This current study investigates the biological efficiency of essential oils extracted from Bistorta affinis and Malcolmia cabulica on human and foodborne pathogens as well as against insects and termites. The oils were obtained by steam distillation using a Clevenger-type system and analyzed for their constituents using GC-MS. Twenty compounds were identified, wherein carvacrol and thymol were the main constituents in both plants. Essential oils exhibited significant activity against all tested foodborne bacteria, fungi, and other pathogens. In addition, essential oils from both plants have shown promising activity against all tested insects, which is a positive sign of being used as an insect repellent. In contrast, no activity was observed against termites. The bioactivities are mainly due to carvacrol and thymol’s levels in the essential oils that known for their effectiveness against foodborne pathogens and pests. The present study constitutes a basis for further investigation and development of bioactive compounds in Bistorta affinis and Malcolmia cabulica.

Congress grass (Parthenium hysterophorus L., family Asteraceae) is found generally in agricultural and vacant lands across the globe. This study investigated the fatty acids and elemental profiles of different parts including roots, stem and leaves of P. hysterophorus in the response to seasonal variation. The elemental analysis and fatty acid were determined in both the winter and summer seasons. The root, stem, and leaves powder were subjected to extraction with acetone, water, chloroform, diethyl ether, ethanol, methanol, and n-hexane to measure the extractive values. Among all the plant parts, leaves recorded the highest extractive value of 29%, and 28% with acetone and diethyl ether, respectively, while the root recorded the lowest (10.3%) extractive value with chloroform. Higher concentrations of saturated and unsaturated fatty acids were detected in all the plant parts in the winter collection than in the summer season. Parthenium hysterophorus is multi medicinal applications such as used for rheumatic pain, urinary tract infections, inflammation, diarrhea, neuralgia, and malaria. The study highlights the importance of collection time (seasons) of different parts of P. hysterophorus for the maximum extraction of both elements and fatty acids either saturated or unsaturated from the pharmacogenetic point of view.

Urease is a metallo-protein in which two nickel (Ni) atoms are present at the active site. It is found in all the organisms such as, bacteria, fungi, plants, and in algae. Urease enzymes are also involved in catalyzing the hydrolysis of urea into CO2 and NH3. Urease is a virulence factor found in various pathogenic bacteria. It is essential in colonization of a host organism and in maintenance of bacterial cells in tissues. Due to its enzymatic activity, urease has a toxic effect on human cells. Current research work aimed to purify the three di-naphthodiospyrols namely; 5,4 -dihydroxy-1-methoxy-6,6 -dimethyl7,3-binaphthyl-1,4,5,8-tetraone (1), 5,8 -dihydroxy-5-methoxy-6,6-dimethyl-7,3-binaphthyl-1,4,1,4 -tetraone (2) and 8,5,8-trihydroxy-6,6-dimethyl-7,3-binaphthyl-1,4,1,4-tetraone (3) from the chloroform fraction of Diospyros lotus roots. The pure constituents (1-3) were evaluated for their urease inhibitory activity. Compounds (1-3) exhibited potent activity with IC50 value of 28.27 ± 1.68, 20.57 ± 2.00, 27.12 ± 2.45 µM respectively, when compared with standard thiourea (IC50 = 21.23 ± 0.12 µM). The pure isolated compounds (1-3) were also subjected to in silico studies to know their binding pattern with enzyme. The binding orientation and interactions with the important residues of urease enzyme revealed that the three compounds inactivate the enzyme activity by interacting with Ni bi-center as well as with the tailpiece residues.

Syzygium cumini L. (Jamun) is a Pakistani indigenous plant, which exhibits various diseases biological activities including antidiabetic, antiulcer, hepatoprotective, and even diuretic. The present work was carried out to provide a scientific justification to its use in folk medicine. In the current research work, the crude methanol extract and subsequent fractions of S. cumini fruits were subjected to phytochemical studies, along with urease, α-glucosidase, and phosphodiesterase inhibition profiling. Results showed that the plant is a rich source of various classes of compounds such as alkaloids, steroids, flavonoids, carbohydrates, phenols, glycosides, saponins, and tannins. In addition, results indicated that the crude extract and fractions significantly inhibit the aforementioned enzymes at tested concentrations. The observed urease inhibitory activity (IC50 values) of methanol, ethyl acetate, n-butanol, and chloroform were 10.17 ± 1.00, 7.09 ± 2.00, 8.32 ± 1.76, and 12.23 ± 1.09 µg, respectively. Similarly, the crude methanol extract, ethyl acetate, chloroform, and n-butanol factions exhibited significant α-glucosidase inhibitory potential with IC50 values of 129.98 ± 1.23, 702.74 ± 1.65, 430.21 ± 1.98, and 650.11 ± 1 0.00 µg, respectively, as compared to the standard. On the other hand, the ethyl acetate and n-butanol factions showed excellent phosphodiesterase inhibitory effect with IC50 values of 270.77 ± 1.98 and 87.09 µg, respectively. The antagonistic effect of these samples is attributed to the presence of different phytochemicals. Moreover, the inhibition of these enzymes provided a sound scientific backup to the mentioned folklores.

