Human-in-the-loop topic modelling incorporates users' knowledge into the modelling process, enabling them to refine the model iteratively. Recent research has demonstrated the value of user feedback, but there are still issues to consider, such as the difficulty in tracking changes, comparing different models and the lack of evaluation based on real-world examples of use. We developed a novel, interactive human-in-the-loop topic modeling system with a user-friendly interface that enables users compare and record every step they take, and a novel topic words suggestion feature to help users provide feedback that is faithful to the ground truth. Our system also supports not only what traditional topic models can do, i.e., learning the topics from the whole corpus, but also targeted topic modelling, i.e., learning topics for specific aspects of the corpus. BACKGROUND: This study aimed to measure the Cobb angle of the fetal spine using three-dimensional ultrasound (3D-US) and to assess the relationship between the Cobb angle and the prognosis of congenital scoliosis. METHODS: From March 2015 to June 2019, 77 pregnant women whose fetuses had suspected spinal skeletal dysplasia consented to undergo 3D-US examinations, and 54 fetuses were selected for the analysis group. The study protocol was approved by the review board of the Institutional Ethics Committee for Fetal Medicine. 3D-US was used to show the structure of the fetal spine in 3 planes, and the Cobb angle was measured on the coronal plane. The diagnostic efficacy of 3D-US was compared to that of X-ray for 33 fetuses. RESULTS: In the diagnosis of congenital scoliosis, the sensitivity, specificity, accuracy, positive predictive value (PPV), and negative predictive value (NPV) of 3D-US were 91.7%, 90.0%, 90.7%, 88.0%, and 93.1%, respectively. The area under the receiver operating characteristic (ROC) curve with 3D-US was 0.908. This experiment was conducted to investigate the effects of lactic acid bacteria preparations on microbial diversity and community structure of calves. On days 1 and 7 of the trial period, feces were collected into sterile tubes and labeled (Day 1: control group D1DZ, experimental group D1SY, and Day 7: control group D7DZ, experimental group D7SY). Twenty Angus calves (150±10 kg) were selected and randomly divided into two groups of 10 calves each. The control group fed a basal diet. In addition to feeding the basal diet, the experimental group was given 15 mL lactobacillus preparation orally at 09:00 and 16:00 h every day. Calves were allowed free feeding and drinking water. All other feeding environments and management conditions remained consistent with the experiment lasting for seven days. At the end of the experiment, the fecal microflora of the calves was analyzed using 16S rRNA sequencing techniques. The 16S rRNA analysis data were processed using the Excel 2007 software and analyzed by the IBM SPSS statistical software (Statistical Analysis System, version 22). The Alpha diversity index analysis showed that the Chao and the Ace indices were significantly different after feeding supplemented with lactic acid bacteria. The PCA analysis showed that the fecal flora structure differed significantly after supplementation with the lactic acid bacteria preparation. Further analysis showed that the lactic acid bacteria increased Firmicutes, Patescibacteria, Rikenellaceae_RC9_gut_group, Clostridium_sensu_stricto_1, and prevotellaceae_UCG-003 in the feces. Therefore, we speculate that lactic acid bacteria preparations play an important role in animal production and are beneficial to the diversity of the fecal microflora of the calves.

In the present study, we investigated the effects of cellulase and Lactiplantibacillus plantarum (L. plantarum) on the fermentation quality, microbial diversity, gene function prediction, and in vitro rumen fermentation parameters of the Caragana korshinskii (C. korshinskii) silage. The experimental treatment groups included control (CK), cellulase (C), L. plantarum (L), and cellulase + L. plantarum (CL). Triplicate silos were sampled after 14 and 56 days of ensiling. The microbial diversity of C. korshinskii silage at 56 days was analyzed using Illumina MiSeq sequencing technology, and the effects of C. korshinskii silage on rumen fermentation were evaluated by the in vitro artificial rumen gas production method. The results showed that the addition of cellulose and L. plantarum treatments reduced ammonia-nitrogen (NH3-N), dry matter (DM), neutral detergent fiber (NDF), and acid detergent fiber content during ensiling. Compared with the CK group, higher lactic acid content was observed in the silage of the C and L groups, but the CL group had a higher acetic acid (AA) content. Compared with the CK group, the relative abundance of Lactiplantibacillus significantly increased, but that of Enterococcus, Weissella, Enterobacter, and Pediococcus significantly decreased in all other groups except the C group (p < 0.05). The results of gene function prediction were as follows: pyruvate kinase, 1-phosphofructokinase, and fructokinase were lactate production enzymes, which suggested the association of the high AA content in the CL group to the high abundance of 6-phosphate glucose dehydrogenase. The abundance of H + -transporting ATPase and ATP-binding cassette (ABC) transporters in the L and CL groups was higher than that in the CK groups. Metabolism of terpenoids and polyketides, mismatch repair, DNA replication, nucleotide excision repair, and homologous recombination increased in the CL group compared with those in the CK group. Compared with the CK group, NH3-N and microbial protein content and the degradation rates of DM and NDF increased in the L and CL groups. To conclude, the L and CL groups had increased Lactiplantibacillus abundance, improved fermentation quality, and high DM degradation rates compared with the CK group.

Background Estimating surgical case duration accurately is an important operating room efficiency metric. Current predictive techniques in spine surgery include less sophisticated approaches such as classical multivariable statistical models. Machine learning approaches have been used to predict outcomes such as length of stay and time returning to normal work, but have not been focused on case duration. Objective The primary objective of this 4-year, single-academic-center, retrospective study was to use an ensemble learning approach that may improve the accuracy of scheduled case duration for spine surgery. The primary outcome measure was case duration. Methods We compared machine learning models using surgical and patient features to our institutional method, which used historic averages and surgeon adjustments as needed. We implemented multivariable linear regression, random forest, bagging, and XGBoost (Extreme Gradient Boosting) and calculated the average R2, root-mean-square error (RMSE), explained variance, and mean absolute error (MAE) using k-fold cross-validation. We then used the SHAP (Shapley Additive Explanations) explainer model to determine feature importance. Results A total of 3189 patients who underwent spine surgery were included. The institution’s current method of predicting case times has a very poor coefficient of determination with actual times (R2=0.213). On k-fold cross-validation, the linear regression model had an explained variance score of 0.345, an R2 of 0.34, an RMSE of 162.84 minutes, and an MAE of 127.22 minutes. Among all models, the XGBoost regressor performed the best with an explained variance score of 0.778, an R2 of 0.770, an RMSE of 92.95 minutes, and an MAE of 44.31 minutes. Based on SHAP analysis of the XGBoost regression, body mass index, spinal fusions, surgical procedure, and number of spine levels involved were the features with the most impact on the model. Conclusions Using ensemble learning-based predictive models, specifically XGBoost regression, can improve the accuracy of the estimation of spine surgery times.

