Kurzfassung Einleitung: Das Mamma Karzinom ist die weltweit häufigste Krebsart bei Frauen* (WHO, 2021). Die Diagnose Brustkrebs bereitet den Betroffenen Ängste aber auch Schmerzen. Diese beiden Phänomene können sich auch gegenseitig beeinflussen (Xiao, Li, Xie, Xu, & Liu, 2018). Die Aromapflege zählt zu den komplementären Pflegemethoden, welche mit ätherischen Ölen angewendet wird. Einige Öle können angstlösende und schmerzlindernde Eigenschafen bieten (Deutsch-Grasl, Buchmayr, & Fink, 2018). Ob und welchen Einfluss die Anwendung der Aromapflege auf Ängste und Schmerzen bei Brustkrebspatientinnen* hat soll diese Arbeit aufzeigen. Methodik: Bei dieser Bachelorarbeit handelt es sich um eine systematische Literaturarbeit. Die Methodik wird anhand vier Phasen beschrieben. Die Vorgehensweise der Literaturrecherche und die Bewertung der Literatur wird erläutert. Anschließend wird die endgültige Auswahl der verwendeten Literatur dargestellt. Auch das Vorgehen der Synthesenbildung wird aufgezeigt. Um die Forschungsfrage beantworten zu können wurden eine Online-Recherche von Oktober 2021 bis Jänner 2022 durchgeführt und dabei mithilfe ausgewählter Ein- und Ausschlusskriterien 11 quantitative und eine qualitative wissenschaftliche Studie ausgewählt, die zuvor mittels Bewertungsbogen von Brandenburg, Panfil, und Mayer (2007) bewertet wurden. Ergebnisse: Die Schmerzintensität und das Angstniveau von Brustkrebspatientinnen* wurden in den Studien von (Keun Joo et al., 2005) und (Xiao et al., 2018) signifikant reduziert. Die Ergebnisse der Studien von (Beyliklioğlu & Arslan, 2019), (Trambert, Kowalski, Wu, Mehta, & Friedman, 2017), (Ozkaraman, Dügüm, Özen Yılmaz, & Usta Yesilbalkan, 2018), (Deng, Xie, Liu, Li, & Xiao, 2022) und (Franco et al., 2016) zeigten eine Minderung des Angstniveaus. Kein positiver Einfluss der Aromaanwendung wurde hingegen in der Studie von (Halm, Baker, & Harshe, 2014) gefunden. Der Schmerzscore in den Studien von (Clemo-Crosby, Day, Stidston, McGinley, & Powell, 2018) und (Tamaki et al., 2017) konnte mithilfe der Aromaanwendung nicht reduziert werden. In den Studien von (Lua, Salihah, & Mazlan, 2015) und (Deng et al., 2022) wurde jedoch eine signifikante Verbesserung der Schmerzintensitäten festgestellt. Der qualitative Ansatz der Studie von (Ho, Kwong, Wan, Ho, & Chow, 2017) zeigte, dass Brustkrebspatientinnen* die Aromaanwendung als positive Erfahrung mit sofortigen Veränderungen empfanden. Schlussfolgerung: Zusammenfassend lässt sich sagen, dass die Studienergebnisse der Anwendungen der Aromapflege bei Brustkrebspatientinnen* mit Schmerzen und Ängsten eine geringe Evidenz haben. Trotzdem kann die Aromapflege zur Anwendung kommen, da keine negativen, sondern viele positive Erfahrungen (entspannend, wohltuend, angenehm, zu Ruhe kommen) berichtet wurden. Als komplementäre Pflegemethode soll und kann die Aromapflege bei Brustkrebspatientinnen* angewendet werden, um das subjektive Erleben zu fördern. Bei weiteren Forschungsarbeiten sollte jedoch ein quantitativer Ansatz verwendet werden, um die Anwendung der Aromapflege mit einer wissenschaftlich höheren Evidenz zu begründen. Abstract Introduction: Breast cancer is the world’s most common type of cancer in women* (WHO, 2021). The diagnosis of breast cancer causes anxiety but also pain. These two phenomena can also influence each other (Xiao et al., 2018). Aroma care is one of the complementary