Previous studies reported disrupted hepatic function and structure following the administration of cyclosporine A (CsA) in humans and animals. Recently, we found that avocado seeds (AvS) ameliorated CsA-induced nephrotoxicity in rats. As a continuation, herein we checked whether AvS could also attenuate CsA-induced hepatotoxicity in rats. Subcutaneous injection of CsA (5 mg/kg) for 7 days triggered hepatotoxicity in rats, as indicated by liver dysfunction, redox imbalance, and histopathological changes. Oral administration of 5% AvS powder for 4 weeks ameliorated CsA-induced hepatotoxicity, as evidenced by (1) decreased levels of liver damage parameters (alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), and bilirubin), (2) resumed redox balance in the liver (reduced malondialdehyde (MDA) and increased superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx)), (3) downregulated hepatic expression of endoplasmic reticulum (ER) stress-related genes (X-box binding protein 1 (XBP1), binding immunoglobulin protein (BIP), C/EBP homologous protein (CHOP)), and apoptosis-related genes (Bax and Casp3), (4) upregulated expression of the anti-apoptotic gene Bcl2, (5) reduced DNA damage, and (6) improved liver histology. These results highlight the ability of AvS to ameliorate CsA-induced hepatotoxicity via the inhibition of oxidative stress and proapoptotic ER stress.

The goal of this study was to investigate whether Ducrosia flabellifolia and Savignya parviflora methanol extract the have effect on colon and prostate cancer cell lines. Analysis of total content of phenolics and flavonoids of each plant extract was carried out. Cytotoxic effect, cell cycle analysis, induction of apoptosis and gene expression of Bcl-2 and Bax genes were studied. Obtained results indicated that, the plant extracts exhibit growth inhibition of used cancer cell lines and induced apoptosis as well as arresting of cell cycle. At the molecular level, changes in gene expression were detected via qPCR and confirmed by western blotting. The exhibited anticancer potentialities of plant extracts against utilized cancer cell lines are due to its containing bioactive compounds. Further detailed isolation, fractionation and characterization of bioactive compounds are needed.

Diabetes is a chronic condition which is locally managed through the stem of Ficus foveolata. To find the exact chemical constituent responsible for this activity, a triterpene lactone (ficusonolide) isolated from F. foveolata was studied for antidiabetic potential through the in vitro antidiabetic paradigm employing L-6 cells and an in vivo antidiabetic assay against non-insulin-dependent rats. The results on glucose uptake in the L-6 cell line indicated that ficusonolide has enhanced the uptake of glucose by 53.27% over control at a dose of 100 μg/mL, while at doses of 50 and 25 μg/mL, the glucose uptake was enhanced by 22.42 and 14.34%, respectively. The extract of F. foveolata (100 mg/kg) and ficusonolide (50 mg/kg) demonstrated a significant (p < 0.001) decline in streptozotocin-induced hyperglycemia of diabetic rats. Ficusonolide displayed conspicuous inhibitory activity against the molecular docking studies with proteins such as dipeptidyl peptidase-IV (DPP-IV), protein tyrosine phosphatase 1B (PTP-1B), α-glucosidase, and α-amylase subjected to molecular targets. Detailed computational and structural insights affirmed promising interactions between target proteins and ficusonolide. In conclusion, the plant and its isolated compound have significant antidiabetic activity with a possible mechanism of interaction with DPP-IV, PTP-1B, α-glucosidase, and α-amylase.

Tobacco (Nicotiana tabacum) is an herbaceous plant originating from South America and processed into cigarettes for consumption. Polyphenols are considered vital components of tobacco in view of their contribution to antioxidant properties. This study aimed to determine the phenolic compounds in different tobacco varieties by applying cold extraction with methanol and distilled water. The extracts were screened for phenolic compound diversity and distribution as well as their antioxidant potential in different tobacco varieties. The results showed that the methanolic extract of tobacco SP-28 exhibited the highest value in the total phenolic content (24.82 ± 0.07 mg GAE/gd.w.) and total flavonoid content (4.42 ± 0.01 mg QE/gd.w.), while the water extract of tobacco SN-2 exhibited the highest value in the total condensed tannin (1.12 ± 0.03 mg CE/gd.w.). The radical scavenging capacities of tobacco SP-28 were relatively high in DPPH (18.20 ± 0.01 mg AAE/gd.w.) and FRAP (3.02 ± 0.10 mg AAE/gd.w.), whereas the ABTS value was the highest in tobacco SN-2 (37.25 ± 0.03 mg AAE/gd.w.), and the total antioxidant capacity was the highest in tobacco SN-1 (7.43 ± 0.18 mg AAE/gd.w.). LC-ESI-QTOF-MS/MS identified a total of 49 phenolic compounds, including phenolic acids (14), flavonoids (30), and other polyphenols (5) in four different tobacco varieties. Tobacco SP-28 showed the highest number of phenolic compounds, especially enriched in flavones. Our study highlights the antioxidant potential of tobacco extracts and reveals the phenolic distribution among different tobacco varieties that could support tobacco utilization in different pharmaceutical industries.

