An aqueous crude extract from Solanum nigrum lyophilized leaves was evaluated for its phytochemical composition and antifungal activity against Penicillium digitatum, the causative agent of green mold of citrus fruit. Phytochemical analysis of the extract revealed the presence of some bioactive substances such as alkaloids, tannins, flavonoids, saponins, etc. The extract showed a remarkable inhibition zone against the pathogen in agar well diffusion assays carried out in Petri plates. Storage of the extract at 4 °C for 60 days had no effect on its in vitro antifungal activity. Further, the extract was tested for its in vivo (preventive- and curative treatments) antifungal activity on lemons wound-inoculated with P. digitatum. An important preventive antifungal effect was observed after 7 days of storage (100% of inhibition), although this activity decreased after 14 and 21 days (85.71 and 57.14% of inhibition, respectively). A slight curative antifungal activity was observed only after 7 days of storage (14.29% of inhibition). Preliminary findings from this study may contribute to the development of new antifungal agents to protect the lemon fruits from postharvest fungal diseases.

Aim of the Important Plant Area (IPA) program - integral part of the CBD Global Strategy for Plant Conservation - is the constitution of a network of the most important natural or semi-natural sites for wild plant and fungi conservation. Giving lower plants and fungi the same importance as vascular plants, the program provides to individual countries a considerable freedom of action in the implementation of IPAs number. Three main principles rule the site identification: Criterion A, based on species contribution; Criterion B, based on floristic richness in relation to the biogeographic zone; Criterion C, based on threatened habitats. Italian botanists from different areas of research responded to the call comprehensively, including all the groups of plants in the definition of the IPAs. Nevertheless, experts in lichens, fungi and freshwater algae found some operational difficulties, mostly due to the incomplete knowledge of species distribution, the lack of updated databases and official red lists. In this paper, the methodological approach used to include Italian lichens in the IPA definition is discussed. Seventy two taxa were selected as species of national interest and 21 areas critical for long-term preservation of lichen populations were proposed, including an archaeological area considered representative of cultural heritages as a lichen complex habitat.

