Im Rahmen der Studie zur Erstellung eines UKT-spezifischen, gesunden Normkollektives, wurden 30 Probanden zwischen 20 und 70 Jahren, mit gleichem Anteil von Frauen und Männern, rekrutiert. Nach klinischer Untersuchung des Gesundheitszustandes wurden die Probanden am 3T-MRT-Scanner gemessen. Die MRT-Daten wurden von zwei Ärzten auf Vorhandensein von Gehirnkrankheiten oder - anomalien neuroradiologisch überprüft. Danach wurden die Basalganglien per Hand mithilfe 3D Slicer sowie automatisiert im FreeSurfer-Programm segmentiert. Bei beiden Methoden wurde das absolute sowie relative Volumen als Prozentsatz des Gehirnvolumens für jeden einzelnen Basalkern berechnet. Wie auch in vielen anderen Studien konnte bestätigt werden, dass ein negativer, mittelstarker linearer Zusammenhang zwischen dem Alter und dem Volumen des N. caudatus sowie des Putamens besteht. Das Alter zeigte im Rahmen dieser Untersuchung keinen signifikanten Einfluss auf das Pallidumvolumen. In der Literatur wird für das Pallidum eine geringere Atrophierate mit großer Varianz beschrieben, so dass hier vermutlich nur in Längsschnittstudien und in großen Querschnittsstudien Alterseffekte gezeigt werden können. Die altersbedingte Schrumpfung des Striatums findet bei Männern schneller statt als bei Frauen, wobei die Volumen-Alter-Korrelationen bei Frauen nicht signifikant sind. Insgesamt stimmen die Ergebnisse gut mit anderen Querund Längsschnittstudien überein. Die Korrelationen zwischen dem Alter und den absoluten sowie relativen Intensitätswerten der Basalkerne wurden jeweils in den T1-, T2- und FLAIR-Sequenzen ausgewertet. Die relativen Intensitätswerte (als Relation vom Intensitätsmittelwert des jeweiligen Kernes zum Intensitätsmittelwert des Spleniums als Kontrollwert) zeigten keine relevanten Korrelationen im Gegensatz zu den absoluten Werten. Trotz großer Streubreite der Werte wurde jedoch bilateral in der FLAIR-Sequenz eine signifikante, altersbedingte Intensitätsminderung beim Nucleus caudatus und beim Putamen und in der T2-Sequenz beim Putamens beobachtet. Wenn man die Mittelvolumina der Basalganglien aus den in 3D Slicer handsegmentierten und in FreeSurfer automatisiert segmentierten Ergebnissen vergleicht, wird in der automatisierten Methode das Volumen des beidseitigen Nucleus caudatus kleiner, das des rechten Putamens und des linken Pallidums signifikant größer berechnet als in der manuellen Methode. Trotz dieser Volumenunterschiede sind die positiven Korrelationen zwischen automatischer und manueller Segmentierung stark und hoch signifikant für den bilateralen Nucleus caudatus und das Putamen. Obwohl beide Methoden gut miteinander korrelieren und die automatisierte Segmentierungstechnik insbesondere wegen der Zeitersparnis interessanter für die Forschung erscheint, bleibt die Handsegmentierung weiterhin der Goldstandard. Die Alterungseffekte wurden auch in den automatisch abgegrenzten Basalganglien ausgewertet und mit handsegmentierten Resultaten verglichen. Insbesondere mit zunehmendem Alter laufen die Ergebnisse der Handsegmentierung und der automatischen Segmentierung auseinander, was sich in einer geringen Volumenunterschätzung im bilateralen Caudatus bei der automatischen Segmentierung und einer stärkeren jährlichen Volumenabnahme im bilateralen Putamen bei Durchführung der Handsegmentierung äußert. Diese Ergebnisse korrelieren gut mit anderen Studien. Eine mögliche Erklärung für diesen Effekt ist die gezeigte Intensitätsentwicklung mit dem Alter: Durch die zunehmende Intensitätsänderung kommt es für entsprechende Algorithmen zu einer schlechteren Abgrenzbarkeit oder Erkennbarkeit. Andere Erklärungen wie vermehrte Bewegung im Alter sind im vorliegenden Kollektiv nicht zu erwarten. Für zukünftige Studien zum Volumen der Basalganglien wird daher empfohlen, die Veränderung der Intensitätswerte und die dadurch eventuell bedingte systematische Abweichung bei der automatischen Segmentierung zu berücksichtigen.