The Sophora mollis is one of the best anti-inflammatory, antioxidant, and anticancerous plant; therefore, the isolated chemical constituents, that is, scopoletin (1), pinitol (2), 2-propenoic acid, 3-(3,4-dihydroxyphenyl)-octacosyl ester (3), betulin (4), and β-sitosterol glucoside (5) were tested for these folklores. The structures of the isolated compounds were confirmed by 1H NMR, 13C NMR, 2D-NMR, and mass spectral data. The anti-inflammatory, anticancer, antiglycation, and antioxidant activities of compounds 1-5 were evaluated using different assays. Compound 1 exhibited significant anti-inflammatory effect as it reduced edema of the paw (83.98%), which is more potent than the standard drug (ibuprofen) (which showed an inhibition percentage of 73.22% a), followed by compound 3. Furthermore, compound 3 showed significant free-radical scavenging activity using the 2,2-diphenyl-1-picryl-hydrazyl-hydrate (DPPH) free-radical assay. Percentage inhibition of DPPH recorded was 95.646 ± 0.003, 94.766 ± 0.014, and 94.516 ± 0.011% at concentrations of 400, 200, and 100 μg/mL, respectively. Evaluation of anticancer activity of isolated compounds reveals weak effect against HeLa and 3T3 cell lines. Docking studies of the most active compound into the binding sites of cyclooxygenase isoforms showed a better antagonistic potential against COX-1 than the COX-2 isoform.

The inadequate role of silver nanoparticles is evident from their underutilization in myriad applications in the modern era. Herein we report the environment-friendly, facile, and robust synthesis of silver nanoparticles (Ag-NPs) from alcoholic extracts of black cumin (Bunium persicum) seeds to reduce and stabilise agents. The extract principally consists of nigellon, terpenoid, thymoquinone, polyene, phenylpropanoids, and phototoxic furanocoumarins. The effect of pH revealed that the BCS@Ag-NPs were more stable at neutral pH than acidic and basic. A change in the size and number of BCS@Ag-NPs was observed at high sodium chloride solution concentrations showing their instability in a concentrated salt solution. The synthesized BCS@Ag-NPs were considered significant catalysts showed by approximate 99% conversion of the 4-nitrophenolate anion into 4-aminophenol in 24 min. The surface plasmon resonance of BCS@Ag-NPs at 430 nm was recorded on UV–Vis spectrometer, and particle size ranging from 35 to 77 nm was measured through scanning electron microscopic (SEM) analysis. FTIR spectra confirmed the presence of amines, alcohols and amide functional groups at 3365 cm−1, 1646 cm−1 and 1026 cm−1 capped along with the reduction of BCS@Ag-NPs. The firm peaks for Ag atoms in BCS@Ag-NPs at almost 0.4, 3.1 and 3.2 keV in EDX analysis showed the presence of elemental silver. Nevertheless, the alcoholic seed extract and synthesized BCS@Ag-NPs exhibited excellent various pharmacological activities. Both the tested samples significantly inhibited Urease, CA-II, PDE-I. The inhibitory potential against tyrosinase was appreciated against extract as compared to BCS@Ag-NPs. The alcoholic extract was effective in the inhibition of E. coli. Both of the tested samples proved significant analgesia in the acetic acid-induced writhing model. A noticeable sedative response was observed as a compared extract. No mortality was detected in acute toxicity. In conclusion, these results provide the scientific rationale to the folklore of black cumin for GIT problems, hypertension, bronchodilation and analgesic.

The global water shortage alert has been upgraded to a higher risk level. Consequently, a sustainable approach for ecofriendly, energy efficient water desalination is required for agricultural and municipal water reuse. In this study, an energy-efficient biological desalination process was used to treat chloride anions, which are the most abundant anion salt in seawater. Three algal species were studied: Scenedismus arcuatusa (S. arcuatusa), Chlorella vulgaris (C. vulgaris), and Spirulina maxima (Sp. maxima), under different operating conditions (saline concentrations, contact time, high light intensity, and CO2 supply), and two kinetic models were used. It was identified that under a high light intensity and CO2 supply, S. arcuatusa enhanced chloride removal from 32.42 to 48.93%; the daily bioaccumulation capacity (Qe), according to the kinetic models, was enhanced from 124 to 210 mg/g/day; and the net biomass production was enhanced from 0.02 to 0.740 g/L. The EDX analysis proved that salt bioaccumulation may be attributed to the replacement of Ca2+ and Mg2+ with Na+ and K+ through algal cells. The study’s findings provide promising data that can be used in the search for novel energy-efficient alternative ecofriendly desalination technologies based on algae biological systems with biomass byproducts that can be reused in a variety of ways.

