Introduction There is a substantial risk of developing stenosis and dysfunction in the arteriovenous fistula (AVF) in patients on hemodialysis (HD). Far infrared radiation (FIR) is a non-invasive local intervention with a potentially beneficial effect on AVF patency. The underlying mechanism is not clear. It was hypothesized that a single FIR treatment reduces factors of inflammation and promotes endothelial vasodilators in the AVF. Methods Forty HD patients with an AVF were included in an open-label intervention study. Patients were randomized to receive either FIR (FIR group) or no FIR (control group). Blood samples were drawn directly from the AVF and from a peripheral vein in the non-AVF arm before (T0) and 40 min after (T40) treatment during a HD session. The changes [median (interquartile range)] in circulating factors of inflammation, endothelial function and vasoreactivity during FIR were measured. Results In the AVF a single FIR treatment during dialysis resulted in a significantly diminished decrease in soluble vascular cell adhesion molecule, sVCAM [−31.6 (−54.3; 22.1) vs −89.9 (−121.6; −29.3), P = .005] and soluble intercellular adhesion molecule, sICAM [−24.2 (−43.5; 25.3) vs −49 (−79.9; −11.6), P = .02] compared with the control group. Other factors, such as interleukins, nitrite, nitrate and tumor necrosis factor 1, also declined during dialysis, but with no significant differences related to FIR in either the AVF or the non-AVF arm. Conclusion A single FIR treatment attenuated the decrease in sVCAM and sICAM in the AVF compared with a control group during HD. Findings do not support the hypothesis of a vaso-protective effect of FIR. The long-term effects of FIR on the AVF are unknown.

SP140 is an epigenetic reader protein expressed predominantly in immune cells. GWAS studies have shown an association between SP140 single nucleotide polymorphisms (SNPs) and diverse autoimmune and inflammatory diseases, suggesting a possible pathogenic role for SP140 in immune-mediated diseases. We previously demonstrated that treatment of human macrophages with the novel selective inhibitor of the SP140 protein (GSK761) reduced the expression of endotoxin-induced cytokines, implicating a role of SP140 in the function of inflammatory macrophages. In this study, we investigated the effects of GSK761 on in vitro human dendritic cell (DC) differentiation and maturation, assessing the expression of cytokines and co-stimulatory molecules and their capacity to stimulate T-cell activation and induce phenotypic changes. In DCs, lipopolysaccharide (LPS) stimulation induced an increase in SP140 expression and its recruitment to transcription start sites (TSS) of pro-inflammatory cytokine genes. Moreover, LPS-induced cytokines such as TNF, IL-6, and IL-1β were reduced in GSK761- or SP140 siRNA- treated DCs. Although GSK761 did not significantly affect the expression of surface markers that define the differentiation of CD14+ monocytes into immature DCs (iDCs), subsequent maturation of iDCs to mature DCs was significantly inhibited. GSK761 strongly reduced expression of the maturation marker CD83, the co-stimulatory molecules CD80 and CD86, and the lipid-antigen presentation molecule CD1b. Finally, when the ability of DCs to stimulate recall T-cell responses by vaccine-specific T cells was assessed, T cells stimulated by GSK761-treated DCs showed reduced TBX21 and RORA expression and increased FOXP3 expression, indicating a preferential generation of regulatory T cells. Overall, this study suggests that SP140 inhibition enhances the tolerogenic properties of DCs, supporting the rationale of targeting SP140 in autoimmune and inflammatory diseases where DC-mediated inflammatory responses contribute to disease pathogenesis.

Bulletin, Acquiring higher sprint and jump abilities are advantageous toward future success for young soccer players. The aim of the present study was to investigate how maturity status is correlated with the development of sprint and jump abilities in adolescent male soccer players during the period of height growth spurts. We identified the age at peak height velocity（ APHV） for 76 young soccer players aged between 12.1 and 15.7 years. Furthermore, maturity offset （MO）, which indicates years from APHV, was calculated as an index for maturity status of each player. The correlation coefficients for MO with 20-m sprinting and vertical jumping tests were -0.722 and 0.668, respectively, which were significantly higher than those for chronological age. Partial correlation analysis controlling for maturity status indicated that height had only a small influence on the development of sprint and jump abilities at this age. Contrary to previous studies on non-athletic boys, body mass had a moderately positive effect on sprint and jump abilities of adolescent soccer players when maturity status was controlled for, suggesting body mass increase with fat-free mass is a factor that enhances both abilities in adolescent soccer players. Coaches should be aware of APHV and maturity of each player and that wide inter-individual variations of maturity exist within players even at same chronological age during adolescence.

The advent of methods for efficient generation and cardiac differentiation of pluripotent stem cells opened new avenues for disease modelling, drug testing, and cell therapies of the heart. However, cardiomyocytes (CM) obtained from such cells demonstrate an immature, foetal-like phenotype that involves spontaneous contractions, irregular morphology, expression of embryonic isoforms of sarcomere components, and low level of ion channels. These and other features may affect cellular response to putative therapeutic compounds and the efficient integration into the host myocardium after in vivo delivery. Therefore, novel strategies to increase the maturity of pluripotent stem cell-derived CM are of utmost importance. Several approaches have already been developed relying on molecular changes that occur during foetal and postnatal maturation of the heart, its electromechanical activity, and the cellular composition. As a better understanding of these determinants may facilitate the generation of efficient protocols for in vitro acquisition of an adult-like phenotype by immature CM, this review summarizes the most important molecular factors that govern CM during embryonic development, postnatal changes that trigger heart maturation, as well as protocols that are currently used to generate mature pluripotent stem cell-derived cardiomyocytes.

PurposeThe aim of this retrospective research is to compare frontal sinus dimensions in skeletal Class I, skeletal Class II, and skeletal Class III individuals and to evaluate the relationship of these dimensions with anterior skull base length and some cephalometric values.Materials and MethodsIn this research, we used lateral cephalometric radiographs of 60 people aged 17 to 25. In individuals with skeletal Class I malocclusion, skeletal Class II malocclusion due to mandibular insufficiency, and skeletal Class III malocclusion due to mandibular excess, measurements of frontal sinus length and height as well as S-N, Co-A and Co-Gn lengths, ANB0, FMA0, SN-GoGn0 angles values were performed. The length between the highest point and the lowest point of the frontal sinus was calculated as the height of the frontal sinus, and the length between the most anterior and the most posterior points of the frontal sinus was calculated as the length of the frontal sinus.ResultsThe frontal sinus length and height were found to be higher in skeletal Class III individuals than in skeletal Class I and skeletal Class II individuals, however, there was no significant difference between skeletal Class I and Class II individuals.ConclusionThe increase in frontal sinus height and length correlated positively with the decrease in the ANB angle and the increase in the SN and Co-Gn lengths. The dimensions of the frontal sinus may be an indicator for the remaining mandibular growth potential.

The primary aim of this study is to examine whether bursting interhemispheric synchrony (bIHS) in the first week of life of infants born extremely preterm, is associated with microstructural development of the corpus callosum (CC) on term equivalent age magnetic resonance imaging scans. The secondary aim is to address the effects of analgesics such as morphine, on bIHS in extremely preterm infants. A total of 25 extremely preterm infants (gestational age [GA] < 28 weeks) were monitored with the continuous two-channel EEG during the first 72 h and after 1 week from birth. bIHS was analyzed using the activation synchrony index (ASI) algorithm. Microstructural development of the CC was assessed at ~ 30 and ~ 40 weeks of postmenstrual age (PMA) using fractional anisotropy (FA) measurements. Multivariable regression analyses were used to assess the primary and secondary aim. Analyses were adjusted for important clinical confounders: morphine, birth weight z-score, and white matter injury score. The serrano artisanal cheese (SAC) is a traditional raw milk dairy produced in southern Brazil. The maturation temperature plays a crucial role regarding the cheeses’ microbiological and physicochemical features and has been underexplored when it comes to unpasteurized milk dairy in Brazil. The objective of the present work is to evaluate the effect of ripening temperature on foodborne bacteria (total and thermotolerant coliforms, positive coagulase Staphylococcus and the production of its enterotoxin, Listeria monocytogenes, Escherichia coli and Salmonella spp.) and physicochemical attributes (moisture, water activity, pH, acidity and sodium chloride concentration) of SAC during maturation at 5ºC-20ºC for up to 60 days. Metagenomic analysis was also evaluated through the rDNA 16S method at ripening time of 0-60 days. The samples were safe for L.monocytogenes, E.coli, Salmonella spp. and staphylococcal toxin. The physicochemical parameters reached legal boundaries as follows: moisture, after 30 days at 5ºC; fat, after 30 days at 12.5ºC and 20ºC; fat at dry extract, after 30 days at 12.5ºC or 15 days at 5ºC. The microbiological features indicate that positive coagulase Staphylococcus reached legal conditions after 30 days when cheeses were ripped at 12.5ºC and 20ºC; thermotolerant coliforms after 30 days at 12.5ºC and 20ºC; and total coliforms after 15 days at 20ºC. Lower temperatures lead to lower inactivation rates, and Staphylococcus did not reach legal parameters at 5ºC positive coagulase regarding moisture, water activity and pH reduction. Meanwhile, sodium chloride concentration and enhanced acidity played important role regarding foodborne pathogen inactivation. The main lactic acid bacteria within the cheeses during ripening were Enterococcus sp. and Lactococcus sp.

