Summary Differentiation of Trypanosoma brucei, a flagellated protozoan parasite, between life cycle stages typically occurs through an asymmetric cell division process, producing two morphologically distinct daughter cells. Conversely, proliferative cell divisions produce two daughter cells, which look similar but are not identical. To examine in detail differences between the daughter cells of a proliferative division of procyclic T. brucei we used the recently identified constituents of the flagella connector. These segregate asymmetrically during cytokinesis allowing the new‐flagellum and the old‐flagellum daughters to be distinguished. We discovered that there are distinct morphological differences between the two daughters, with the new‐flagellum daughter in particular re‐modelling rapidly and extensively in early G1. This re‐modelling process involves an increase in cell body, flagellum and flagellum attachment zone length and is accompanied by architectural changes to the anterior cell end. The old‐flagellum daughter undergoes a different G1 re‐modelling, however, despite this there was no difference in G1 duration of their respective cell cycles. This work demonstrates that the two daughters of a proliferative division of T. brucei are non‐equivalent and enables more refined morphological analysis of mutant phenotypes. We suggest all proliferative divisions in T. brucei and related organisms will involve non‐equivalence.

Glioblastoma (GB) is the most prevalent and aggressive brain tumor with high morbidity. Determination of tumor initiative and prognostic factors are ultimately needed for therapy options. The effects of Serine Protease Inhibitor (SERPIN) B1 on tumor progression have been reprted in several cancer types including breast and lung cancers. However, the relation between SERPINB1 expression levels and glioma progression is still to be elucidated.

Parageobacillus thermoglucosidasius is a metabolically versatile, facultatively anaerobic thermophile belonging to the family Bacillaceae. Previous studies have shown that this bacterium harbours co-localised genes coding for a carbon monoxide (CO) dehydrogenase (CODH) and Ni-Fe hydrogenase (Phc) complex and oxidises CO and produces hydrogen (H2) gas via the water-gas shift (WGS) reaction. To elucidate the genetic events culminating in the WGS reaction, P. thermoglucosidasius DSM 6285 was cultivated under an initial gas atmosphere of 50% CO and 50% air and total RNA was extracted at ~8 (aerobic phase), 20 (anaerobic phase), 27 and 44 (early and late hydrogenogenic phases) hours post inoculation. The rRNA-depleted fraction was sequenced using Illumina NextSeq, v2.5, 1x75bp chemistry. Differential expression revealed that at 8 vs 20, 20 vs 27 and 27 vs 44 hours post inoculation, 2190, 2118 and 231 transcripts were differentially (FDR &lt, 0.05) expressed. Cluster analysis revealed 26 distinct gene expression trajectories across the four time points. Of these, two similar clusters, showing overexpression at 20 relative to 8 hours and depletion at 27 and 44 hours, harboured the CODH and Phc transcripts, suggesting possible regulation by O2. The transition between aerobic respiration and anaerobic growth was marked by initial metabolic deterioration, as reflected by up-regulation of transcripts linked to sporulation and down-regulation of transcripts linked to flagellar assembly and metabolism. However, the transcriptome and growth profiles revealed the reversal of this trend during the hydrogenogenic phase.

Like physics in the 19th century, biology and molecular biology in particular, has been fertilized and enhanced like few other scientific fields, by the incorporation of mathematical methods. In the last decades, a whole new scientific field, bioinformatics, has developed with an output of over 30,000 papers a year (Pubmed search using the keyword “bioinformatics”). Huge databases of mass throughput data have been established, with ArrayExpress alone containing more than 2.7 million assays (October 2019). Computational methods have become indispensable tools in molecular biology, particularly in one of the most challenging areas of cancer research, multidrug resistance (MDR). However, confronted with a plethora of different algorithms, approaches, and methods, the average researcher faces key questions: Which methods do exist? Which methods can be used to tackle the aims of a given study? Or, more generally, how do I use computational biology/bioinformatics to bolster my research? The current review is aimed at providing guidance to existing methods with relevance to MDR research. In particular, we provide an overview on: a) the identification of potential biomarkers using expression data; b) the prediction of treatment response by machine learning methods; c) the employment of network approaches to identify gene/protein regulatory networks and potential key players; d) the identification of drug-target interactions; e) the use of bipartite networks to identify multidrug targets; f) the identification of cellular subpopulations with the MDR phenotype; and, finally, g) the use of molecular modeling methods to guide and enhance drug discovery. This review shall serve as a guide through some of the basic concepts useful in MDR research. It shall give the reader some ideas about the possibilities in MDR research by using computational tools, and, finally, it shall provide a short overview of relevant literature. © 2019