Several plants in the genus Pistacia are used in the treatment of various pathogenic and non-pathogenic disorders. Especially important are the major species belonging to this genus such as Pistacia lentiscus, Pistacia atlantica, Pistacia vera, Pistacia terebinthus, and Pistacia khinjuk, among others; these have been reported for their potential benefits both in medical and commercial purposes. In addition, members of this genus exhibit numerous ethnomedicinal uses, such as analgesic, anti-inflammatory, anticancer, antimicrobial, antihypertension, antihyperlipidemic, antiviral, and antiasthma. In light of these potential uses, the present chapter aimed to collect and summarize the literature about all of this medicinal information. Accordingly, this chapter focuses on the pharmacological uses and benefits of the genus Pistacia, especially those related to health issues.

Fast, simple, and environmentally friendly gold nanoparticles (Au-NPs) capped and stabilized with black cumin (Bunium persicum) seed alcohol extract are reported. The aqueous gold ions (Au3+) were treated with B. persicum (BP) seed extract, which resulted in a rapid color change to red, indicating the synthesis of Au-NPs. UV-Vis spectroscopy, FTIR, SEM, energy dispersive X-ray (EDX), and X-ray diffraction (XRD) techniques were used to further characterize the Au-NPs. Its stability was assessed against various pH levels and sodium chloride levels (NaCl), different salts of same concentration as well as at a range of temperature (30–100°C). The UV-Vis spectrum in the Au-NPs produced a 540 nm plasmon surface resonance, and a 25–50 nm range of particulates was shown in the SEM analysis. In addition, the FTIR spectra confirmed the inclusion in the capping and decrease of Au-NPs of amines, amide groups, and alcohols. The EDX analysis confirmed the presence of element Au. Furthermore, Au-NPs were tested for enzyme inhibition and antibacterial and antifungal activities and showed remarkable response. These findings have concluded that BP seed extract is an effective bio-reductant of gold nanoparticle synthesis, which can be further applied in different biomedical and pharmaceutical industries.

Background During the last few decades, patients worldwide have been interested in using alternative medicine in treating diseases to avoid the increased side effects of chemical medications. Green coffee is unroasted coffee seeds that have higher amounts of chlorogenic acid compared to roasted coffee. Green coffee was successfully used to protect against obesity, Alzheimer disease, high blood pressure and bacterial infection. Methods This study aimed to investigate the probable protective activity of the green coffee methanolic extract, silymarin and their combination on CCl4-induced liver toxicity in male rats. Thirty Sprague – Dawley male albino rats were divided into 5 groups; control negative (G1) just got the vehicle (olive oil) and the other four groups received CCl4 dissolved in olive oil through an intraperitoneal injection and were divided into untreated control positive group (G2), the third group (G3) was treated with green coffee methanolic extract, the fourth group (G4) was treated with silymarin, and the fifth group (G5) was treated with a combination of green coffee methanolic extract and silymarin. Results In the positive control group treated with CCl4 (G2), the CCl4-induced toxicity increased lipid peroxidation, IL-6, kidney function parameters, liver function enzymes, total cholesterol, triglycerides and low-density lipoproteins, and decreased irisin, antioxidants, CYP450 and high-density lipoprotein levels. Hepatic tissues were also injured. However, treating the injured rats in G3, G4 and G5 significantly improved the altered parameters and hepatic tissues. Conclusions Green coffee methanolic extract, silymarin, and their combination succeeded in protecting the male rats against CCl4 hepatotoxicity due to their antioxidant activity. Effect of green coffee methanolic extract mixed with silymarin in G5 was more efficient than that of green coffee methanolic extract in G3 or silymarin in G4.

Fernandoa adenophylla (Wall. ex G. Don) Steenis is traditionally used to cure various diseases and can be included as an ingredient in massage oils, which are supposed to comfort muscular tension and pain. This study was designed to assess the antinociceptive, muscle relaxant, and molecular docking properties of a novel compound, namely, (5aR,5a1R,6R,7aS,14bR,15R)15-hydroxy-7a-methyl-6-(2-methylprop-1-en-1-yl)-7,7a,14b,15-tetrahydro-5H-t-5a,15methanobenzo[g]benzo[5,6]azuleno[1,8-bc]chromene-5,9,14,16(5a1H,6H)- tetraone (peshawaraquinone), isolated from the methanolic extract of F. adenophylla in an animal model. The chemical structure of the isolated compound was elucidated using advanced spectroscopic techniques and further confirmed by XRD analysis. Compound 1 was tested against hot plate-induced noxious stimuli at various doses (2.5, 5, 10, and 15 mg/kg i.p.). The muscle relaxation potency of compound 1 was evaluated in the inclined and traction test, while the open-field test was used for the determination of sedative potential. The isolated compound was also subjected to acute toxicity analysis. The compound was then subjected to molecular docking analysis to determine the exact mechanism of action. Compound 1 demonstrated significant (p < 0.05) analgesic effect in a dose-dependent manner. A noticeable muscle relaxant effect was observed with the passage of time in both experimental models. The compound 1 showed a significant (p < 0.05) sedative effect, and in an acute toxicity study, the compound 1 was devoid of any noxious effects. The docking studies showed preferential affinity for μ-opioid and GABAA receptors. Hence, the prospective antinociceptive and muscle relaxant and sedative properties are probably mediated through these two target interactions.