Augmented Reality (AR) has been used to facilitate surgical guidance during External Ventricular Drain (EVD) surgery, reducing the risks of misplacement in manual operations. During this procedure, the key challenge is accurately estimating the spatial relationship between pre-operative images and actual patient anatomy in AR environment. This research proposes a novel framework utilizing Time of Flight (ToF) depth sensors integrated in commercially available AR Head Mounted Devices (HMD) for precise EVD surgical guidance. As previous studies have proven depth errors for ToF sensors, we first assessed their properties on AR-HMDs. Subsequently, a depth error model and patient-specific parameter identification method are introduced for accurate surface information. A tracking pipeline combining retro-reflective markers and point clouds is then proposed for accurate head tracking. The head surface is reconstructed using depth data for spatial registration, avoiding fixing tracking targets rigidly on the patient's skull. Firstly, $7.580\pm 1.488 mm$ depth value error was revealed on human skin, indicating the significance of depth correction. Our results showed that the error was reduced by over $85\%$ using proposed depth correction method on head phantoms in different materials. Meanwhile, the head surface reconstructed with corrected depth data achieved sub-millimetre accuracy. An experiment on sheep head revealed $0.79 mm$ reconstruction error. Furthermore, a user study was conducted for the performance in simulated EVD surgery, where five surgeons performed nine k-wire injections on a head phantom with virtual guidance. Results of this study revealed $2.09 \pm 0.16 mm$ translational accuracy and $2.97\pm 0.91$ degree orientational accuracy.

Rehabilitation training of patients who received total hip arthroplasty (THA) operation is necessary for their rebuilding of motor function. However, most existing exoskeleton devices for hip rehabilitation have an anthropomorphic structure. Misalignment between the mechanical and planted prothesis center is a problem that can cause additional stress in the hip for anthropomorphic exoskeletons, which are thus not applicable to THA rehabilitation training process. Also, the parasitic force due to the cable pulling of soft exoskeletons is also regarded as a shortcoming for users. To address these limitations and to provide training assistance for THA patients for better recovery, a novel hip exoskeleton with parallel structure is presented in this paper. The proposed exoskeleton has a remote actuation and Cable-conduit transmissions and is hence light in weight and can provide bidirectional assistive/resistive torque in the hip without generating stress in the hip, which is significant for THA patients having weak and sensitive planted hip joints. A controller is presented for a stable and safe human-machine interfacing during training with desired assistance delivered. Experiment results based on a benchmark platform verify the performance of the proposed exoskeletons system.

On k-folds cross-validation, the linear regression model had an R2 of 0.34, RMSE of 165.3, and MAE of 128.4. Among all models, the XGBoost regressor performed the best with an R2 of 0.70, RMSE of 110.9, and MAE of 75.8. Based on SHAP analysis of the XGBoost regression, body mass index, spinal fusions, surgical procedure, and number of spine levels involved were the features with the most impact on the model. CONCLUSIONS Utilizing ensemble learning-based predictive models, specifically XGBoost regression, can improve accuracy of the estimation of spine surgery times. CLINICALTRIAL N/A

To establish methodological criteria for the clinical application of E-cervix technology in non-pregnant women, evaluate cervical differences between non-pregnant CI patients and normal women, and explore the predictive value of E-cervix for CI in non-pregnant women.In all, 46 non-pregnant women with CI and 46 non-pregnant women with full-term pregnancy experience were included. Elastic parameters (HR, IOS, EOS, IOS/EOS, CL) of the cervix were measured by transvaginal examination with a mid-sagittal section as the ROI, followed by examination of the whole cervix. The pressure of the probe on the cervix was divided into three levels. The elastic data of three consecutive images were separately obtained by two doctors for three pressure levels. Image stability and the effects of pressure were evaluated and the repeatability of the measurements was tested. In the case and control groups, cervical elasticity images were selected by standardized operation methods. Differences in the parameters between the two groups were determined and the diagnostic efficiency of each parameter was analyzed.The HR, IOS, and EOS showed significant differences when measured on different sequential images obtained by continuous mapping (p 0.05). The elastic parameters obtained from the first and last two images were significantly different (p 0.05) and there were no significant differences between the elastic parameters obtained from the last two images (p 0.05). The HR, IOS, EOS, and IOS/EOS showed significant differences among the pressure levels (p 0.05), while the IOS showed a significant difference only between grades 0 and 2 (p 0.05). E-cervix showed good repeatability within and among operators. The elastic parameters obtained by E-cervix in the CI group and control group were significantly different. ROC curve analysis showed that the HR was the most sensitive index for diagnosing CI.In E-cervix data acquisition, the first image should be excluded and the probe should not put pressure on the cervix. E-cervix has good retest and inter-observer reliability. Compared with normal women, CI patients showed a relatively softer and shorter cervix. E-cervix can be used as an important diagnostic tool for assessing cervical strain.

BACKGROUND: The precise pathogenesis of anophthalmia/microphthalmia remains unknown. Prenatal observation of the optic chiasm in fetuses with this malformation would assist in understanding the embryonic development of the condition. The present study aimed to establish the normal fetal size ranges of decussation of the optic chiasm, optic nerves, and optic tracts in the axial plane using two-dimensional transabdominal ultrasound throughout gestation and to compare these ranges to the corresponding values in fetuses with anophthalmia/microphthalmia METHODS: In total, 310 normal fetuses and 16 fetuses with anophthalmia/microphthalmia were included in this study. The widths of the decussation of the optic chiasm, optic nerves, and optic tracts of normal fetuses at 19–40 weeks' gestation were measured in the axial plane by two-dimensional transabdominal ultrasound. The same widths were retrospectively measured in the axial plane using three-dimensional ultrasound in fetuses with anophthalmia/microphthalmia and compared to the results from the normal fetuses. RESULTS: The decussation, optic nerves, and optic tracts of 310 normal fetuses were measured. The normal widths of the decussation of the optic chiasm, optic nerves, and optic tracts increased linearly with gestational age. The interobserver and intraobserver reproducibility was excellent for the decussation but relatively low for the optic nerves and optic tracts. To analyze the normal development of fetal gallbladder (GB) and evaluate the postnatal outcomes of fetuses with isolated GB anomalies without other structural abnormalities by ultrasound.This was a retrospective study. We collected normal cases during 2016 between 15 and 40 gestational weeks and measured the length and width of GB to establish a normal reference range of GB. Using the 10th and 90th percentiles of the normal reference range, isolated cases of enlarged or small GB without other structural anomalies were selected between January 2017 and December 2019. Simultaneously, cases of echogenic material in the GB were included. Fetuses with non-visualization of the fetal gallbladder (NVFGB) were collected between January 2010 and December 2019. Postnatal outcomes of these cases were followed up and analyzed.There were 670 cases in the normal group and a linear correlation was found between the dimensions of GB and the gestational age, including the length (The postnatal outcomes of most fetuses (96.8%) with isolated GB anomalies such as enlarged GB, echogenic material in the GB were favorable. When it comes to cases of small GB or NVFGB in prenatal ultrasound diagnosis, we should alert biliary atresia in prenatal counseling.