Among alternative perovskite device architectures, mesoporous scaffolds based on metal oxide and carbon look highly promising due to their inherent high stability. In this study, the perovskite deposition has been realized on a 1.5 cm2 active surface area using inkjet infiltration through the mesoporous scaffold, affording a clean industrial process for large-scale and stable perovskite devices. A spectacular enhancement of photovoltaic performances from 10% to 14% was then obtained through the implementation of a maturation step. A study of the occurring mechanisms was conducted using a full set of characterization techniques including J-V measurements, UV–visible and PL spectroscopies, LBIC and PL imaging, XRD, and surface roughness measurements. In addition, the impact of this maturation step on the durability of the performances was investigated.

Electrospun nanofiber constructs represent a promising alternative for mimicking the natural extracellular matrix in vitro and have significant potential for cardiac patch applications. While the effect of fiber orientation on the morphological structure of cardiomyocytes has been investigated, fibers only provide contact guidance without accounting for substrate stiffness due to their deposition on rigid substrates (e.g., glass or polystyrene). This paper introduces an in situ fabrication method for suspended and well aligned nanofibrous scaffolds via roller electrospinning, providing an anisotropic microenvironment with reduced stiffness for cardiac tissue engineering. A fiber surface modification strategy, utilizing oxygen plasma treatment combined with sodium dodecyl sulfate solution, was proposed to maintain the hydrophilicity of polycaprolactone (PCL) fibers, promoting cellular adhesion. Human-induced pluripotent stem cell (hiPSC)-derived cardiomyocytes (CMs), cultured on aligned fibers, exhibited an elongated morphology with extension along the fiber axis. In comparison to Petri dishes and suspended random fiber scaffolds, hiPSC-CMs on suspended aligned fiber scaffolds demonstrated enhanced sarcomere organization, spontaneous synchronous contraction, and gene expression indicative of maturation. This work demonstrates the suspended and aligned nano-fibrous scaffold provides a more realistic biomimetic environment for hiPSC-CMs, which promoted further research on the inducing effect of fiber scaffolds on hiPSC-CMs microstructure and gene-level expression.

In China, the wild population of Nanjing Linden (Tilia miqueliana M.) is experiencing a drastic decline, primarily due to high levels of empty seeds. This study aimed to measure the biochemical and physiological changes during fruit and seed development in T. miqueliana to determine the developmental mechanism. The weight method and photosynthetic respiration were used to determine the biological aspects of both fruits and embryos, while transmission electron microscopy and the anthrone colorimetric method were used to determine the endosperm content, including sugar, starch, protein, and fat. Enzyme-linked immunosorbent assays were conducted to determine the levels of endogenous plant hormones such as indole-3-acetic acid (IAA), gibberellic acid 3 (GA3), zeatin riboside (ZR), and abscisic acid (ABA). The nonlinear least-squares method was used to fit the model of nutrient and hormone levels, revealing that fruit size expanded from the 5th to the 65th day and that fruit moisture content exhibited a downward trend, along with a decrease in fruit respiration intensity. Embryos were found to be fully developed between 35 DAF and 65 DAF, while the nutrients in the endosperm, i.e., sugar, starch, protein, and fat, continuously accumulated after 50 DAF. Additionally, ABA, IAA, GA3, and ZR contents were found to synergistically regulate seed development and maturation.

Human-induced pluripotent stem cell-derived cardiomyocytes (hiPSC-CMs) are increasingly used for personalised medicine and preclinical cardiotoxicity testing. Reports on hiPSC-CM commonly describe heterogenous functional readouts and underdeveloped or immature phenotypical properties. Cost-effective, fully defined monolayer culture is approaching mainstream adoption; however, the optimal age at which to utilise hiPSC-CM is unknown. In this study, we identify, track and model the dynamic developmental behaviour of key ionic currents and Ca2+-handling properties in hiPSC-CM over long-term culture (30–80 days). hiPSC-CMs > 50 days post differentiation show significantly larger ICa,L density along with an increased ICa,L-triggered Ca2+-transient. INa and IK1 densities significantly increase in late-stage cells, contributing to increased upstroke velocity and reduced action potential duration, respectively. Importantly, our in silico model of hiPSC-CM electrophysiological age dependence confirmed IK1 as the key ionic determinant of action potential shortening in older cells. We have made this model available through an open source software interface that easily allows users to simulate hiPSC-CM electrophysiology and Ca2+-handling and select the appropriate age range for their parameter of interest. This tool, together with the insights from our comprehensive experimental characterisation, could be useful in future optimisation of the culture-to-characterisation pipeline in the field of hiPSC-CM research.

In den letzten Jahrzehnten ist die Zahl der Erwachsenen und Jugendlichen, die an nicht organisierten oder wettbewerbsorientierten Sportarten teilnehmen, gestiegen. In der Pubertät beeinflussen sowohl hormonelle Veränderungen als auch mechanische Belastungen die Entwicklung von Muskeln und Sehnen. Derzeit gibt es keine Informationen über die Interaktion dieser beiden Stimuli, aber es besteht die Vermutung, dass im Zuge der Pubertät bei Athleten und Nicht-Athleten im Vergleich zu Muskelkraft und Sehnensteifigkeit ungleiche Verhältnisse auftreten können. Die Folge dieses Ungleichgewichts könnte eine höhere mechanische Belastung für die Sehnen sein, was weiter zu Verletzungen führen kann. Angesichts der unzureichenden Beweise für die Entwicklung des muskulotendinösen Gewebes während der Pubertät und des Mangels an Kenntnissen darüber, wie die Reifung die Muskel-Sehnen-Einheit, insbesondere die Interaktion mit überlagerten mechanischen Belastungen, beeinflusst, untersucht diese Arbeit die morphologische und mechanische Entwicklung von Knieextensoren und Patellasehne, indem Nicht-Athleten und Athleten aus drei Altersgruppen verglichen werden (frühe Pubertät: 12–14 Jahre, engl.:EA; späte Pubertät: 16–18 Jahre, engl.: LA; Erwachsene: 20–35 Jahre, engl.: YA). Athleten erreichten häufiger Dehnungsgrößen von mehr als 9% Dehnung im Vergleich zu Nicht-Athleten, was auf einen erhöhten mechanischen Bedarf an der Sehne hinweist. Obwohl das Training die Eigenschaften der M. quadriceps femoris-Sehnen-Einheit verbessert, bleibt ihre Entwicklung von früher Pubertät bis zum Erwachsenenalter bei Athleten und Nicht-Athleten ähnlich, mit dem Hauptunterschied zwischen früher Pubertät und später Pubertät. Alter und sportliches Training waren jedoch mit einer höheren Prävalenz von Ungleichgewichten innerhalb der Muskel-Sehnen-Einheit und einer damit einhergehenden erhöhten mechanischen Belastung oder Beanspruchung für die Patellasehne verbunden. In recent decades, the number of adults and especially adolescents who participate in some kind of non-organized or competitive sports has been increasing. During adolescence, the development of muscle and tendons is affected both by maturation, due to hormonal changes, and by mechanical loading. However, there is no information currently on the interaction of this double fold stimulus although there is reason to believe that during adolescence in athletes and non-athletes there may be imbalances developing between muscle strength capacity and tendon stiffness. The result of this imbalance could be the tendon exposure to high mechanical demand by the associated working muscles, which might further lead to tendon injury. Considering the not satisfactory evidence of the musculotendinous tissue development during adolescence, and the lack of knowledge about how maturation affects the muscle-tendon unity, especially in interaction with superimposed mechanical loading, this thesis investigates the morphological and mechanical development of the knee extensors and patellar tendon, by comparing non-athletes and athletes in three different age groups (i.e., early adolescents: EA 12–14 years; late adolescents: LA 16–18 years, and young adults: YA 20–35 years). Athletes were more likely to reach strain magnitudes higher than 9% strain compared to non-athlete controls indicating an increased mechanical demand for the tendon. Although athletic training enhances the properties of the quadriceps femoris muscle-tendon unit, their development from early-adolescence to adulthood remains similar in athletes and non-athletes with the major alterations between early and LA. However, both age and athletic training were associated with a higher prevalence of imbalances within the muscle-tendon unit and a resultant increased mechanical demand for the patellar tendon.