Oxidative stress (OS), mitochondrial dysfunction, and dysregulation of alpha-synuclein (aSyn) homeostasis are key pathogenic factors in Parkinson’s disease. Nevertheless, the role of aSyn in mitochondrial physiology remains elusive. Thus, we addressed the impact of aSyn specifically on mitochondrial response to OS in neural cells. We characterize a distinct type of mitochondrial fragmentation, following H2O2 or 6-OHDA-induced OS, defined by spherically-shaped and hyperpolarized mitochondria, termed “mitospheres”. Mitosphere formation mechanistically depended on the fission factor Drp1, and was paralleled by reduced mitochondrial fusion. Furthermore, mitospheres were linked to a decrease in mitochondrial activity, and preceded Caspase3 activation. Even though fragmentation of dysfunctional mitochondria is considered to be a prerequisite for mitochondrial degradation, mitospheres were not degraded via Parkin-mediated mitophagy. Importantly, we provide compelling evidence that aSyn prevents mitosphere formation and reduces apoptosis under OS. In contrast, aSyn did not protect against Rotenone, which led to a different, previously described donut-shaped mitochondrial morphology. Our findings reveal a dichotomic role of aSyn in mitochondrial biology, which is linked to distinct types of stress-induced mitochondrial fragmentation. Specifically, aSyn may be part of a cellular defense mechanism preserving neural mitochondrial homeostasis in the presence of increased OS levels, while not protecting against stressors directly affecting mitochondrial function. peerReviewed

The symptomatic treatment options that are currently available do not slow or halt disease progression. This highlights the need of a better understanding of disease mechanisms and disease models. The generation of newborn neurons in the adult hippocampus and in the subventricular zone/olfactory bulb system is affected by many different regulators and possibly involved in memory processing, depression, and olfaction, symptoms which commonly occur in PD. The pathology of the adult neurogenic niches in human PD patients is still mostly elusive, but different preclinical models have shown profound alterations of adult neurogenesis. Alterations in stem cell proliferation, differentiation, and survival as well as neurite outgrowth and spine formation have been related to different aspects in PD pathogenesis. Therefore, neurogenesis in the adult brain provides an ideal model to study disease mechanisms and compounds. In addition, adult newborn neurons have been proposed as a source of endogenous repair. Herein, we review current knowledge about the adult neurogenic niches in PD and highlight areas of future research.

Rheumatoid arthritis (RA) is a chronic inflammatory systemic disease that primarily affects the joints but may involve every organ of the human organism. RA has a prevalence of nearly 1% in the worldwide population. The disease is associated with significant disability and increased mortality if insufficiently treated. The pathogenesis of RA is not fully understood but is characterized by a strong genetic contribution and by abnormalities in the function of CD4 T cell subsets, which drive an adaptive autoimmune response. The cytokine interleukin 4 (IL-4), acting through the IL-4 receptor (IL-4R), crucially regulates CD4 T cell subset development and function, thereby contributing to RA development and/or progression. To delineate pathogenic mechanisms of RA and to identify early risk factors associated with the disease, the contribution of the IL-4R and the immunomodulatory role of IL-4 signaling in RA were investigated in this thesis. An association of RA with a region on the short arm of chromosome 16 containing the IL4R gene was previously shown. Therefore, in the first part of this work, the association of two single nucleotide polymorphisms (SNPs) in the IL-4 receptor gene (IL4R) with RA susceptibility and severity was investigated in a case-control study. The two IL4R SNPs analyzed lead to amino acid substitutions of isoleucine (I) to valine (V) at position 50 (I50V SNP) and of glutamine (Q) to arginine (R) at position 551 (Q551R SNP). They are located within the functionally important IL- 4- and STAT6-binding regions of the