ObjectivesThe objective of the study was to explore the added value of whole-exome sequencing (WES) in abnormal fetuses with detailed prenatal ultrasound and postnatal phenotype with normal karyotype and chromosomal microarray analysis (CMA).MethodsParents of fetuses with structural abnormalities by prenatal ultrasound who consented to provide fetal samples were prospectively recruited from January 2017 to December 2019. With aneuploidies or cases with copy number variations (CNVs) excluded, WES was performed for cases with normal karyotype and CMA results. Detailed prenatal ultrasound and postnatal imaging or pathology features were recommended for further interpretation of genetic variants.ResultsWES was performed for 94 eligible fetuses, DNA samples of which were extracted from 53 parent–fetus trios and 41 proband-only fetal tissues. A diagnostic genetic variant was identified in 37 (39.4%) of 94 fetuses, and 34 (64.2%) were detected in 53 trios, which was significantly greater than 3 (7.3%) in 41 proband-only cases (p < 0.001). In 34 trios with diagnostic genetic variants, 23 (67.6%) were de novo and 11 (32.4%) were inherited with two homozygous and nine heterozygous variants. Fourteen (14.9%) of 94 fetuses had a variant of uncertain significance (VUS). Among 94 cases, six affected pregnancies continued and 88 terminated, and 57 of 88 terminated cases underwent postmortem examinations. With accurate phenotypes demonstrated by prenatal ultrasound and postnatal autopsies, the clinical phenotypes were correlated in 33 (89.2%) of 37 cases with specific genotypes, with the highest matching ratio in skeletal diseases (20/33, 60.6%).ConclusionWES has added value in the genetic diagnosis of abnormal fetuses with normal karyotypes and CMA, particularly in skeletal diseases. Using WES in various anomalous fetuses can broaden the understanding of prenatal phenotypes and genetic variants.

In order to understand the source of groundwater inrush and its hydrogeochemical evolution path truly, determine the evolution characteristics and development trend of water cycle, and effectively solve the problem of water disaster in mining area. In this paper, Jiaozuo mining area, a typical large water mining area of North China type, is selected as the study area. Taking the Ordovician aquifer in the study area as the research object, the Ca2+ hydrochemical equilibrium model of Ordovician limestone water system is established by analyzing the characteristics of concentration gradient field and the relationship between the hydrochemical ions (Ca2+、K++Na+、Mg2+、Cl-、SO42-、HCO3-) and TDS in the mining area, it made a systematic study on the characteristics of Ordovician limestone water circulation, and analyzed the recharge, runoff and discharge characteristics of groundwater in different areas of the mining area. The results show that the Ordovician limestone water in the Jiaozuo mining area has a weak leaching effect in the north and southwest, but a strong leaching effect in the southeast, which shows a runoff characteristic from the north and southwest to the southeast.

Dysregulation of ubiquitin-conjugating enzyme E2S (UBE2S) contributes to tumor progression. However, its clinical significance and biological function in endometrial cancer (EMC) remain unclear. Here, we show that UBE2S is upregulated in EMC and exhibits oncogenic activities via activation of SOX6/β-Catenin signaling. High expression of UBE2S is significantly associated with poor prognosis in two independent cohorts consisting of a total of 773 patients with EMC. in vitro studies demonstrate that ectopic expression of UBE2S promotes cell proliferation and migration, whereas knockdown of UBE2S results in opposite phenotypes. Overexpression of UBE2S in EMC cells enhances the nuclear translocation of β-Catenin, and subsequently induces the expression of c-Myc and Cyclin D1. Inhibition of β-Catenin by XAV-939 markedly attenuates UBE2S-promoted cell growth. Mechanistically, UBE2S suppresses the expression of SOX6 to trigger β-Catenin signaling. Re-expression of SOX6 in UBE2S-expressing EMC cells abolishes the nuclear localization of β-Catenin. Collectively, these data suggest UBE2S may serve as a promising prognostic factor and function as an oncogene in EMC. The newly identified UBE2S/SOX6/β-Catenin axis represents a new potential therapeutic target for EMC intervention.

AIM To explore the effectiveness of cavum septi pellucidi (CSP) width to anteroposterior cerebellar diameter (APCD) ratio as a diagnostic adjunct for prenatal diagnosis of trisomy 18. METHODS Images of normal fetal brain within 15 and 35 weeks were stored in our center from 2016 to 2017. Images of aneuploid fetuses were retrospectively collected from 2004 to 2017. The transverse cerebellar diameter, APCD and CSP width were measured. CSP/APCD and APCD/transverse cerebellar diameter ratios were calculated and compared between euploid and aneuploid fetuses. RESULTS One thousand and forty one fetuses were analyzed, including 817 euploid fetuses and 224 aneuploid fetuses (trisomy 21 117 cases, trisomy 18 82 cases, trisomy 13 9 cases, sex-linked 16 cases). No correlation had been found between both ratios and gestational weeks (P > 0.05). Converting from conventional tillage to no-tillage influences the soil aggregate-size distribution and thus soil organic carbon (SOC) stabilization. However, the dynamics of soil aggregation and the straw-derived carbon (C) incorporation within aggregate fractions are not well understood. An experiment was established in 2004 to test the effects of two treatments, no-tillage with residue (NT) and conventional tillage without residue (CT), on the soil aggregate-size distribution and SOC stabilization in a continuous maize (Zea mays L.) cropping system located in the semiarid region of northern China. Soil samples were collected from the 0–10 cm layer in 2008, 2010 and 2015, and were separated into four aggregate-size classes (>2, 0.25–2, 0.053–0.25, and 2 and 0.25–2 mm) and associated SOC concentration compared with CT. Additionally, to compare straw-derived C incorporation within NT and CT aggregate fractions, 13C-labeled straw was incubated with intact NT and CT soils. After 90 days, the highest proportion of 13C-labeled straw-derived C was observed in the >2 mm fraction, and this proportion was lower in NT than that in CT soil. Overall, we conclude that long-term continuous NT increased the proportion of macroaggregates and the C concentration within macroaggregates, and the physical protection provided by NT is beneficial for soil C sequestration in the continuous maize cropping system in semiarid regions of northern China.

This paper describes a bilateral filter that is being proposed as a coding tool for the Versatile Video Codec (VVC). The filter acts as a loop filter in parallel with the sample-adaptive offset (SAO) filter. Both the proposed filter and SAO act on the same input samples, each filter produces an offset, and these offsets are then added to the input sample to produce an output sample that, after clipping, goes to the next stage. The method has been implemented and tested according to the common test conditions in VVC test model version 5.0. For the all-intra configuration, we report a BD rate figure of -0.4% with an encoder run time increase of 6% and a decoder run time increase of 4%. For the random access configuration, the BD rate figure is -0.5% with an encoder run time increase of 2% and a decoder run time increase of 2%.