The prospective use of human-induced pluripotent stem cell-derived cardiomyocytes (hiPSC-CM) for cardiac regenerative medicine strongly depends on the electro-mechanical properties of these cells, especially regarding the Ca2+-dependent excitation–contraction (EC) coupling mechanism. Currently, the immature structural and functional features of hiPSC-CM limit the progression towards clinical applications. Here, we show that a specific microarchitecture is essential for functional maturation of hiPSC-CM. Structural remodelling towards a cuboid cell shape and induction of BIN1, a facilitator of membrane invaginations, lead to transverse (t)-tubule-like structures. This transformation brings two Ca2+ channels critical for EC coupling in close proximity, the L-type Ca2+ channel at the sarcolemma and the ryanodine receptor at the sarcoplasmic reticulum. Consequently, the Ca2+-dependent functional interaction of these channels becomes more efficient, leading to improved spatio-temporal synchronisation of Ca2+ transients and higher EC coupling gain. Thus, functional maturation of hiPSC-cardiomyocytes by optimised cell microarchitecture needs to be considered for future cardiac regenerative approaches.

Pit-wall mud (PWM) fosters bacterial communities involved in Baijiu production. PWM varies depending on pit age and height. In this study, we explored the bacterial communities in PWM and factors driving their evolution. The abundance and diversity of bacterial communities were low in new PWM (NPWM). In old PWM (OPWM), similar but diverse bacterial communities were observed at different heights. Lactobacillus was the predominant genus in NPWM, and Caproiciproducens, Aminobacterium, Hydrogenispora, Lactobacillus, Petrimonas, Syntrophomonas, and Sedimentibacter were the dominant genera in OPWM. A decrease was noted in the abundance of Lactobacillus, which indicated evolution. Among all the physicochemical properties, pH had the highest degree of interpretation with an R2 value of 0.965. pH also exerted the strongest effect on bacterial communities. The path coefficients of pH on bacterial community diversity and abundance were 0.886 and 0.810, respectively. Caproiciproducens and Clostridium sensu stricto 12 metabolized lactic acid, inhibiting the growth of Lactobacillus at a suitable pH, which led to the maturation of PWM. Our findings enrich the literature on the evolution of bacterial communities in PM and the maturation of PM.

Dissertação de Mestrado em Engenharia Agronómica apresentada na Escola Superior Agrária de Ponte de Lima O cultivo do mirtilo em Portugal tem vindo a crescer nos últimos anos. Trata-se de uma espécie que despertou grande interesse comercial, principalmente ao nível das exportações para países no norte da Europa. Nas últimas décadas, tem-se assistido a um aumento da temperatura média à escala global, com invernos cada vez mais amenos, com consequências ao nível da quebra da dormência das espécies particularmente exigentes em frio invernal, traduzindo-se em abrolhamentos tardios, irregulares, florações fracas e muito escalonadas, com influência negativa na produtividade do mirtilo. O objetivo deste trabalho, consistiu na avaliação de uma formulação com vários produtos fertilizantes e um fitofarmacêutico, na quebra da dormência em três cultivares de mirtilo: Duke, Bluecrop e Chandler. O estudo consistiu em três ensaios (um por cultivar) e num delineamento completamente casualizado, com quatro repetições. No campo, avaliou-se a fenologia das cultivares em estudo no período de 6 de março de 2021 até 22 de maio de 2021, tendo as observações incidido em 2 ramos por cada duas plantas, em cada repetição. Após cada colheita, mediu-se o calibre dos frutos e colocaram-se 20 frutos por repetição na arca frigorífica para as determinações analíticas. Com as colheitas terminadas, realizou-se as análises em laboratório do ºBrix, acidez titulável, pH e matéria seca. Dos resultados obtidos, verificou-se que não houve diferenças estatisticamente significativas entre as modalidades ensaiadas, mas a formulação testada nestas plantas ajudou a que as fases fenológicas fossem mais uniformes e precoces, bem como um início de colheita ligeiramente mais cedo e um número superior de frutos e calibre nas plantas tratadas. No geral, os valores das determinações analíticas encontraram-se numericamente superiores às plantas da modalidade testemunha, no entanto, teria interesse fazer novo ensaio com a mesma formulação ou com novos ajustes e comparar com um produto diferente de forma a confirmar resultados. The crop of blueberry in Portugal has been growing in recent years. It is a species that has aroused great commercial interest, mainly in terms of exports to countries in northern Europe. In recent decades, there has been an increase in the average temperature on a global scale, with increasingly mild winters, with consequences in terms of the breakdown of the dormancy of particularly demanding species in winter cold, resulting in late, irregular brats, weak blooms and very with an influence on blueberry productivity. The purpose of this the work consisted of the evaluation of a formulation with several products fertilisers and a plant protection plant, in the breaking of dormancy in three blueberry cultivars: Duke, Bluecrop and Chandler. The study consisted of three trials (one per cultivar) and consisted of a completely casualized design, with four repetitions. In the field, the phenology of the cultivars under study was evaluated from March 6, 2021 to May 22, 2021, observations were based on 2 branches per two plants in each repetition. After each harvest, the size of the fruits was measured and 20 fruits were placed per repetition in the freezer for analytical determinations. With the harvests completed, the analyses were carried out in laboratory of ºBrix, titratable acidity, pH and dry matter. From the results obtained, it was found that there were no statistically significant differences between the modalities tested, but the tested formulation had phases more uniform and early phenological, as well as a harvest start slightly earlier and a higher number of fruits and size in treated plants. Overral, the values of the analytical determinations were numerically superior to the plants of control modality, however, it would be in the interest to do a new essay with the same formulation or with new adjustments and compare with a product different in order to confirm results.

Chlamydia trachomatis usually causes mucosal infections, bringing considerable morbidity and socioeconomic burden worldwide. We previously revealed that IL-27/IL-27R mediates protection against chlamydial invasion by promoting a protective Th1 response and suppressing neutrophilic inflammation. Here, we used the mouse model of Chlamydia muridarum (C. muridarum) respiratory infections to further investigate the impact of IL-27 signaling in the DCs-regulated immune response, since an elevated IL-27/IL-27R expression in DCs was identified following chlamydial infection. An adoptive transfer of Chlamydia muridarum-stimulated DCs to wild-type mice approach was subsequently used, and the donor-DCs-promoted resistance with a higher Th1 response against chlamydial infection was attenuated when DCs lacking IL-27R were used as donor cells. Flow cytometry analysis revealed the suppression of IL-27 signaling on DCs phenotypic maturation. A further functional maturation analysis of DCs revealed that IL-27 signaling restricted the protein and mRNA expression of IL-10 from DCs following infection. Thus, these findings suggest that IL-27 signaling could support the Th1 response via inhibiting IL-10 production in DCs, thus mediating the protective host defense against chlamydial respiratory infection.

The enzymes of microorganisms that live in cold environments must be able to function at ambient temperatures. Cold-adapted enzymes generally have less ordered structures that convey a higher catalytic rate, but at the cost of lower thermodynamic stability. In this study, we characterized P355, a novel intracellular subtilisin protease (ISP) derived from the genome of Planococcus halocryophilus Or1, which is a bacterium metabolically active down to −25°C. P355′s stability and activity at varying pH values, temperatures, and salt concentrations, as well as its temperature-dependent kinetics, were determined and compared to an uncharacterized thermophilic ISP (T0099) from Parageobacillus thermoglucosidasius, a previously characterized ISP (T0034) from Planococcus sp. AW02J18, and Subtilisin Carlsberg (SC). The results showed that P355 was the most heat-labile of these enzymes, closely followed by T0034. P355 and T0034 exhibited catalytic constants (kcat) that were much higher than those of T0099 and SC. Thus, both P355 and T0034 demonstrate the characteristics of the stability-activity trade-off that has been widely observed in cold-adapted proteases.

(1) Background: The selection of raw material and the postmortem processing of beef influence its quality, such as taste. In this study, the metabolome of beef from cows and heifers is examined for differences during aging. (2) Methods: Thirty strip loins from eight heifers and seven cows (breed code: 01–SBT) were cut into ten pieces and aged for 0, 7, 14, 21 and 28 days. Samples from the left strip loins were wet-aged in vacuum, while samples from right strip loins were dry-aged at 2 °C and 75% relative humidity. The beef samples were extracted with methanol–chloroform–water, and the polar fraction was used for 1H NMR analysis. (3) Results: The PCA and OPLS-DA showed that the metabolome of cows and heifers varied. Eight metabolites revealed significant differences (p < 0.05) in the samples from cows and heifers. The aging time and aging type of beef also affected the metabolome. Twenty-eight and 12 metabolites differed significantly (p < 0.05) with aging time and aging type, respectively. (4) Conclusions: The variations between cows and heifers and aging time affect the metabolome of beef. By comparison, the influence of aging type is present but less pronounced.