© 2021 The Authors., Aims: Understanding fine-grain diversity patterns across large spatial extents is fundamental for macroecological research and biodiversity conservation. Using the GrassPlot database, we provide benchmarks of fine-grain richness values of Palaearctic open habitats for vascular plants, bryophytes, lichens and complete vegetation (i.e., the sum of the former three groups). Location: Palaearctic biogeographic realm. Methods: We used 126,524 plots of eight standard grain sizes from the GrassPlot database: 0.0001, 0.001, 0.01, 0.1, 1, 10, 100 and 1,000 m and calculated the mean richness and standard deviations, as well as maximum, minimum, median, and first and third quartiles for each combination of grain size, taxonomic group, biome, region, vegetation type and phytosociological class. Results: Patterns of plant diversity in vegetation types and biomes differ across grain sizes and taxonomic groups. Overall, secondary (mostly semi-natural) grasslands and natural grasslands are the richest vegetation type. The open-access file ”GrassPlot Diversity Benchmarks” and the web tool “GrassPlot Diversity Explorer” are now available online (https://edgg.org/databases/GrasslandDiversityExplorer) and provide more insights into species richness patterns in the Palaearctic open habitats. Conclusions: The GrassPlot Diversity Benchmarks provide high-quality data on species richness in open habitat types across the Palaearctic. These benchmark data can be used in vegetation ecology, macroecology, biodiversity conservation and data quality checking. While the amount of data in the underlying GrassPlot database and their spatial coverage are smaller than in other extensive vegetation-plot databases, species recordings in GrassPlot are on average more complete, making it a valuable complementary data source in macroecology., GrassPlot development has been supported by the Bavarian Research Alliance (BayIntAn_UBT_2017_58), the Eurasian Dry Grassland Group (EDGG) and the International Association for Vegetation Science (IAVS); IB, CorM, JAC, IGM, DGM, MHe, DL and MTo were supported by the Basque Government (IT936‐16); CorM, IAx, MCh, JDa, PD, MHá, ZL, ZPr, EŠ and LT were supported by the Czech Science Foundation (19‐28491X); TR was supported by the Estonian Research Council (PUT1173); RJP was funded by the Strategic Research Programme of the Scottish Government’s Rural and Environmental Science and Analytical Services Division”; SBa was supported by the GINOP‐2.3.2‐15‐2016‐00019 project; GFi was partially supported by the MIUR initiative “Department of excellence” (Law 232/2016)"; BJA was funded by the Spanish Research Agency (grant AEI/ 10.13039/501100011033); AK, VB, IM, DS, IV and DV were supported by the National Research Foundation of Ukraine (2020.01/0140); MP and AH were supported by the Estonian Research Council (PRG874, PRG609), and the European Regional Development Fund (Centre of Excellence EcolChange); Data collection of HCP was funded by FORMAS (Swedish Research Council for Environment, Agricultural Science and Spatial Planning) and The Swedish Institute; JR was supported by the Czech Science Foundation (grant No. 20‐09895S) and the long‐term developmental project of the Czech Academy of Sciences (RVO 67985939); ATRA was funded by the Grant of Excellence Departments, MIUR‐Italy (ARTICOLO 1, COMMI 314 – 337 LEGGE 232/2016); JMA was supported by Carl Tryggers stiftelse för vetenskaplig forskning and Qatar Petroleum; AAli was supported by the Jiangsu Science and Technology Special Project (Grant No. BX2019084), and Metasequoia Faculty Research Startup Funding at Nanjing Forestry University (Grant No. 163010230), and he is currently supported by Hebei University through Faculty Research Startup Funding Program; ZB was supported by the NKFI K 124796 grant; The GLORIA‐ Aragón project of JLBA was funded by the Dirección General de Cambio Climático del Gobierno de Aragón (Spain); MCs and LDem were supported by DG Environment through the European Forum on Nature Conservation and Pastoralism and Barbara Knowles Fund, in collaboration with Pogány‐havas Association, Romania; JDa was partially supported by long‐term research development project no. Aim Species-area relationships (SARs) are fundamental scaling laws in ecology although their shape is still disputed. At larger areas, power laws best represent SARs. Yet, it remains unclear whether SARs follow other shapes at finer spatial grains in continuous vegetation. We asked which function describes SARs best at small grains and explored how sampling methodology or the environment influence SAR shape. Location Palaearctic grasslands and other non-forested habitats. Taxa Vascular plants, bryophytes and lichens. Methods We used the GrassPlot database, containing standardized vegetation-plot data from vascular plants, bryophytes and lichens spanning a wide range of grassland types throughout the Palaearctic and including 2,057 nested-plot series with at least seven grain sizes ranging from 1 cm(2) to 1,024 m(2). Using nonlinear regression, we assessed the appropriateness of different SAR functions (power, power quadratic, power breakpoint, logarithmic, Michaelis-Menten). Based on AICc, we tested whether the ranking of functions differed among taxonomic groups, methodological settings, biomes or vegetation types. Results The power function was the most suitable function across the studied taxonomic groups. The superiority of this function increased from lichens to bryophytes to vascular plants to all three taxonomic groups together. The sampling method was highly influential as rooted presence sampling decreased the performance of the power function. By contrast, biome and vegetation type had practically no influence on the superiority of the power law. Main conclusions We conclude that SARs of sessile organisms at smaller spatial grains are best approximated by a power function. This coincides with several other comprehensive studies of SARs at different grain sizes and for different taxa, thus supporting the general appropriateness of the power function for modelling species diversity over a wide range of grain sizes. The poor performance of the Michaelis-Menten function demonstrates that richness within plant communities generally does not approach any saturation, thus calling into question the concept of minimal area. We thank all vegetation scientists who carefully collected multi‐ scale plant diversity data from Palaearctic Grasslands available in GrassPlot. Despite the importance for scientific and conservation purposes, the knowledge of the Italian territory is far from exhaustive. New chorological data for 87 vascular taxa regarding the central-southern part of Italy and its two main islands (Sicilia and Sardegna) are presented. Among these taxa, Epilobium nummularifolium, Metrosideros excelsa, and Salvinia minima are recorded as casual aliens for the first time in Europe (excluding Azores and Madeira for M. excelsa), while Cyclamen balearicum and Polygala rupestris are reported for the first time and confirmed for Italian native flora, respectively. Furthermore, several taxa are new or confirmed at regional level. Finally, Lathyrus cirrhosus, Urginea fugax, and Linum tenuifolium are excluded from Italy, continental and peninsular Italy, and Sardegna, respectively.