This study was aimed to evaluate the impact of different monochromatic LED light colour (white ‘WL’, green ‘GL’, blue ‘BL’ and mixed green and blue ‘GL × BL’) and two levels of light intensity (5 lx and 20 lx) on growth performance, physiological responses (haematological and biochemical), behaviour and fear reactions in broiler. A 4 × 2 factorial treatment structure was used to evaluate the impact of 4 LED light colours (WL, GL, BL and GL × BL) and 2 levels of light intensity (5 lx and 20 lx) in triplicates. One-day-old Cobb Avian-48 chicks (N = 384, 36.29 ± 0.092 g) were used. Results revealed a significant effect of light colour on performance (final weight, weight gain and feed conversion ratio [FCR]), haematological parameters (haemoglobin, RBCs, PCV, TLC and H/L ratio), biochemical parameters (TP, albumin (AL), and AL/globulin (GL) ratio), tonic immobility (TI) duration and open field (OF) test (latency to first step, mobility duration, immobility duration, immobility sitting, immobility standing, pecking and dropping) with the best results observed in GL × BL. Light intensity had no effects on studied parameters except for H/L ratio, TI duration, latency to first step, immobility sitting, immobility standing and pecking with better findings detected in 5 lx. There was a significant light colour × light intensity interaction on final weight, weight gain, AL, TI duration, mobility duration, immobility duration, immobility sitting, immobility standing and pecking. Conclusively, monochromatic LED light (GL × BL, BL and GL) particularly of low intensity could be a source of light in broiler houses that might have the potential to improve broiler welfare.Highlights LED light is an important factor in the broiler production. Mixed blue and green, blue or green light of 5 lx improves broiler performance and health status. Mixed blue and green, blue or green light of low intensity reduced fear reaction in broilers.

Introduction: The genus Euphorbia is known to contain diterpenoids, and several isolated compounds which exhibited biological activities including significant multidrug resistance reversal effects. This work is focused on the isolation, in vitro and in silico studies of two natural bio-active flavonoids (1 & 2) isolated from Euphorbia pulcherrima bark for the very first time. Methods: The phytochemical investigation resulted in the identification of two flavonoids: 3,5,7-trihydroxy-2-(4-hydroxy-3-methoxyphenyl)-6-methoxy-4H-chromen-4-one (1) and 2-(3,4-dihydroxyphenyl)-3,5,7-trihydroxy-6-methoxy-4H-chromen-4-one (2), which were isolated for the first time from Euphorbia pulcherrima. Results: The chemical structures of the two isolated compounds were confirmed by H-1 NMR, C-13 NMR, and ESI-HRMS spectral data. The Bioactivity activity of these compounds was evaluated; results revealed that compounds 1 & 2 exhibit promising urease inhibitory potential with IC50 values of 15.3 +/- 2.13 mu M and 19.0 +/- 2.43 mu M, respectively, whereas the positive control thiourea had an IC50 of 21.0 +/- 0.23 mu M. Similarly, these compounds were also evaluated against the tyrosinase enzyme; results showed that compound 1 displays significant inhibitory activity with an IC50 value of 48.7 +/- 2.19 mu M, whereas compound 2 exhibited a moderate effect with an IC50 value of 74.8 +/- 1.79 mu M, when compared with the standard (alpha-kojic acid, IC50 = 47.6 +/- 0.67 mu M). Additionally, compounds 1 and 2 also exhibited anti-glycation and phosphodiesterase inhibitory activities. Conclusion: Studies dealing with the drug like properties such as in silico screening (docking study) was also carried out to discover the structural features of both compounds 1 and 2. Results indicated that the docking scores of compounds 1 and 2 are in agreement with their IC50 values. Key messages Isolation and characterization of two bioactive flavonoids (1 and 2) from Euphorbia pulcherrima. In silico and in vitro enzyme inhibition studies were conducted to identify the therapeutic potential of flavonoids 1 and 2. Drug-like properties were calculated to discover important pharmacophoric features.

In the current study, the folklore medicine

In the current study, the folklore medicine, Syzygium cumini, was experimentally evaluated for anti-inflammatory, analgesic, sedative, and muscle relaxant effects. The extract and fractions of S. cumini were found safe up to 1000 mg/kg with no mortality, except for slight sedation as a minor side effect. Both, the extract and various fractions of S. cumini demonstrated significant inhibition (86.34%) of carrageenan-induced inflammation in mice. Acetic acid induced writhes were attenuated ( p < 0.001 ) by S. cumini in a dose-dependent manner, except for the n-hexane fraction. The maximum effect was observed at a dose of 500 mg/kg in mice. The maximum muscle relaxant effect of all tested samples was recorded at a dose of 500 mg/kg bodyweight, where the percent inhibition exhibited by dichloromethane fraction was 82.34%, followed by chloroform fractions (71.43%) and methanolic extract (70.91%). Our findings validate the folklore medicinal claims of S. cumini, as an analgesic and anti-inflammatory agent.