Introduction: Enamel mineralization requires calcium transport into the extracellular matrix for the synthesis of hydroxyapatite (HA) crystals. Formation of HA releases protons into the matrix, which are then neutralized when ameloblasts modulate from cells with apical invaginations, the so-called ruffle-ended ameloblasts (RE), to smooth-ended ameloblasts (SE). Ameloblast modulation is associated with the translocation of the calcium exchanger Nckx4 to the apical border of RE, to remove Na+ from the enamel matrix in exchange for Ca2+ and K+. As enamel matures, Na+ and K+ in the matrix progressively decrease. However, the transporter to remove K+ from mineralizing enamel has not been identified.Methods: Expression of K+ exchangers and channels in secretory and maturation stage of enamel organs were compared following an RNA-seq analysis. Kcnj15, which encodes the Kir4.2 inwardly rectifying K+ channel, was found to be the most upregulated internalizing K+ transporter in maturation stage of enamel organs. Kir4.2 was immunolocalized in wt, Nckx4−/−, Wdr72−/−, and fluorosed ameloblasts. Regulation of Wdr72 expression by pH was characterized in vitro and in vivo.Results: Kir4.2 immunolocalized to the apical border of wild type (wt) mouse RE and cytosol of SE, a spatial distribution pattern shared by NCKX4. In Nckx4−/− ameloblasts, Kir4.2 also localized to the apical surface of RE and cytosol of SE. However, in fluorosed and Wdr72−/− ameloblasts, in which vesicle trafficking is disrupted, Kir4.2 remained in the cytosol. In vitro, Wdr72 was upregulated in LS8 cells cultured in medium with a pH 6.2, which is the pH of the enamel matrix underlying RE, as compared to pH 7.2 under SE.Conclusion: Taken together these results suggest that Kir4.2 participates in K+ uptake by maturation ameloblasts, and that K+ and Na+ uptake by Kir4.2 and Nckx4, respectively, may be regulated by pH through WDR72-mediated endocytosis and membrane trafficking.

The effect produced by the application of methyl jasmonate (MeJA) in vineyards is clear, but this is a product that, despite its efficiency, is expensive, volatile and difficult to dissolve. Regarding increasing the MeJA use efficiency, new forms of application are proposed in this article, such as the use of calcium phosphate nanoparticles with two different morphologies: amorphous (ACP) and crystalline (Ap). In addition, few of the studies addressed so far have assessed MeJA’s effect during the ripening period of the grapes. As a result of this, in this article, we evaluated/studied for first time the effect of the different MeJA formats on the phenolic composition of the grape during the ripening period. The results showed small differences between the two morphologies of the nanoparticles, which promoted a significant a delay in the sugar accumulation and an increase in the different phenolic compounds compared to the control. Such improvements were not as significant as those induced by the conventional MeJA treatment. However, it is remarkable that when the nanoparticles were applied, we used a concentration 10 times lower than when it is used conventionally. Recent developments in clinical flow cytometry allow the simultaneous assessment of proliferative and anti-apoptotic activity in the different hematopoietic cell lineages and during their maturation process. This can further advance the flow cytometric diagnosis of myeloid malignancies. In this study we established indicative reference values for the Ki-67 proliferation index and Bcl-2 anti-apoptotic index in blast cells, as well as maturing erythroid, myeloid, and monocytic cells from normal bone marrow (BM). Furthermore, the cell fractions co-expressing both proliferation and anti-apoptotic markers were quantified. Fifty BM aspirates from femoral heads of patients undergoing hip replacement were included in this study. Ten-color/twelve-parameter flow cytometry in combination with a software-based maturation tool was used for immunophenotypic analysis of Ki-67 and Bcl-2 positive fractions during the erythro-, myelo-, and monopoiesis. Indicative reference values for the Ki-67 and Bcl-2 positive fractions were established for different relevant hematopoietic cell populations in healthy BM. Ki-67 and Bcl-2 were equally expressed in the total CD34 positive blast cell compartment and 30% of Ki-67 positive blast cells also showed Bcl-2 positivity. The Ki-67 and Bcl-2 positive fractions were highest in the more immature erythroid, myeloid and monocytic cells. Both fractions then gradually declined during the subsequent maturation phases of these cell lineages. We present a novel application of an earlier developed assay that allows the simultaneous determination of the Ki-67 proliferative and Bcl-2 anti-apoptotic indices in maturing hematopoietic cell populations of the BM. Their differential expression levels during the maturation process were in accordance with the demand and lifespan of these cell populations. The indicative reference values established in this study can act as a baseline for further cell biological and biomedical studies involving hematological malignancies.

Long noncoding RNA LINRIS (LINC00920) is known to participate in colorectal cancer. This study aimed to explore the role of LINRIS in non-small cell lung cancer (NSCLC). NSCLC and adjacent non-tumor tissues were collected from 62 NSCLC patients. LINRIS expression was detected using real-time quantitative PCR (RT-qPCRs). The 62 NSCLC patients were monitored every month for 5 years to evaluate the role of LINRIS in predicting the prognosis of NSCLC. The effects of LINRIS silencing on microRNA-10a (miR-10a) precursor and mature miR-10a levels were assessed by RT-qPCR. Cell proliferation was measured using Cell Counting Kit-8 (CCK-8) assays. LINRIS expression was upregulated in NSCLC tissues. High LINRIS levels predicted poor survival of NSCLC patients. LINRIS were positively correlated with mature (miR-10a) levels but not miR-10a precursor. In NSCLC cells, LINRIS silencing showed no role in miR-10a precursor accumulation but downregulated mature miR10a level. Moreover, LINRIS silencing inhibited cell proliferation, while miR-10a overexpression increased cell proliferation and inhibited the role of LINRIS silencing. Overall, LINRIS silencing may inhibit NSCLC cell proliferation by suppressing miR-10a maturation. Astrocytes are critical for the development and function of synapses. There are notable species differences between human astrocytes and commonly used animal models. Yet, it is unclear whether astrocytic genes involved in synaptic function are stable or exhibit dynamic changes associated with disease states and age in humans, which is a barrier in understanding human astrocyte biology and its potential involvement in neurologic diseases. To better understand the properties of human astrocytes, we acutely purified astrocytes from the cerebral cortices of over 40 humans across various ages, sexes, and disease states. We performed RNA sequencing to generate transcriptomic profiles of these astrocytes and identified genes associated with these biological variables. We found that human astrocytes in tumor-surrounding regions downregulate genes involved in synaptic function and sensing of signals in the microenvironment, suggesting involvement of peritumor astrocytes in tumor-associated neural circuit dysfunction. In aging, we also found downregulation of synaptic regulators and upregulation of markers of cytokine signaling, while in maturation we identified changes in ionic transport with implications for calcium signaling. In addition, we identified subtle sexual dimorphism in human cortical astrocytes, which has implications for observed sex differences across many neurologic disorders. Overall, genes involved in synaptic function exhibit dynamic changes in the peritumor microenvironment and aging. These data provide powerful new insights into human astrocyte biology in several biologically relevant states that will aid in generating novel testable hypotheses about homeostatic and reactive astrocytes in humans.SIGNIFICANCE STATEMENTAstrocytes are an abundant class of cells playing integral roles at synapses. Astrocyte dysfunction is implicated in a variety of human neurologic diseases. Yet our knowledge of astrocytes is largely based on mouse studies. Direct knowledge of human astrocyte biology remains limited. Here, we present transcriptomic profiles of human cortical astrocytes, and we identified molecular differences associated with age, sex, and disease state. We found that peritumor and aging astrocytes downregulate genes involved in astrocyte–synapse interactions. These data provide necessary insight into human astrocyte biology that will improve our understanding of human disease.

Summary The maturation and functional characteristics of human induced pluripotent stem cell (hiPSC)-cortical neurons has not been fully documented. This study developed a phenotypic model of hiPSC-derived cortical neurons, characterized their maturation process, and investigated its application for disease modeling with the integration of multi-electrode array (MEA) technology. Immunocytochemistry analysis indicated early-stage neurons (day 21) were simultaneously positive for both excitatory (vesicular glutamate transporter 1 [VGlut1]) and inhibitory (GABA) markers, while late-stage cultures (day 40) expressed solely VGlut1, indicating a purely excitatory phenotype without containing glial cells. This maturation process was further validated utilizing patch clamp and MEA analysis. Particularly, induced long-term potentiation (LTP) successfully persisted for 1 h in day 40 cultures, but only achieved LTP in the presence of the GABAA receptor antagonist picrotoxin in day 21 cultures. Chub mackerel, Scomber japonicus, is heavily farmed and harvested due to its demand as a high-quality protein source rich in fatty acids. However, the effects of environmental cues on sexual maturation of the fish remain understudied. We aim to elucidate the effect of light manipulation on the hormones related to reproduction and on the stress response in the species. Mackerel were exposed to different photoperiods (12 h light:12 h dark or 14 h light:10 h dark) and light wavelengths (provided by white fluorescent bulbs or green LEDs). Total RNA extracted from the brain was assayed with quantitative polymerase chain reaction (a powerful technique for advancing functional genomics) and blood plasma was analyzed via immunoassay using ELISA kits. The mRNA expression of gene-encoding gonadotropin-releasing hormone, gonadotropin hormone, follicle-stimulating hormone, and luteinizing hormone were significantly increased through the use of an extended photoperiod and green wavelength, which also increased testosterone and 17β-estradiol plasma levels. Plasma levels of cortisol and glucose, which are indicators of a stress response, were significantly decreased through green LED exposure. Our results indicate that environmental light conditions affect the production of pituitary and sex hormones, and reduce the stress response in S. japonicus.