care methods, which is applied with essential oils. Some oils may offer anxiety-relieving and pain-relieving properties (Deutsch-Grasl et al., 2018). This work is intended to show whether and what effect the use of aroma care has on anxiety and pain in breast cancer patients*. Methodology: This Bachelor’s thesis is a systematic literature thesis. The methodology is described in four phases. The procedure of literature research and the evaluation of literature is explained. The final selection of the literature used is then presented. The process of synthesis formation is also shown. To answer the research question, online research was conducted from October 2021 to January 2022. Eleven quantitative and one qualitative scientific study were selected using selected inclusion and exclusion criteria, which were previously evaluated by means of evaluation forms from Brandenburg, Panfil, and Mayer (2007). Results: The pain intensity and anxiety levels of breast cancer patients* were evaluated in the studies by Keun Joo et al. (2005) and Xiao et al. (2018) significantly reduced. The results of the studies by Beyliklioğlu & Arslan (2019), Trambert et al. (2017), Ozkaraman et al. (2018), Deng et al. (2022) and Franco et al. (2016) showed a decrease in the level of anxiety. However, no positive influence of flavour application was found in the study by Halm, Baker, & Harshe (2014). The pain score in the studies by Clemo-Crosby et al. (2018) and Tamaki et al. (2017) could not be reduced using the flavour application. However, the studies of Lua et al. (2015) and Deng et al. (2022) showed a significant improvement in pain intensities. The qualitative approach of the study by Ho et al. (2017) showed that breast cancer patients* perceived flavor application as a positive experience with immediate changes. Conclusion: In summary, the study results of aroma care applications in breast cancer patients* with pain and anxiety have little evidence. Nevertheless, aroma care can be used, as there are not negative experiences reported, but many positive ones (relaxing, soothing, pleasant, relaxing). Aroma care should and can be used as a complementary care method in breast cancer patients* to promote subjective experience. However, further research should use a quantitative approach to justify the use of essential care on the basis of a scientifically higher level of evidence.

ZusammenfassungTechnologien der Digitalisierung haben großes Potenzial, die Art und Weise, wie wir wirtschaften und arbeiten, grundlegend zu verändern. Vor allem in der Produktentstehung verspricht die Digitalisierung ein hohes Nutzenpotenzial. Das Projekt „Instrumentarium zur Gestaltung individualisierter virtueller Produktentstehungsprozesse in der Industrie 4.0“ (IviPep) hat das Ziel der menschengerechten Gestaltung der digitalen Arbeitswelt bei gleichzeitiger Effizienzsteigerung der Produktentstehung. In dem vorliegenden Beitrag werden ausgehend von der Zielsetzung die Ergebnisse des Forschungsvorhabens beschrieben. Hierzu werden sowohl die elementaren Forschungsergebnisse, als auch die Anwendungsbeispiele der Partnerunternehmen vorgestellt. Abschließend wird das Instrumentarium beschrieben. Das Verbundprojekt wurde im Rahmen des Programms „Zukunft der Arbeit“ vom Bundesministerium für Bildung und Forschung (BMBF) und dem Europäischen Sozialfonds (ESF) gefördert (Förderkennzeichen 01FI16133).