IL-4R, respectively, and are thought to modulate IL-4 signaling. Allelic distribution at the I50V and Q551R IL4R SNPs, determined by allele specific PCR analysis in 471 RA patients and 371 healthy controls, revealed no major differences in allele, genotype, and haplotype frequencies, supporting the conclusion that IL4R is not associated with RA susceptibility. However, a significant impact of the I50V SNP on disease severity was determined. The V50 allele was strongly associated with early aggressive erosive RA independently of other diagnostic factors previously associated with severe RA (e.g. the presence of the shared epitope [SE] and of the rheumatoid factor [RF]) and showed a high predictive power for developing erosions within the first two years of RA. These data identified the IL4R gene as a novel genetic marker for early severe RA. IL-4 controls T cell differentiation via the IL-4R by inhibiting Th1 and promoting Th2 and Treg cell differentiation. Restricted IL-4 signaling in T cells might cause uncontrolled expansion of pro-inflammatory Th1 effector cells and impaired Th2 and Treg cell development that together may drive persistent rheumatoid inflammation. Therefore, in the second part of this work, the functional importance of the I50V and Q551R IL4R SNPs for IL-4 signaling in CD4 T cells was investigated. The effect of IL-4 on the function of CD4 T cells from healthy individuals expressing different IL4R alleles was assessed in vitro. The Q551R SNP only slightly influenced IL-4-mediated functions of CD4 T cells. In marked contrast, the I50V SNP had a profound effect on IL-4 signaling in CD4 T cells. While not affecting IL-4R expression and cell proliferation in response to IL-4, the V50 allele of the I50V IL4R SNP was associated with decreased responsiveness of CD4 T cells to IL-4 in terms of STAT6 phosphorylation, GATA3 mRNA upregulation, and IL-12Râ2 mRNA and protein downmodulation. These events are central mechanisms by which IL-4 regulates CD4 T cell differentiation. Consequently, V50 homozygosity was associated with significantly diminished IL-4-mediated inhibition of Th1 cell development and an impaired supportive effect of IL-4 on Th2 cell differentiation. Together, these data indicate a regulation of CD4 T cell function by the I50V IL4R SNP alleles that might underline the association of the V50 allele with early erosive RA. To investigate yet largely unknown mechanisms underlying regulatory T cell (Treg) development controlled by IL-4, gene expression profiling of developing Treg cells induced by IL-4 in vitro was analyzed in the third part of the thesis using DNA microarray technology. A number of genes were identified during different stages of Treg development that were specifically expressed in Tregs. The majority of the genes differently regulated in developing Treg cells at early stages are involved in cell cycle and cell proliferation control, whereas genes differently regulated later in development encode for surface receptors, enzymes, transcription factors, and signal transduction molecules. The adhesion molecule CEACAM1 and the prohormone PMCH were identified as early and late upregulated genes during Treg development, respectively. In line with the gene chip analysis, PMCH and CEACAM1 were detectable on mRNA level by real-time PCR in independent experiments, and CEACAM1 was highly expressed on activated mature Tregs as determined by flow cytometry. The results show that IL-4 controls a broad spectrum of genes at different stages of Treg development, suggesting their importance for immune homeostasis. The precise role of the identified genes in Treg development and/or functions has to be further analyzed. In summary, this study provides new aspects on the immunomodulatory role of IL-4: a number of IL-4-regulated genes participating in Treg cell development and an intriguing regulatory mechanism of CD4 T cell function by the alleles of the I50V SNP of the IL4R gene were delineated. The I50V IL4R SNP was identified as the first genetic marker in RA with a high predictive value for early joint destruction. It might be used to identify high-risk RA patients early in disease, before irreversible joint damage has occurred, and to adapt therapy individually. Die rheumatoide