Objectives To investigate the role of ultrasound in the preoperative diagnosis of adnexal torsion and describe its histological spectrum and outcomes at different ages. Methods This study comprised a retrospective investigation of a data set that included 195 female patients with clinically suspected adnexal torsion between January 2010 and July 2015. Each patient received a detailed pelvic ultrasound examination by an experienced ultrasound examiner, and a definitive diagnosis was achieved via surgery. The accuracy of an ultrasonic diagnosis of adnexal torsion was assessed. The patients were divided into three groups: female children and adolescents, fertile women, and postmenopausal women. The size, type, histological spectrum, and outcome of adnexal torsion in the different groups were also described. Results The sensitivity, specificity, and accuracy of ultrasound were 0.84, 0.77, and 0.81, respectively. There were significant differences in the presence of ultrasonic hallmarks among the true positive, false positive, and false negative cases of adnexal torsion. Ovarian torsion was identified in 94 cases, and isolated tubal torsion was identified in 15 cases. The most common histologic diagnoses of the pediatric group and the other two groups were a normal ovary and teratoma, respectively. The most common histologic diagnoses in the cases of isolated tubal torsion were mesosalpinx cyst and oviduct inflammation. Conclusions Ultrasonography plays an important role in the preoperative diagnosis of adnexal torsion. Despite ovarian involvement in most of the cases, isolated oviduct torsion was not uncommon. The spectrum of histological diagnoses varied among the age groups.

Objectives To evaluate the usefulness of chromosomal microarray analysis (CMA) in fetuses with ventricular septal defects (VSDs) with or without associated anomalies and normal karyotype. Methods Fetuses with VSDs and normal karyotypes were investigated using an Affymetrix CytoScan HD array. The cases were classified as isolated or non-isolated VSDs. Results Among the 52 VSD fetuses, 22 (42.3%) had isolated defects, 30 (57.7%) had additional other ultrasound anomalies. Twenty-six CNVs were identified in 18 fetuses (34.6%), 15 benign CNVs were detected in 11 (21.2%) fetuses, 8 pathogenic CNVs were detected in 6 (11.5%) fetuses. After excluding 2 fetuses with 22q11.2 deletion syndrome, the rate of pathogenic CNVs was 7.7%. The proportion of variants of unknown significance was 5.8% (3/52). In five cases, additional malformations were detected after birth or abortion, and one case had a prenatal isolated VSD. The detection rate of pathogenic CNVs in non-isolated VSDs was non-significantly higher than that in prenatal or postnatal isolated VSDs (4.5%, 1/22 vs. 16.7%, 5/30, P = 0.226; 0/21 vs. 19.4%, 6/31, P = 0.07). Conclusions The results demonstrated the value of CMA in the prenatal diagnosis of VSDs. The complexity of other defects enhanced the frequency of pathogenic CNVs, although the results were not significantly different.

In this research, diffusion brazing of monocrystal silicon was performed using germanium powders as interlayer. The effects of brazing temperature (950–1050 °C) and time (1–3 h) were studied on the joint microstructure and its mechanical properties. The results indicated that, with the rise in brazing time and temperature, the morphology near the sample edge turned from sintered structures to bridge connections. While those in the center evolved from straight Ge layers to symmetric layer structures with the sequence of “Si base/Isothermal solidification zone/Athermal solidification zone/Ge laye r ”. Withal, the crushing and interlocking morphologies appeared. Bond strength was evaluated and maximum tensile strength over 15.8 MPa was obtained for the joint processed at 1000 °C for 3 h. Results showed that, thickness of diffusion layer (i.e., the isothermal solidification zone and athermal solidification zone) was proved to be the controlling factor for the tensile strength of diffusion brazed monocrystal silicon.

Polyhydramnios is sometimes associated with genetic defects. However, establishing an accurate diagnosis and pinpointing the precise genetic cause of polyhydramnios in any given case represents a major challenge because it is known to occur in association with over 200 different conditions. Whole exome sequencing (WES) is now a routine part of the clinical workup, particularly with diseases characterized by atypical manifestations and significant genetic heterogeneity. Here we describe the identification, by means of WES, of novel compound heterozygous truncating variants in the LMOD3 gene [i.e., c.1412delA (p.Lys471Serfs*18) and c.1283dupC (p.Gly429Trpfs*35)] in a Chinese family with two successive fetuses affected with polyhydramnios, thereby potentiating the prenatal diagnosis of nemaline myopathy (NM) in the proband. LMOD3 encodes leiomodin-3, which is localized to the pointed ends of thin filaments and acts as a catalyst of actin nucleation in skeletal and cardiac muscle. This is the first study to describe the prenatal and postnatal manifestations of LMOD3-related NM in the Chinese population. Of all the currently reported NM-causing LMOD3 nonsense and frameshifting variants, c.1412delA generates the shortest truncation at the C-terminal end of the protein, underscoring the critical role of the WH2 domain in LMOD3 structure and function. Survey of the prenatal phenotypes of all known LMOD3-related severe NM cases served to identify fetal edema as a novel presenting feature that may provide an early clue to facilitate prenatal diagnosis of the disease.

OBJECTIVES The aim of this study was to determine whether cervical elastographic parameters in addition to cervical length (CL) during the 3 trimesters of pregnancy would be predictive of spontaneous preterm birth (sPTB) among low-risk women. METHODS This work was a prospective nested case-control study evaluating cervical elastographic parameters and CL in low-risk women during the 3 trimesters of pregnancy. A binary logistic regression analysis was used to calculate significant covariates for prediction of sPTB. The area under the curve of the prediction model was calculated by using a receiver operating characteristic curve. RESULTS There were 286 women (26 cases and 260 controls) included in the analysis. The parameters of cervical elasticity became softened and heterogeneous during the 3 trimesters of pregnancy in both women with and without sPTB. AIM To assess the reproducibility of a semiautomatic quantification tool for cervical stiffness and evaluate the normal changes in cervical elasticity during the three trimesters of pregnancy. METHODS This longitudinal prospective pilot study evaluated cervical elasticity during the three trimesters of pregnancy (11-14, 20-24 and 28-32 weeks) in women with singleton pregnancies. Women with a history of conization, cerclage, cervical Naboth cysts (diameter > 10 mm), cervical tumors, or uterine malformation were excluded. A semiautomatic tool was used to evaluate the stiffness of the whole cervix and the internal and external cervical os with multiple quantitative elasticity parameters and the cervical length (CL) on the sagittal view via transvaginal elastography. Intraclass correlation coefficients (ICC) and Bland-Altman analysis were used to assess intra- and interobserver variability. E-Cervix parameters during the three trimesters were compared using the Friedman test. RESULTS In total, 217 women with 651 strain examinations during the three trimesters were included. The intra- and interobserver ICC for the E-Cervix parameters ranged from 0.947 to 0.991 and 0.855 to 0.989, respectively. There were significant differences in all parameters among the three trimesters. Cervical elasticity showed significant softening and became heterogeneous during the three trimesters. This paper analyses the causes of jump tripping. Then, the zero sequence protection is set and inter-stage cooperation is improved. Finally, a novel relay protection scheme combining zero-sequence current protection and trip unit is designed. Field test shows that the scheme proposed in this paper can effectively solve the problem of over-step tripping, and avoid the expansion of power outage. It can effectively improve the reliability of power supply. The device can be widely used in intelligent distribution network and substation.

