Vaccines have become effective in protecting the health of a population. Vaccine development is a challenge, ensuring safety of the vaccine from its source until it is administered is another. One crucial part in the cold chain system is the transportation of the vaccine carriers from the health centers to poorly accessible communities. The Philippines’ Department of Health (DOH) Vaccine, Cold Chain and Logistics Management Manual of Operations recommends actions in order to safely deliver the vaccines with the use of WHO prequalified passive containers such as vaccine transport box and vaccine carrier. This research provides an option to vaccine carrier for use when delivering vaccines from health centers to poorly accessible communities where government health personnel are deployed to administer vaccines to newborn, bedridden, disabled and elderly with mobility issues. The use of the current vaccine carrier requires the use of at least six pieces of ice packs, trained and highly skilled health care personnel capable of manually taking, monitoring and recording vaccine temperature in the course of delivery to the vaccination site. This research output provides not only an alternative design to the vaccine carrier but also a prototype. The prototype is capable of maintaining vaccine temperature within the target range, minimizing risk of temperature damage; taking and recording of the temperature of the built-in dummy vaccine; and eliminating the opening and re-opening of the vaccine carrier’s lid in the process. The capability of the prototype in taking, monitoring and recording of the temperature was tested to last for at least a continuous 12-hour duration at a 3-second interval. The prototype can support the cold chain system requirement of temperature monitoring every 30 minutes during the delivery of vaccines from the health centers to poorly accessible communities.

NSTEACS is subset of ACS that may present with a wide degree of stenosis from normal vessels to severe obstruction. Identification of which population of NSTEACS that has normal vessels has attracted a great attention. Several trials on non-invasive imaging such as coronary CT have been largely investigated. Current available trials have showed that coronary CT is accurately identify significant stenosis in patients with NSTEACS thus can be used to rule out the disease and reduce the need and duration of unneeded antithrombotic. However, several limitations of the studies has to be taken into account when translating into clinical practice. Nevertheless, current evidence are showing promising results on the role of coronary CT in management of NSTEACS.

The ability to build in-depth cell signaling networks from vast experimental data is a key objective of computational biology. The spleen tyrosine kinase (Syk) protein, a well-characterized key player in immune cell signaling, was surprisingly first shown by our group to exhibit an onco-suppressive function in mammary epithelial cells and corroborated by many other studies, but the molecular mechanisms of this function remain largely unsolved. Based on existing proteomic data, we report here the generation of an interaction-based network of signaling pathways controlled by Syk in breast cancer cells. Pathway enrichment of the Syk targets previously identified by quantitative phospho-proteomics indicated that Syk is engaged in cell adhesion, motility, growth and death. Using the components and interactions of these pathways, we bootstrapped the reconstruction of a comprehensive network covering Syk signaling in breast cancer cells. To generate in silico hypotheses on Syk signaling propagation, we developed a method allowing to rank paths between Syk and its targets. We first annotated the network according to experimental datasets. We then combined shortest path computation with random walk processes to estimate the importance of individual interactions and selected biologically relevant pathways in the network. Molecular and cell biology experiments allowed to distinguish candidate mechanisms that underlie the impact of Syk on the regulation of cortactin and ezrin, both involved in actin-mediated cell adhesion and motility. The Syk network was further completed with the results of our biological validation experiments. The resulting Syk signaling sub-networks can be explored via an online visualization platform., Author summary The complex nature of cancer hampers traditional biological approaches to unravel its molecular mechanisms and develop targeted drug therapies. Cancer affects a number of “hallmark” cellular processes controlled by multiple signaling pathways. Our goal is to identify the pathways that negatively affect tumor development and progression. We established that the Syk protein tyrosine kinase exhibits a tumor-suppressive function in breast cancer. Large scale global biochemical analyses allowed to identify Syk targets in cancer cells, but their mechanisms and interrelationships remain unknown. Our main goal was to pinpoint a limited number of biologically realistic molecular “paths” from Syk to its effectors. We therefore developed a new methodology combining graph theoretical methods allowing to reveal the shortest “paths” between “nodes” in a graph including an approach that investigates also longer “paths”. Applied to the Syk network, this method allowed us to propose and validate new signaling axes relating Syk to major effectors of the cell adhesion and mobility that are crucial cancer hallmarks.

Hypertrophic Obstructive Cardiomyopathy (HOCM) prevalence is about 1 among 500 population. It is caused by genetic malformation. Dyspnoe, angina pectoris and syncope are clinical manisfestation. Pharmacological and non-pharmacological treatments can be performed to relieve the obstructive symptoms. Recently alcohol septal ablation has been proven improve haemodynamic and functional status.We report a case of HOCM patient with refractory lung oedema and intubated due to respiratory failure that was immediatelly improved afterseptal ablation.