Arthritis (RA) ist eine chronisch entzündliche Systemerkrankung, die sich vor allem an den Gelenken manifestiert, aber jedes Organ des menschlichen Körpers befallen kann. Die Erkrankung tritt bei ca. 1% der Weltbevölkerung auf. Bei unzureichender Behandlung kann die Erkrankung zu erheblichen Funktionseinschränkungen und erhörter Mortalität führen. Die Pathogenese der RA ist noch nicht vollständig verstanden. Es gilt allerdings als sicher, dass die Krankheit eine sehr starke genetische Komponente hat und durch funktionelle Abnormalitäten der CD4 T-Zellsubtypen gekennzeichnet ist. Das Zytokin Interleukin-4 (IL-4), das über seinen IL-4- Rezeptor (IL-4R) wirkt, reguliert entscheidend die Differenzierung und die Funktion von CD4 TZellsubtypen und kann damit zur Entwicklung der RA beitragen. Um die pathologischen Mechanismen der RA einzugrenzen und die frühen Risikofaktoren, die mit der Erkrankung assoziiert sind, zu identifizieren, wurden in dieser Arbeit die Rolle des IL-4Rs und die immunomodulierende Rolle des IL-4 Signalwegs bei der RA untersucht. Die Assoziation zwischen der RA und einer Region auf dem kurzen Arm des Chromosoms 16, in der sich das IL-4-Rezeptor-Gen (IL4R) befindet, wurde in mehreren Studien gezeigt. Im ersten Teil dieser Arbeit wurde deshalb die Assoziation zwischen zwei Single Nucleotide Polymorphismen (SNP) in dem IL4R und der Prädisposition und dem Verlauf der RA in einer Fall-Kontroll-Studie untersucht. Die zwei untersuchten IL4R SNP führen zu einem Aminosäuren Austausch von Isoleuzin (I) zu Valin (V) in der Position 50 (I50V SNP) und von Glutamin (Q) zu Arginin (R) in der Position 551 (Q551R SNP). Sie befinden sich innerhalb der funktionell wichtigen IL-4- und STAT6-bindenden Regionen des IL-4Rs und sind vermutlich am IL-4- Signalweg beteiligt. Die Allel-Verteilung der IL4R SNP I50V und Q551R, die durch Allelspezifische PCR bei 471 RA-Patienten und 371 gesunden Kontrollen bestimmt wurde, zeigte keine signifikanten Unterschiede in Allel-, Genotyp- und Haplotyp-Frequenzen zwischen Patienten und Kontrollen. Aus diesem Ergebnis lässt sich schlussfolgern, dass der IL4R nicht mit einer Prädisposition für die RA assoziiert ist. Es wurde jedoch ein signifikanter Einfluss des I50V SNPs auf den Verlauf der Erkrankung gefunden. Für das V50 Allel wurde eine starke Assoziation mit früher erosiver RA nachgewiesen. Diese Assoziation war unabhängig von anderen diagnostischen Markern, die mit einem schweren Verlauf der RA assoziiert sind (shared epitope [SE] und Rheumafaktor [RF]), und hat einen hohen prognostischen Wert für die Voraussage einer erosiven Erkrankung innerhalb der ersten zwei Jahren der RA. Durch diese Ergebnisse wurde das IL4R Gen als neuer genetischer Marker für die frühe erosive RA identifiziert. IL-4 kontrolliert die T-Zelldifferenzierung mittels IL-4R durch Hemmung der Th1- und Förderung der Th2- und Treg-Zelldifferenzierung. Verminderte IL-4-Signalgebung in T-Zellen kann unkontrollierte Vermehrung von pro-inflammatorischen Th1-Effektorzellen und verringerte Th2- und Treg-Zellentwicklung verursachen, die zu persistierender rheumatoider Entzündung führen können. Deshalb wurde im zweiten Teil der Arbeit die funktionelle Bedeutung der I50V und Q551R IL4R SNP für die IL-4-Signalgebung in CD4 T-Zellen untersucht. Der Effekt von IL- 4 auf die CD4 T-Zellenfunktion von gesunden Spendern, die unterschiedliche IL4R-Allele exprimieren, wurde in vitro getestet. Der Q551R SNP beeinflusste die IL-4-gesteuerten Funktionen der CD4 T-Zellen nur in geringem Maße. Im Gegensatz dazu hatte der I50V SNP einen starken Effekt auf die IL-4-Signalgebung in CD4 T-Zellen. Während die IL-4R-Expression und Zellproliferation in Antwort auf IL-4 nicht beeinflusst wurden, war das V50-Allel des I50V IL4R SNPs mit verringerter Empfindlichkeit der CD4 T-Zellen auf IL-4 assoziiert. Dieser Effekt zeigte sich hinsichtlich der STAT6-Phosphorylierung, der GATA3 mRNA-Erhöhung und der Verringerung der IL-12Rß2 mRNA und des IL-12Rß2 Proteins. Durch diese zentralen Mechanismen reguliert IL-4 die CD4 T-Zelldifferenzierung. Die IL-4-gesteuerte Hemmung der Th1- und die Förderung der Th2-Zellentwicklung sind daher bei V50-Homozygotie signifikant vermindert. Diese Ergebnisse weisen auf eine Regulation der CD4 T-Zellfunktion