Land use change is an important factor affecting soil carbon and nitrogen cycle, and this is a hot spot in the study of global climate change. Based on the fixed depth method (FD) and the equivalent mass method (ESM), we explored the effects of land use change on soil organic carbon (SOC) and total nitrogen (TN) storage from the perspectives of forest reclamation and farmland vegetation restoration in Karst area. The storage of SOC and TN significantly decreased after the fore-sts were converted to grassland, eucalyptus and farmland. On average, the SOC and TN losses calculated by the FD method were 47.4% and 41.6%, respectively, the losses of SOC and TN calculated by ESM method were 54.8% and 49.7%. The SOC and TN storage increased significantly after abandonment of farmland to grassland or planting eucalyptus. On average, the SOC and TN stocks calculated based on the FD method increased by 60.5% and 49.7%, while the SOC and TN calculated by the ESM method increased by 85.5% and 70.8% respectively. The difference between FD method and ESM method was analyzed, it was concluded that FD method ignored the difference of soil bulk density after land use change. Soil bulk density was significantly increased after forest destruction. The loss of SOC and TN storage was overestimated by the FD method. Soil bulk density decreased after vegetation restoration, FD method will underestimate the enhancement of SOC and TN storage. It is suggested that the ESM method should be used to estimate the impacts of land use change on SOC and TN storage.土地利用变化是影响土壤碳、氮循环的重要因素,也是研究全球气候变化的热点.本研究基于固定深度法(FD)和等效质量法(ESM),从森林开垦和退耕还林还草两个角度探讨喀斯特移民迁入区土地利用变化对土壤有机碳(SOC)和全氮(TN)储量的影响.结果表明: 原始森林被开垦为草地、桉树林和农田后,SOC和TN储量均显著减少;基于FD方法计算的SOC和TN储量分别损失了47.4%、41.6%,而通过ESM方法计算的SOC和TN的损失率分别为54.8%、49.7%.农田撂荒为草地及种植桉树后,SOC和TN储量显著增加;基于FD方法计算的SOC和TN储量提高了60.5%、49.7%,通过ESM方法计算的SOC和TN分别增加85.5%和70.8%.FD方法忽略了土地利用变化后土壤容重的差异,而森林开垦后会显著增加土壤容重,因此,FD方法高估了SOC和TN储量;农田恢复后土壤容重减小,FD方法则会低估SOC和TN储量的增加.建议相关研究选择ESM方法测算土地利用变化对SOC和TN储量的影响.

Biochar is widely and increasingly applied to farmlands. However, it remains unclear how long-term biochar addition alters the characteristics and chlorine reactivity of soil-derived dissolved organic matter (DOM), an important terrestrial disinfection byproduct (DBP) precursor in watersheds. Here, we analyzed the spectroscopic and molecular-level characteristics of soil-derived DOM and the formation and toxicity of DBP mixtures from DOM chlorination for two long-term (5 and 11 years) biochar addition experimental farmlands. As indicated by spectroscopic indices and Fourier transform ion cyclotron resonance mass spectrometry analyses, 11 years of biochar addition could increase the humic-like and aromatic and condensed aromatic DOM and decrease the microbial-derived DOM, while 5 years of biochar addition at the other site did not. The response of condensed aromatic dissolved black carbon did not increase with increasing cumulative biochar dose but appeared to be affected by biochar aging time. Despite the possible increase in aromatic DOM, biochar addition neither increased the reactivity of DOM in forming trihalomethanes, haloacetonitriles, chloral hydrates, or haloketones nor significantly increased the microtoxicity or genotoxicity of the DBP mixture. This study indicates that biochar addition in watersheds may not deteriorate the drinking water quality via the export of terrestrial DBP precursors like wildfire events.

With this method, optically active chiral bicyclic tetrahydropyridazine derivatives could be obtained in one step from readily available starting materials.

Background: Whole-exome sequencing (WES) over the last few years has been increasingly employed for clinical\ud diagnosis. However, one caveat with its use is that it inevitably fails to detect disease-causative variants that occur\ud within noncoding RNA genes. Our experience in identifying pathogenic variants in the noncoding RNU4ATAC gene,\ud in a Chinese family where two successive foetuses had been affected by severe microcephaly, is a case in point.\ud These foetuses exhibited remarkably similar phenotypes in terms of their microcephaly and brain abnormalities;\ud however, the paucity of other characteristic phenotypic features had made a precise diagnosis impossible. Given\ud that no external causative factors had been reported/identified during the pregnancies, we sought a genetic cause\ud for the phenotype in the proband, the second affected foetus.\ud Results: A search for chromosomal abnormalities and pathogenic copy number variants proved negative. WES was\ud also negative. These initial failures prompted us to consider the potential role of RNU4ATAC, a noncoding gene\ud implicated in microcephalic osteodysplastic primordial dwarfism type-1 (MOPD1), a severe autosomal recessive\ud disease characterised by dwarfism, severe microcephaly and neurological abnormalities. Subsequent targeted\ud sequencing of RNU4ATAC resulted in the identification of compound heterozygous variants, one being the most\ud frequently reported MOPD1-causative mutation (51G>A), whereas the other was a novel 29T>A variant. Four\ud distinct lines of evidence (allele frequency in normal populations, evolutionary conservation of the affected nucleotide,\ud occurrence within a known mutational hotspot for MOPD1-causative variants and predicted effect on RNA secondary\ud structure) allowed us to conclude that 29T>A is a new causative variant for MOPD1.\ud Conclusions: Our findings highlight the limitations of WES in failing to detect variants within noncoding RNA genes\ud and provide support for a role for whole-genome sequencing as a first-tier genetic test in paediatric medicine.\ud Additionally, the identification of a novel RNU4ATAC variant within the mutational hotspot for MOPD1-causative variants\ud further strengthens the critical role of the 5′ stem-loop structure of U4atac in health and disease. Finally, this analysis\ud enabled us to provide prenatal diagnosis and genetic counselling for the mother’s third pregnancy, the first report of\ud its kind in the context of inherited RNU4ATAC variants.