Zirkulierende Tumorzellen (CTCs) sind maligne Zellen, die in sehr geringer Anzahl im peripheren Blut von Tumorpatienten zu finden sind. Sie können entweder vom Primär-tumor oder von Metastasen in sekundären Organen stammen und sind in der Lage, sich nach dem Verlassen der Blutbahn in verschiedenen Geweben anzusiedeln und neue Me-tastasen auszubilden. Der Nachweis dieser CTCs im Blut ist mittlerweile zu einem viel-versprechenden und viel beforschtem Gebiet der Onkologie geworden. Das Vorhanden-sein von CTCs im Blut von Tumorpatienten kann ein früher Indikator für eine Metasta-sierung sein, als zuverlässiger Vorhersageparameter für die Prognose dienen und die Effektivität einer Therapie aufzeigen. In der vorliegenden Arbeit wurde ein immunologisches Verfahren entwickelt, womit CTCs in venösem Vollblut schnell und einfach als EpCAM-und Zytokeratin-positive und CD45-negative Zellen nachgewiesen werden können. Dabei erfolgt nach einer Ly-se der Erythrozyten in der Probe eine Leukozytendepletion mittels MACS (magnetic-activated cell sorting) und die Identifikation der Tumorzellen mittels FACS-Analyse mit einer festgelegten Gatingstrategie. Mit diesem Verfahren wurden Blutproben von 42 Patienten mit metastasierten gastrointestinalen Tumoren und von 10 gesunden Normal-spendern auf die Anzahl von CTCs/3,75ml Blut untersucht. Dabei wiesen die Patienten annähernd signifikant (p=0,076) mehr CTCs auf als die Normalspender. In 43% der Fäl-le hatten die Patienten >2 CTCs/3,75ml Vollblut und galten damit als CTC-positiv. Pati-enten mit Kolorektalem Karzinom zeigten mit 71% den höchsten Anteil an CTC-Positivität. Bei den gesunden Normalspendern ließen sich in keinem Fall >2 CTCs nachweisen. Nach einer Evaluation dieser Methode in einer weiteren Studie mit einem größeren Pati-enten- und Vergleichskollektiv könnte eine höhere Beweisebene erreicht werden, die den Einsatz in der klinischen Routine ermöglicht. Daneben sind eine weitere Optimie-rung des FACS-Verfahrens und eine Erweiterung des Antigenspektrums bei der CTC-Detektion zur Erhöhung der Sensitivität und Spezifität erforderlich. Patienten würden von diesem einfachen und wenig invasivem CTC-Detektionsverfahren vor allem im Hinblick auf eine Individualisierung der Therapie und auf das Vermeiden einer Überthe-rapie deutlich profitieren., Circulating tumor cells are malignant cells which can be found in very low number in the peripheral blood of patients with malignant diseases. They originate either from the primary tumor or from metastases in secondary organs and are able to settle down in different tissues and form new metastases after leaving circulation. Detection of CTCs in the blood has come to a promising and much researched field in oncology. The presence of CTCs in the blood of patients with malignant diseases can be an early indicator for metastasizing, serve as a reliable prediction parameter for prognosis and show the efficacy of therapy. In this paper an immunological method was developed to detect CTCs in venous whole blood fast and easily as EpCAM and cytokeratin positive and CD45 negative cells. In this method after lysis of erythrocytes in the sample a depletion of the leucocytes by MACS (magnetic activates cell sorting) and the identification of the tumor cells by FACS analysis with a determined gating strategy is accomplished. With this method blood samples of 42 patients with metastatic gastrointestinal tumors and 10 healthy donors were tested for the number of CTCs/3,75 ml blood. Patients had more CTCs than the healthy donors with approaching significance (p=0,076). 43% of the patients had > 2 CTCs/3,75 ml whole blood and were considered to be CTC positive. Patients with colorectal cancer showed with 71% the highest ratio of CTC positivity. In the group of healthy donors > 2 CTCs could not be detected. After evaluating this method in further studies with a bigger collective of patients and healthy donors evidence with higher level could be reached so that the use in clinical routine is possible. Therefore a further improvement of the FACS and an extension of the antigen spectrum within the CTC detection is necessary for increase of sensitivity and specificity. Patients would clearly profit from this easy and low invasive CTC detection method especially in terms of individualization of therapy and avoiding an overtherapy.