Organisms utilize varying lipid resource allocation strategies as a means to survive seasonal environmental changes and life-history stage transitions. In Atlantic salmon, a certain lipid threshold is needed to initiate sexual maturation. Because of this, an individual’s maturation schedule may be affected by changes in temperature and food availability across the seasons that create natural fluctuations of lipid reserves.Recent studies have found a genome region, including the genevgll3,that explains a large proportion of variation for size and age at maturity. Vgll3encodes a transcription co-factor that acts as an inhibitor of adipogenesis in mice and also affects condition factor and other phenotypes in juvenile salmon. However, even with many studies investigating varying temperature effects, there is a lack of temporal studies examining the effects of seasonality on such phenotypes, nor have the effects ofvgll3genotype on condition factor and maturation in different temperatures at different life stages.Here, we investigate the influence of different larval and juvenile incubation temperatures,vgll3genotype and their interactions on juvenile salmon phenotypes including body condition, and sexual maturation rate. We reared Atlantic salmon for 2 years in varying temperatures with an average 1.76 °C difference between warm and cold treatments in four different larval-juvenile phase treatment groups (Warm-Warm, Warm-Cold, Cold-Warm, and Cold-Cold) until the first occurrence of maturation in males.We found no effect of larval temperature on the measured phenotypes or maturation rate, suggesting the occurrence of growth compensation over the course of the experiment. Agreeing with previous studies, an increased maturation rate was observed in individuals of the warm juvenile temperature treatment.In addition, we observed differences in condition factor associated withvgll3genotype, wherebyvgll3*EEindividuals (the genotype associated with early maturation) had a less variable condition factor across the seasons compared to thevgll3*LL(associated with late maturation) individuals.This result suggests avgll3influence on resource acquisition and allocation strategies, possibly linked with the early maturation process, with individuals carrying the early maturationvgll3genotype having a higher early maturation rate and a higher condition factor in the spring.

Introduction: Hydrocortisone is the standard of care in cortisol replacement therapy for congenital adrenal hyperplasia patients. Challenges in mimicking cortisol circadian rhythm and dosing individualization can be overcome by the support of mathematical modelling. Previously, a non-linear mixed-effects (NLME) model was developed based on clinical hydrocortisone pharmacokinetic (PK) pediatric and adult data. Additionally, a physiologically-based pharmacokinetic (PBPK) model was developed for adults and a pediatric model was obtained using maturation functions for relevant processes. In this work, a middle-out approach was applied. The aim was to investigate whether PBPK-derived maturation functions could provide a better description of hydrocortisone PK inter-individual variability when implemented in the NLME framework, with the goal of providing better individual predictions towards precision dosing at the patient level.Methods: Hydrocortisone PK data from 24 adrenal insufficiency pediatric patients and 30 adult healthy volunteers were used for NLME model development, while the PBPK model and maturation functions of clearance and cortisol binding globulin (CBG) were developed based on previous studies published in the literature.Results: Clearance (CL) estimates from both approaches were similar for children older than 1 year (CL/F increasing from around 150 L/h to 500 L/h), while CBG concentrations differed across the whole age range (CBGNLME stable around 0.5 μM vs. steady increase from 0.35 to 0.8 μM for CBG PBPK). PBPK-derived maturation functions were subsequently included in the NLME model. After inclusion of the maturation functions, none, a part of, or all parameters were re-estimated. However, the inclusion of CL and/or CBG maturation functions in the NLME model did not result in improved model performance for the CL maturation function (ΔOFV > −15.36) and the re-estimation of parameters using the CBG maturation function most often led to unstable models or individual CL prediction bias.Discussion: Three explanations for the observed discrepancies could be postulated, i) non-considered maturation of processes such as absorption or first-pass effect, ii) lack of patients between 1 and 12 months, iii) lack of correction of PBPK CL maturation functions derived from urinary concentration ratio data for the renal function relative to adults. These should be investigated in the future to determine how NLME and PBPK methods can work towards deriving insights into pediatric hydrocortisone PK.

Although aluminium-based vaccines have been used for almost over a century, their mechanism of action remains unclear. It is established that antigen adsorption to the adjuvant facilitates delivery of the antigen to immune cells at the injection site. To further increase our understanding of aluminium-based vaccines, it is important to gain additional insights on the interactions between the aluminium and antigens, including antigen distribution over the adjuvant particles. Immuno-assays can further help in this regard. In this paper, we evaluated how established formulation strategies (i.e., sequential, competitive, and separate antigen addition) applied to four different antigens and aluminium oxyhydroxide, lead to formulation changes over time. Results showed that all formulation samples were stable, and that no significant changes were observed in terms of physical-chemical properties. Antigen distribution across the bulk aluminium population, however, did show a maturation effect, with some initial dependence on the formulation approach and the antigen adsorption strength. Sequential and competitive approaches displayed similar results in terms of the homogeneity of antigen distribution across aluminium particles, while separately adsorbed antigens were initially more highly poly-dispersed. Nevertheless, the formulation sample prepared via separate adsorption also reached homogeneity according to each antigen adsorption strength. This study indicated that antigen distribution across aluminium particles is a dynamic feature that evolves over time, which is initially influenced by the formulation approach and the specific adsorption strength, but ultimately leads to homogeneous formulations.

The purpose of this study was to examine the presence of the relative age effect (RAE) in young tennis players. Data from a sample of all ranked (n= 3463) and licensed (n= 29150) players in consecutive age groups (10- to 17-year-old) were collected from the official web page of the national federation. They were assigned to four quartiles according to the month of birth: a) Quartile 1 = January-February-March, b) Quartile 2 = April-May-June, c) Quartile 3 = July-August-September, d) Quartile 4 = October-November-December. The players aged between 10 and 17 years affiliated with the national federation were assigned as theoretically expected distribution. The results revealed that 70%, 60.1%, and 56.6% of the top 10, 50, and all ranked players were born in the first half of the year, respectively. However, the results from the chi-square test of goodness-of-fit showed significant RAEs only in the distributions of all ranked and top 50 players at the age of 10 and 11 correspondingly. These results may suggest that RAE exists among young tennis players who are in their early adolescence years.

The aim of this study was to quantify and compare various external match load measures in three age groups and leagues in male soccer (U18 in highest league of their age group vs U21 in fourth highest league vs first team in highest league). In this retrospective observational cohort study accelerations, decelerations, absolute and relative high-speed running as well as sprint distance, dynamic stress load, explosive distance, high intensity bursts total distance, high metabolic load (HML) distance, speed intensity, total distance, total time, and total loading were assessed in 416 individual player matches of 59 players. All these external load measures showed large inter-individual variability. At a group level, one-way ANOVAs or Kruskal–Wallis tests revealed statistically significant differences between the three teams for all measures analyzed (all p < 0.05), except accelerations. The first team displayed statistically significant higher dynamic stress load, explosive distance, HML distance, speed intensity, total distance and total loading compared to the two youth teams (all p < 0.05). The U18 featured statistically significant higher number of decelerations, absolute and relative high-speed running distance, high metabolic load distance, speed intensity, relative sprint distance, total distance, and total time than the U21, while for U21 higher dynamic stress load was observed than for U18 (all p < 0.05). Based on our data we conclude a routinely monitoring of match loads of different age groups and competitive settings to be required to 1) provide an indication of what players need to be prepared for, 2) track the athletic and match evolution, and 3) individually tailor training programs allowing players to fulfill the short- and long-term sport-specific requirements.

Ischemic heart failure is due to irreversible loss of cardiomyocytes. Preclinical studies showed that human pluripotent stem cell (hPSC)-derived cardiomyocytes could remuscularize infarcted hearts and improve cardiac function. However, these cardiomyocytes remained immature. Incorporating hPSC-derived epicardial cells has been shown to improve cardiomyocyte maturation, but the exact mechanisms are unknown. We posited epicardial fibronectin (FN1) as a mediator of epicardial-cardiomyocyte crosstalk and assessed its role in driving hPSC-derived cardiomyocyte maturation in 3D-engineered heart tissues (3D-EHTs). We found that the loss of FN1 with peptide inhibition F(pUR4), CRISPR-Cas9-mediated FN1 knockout, or tetracycline-inducible FN1 knockdown in 3D-EHTs resulted in immature cardiomyocytes with decreased contractile function, and inefficient Ca2+ handling. Conversely, when we supplemented 3D-EHTs with recombinant human FN1, we could recover hPSC-derived cardiomyocyte maturation. Finally, our RNA-sequencing analyses found FN1 within a wider paracrine network of epicardial-cardiomyocyte crosstalk, thus solidifying FN1 as a key driver of hPSC-derived cardiomyocyte maturation in 3D-EHTs.