In this thesis, we propose and discuss a class of motion-adaptive transforms (MAT) to describe the temporal correlation in image sequences for compression. The temporal correlation is based on motion models, and undirected graphs are used to represent this correlation in image sequences. The transforms are adaptive to general motion fields. Hence, they avoid the predict-update mismatch of the classic block-motion lifting schemes in processing connected and disconnected pixels. Moreover, the proposed transforms are orthonormal for general motion field, and thus, they permit energy conservation and perfect reconstruction. As we represent the motion-connected signals by graphs, we introduce a graph-based covariance matrix model and use the associated eigenvector matrix for compression. As the proposed covariance model is closely related to the graph, the relation between the covariance matrix and theLaplacian matrix is studied and the associated eigenvector matrices are discussed. The class of MAT is constructed by using so-called scale factors.We show that the scale factors determine a relevant subspace of the signal representation.Hence, we propose a subspace-constrained transform, which achieves optimal energy compaction given the subspace constraint. On the other hand, the resulting basis vectors are signal dependent. To construct practical transforms without using covariance matrices, we consider two types of incremental transforms over graphs, namely the uni-directional orthogonal transform (Uni-OT) and the bidirectional orthogonal transform (Bi-OT). In addition, fractional-pel MAT is proposed to further extend the class of MAT. Our fractional-pel MAT can incorporate a general interpolation filter into the basis vectors, while offering perfect reconstruction, orthogonality, and improved coding efficiency. QC 20180305

A highly efficient chiral gold phosphate-catalyzed tandem hydroamination/asymmetric transfer hydrogenation reaction is described. A series of chiral tetrahydroquinolines were obtained in excellent yields and enantioselectivities. In this reaction, the gold catalyst enables both the hydroamination step as a π-Lewis acid and the asymmetric hydrogen-transfer process as an effective chiral Lewis acid.

To establish formulae for the calculation of fetal cardiovascular Z-scores based on femur length (FL), and to compare cardiovascular parameters between fetuses with tetralogy of Fallot (TOF) and normal fetuses in order to assess their value for the prenatal diagnosis of TOF.A total of 329 normal fetuses and 43 fetuses with TOF were included in this study. Eleven cardiovascular dimensions were measured offline after cardiac spatiotemporal image correlation volume acquisition. Normal cardiovascular Z-score formulae were constructed for these measurements based on FL by performing a standard regression analysis followed by weighted regression of absolute residual values. The following ratios were calculated: right ventricular internal diameter (RVID) to left ventricular internal diameter (LVID) and pulmonary artery diameter (PA) to aorta diameter (Ao). Subsequently, all parameters were compared between the normal and TOF groups.Formulae for calculating Z-scores for the 11 cardiovascular dimensions were constructed. FL was significantly correlated with all cardiovascular dimensions assessed (r = 0.816-0.944, P 0.001 for all). RVID, Ao, PA, aortic arch isthmus, and ductus arteriosus mean Z-scores and PA/Ao ratio were statistically significantly different between normal fetuses and those with TOF. In the TOF group, all Ao Z-scores (43/43) were + 2 and all PA/Ao ratios (43/43) were below the normal 95% range. However, only 48.8% (21/43) of the PA Z-scores were -2.The cardiovascular Z-score formulae developed can provide a quantitative basis for the prenatal diagnosis of TOF. Aortic dilatation and abnormal PA/Ao ratio may be markers for the antenatal diagnosis of TOF.

Organic amendment is considered as an effective way to increase soil organic carbon (SOC) stock in croplands. To better understand its potential for SOC sequestration, whether SOC saturation could be observed in an intensive agricultural ecosystem receiving long-term composted manure were examined. Different SOC pools were isolated by physical fractionation techniques of a Cambisol soil under a long-term manure experiment with wheat-maize cropping in North China Plain. A field experiment was initiated in 1993, with 6 treatments including control (i.e., without fertilization), chemical fertilizer only, low rate of traditional composted manure (7.5 t ha−1), high rate of traditional composted manure (15 t ha−1), low rate of bio-composted manure (7.5 t ha−1) and high rate of bio-composted manure (15 t ha−1). The results showed that consecutive (for up to 20 years) composted manure amendments significantly improved soil macro-aggregation, aggregate associated SOC concentration, and soil structure stability. In detail, SOC concentration in the sand-sized fraction (>53 μm) continued to increase with manure application rate, while the silt (2–53 μm) and clay (>2 μm) particles showed no further increase with greater C inputs, exhibiting the C saturation. Further physical separation of small macro-aggregates (250–2 000 μm) into subpools showed that the non-protected coarse particulate organic matter (cPOM, >250 μm) was the fraction in which SOC continued to increase with increasing manure application rate. In contrast, the chemical and physical protected C pools (i.e., micro-aggregates and silt-clay occluded in the small macro-aggregates) exhibited no additional C sequestration when the manure application rate was increased. It can be concluded that repeated manure amendments can increase soil macro-aggregation and lead to the increase in relatively stable C pools, showing hierarchical saturation behavior in the intensive cropping system of North China Plain.

Objectives To analyze the characteristics, associations, and outcomes of prevalence of the right heart and to evaluate the use of sonography to predict the risk of a poor prognosis in fetuses with this condition. Methods We conducted a retrospective cohort study of 182 fetuses with prevalence of the right heart. At the initial evaluation, the left ventricle (LV) and right ventricle (RV) sizes, left atrium and right atrium sizes, great artery diameters, appearance of the aortic arch and ductus arteriosus arch, and flow direction across the foramen ovale and aortic arch were documented. Malformations were documented in fetuses with intracardiac anomalies, who were divided into groups with and without coexisting extracardiac defects. The RV-LV diameter ratio and main pulmonary artery-to-aortic root diameter ratio were calculated and compared. The following variables were analyzed: coexisting intracardiac anomalies, associated extracardiac anomalies, diagnosis before 24 gestational weeks, fetal growth restriction, reversed flow across the foramen ovale, reversed flow in the aortic arch, and RV-LV ratio greater than 2.0. Relationships between these sonographic findings and the prognosis were evaluated by multivariable logistic regression. Results Of the 182 fetuses, 1 (0.5%) had intrauterine death, and 25 (13.7%) had neonatal death; 1 (0.5%) had selective reduction, and 106 (58.2%) underwent termination of pregnancy; 49 (26.9%) survived at this writing. The mean RV-LV ratio was 1.67. Multivariable logistic regression revealed that only 2 sonographic parameters, coexisting intracardiac anomalies (odds ratio, 17.75; 95% confidence interval, 4.18-75.26) and diagnosis before 24 weeks (odds ratio, 17.26; 95% confidence interval, 1.80-165.39) were significantly associated with a poor prognosis. Conclusions The sonographic parameters of coexisting intracardiac anomalies and diagnosis before 24 gestational weeks are significant independent predictors of a poor prognosis in fetuses with prevalence of the right heart. Combinations of these risk factors may be useful in prenatal consultation.