Die Regulation der Nahrungsaufnahme ist vielschichtig und seit Jahrzehnten im Fokus der Forschung, da die Erfolge einer Pharmakotherapie bei Störungen des Nahrungsaufnahmeverhaltens bisher unzureichend sind. Die Komplexität der beteiligten regulatorischen Systeme und deren enge Verzahnung untereinander bedingen, dass die genauen Mechanismen bisher nicht bis ins Detail geklärt werden konnten. Obwohl gesichert ist, dass das zentrale serotonerge System einen Einfluss auf die Regulation der Nahrungsaufnahme ausübt, ist die Wirkungsweise des 5-HT1A-Rezeptors nicht vollständig aufgeklärt. Durch die vorliegende Arbeit sollen die Kenntnisse zur Beteiligung des serotonergen Systems und insbesondere des 5-HT1A-Rezeptors an der Regulation der Nahrungsaufnahme bei der Maus durch mehrere Untersuchungen erweitert werden. Die beiden Mausstämme NMRI und C57BL/6 und die transgene Mauslinie L35, die sich durch eine Überexpression des postsynaptischen 5-HT1A-Rezeptors auszeichnet, wurden nach Gabe des 5-HT1A-Rezeptoragonisten 8-OH-DPAT untersucht. Die Experimente wurden unter verschiedenen Versuchsbedingungen durchgeführt: (1) ad libitum-Fütterung, der Test erfolgte während der Hellphase, (2) nach 16h Futterdeprivation, der Test erfolgte wiederum während der Hellphase und (3) unter ad libitum-Fütterung, der Test erfolgte während der Dunkelphase. Dabei wurden beide Geschlechter in zwei Altersstufen durch Verwendung verschiedener Dosierungen bzw. isotonischer Kochsalzlösung untersucht. Mit Hilfe des 5-HT1A-Rezeptorantagonisten WAY100635 sollen die 8 -OH-DPAT-Wirkungen am 5-HT7- und alpha2-adrenergen Rezeptor als Ursache für die beobachteten Effekte ausgeschlossen werden. Zunächst wurde jeweils die Menge an aufgenommener Nahrung in bestimmten Zeiträumen (0-30min, 30min-60min, 60min-120min; nach 24h) nach einer systemischen Injektion von 8-OH-DPAT bestimmt. Parallel dazu wurden die aufgenommene Wassermenge und die motorische Aktivität im Haltungskäfig ermittelt. Anschließend erfolgte eine nähere Charakterisierung der beobachteten Effekte mit Hilfe der Mikrostruktur des Fressens. Neben der Bestätigung einer unter bestimmten Voraussetzungen durch 8 -OH-DPAT induzierten Hyperphagie bei der Maus, wurde ein Einfluss von 8-OH- DPAT auf die Futteraufnahme in Abhängigkeit vom Mausstamm, Geschlecht, Fütterungsstatus, Altersstufe und Tagesphase dargestellt. Durch den 5-HT1A- Rezeptorantagonisten WAY100635 konnte eine Aktivierung des 5-HT1A-Rezeptors als Ursache für die beobachteten Effekte auf die Menge an aufgenommenem Futter identifiziert und eine 8-OH-DPAT-Wirkung am 5-HT7 und alpha2-adrenergen Rezeptor ausgeschlossen werden. Bei ad libitum-gefütterten männlichen NMRI- Mäusen konnte während der Hellphase 60min-120min nach einer 8-OH-DPAT-Gabe eine Hyperphagie beobachtet werden. Bei C57BL/6-Mäusen gelang dies nur in der adulten Altersstufe und nach einer initialen Hypophagie. Denkbar für die Diskrepanz könnten Stammesunterschiede sein. Beim Vergleich der Wirkungen der Wildtyp-Tiere vom NMRI-Stamm und der transgenen Mäuse der Linie L35, die durch eine Überexpression des postsynaptischen 5-HT1A-Rezeptors charakterisiert sind, konnte erstmals ein Einfluss des postsynaptischen 5-HT1A-Rezeptors auf die Regulation der Futteraufnahme gezeigt werden. Für alle getesteten Gruppen der transgenen Mäuse war nach der Gabe des 5-HT1A-Rezeptoragonisten 8-OH-DPAT ein Einfluss des postsynaptischen 5-HT1A-Rezeptors auf die Futteraufnahme nachweisbar. Der Umfang der Beeinflussung variierte in Abhängigkeit vom Geschlecht und der Altersstufe. Erstmals konnte eine 8-OH-DPAT-induzierte Hyperphagie auch bei weiblichen Mäusen nachgewiesen werden. Bei adulten ad libitum-gefütterten Tieren war jedoch in beiden Geschlechtern kein Hyperphagie-Effekt mehr nachweisbar. Weitere Untersuchungen müssen zeigen, ob und inwiefern sich der postsynaptische 5-HT1A-Rezeptor während der Individualentwicklung und Reifung verändert. Die Ergebnisse der vorliegenden Arbeit lassen somit Schlüsse auf die Relevanz des serotonergen Systems im Allgemeinen und des 5-HT1A-Rezeptors im Speziellen für die Regulation der Nahrungsaufnahme zu. Der dargestellte Einfluss des postsynaptischen 5-HT1A- Rezeptors zeigt dabei die