A cobertura do solo com material inorgânico é uma técnica muito utilizada em algumas culturas criando um microclima no local a fim de melhorar a qualidade dos produtos. Porém, cada cultura tem um efeito diferente conforme o clima, a localização e o manejo. Sendo assim, o objetivo deste trabalho foi cobrir o solo com tecido de diferentes cores em uvas Tannat a fim de melhorar a qualidade da fruta e do vinho elaborado. O delineamento experimental foi inteiramente casualizado, onde as plantas foram cobertas no período de maturação das uvas, composto por quatro tratamentos e quatro repetições: cobertura do solo com tecido-não-tecido (TNT) nas cores azul, amarela e vermelha e o tratamento testemunha sem cobertura. Após a colheita, foram realizadas as análises físicas das uvas e químicas do mosto, vinificação, e posteriormente a análise química do vinho, os dados foram submetidos ao teste de Tukey a 5% de significância. Conforme os resultados encontrados, podemos observar que a cobertura do solo com TNT amarelo proporcionou menor tonalidade de cor do mosto, o que é mais desejável, e influenciou na maior graduação alcoólica do vinho. Já a o TNT azul intensificou a tonalidade do mosto. Portanto, as diferentes cores em cobertura do solo podem modificar algumas características da uva e do vinho Tannat. Covering the soil with plastics is a technique widely used in some cultures, creating a microclimate on site to improve product quality. However, each culture has a different effect depending on the climate, location and management. Therefore, the objective of this work was to cover the soil with fabric of different colors in Tannat grapes in order to improve the quality of the fruit and the elaborated wine. The experimental design was completely randomized, where the plants were covered during the maturation period of the grapes, consisting of four treatments and four repetitions: soil cover with non-woven fabric (TNT) in blue, yellow colors and red and the witnesses without coverage. After the harvest, physical analyzes of the grapes and chemical analysis of the must, vinification, and later the chemical analysis of the wine were carried out. The data obtained were submitted to the Tukey test at 5% significance level. According to the results found, can observe that the ground cover with blue TNT provided a greater color shade of the must and a lower pH. The yellow soil cover, in turn, influenced the higher alcohol content of wine. Therefore, the different colors in soil cover can modify the quality of the Tannat grape and wine. Sin embargo, cada cultivo tiene un efecto diferente según el clima, la ubicación y el manejo. Por lo tanto, el objetivo de este trabajo fue cubrir el suelo con tela de diferentes colores en uva Tannat con el fin de mejorar la calidad de la fruta y el vino producido. El diseño experimental fue completamente al azar, donde las plantas fueron cubiertas durante el período de maduración de la uva, constando de cuatro tratamientos y cuatro repeticiones: cubierta de suelo con tela no tejida (TNT) en colores azul, amarillo y rojo y el tratamiento control sin cobertura. Después de la cosecha, el análisis físico de las uvas y el análisis químico del mosto, vinificación y posteriormente el análisis químico del vino, los datos fueron sometidos a la prueba de Tukey al 5% de significación. De acuerdo con los resultados encontrados, podemos observar que cubrir el suelo con TNT amarillo proporcionó una menor tonalidad de color del mosto, que es más deseable, e influyó en el mayor grado alcohólico del vino. El TNT azul intensificó la tonalidad del mosto. Por lo tanto, los diferentes colores en la cobertura del suelo pueden modificar algunas características de la uva y del vino Tannat.

Grapevine (Vitis vinifera L.) is one of the most widely cultivated fruit crops because the winemaking industry has huge economic relevance worldwide. Uncovering the molecular mechanisms controlling the developmental progression of plant organs will prove essential for maintaining high-quality grapes, expressly in the context of climate change, which impairs the ripening process. Through a deep inspection of transcriptomic data, we identified VviNAC60, a member of the NAC transcription factor family, as a putative regulator of grapevine organ maturation. We explored VviNAC60 binding landscapes through DNA affinity purification followed by sequencing and compared bound genes with transcriptomics datasets from grapevine plants stably and transiently overexpressing VviNAC60 to define a set of high-confidence targets. Among these, we identified key molecular markers associated with organ senescence and fruit ripening. Physiological, metabolic, and promoter activation analyses showed that VviNAC60 induces chlorophyll degradation and anthocyanin accumulation through the upregulation of STAY-GREEN PROTEIN 1 (VviSGR1) and VviMYBA1, respectively, with the latter being upregulated through a VviNAC60–VviNAC03 regulatory complex. Despite sharing a closer phylogenetic relationship with senescence-related homologs to the NAC transcription factor AtNAP, VviNAC60 complemented the nonripening(nor) mutant phenotype in tomato (Solanum lycopersicum), suggesting a dual role as an orchestrator of both ripening- and senescence-related processes. Our data support VviNAC60 as a regulator of processes initiated in the grapevine vegetative- to mature-phase organ transition and therefore as a potential target for enhancing the environmental resilience of grapevine by fine-tuning the duration of the vegetative phase.

Diamondoid hydrocarbons were investigated in gas-condensates reservoirs from the Drava Depression, Croatian part of the Pannonian Basin, to define thermal maturity of condensates. The largest gas and gas-condensate fields in Croatia are Molve, Kalinovac, and Stari Gradac, located in the northwestern part of the Drava Depression. This area has extremely complex structural, lithological, and stratigraphic features that have resulted in the specific physical characteristics of the discovered hydrocarbon fluids. Bulk properties reveal that samples have high concentration of light aromatic hydrocarbon (BTEX) and low sulphur, and some of them are waxy; Stari Gradac and Severovci. Origin and maturity parameters of condensate were investigated and correlated based on gas chromatography (GC) and gas chromatography coupled to mass spectrometry (GC-MS) techniques. The value of diamondoid based maturity parameters, like methydiamantane index 40–50 % indicate high level of thermal maturity corresponding to vitrinite reflectance 1.1–1.6 %. Condensate samples were not biodegraded, as shown by high API density values and the absence of unresolved complex mixture (UCM)., Diamantoidni ugljikovodici ispitivani su u plinsko-kondenzatnim ležištima Dravske depresije, hrvatski dio Panonskog bazena, da bi se odredila termička zrelost kondenzata. Osnovna svojstva pokazuju visoku koncentraciju laganih aromatskih ugljikovodika (BTEX) i nisku koncentraciju sumpora, a neki od njih sadrže visok udio krutih parafina; Stari Gradac i Severovci. Parametri porijekla i zrelosti kondenzata karakterizirani su i korelirani metodama plinske kromatografije (GC) i vezanim sustavom plinska kromatografija – masena spektrometrija (GC-MS). Parametar zrelosti na bazi dijamantoida, poput metildiamantanskog indeksa 40–50 % ukazuje na visoku razinu termičke zrelosti koja odgovara refleksiji vitrinita 1,1–1,6 %. Uzorci kondenzata nisu biodegradirani, što pokazuju visoke vrijednosti API gustoće i odsutnost kromatografski nerazdvojenih spojeva (engl. unresolved complex mixture, UCM).

This study presents data on the seasonal changes in population structure, maturation, fecundity and reproduction of the Lepidorhombus boscii from the central-eastern Adriatic Sea. This species is a commercially valuable fish in the Adriatic Sea; still, knowledge about its reproductive biology is very scarce. The analyses are based on the data collected between July 2020 and June 2021 by bottom trawls. In total, 963 individuals were collected out of which 508 were females (47.24%) and 455 were males (52.75%). Throughout the year, oocytes in the previtellogenic and vitellogenic stages were present in the ovaries, and the development of the ovaries followed the values of the gonadosomatic index. L. boscii has asynchronous ovarian development and a prolonged spawning season in the Adriatic Sea with the main spawning season between November and March. The oocyte diameter ranged from 34.01 to 562.72 μm (178.49 ± 129.83 μm). This study presents the first information on the reproductive cycle of L. boscii in the eastern Adriatic Sea and contributes to the understanding of the biology of this species necessary for its sustainable management.

