BackgroundObservational studies have shown that central obesity is associated with adverse cardiac structure and function. However, causal association between central obesity and left ventricular (LV) structure and function in preserved ejection fraction (EF) population is still uncertain.MethodsGenome-wide association studies summary data of waist circumference adjusted for body mass index (WCadjBMI) and waist-to-hip ratio adjusted for body mass index (WHRadjBMI) were selected as instrumental variables from the Genetic Investigation of Anthropometric Traits (GIANT) Consortium (n = 224,459). Outcome datasets for LV parameters including LV end-diastolic volume (LVEDV), LV end-systolic volume (LVESV), LV ejection fraction (LVEF), LV mass (LVM), and LV mass-to-end-diastolic volume ratio (LVMVR) were obtained from the participants without prevalent myocardial infarction or heart failure (LVEF ≥ 50%) in UK Biobank Cardiovascular Magnetic Resonance sub-study (n = 16,923). Two-sample Mendelian randomization (MR) was performed with the inverse-variance weighted (IVW) method as the primary estimate and with the weighted median and MR-Egger as the supplemental estimates. Sensitivity analysis was used to assess the heterogeneity and pleiotropic bias in the MR results.ResultsIn the IVW analysis, every 1-standard deviation (SD) higher WHRadjBMI was significantly associated with higher LVMVR (β = 0.4583; 95% confidence interval [CI]: 0.2921 to 0.6244; P = 6.418 × 10–8) and lower LVEDV (β = –0.2395; 95% CI: –0.3984 to –0.0807; P = 0.0031) after Bonferroni adjustment. No heterogeneity and horizontal pleiotropy were detected in the analysis. No association of WCadjBMI was found with LVEF, LVEDV, LVESV, LVM, or LVMVR.ConclusionOur findings provide evidence of significant causal association between WHRadjBMI and adverse changes in LV structure and function in preserved EF population.

Introduction: Coronary heart disease (CHD) is a dynamic process in which there are interactions between endothelial dysfunction, oxidative stress, and inflammatory responses. The aim of the present study was to investigate the function and mechanism of HSCARG in the treatment of CHD. Methods: Male apolipoprotein E/low-density lipoprotein receptor-deficient mice were given a high-fat diet with 21% fat and 0.15% cholesterol for the in vivo model. Human umbilical vein endothelial cells were incubated with angiotensin II for the in vitro model. HSCARG expression was inhibited in patients or mice with CHD. Results: HSCARG reduced oxidative stress in mice with CHD. HSCARG also reduced reactive oxygen species (ROS)-oxidative stress in the in vitro model. HSCARG induced p47phox expression in the in vitro model by NF-κB activity. The regulation of nuclear factor kappa B (NF-κB) activity or p47phox expression participates in the effects of HSCARG in CHD. Conclusion: Altogether, our data indicate that HSCARG reduced ROS-oxidative stress in in vivo and in vitro models of CHD via p47phox by NF-κB activity and may be a clinical target for CHD.

Biological applications of nanomaterials as delivery carriers have been embedded in traditional biomedical research for decades. Despite lagging behind, recent significant breakthroughs in the use of nanocarriers as tools for plant biotechnology have created great interest. In this Perspective, we review the outstanding recent works in nanocarrier-mediated plant transformation and its agricultural applications. We analyze the chemical and physical properties of nanocarriers determining their uptake efficiency and transport throughout the plant body.

Background. Studies have shown that the conventional left ventricular mechanical dyssynchrony (LVMD) parameters have their own statistical limitations. The purpose of this study is to extract new LVMD parameters from the phase analysis of gated SPECT MPI by deep learning to help CRT patient selection. Methods. One hundred and three patients who underwent rest gated SPECT MPI were enrolled in this study. CRT response was defined as a decrease in left ventricular end-systolic volume (LVESV) >= 15% at 6 +- 1 month follow up. Autoencoder (AE), an unsupervised deep learning method, was trained by the raw LV systolic phase polar maps to extract new LVMD parameters, called AE-based LVMD parameters. Correlation analysis was used to explain the relationships between new parameters with conventional LVMD parameters. Univariate and multivariate analyses were used to establish a multivariate model for predicting CRT response. Results. Complete data were obtained in 102 patients, 44.1% of them were classified as CRT responders. AE-based LVMD parameter was significant in the univariate (OR 1.24, 95% CI 1.07 - 1.44, P = 0.006) and multivariate analyses (OR 1.03, 95% CI 1.01 - 1.06, P = 0.006). Moreover, it had incremental value over PSD (AUC 0.72 vs. 0.63, LH 8.06, P = 0.005) and PBW (AUC 0.72 vs. 0.64, LH 7.87, P = 0.005), combined with significant clinic characteristics, including LVEF and gender. Conclusions. The new LVMD parameters extracted by autoencoder from the baseline gated SPECT MPI has the potential to improve the prediction of CRT response., 10 pages, 3 figures, will submit to journal of nuclear cardiology

A new type of biodegradable polyesteramide P(LA/AU) based on lactic acid and aminoundecanoic acid was synthesized by the melt polycondensation method. The copolymers obtained were characterized by FTIR, 1H-NMR, DSC, and XRD. The in vitro degradation behavior of the copolymers was studied using weighing, 1H-NMR, FTIR, DSC, and SEM. With the increase in aminoundecanoic acid units, the melting temperature and crystallinity increased, but the water absorption and degradation rate decreased accordingly. During degradation in vitro, the ester moiety decreased due to ester cleavage along the macromolecular main chain. As degradation proceeded, the melting temperature and crystallinity increased at first, then started to decrease because the crystalline phase was destroyed.
