The Ganymede Laser Altimeter (GALA) is an instrument onboard JUICE, whose main scientific goals are to understand ice tectonics based on topographic data, the subsurface structure by measuring tidal response, and small-scale roughness and albedo of the surface. In addition, from the perspective of astrobiology, it is imperative to study the subsurface ocean scientifically. The development of GALA has proceeded through an international collaboration between Germany (the lead), Japan, Switzerland, and Spain. Within this framework, the Japanese team (GALA-J) is responsible for developing three receiver modules: the Backend Optics (BEO), the Focal Plane Assembly (FPA), and the Analog Electronics Module (AEM). Like the German team, GALA-J also developed software to simulate the performance of the entire GALA system (performance model). In July 2020, the Proto-Flight Models of BEO, FPA, and AEM were delivered from Japan to Germany. This paper presents an overview of JUICE/GALA and its scientific objectives and describes the instrumentation, mainly focusing on Japan's contribution. © 2021 COSPAR., NSO/Kitt Peak data (synoptic maps of He I Å intensity) used here are produced cooperatively by NSF/NOAO, NASA/GSFC, and NOAA/SEL. We aimed to reanalyze the differences in the pharmacokinetics (PKs) of meloxicam in East Asian populations based on a population approach using previously published data and to investigate the factors found in population PK analysis that affect the pharmacodynamics (PDs) of meloxicam. Population PK analysis was performed in 119 healthy male subjects (30 Japanese, 30 Chinese, 29 Korean, and 30 white) under strictly controlled trial conditions with regulated meals and a single lot of the drug. We found that CYP2C9 genotype and lean body mass were statistically significant predictors of clearance and volume of distribution, respectively. A statistical significant difference in the PK parameters between ethnic groups could not be identified. Simulations using PK/PD models showed that CYP2C9 genotype is the factor that affects the PDs of meloxicam. The genetic polymorphisms highlighted in this study would be beneficial for conducting clinical trials in East Asians with similar genetic backgrounds.

本稿では、1996年2月12日に打ち上げられた極超音速飛行実験機(HYFLEX)の航法・誘導・制御系解析に用いた飛行シミュレーションプログラムと飛行シミュレーション解析結果について報告する。従来、航空宇宙技術研究所(NAL)では、再突入機、あるいはスペース・プレーンの飛行シミュレーションを行う際の運動方程式は、地球を球と仮定して導出されていた。また、HYFLEXの基本設計段階においても、地球の取り扱いを球とした仮定の基で導かれた運動方程式で飛行シミュレーションを行っていた。しかし、HYFLEXの回収に対する信頼性、成功率を上げること、および飛行安全性確保の観点からより精度の高い飛行シミュレーション解析の要求が生じた。そこで本稿では、地球をより精度の高い回転楕円体モデルと仮定して取り扱う。運動方程式導出に際しては、地球モデルの変更に伴い局所水平座標系の定義が従来のものと一致しなくなるため、新たに地心局所座標系と対気速度座標系を定義する。さらに、地球モデルの変更に伴い機体に作用する外力である重力ベクトルも変化する。このため、重力モデルも逆自乗場のモデルから、重力ポテンシャルによるモデルへと変更した。また、計算効率、計算精度を上げるため、新たに2つの状態量を導入した。本稿ではまず最初に、上記に伴い新たに開発された飛行シミュレーションプログラムについて述べる。そして、開発した飛行シミュレーションプログラムにより行った、打ち上げ前の最終的な飛行シミュレーション解析結果についても報告する。得られた飛行シミュレーション解析結果により、設計されたHYFLEXの航法・誘導・制御系が所望の性能を有していることが確認された。また、着水点、および着水点分散域に関するより精度の高いデータを得た。, This paper describes a flight simulation program, used to analyze the navigation, guidance and control system and for flight analysis of the Hypersonic Flight Experiment vehicle (HYFLEX). Up to this time, the earth had been considered as a sphere at NAL. The flight simulation was carried out using the model at the predesign phase of the HYFLEX. However, more precise flight analysis had to be carried out to predict the splash down point as planned to recover the HYFLEX vehicle. In this paper, two coordinate systems were newly introduced to deal with the earth as a ellipsoid of revolution and new equations of motion were derived. Also, the gravity model was changed to the model mentioned in reference. To maintain precision, two state vectors were newly defined. The designed navigation, guidance and control system of the HYFLEX is proved to satisfy the mission requirements through the results of flight analysis. To assess the tolerability, pharmacokinetics and pharmacodynamics of a novel nonsteroidal and noncompetitive inhibitor of type I and type II 5alpha-reductases, (-)-(S)-4-[1-[4-[1-(4-isobutylphenyl) butoxy]benzoyl]indolizin-3-yl]butyric acid (TF-505), after single and multiple oral doses in healthy volunteers.In the single-dose study, six young adult males in each dose group received 25 mg or 50 mg of TF-505, and six older males (or= 40 years) in each dose group received 75 mg or 100 mg of TF-505. The subjects were given the drug in ascending dose and in the fasting state. Six subjects also received 50 mg of TF-505 after breakfast in a two-period crossover manner. In the multiple-dose study, six older males in each dose group received 12.5 mg or 25 mg TF-505 after breakfast daily for 7 days. Plasma concentrations of TF-505, dihydrotestosterone (DHT) and testosterone were measured. The pharmacokinetics of TF-505 were analysed by a compartment model with first-order absorption, first-order elimination and a lag time. Pharmacokinetic and pharmacodynamic relationships were evaluated by indirect response modelling with inhibition of input.Maximum plasma concentration (Cmax) and the area under the concentration-time curve (AUC) increased proportionately after the single dose up to 50 mg and with the multiple doses. Linearity was not detected between 75 and 100 mg of TF-505. Dose dependency was also noted for the effect of TF-505 on DHT concentrations following single doses up to 50 mg and multiple doses. Plasma DHT concentrations decreased maximally to 58.2, 49.5, 54.2 and 49.8% of basal values at 8-12 h after single administration of 25, 50, 75 and 100 mg TF-505, respectively, and to 60.5 and 49.4% at the 7th and 5th dose following multiple doses of 12.5 and 25 mg TF-505, respectively. The predicted effect curves matched the observed data when the indirect response model was applied to the time course of the suppressant effect of TF-505 on plasma DHT concentrations after both the single and multiple studies. Fifty percent inhibitory concentrations (IC50) of 0.82, 1.48, 1.31 and 0.88 micro g ml(-1), zero-order rate constants for the onset of plasma DHT concentration changes (kin) of 17.8, 17.4, 17.0 and 10.7% h(-1) and first-order rate constants for increase in plasma DHT concentrations to basal values (kout) of 0.17, 0.16, 0.17 and 0.10 h(-1) for the single study at doses of 25, 50, 75 and 100 mg, respectively, were attained. In the multiple-dose study, IC50s were 1.74 and 1.49 micro g ml(-1) for the 12.5 and 25 mg doses, respectively. No serious adverse events related to TF-505 were observed.TF-505 was well tolerated in healthy male volunteers. Accumulation of TF-505 in plasma was not observed during multiple dosing. The indirect response model described the relationships between pharmacokinetics and pharmacodynamics of TF-505. Such modelling is expected to yield an appropriate dosage regimen in subsequent clinical trials.