Cumulus cells in the ovarian follicles emerge by differentiation from somatic granulosa cells. They are located around the oocytes in stratified clusters and their innermost layer is called the corona radiata. Cytoplasmic microvilli of cumulus cells pass through zona pellucida to the proximity of the oolema, where formation of gap junctions enables signalling and metabolic codependency. Oocytes produce factors affecting processes in cumulus cells, including their metabolism and viability. By modulating the abundance of cyclic nucleotides in oocytes, cumulus cells allow the maintenance of meiotic arrest, providing time for oocytes to finish their growth and gain competence for maturation. Because oocytes have a very limited ability to process glucose, metabolic cooperation with cumulus cells enables them to gain the energy needed for processes associated with growth and maturation. Several metabolic pathways of glucose processing create energy substrates for the formation of ATP, which they then transport to the oocytes. They also regulate the amount of lipid droplets in oocytes and protect them from cellular toxicity. Biological maturity can affect performance on motor tests, thus young players can have advantages or disadvantages during testing by being more or less mature than their peers of the same chronological age. The aim of this study was to investigate the effects of biological age on speed, agility, and explosive power in young tennis players. Fifty tennis players (age 12.3 ± 1.2 years, height 156.7 ± 12.8 cm, body mass 45.9 ± 8.9 kg) who were ranked up to 50th place in the ranking of the National Tennis Association, as well as up to 300th place on the international “Tennis Europe” ranking, participated in the study. They were divided into three groups according to their maturation status, that is, the peak height velocity (PHV) maturity offset: pre-PHV [1.5] (n = 25). They performed tests of speed (5, 10, and 20 m sprints), agility (20 yards, 4 × 10 yards, T-test, TENCODS, and TENRAG), and explosive power (countermovement jump, one-leg countermovement jump, squat jump, long jump, and one-leg triple jump). Results showed significantly higher height of the vertical jump in the post-PHV group compared to the pre-PHV group, in the range of ~16% to ~27%. Moreover, linear and change of direction speed was significantly faster in the post-PHV group compared to the pre-PHV group, in the range of ~5% to ~8%. Height of the squat jump and speed in the T-test were significantly better in the post-PHV group compared to the circa-PHV group, in the range of ~7% to ~15%, while height of the single-leg triple jump was significantly higher in the circa-PHV group compared to the pre-PHV group by ~7%. This study showed that tennis players of older biological age achieve better results in almost all variables of speed, agility, and explosive power compared to players of younger biological age. Coaches should be aware of the differences found in physical performance and consider the practical implications that maturation can have in the long-term development of young tennis players.

The application of a no-touch technique to create an autologous radiocephalic arteriovenous fistula might improve the patency rate. In the present report, we have expanded the concept of the no-touch technique by introducing a modified no-touch technique in which we preserve the perivenous vascular tissue, followed by a functional end-to-side anastomosis to create a radiocephalic arteriovenous fistula with early maturation for hemodialysis.

Gene expression programs and concomitant chromatin regulation change dramatically during the maturation of postmitotic neurons. Subnuclear positioning of gene loci is relevant to transcriptional regulation. However, little is known about subnuclear genome positioning in neuronal maturation. Using cultured murine hippocampal neurons, we found genomic locus 14qD2 to be enriched with genes that are upregulated during neuronal maturation. Reportedly, the locus is homologous to human 8p21.3, which has been extensively studied in neuropsychiatry and neurodegenerative diseases. Mapping of the 14qD2 locus in the nucleus revealed that it was relocated from the nuclear periphery to the interior. Moreover, we found a concomitant decrease in lamin B1 expression. Overexpression of lamin B1 in neurons using a lentiviral vector prevented the relocation of the 14qD2 locus and repressed the transcription of the Egr3 gene on this locus. Taken together, our results suggest that reduced lamin B1 expression during the maturation of neurons is important for appropriate subnuclear positioning of the genome and transcriptional programs.

HCC patients were followed up for 5 years to analyze the prognostic value of DNAJC3-AS1 for HCC. The direct interaction between DNAJC3-AS1 and premature miR-27b was analyzed with RNA pull-down assay. Subcellular analysis of DNAJC3-AS1 was explored by subcellular fractionation assay. DNAJC3-AS1 overexpression and knockdown were carried out to analyze the role of DNAJC3-AS1 in miR-27b maturation. Cell proliferation was analyzed by BrdU assay.Results: DNAJC3-AS1 was overexpressed in HCC and predicts the poor survival. MiR-27b was downregulated at mature miRNA level, but upregulated at premature level. DNAJC3-AS1 directly interacted with premature miR-27b and was localized to both nuclear and cytoplasm. DNAJC3-AS1 overexpression upregulated premature miR-27b and downregulated mature miR-27b, while DNAJC3-AS1 knockdown led to the opposite results. DNAJC3-AS1 suppressed the role of miR-27b in inhibiting cell proliferation.Conclusion: DNAJC3-AS1 promotes HCC by sponging premature miR-27b and might be a biomarker and therapeutic target for HCC.Keywords: DNAJC3-AS1, miR-27b, hepatocellular carcinoma, maturation

Like in other sports systems, in kayaking, young athletes are commonly grouped by their birth years. This study analyzed maturity status, relative age, and constituent age effects in young Under 14 (U14) and 16 (U16) Iberian male kayakers. One hundred and thirty (U14: n =80; U16: n = 50) young kayakers aged 14.10 ± 1.06 years were assessed for anthropometry, performance, maturity, and sport experience. The year was divided into four birth quarters (BQ). There were no significant differences in the kayaker’s distribution by BQ in both categories (U14, p = 0.348; U16, p = 0.709) or total sample (p = 0.783). Six of the ten best kayakers in the U14 category were born in the year’s first half, and eight were among the U16 kayakers. Talent detection and selection systems based solely on the young kayakers’ performances may imply some bias. One of the ways to get around this situation would be to adopt a grouping system similar to bio-banding, allowing the maximum number of practitioners to experience the possibility of obtaining competitive success.

The champa (Campomanesia lineatifolia) is a very perishability berry with a pleasant taste. The objective of this research was to study the regulation and prolongation of the postharvest conservation of champa fruits using 1-methylcyclopropene (1-MCP) and refrigeration. Two experiments were carried out: In experiment 1, four treatments were evaluated to understand the regulation of maturation. The treatments were: control, 1-MCP, ethylene and 1-MCP + ethylene. The fruits were stored at room temperature. In experiment 2, the combination of the application of 1-MCP and two storage temperatures (room temperature [16±2°C] as well as 2±0.4°C) were used. In the two experiments, four repetitions were used, each one with approximately 500 g of fruits harvested at stage 2 of maturity (25% yellow and 75% green). The results of experiment 1 indicated that the fruits with 1-MCP presented the longest postharvest duration (17 days), firmness, and titratable acidity. They also had the lowest respiratory rate and weight loss during the 10 days. However, the fruits treated with ethylene had the opposite behavior, indicating that the changes evaluated during ripening were associated with this hormone. In experiment 2, the fruits refrigerated at 2°C, with or without the application of 1-MCP, had the longest postharvest duration, lasting 26 days, as compared to 12 days in the control fruits (without refrigeration and with 1-MCP). The fruits that lasted longer had a low respiratory rate during storage, as well as lower values for weight loss and, color index but, greater firmness and acidity. The 1-MCP was efficient when the fruits were at room temperature. RESUMEN La champa (Campomanesia lineatifolia) es una baya con sabor agradable y muy perecedera. El objetivo de este trabajo fue estudiar la regulación y prolongación de la vida poscosecha de los frutos de champa mediante el uso de 1-metilciclopropeno (1-MCP) y la refrigeración. Se realizaron dos experimentos, en el experimento 1 se evaluaron cuatro tratamientos para entender la regulación de la maduración, estos fueron: testigo, 1-MCP, etileno y 1-MCP+etileno, los frutos fueron almacenados a temperatura ambiente. En el experimento 2 se realizó la combinación de la aplicación o no de 1-MCP y dos temperaturas de almacenamiento (ambiente [16±2°C] y 2±0,4°C). En los dos experimentos se utilizaron cuatro repeticiones, cada una con aproximadamente 500 g de frutos cosechados en estado 2 de madurez (25% amarillos, 75%verdes). Los resultados del experimento 1 indicaron que los frutos con 1-MCP presentaron la mayor duración poscosecha (17 días), firmeza, acidez titulable, también la menor tasa respiratoria y pérdida de peso durante los 10 días que estuvieron los frutos de todos los tratamientos, los frutos tratados con etileno tuvieron el comportamiento contrario, indicando que los cambios durante la maduración evaluados están asociados a esta hormona. En el experimento 2, los frutos refrigerados a 2°C con o sin aplicación de 1-MCP tuvieron la mayor duración poscosecha con 26 días, en comparación con 12 días que duraron los frutos testigo (sin refrigeración y son 1-MCP), estos mismos frutos con mayor duración presentaron una muy baja tasa respiratoria durante el almacenamiento, menores valores de la pérdida de peso, menor índice de color, mayor firmeza y acidez. También se encontró que el 1-MCP es eficiente cuando los frutos están a temperatura ambiente.

