Mitochondrial dysfunction, oxidative stress, inflammation and glucose dysmetabolism are pathological signs of Alzheimer's disease (AD). Dietary aluminum (Al) overload is often used to induce AD in rodents and trigger the onset of oxidative-stress hallmarks resembling those of the human disease. The Nuclear factor erythroid 2–related factor 2 (Nrf2), owing to its key role in redox homeostasis, mitochondrial function and inflammation, is a promising drug target for neurological disorders, but only a few data are available on its modulatory effects on glucose transporter expression levels. While it has been found that the protective effect of Conjugated linoleic acid (CLA) occurs through the activation of an Nrf2-mediated adaptive response, its beneficial effect on the considered pathological signs in the Al-induced model has not been established yet. Thirty-five male BalbC mice were divided into 5 groups: two Al-intoxicated groups were treated for 5 weeks with low or high Al doses (8 or 100 mg/kg/day in drinking water, respectively; L or H). Two groups of animals, orally supplemented with CLA (600 mg/kg bw/day) for 7 weeks (2 preliminary weeks plus the 5-week treatment with Al; CLA + L, CLA + H) were used to investigate its protective effect, while untreated mice were used as control (Cntr). We provide evidence that mitochondrial dysfunction, Nrf2 alteration, inflammation and Acetylcholinesterase (AChE) hyperactivation can occur even from L exposure. Interestingly, animal pre-treatment with an allometric CLA dose led to significant downregulation of the toxic effects elicited by L or H, likely through the activation of an adaptive response. In conclusion, CLA ability to increase the level of glucose transporters - along with its antioxidant and anti-inflammatory effect - expands the therapeutic targets of these molecules and comes out as an intriguing suitable candidate for the treatment of multifactorial disease.

Oogenesis was examined in nine species of Antarctic fish to verify the existence of morphological peculiarities. The analyses were carried out on specimens belonging to three different families of Notothenioids (Nototheniidae, Channichthyidae and Bathydraconidae), all captured in the Ross Sea, in front of the Italian Station of Terra Nova Bay. Following dissection, the ovaries were processed and examined at the light and electron microscopes to determine the oocyte gross and fine morphology. The attention, in particular, was focused on the presence of cytoplasmic round bodies and on the organization of the cortical alveoli and the vitelline envelope. Results reveal significant specie-specific differences that could be partly correlated to the phylogenetic radiation but not to the peculiar environmental conditions being essentially comparable to those observed among temperate species.

Various in vitro and in vivo studies have highlighted the role of epigenetics on chromatin conformation and gene expression occurring upon neural development. KRAB-containing zinc finger protein ZFP57 is involved in the maintenance of parent-of-origin-dependent epigenetic state, DNA methylation and histone-tail-specific modifications of hundreds of imprinted loci in mouse. ZFP57 associates with KAP1 (general co-repressor of the KRAB-ZFPs) that acts as a recruiter of chromatin modifiers including histone H3 methyltransferase SETDB1 and heterochromatin protein-1(HP1), thus promoting H3 trimethylation at lysine9 and chromoprotein recruitment at targeted sites. DNA methyltransferases are also present in KAP1 complexes. This finding correlates with the requirement for ZFP57 to maintain DNA methylation at imprinting control regions (ICRs). This study focuses on the role of ZFP57/KAP1 in imprinting maintenance and epigenome regulation during neurogenesis and differentiation in a mouse model. We have established the genome wide occupancy of ZFP57 and associated chromatin modifiers KAP1, Hp1g and H3K9me3 and correlated with DNA methylation in murine embryonal stem cells (ESC). ZFP57, while strongly expressed in ESCs and down regulated during differentiation, was observed to be highly expressed during embryogenesis notably in the developing brain. The strong and sustained expression in the nervous system of the developing murine embryo is related to its association to ICRs, extending its potential role beyond that of maintenance of imprints during pre-implantation. We report data regarding initial studies detailing the influence and dynamics of ZFP57/KAP1 epigenetic/omic dependent signature and gene expression studies on ESCs (undifferentiated and differentiated cells toward neural fate) and developing/post-natal brain.