Vielschichtigkeit der Regulation der Nahrungsaufnahme und verdeutlicht, dass bei der Entwicklung von Pharmaka mit Wirkung auf die Nahrungsaufnahme das serotonerge System mitberücksichtigt werden sollte., The success of pharmacotherapy in disorders of eating behavior has been insufficient. This is why the complex regulation of food intake is in the focus of research for decades. The involved multipart regulatory systems show a close interaction with each other. Because of this the exact mechanisms have not yet been elucidated in detail. Although it is ensured that the central serotonergic system exerts an influence on the regulation of food intake, the impact of the serotonin (5-HT)1A receptor is not completely understood. The present work enlarges the knowledge of the participation of the serotonergic system, and in particular of the 5-HT1A-receptor in the regulation of food intake in the mouse while using various tests. In the experiments the food intake of two mouse-strains, NMRI and C57BL/6, and the transgenic mouse L35, with an overexpression of the postsynaptic 5-HT1A-receptor, after a systemic injection of the 5-HT1A-receptor-agonist 8-OH-DPAT has been investigated. The tests were conducted under three different conditions. First, mice have been fed ad libitum while the test started in the light phase. Second, mice have been food deprived for 16h and again tested during the light phase. Last, mice have been fed ad libitum and the experiment started at the beginning of the dark phase. With the help of the 5-HT1A-receptorantagonist WAY100635 an 8-OH- DPAT-effect via the 5-HT7- and the alpha2-adrenergic-receptor should be ruled out. After systemic injection of 8-OH-DPAT the amount of eaten food has been detected in different intervals (0-30min, 30min-60min, 60min-120min). In addition to this, water intake and motor activity in the home cage have been determined. For further characterization of the 8-OH-DPAT induced effects on food intake the microstructure of feeding was used. As the most important result an effect of 8-OH-DPAT on food intake in mice has been shown. However, this effect was depending on the mouse strain, sex, feeding status, age, and time of testing. Specifying this, male ad libitum fed NMRI mice show a hyperphagia but C57BL/6 males do so only in the adult group and after a hypophagic response at the beginning of the trial. This implicates strain differences. Because of tests with the 5-HT1A-receptor-antagonist WAY100635 it is clear that the effects of 8-OH-DPAT are due to its impact at the 5-HT1A- receptor. The assessment of the effects of wild type animals, and transgenic mice shows a distinct effect of the postsynaptic 5-HT1A-receptor on food intake for all tested transgenic groups. First, the injection of NaCl leads to a different “basal”-food intake. This could be shown due to reduced food intake 30min-60min after testing begun. Second, food intake was influenced by application of 8-OH-DPAT. The degree of influence has been varied depending on gender and age. For the first time, a hyperphagia in female mice has been shown. Conversely, in adult transgenic mice there was no effect of 8-OH-DPAT on food intake. Further tests have to clarify whether and to which extend the postsynaptic 5-HT1A-receptor is subject of changes during maturation. The results of the present work allow conclusions about the importance of the serotonergic system in general and of the 5-HT1A-receptor in particular in the regulation of food intake. The presented impact of the postsynaptic 5-HT1A- receptor on food intake clearly shows the complexity of the regulation of food intake.

Der Mensch ist seinem Selbstverständnis nach ein Wesen, das sich im Wahrnehmen und Handeln spontan, d.h. aus eigenem, selbst gewählten Antrieb, bestimmen kann. Schon Aristoteles und Kant versuchten dies erkenntnis- und handlungstheoretisch zu begründen. In den Neurowissenschaften hingegen behauptet man, dass die Möglichkeit, sich seinen Antrieb zum Wahrnehmen und Handeln selbst wählen und geben zu können, nicht besteht. Dass eine Synthese beider Standpunkte möglich ist, versucht der bisher kaum rezipierte, empirisch fundierte 'kritische Personalismus' William Sterns zu zeigen.