Summary We attempted to elucidate the roles of β-diversity components and similarity in shaping Lobaria pulmonaria lichen communities in Italian forests to provide scientific tools for improving their long-term conservation. A total of 20 sites were selected by stratified random sampling, including five chestnut forests, seven beech–silver fir forests and eight oak forests distributed across 10 administrative regions of Italy. Site selection was based on a national data base including all the available records of L. pulmonaria in Italy. In each forest, four plots were randomly placed, and in each plot, 5–6 trees were randomly selected for lichen sampling. For each forest and plot, meaningful predictors of lichen patterns were quantified. In particular, factors indicative of both geographic conditions and forests structure were considered. The SDR simplex approach was used to estimate the relative importance of similarity (S), relative species replacement (R) and relative richness difference (D) to β-diversity and similarity, while a hierarchical partitioning (HP) method was used to evaluate the relative importance of environmental predictors in explaining their patterns. A total of 201 lichens and three nonlichenized fungi were found, including 51 species of conservation concern. The components of β-diversity and similarity contributed to shaping L. pulmonaria communities at both forest and plot scales across Italy, resulting in nearly random compositions, that is, the species set on each tree was a random sample from the available species pool. Species replacement and similarity were generally associated with forest structure predictors, while richness difference was mainly associated with geographic predictors. The implication of this is that the long-term conservation of L. pulmonaria communities could be promoted by maintaining scattered nodes and appropriate habitat traits, especially in large forested landscapes where species turnover is higher. Synthesis. In this work, we used a new approach for analysing a countrywide data set improving the ecological understanding of the dynamics regulating epiphytic communities. In particular, this study improves the understanding of the contribution of different components of diversity across two spatial scales and evaluates the relative importance of environmental predictors in explaining variation of each diversity component.

Similarly to other Mediterranean regions, Italy is expected to experience dramatic climatic changes in the coming decades. Do to their poikilohydric nature, lichens are among the most sensitive organisms to climate change and species requiring temperate-humid conditions may rapidly decline in Italy, such in the case of the epiphytic Lobaria species that are confined to humid forests. Our study, based on ecological niche modelling of occurrence data of three Lobaria species, revealed that in the next decades climate change will impact their distribution range across Italy, predicting a steep gradient of increasing range loss across time slices. Lobaria species are therefore facing a high extinction risk associated with reduction of their range. The current patterns indicate that only L. pulmonaria still has a continuous distribution across Italy, with potential contact between Apennine and Alpine populations. This situation is consistent with the wider climatic niche of this species, still offering a major opportunity for its successful long-term conservation. Results (a) support the inclusion of the three Lobaria species in European conservation policies, such as the Habitat Directive, and (b) warn against an over-estimation of the indicator power of single flagship species to establish conservation priorities for lichens, indicating that even lichens with peculiar and similar climatic envelopes may fail to co-occur within a given forest stand. A multiple indicator approach could provide more useful tools for a community-based conservation strategy for epiphytes.

This work aimed to investigate whether the insecticide acephate (125 or 250 mg/kg b.w.) or diflubenzuron (752 or 1075 mg/kg b.w.), two of the most widely used pesticides worldwide, impairs CYP-linked murine metabolism in liver, kidney and lung microsomes after repeated (daily, for three consecutive days) i.p. administration. The regio- and stereo-selective hydroxylation of testosterone was used as multibiomarker of different CYP isoforms. Both gender and tissue specific effects were observed. Lung was the most responsive tissue to induction by lower diflubenzuron dose, as exemplified by the marked increase of testosterone 7alpha-hydroxylation (CYP2A) (up to 13-fold) in males. Higher dose produced a generalized inactivation. At the lower dose acephate induced 6beta- (CYP3A1/2, liver) as well as 2beta- (CYP2B1/2, kidney) hydroxylase activities ( approximately 5 and approximately 4-fold increase, respectively) in males. In females, a marked suppression of the various hydroxylations was observed. At 250 mg/kg of acephate, animals did not survive. Induction of the most affected isoforms was sustained by immunoblotting analysis. Corresponding human CYP modulations might disrupt normal physiological functions related to these enzymes. Furthermore, the co-mutagenic and promoting potential of these pesticides, phenomena linked to CYP upregulation (e.g. increased bioactivation of ubiquitous pollutants and generation of oxygen free radicals) are of concern for a more complete definition of their overall toxicological potential.