Several lines of evidence have provided clues for the pivotal role played by the KRAB-containing zinc finger protein ZFP57 in the maintenance of the parent of origin-dependent epigenetic state, DNA methylation and histone tails specific modifications, of the hundreds imprinted loci in mouse which include several non coding RNAs. ZFP57 associates with KAP1, a general co-repressor of the abundant KRAB-ZFPs family, which acts as a scaffold for the recruitment of chromatin modifiers that include the histone H3 methyltransferase SETDB1 and heterochromatin protein-1 (HP1) isoforms, thus promoting H3 trimethylation at lysine9 and chromoproteins recruitment at targeted sites. DNA methyltransferases are also present in KAP1 complexes. This finding correlates with the requirement for ZFP57 to maintain DNA methylation at imprinting control regions (ICRs). In this work we established the genome wide occupancy of ZFP57, along with associated chromatin modifiers KAP1, Hp1 and tri-methyl H3K9 and correlated it with the DNA methylation profile in murine embryonal stem cells (ESC). The data show that the majority of ZFP57 bound sites reside at DNA methylated non-imprinted loci, of which several encompass non-coding RNAs, implying a wider role for this factor in the shaping of the epigenome, notably at germ-line DNA methylated CpG islands and repeated retrotrasposons. Further, we show that ZFP57, while strongly expressed in ES cells and down regulated during differentiation, is also expressed at several sites during embryo development. The strong and sustained expression in the nervous system of the developing murine embryo is paralleled by association of the factor to ICRs extending its potential role beyond that of imprints maintenance at pre-implantation. We will report also on data suggesting a potential role for ZFP57 in the in vitro differentiation of ES cells toward the neural fate.

Glucose-6-phosphate dehydrogenase-deleted embryonic stem (ES) cells (G6pdD) proliferate in vitro without special requirements, but when challenged with oxidants fail to sustain glutathione disulphide reconversion to reduced glutathione (GSH), entering a condition of oxidative stress. Here, we investigate the signalling events downstream of GSH oxidation in G6pdD and wild-type (wt) ES cells. We found that G6pdD ES cells are very sensitive to oxidants, activating an apoptotic pathway at oxidant concentrations otherwise sublethal for wt ES cells. We show that the apoptotic pathway activated by low oxidant concentrations is accompanied by mitochondria dysfunction, and it is therefore blocked by the overexpression of Bcl-XL. Bcl-XL does not inhibit the decrease in cellular GSH and reactive oxygen species formation following oxidant treatment. We also found that oxidant treatment in ES cells is followed by the activation of the MEK/ extracellular signal-regulated kinase (ERK) pathway. Interest¬ingly, ERK activation has opposite outcomes in G6pdD ES cells compared to wt, which has a proapoptotic function in the first and a prosurvival function in the latter. We show that this phenomenon can be regulated by the cellular GSH level.

Mouse embryonic stem (ES) glucose-6-phosphate (G6P) dehydrogenase-deleted cells (G6pd Delta), obtained by transient Cre recombinase expression in a G6pd-loxed cell line, are unable to produce G6P dehydrogenase (G6PD) protein (EC 1.1.1.42). These G6pd Delta cells proliferate in vitro without special requirements but are extremely sensitive to oxidative stress. Under normal growth conditions, ES G6pd Delta cells show a high ratio of NADPH to NADP+ and a normal intracellular level of GSH. In the presence of the thiol scavenger oxidant, azodicarboxylic acid bis[dimethylamide], at concentrations lethal for G6pd Delta but not for wild-type ES cells, NADPH and GSH in G6pd Delta cells dramatically shift to their oxidized forms. In contrast, wild-type ES cells are able to increase rapidly and intensely the activity of the pentose-phosphate pathway in response to the oxidant. This process, mediated by the [NADPH]/[NADP+] ratio, does not occur in G6pd Delta cells. G6PD has been generally considered essential for providing NADPH-reducing power. We now find that other reactions provide the cell with a large fraction of NADPH under non-stress conditions, whereas G6PD is the only NADPH-producing enzyme activated in response to oxidative stress, which can act as a guardian of the cell redox potential. Moreover, bacterial G6PD can substitute for the human enzyme, strongly suggesting that a relatively simple mechanism of enzyme kinetics underlies this phenomenon.

