zustand des DMS. Dieser Magnetowiderstandseffekt wurde durch elektrische Spininjektion in den NMS erklärt. In dieser Arbeit wird zunächst anhand von 4-Punkt Transportmessungen an miniaturisierten, elektronenstrahllithographisch gefertigten DMS/NMS/DMS Strukturen dieser Widerstandseffekt näher untersucht, um eine Bestimmung der Spinrelaxationslänge im nichtmagnetischen II-VI Halbleiter zu erlauben. Aufgrund der im Rahmen dieser Experimente erhaltenen Ergebnisse muss jedoch die Verknüpfung des positiven Magnetowiderstandseffekts mit der elektrischen Spininjektion in den NMS des Multischichtsystems revidiert werden. Im weiteren Verlauf der Arbeit werden Strukturen mit Abmessungen in der Größenordnung von 1 µm hergestellt und gemessen, mit deren Hilfe ein eindeutiger Nachweis der elektrischen Spininjektion in einen nichtmagnetischen Halbleiter mittels Transportmessungen ermöglicht wird. Mit diesen Resultaten kann eine oberer Grenzwert für die Spinfliplänge in ZnBeSe von 100 nm abgeschätzt werden., 2-probe transport measurements that were performed in the past on ZnSe-based DMS/NMS/DMS multilayer-systems showed a 25% increase of the resistance during the transition of the DMS from its unpolarized to its polarized state. This magnetoresistance effect was described by spin injection into the NMS. In this work, this resistance effect is further investigated by 4-probe transport measurements on miniaturized DMS/NMS/DMS structures that were fabricated by electron beam lithography to allow the determination of the spin-relaxation length in the non-magnetic II-VI semiconductor. On the basis of the results that were obtained in the framework of these experiments the connection of the positive magnetoresistance effect with spin injection into the NMS of the multilayer-system has to be revised. In the further course of this work structures with dimensions of the order of 1 µm are fabricated und measured that allow the unambiguous detection of electrical spin injection into a non-magnetic semiconductor by transport measurements. With these results an upper limit of the spin-flip length in ZnBeSe of 100 nm can be estimated.

Activation of microglial cells, the resident macrophages of the brain, occurs rapidly following brain injury. De-ramification, i.e. transformation from ramified into amoeboid morphology is one of the earliest manifestations of microglial activation. In the present study, we identified the physiological mechanisms underlying microglial de-ramification induced by lysophosphatidylcholine (LPC). Patch-clamp experiments revealed activation of non-selective cation currents and Ca(2+)-dependent K(+) currents by extracellular LPC. LPC-activated non-selective cation channels were permeable for monovalent and divalent cations. They were inhibited by Gd(3+), La(3+), Zn(2+) and Grammostola spatulata venom, but were unaffected by diltiazem, LOE908MS, amiloride and DIDS. Ca(2+) influx through non-selective cation channels caused sustained increases in intracellular Ca(2+) concentration. These Ca(2+) increases were sufficient to elicit charybdotoxin-sensitive Ca(2+)-dependent K(+) currents. However, increased [Ca(2+)](i) was not required for LPC-induced morphological changes. In LPC-stimulated microglial cells, non-selective cation currents caused transient membrane depolarization, which was followed by sustained membrane hyperpolarization induced by Ca(2+)-dependent K(+) currents. Furthermore, LPC elicited K(+) efflux by stimulating electroneutral K(+)-Cl(-) cotransporters, which were inhibited by furosemide and DIOA. LPC-induced microglial de-ramification was prevented by simultaneous inhibition of non-selective cation channels and K(+)-Cl(-) cotransporters, suggesting their functional importance for microglial activation.

The invention relates to a fluid cooling device comprising a combined (10) cooling, filtering, pumping and control device, whereby fluid pumped by a pump (16) in a fluid circuit (18) can be filtered by a filtering device (14) and can be cooled by a cooling device (12). In said invention, a control device (42) has a fluid flow distributor (44). Said fluid flow distributor (44) is located in the secondary arm (46) of the cooling device (12) and said device can be controlled independently of the temperature of the fluid to be cooled in such a way that at lower fluid temperatures below a predefinable temperature threshold value (54), the cooling device is at least (12) partially supplied with fluid, thereby improving upon known fluid cooling devices and thereby subjecting the cooling device to a lower load during operation.

Polysaccharides play critical roles in bacteria, including the formation of protective capsules and biofilms and establishing specific host cell interactions. Their transport across membranes is often mediated by ATP-binding cassette (ABC) transporters, which utilize ATP to translocate diverse molecules. Cyclic beta-glucans (C beta Gs) are critical for host interaction of the Rhizobiales, including the zoonotic pathogen Brucella. C beta Gs are exported into the periplasmic space by the cyclic glucan transporter (Cgt). The interaction of an ABC transporter with a polysaccharide substrate has not been visualized so far. Here we use single-particle cryoelectron microscopy to elucidate the structures of Cgt from Brucella abortus in four conformational states. The substrate-bound structure reveals an unusual binding pocket at the height of the cytoplasmic leaflet, whereas ADP-vanadate models hint at an alternative mechanism of substrate release. Our work provides insights into the translocation of large, heterogeneous substrates and sheds light on protein-polysaccharide interactions in general.