Introduction Myelodysplastic syndromes (MDS) are clonal hematologic disorders characterized by ineffective hematopoiesis, dysplasia and increased risk of progression to acute myeloid leukemia. The development of targeted therapies for MDS has been lagging behind and remains a key clinical challenge that has been hampered, at least in part, by difficulties to establish in vivo model systems that recapitulate disease heterogeneity and complexity. Attempts to generate a xenograft model of lower risk MDS have only achieved low and often transient levels of engraftment. Recent evidence from mouse studies suggests that MDS is a disease in which both the hematopoietic system and the bone marrow microenvironment might be involved. Thus, we hypothesized that a specific MDS microenvironment might be required for the successful modeling of low risk MDS in mice, proposing a dependency of the “disease propagating cells“ on their corresponding niche cells in human MDS. Methods Our study is based on xenotransplantation of material from 19 MDS patients classified as follows: IPSS low risk (n=6), intermediate-1 risk (n=13), WHO 2008 classification: MDS 5q- (n=7), MDS RCMD (n=7), MDS RAEB I (n=3), MDS-U (n=1), MDS RARS (n=1). MDS CD34+ cells were co-injected with patient-derived mesenchymal stromal cells (MSCs) directly in the bone marrow cavity (i.f) of NOD.Cg-Prkdscid Il2rgtm1Wjl/Szj (NSG) or NSGS (NSG mice expressing human SCF, IL3 and GM-CSF) mice. Molecular tracking of MDS cells was carried out by copy number analysis (Affymetrix SNP 6.0 Arrays), metaphase cytogenetics, interphase FISH, Roche 454 deep sequencing and pyrosequencing of known mutations. Mice were analyzed after a minimum of 16 weeks post transplantation. Results We show that co-injection of MDS CD34+ cells with their corresponding MSCs leads to significant and long-term engraftment of over 77% of the MDS patients analyzed, both in NSG (10/13 patients, range hCD45+= 1-18%) and NSGS mice (7/8 patients, range hCD45+=2.2-74%). In contrast, absence of MSCs or co-injection of healthy age-matched MSCs only gave rise to limited engraftment in NSG mice (2/7 patients (hCD45+=1-3.8%) and 1/2 patients (hCD45+=2%), respectively). Transplanted samples exhibited a clear myeloid bias and significant engraftment of cells with progenitor (CD34+CD38+) and stem cell phenotype (CD34+CD38-) that could be serially transplanted. In addition, presence of morphologically dysplastic cells was readily detectable in NSGS mice. Importantly, molecular analysis of the engrafted cells confirmed their “diseased” origin as they carried identical lesions to the ones present in the original MDS patient. Furthermore, we could demonstrate that disease-propagating stem cells in lower risk MDS exclusively reside within the lin-CD34+CD38- stem cell fraction. Finally, RNA sequencing analysis comparing MDS and age-matched healthy control MSCs revealed altered expression of key genes involved in cellular adhesion, extra-cellular matrix (ECM) remodeling and cellular cross-talk in diseased MSCs, strongly supporting the notion of a complex interplay between MDS hematopoietic cells and their corresponding stroma. In addition, patient MSCs exhibited clear molecular features of fibrosis, a clinical feature often associated with MDS. Conclusion In this study we have identified patient-derived MSCs as a critical functional component of lower risk MDS. Together with MDS stem cells, these patient MSCs form a functional stem cell-niche unit, which allows the propagation of the disease in a xenograft recipient. The striking changed expression in diseased MSCs of genes involved in processes like cytokine-cytokine receptor interaction, cellular adhesion, ECM remodeling as well as hypoxia further suggests that diseased MDS cells might alter the function of the normal HSC niche into one that can support the requirement of MDS cells. Studying the interaction of MDS stem cells and MSCs at the cellular and molecular level will provide a platform for unraveling the molecular basis of clonal dominance in MDS as well as allow the design of targeted strategies aimed to disrupt the MDS stem cell-MSC niche interactions. Water vortex power plants are presumed to combine energy production with a fish way for up and down-stream migration at the same time. The authors present the construction of a water vortex power plant in a lab with special design to investigate the upstream and downstream fish migration. 3D Particle Tracking Velocimetry (PTV) offers a flexible technique for the determination of velocity fields and surface heights in flows. In this project, a prior analysis of the PTV methodology and the software used has been carried out, the MoveInspect of the AICON company. Posteriori has been carried out a study of the surface velocities of the water and of its heights in the 1: 1 prototype of a water vortex turbine, using MoveInspect. The PTV methodology has already been proven to be able to study with some good results flows with turbulences and vortices, but for small laboratory researches, with a controlled environment.