More than 80 genetic variants of glucose-6-phosphate dehydrogenase (G6PD) are associated with chronic non-spherocytic haemolytic anaemia (CNSHA). In order to help clarify the molecular basis of this association, we have carried out a detailed biochemical and genetic characterization of two G6PD deficient brothers affected by CNSHA. The G6PD from the two patients has altered electrophoretic mobility, abnormally elevated Michaelis constant (Km) for G6P, and extreme instability in vivo and in vitro. By comparison with published information we found that this is a new G6PD variant which we have designated G6PD Portici. The entire coding region of the gene has been sequenced, and a single point mutation, a G----A transition, was found at position 1178 in exon X, causing a substitution of histidine for arginine at residue 393 in the polypeptide chain. By polymerase chain reaction (PCR) amplification followed by diagnostic restriction enzyme analysis and allele-specific oligonucleotide hybridization we have demonstrated the inheritance of this mutation in the patient's family. Our results support the notion of a causative link between this mutation in the G6PD gene and CNSHA. Our data, in combination with previous data in the literature, suggest that the three-dimensional structure of G6PD is such as to cause interaction in the binding of its two substrates, G6P and NADP.

To elucidate the molecular basis of G6PD deficiency in the Han and Li nationalities in Hainan, China.Polymerase chain reaction and restriction enzyme digestion were used to screen the mutations 1388G--A, 1360C--T, 1024C--T, 592C--T, 517T--C, 493A--G, 487G--A, 392G--T and 95A--G. Single strand conformation polymorphism analysis was used to screen the other mutations followed by DNA sequencing to characterize the mutations of the samples with abnormal SSCP bands.Of the fifty-nine Han cases with G6PD deficiency, fourteen with 1388G--A (23.7%), three with 871G--A(5.1%), one with 835A--T(1.7%), one with 517T--C (1.7%), three with 392G--T(5.1%), and four with 95A--G(6.8%) were found. Of the thirty-two Li cases with G6PD deficiency, six with 1388G--A(18.8%), three with 871G--A(9.4%), and two with 95A--G(6.3%) were found. A new mutation 835A--G which causes the substitution of Ala for Thr at 279 in a Han case was identified and named as G6PD Haikou. The enzyme activity of the variant is about 10% of the normal and lower than the activity of the variant 835A--T with about 40% of the normal. Analysis of the 3D model of human G6PD has revealed that the hydroxyl group of Thr at 279 is a group in maintaining the interaction of the G6PD subunits.The most common mutations of G6PD deficiency in Han and Li nationalities in Hainan are similar. Compared with the mutation spectrum of G6PD gene in the populations in other regions of China, the results indicate that some G6PD gene mutations are widespread in the populations of different regions in the southern part of China. The hydroxyl group of the Thr at 279 of human G6PD may be a necessary group for maintaining the interaction of the G6PD subunits and the enzyme activity.

In the present study we analyse the nature and the functional significance of the spherical and fibrillo-granular structures appearing in the oocyte nucleus of the lizard Podarcis sicula, following the disappearance of the typical nucleolus. By LM and TEM approaches, we demonstrate that the fibrillo-granuli, containing DNA, RNA and nucleolar proteins, are micronucleoli transcriptionally active and that their DNA is probably derived from nucleolar fragmentation. By contrast, we could not explain the origin and role of the so-called spherical bodies, appearing earlier in oocyte growth; these, in fact, do not contain nucleic acids or nucleolar proteins and do not incorporate uridine. Different possible explanations of their significance are discussed.

The isolation of effective probes is of paramount importance in studies aimed at elucidating the molecular mechanisms underlying the development of ectomycorrhizal fungi, as well as in the creation of biotechnological tools for improving truffle harvest and species identification. In one strategy which we are using to isolate DNA probes, recombinant plasmids are randomly isolated from genomic libraries and their nucleotide sequences compared with available data banks of nucleotide and aminoacid sequences. As a first step in this strategy, we have analyzed 56 clones from a genomic library of plasmids bearing small inserts of DNA from Tuber albidum. Several clones were singled out on the basis of sequence similarity with known genes. Among these was a recombinant plasmid carrying nucleotide sequences potentially coding for a protein extremely similar to the a subunit of Fatty Acid Synthase of other fungal species. The potentially coding region appeared to be interrupted by an intron and its sequence has allowed the construction of an evolutionary tree including Tuber magnatum and three different fungal species.