Moderate-to-high intensity of exercise training within 2 to 3 months decreases oxygen free radicals (reactive oxygen species, ROS) and increases nitric oxide (NO) in outpatients with myocardial infarction. There is no data about the association of ROS and NO after short-term low-intensity exercise training within 5 days in patients hospitalized with acute myocardial infarction (AMI). A total of 32 male patients with AMI were randomized into two groups: 15 patients with short-term low-intensity exercise training within 5 days formed the training group and 17 patients without such exercise training formed the control group. All patients performed exercise treadmill test with modified Bruce protocol before and after the study. F2-isoprostane and NO concentration of the training group increased slightly after modified Bruce exercise treadmill test. Compared with the control group, NO of the training group was also slightly higher. Baseline NO and uric acid were negative predictor variables for F2-isoprostane in all patients hospitalized with AMI, and triglyceride was a positive predictor variable. After the study, physical capacity of the training group was higher; but heart rate and systolic blood pressure were lower significantly. This study showed that short-term low-intensity exercise training for patients hospitalized with AMI did not change ROS and NO productions, but it improved physical capacity and lowered heart rate and systolic blood pressure. NO was negative predictor variable for F2-isoprostane in controlling ROS changes in dynamic compensation mechanism.

Known amounts of radioactivity, across 8 logs of specific activity, were counted in a scintillation counter. The same amount of radioactivity was spotted on a nylon membrane and exposed to a phosphorimager screen for various exposure times. Mathematical models displaying travelling waves appear in a variety of domains. These waves are often faced with different kinds of perturbations. In some cases, these perturbations result in propagation failure, also known as wave-blocking. Wave-blocking has been studied in the case of several specific models, often with the help of numerical tools. In this thesis, we will display a technique that uses symmetry and a center manifold reduction to find a criterion which defines regions in parameter space where a wave will be blocked. We focus on waves with low velocity and small symmetry-breaking perturbations, which is where the blocking initiates; the organising center. The range of the tools used makes the technique easily generalizable to higher dimensions. In order to demonstrate this technique, we apply it to the bistable equation. This allows us to do calculations explicitly. As a result, we show that wave-blocking occurs inside a wedge originating from the organising center and derive an expression for this wedge to leading order. We verify our results with some numerical simulations.

We consider two parameter families of differential equations on a Banach space X, where the parameters c and $\epsilon$ are such that: (1) when $\epsilon=0$, the differential equations are symmetric under the action of the group of one-dimensional translations SE(1) acting on X, whereas when $\epsilon\neq 0$, this translation symmetry is broken, (2) when $\epsilon=0$, the symmetric differential equations admit a smooth family of relative equilibria (travelling waves) parametrized by the drift speed c, with $c=0$ corresponding to steady-states. Under certain hypotheses on the differential equations and on the Banach space X, we use the center manifold theorem of Sandstede, Scheel and Wulff to study the effects of the symmetry-breaking perturbation on the above family of relative equilibria. In particular, we show that the phenomenon commonly referred to as propagation failure, or wave blocking occurs in a cone in the $(c,\epsilon)$ parameter space which emanates from the point $(c,\epsilon)=(0,0)$. We also discuss how our methods can be adapted to perturbations of parameter-independent differential equations (such as the Fisher-KPP) which admit families of relative equilibria parametrized by drift speed., Comment: 30 pages, 7 figures

Succinct Report on the Analysis of the AISLF Congress (Tour 2004): Following the line of work of the January 2005 issue of the BMS (n. 85, based on the analysis of the abstracts of the February 2004 Association Française de Sociologie Congress), we try to demonstrate the optimal use that can be made of text analysis software in human sciences. But in this case, we use the reports from the most recent meeting ot the Association Internationale des Sociologues de Langue Française (AISLF) in July 2004. We use the ALCESTE software in a first step to extract a maximum of “terminal”lexical classes (15 in this case). The second step uses the stages of ALCESTE’s hierarchically descending classification analysis to examine the possible origins of these successive partitions. Dans la ligne de ce que visait le numéro de janvier 2005 du BMS (n. 85, à partir des résumés du congrès de l’Association Française de Sociologie - février 2004 ), on cherche dans cet article à optimiser l’usage qu’on peut avoir, en sciences humaines, des logiciels d’analyse textuelle. International audience; CDP-diacylglycerol synthase (CDS) is a key rate-limiting enzyme in the phospholipid metabolism of Plasmodium falciparum, converting phosphatidic acid to CDP-diacylglycerol. The CDS gene is predominantly expressed in the mature intraerythrocytic stages. Consequently, we physically and functionally characterized the CDS gene promoter. The mRNA transcription initiation site was mapped 121 bp upstream of the CDS gene translation start site. A 1909 bp 5' upstream sequence was isolated and found to be transcriptionally active thus constituting a functional CDS promoter. Mapping of this promoter identified a 44 bp cis-acting sequence, located between -1640 and -1596 bp upstream of the ATG codon, essential for efficient transcriptional activity. This 44 bp sequence binds specifically to nuclear factors from trophozoite stage parasites. We further showed that a 24 bp element, lying within the 44 bp sequence, mediates the specific binding to nuclear proteins and shows no significant homology to known eukaryotic DNA consensus sequence elements that bind transcription factors. The deletion of the 24 bp element abrogated promoter activity, indicating that this cis-acting sequence element is essential for efficient transcription of the CDS gene.