Objectives The objectives of this study are to determine and compare the prevalence of persistent left superior vena cava (PLSVC) in chromosomally normal and abnormal fetuses and to evaluate the potential of PLSVC as a screening marker for chromosomal abnormalities. Methods Women undergoing routine fetal sonographic examinations were evaluated once for the presence of PLSVC. PLSVC was diagnosed on the basis of the identification of an additional vessel in the left of the pulmonary artery in a three-vessel trachea view. Associated abnormalities, karyotypes, and outcomes were analyzed. Results A total of 164 (0.7%, 164/25 171) cases of PLSVC were detected and successfully followed-up. The detection rates were 0.5% (81/17 535) and 1.1% (83/7636) in the low-risk and high-risk cases, respectively. Objectives This study aimed to assess the fetal conus medullaris (CM) position with three-dimensional (3D) ultrasonography and its use in detecting and diagnosing spina bifida occulta (SBO). Methods We examined 150 normal fetuses (gestational age, 20–38 weeks) and 14 fetuses with SBO. All fetuses underwent prenatal 3D ultrasonography, and the CM position in relation to the vertebral body was evaluated using volume contrast imaging. Intra- and inter-observer agreements were assessed using weighted kappa. The correlation between gestational age and CM level was determined by linear regression analysis. Results Operators successfully located the CM position in 145 cases. The CM was at L3 or higher in all normal group cases. To investigate the role of ultrasound in the preoperative diagnosis of adnexal torsion and describe its histological spectrum and outcomes at different ages.This study comprised a retrospective investigation of a data set that included 195 female patients with clinically suspected adnexal torsion between January 2010 and July 2015. Each patient received a detailed pelvic ultrasound examination by an experienced ultrasound examiner, and a definitive diagnosis was achieved via surgery. The accuracy of an ultrasonic diagnosis of adnexal torsion was assessed. The patients were divided into three groups: female children and adolescents, fertile women, and postmenopausal women. The size, type, histological spectrum, and outcome of adnexal torsion in the different groups were also described.The sensitivity, specificity, and accuracy of ultrasound were 0.84, 0.77, and 0.81, respectively. There were significant differences in the presence of ultrasonic hallmarks among the true positive, false positive, and false negative cases of adnexal torsion. Ovarian torsion was identified in 94 cases, and isolated tubal torsion was identified in 15 cases. The most common histologic diagnoses of the pediatric group and the other two groups were a normal ovary and teratoma, respectively. The most common histologic diagnoses in the cases of isolated tubal torsion were mesosalpinx cyst and oviduct inflammation.Ultrasonography plays an important role in the preoperative diagnosis of adnexal torsion. Despite ovarian involvement in most of the cases, isolated oviduct torsion was not uncommon. The spectrum of histological diagnoses varied among the age groups.

The influences of tillage systems on soil carbon (C) stocks have been studied extensively, but the distribution of soil C within aggregate fractions is not well understood. The objective of this study was to determine the influences of various tillage systems on soil aggregation and aggregate-associated C under wheat ( Triticum aestivum L.) and corn ( Zea mays L.) double cropping systems in the North China Plain. The experiment was established in 2001, including four treatments: moldboard plow (MP) with residue (MP+R) and without residue (MP-R), rotary tillage with residue (RT), and no-till with residue (NT). In 2007 soil samples were collected from the 0–5, 5–10, and 10–20 cm depths, and were separated into four aggregate-size classes (>2 000, 250–2 000, 53–250, and >53 μm) by wet-sieving method. Aggregate-associated C was determined, and the relationships between total soil C concentration and aggregation-size fractions were examined. The results showed that NT and RT treatments significantly increased the proportion of macroaggregate fractions (>2 000 and 250–2 000 μm) compared with the MP-R and MP+R treatments. Averaged across all depths, mean weight diameters of aggregates (MWD) in NT and RT were 47 and 20% higher than that in MP+R. The concentration of bulk soil organic C was positively correlated with MWD ( r =0.98; P =0.024) and macroaggregate fraction ( r =0.96; P =0.036) in the 0–5 cm depth. In the 0–20 cm depth, comparing with MP+R, total C occluded in the >2 000 μm fraction was increased by 9 and 6% under NT and RT, respectively. We conclude that adoption of conservation tillage system, especially no-till, can increase soil macro-aggregation and total C accumulation in macroaggregates, which may improve soil C sequestration in the intensive agricultural region of the North China Plain.

Objectives To determine whether the use of 4-dimensional (4D) sonography with spatiotemporal image correlation (STIC) and tomographic ultrasound imaging (TUI) can provide additional information with respect to 2-dimensional (2D) echocardiography in the prenatal diagnosis of anomalous pulmonary venous connections. Methods The study population consisted of 10 cases that were initially suspected to have total or partial anomalous pulmonary venous connections by prenatal 2D echocardiography between January 2008 and April 2011. All 10 cases were further examined and analyzed by 4D sonography with STIC-TUI. Detailed postnatal surgery or autopsy was performed on all 10 fetuses. Results Total anomalous pulmonary venous connections were found in 5 cases, and a partial connection was diagnosed in 1 fetus postnatally. The remaining 4 cases were confirmed to have normal pulmonary venous connections. Four of the 5 fetuses with anomalous pulmonary venous connections had an additional major cardiac defect; 1 fetus had an isolated connection. Anomalous drainage was supracardiac to the superior vena cava in 2 cases, cardiac to the coronary sinus in 3, and partially infracardiac to the portal vein in remaining case. The pulmonary venous connections were completely and correctly visualized with 2D echocardiography in 2 of the 10 cases, partially identified in 4, and not distinguished completely in 4. Four-dimensional sonography imaging with STIC-TUI clearly visualized the connections in 9 of the 10 cases, and the remaining case was partially identified. Conclusions Four-dimensional sonography with STIC-TUI facilitates visualization of pulmonary venous connections, thus supplying additional information with respect to 2D echocardiography in the prenatal diagnosis of anomalous pulmonary venous connections.

To evaluate the usefulness of chromosomal microarray analysis in fetuses with ventricular septal defects (VSDs) with or without associated anomalies and normal karyotype.Fetuses with VSDs and normal karyotypes were investigated by using an Affymetrix CytoScan HD array. The cases were classified as isolated or nonisolated VSDs.Among the 52 VSD fetuses, 22 (42.3%) had isolated defects and 30 (57.7%) had additional other ultrasound anomalies. Twenty-six CNVs were identified in 18 fetuses (34.6%), 15 benign CNVs were detected in 11 (21.2%) fetuses, and 8 pathogenic CNVs were detected in 6 (11.5%) fetuses. After excluding 2 fetuses with 22q11.2 deletion syndrome, the rate of pathogenic CNVs was 7.7%. The proportion of variants of unknown significance was 5.8% (3/52). In five cases, additional malformations were detected after birth or abortion, and one case had a prenatal isolated VSD. The detection rate of pathogenic CNVs in nonisolated VSDs was nonsignificantly higher than that in prenatal or postnatal isolated VSDs (4.5%, 1/22 vs 16.7%, 5/30, P = 0.226; 0/21 vs 19.4%, 6/31, P = 0.07).The results demonstrated the value of chromosomal microarray analysis in the prenatal diagnosis of VSDs. The complexity of other defects enhanced the frequency of pathogenic CNVs, although the results were not significantly different. © 2016 John WileySons, Ltd.