During hibernation, ribosomal bodies are present in the germ cells of Lacerta sicula , which have undergone meiotic prophase, but neither in oogonia nor in stroma cells. These bodies are similar in structure, although smaller in size, to those which have been described in the cytoplasm of growing oocytes; in the germ cells at zygopachytene, they are frequently associated with the nuclear envelope.

The follicular epithelium of the lizard oocytes undergoes structural and morphological modifications throughout oocyte growth. During this process the number of follicle cells increases and the epithelium acquires a multilayered and polymorphic organization which is characterized by the appearance of large follicle cells (intermediate and pyriform’cells). The number of large cells also increases during oocyte growth and this increase parallels that of small cells. However, only the small cells become labelled one hour after [3H-]thymidine administration. Large cells have been found labelled after a longer period of time, i.e. 4–5 months after isotope injection. All these results together indicate that large follicle cells arise from the differentiation of small cells.

Intercellular bridges first appear during lizard oogenesis when follicles are rather small (150 pm in diameter); at this stage they form connecting links between the oocyte and follicle cells, which have not yet differentiated into pyriform cells. Later on, when the follicles have become larger (1 mm) and the follicular epithelium appears constituted by 3 types of cells (small, intermediate and pyriform cells) they form connecting links between the oocyte and both intermediate and pyriform cells. The establishment of intercellular bridges between pyriform cells and the oocyte precedes the complete differentiation of the former, which excludes the possibility that the fusion between pyriform cells and oocyte occurs only after these cells are completely differentiated. In still larger follicles (up to 2 mm in diameter), during the degeneration of the pyriform cells, the occurrence, inside the bridges, of mitochondria and other cytoplasmic material suggests that these cells at the end of their function transfer their contents into the oocyte.

In the present paper we investigated the role played by apoptosis during oogenesis in the cartilagineous fish Torpedo marmorata. TEM, TUNEL and immunohistochemical techniques were employed to specifically reveal morphological and biochemical hallmarks of apoptosis in specimens from birth to sexual maturity. Data obtained demonstrate that apoptosis occurs in prefollicular oocyte selection, in maintaining the homeostasis of granulosa in healthy growing oocyte and in resorbing atretic follicles. In this respect, the involvement of apoptosis in Torpedo marmorata oogenesis closely parallels that found in mammals, thus confirming that strategies of germ cell selection among vertebrates have been evolutively preserved. In the present report, we investigated zinc, copper and metallothionein (MT) contents in zebrafish oocytes and embryos. Our results demonstrate that the metal content increases during oocytes maturation. Zinc increases from 30 ng/oocyte (stage-1 oocytes) to 100 ng/oocyte (stage-3 oocytes); copper varied from 1 ng/oocyte (stage-1 oocytes) to 3.5 ng/oocyte (stage-3 oocytes). During embryogenesis, zinc and copper contents dramatically increase after fertilisation around the 512-cells stage, then slowly decrease until the mid-gastrula stage. During oocyte growth, the changes in the MT level are proportional to metal content, whereas during embryogenesis the pattern of MT accumulation does not parallel that of the two metals. Indeed, the maternal pool of MT decreases steadily during the early stages of the development until the gastrula stage. We have examined the effect of cadmium on the expression of MT during zebrafish development. After cadmium exposure, MT content increases in embryos at the blastula stage, whereas no induction occurs in embryos at the gastrula stage. However, pre-treatment of embryos at the gastrula stage with 5-aza-2'-deoxycytidine induces MT synthesis following exposure to cadmium. These observations show that changes in metal levels are not correlated to MT content in the embryo, whereas DNA methylation is one of the factors regulating MT expression.