The objective of this research was to carry out an analysis of growth and physicochemical changes in cv. Anna apple in the Colombian high-altitude tropics using on the accumulation of growing degree days (GDD). Fruit samplings were taken every 15 days after anthesis (DAA) until harvest at 100 DAA (892.37 GDD). The dry and fresh weight and the equatorial and polar diameters followed a simple sigmoidal pattern. This was confirmed with the behavior of the growth rates. The equatorial and polar diameters increased drastically between 455.39 and 589.32 GDD (45 and 60 DAA), while the weight did not, indicating that the void spaces increased in the pulp during this period. The respiratory rate had the highest value (61.93±6.79 mg CO2 kg-1 h-1) at 159.61 GDD (15 DAA) and then decreased continuously until harvest. The firmness increased from 159.61 to 455.39 GDD and, then continuously decreased, at harvest, it was 38.38±3.48 N. The total soluble solids increased and had an ending value of 8.58±0.37ºBrix. The total titratable acidity increased from 159.61 to 319.79 GDD (30 DAA), and then decreased until the end of the study with an acidity of 0.71±0.03%. Objectives The purpose of the present study was two-fold: (1) To analyse physical fitness changes of youth football players after a full-season; and (2) to examine whether physical fitness changes are explainable by estimated maturity status, 2digit:4digit ratio (2D:4D) from each hand and training load (TL) measures. Methods Twenty-seven youth elite Under-15 football players were daily monitored for training load measures during 38 weeks. At the beginning and at the end of the season, all players were assessed for physical fitness. Also, the maturity status estimation and the length of the second and fourth digits of both hands were collected at the beginning of the season. Results Significant differences were found for all physical fitness measures after the season. The second and fourth digits of left and right hands had negative moderate correlations with change of direction (COD) changes (r=-.39 to − 0.45 | p = .05 to 0.02). Also, the maturity offset measure had negative moderate correlations with COD changes (r=-.40 | p = .04). From the reported significant correlations, the maturity offset, Left 4D, Right 2D and Right 4D significantly predicted the Mod.505 COD test changes (β = 0.41, p = .04; β = -0.41, p = .04; β = -0.45, p = .02; and β = -0.44, p = .03, respectively). Conclusion The maturity offset and the 2D:4D measures have the potential to predict COD performance changes over-time in youth football players. Given the lack of associations between the maturity estimation, 2D:4D and training load measures, with the overall physical fitness measures, coaches should rely only at COD changes.

The article is focused on the behaviour of the basic properties of dairy cow's solid manure over the maturation period. The manure was exposed to mechanized aerobic processing by mechanically turning-over. Performed analysis comprehended measurements of mass participation percentages of organic components, mineral matters, H2O, N, P2O5 and K2O, as well as measurements of PH-values during the seven weeks of supervised test period of effective aerobic preparation of the composting material. Through maturation, large growth of averaged mass percentages of nitrogen, phosphorous-pentoxide and potassium-oxide, more than triple in comparison to the initial levels in a fresh solid, has been verified. Following a possible common approach in the scientific and technical practice, an analytical semi-empirical approach was used in describing the changes over the time of manure properties during composting, supported by prototype of the machine Kompomat 1. Combining the analytical theory of diffusion partial differential equations and application of numerical methods to non-linear fitting of the experimental output data, it was verified that all acquired manure parameters follow an identical general exponential law (R2 all properties = 0,906-0,990; RMSEall properties = 0,010-2,544). Clear relationships between the values of basic parameters of solid manure under mechanized aerobic fermentation (y) and composting duration lap time (x) have been established. © 2022, Strojarski Facultet. Among fishery biologists and even ichthyologists, maturation and spawning of fish are viewed as processes that use “energy” that would otherwise be applied to somatic growth, which is supposed to explain why post-maturity growth in length tends to decline. This widespread conceptualization may be called the “reproductive drain hypothesis” (RDH). However, when growth is correctly viewed as involving body mass, and is thus expressed in weight, post-maturity turns out (in iteroparous bony fish whose maximum length exceeds 10 cm) to accelerate after first maturity, despite its energy cost. This, and other common observations flatly contradict the RDH, and the time has come to withdraw this hypothesis. As a contribution towards this task, we propose an alternative reconceptualization of fish spawning consistent with what is known about fish biology.&nbsp, Los biólogos pesqueros e incluso los ictiólogos tienden a ver la maduración y el desove de los peces como procesos que usan “energía” que, de otro modo, sería aplicada al crecimiento somático, lo cual se supone que explica por qué el crecimiento post-maduración, en lo que se refiere a longitud, tiende a declinar. Esta popular conceptualización puede ser denominada “Hipótesis del Drenaje Reproductivo” (RDH). Sin embargo, cuando el crecimiento es visto de manera correcta como un proceso que involucra el volumen o masa corporal y, por lo tanto, se expresa en peso, la post-maduración (en peces óseos iteróparos cuya longitud máxima supera los 10 cm) se acelera luego de la primera maduración, a pesar de su costo energético. In recalcitrant woody species, such as chestnut (Castanea sativa Mill.), one of the major maturation-related shifts is the loss of the ability to form adventitious roots in response to auxin treatment as the plant ages. To analyze the molecular mechanisms underlying this phenomenon, an in vitro model system of two different lines of microshoots derived from the same field-grown tree was established. While juvenile-like shoots root readily when treated with exogenous auxin, microshoots established from the crown of the tree rarely form roots. In the present study, a transcriptomic analysis was developed to compare the gene expression patterns in both types of shoots 24 h after hormone and wounding treatment, matching the induction phase of the process. Our results support the hypothesis that the inability of adult chestnut tissues to respond to the inductive treatment relies in a deep change of gene expression imposed by maturation that results in a significant transcriptome modification. Differences in phytohormone signaling seem to be the main cause for the recalcitrant behavior of mature shoots, with abscisic acid and ethylene negatively influencing the rooting ability of the chestnut plants. We have identified a set of related MADS-box genes whose expression is modified but not suppressed by the inductive treatment in mature shoots, suggesting a putative link of their activity with the rooting-recalcitrant behavior of this material. NK cells are innate lymphocytes involved in a large variety of contexts and are crucial in the immunity to intracellular pathogens as well as cancer due to their ability to kill infected or malignant cells. Thus, they harbor a strong potential for clinical and therapeutic use. NK cells do not require antigen exposure to get activated; their functional response is rather based on a balance between inhibitory/activating signals and on the diversity of germline-encoded receptors they express. In order to reach optimal functional status, NK cells go through a step-wise development in the bone marrow before their egress, and dissemination into peripheral organs via the circulation. In this review, we summarize bone marrow NK cell developmental stages and list key factors involved in their differentiation before presenting newly discovered and emerging factors that regulate NK cell central and peripheral maturation. Lastly, we focus on the impact inflammatory contexts themselves can have on NK cell development and functional maturation.

Picea pungens (Engelm.), known for its blue-green needles, has become a likable ornamental species in northeast China since 2000. Nonetheless, a lack of propagation methods that can maintain genetic fidelity and develop seedlings at a large scale prevents the further expansion of the species. Somatic embryogenesis (SE), paired with cryopreservation technologies, may provide a valid alternative. Picea pungens SE is not new, but its practical application has been limited due to low efficiencies in SE initiation and maturation as well as a lack of effective cryopreservation technology. In this study, experiments were carried out to overcome the limitations by modifying culture media. For initiation, the efficiency was enhanced by adjusting concentrations of 2.4-dichlorophenoxy acetic acid (2,4-D), 6-benzyl amino–purine (6-BA) or sucrose supplemented to the induction medium. The concentrations of 4.0 mg/L 2,4-D, 2 mg/L 6-BA, and 5 to 10 g/L sucrose were found optimal in maximizing initiation efficiency. For maturation, the efficiency, expressed as the number of mature somatic embryos per gram of fresh mass cultured (E/gFM), varied greatly with the choices of the basal medium and concentration of abscisic acid (ABA) of the maturation medium. Based on our results, the judicial choices were using the DCR medium as the basal medium and 10 mg/L ABA. The maturation efficiency could also be improved by adjusting the maturation medium’s osmotic pressure by manipulating the concentrations of carbohydrate and Gelrite and culture density. Background Human induced pluripotent stem cell (iPSC)-derived cardiomyocytes (iPSC-CMs) do not display all hallmarks of mature primary cardiomyocytes, especially the ability to use fatty acids (FA) as an energy source, containing high mitochondrial mass, presenting binucleation and increased DNA content per nuclei (polyploidism), and synchronized electrical conduction. This immaturity represents a bottleneck to their application in (1) disease modelling—as most cardiac (genetic) diseases have a middle-age onset—and (2) clinically relevant models, where integration and functional coupling are key. So far, several methods have been reported to enhance iPSC-CM maturation; however, these protocols are laborious, costly, and not easily scalable. Therefore, we developed a simple, low-cost, and rapid protocol to promote cardiomyocyte maturation using two small molecule activators of the peroxisome proliferator-activated receptor β/δ and gamma coactivator 1-alpha (PPAR/PGC-1α) pathway: asiatic acid (AA) and GW501516 (GW). Methods and Results Monolayers of iPSC-CMs were incubated with AA or GW every other day for ten days resulting in increased expression of FA metabolism-related genes and markers for mitochondrial activity. AA-treated iPSC-CMs responsiveness to the mitochondrial respiratory chain inhibitors increased and exhibited higher flexibility in substrate utilization. Additionally, structural maturity improved after treatment as demonstrated by an increase in mRNA expression of sarcomeric-related genes and higher nuclear polyploidy in AA-treated samples. Furthermore, treatment led to increased ion channel gene expression and protein levels. Conclusions Collectively, we developed a fast, easy, and economical method to induce iPSC-CMs maturation via PPAR/PGC-1α activation. Treatment with AA or GW led to increased metabolic, structural, functional, and electrophysiological maturation, evaluated using a multiparametric quality assessment.