Studies of the ethyl acetate extract bark extract of Olea ferruginea led to the isolation of one new compound Ferruginan A (1) in addition to two known compounds, Ferruginan (2) and cycloolivil (3). Structures of the isolated compounds were confirmed by mass spectrometry (MS) and NMR spectral data. The ethyl acetate fraction and compounds (1–3) were evaluated against breast cancer cell line (MCF-7) and as antioxidants using the free radical scavenging assay. Results revealed that compound 2 exhibits significant antioxidant activity with an I C 50 value of 21.74 μg/mL. In addition, the ethyl acetate fraction showed good cytotoxic activity (79.31% inhibition at 250 μg/mL), whereas compounds 1–3 exerted mild cytotoxic activity ( I C 50 = 8.03 – 12.01 μ g / mL ) as compared to the standard ( I C 50 = 4.41 μ g / mL ) against MCF-7. Docking studies suggested that antioxidant activity is due to the chelation of compounds with copper present in the active site of tyrosinase. Among the different biological sources, seaweeds have lot of biotechnological applications. Saudi Arabia is bounded by three bodies of water. With a coastal border of almost 1,800 km. This area high species richness caused by its complex geological history has encompassed genetic and morphological diversity studies for decades. The DNA-barcoding using rbcL gene has proved its usefulness in studying seaweeds phylogenetic diversity, multiple cryptic introductions, environmental modulation the geographical distribution and species identification in different seaweed species. Eight algae samples were collected from different locations in Saudi Arabia. The rbcL gene was used through PCR protocol for species identification. A total number of 8 sequences were obtained with a total sequence length of 5263 bp. where it ranged from 610 to 753 with an average length of 658 bp. The species identification revealed that the specimens samples 1,2,3,4,5,6,7 and 8 belongs to Padina pavonica, Turbinaria gracilis, Carpomitra costata, Pterocladiella capillacea, Cladostephus spongiosus, Ulva lactuca, Sporochnus comosus and Sargassum muticum respectively. The rbcL-based DNA bar-coding was almost successful to identify different seaweeds specimens according to species and genus. Some specimens rbcL was not adequate to identify genus level and failed to differentiate between highly similar species. We suggest to use more DNA barcoding techniques in addition to rbcL to ad more resolution to the species identification.

BACKGROUND AND OBJECTIVE Carissa edulis (CE) (Apocynaceae) is distributed in tropical Africa and Asia and commonly used in folk medicine to treat many diseases such as headache, cough, rheumatism and fever. The purpose of this study was to evaluate the protective role of ethanolic extract of CE, a medicinal plant locally called "Al-Arm" in Yemen, against liver injury induced by dimethoate (DM) intoxication in male guinea pigs. MATERIALS AND METHODS Animals were divided randomly into 5 groups and kept at 5 animals per group. The first group was served as a control group and administered with vehicle orally; the group II administered with DM (14 mg kg-1; 1/25 LD50) orally. Animals of group III, IV and V were administered with 100 mg kg-1 of CE extract, 200 mg kg-1 of CE extract and 100 mg kg-1 Liv-52 orally half hour before DM administration, respectively. All the previous administrations were repeated daily for 21 days. Data were analyzed by one-way ANOVA using SPSS. RESULTS The DM caused a statistically significant increase in the serum level of liver enzymes (AST, ALT, ALP) when compared to control animals, whereas CE and Liv-52 pre-treatment to the DM-intoxicated animals resulted in a significant normalization of the activities of enzymes. Similarly, a significant increase in lipid peroxidation (LPO) level, while induced significant decreases in the activities of liver catalase (CAT) and glutathione-S-transferase (GST). In contrast, co-administration of CE and Liv-52 to DM-treated animals restored most of these biochemical parameters to nearly normal levels. Histopathological examination of intoxicated animals showed many tissues alterations such as; vasodilation, hemorrhage, cytoplasmic vacuolization, inflammation and nuclear pyknosis indicating liver damage, while the animals received CE or Liv-52 showed less pathological effects when compared to animals treated with DM alone. CONCLUSION The biochemical and histological results confirmed the hepatoprotective effect of ethanolic extract of CE against DM-induced hepatotoxicity in male guinea pigs.

Investigation on medicinal plants' therapeutic potential has gained substantial importance in the discovery of novel effective and safe therapeutic agents. The present study is aimed at investigating the hepatoprotective potential of Seriphidium kurramense methanolic extract (SKM) against carbon tetrachloride- (CCl4-) induced hepatotoxicity in rats. S. kurramense is one of the most imperative plants for its various pharmacological activities. Therefore, this study was aimed at evaluating the hepatoprotective potential against CCl4-induced liver toxicity. The serum samples were analyzed for alanine aminotransferase (ALT) and aspartate aminotransferase (AST) together with the oxidative stress mediator levels as nitric oxide (NO), malondialdehyde (MDA), glutathione (GSH), reduced glutathione (GSH), and superoxide dismutase (SOD) as well as peroxidation and H2O2 activity. CCl4 administration resulted in an elevated free radical generation, altered liver marker (AST and ALT) enzymes, reduced antioxidant enzyme, and increased DNA damage. Methanolic extract of S. kurramense decreased CCl4-induced hepatotoxicity by increasing the antioxidant status and reducing H2O2 and nitrate content generation as well as reducing DNA damage. Additionally, SKM reversed the morphological alterations induced by CCl4 in the SKM-treated groups. These results demonstrated that SKM displayed hepatoprotective activity against CCl4-induced hepatic damage in experimental rats.