Copper (Cu) is an essential element required in many biological processes including cellular growth and development. The molecular mechanisms involved in copper homeostasis include proteins that play a role in Cu uptake. Genes encoding high affinity copper transporters (Ctr) have been identified in yeast, plant and mammalian cells. Analysis of copper and zinc content in growing ovarian follicles and ovulated eggs of the reptilian Podarcis sicula demonstrated that the levels of both metals rise during oocyte growth, reaching the maximum in ovulated eggs. By exploiting the remarkable evolutionary conservation of the primary structure of Ctr proteins, cDNA encoding a Ctr was isolated from the liver of the lizard P. sicula by reverse transcriptase PCR and RACE strategy by using primers designed based on consensus motifs present in mammalian Ctr. The predicted protein sequence contains three transmembrane domains and a putative hydrophilic extracellular amino-terminal domain. Besides complementing the respiratory deficiency of yeast cells defective in high affinity Cu transport, expression of lizard Ctr1(1) in Hek293 cells stimulates Cu uptake.Gene expression assessed by Northern blot hybridization of RNA from different tissues of P. sicula shows the highest levels of transcript in both intestine and liver. The profile of Ctr1 mRNA in growing ovarian follicles and eggs demonstrates that the transcript accumulates during the oocyte growth and reaches the highest levels in ovulated eggs. These results suggest that lizard Ctr1 protein may function in Cu acquisition in growing oocytes and eggs.

We have carried out a systematic study of the molecular basis of glucose-6-phosphate dehydrogenase (G6PD) deficiency on a sample of 53 male subjects from Calabria, in southern Italy. Our sequential approach consisted of the following steps: (1) Partial biochemical characterization was used to pinpoint candidate known variants. The identity of these was then verified by restriction-enzyme or allele-specific oligonucleotide hybridization analysis of the appropriate PCR-amplified fragment. (2) On samples for which there was no obvious candidate mutation, we proceeded to amplify the entire coding region in eight fragments, followed by single-strand conformation polymorphism (SSCP) analysis of each fragment. (3) The next step was M13 phage cloning and sequencing of those individual fragments that were found to be abnormal by SSCP. Through this approach we have identified the molecular lesion in 51 of the 53 samples. In these we found a total of nine different G6PD-deficient variants, five of which (G6PD Mediterranean, G6PD A-, G6PD Coimbra, G6PD Seattle, and G6PD Montalbano) were already known, whereas four are new (G6PD Cassano, G6PD Cosenza, G6PD Sibari, and G6PD Maewo). G6PD Mediterranean is the commonest variant, followed by G6PD Seattle. At least seven of the variants are present, at polymorphic frequencies, in the Calabria region, and some have a nonrandom distribution within the region. This study shows that the genetic heterogeneity of G6PD deficiency in Calabria, when analyzed at the DNA level, is even greater than had been anticipated from biochemical characterization. The sequential approach that we have followed is fast and efficient and could be applied to other populations.

Glucose-6-phosphate dehydrogenase (G6PD) has been analyzed by gel electrophoresis and by quantitative assay in an unselected sample of 1524 schoolboys from the province of Matera (Lucania) in southern Italy. We have identified 43 subjects with a G6PD variant. Of these, 31 had severe G6PD deficiency, nine had mild to moderate deficiency, and three had a non-deficient electrophoretic variant. The overall rate of G6PD deficiency was 2.6%. The frequency of G6PD deficiency, ranging from 7.2% on the Ionian Coast to zero on the eastern side of the Lucanian Apennines, appears to be inversely related to the distance of each town examined from the Ionian Coast, suggesting that this geographic distribution may reflect, at least in part, gene flow from Greek settlers. Biochemical characterization has shown that most of the G6PD deficiency in this population is accounted for by G6PD Mediterranean. In addition, we have found several examples of two other known polymorphic variants (G6PD Cagliari and G6PD A−): three new polymorphic variants. G6PD Metaponto (class III), G6PD Montalbano (class III), and G6PD Pisticci (class IV); and two sporadic variants, G6PD Tursi (class III) and G6PD Ferrandina (class II). These data provide further evidence for the marked genetic heterogeneity of G6PD deficiency within a relatively narrow geographic-area and they prove the presence in the Italian peninsula of a sene (Gd A−) regarded as characteristically African.