durch die Allele des I50V IL4R SNPs hin und unterstreichen damit die Assoziation des decrease-in-function V50-Alleles mit früh erosiver RA. Um bisher weitgehend unbekannte Mechanismen der IL-4-gesteuerten regulatorischen T-Zell (Treg)-Entwicklung zu untersuchen, wurde im dritten Teil dieser Arbeit die Genexpression von sich unter IL-4 entwickelnden Treg-Zellen mittels DNA-Microarrays analysiert. Während der verschiedenen Stadien der Treg-Entwicklung konnte eine Vielzahl von Genen identifiziert werden, die spezifisch in sich entwickelnden Treg-Zellen exprimiert werden. Die Mehrheit der Gene, die an frühen Zeitpunkten der Treg-Entwicklung exprimiert werden, ist an der Regulation des Zellzyklus und der Zellproliferation beteiligt. Die Gene, die in späteren Entwicklungsstadien reguliert wurden, kodierten dagegen Oberflächenmoleküle, Enzyme, Transkriptionsfaktoren und Signaltransduktionsmoleküle. In der Treg-Entwicklung wurden das Adhäsionsmolekül CEACAM1 als früh hochreguliertes Gen und das Prohormon PMCH als spät hochreguliertes Gen gefunden. Die Hochregulation der PMCH und CEACAM1 mRNA ließ sich in unabhängigen Experimenten durch real-time PCR bestätigen. Außerdem wurde eine hohe CEACAM1- Expression auf aktivierten reifen Treg-Zellen mittels Durchflusszytometrie nachgewiesen. Die Ergebnisse zeigen deutlich, dass IL-4 an verschiedenen Zeitpunkten der Treg-Entwicklung ein breites Spektrum von Genen kontrolliert, was auf deren Bedeutung für die Immunhomeostase hindeutet. Die genaue Rolle der identifizierten Gene für die Treg-Entwicklung und/oder Funktion muss noch weiter untersucht werden. Diese Arbeit liefert neue Einblicke in die Immunmodulation durch IL-4. Eine Vielzahl von IL-4 regulierten Genen, die an der Treg-Zellentwicklung beteiligt sind, wurde bestimmt. Außerdem wurden neue Aspekte der IL-4-vermittelten Regulation der CD4 T-Zellfunktion identifiziert. Der I50V IL4R SNP ist der erste genetische Marker mit hohem prognostischem Wert für die frühe Gelenkzerstörung bei der RA. Dieser Marker könnte daher sehr nützlich sein, um das Risiko von Früherosionen bei RA Patienten zu diagnostizieren und entsprechende präventive therapeutische Maßnahmen zu ergreifen.

Background Synucleinopathies comprise a group of neurodegenerative diseases associated with abnormal accumulation of α-synuclein. One of the key factors that contribute to the progression of synucleinopathies is neuroinflammation. However, the role of lymphocytes in synucleinopathies like Parkinson’s disease (PD) remains largely unclear. Methods To investigate how lymphocytes impact synucleinopathies, human wild-type α-synuclein (WTS) transgenic mice were crossed with mice lacking mature lymphocytes (Rag2−/−). In this in vivo model, we quantified α-synuclein aggregation in the substantia nigra (SN) and striatum and determined the numbers of innate and adaptive immune cells in the central nervous system (CNS). The activation state of resident and infiltrated CNS myeloid cells (M1 vs. M2) was further classified by gene and protein expression analyses. The impact of T and B lymphocytes on the phagocytic activity of microglia in the presence of α-synuclein aggregates was addressed in BV2 microglia in vitro. Results Compared to WTS+ Rag2+/+ mice, where T but not B lymphocytes infiltrated the CNS, decreased amounts of α-synuclein aggregates were found in WTS+ Rag2−/− mice devoid of mature lymphocytes. The presence of T lymphocytes did not alter the number of Iba1+ microglia but increased the frequency of the CD11b+ CD45hi population in the CNS, indicative of an increased number of infiltrated macrophages. Moreover, the M1 phenotype was more prominent in WTS+ Rag2+/+ mice, whereas the M2 activation state was dominating in the absence of lymphocytes in WTS+ Rag2−/− mice. In vitro, in the presence of T but not B lymphocytes, significantly less α-synuclein was phagocytosed by BV2 microglia, further supporting the prevalence of the M1 phenotype in the presence of T lymphocytes. Conclusions Peripheral T lymphocytes strongly contribute to increased α-synuclein pathology via modulation of CNS myeloid cell function. In the presence of T lymphocytes, microglia phagocytosis of aggregated α-synuclein is reduced, which increases the severity of synucleinopathy.