The eddy covariance(EC) technique is generally regarded as a standard method for crop evapotranspiration measurements.However,the imbalance of energy closure prevails in the EC observations.Evaluating the effect of energy balance closure on EC measurement is critical for improving the accuracy of this method.In this paper,a weighting method(the Lysimeter method) was used as reference to evaluate the effect of energy balance ratio(EBR) on EC evapotranspiration.The results revealed that daytime EBR varied seasonally in the field,where wheat and maize were rotated in the winter and summer,respectively.The EBR was higher in autumn and winter but lower in the spring and summer.For the wheat field,mean daytime EBR varied from 0.26 to 2.84 with an average of 1.15.As for the maize field,EBR varied from 0.19 to 2.59 with a mean value of 0.78.The evapotranspiration(ET) using EC(ETec) was clearly lower than derived by Lysimeter(ETL).The mean ratio of ETec to ETL(ETec/ETL) was 0.61 and 0.50 during the wheat growing season and the maize season,respectively.The ET observed with these two approaches significantly correlated with each other(P 0.01),with their characteristics of seasonal variation performing in a similar manner.The ETec/ETL was found to be proportional to EBR(P 0.01) in both the winter wheat field and the summer maize field.Furthermore,the effect of the leaf area index(LAI) on the relationship between ET ratios and EBR was significant in the even crop field during the entire growing season of winter wheat and the maize growing stage when the LAI was higher than 1.However,the effect was insignificant in the uneven maize field when the LAI was less than 1.On the other hand,friction velocity(u*) exerted a strong impact on EBR and its relationship with ET ratios.EBR was observed to be proportional to u* in both the winter wheat field and the summer maize field.ET ratios were proportional to EBR when u* was small.Nonetheless,the correlation between these two parameters showed recession while u* increasing,especially in summer maize field,ETec/ETL was not significantly correlated with EBR when u* was bigger than 0.3 m/s.Due to the weak turbulent intensity and the large eddy under the condition of low friction velocity,the EC sensor could not catch enough turbulent energy in a limited observation period(30 minutes),which caused lower measured turbulent fluxes and imbalanced energy closure.The results offer a possible way to correct the underestimated ETec using EBR.

Background Groundwater contaminated with arsenic imposes a big challenge to human health worldwide. Using natural compounds to subvert the detrimental effects of arsenic represents an attractive strategy. The transcription factor nuclear factor erythroid 2-related factor 2 (Nrf2) is a critical regulator of the cellular antioxidant response and xenobiotic metabolism. Recently, activation of the Nrf2 signaling pathway has been reported to confer protection against arsenic-induced toxicity in a cell culture model. Objectives The goal of the present work was to identify a potent Nrf2 activator from plants as a chemopreventive compound and to demonstrate the efficacy of the compound in battling arsenic-induced toxicity. Results Oridonin activated the Nrf2 signaling pathway at a low subtoxic dose and was able to stabilize Nrf2 by blocking Nrf2 ubiquitination and degradation, leading to accumulation of the Nrf2 protein and activation of the Nrf2-dependent cytoprotective response. Pretreatment of UROtsa cells with 1.4 μM oridonin significantly enhanced the cellular redox capacity, reduced formation of reactive oxygen species (ROS), and improved cell survival after arsenic challenge. Conclusions We identified oridonin as representing a novel class of Nrf2 activators and illustrated the mechanism by which the Nrf2 pathway is activated. Furthermore, we demonstrated the feasibility of using natural compounds targeting Nrf2 as a therapeutic approach to protect humans from various environmental insults that may occur daily.

In reliability-based robust design optimization (RBRDO) formulation, the product quality loss function is minimized subject to probabilistic constraints. Since the quality loss function is expressed in terms of the first two statistical moments, mean and variance, three methods have been recently proposed to accurately and efficiently estimate the moments: the univariate dimension reduction method (DRM), performance moment integration (PMI) method, and percentile difference method (PDM). In this paper, a reliability-based robust design optimization method is developed using DRM and compared to PMI and PDM for accuracy and efficiency. The numerical results show that DRM is effective when the number of random variables is small, whereas PMI is more effective when the number of random variables is relatively large.

The reliability-based design optimization (RBDO) using performance measure approach for problems with correlated input variables requires a transformation from the correlated input random variables into independent standard normal variables. For the transformation with correlated input variables, the two most representative transformations, the Rosenblatt and Nataf transformations, are investigated. The Rosenblatt transformation requires a joint cumulative distribution function (CDF). Thus, the Rosenblatt transformation can be used only if the joint CDF is given or input variables are independent. In the Nataf transformation, the joint CDF is approximated using the Gaussian copula, marginal CDFs, and covariance of the input correlated variables. Using the generated CDF, the correlated input variables are transformed into correlated normal variables and then the correlated normal variables are transformed into independent standard normal variables through a linear transformation. Thus, the Nataf transformation can accurately estimates joint normal and some lognormal CDFs of the input variable that cover broad engineering applications. This paper develops a PMA-based RBDO method for problems with correlated random input variables using the Gaussian copula. Several numerical examples show that the correlated random input variables significantly affect RBDO results.

Structural analysis and design optimization have recently been extended to consider various uncertainties. If the statistical data for the uncertainties are sufficient to construct the input distribution function, the uncertainties can be treated as random variables and RBDO is used; otherwise, the uncertainties can be treated as fuzzy variables and PBDO is used. However, many structural design problems include both uncertainties with sufficient data and uncertainties with insufficient data. For these problems, RBDO will yield an unreliable design since the distribution functions of uncertainties are not believable. On the other hand, treating the random variables as fuzzy variables and invoking PBDO may yield too conservative design with a higher optimum cost. This paper proposes a new design formulation using the performance measure approach (PMA). For the inverse analysis, this paper proposes a new most probable/possible point (MPPP) search method called maximal failure search (MFS), which is an integration of the enhanced hybrid mean value method (HMV+) and maximal possibility search (MPS) method. Some mathematical and physical examples are used to demonstrate the proposed inverse analysis method and design formulation.

A highly efficient chiral gold phosphate-catalyzed tandem hydroamination/asymmetric transfer hydrogenation reaction is described. A series of chiral tetrahydroquinolines were obtained in excellent yields and enantioselectivities. In this reaction, the gold catalyst enables both the hydroamination step as a π-Lewis acid and the asymmetric hydrogen-transfer process as an effective chiral Lewis acid.