Tobacco (Nicotiana tabacum) is an herbaceous plant originating from South America and processed into cigarettes for consumption. Polyphenols are considered vital components of tobacco in view of their contribution to antioxidant properties. This study aimed to determine the phenolic compounds in different tobacco varieties by applying cold extraction with methanol and distilled water. The extracts were screened for phenolic compound diversity and distribution as well as their antioxidant potential in different tobacco varieties. The results showed that the methanolic extract of tobacco SP-28 exhibited the highest value in the total phenolic content (24.82 ± 0.07 mg GAE/gd.w.) and total flavonoid content (4.42 ± 0.01 mg QE/gd.w.), while the water extract of tobacco SN-2 exhibited the highest value in the total condensed tannin (1.12 ± 0.03 mg CE/gd.w.). The radical scavenging capacities of tobacco SP-28 were relatively high in DPPH (18.20 ± 0.01 mg AAE/gd.w.) and FRAP (3.02 ± 0.10 mg AAE/gd.w.), whereas the ABTS value was the highest in tobacco SN-2 (37.25 ± 0.03 mg AAE/gd.w.), and the total antioxidant capacity was the highest in tobacco SN-1 (7.43 ± 0.18 mg AAE/gd.w.). LC-ESI-QTOF-MS/MS identified a total of 49 phenolic compounds, including phenolic acids (14), flavonoids (30), and other polyphenols (5) in four different tobacco varieties. Tobacco SP-28 showed the highest number of phenolic compounds, especially enriched in flavones. Our study highlights the antioxidant potential of tobacco extracts and reveals the phenolic distribution among different tobacco varieties that could support tobacco utilization in different pharmaceutical industries.

Traditionally, Diospyros lotus Linn is used for insomnia and other associated disorders. Insomnia is a worldwide disorder with different etiology which is treated with different synthetic medicine associated with addiction. Natural products are generally devoid of such addition with good efficacy. Current research was conducted to evaluate the sedative and hypnotic effects of dimeric naphthoquinones such as dinaphthodiospyrol A (1), dinaphthodiospyrol B (2), dinaphthodiospyrol C (3), dinaphthodiospyrol D (4), dinaphthodiospyrol E (5) and dinaphthodiospyrol F (6) isolated from the chloroform fractions of D. lotus. The sedative and hypnotic effects at the dose of 5 and 10 mg/kg (each compound) were assessed through open field and phenobarbital induced sleep test, respectively. In the case of open field test the administration of tested compounds significantly hindered the movement of animals, while in case of hypnotic effect the tested samples significantly improved the onset and duration of sleep as compared to control. The overall effects were in a dose dependent manner. The compounds were also assessed for acute toxicity, but no toxicity was observed. In this regard, our research triumphantly announced the strong chemical base for the folkloric values of the plant with their fringe benefits and implemented a platform for further aspects of mechanistic and clinical studies. A possible mechanism of in vivo inhibition was studied by using docking simulations on GABA receptors. Binding orientations and types of interactions revealed that a possible mechanism behind these pharmacological actions might be interaction with GABA receptors.

This study was aimed to evaluate the efficiency of Sargassumpolycystum and nucleotides- supplemented diets to improve immune response and cold-tolerance of juvenile Litopenaeus vannamei. Four treatments were evaluated: T1, the control, shrimp received only a basal diet, T2, a basal diet with 500 ppm nucleotides, T3, a basal diet with 500 ppm S.polycystum powdered, T4, a basal diet with 500 ppm nucleotides and 500 ppm S.polycystum powdered. Shrimp were fed experimental diets for 56 days. Results revealed shrimp fed T4 diet exhibited the best significant improvement in water quality, survival, growth, and feed utilization indices followed by T2, and T3, while T1 showed the worst values. Additionally, nonspecific immune responses (phagocytosis (%), lysozyme, phenoloxidase, super oxide dismutase (SOD) activity, total nitric oxide) were improved with 1.7–3.2-fold in T4 higher than T1. Histomorphology of hepatopancreas in T4 showed the most increased activation of the hepatic glandular duct system compared with the other treatments. Moreover, nucleotides/seaweed-supplemented diets upregulated relative expression of cMnSOD, Penaeidin4, and heat shock protein70 (HSP70) genes, while translationally controlled tumor protein (TCTP) was downregulated. In conclusion, the synergistic effects of both S. polycystum and nucleotides have many advantages as a growth promoter, immunostimulant, antimicrobial, and cold-tolerant stimulant to L. vannamei.