The aim of this study was to compare the outcomes of consecutive IHCA with out-of-hospital cardiac arrest (OHCA) patients treated with TTM. Design, setting, and patients: Retrospectively collected data on all consecutive adult patients treated with TTM at a university tertiary heart center between 2005 and 2011 were analyzed. Measurements: Primary endpoints were survival to hospital discharge and long-term survival. Secondary endpoint was neurological outcome assessed using the Pittsburgh cerebral performance category (CPC). Results: A total of 282 patients were included in this study; 233 (83%) OHCA and 49 (17%) IHCA. The IHCA group presented more often with asystole, received bystander cardiopulmonary resuscitation (CPR) in all cases, and had shorter time to return of spontaneous circulation (ROSC). Survival to hospital discharge was 54% for OHCA and 53% for IHCA (adjusted odds ratio 0.98 [95% confidence interval {CI}; 0.43–2.24]). Age ≤60years, bystander CPR, time to ROSC ≤10 min, and shockable rhythm at presentation were associated with survival to hospital discharge. Good neurologic outcome among survivors was achieved by 86% of OHCA and 92% of IHCA (P=0.83). After a median follow-up time of >5 years, 83% of OHCA and 77% of IHCA were alive (adjusted hazard ratio [HR] 1.51 [95% CI; 0.59–3.91]). Age ≤60 years was the only factor associated with long-term survival (adjusted HR 2.73 [95% CI; 1.36–5.52]). Conclusion: There was no difference in short- and long-term survival and no difference in neurologic outcome to hospital discharge between IHCA and OHCA patients treated with TTM despite higher frequency of asystole in IHCA. A total of 237 patients were treated over a period of five years for anorectal abscesses. Wide circular opening with removal of all purulent foci and necroses from outside had been preferred, because almost only "late forms" of anorectal inflammatory processes (perianal and ischiorectal abscesses) were seen. Sphincterotomy was possible in only a few patients. Postoperative fistulae occurred in 14% of the patients.

The Dvurechenskii mud volcano (DMV) is located in permanently anoxic waters at 2060 m depth (Sorokin Trough, BlackSea). The DMV was studied during the RV Meteor expedition M72/2 as an example of an active mud volcano system, to inves-tigate the significance of submarine mud volcanism for the methane and sulfide budget of the anoxic Black Sea hydrosphere.Our studies included benthic fluxes of methane and sulfide, as well as the factors controlling transport, consumption and pro-duction of both compounds within the sediment. The pie-shaped mud volcano showed temperature anomalies as well as soluteand gas fluxes indicating high fluid flow at its summit north of the geographical center. The anaerobic oxidation of methane(AOM) coupled to sulfate reduction (SR) was repressed in this zone due to the upward flow of sulfate-depleted fluids throughrecently deposited subsurface muds, apparently limiting microbial methanotrophic activity. Consequently, the emission of dis-solved methane into the water column was high, with an estimated rate of 0.46 mol m2d1. On the wide plateau and edge ofthe mud volcano surrounding the summit, fluid flow and total methane flux were lower, allowing higher SR and AOM ratescorrelated with an increase in sulfate penetration into the sediment. Here, between 50% and 70% of the methane flux (0.07–0.1 mol m2d1) was consumed within the upper 10 cm of the sediment. The overall amount of dissolved methane releasedfrom the entire mud volcano structure into the water column was significant with a discharge of 1.3107mol yr1. TheDMV maintains also high areal rates of methane-fueled sulfide production and emission of on average 0.05 mol m2d1. Thisis a difference to mud volcanoes in oxic waters, which emit similar amounts of methane, but not sulfide. However, based on acomparison of this and other mud volcanoes of the Black Sea, we conclude that sulfide and methane emission into the hydro-sphere from deep-water mud volcanoes does not significantly contribute to the sulfide and methane inventory of the Black Sea.

The use of growth estimation methods that depend on unreliable age data has previously hindered the quantification of perceived differences in growth rates between the two cod stocks inhabiting the Baltic Sea. Data from cod tagged in different regions of the Baltic Sea during 2007–2019 were combined, and general linear models were fit to investigate inter-regional (defined as area of release) and inter-stock (assigned to a subset of recaptures using genetic and otolith shape analyses) differences in individual growth. An average-sized cod (364 mm) caught in the western Baltic Sea and assigned to the western Baltic cod stock grew at more than double the rate (145 mm year−1) on average than a cod of the same size caught in the eastern Baltic Sea and assigned to the eastern Baltic cod stock (58 mm year−1), highlighting the current poor conditions for the growth of cod in the eastern Baltic Sea. The regional differences in growth rate were more than twice as large (63 mm year−1) as the stock differences (24 mm year−1). Although the relative importance of environmental and genetic factors cannot be fully resolved through this study, these results suggest that environmental experience may contribute to growth differences between Baltic cod stocks.
