A pesquisa busca aprofundar as reflexões sobre Patrimônio Cultural através da análise das redes em torno do inventariamento dos bens culturais das Comunidades Quilombolas do Vale do Ribeira - São Paulo, relacionando de maneira cruzada estudos históricos e antropológicos sobre o território e conflitos sociais. Visando construir conhecimento colaborativo com os grupos locais e o terceiro setor na região, investiga a importância das significações à política de preservação do seu patrimônio, pauta sobre a relação com a titulação do território quilombola e identifica possíveis desafios da aplicabilidade da metodologia de referenciamento. This research seeks to deepen reflections about the Cultural Heritage by reviewing the networks around inventory of cultural goods of Quilombola Commnunities in Vale do Ribeira, São Paulo, relating historical and anthropological studies on land and social conflicts. Aiming to create collaborative knowledge with local groups and the third sector in the region, this project investigates the importance of cultural meanings for the policy of preserving their heritage. It also addresses the issue about the relationship with the quilombola territorial naming and identifies and potential challenges to the applicability of the referencing methodology. Die mechanischen Eigenschaften lebender Zellen sind grundlegend für biologische Prozesse, wie Wundheilung oder die Embryogenese. Zellen zeigen eine komplexe, nichtlineare mechanische Antwort, die sich sowohl als zunehmende als auch als abnehmende Steifigkeit äußern kann. Die auftretenden Nichtlinearitäten in der mechanischen Zellantwort und im besonderen ihre Abhängig-keit von Stimulus und Messmethode sind noch nicht verstanden. Im Rahmen dieser Arbeit wurde die nichtlineare mechanische Antwort lebender Fibroblasten auf oszillatorische Stimuli mit stei-gender Amplitude charakterisiert. Die Resultate zeigen, dass die nichtlineare Zellantwort von der Verformungsgeschichte abhängt. Je nach Vorgeschichte zeigen lebende Zellen unterschiedliche Antworten auf den gleichen oszillatorischen Stimulus. Intrazyklische Versteifungen und interzy-klisch abnehmende Steifigkeiten können im gleichen Experiment beobachtet werden. Analogien zu vernetzten Biopolymergelen legen eine inelastische Umordnung des Zytoskeletts und eine re-sultierende pfadabhängige mechanische Adaption der Zellen als Grundlage der Abhängigkeit der Zellantwort von der Verformungsgeschichte nahe. Eine Modellierung der Ergebnisse mit einem elastoplastischen Kontinuumsmodell und einem dynamischen Filamentmodell unterstützt das mi-kroskopische Modell der inelastischen Umordnung. Adaptionen des Zytoskeletts können eine wich-tige Rolle für einzelne Zellen, Gewebe und Organismen spielen. The mechanical properties of living cells are a major determinant for biological processes, such as wound healing and embryogenesis. Cells exhibit a complex nonlinear mechanical response, including stiffening, softening and an adaptation of cytoskeletal structure. Nonlinearities in the cell mechanical response and especially their dependence on the stimulus and the experimental technique are not yet understood. In this thesis, the nonlinear response of living fibroblast cells is characterized using large amplitude oscillatory shear. The results show that nonlinear cell re-sponse depends on the strain history. Cells can exhibit different mechanical responses to identical oscillatory excitations. Fibroblast cells exhibit a superposition of intercycle softening and intra-cycle stiffening. Analogies to experimental and theoretical work on crosslinked biopolymer gels suggests that inelastic rearrangements of the cytoskeleton and resulting path-dependent mecha-nical adaptations are responsible for the strain history dependence. Modeling approaches using an elastoplastic continuum model and a dynamic, crosslinked filament model support the microscopic picture of inelastic rearrangements. Adaptations of the cytoskeleton can have an important role for single cells, tissues as well as whole organisms.