The Sophora mollis is one of the best anti-inflammatory, antioxidant, and anticancerous plant; therefore, the isolated chemical constituents, that is, scopoletin (1), pinitol (2), 2-propenoic acid, 3-(3,4-dihydroxyphenyl)-octacosyl ester (3), betulin (4), and β-sitosterol glucoside (5) were tested for these folklores. The structures of the isolated compounds were confirmed by 1H NMR, 13C NMR, 2D-NMR, and mass spectral data. The anti-inflammatory, anticancer, antiglycation, and antioxidant activities of compounds 1–5 were evaluated using different assays. Compound 1 exhibited significant anti-inflammatory effect as it reduced edema of the paw (83.98%), which is more potent than the standard drug (ibuprofen) (which showed an inhibition percentage of 73.22% a), followed by compound 3. Furthermore, compound 3 showed significant free-radical scavenging activity using the 2,2-diphenyl-1-picryl-hydrazyl-hydrate (DPPH) free-radical assay. Percentage inhibition of DPPH recorded was 95.646 ± 0.003, 94.766 ± 0.014, and 94.516 ± 0.011% at concentrations of 400, 200, and 100 μg/mL, respectively. Evaluation of anticancer activity of isolated compounds reveals weak effect against HeLa and 3T3 cell lines. Docking studies of the most active compound into the binding sites of cyclooxygenase isoforms showed a better antagonistic potential against COX-1 than the COX-2 isoform.

At the same time, nutrient composition and antioxidant compounds are different between lettuce varieties, especially for green and red lettuce types. The benefit of lettuce consumption depends on its composition, particularly antioxidants, which can function as nutrients. The health benefits rely on their biochemical effect when reaching the bloodstream. Some components can be released from the food matrix and altered in the digestive system. Indeed, the bioaccessibility of lettuce is measuring the quantity of these compounds released from the food matrix during digestion, which is important for health-promoting features. Extraction of bioactive compounds is one of the new trends observed in lettuce and is necessarily used for several application fields. Therefore, this review aims to demonstrate the nutritional value of lettuce and its pharmacological properties. Due to their bioaccessibility and bioavailability, the consumer will be able to comprehensively understand choosing a healthier lettuce diet. The common utilization pattern of lettuce extracted nutrients will also be summarized for further direction.

Dracaena cinnabari (D. cinnabari) is an endemic plant located in Socotra Island, Yemen. Deep red resin attained from different plant species including D. cinnabari is commonly known as dragon’s blood. In folk medicine, it is prescribed for the treatment of traumatic dermal, dental, and eye injuries as well as blood stasis, pain, and gastrointestinal diseases in humans. Numerous studies have investigated that this resinous medicine has antidiarrheal, antiulcer, antimicrobial, antiviral, antitumor, anti-inflammatory, analgesic, wound healing, and antioxidant activity. Several phytochemicals have been isolated from D. cinnabari, including the biflavonoid cinnabarone, triflavonoids, metacyclophanes, chalcones, chalcanes, dihydrochalcones, sterols, and terpenoids. The present review highlights the structures and bioactivities of main phytochemicals isolated from D. cinnabari regarding the botany and pharmacological effects of the resin derived from this plant.

The use of insects as a feasible and useful natural product resource is a novel and promising option in alternative medicine. Several components from insects and their larvae have been found to inhibit molecular pathways in different stages of cancer. This study aimed to analyze the effect of aqueous and alcoholic extracts of Vespa orientalis larvae on breast cancer MCF7 cells and investigate the underlying mechanisms. Our results showed that individual treatment with 5% aqueous or alcoholic larval extract inhibited MCF7 proliferation but had no cytotoxic effect on normal Vero cells. The anticancer effect was mediated through (1) induction of apoptosis, as indicated by increased expression of apoptotic genes (Bax, caspase3, and p53) and decreased expression of the anti-apoptotic gene Bcl2, (2) suppression of intracellular reactive oxygen species, (3) elevation of antioxidant enzymes (CAT, SOD, and GPx) and upregulation of the antioxidant regulator Nrf2 and its downstream target HO-1, (4) inhibition of migration as revealed by in vitro wound healing assay and downregulation of the migration-related gene MMP9 and upregulation of the anti-migratory gene TIMP1, and (5) downregulation of inflammation-related genes (NFκB and IL8). The aqueous extract exhibited the best anticancer effect with higher antioxidant activities but lower anti-inflammatory properties than the alcoholic extract. HPLC analysis revealed the presence of several flavonoids and phenolic compounds with highest concentrations for resveratrol and naringenin in aqueous extract and rosmarinic acid in alcoholic extract. This is the first report to explain the intracellular pathway by which flavonoids and phenolic compounds-rich extracts of Vespa orientalis larvae could induce MCF7 cell viability loss through the initiation of apoptosis, activation of antioxidants, and inhibition of migration and inflammation. Therefore, these extracts could be used as adjuvants for anticancer drugs and as antioxidant and anti-inflammatory agents.

The present study evaluated the possible synergistic impacts of sodium butyrate and Spirulina platensis as a feed additive on growth performance, carcass composition, blood health, and intestinal histomorphology of Nile tilapia (Oreochromis niloticus).