Die Akutfolgen ionisierender Strahlung wurden in zahlreichen Studien gut untersucht, ein Zusammenhang zwischen Hochdosisbestrahlung und deterministischen Spätschäden wurde vielfach belegt. Über die Langzeitfolgen niedrig dosierter ionisierender Strahlung hingegen ist bislang weniger bekannt, obwohl epidemiologische Studien auf ein erhöhtes Risiko strahlenassoziierter Spätfolgen hinweisen, sogar bei sehr geringen Dosen. Ionisierende Strahlung bedingt dosis- und zeitabhängigen Veränderungen in zahlreichen Geweben. Mittlere bis hohe Strahlendosen führen unter anderem zur strahleninduzierten koronaren Herzkrankheit mit ihren vielfältigen Folgeerkrankungen. Die vorliegende Studie beschäftigt sich mit den inflammatorischen, thrombotischen und fibrotischen Spätfolgen des murinen ApoE-/- -Herzens 3 bzw. 6 Monate nach Niedrig-Dosis-Bestrahlung. Dazu wurde die Expression ausgewählter Markerproteine an gefroren Herzschnitten und Plasmaproben mittels Immunfluoreszenzanalyse bzw. ELISA quantifiziert. Es konnte gezeigt werden, dass bereits sehr niedrige Bestrahlungsdosen (0,025 – 0,5 Gy) zu kompensatorischen Langzeiteffekten wie beispielsweise einer erhöhte Kapillardichte und Änderungen von Kollagen-IV und Thy-1-Expression führen. Dabei finden sich sowohl pro- als auch antiinflammatorische Effekte. Aus den Erkenntnissen multipler inflammatorischer und thrombotischer Veränderungen nach Niedrigdosisbestrahlung ließen sich möglicherweise wichtige Rationalen der anti-inflammatorischen Strahlentherapie ableiten. Darüber hinaus könnte anhand systemischer Plasmamarker die individuelle Evaluation strahlenassoziierter Spätfolgen und Einleitung adäquater Interventionsstrategien ermöglicht werden. Ob die hier gezeigten Effekte nur in Patienten mit besonderem Risikoprofil, wie erhöhten Cholesterin-Spiegeln, oder auch metabolisch gesunden relevant sind, bleibt weiteren Untersuchungen vorbehalten.

Epidemiological studies indicate long-term risks of ionizing radiation on the heart, even at moderate doses. In this study, we investigated the inflammatory, thrombotic and fibrotic late responses of the heart after low-dose irradiation (IR) with specific emphasize on the dose rate. Hypercholesterolemic ApoE-deficient mice were sacrificed 3 and 6 months after total body irradiation (TBI) with 0.025, 0.05, 0.1, 0.5 or 2 Gy at low (1 mGy/min) or high dose rate (150 mGy/min). The expression of inflammatory and thrombotic markers was quantified in frozen heart sections (CD31, E-selectin, thrombomodulin, ICAM-1, VCAM-1, collagen IV, Thy-1, and CD45) and in plasma samples (IL6, KC, MCP-1, TNFα, INFγ, IL-1β, TGFβ, INFγ, IL-10, sICAM-1, sE-selectin, sVCAM-1 and fibrinogen) by fluorescence analysis and ELISA. We found that even very low irradiation doses induced adaptive late responses, such as increases of capillary density and changes in collagen IV and Thy-1 levels indicating compensatory regulation. Slight decreases of ICAM-1 levels and reduction of Thy 1 expression at 0.025–0.5 Gy indicate anti-inflammatory effects, whereas at the highest dose (2 Gy) increased VCAM-1 levels on the endocardium may represent a switch to a pro-inflammatory response. Plasma samples partially confirmed this pattern, showing a decrease of proinflammatory markers (sVCAM, sICAM) at 0.025–2.0 Gy. In contrast, an enhancement of MCP-1, TNFα and fibrinogen at 0.05–2.0 Gy indicated a proinflammatory and prothrombotic systemic response. Multivariate analysis also revealed significant age-dependent increases (KC, MCP-1, fibrinogen) and decreases (sICAM, sVCAM, sE-selectin) of plasma markers. This paper represents local and systemic effects of low-dose irradiation, including also age- and dose rate-dependent responses in the ApoE-/- mouse model. These insights in the multiple inflammatory/thrombotic effects caused by low-dose irradiation might facilitate an individual evaluation and intervention of radiation related, long-term side effects but also give important implications for low dose anti-inflammatory radiotherapy.

Este TFG presenta un análisis de la evolución de los efectos del cambio climático en El Salvador, además se observa la evolución que siguen las emisiones de CO2, TPES (Total Primary Energy Supply), PIB (Producto Interior Bruto) y Población de este país, para el periodo que comprende entre el 1990 y el 2015 con el objetivo de cuantificar los factores conductores resultantes de estos datos mencionados. Todos los análisis efectuados y los resultados obtenidos son comparados, a su vez, con la realidad que atraviesa El Salvador en este periodo indicado. Los cuatro factores conductores del cambio climático de un país son la intensidad de carbono, la intensidad energética, el PIB per cápita y la población. La multiplicación de todos ellos, da como resultado las emisiones de CO2 del país, a través de la relación que existe según la expresión matemática llamada identidad de Kaya. Una vez se tienen a disposición estos factores conductores se pueden realizar futuros escenarios para el país, que sigan una proyección basada en el histórico. O, por otro lado, escenarios alternativos que hagan que los factores conductores tiendan a valores fijados de antemano, y así poder ver de qué forma varían las emisiones del país, según las variaciones impuestas, gracias a la Identidad de Kaya. Se analiza también, que relación guardan estas emisiones resultantes de los escenarios futuros con las que deberían haber en el año 2050 para que se pueda cumplir con el objetivo de los 2º C fijado en la cumbre de París.
